The relation of the net uptake or output of carbon dioxide bydetached leaves of Pelargonium zonale or wheat to ambient carbondioxide concentration was investigated at two or three lightintensities. The statistics of fitted curves were used as datain analysis of variance. For Pelargonium the results were consistentwith a hypothesis of a ‘straight balance’ betweena rate of photosynthetic uptake proportional to concentrationand a constant rate of respiratory production. For wheat, however,the curve was steeper near than at lower carbon dioxide levels;this disproved the ‘straight balance’ hypothesisand was consistent with an increase in carbon dioxide fixationor a reduction in output over a limited concentration rangeon either side of . The characteristics of light respiration, possibly carbon dioxidedependent, were investigated further in an experiment on theeffect of oxygen concentrationupon r values for two species(P. zonale and Hydrangea sp.) at two light intensities; theywere also studied by measuring the ‘burst’ of carbondioxide output when leaves were darkened after illuminationat four different light intensities and at two temperatures. was linearly related to oxygen tension up to 610 mm partialpressure of mercury and a fivefold increase in light intensityhad only a very small effect indicating that photoxidation wasnot important. The carbon dioxide burst on darkening showedrelations to temperature and previous light intensity quitedifferent from those of which should be proportional to lightrespiration. These results, therefore, do not support the viewthat the burst represents the persistence of an enhanced lightrespiration.  相似文献

The rates of CO2 incorporation into the epidermis of C. communiswere linear and were similar during the completion of opening(2 h) and closing (1 h) movements of stomata. The kinetics of14C turnover between metabolites and the rates of ‘leakage’of metabolites were determined for opening and closing movements.When stomata were opening there was a slow turnover of 14C frommalate chiefly into sugars. Upon stomatal closure 14C was initiallymainly in sugars, malate, and sugar phosphates. Thereafter,there was a slight loss of label from sugar phosphates witha corresponding increase in malate. Starch became labelled duringopening and closing movements. Rates of incorporation of CO2found in the ‘leakage’ fraction were greatest whenstomata were opening. Of the labelled compounds Most‘from the tissue, malate was the most highly labelled whetherstomata were opening or closing. Although interpretation of the turnover patterns is difficultwithout knowledge of pool sizes for the metabolites it is suggestedthat a pool of sugars exists within the guard cells, which havefairly direct and reversible access to carbon from starch andmalate. The implications of loss of malate from guard cellsduring stomatal opening and closing are discussed.  相似文献

Two experiments are described in which stomatal sensitivityto low-intensity white light was studied for Xanthium pennsylvanicumWall. In the first experiment a daylength extension for 7, 9, or 15hrs. was given using 10, 40, or 160 lux to shorten a basic 16-hr.night, which was also given at its full length as a tenth treatment.Measurements were made of stomatal opening ability on the morningfollowing the different treatments. With a 15-hr. extensionthere was at all intensities a significant response, shown bya reduced rate of opening in the morning. With a 9-hr. extensionusing 40 or 160 lux, opening ability was reduced, but 9 hrs.of 10 lux was insufficinet to produce a detectable effect. The7-hr. extension was ineffective at all three intensities. In the second experiment stomatal behaviour was observed during20 hrs. of either darkness or 10 lux at four temperatures (15,22, 29, and 36°C.). During 20 hrs. of darkness there wasnight opening at all temperatures, but at lower temperaturesit began sooner and lasted longer. These responses to temperaturedid not fit a simple linear relationship, there being a significantcubic term revealed by non-linear regression analysis. Thiscould be explained if the response was considered in terms ofthe magnitude of the change in temperature (from 25°C.)at the beginning of the experiment; there appeared to be sometemperature compensation over a limited range. in 10 lux, nightopening was suppressed at 29° and 36°, but at 15°it was apparently unaffected by the light; at 22° it wasnot completely suppressed by 10 lux but the time of its occurrencewas delayed. Effects of light and temperature are discussed in relation toan endogenous rhythm in darkness which was previoulsy shownto operate in Xanthium pennsylvanicum (Part IX). It is considered that to explain effects of very low intensitylight it will be necessary to recognize a ‘low intensityresponse’ by stomata, which does not operate via changesin guard-cell carbon dioxide.  相似文献

Eight detached wheat leaves were each subjected to varying degreesof controlled water strain, in a specially constructed leafchamber (which is described in detail) connected to a closedsystem incorporating an infra-red gas analyser. The water strainwas varied by applying successively to their leaf sheaths twoconcentrations of mannitol solution (0·2 M. and 0·4M.) each preceded and followed by water. Conditions were maintainedconstant for light (900 f.c.), leaf temperature (25° C.)and humidity (50 per cent. RH) of the circulating air as itentered the leaf chamber. Steady-state values (gamma) of thecarbon dioxide concentration in the closed system were recordedboth for the water and mannitol treatments and the data analysedstatistically. The two mannitol treatments were found to increasegamma from 80 p.p.m. with water to 94 and 116 p.p.m. respectively,all differences being highly significant. Concomitant measurementswere made of stomata1 resistance with a porometer and of transpirationby means of wet and dry thermocouples; the stomata showed markedclosure in response to mannitol treatment (complete closurewith the higher concentration) and partial reopening with theleaf sheath in water once more, transpiration also falling andrising inversely with water strain. It is concluded that part of the closing response of wheat stomatato water strain under natural conditions must be operated byincrease in the intercellular-space carbon dioxide concentration,but a comparison with previous experimental results suggeststhat this can only be a small part unless sensitivity to carbondioxide is greatly increased by water loss. The initial response to mannitol treatment was a preliminaryopening of the stomata and a fall of about 10 per cent. in carbondioxide concentration. These results imply that under naturalconditions the preliminary opening of wheat stomata with wiltingmay be partly passive and partly active with a carbon dioxide-operatedmechanism. A subsidiary experiment with water strain varied by removingand restoring the water supply to the leaf sheath gave increasesand decreases in carbon dioxide similar to those in the mainexperiment; the results of the latter need not, therefore, beattributed to metabolism of the mannitol. It is suggested that values of gamma give a good measure ofthe efficiency of the mesophyll for net carbon dioxide absorption,with the stomatal factor eliminated, under the given conditionsand with carbon dioxide ‘limiting’ in the senseused by Maskell (1928).  相似文献

It was found that stomata on illuminated leaves, both of Pelargoniumand wheat, opened much wider where the leaf surface was enclosedin a small volume of air, as in a normal porometer cup, thanelsewhere. This was shown for both species by the infiltrationmethod, and for Pelargonium by Lloyd's method and direct microscopicalobservation also. The effect was shown not to be due to pressure of the porometercup or glass plate on the leaf, or to temperature differences,nor directly to the lack of movement or high humidity of theenclosed air. A considerable body of data was collected which appeared tosupport the hypothesis that the wide opening was due to accumulationof some volatile substance produced by the leaf, but all theresults were also consistent with the view that it was causedby reduction in the carbon dioxide content of the enclosed airbelow the normal 0·03 per cent. owing to photosynthesis.Further crucial experiments with both the porometer and infiltrationmethods left virtually no doubt that the latter hypothesis wascorrect. This extreme sensitivity of stomata to carbon dioxide concentrationwithin the range 0·03 per cent. to zero is discussedin relation to their operation in nature, and a possible biologicaladvantage is suggested. The bearing of the effect upon porometer investigations is alsodiscussed and it is concluded that for all quantitative or semi-quantitativeexperimentation it is essential to use a cup detached betweenreadings, or at least swept with air such as surrounds the restof the leaf, and to have the upper leaf surface above the cuparea freely exposed or similarly swept. For qualitative investigationof the light response of stomata the traditional form of cupmay be used. The importance is stressed of allowing porometer readings toreach equilibrium under one set of conditions before changingto another, when investigating the ‘closing’ or‘opening’ effects of external factors. Several subsidiary effects, observed in the course of the investigation,are discussed; in particular an effect of humidity upon therate of response to other factors.  相似文献

An investigation of the effects of different day-length treatmentson stomatal behaviour in Xanthium pennsylvanicum Wall. has shownthat there are differences in long-and short-day treatmentssimilar to those first reported by Schwabe (1952) for Chrysanthemumand Kalanchoe, viz. stomatal opening towards the end of thenight in short days, but not in long days (short night precededby a period of low intensity illumination). In Xanthium therewere in addition very marked differences in rates of stomatalopening in the morning after different lengths of night. Theseeffects were not persistent, there being an immediate reversalupon a change from long- to shrot-day treatment, or vice versa. Further investigation showed that there was an endogenous rhythmaffecting the stomata in continuous darkness; rate of openingwas slow after very short nights, but became greater with extensionof the night to 14–16 hrs., this being the first ‘peak’of an ‘opening ability’ rhythm. The rhythm diedout rapidly and the second cycle was much reduced in amplitude.The period of the rhythm appeared to be approximately 24 hrs.The phase was set mainly by the time of onset of darkness, butthe duration of the pretreatment with low intensity illuminationwas also important—prolonging this was found to reudcethe time in darkness before the first peak. Each hour of lightof 1,500 lux given before darkness was found to be equivalentot approximately 0.3 hr. to darkness. The predominant effect of the length of the preceding nightwas on the slope of the opening curves rather than on the timeof onset of opening following illumination. The first peak of the opening ability rhythm was often manifestedeven in continuous darkness by a period of ‘night opening’of the stomata. The results are discussed and compared with those of other authorsand attempts are made to relate them to theories of the stomatalmechanism.  相似文献

The success of Triticum aestivumxZea mays crosses, used to producewheat doubled haploids, is influenced by light intensity. Toexamine the basis for this response, pollen tube growth, embryosurvival and indicators of photosynthetic rate were measuredin two wheat cultivars (‘Karamu’ and ‘Kotuku’)crossed with maize at two irradiance levels (250 or 750 µmolm-2s-1, PAR). Pollen tube growth was significantly affectedby light intensity in ‘Karamu’ plants but not in‘Kotuku’ plants, despite both cultivars being pollinatedby the same maize source. The percentage of pollen tubes reachingthe cavity between the ovarian wall and integuments, or in themicropyle of ‘Karamu’ plants at high light intensity(65%) was nearly three-times greater than that at low lightintensity (22%). Thus, either low light intensity can affectthe maternal wheat plant in a way that inhibits pollen tubegrowth and/or high light intensity may promote pollen tube growthin ‘Karamu’ plants. Significant differences in ratesof electron transport in plants grown at the two light intensitiesindicated that the rate of photosynthesis may also have an effecton pollen tube growth. These results have importance for improvingthe efficiency of wheat x maize crosses and other wide cerealcrosses. Extracts of small and mature-size lupin pods yielded four substancesaffecting the growth of wheat-coleoptile sections: one acidpromotor (A), two acid inhibitors(B and X), and one neutralinhibitor(Y). Inhibitor B was extremely active, however, coleoptile sectionsshowed no signs of toxic effects; they resumed growth at a rapidrate after rinsing them and adding ß-indolylaceticand (IAA) to the medium. 1 µg of IAA was required to counteractthe effect of ‘B’ extracted from 230 mg. Of tissue.On an equal fresh weight basis the inhibiting action of ‘B’in lupin pods was 500–1,500 times more potent than thatof ‘inhibitor ß’ in etiolated pea seedlings. Small pods of plants infected with pea-mosaic virus yielded3 times the amount of ‘A’ of healthy plants (equivalentto 1 µg. IAA 0.3 µg. IAA per 25 g. of tissue respectively),and approximately the amount of ‘B’. Mature podsof virus-infected plants again yielded more‘A’,but also 2? times more ‘B’ than pods of healthyplants. Healthy pods yielded more ‘A’ than virus-infectedpods, and there was no difference in ‘X’. A lupin abscission test was developed and the effects of proximaland distal application of -naphthyl acetic acid (NAA) are presented,and discussed with respect to results of other abscission tests. ‘A’ accelerated abscission when applied proximally,and delayed or prevented it when applied distally. ‘B’strongly accelerated abscission when applied in either way.A possible mechanism explaining the abscission-inducing effectof developing pods on later flowers is discussed in terms ofthe substances ‘A’ and ‘B’. The influence of shaking rates (expressed as revolutions permin) on orbital shaking platforms (1 in (2.54 cm) diam. rotarymotion) on the growth of cell suspension cultures of Acer pseudoplatanusL. and Atropa belladonna cultivar lutea Döll are described.By following cell growth and respiration and the levels of oxygenand carbon dioxide in the media during the progress of incubationit is concluded that the reduction of growth at sub-optimalshaking rates is not due to oxygen deficiency or toxic accumulationof carbon dioxide. The growth of the Atropa cell suspensionin ‘closed systems’ has been studied by the developmentof modified culture vessels and evidence obtained that the reducedgrowth in the systems is due to the formation by the culturesof an unidentified volatile growth inhibitor and not to eitheroxygen depletion or toxic accumulation of either carbon dioxideor ethylene. It is suggested that the reduced growth in ‘opensystems’ cultures at sub-optimal shaking speeds is eitherdue to retention of this volatile inhibitor or to restrictionof nutrient uptake by the existence of a stationary liquid-phaseboundary to the cells.  相似文献

Impedance Spectroscopy in Frost Hardiness Evaluation of Rhododendron Leaves   总被引:5,自引:0,他引:5  
Impedance spectroscopy was used in studying frost hardinessof leaves of two diploid rhododendron cultivars, RhododendronL. ‘PJM’ and R. ‘Cunningham's White’,and their tetraploid derivatives, R. ‘Northern Starburst’(NSB) and CW4. After the growing season and initial hardeningin a greenhouse, plants were subjected to an acclimation regimein a phytotron: 3 consecutive weeks at +5, +1 and -2°C each.Hardiness was studied with controlled freezing tests beforeeach decrease in temperature and at the end of the experiment,based on data of extracellular resistance reand relaxation time of the frost-exposed leaves. The correlation of the two estimateswas 0.92. Generally, the diploid clones had better frost hardinessthan the tetraploid clones. At the end of the experiment, frosthardiness of the diploid ‘PJM’ was -28.7°C andthat of the tetraploid NSB -20.6°C. Leaves of the diploid‘Cunningham's White’ and of the tetraploid CW4 hardenedto -32.0°C and -20.9°C, respectively. Frost hardinessestimated by impedance spectroscopy correlated well with earlierresults based on visual scoring (r = 0.81–0.86) and electrolyteleakage tests (r = 0.84–0.90), but results from impedancespectroscopy indicated weaker hardiness than the other tests.The difference between the results from impedance spectroscopyand the other tests was smaller and more coherent within the‘Cunningham's White’ clones than within ‘PJM’and NSB. Changes in the major alcohol-soluble, low molecular weight carbohydratesof P. purpurea, O--D-galactopyranosyl-(1-2)-glycerol (‘floridoside’)and O--D-galactopyranosyl-(1-1)-glycerol (‘isofloridoside’),have been examined in response to salinity variation. ‘Floridoside’is shown to vary in absolute amount, increasing in hypersalineand decreasing in hyposaline media. ‘Isofloridoside’content per cell does not change in a similar manner. Responsesare almost identical under light or dark conditions, ‘floridoside’changes being complete within 24 h. Decreasing the externalwater potential using ionic and non-ionic solutes has the sameeffect upon galactosyl-glycerol content. The amount of ‘floridoside’synthesized, and degraded under hypersaline and hyposaline conditionsrespectively is shown to be insufficient to restore cell volumeto its original value. It is therefore suggested that the primaryfunction of ‘floridoside’ increases in concentratedsea-waters is that of a compatible solute, serving to protectthe cell during periods when the external salt content is increaseddramatically.  相似文献

The line of closure in the stoma in fully grown adult leavesof certain eucalypts is formed by special ridges of cuticlecalled ‘stomatal bars’ developed at the line ofclosure itself or from upgrowth of the cuticle of the lowersurface of the guard cells. Stomatal bars, previously discoveredin three members of the informal group ‘Bisectae’are shown to be restricted to certain species in that group.Possession of stomatal bars may affect stomatal performancebut does not appear to be a general adaptive response to thehabitats of the species which possess them. More probably, thepossession of stomatal bars is an inherited character with taxonomicvalue. The presence of stomatal bars in other genera is discussed. The effects of exposure of up to 2 h with sulphur dioxide ona range of plant species was observed by measuring changes inthe rate of net photosynthesis under closely controlled environmentalconditions. Ryegrass, Lolium perenne ‘S23’ was thespecies most sensitive to SO2; significant inhibition was detectedat 200 nl l–1. Fumigations at 300 nl l–1 also inhibitedphotosynthesis in field bean (Vicia faba cv. ‘Three FoldWhite’ and ‘Blaze’) and in barley (Hordeumvulgare cv. ‘Sonja’). No effect was detected inwheat (Triticum aestivum cv. ‘Virtue’) at concentrationsup to 600 nl l–1 SO2, or in oil-seed rape (Brassica napuscv. ‘Rafal’) except at 800 nl l–1 SO2). Recoverycommenced immediately after the fumigation was terminated andwas complete within 2 h when inhibition had not exceeded 20%during the SO2 treatment. The effect of a temperature increase from 25 to 35°C onstomatal opening in darkness (‘night opening’) onexcised, turgid leaves of Stachytarpheta indica was investigatedby microscopic examination of a baxial epidermis fixed in absoluteethanol. An appreciable degree of opening occurred towards theend of a 14-h night at 25°C, and this was substantiallyenhanced by the temperature increase to 35°C in the dark,which also promoted a marked increase in starch hydrolysis andaccumulation of potassium in the guard cells. The degree oftemperature-induced night opening was somewhat smaller thanthat of light-induced opening, and was higher in CO2-free airthan in normal air. 2,4-dinitrophenol (DNP) was effective inarresting stomatal opening and suppressing starch hydrolysisand increase in stomatal potassium. The temperature-inducednight opening is related, to a great extent, to the enhancementby high temperature of starch hydrolysis and potassium accumulationin the guard cells, and the inhibitory effect of DNP on stomatalopening is attributed largely to its suppression of these twometabolic processes. The importance of oxidative phosphorylationas a possible source of energy for stomatal opening is brieflydiscussed.  相似文献

Plants of Vicia faba and Hordeum vulgare were grown in growthboxes with 7 mW cm–2 PAR, 14 h day/10 h night, at 22/20°C. Stomata of attached leaves were measured under controlledconditions by means of an optical microscope and the distributionfunctions of the widths of pores were established. For Viciafaba they appeared to be symmetrical bell-shaped functions.In the process of stomatal opening or closure the shape of thedistribution remained constant, its maximum sliding left andright along the aperture axis. This result has been interpretedto mean that increments or decrements of apertures were equalfor all stomata independent of their individual apertures. Theconclusion has been drawn that the ‘driving force’is evenly distributed, equal for all stomata, and varies withinwider limits than is possible for stomatal apertures. Stomatalopening is limited by the closed state from below and by ananatomically possible maximum aperture from above.  相似文献

Studies on the stomatal regulation in the root (wilt) affectedcoconut palms (Cocos nucifera L.) revealed that the diseasedpalms had low stomatal resistance compared to the healthy palms,irrespective of their age. The same trend was observed whetherthe determinations were made at different times of the day (6–18h) or under irrigated and unirrigated conditions or in differentseasons (‘dry’ and ‘wet’). Thus, thestomatal regulation was significantly impaired in the diseasedpalms resulting in excessive water loss compared to the healthypalms. Results are discussed with the available literature onother similar disease caused by fungi, bacteria and mycoplasma-likeorganisms in different plants. In vitrorates of gross and net oxygen production were measuredas a function of light intensity in some plankton communitiescollected from Bedford Basin, Nova Scotia, and in a monoclonalculture of Synechococcus. The rate of gross oxygen productionwas measured by a technique in which the stable oxygen isotope,18O, serves as a photosynthetic tracer Net oxygen productionwas measured by automated Winkler technique. The rate of communityrespiration in the light was then determined by the differencebetween gross and net rates of oxygen production. In the naturalpopulations examined, neither gross nor net oxygen productionrates were significantly inhibited at the highest light intensitymeasured (500–800 µE m–2 s–1) In a samplein which the dark respiration rate was small relative to themaximal rate of production [Pmax;sensu Platt et al (1980) JMar. Res., 38, 687–701] the rates of ‘light’respiration were 3 times greater. In two other communities,with high rates of dark respiration relative to Pmaxthe ratesof ‘light’ respiration were closer to rates of darkrespiration. In the Synechococcus clone, both gross and netoxygen production rates were inhibited at high light intensities.Rates of ‘light’ respiration were found to varyas a function of light intensity. The greatest rates of respirationwere measured in samples incubated at light intensities thatwere just saturating (100 µE m–2 s–1). When nitrate, ammonium, or urea were the sole nitrogen sourcesfor the growth of Platymonas striata there was a short initiallag period, after which uptake/assimilation occurred at a constantrate. Glycine ‘uptake’ was however non-linear withtime. Addition of ammonium or urea markedly inhibited nitrate‘uptake’ whereas although glycine was initiallytotally inhibitory, nitrate ‘uptake’ recommencedabout 1 h after addition, but at a greatly reduced rate. Ammoniumtotally inhibited urea ‘uptake’, but did not inhibitglycine ‘uptake’. When ammonium, urea, or glycinewere present at the same time as nitrate the total rate of nitrogen'uptake/assimilation was within 40% (or closer) of that of nitratenitrogen ‘uptake’ alone. In the first two casesthis reflected a total switching off of nitrate ‘uptake’whilst the new substrate was assimilated at a rate similar tothe initial nitrate rate, whereas in the case of glycine, bothsubstrates were used simultaneously. Once the concentrationof any of the inhibitory nitrogen sources had been reduced toa low level the ‘uptake’/assimilation of nitraterecommenced. It is suggested that the ability of P. striatato maintain a more or less constant rate total nitrogen ‘uptake’/assimilationirrespective of the mixture of utilizable nitrogen sources presentedto it, is not due to controls at the level of the entry of substrateinto the cell, but to intracellular actions at the enzyme level.The relative constancy of, and high rate of ‘uptake’,irrespective of nitrogen source and over a wide range of extracellularconcentrations, is tentatively considered to be due to saturationof the assimilatory mechanisms under the conditions employed;all the sources lead, either directly or indirectly, to theproduction of amino groupings for entry into nitrogenous metabolism.This homeostasis of nitrogen ‘uptake’/assimilationwould clearly be of considerable benefit to the alga in nature,enabling it to maximize growth in changing environmental conditions. As in the previous investigation (Part I), stomatal responsesin wheat were followed by means of porometer readings; betweenthese, air was forced through the stomata to sweep out the substomatalcavities. Instead of forcing the whole of a relatively slowair-flow in through the stomata, however, a fast flow was sweptacross the leaf surface and a portion of this passed into theintercellular space system. This helped to maintain the desiredcarbon dioxide content close to the stomatal openings and thestomata could thus be treated with different carbon dioxideconcentrations. A factorial experiment was carried out in which6 carbon dioxide concentrations from zero to 0084 per cent,were combined with 3 light intensities from 90 to 800 f.c, the18 treatments being replicated 6 times in three ‘66 latinsquares’. A supplementary experiment in which the same6 carbon dioxide treatments were given in darkness was carriedout in duplicate. The results confirmed previous findings inthat the stomata showed no appreciable response to variationof external carbon dioxide supply between zero and 001 percent, concentration and that a large and highly significantlight response was found at each of these two carbon dioxidelevels. This is held to indicate the existence of a light effect(direct or indirect) not operating by reduction of the internalcarbon dioxide content of the guard cells. On the assumptionthat the rates of air flow through the stomata were adequate,the combined results of the two experiments are interpretedmainly in terms of two direct effects of light upon the stomata,one depending on photosynthetic reduction of carbon dioxidein the guard cells and the other, mentioned above, not so operating.To these must be added the indirect effect responsible for the‘transmission’ described in Part 1, which mightpossibly account for the whole of the second direct effect postulated.Finally, if the rates of flow were in fact grossly inadequate,the trends attributed to the first direct effect mentioned abovemight have been due to indirect control by the mesophyll viathe intercellular space atmosphere. Such control must accountfor part of the light responses which occur under natural conditions.The natural light response is thus made up of at least two andmore probably four components—two direct and two indirect,in each case one operating by the internal carbon dioxide andthe other not. A highly significant diurnal rhythm of stomatalmovement under constant illumination and temperature is shownto occur in wheat. The measure of stomatal resistance used (logresistance per 10,000 stomata) is discussed, and it is concludedthat although it is empirical a much more satisfactory measureis not at present available.  相似文献
