The commercial Zantedeschia industry faces a potentially serious problem with the cultivar Florex Gold periodically producing weak, poor performing “bushy” plants in their second growing season. As these plants are derived from tissue culture, it had been suggested that the use of cytokinin during the tissue culture process might be a causative factor. Endogenous cytokinin concentration and profile in dormant and sprouting Zantedeschia tuber eyes (buds) of the cultivars Florex Gold (prone to bushiness) and Best Gold (not prone to bushiness) were determined by high-performance liquid chromatography and radioimmunoassay. During sprouting the total cytokinin concentration in Florex Gold tuber eyes rose tenfold, with a 40-fold increase in the nucleotides. No role for endogenous cytokinin concentration or profile in the expression of the bushy syndrome was established, as there were no measurable differences between bushy and control Florex Gold samples, whether dormant or sprouting. However, sprouting tuber eyes from the cultivar Florex Gold had nearly twofold more cytokinin than sprouting eyes from the cultivar Best Gold, which may in some way predispose Florex Gold to bushiness.  相似文献

One-year old tubers of two hybrid calla lily (calla) cultivars (Zantedeschia ‘Pot of Gold’ and ‘Majestic Red’) were inoculated with the arbuscular mycorrhizal fungus (AMF), Glomus intraradices, or not, and grown at three different rates of phosphorus (P) supply to asses the effects of AMF-inoculation on plant development (time of shoot emergence and flowering), flowering (number, length and rate of flowering), and tuber biomass and composition over two growing cycles (2002, 2003). Tubers and flowers of calla responded differently to AMF inoculation. Differences in mycorrhizal responsiveness between cultivars was related to differences in P requirements for flower and tuber production, and the influence of P supply on resource allocation to different reproductive strategies. Inoculation increased shoot production and promoted early flowering, particularly in 2003. Inoculated plants also produced larger tubers than non-inoculated plants, but only increased the number of flowers per plant in 2003. High P supply also increased tuber biomass, but decreased the number of flowers per plant in 2002. Plants grown at a moderate P-rate, produced the most flowers in 2003. For ‘Majestic Red’, benefits from AMF were primarily in terms of tuber yield and composition, and AMF effects on marketable flower production could potentially have negative impact on production strategies for growers. Inoculation of ‘Pot of Gold’ primarily influenced flower production and aspects of tuber quality that caused detectable enhancement of tuber yield and flowering in the second growing cycle following inoculation (2003). The results of this study show that the responses of calla to AMF are partially a function of how nutrient supply alters resource allocation to sexual and vegetative reproduction. Whether AMF-induced changes in resource allocation to flowering and tubers significantly alters commercial productivity and quality of calla depends on the crop production goals (e.g. tubers, cut flowers or potted plants). Bushiness in Gerbera is a major problem causing more than 30% loss of production during micropropagation and greenhouse production. We studied two Gerbera cultivars `Rosabella' (bushy) and `Sunset' (non-bushy). Three different types of inocula (main, adventitious and axillary shoots) were investigated. The choice of the inoculum had an important influence on bushiness, the main axis being the most sensitive. Higher KIN concentrations enhanced bushiness. Addition to the fungicide imazalil to the propagation medium reduced bushiness. The appropriate concentration of imazalil depended on the degree of bushiness and the inoculum type.  相似文献

In transplants from in vitro‐derived plantlets from very early potato (Solanum tuberosum) cultivars, a lower degree of tuber induction at the time of field planting is thought to increase tuber production. Leaf‐bud cuttings were used to assess the progress to tuber induction in in vitro‐derived potato plantlets during the transplant production phase and after subsequent transplanting into the field. Induction and initiation of tubers on the same plants were assessed to study the effects of the duration of transplant production and conditions during transplant production for cv. Gloria (very early) and cv. Bintje (mid‐early). In vitro‐produced plantlets were not induced by the time of planting but rapidly progressed to the induced state thereafter. The progress in induction with time and the change in percentage of plants showing tubers fitted typical sigmoid curves. Plantlets achieved 50% induction ca 15 days after planting into in vivo conditions, and 50% tuber initiation usually occurred 10 days later. Shorter transplant production periods reduced the degree of induction of the transplants at field planting. Transplant production for more than 2 weeks was required to allow conditions during that period to affect induction or initiation. Long‐term non‐inducing conditions delayed the progress to tuber induction in cv. Gloria and delayed tuber initiation in both cultivars. Cv. Gloria showed no faster progress to induction than cv. Bintje but initiated tubers earlier. The results suggest that the relation between progress to induction and tuber initiation is cultivar dependent and that leaf‐bud cuttings can be used successfully in very young in vitro‐derived plants for assessing the progress to tuber induction.  相似文献

The effects of the plant growth retardant tetcyclacis on in vitro tuber formation in potatoes was studied, using two different approaches: 1. tuber formation in various lines that did not or hardly form tubers under control conditions, and 2. tuber formation by the variety Bintje, which readily forms tubers. The ABA-deficient (droopy) lines of S. phureja hardly formed tubers without the addition of tetcyclacis. In the presence of this growth retardant tuberization was nearly 100%, within three weeks of in vitro culture, even in the absence of cytokinin. A series of somatic hybrids between S. tuberosum and S. brevidens, that did not form tubers in field and pot experiments, were tested. They all formed tubers in vitro in the presence of tetcyclacis. Stoloniferous shoots formed on single-node cuttings from in vitro grown Solanum tuberosum var Bintje plantlets were transferred to media containing a high level of sucrose. In the presence of tetcyclacis, tuber formation started after 4 days, reaching a maximum level of 80% at day 7. Tubers formed in the presence of tetcyclacis, accumulated starch and expressed several tuber-specific genes. These effects were fully antagonized by gibberellic acid. Due to its high commercial value, many studies on rose (Rosa hybrida L.) micropropagation have been published. However, there are a limited number of studies on rose in vitro flowering. These studies only focused on the roles of plant growth regulators in the formation and morphogenesis of flowers. In this protocol, cytokinin was confirmed to positively function in the induction of in vitro rose flowers. In fact, more than 40% of in vitro shoots were induced to flower when they were grown on a medium supplemented with benzylaminopurine (BA) (2 mg L−1) and IAA (0.1 mg L−1). In addition, this study showed that the growth medium supplemented with only coconut water (15 or 20% v/v) was very efficient to induce flowering of in vitro miniature rose plants (> 70%) after 60 d of subculture. In addition, the in vitro flowers were normal and almost similar to ex vitro flowers in terms of flower shape and color. Based on these results, a detailed procedure for in vitro miniature rose flower production is provided.

In water-culture experiments with potato plants (Solanum tuberosum L. cv. Ostara), changes in cytokinin activity in the stolon tips and newly formed tubers during tuberization were studied. Tuberization was induced by withdrawing nitrogen from the nutrient solution. — The cytokinin activity was low in the stolon tips prior to tuberization, but increased considerably in both stolon tips and young tubers during tuberization. At the same time qualitative changes in the cytokinin spectrum occurred. These qualitative changes are reversible if ‘regrowth’ of young tubers is brought about by a sudden high supply of nitrogen. — Despite the close correlation between tuberization and cytokinin activity, it is assumed that cytokinins are not directly responsible for the onset of tuberization, although they play an important role in tuber growth.  相似文献

Hormonal control of flower induction and inflorescence development in vitro was investigated in photoperiodically day-neutral calla lily (Zantedeschia spp., colored cultivars). The effects of gibberellins (GAs, 5.8–2900 M) and the cytokinin benzyl adenine (BA, 0.4–13.3 M) on inflorescence development were studied in plantlets regenerated in tissue culture. Plantlets were dipped in GA and BA solutions prior to replanting in new media. GA was mandatory for the shift from the vegetative to the reproductive stage. GA3, GA1 and GA4 had the same florigenic effect. Inflorescence development in the apical bud was observed after 30–50 days in GA-treated plantlets grown in vitro and resembled the pattern occurring under natural conditions. The transition from the vegetative to the reproductive phase was characterized by a swollen, dome-shaped apex that transformed into a smooth elongated apex surrounded by the spathe primordium, at the tip of the elongating peduncle primordium. Floret primordia developed in inflorescences at a more advanced stage. The female florets located at the base of the primordial spadix, could be clearly distinguished from male florets located above them. BA did not have an effect on flower induction but, in the presence of GA, BA at concentrations up to 4.4 M enhanced inflorescence differentiation. The results indicate that inflorescence development in Zantedeschia plantlets in tissue culture can serve as a potential model to study the role of GAs and other factors in the flowering process of day-neutral plants that do not require external signals for flower induction.  相似文献

A method for regeneration of plants from tuber explants of a Zantedeschia hybrid via somatic embryogenesis was developed. In vitro cultures were initiated starting from both anthers and tubers. Somatic embryogenesis was only achieved from tuber explants. 6-Benzyladenine (BA) at 0.6 or 2 mg l−1 in combination with 2 mg l−1 α-naphthaleneacetic acid (NAA) yielded the highest number of embryos per explant. The somatic embryos converted into plantlets on Murashige and Skoog basal medium supplemented with vitamins, micro- and macronutrients, 1 mg l−1 6-τ-τ-(dimethylallylamino)-purine (2iP), 3% sucrose and 0.7% agar. This is the first report on induction of somatic embryogenesis in Zantedeschia.  相似文献

In order to establish and optimize an in vitro micropropagation method of Drosera peltata (a tuberous sundew), a carnivorous plant, the effects of medium type, MS medium concentration, pH and cytokinin type on shoot proliferation and tuber formation were investigated, using one month-old shoot tips. The shoot proliferation and tuber formation were most effective on 1/2 MS medium without cytokinins. The optimum pH of the media was pH 5.7. Tubers were planted in plastic pots filled with 1:1 peat moss and sand. The survival rate of the plantlets was almost 100%, and they exhibited normal development. With subculture every 12 weeks, hundreds of the plants were propagated from a single plant within a year.  相似文献

The morphological, cytological and molecular analyses of the plants regenerated after PEG-induced fusion between mesophyll protoplasts from the dihaploid potato clone H-8105 and the wild tuberous disease-resistant species S. pinnatisectum, were performed. A single fusion experiment yielded 313 calli, although only two calli produced shoots. From the rooted shoots, two stable clones (PT-01-1 and PT-01-2) exhibiting different vigor and habitat, were developed. The plants of PT-01-1 clone grew slowly in vitro, produced tubers after transfer to soil but did not set flowers. In contrast, the plants of the vigorous clone PT-01-2 produced both tubers and flowers after transfer to soil. The flower and tuber morphology of PT-01-1 and PT-01-2 regenerants was intermediate in comparison to the parental species. Cytological analysis revealed that the PT-01-1 clone was pentaploid and the PT-01-2 clone was tetraploid. The molecular (RAPD) analysis confirmed hybridity of both clones. The preliminary tests on late blight resistance of the hybrids showed no differences with a potato parent.  相似文献

Phenologic changes and variation in the level of endogenous gibberellins (GAs), abscisic acid (ABA), carbohydrate content, and α-amylase activity were examined in colored Zantedeschia spp. cv. Cala Gold. These changes were examined in the primary bud tissues and in the attached tuber tissue during the growth cycle. Dormant tubers were dry-stored at 20°C for 3 months, planted in a phytotron, and grown under 22/16 ± 1°C. Plant development was monitored under continued irrigation until leaf senescence and tuber dormancy. GAs and ABA were extracted from the primary bud tissues, fractionated by HPLC, and analyzed using GC-SIM. Starch, glucose, soluble protein, and α-amylase activity were monitored in the tuber tissue attached to the primary bud. Endogenous changes in GAs and ABA in the primary bud were correlated with endogenous changes in carbohydrate content and α-amylase activity in the attached tuber tissue. These correlations were observed during the rest and the growth periods and were associated with developmental changes in the plant, that is, bud dormancy relaxation, bud growth, and inflorescence differentiation. ABA content decreased and a transient pulse of GA was measured in the primary bud concomitantly with the onset of shoot elongation in dry tubers during storage, before planting. The sharp increase of GAs in the bud preceded inflorescence differentiation as observed in dissected apices by about 15 days, as well as the increase in α-amylase activity in the attached tuber tissue. A steep decrease in starch level was measured in the tuber after planting, concomitantly with massive plant growth. These findings suggest a possible involvement of gibberellin in the initiation of α-amylase activity during dormancy relaxation in colored Zantedeschia and in the autonomous induction of flowering.  相似文献

We previously obtained somaclonal variants of the important French fry processing cultivar Russet Burbank with significantly enhanced resistance to common scab disease. In this study we have shown the commercial merit of a proportion of these variants through comparison of relative yield and tuber quality with the parent cultivar Russet Burbank. Whilst we showed a weak negative correlation between tuber yield (as assessed by weight of tubers per plant) and relative disease resistance within selected variants, we identified several with equivalent yields to the parent cultivar. Furthermore, two disease-resistant variants (TC-RB8 and NZ-24B) consistently yielded more tuber mass than the parent. The majority of our Russet Burbank variants showed equivalent tuber quality characteristics (occurrence of defects, tuber specific gravity and dry matter content, and flesh colour) and cooking qualities (fry colour and presence of dark end defects) to the parent cultivar. Independent testing by a commercial French fry processor confirmed these quality characteristics. We present data demonstrating that highly common scab disease-resistant somaclonal variants of Russet Burbank have commercially acceptable tuber yield and quality characteristics, comparable to the industry standard and parent Russet Burbank cultivar. We also demonstrate the value of in vitro cell selection techniques for potato cultivar improvement.  相似文献

The restricted flowering of colored cultivars ofZantedeschia is a consequence of developmental constraints imposed by apical dominance of the primary bud on secondary buds in the tuber, and by the sympodial growth of individual shoots. GA3 enhances flowering inZantedeschia by increasing the number of flowering shoots per tuber and inflorescences per shoot. The effects of gibberellin on the pattern of flowering and on the developmental fate of differentiated inflorescences along the tuber axis and individual shoot axes were studied in GA3 and Uniconazole-treated tubers. Inflorescence primordia and fully developed (emerged) floral stems produced during tuber storage and the plant growth period were recorded. Days to flowering, percent of flowering shoots and floral stem length decreased basipetally along the shoot and tuber axes. GA3 prolonged the flowering period and increased both the number of flowering shoots per tuber and the differentiated inflorescences per shoot. Activated buds were GA3 responsive regardless of meristem size or age. Uniconazole did not inhibit inflorescence differentiation but inhibited floral stem elongation. The results suggest that GA3 has a dual action in the flowering process: induction of inflorescence differentiation and promotion of floral stem elongation. The flowering pattern could be a result of a gradient in the distribution of endogenous factors involved in inflorescence differentialtion (possibly GAs) and in floral stem growth. Plants produced vegetatively in tissue culture may differ from the plants from which they have been derived. Two major classes of off-types occur: genetic ones and epigenetic ones. This review is about epigenetic aberrations. We discuss recent studies that have uncovered epigenetic modifications at the molecular level, viz., changes in DNA methylation and alterations of histone methylation or acetylation. Various studies have been carried out with animals, and with plant cells or tissues that have grown in tissue culture but only little work has been done with shoots generated by axillary branching. We present various molecular methods that are being used to measure epigenetic variation. In micropropagated plants mostly differences in DNA methylation have been examined. Epigenetic changes are thought to underlie various well-known tissue-culture phenomena including rejuvenation, habituation, and morphological changes such as flower abnormalities, bushiness, and tumorous outgrowths in, among others, oil palm, gerbera, Zantedeschia and rhododendron.  相似文献

Cassava (Manihot esculenta Crantz), is an important tropical tuber crop with global importance and plays a significant role in the food, nutritional and livelihood security of around 500 million people. In India, the low productivity of cassava attributes to the soil borne disease, particularly tuber rot caused by Phytophthora palmivora (Butl.) which is destructive and the attack is spreading in alarming rate in all the cassava growing regions causing heavy yield loss of more than 50%. Introduction of disease resistant varieties may alleviate the problem to a certain extent. This paper describes the screening procedures and findings on the disease resistant variety of cassava accession against tuber rot. Variety Sree Padmanabha imparted high resistance against tuber rot, while Sree Sahya was moderately resistant and all other accessions studied were found to be susceptible in in vitro and in field trials. In screening studies, a reproducible positive correlation was obtained between attached tubers in live plant with detached tubers which showed that detached tuber part can be used for the prediction of resistance in attached live plants of cassava for cultivar resistance. The procedure described here could be used as a simple, rapid and efficient method for screening of cassava accessions against tuber rot of cassava.  相似文献

Auxins and cytokinins are implicated in a wide variety of developmental and physiological processes in plants. Phytophthora palmivora causes tuber rot in cassava growing regions of Tamil Nadu and Kerala, South India. The in vitro effect of cytokinin, benzyl amino purine (BAP) and auxins, naphthalene acetic acid (NAA) and indole acetic acid (IAA) on P. palmivora mycelium growth was investigated. The inhibitory activity varied among the growth regulators and complete inhibition of the pathogen was observed at 50, 2000 and 2500 ppm by the BAP, IAA and NAA, respectively. The effective growth regulator, BAP was also analysed on tubers before and after the invasion of the pathogen to observe its effect in tuber. Further, it was also checked against the bio-control agent Trichoderma harzianum. The study indicates that the use of BAP could be an important approach in controlling tuber rot pathogen, P. palmivora.  相似文献

The relationships between rain and blight (Phytophthora infestans) were studied in unsprayed crops of cultivars differing widely in foliage and tuber susceptibility. The occasions when tubers were infected depended on rain and not cultivar, but numbers of tubers infected after rain was affected by the blight susceptibility of the cultivar. Infected tubers were first found when less than 5 % (BMS key) of the potato foliage was infected but few fresh infections occurred when 50–75% of the foliage had been destroyed. Some tubers were infected after 8 mm rain (tubers near the surface with even less) but large increases in numbers of tubers infected usually occurred only after 25 mm or more had increased soil moisture to above ‘field capacity’ around the tuber for at least 24 h. The most susceptible cultivars Ulster Ensign and Arran Banner had all plants with some tuber blight, and some plants with all tubers affected and often many lesions per tuber. Cultivars of intermediate susceptibility, King Edward and Up-to-Date, had some plants without blighted tubers, many with a few and very few with all. The more resistant cultivars Majestic and Arran Viking had many plants without infected tubers and many lesions that aborted while still necrotic threads, so that the fungus did not spread. Most infections occurred through tuber eyes, lenticels or sometimes growth cracks. The distribution of blight lesions on tubers differed in the different seasons, for example, lenticels were most commonly infected on Arran Banner and Ulster Ensign and eyes on King Edward, Majestic and Arran Viking. In late or slowly developing attacks, lesions on stems became more numerous and larger than in fast, early attacks and were prolific sources of spores on King Edward and Up-to-Date but not on Majestic and Arran Viking. Because much rain water runs down the stems of Up-to-Date and King Edward, stem lesions can provide an important source of inoculum for tubers.  相似文献

Floral productivity of Zantedeschia is dependent on the conversion rate of buds to shoots, which is controlled by varying intensities of para- (apical dominance), endo- (dormancy), and ecodormancy. We present evidence of cross-talk between cytokinin and gibberellin in their complementary roles to alleviate bud dormancy and enhance flowering in a perennial geophyte. We assessed the impact of cytokinin and gibberellin, applied alone and in sequential combinations, on bud fate during three phases along the ontogeny of growth, which coincide with the progressive transition of buds from apical dominance to dormancy. Given that cytokinin can stimulate branching and gibberellin can induce flowering in Zantedeschia, we measured these phenotypic responses as parameters of bud commitment. The efficacy of cytokinin alone to stimulate branching declined with the transition to dormancy (phase 1 = 3.8 ± 0.2 shoots; phase 3 = 1.0 ± 0.3 shoots). To sustain branching during this transition, a sequential application of gibberellin was necessary. Gibberellin alone failed to stimulate branching. The efficacy of gibberellin alone to stimulate flowering diminished with the transition to dormancy. Any flowering during this transition occurred only after the sequential application of cytokinin. Cytokinin alone failed to stimulate flowering. Alleviating bud dormancy and enhancing flowering in Zantedeschia, achieved by the reciprocal cross-talk between cytokinin and gibberellin, contributes to the pool of evidence drawing common mechanisms between dormancy and flowering and may have commercial implications.  相似文献

The enzymatic and biochemical properties of the proteins encoded by five potato cytokinin oxidase/dehydrogenase (CKX)-like genes functionally expressed in yeast and the effects of tuber dormancy progression on StCKX expression and cytokinin metabolism were examined in lateral buds isolated from field-grown tubers. All five putative StCKX genes encoded proteins with in vitro CKX activity. All five enzymes were maximally active at neutral to slightly alkaline pH with 2,6-dichloro-indophenol as the electron acceptor. In silico analyses indicated that four proteins were likely secreted. Substrate dependence of two of the most active enzymes varied; one exhibiting greater activity with isopentenyl-type cytokinins while the other was maximally active with cis-zeatin as a substrate. [3H]-isopentenyl-adenosine was readily metabolized by excised tuber buds to adenine/adenosine demonstrating that CKX was active in planta. There was no change in apparent in planta CKX activity during either natural or chemically forced dormancy progression. Similarly although expression of individual StCKX genes varied modestly during tuber dormancy, there was no clear correlation between StCKX gene expression and tuber dormancy status. Thus although CKX gene expression and enzyme activity are present in potato tuber buds throughout dormancy, they do not appear to play a significant role in the regulation of cytokinin content during tuber dormancy progression.  相似文献
