Gliding allows mammals to exploit canopy habitats of old-growth forests possibly as a means to save energy. To assess costs of quadrupedal locomotion for a gliding arboreal mammal, we used open-flow respirometry and a variable-speed treadmill to measure oxygen consumption and to calculate cost of transport, excess exercise oxygen consumption, and excess post-exercise oxygen consumption for nine northern flying squirrels (Glaucomys sabrinus) and four fox squirrels (Sciurus niger). Our results indicate that oxygen consumption during exercise by flying squirrels was 1.26–1.65 times higher than predicted based on body mass, and exponentially increased with velocity (from 0.84 ± 0.03 ml O2 kg−1 s−1 at 0.40 m s−1 to 1.55 ± 0.03 ml O2 kg−1 s−1 at 0.67 m s−1). Also, cost of transport in flying squirrels increased with velocity, although excess exercise oxygen consumption and excess post-exercise oxygen consumption did not. In contrast, oxygen consumption during exercise for fox squirrels was similar to predicted, varying from 0.51 (±0.02) ml O2 kg−1 s−1 at 0.63 m s−1 to 0.54 (±0.03) ml O2 kg−1 s−1 at 1.25 m s−1. In addition, the cost of transport for fox squirrels decreased with velocity, while excess exercise oxygen consumption and excess post-exercise oxygen consumption did not. Collectively, these observations suggest that unlike fox squirrels, flying squirrels are poorly adapted to prolonged bouts of quadrupedal locomotion. The evolution of skeletal adaptations to climbing, leaping, and landing and the development of a gliding membrane likely has increased the cost of quadrupedal locomotion by >50% while resulting in energy savings during gliding and reduction in travel time between foraging patches.  相似文献

Natural cartilage remodels both in vivo and in vitro in response to mechanical forces and hence mechanical stimulation is believed to have a potential as a tool to modulate extra-cellular matrix synthesis in tissue-engineered cartilage. Fluid-induced shear is known to enhance chondrogenesis on animal cells. A well-defined hydrodynamic environment is required to study the biochemical response to shear of three-dimensional engineered cell systems. We have developed a perfused-column bioreactor in which the culture medium flows through chondrocyte-seeded porous scaffolds, together with a computational fluid-dynamic model of the flow through the constructs' microstructure. A preliminary experiment of human chondrocyte growth under static versus dynamic conditions is described. The median shear stress imposed on the cells in the bioreactor culture, as predicted by the CFD model, is 3 × 10−3 Pa (0.03 dyn/cm2) at a flow rate of 0.5 ml/min corresponding to an inlet fluid velocity of 44.2 μm/s. Providing a fluid-dynamic environment to the cells yielded significant differences in cell morphology and in construct structure. Received: 22 December 2001 / Accepted: 18 February 2002  相似文献

Bifidobacterium longum grew at 65 L pilot scale of the membrane bioreactor (MBR), externally fitted with ceramic membrane (0.7 m2). Cell mass at the MBR reached 22.18 g L−1 as dry cell weight in 12 h, which is 8.44 times higher than cell mass attained at the vial culture. The growth rate in the vial culture was μ = 0.385 h and at the batch culture was μ = 1.13 h in the exponential period and μ = 0.31 h−1 in the stationary period. In the fed-batch mode was μ = 1.102 h−1 for 6 h with inoculation and declined to μ = 0.456 h−1 with feeding of feed medium. The growth rate at the MBR was μ = 0.134 h−1. The number of viable cells was 6.01 × 1012 cfu L−1 at the batch culture, but increased to 1.15 × 1014 cfu L−1 at the MBR culture. The specific growth rate of viable cell number (colony-forming units per liter, per hour) improved by 6.01 times from the batch to the MBR culture. The wall shear stress mainly generated by the pump, and the membrane incorporated into the MBR was controlled during the cultivation at the MBR. The viability of B. longum declined to under 10% in the first 2 weeks of the 4-week stability test (40°C) as B. longum was exposed to over wall shear stress 713 Pa, but the viability improved to 30–40% in wall shear stress of 260 Pa or STR culture. The loss in the cell viability can be saved by managing with wall shear stress during the cultivation at the MBR.  相似文献

Reported mechanical properties of orbital connective tissue and fat have been too sparse to model strain–stress relationships underlying biomechanical interactions in strabismus. We performed rheological tests to develop a multi-mode upper convected Maxwell (UCM) model of these tissues under shear loading. From 20 fresh bovine orbits, 30 samples of connective tissue were taken from rectus pulley regions and 30 samples of fatty tissues from the posterior orbit. Additional samples were defatted to determine connective tissue weight proportion, which was verified histologically. Mechanical testing in shear employed a triborheometer to perform: strain sweeps at 0.5–2.0 Hz; shear stress relaxation with 1% strain; viscometry at 0.01−0.5 s−1 strain rate; and shear oscillation at 1% strain. Average connective tissue weight proportion was 98% for predominantly connective tissue and 76% for fatty tissue. Connective tissue specimens reached a long-term relaxation modulus of 668 Pa after 1,500 s, while corresponding values for fatty tissue specimens were 290 Pa and 1,100 s. Shear stress magnitude for connective tissue exceeded that of fatty tissue by five-fold. Based on these data, we developed a multi-mode UCM model with variable viscosities and time constants, and a damped hyperelastic response that accurately described measured properties of both connective and fatty tissues. Model parameters differed significantly between the two tissues. Viscoelastic properties of predominantly connective orbital tissues under shear loading differ markedly from properties of orbital fat, but both are accurately reflected using UCM models. These viscoelastic models will facilitate realistic global modeling of EOM behavior in binocular alignment and strabismus.  相似文献

In this paper, a dynamic model is proposed to quantify the relationship between fluid flow and Cl-selective membrane current in vascular endothelial cells (VECs). It is assumed that the external shear stress would first induce channel deformation in VECs. This deformation could activate the Cl channels on the membrane, thus allowing Cl transport across the membrane. A modified Hodgkin–Huxley model is embedded into our dynamic system to describe the electrophysiological properties of the membrane, such as the Cl-selective membrane current (I), voltage (V) and conductance. Three flow patterns, i. e., steady flow, oscillatory flow, and pulsatile flow, are applied in our simulation studies. When the extracellular Cl concentration is constant, the I-V characteristics predicted by our dynamic model shows strong consistency with the experimental observations. It is also interesting to note that the Cl currents under different flow patterns show some differences, indicating that VECs distinguish among and respond differently to different types of flows. When the extracellular Cl concentration keeps constant or varies slowly with time (i.e. oscillates at 0.02 Hz), the convection and diffusion of Cl in extracellular space can be ignored and the Cl current is well captured by the modified Hodgkin–Huxley model alone. However, when the extracellular Cl varies fast (i.e., oscillates at 0.2 Hz), the convection and diffusion effect should be considered because the Cl current dynamics is different from the case where the convection-diffusion effect is simply ignored. The proposed dynamic model along with the simulation results could not only provide more insights into the flow-regulated electrophysiological behavior of the cell membrane but also help to reveal new findings in the electrophysiological experimental investigations of VECs in response to dynamic flow and biochemical stimuli.  相似文献

The red-tide dinoflagellate Protoceratium reticulatum is shown to be protected against turbulence-associated damage by the use of the additives Pluronic F68 (PF68) and carboxymethyl cellulose (CMC) in the culture medium. Relative to agitated controls, these additives had a dose-dependent protective effect at concentrations of up to 0.4 and 0.5 g L−1 for CMC and F68, respectively. In static cultures, these additives inhibited growth directly or indirectly at a concentration of >0.5 g L−1. Compared to CMC, PF68 was a better protectant overall. Cell-specific production of yessotoxins was enhanced under elevated shear stress regimens so long as the turbulence intensity was insufficient to damage the cells outright. Shear-induced production of reactive oxygen species and direct effects of turbulence on the cell cycle contributed to the observed shear effects.  相似文献

Somatic embryogenesis can be induced in the leaves of cork oak (Quercus suber L.) trees. The use of this propagation system in multivarietal forestry requires the mass production of cloned plants at low cost. Investigations were made into the influence of three types of Erlenmeyer flask and three orbiting speeds (60, 110, and 160 rpm) on oxygen transfer rate (KL a), the shear force index (SFI), biomass production, and the proliferation of embryogenic clumps (EMCs) in cultures during the proliferation phase. KL a varied between 0.11 and 1.47 h−1 without biomass production being limited by oxygen availability. The EMCs grew even in hypoxic conditions, although the suppression of gaseous exchange strongly reduced biomass production. Cultures with different levels of hydrodynamic stress and SFI values (1.4·10−3–8.8·10−3 cm min−1) were obtained. Proliferation rates of EMCs increased with agitation rate and the SFI. The largest number of EMCs was obtained in baffled flasks agitated at 160 rpm (KL a of 1.47 h−1, and SFI of 8.8·10−3 cm min−1) with mild hydrodynamic stress enhancing growth. Biomass production increased with agitation and hydrodynamic stress, but only when the SFI value was below 5·10−3 cm min−1. The greatest biomass production was obtained in smooth 100 ml flasks agitated at 160 rpm. The differentiation of embryos was favoured by the lowest KL a (0.11 h−1) and SFI (1.40·103 cm min−1) values, achieved using these flasks when agitated at 60 rpm.  相似文献

The activated sludge process (ASP) has high operational costs due to the need for aeration at dissolved O2 (DO) levels of ≥2 mg l−1 and high capital costs to construct large reactors due to a low organic loading [typically 1 kg chemical oxygen demand (COD) m−3 day−1]. A novel method for improving the energy use and treatment efficiency of the ASP via limited oxygenation (0.4 mg DO l−1) and high organic loading (6.2 kg COD m−3 day−1) is proposed based on a laboratory-scale ASP for ammonia-rich industrial wastewaters. The sludge blanket phenomenon and granulation occurred simultaneously in the upflow microaerobic reactor.  相似文献

Assessment of the importance of internal nutrient loading is essential for managing and restoring eutrophic shallow lakes. To date, studies of internal loads have tended to focus on one of two abiotic processes, either molecular diffusion or sediment/nutrient entrainment (resuspension). This study presents a new approach to determining the non-biological fluxes of nitrogen (N) and phosphorus (P) from the sediment to the water column of shallow lakes. Three mutually exclusive flux processes: (i) molecular diffusion, (ii) turbulent diffusion (eddy diffusivity) and (iii) wind-induced resuspension of N and P, were related to a gradient of benthic shear stress. A model presented here allowed the durations and magnitudes of different non-biological fluxes to be calculated over time, based on benthic shear stress. Two site-specific critical shear stress thresholds determined which of the three flux processes dominated for any benthic shear stress value. The model was calibrated for a shallow lake and the continuous flux of nutrient from the sediment to the overlying water generated by each process during that period was calculated, enabling the estimation of the relative importance of each of the three flux processes over a one-year period. Wind-induced resuspension dominated the internal nutrient flux, operating for 38% of the time and contributing 0.9 T P year−1 and 10.2 T N year−1 to the internal nutrient load. In contrast, molecular diffusion only contributed 0.01–0.02 T P year−1 and 0.12–0.20 T N year−1 to the water column, while turbulent diffusion provided up to 0.6 T P year−1 and 6.2 T N year−1. Our model suggests that turbulent diffusion is a neglected and potentially important process contributing to internal nutrient loading in shallow lakes, whereas molecular diffusion appears to be relatively unimportant in lakes that experience turbulence at the sediment–water interface. The extent of mercury (Hg) toxicity in the heterocystous cyanobacterium Nostoc muscorum grown for 72 h in three different light intensities was tested for various physiological parameters viz. growth, pigment contents, photosynthesis, respiration, reactive oxygen species (ROS), malondialdehyde formation and antioxidants. A general reduction in growth and pigments, whole cell O2-evolution, photosynthetic electron transport activities and 14CO2-fixation was observed in a metal concentration–dependent manner, and this effect was more pronounced in high light (130 μmol photon m−2 s−1)–exposed cells as compared to low (10 μmol photon m−2 s−1) and normal (70 μmol photon m−2 s−1) light intensity–exposed cells; however, carotenoids and respiration showed reverse trend. Among photosynthetic electron transport activities, whole chain activity was found to be most sensitive in comparison with photosystem II (PS II) and photosystem I (PS I). Comparing the different photosynthetic processes, 14CO2-fixation was most affected in cyanobacterial cells when exposed to Hg and different light intensities. After application of various exogenous electron donors, diphenyl carbazide was found to be more effective to restore PS II activity, suggesting that site of damage lies in between oxygen evolving complex and PS II. Level of oxidative stress (superoxide radical and lipid peroxidation) was maximum at 3.0 μM of Hg when coupled with high light intensity (except hydrogen peroxide). A dose-dependent increase in enzymatic antioxidants such as superoxide dismutase, peroxidase and catalase as well as non-enzymatic antioxidants such as proline, ascorbate, cysteine (except under high light intensity) and non-protein thiols [NP-SH] was observed, which further increased with the increase in light intensity. It was noticed that Hg intoxicates N. muscorum through ROS production, which is aggravated along with the increase in light intensity. Overall results suggest that the severity of the metal stress does increase with Hg concentrations but when coupled with light, it was the light intensity that determines the extent of Hg toxicity.  相似文献

All embryonic and fetal amniotes possess a ductus(i) arteriosus(i) that allows blood to bypass the pulmonary circulation and the non-functional lungs. The central hemodynamic of embryonic reptiles are unique, given the additional systemic aorta that allows pulmonary circulatory bypass, the left aorta (LAo). The LAo exits in the right ventricle or ‘pulmonary side’ of reptilian hearts in both embryos and adults, but its functional significance in ovo is unknown. This study investigated the role of the LAo in embryonic American alligators by surgically occluding the LAo and measuring oxygen consumption and, in addition, measured hemodynamic responses to hypoxia in embryonic alligators. We measured systemic cardiac output and primary chorioallantoic membrane (CAM) artery blood flow for normoxic and hypoxic-incubated (10% O2) American alligator embryos (Alligator mississippiensis). Chronic blood flow (1–124 h) in the primary CAM artery for hypoxic-incubated embryos (92 ± 26 ml min−1 kg−1) was elevated when compared with normoxic-incubated embryos (29 ± 14 ml min−1 kg−1, N = 6; P = 0.039). For hypoxic-incubated embryos, acute LAo blood flow (49.6 ± 24.4 ml min−1 kg−1) was equivalent to the combined flow of the three systemic great vessels that arise from the left ventricle, the right aorta, common carotid and subclavian arteries (43.6 ± 21.5 ml min−1 kg−1, N = 5). Similarly, for normoxic-incubated embryos, LAo blood flow (27.3 ± 6.6 ml min−1 kg−1) did not statistically differ from the other three vessels (18.4 ± 4.9 ml min−1 kg−1, N = 5). This study contains the first direct test of LAo function and the first measurements of blood flow in an embryonic reptile. These data support the hypotheses that embryonic alligators utilize the LAo to divert a significant amount of right ventricular blood into the systemic circulation, and that CAM blood flow increases following chronic hypoxic conditions. However, surgical occlusion of the LAo did not affect egg [(V)\dot]\textO2, \dot{V}_{{\text{O}}_{2}}, supporting the hypothesis that the LAo of reptiles is not critical to maintain in ovo oxygen consumption.  相似文献

To study the effect of water flow on coral growth, four series of ten coral nubbins of Galaxea fascicularis were exposed to four different flow regimes (0, 10, 20, and 25 cm s−1, bidirectional flow) for 42 weeks. Buoyant weight, surface area, and polyp number were measured at regular intervals. Net photosynthesis and dark respiration were measured at the corresponding flow speeds, and daily amount of photosynthetic carbon left for coral growth was calculated. Finally, skeletal density and CN content, chlorophyll concentration and dry weight of coral tissue were determined for each coral. Specific growth rate (in day−1) decreased with time in each flow treatment. Absence of flow resulted in significantly lower growth rates. Average specific growth rate calculated over the entire experiment was not significantly different between 10 and 20 cm s−1, while it was significantly higher at 25 cm s−1. From 10 to 25 cm s−1, average net photosynthetic rate decreased and average dark respiration rate did not change significantly. Scope for growth based on phototrophic carbon decreased with increasing flow. Growth was not positively correlated with either photosynthesis or respiration, or scope for growth. It is suggested that higher flow rates reduce the chance of disturbance of coral growth by competing algae or cyanobacteria, allowing corals to grow more readily with the maximum specific growth rate possible under the given environmental conditions. Notably, other effects of increased flow, such as increased respiratory rates and increased (in)organic nutrient uptake, might have been equally responsible for the increased growth of the corals in 25 cm s−1.  相似文献

Remodeling of endothelial basement membrane is important in atherogenesis. Since little is known about the actual relationship between type IV collagen and matrix metalloprotease−2 (MMP-2) in endothelial cells (ECs) under shear stress by blood flow, we performed quantitative analysis for type IV collagen and MMP-2 in ECs under high shear stress. The mRNA of type IV collagen from ECs exposed to high shear stress (10 and 30 dyn/cm2) had a higher expression compared to ECs exposed to a static condition or low shear stress (3 dyn/cm2) (P < 0.01). 3H-proline uptake analysis and fluorography revealed a remarkable increase of type IV collagen under high shear stress (P < 0.01). In contrast, zymography revealed that exposing to high shear stress, however similar positivity was leveled in the intracellular MMP-2 in the control and high shear stress-exposed ECs, reduced the secretion of MMP-2 in ECs. The results of Northern blotting, gelatin zymography and monitoring the intracellular trafficking of GFP-labeled MMP-2 revealed that MMP-2 secretion by ECs was completely suppressed by high shear stress, but the intracellular mRNA expression, protein synthesis, and transport of MMP-2 were not affected. In conclusion, we suggest that high shear stress up-regulates type IV collagen synthesis and down-regulates MMP-2 secretion in ECs, which plays an important role in remodeling of the endothelial basement membrane and may suppress atherogenesis.  相似文献

We examined the hydrologic controls on nitrogen biogeochemistry in the hyporheic zone of the Tanana River, a glacially-fed river, in interior Alaska. We measured hyporheic solute concentrations, gas partial pressures, water table height, and flow rates along subsurface flowpaths on two islands for three summers. Denitrification was quantified using an in situ 15NO3 push–pull technique. Hyporheic water level responded rapidly to change in river stage, with the sites flooding periodically in mid−July to early−August. Nitrate concentration was nearly 3-fold greater in river (ca. 100 μg NO3–N l−1) than hyporheic water (ca. 38 μg NO3–N l−1), but approximately 60–80% of river nitrate was removed during the first 50 m of hyporheic flowpath. Denitrification during high river stage ranged from 1.9 to 29.4 mg N kg sediment−1 day−1. Hotspots of methane partial pressure, averaging 50,000 ppmv, occurred in densely vegetated sites in conjunction with mean oxygen concentration below 0.5 mgOl−1. Hyporheic flow was an important mechanism of nitrogen supply to microbes and plant roots, transporting on average 0.41 gNO3–N m−2 day−1, 0.22 g NH4+–N m−2 day−1, and 3.6 g DON m−2 day−1 through surface sediment (top 2 m). Our results suggest that denitrification can be a major sink for river nitrate in boreal forest floodplain soils, particularly at the river-sediment interface. The stability of the river hydrograph and the resulting duration of soil saturation are key factors regulating the redox environment and anaerobic metabolism in the hyporheic zone.  相似文献

The production of l-phenylalanine is conventionally carried out by fermentations that use glucose or sucrose as the carbon source. This work reports on the use of glycerol as an inexpensive and abundant sole carbon source for producing l-phenylalanine using the genetically modified bacterium Escherichia coli BL21(DE3). Fermentations were carried out at 37°C, pH 7.4, using a defined medium in a stirred tank bioreactor at various intensities of impeller agitation speeds (300–500 rpm corresponding to 0.97–1.62 m s−1 impeller tip speed) and aeration rates (2–8 L min−1, or 1–4 vvm). This highly aerobic fermentation required a good supply of oxygen, but intense agitation (impeller tip speed ~1.62 m s−1) reduced the biomass and l-phenylalanine productivity, possibly because of shear sensitivity of the recombinant bacterium. Production of l-phenylalanine was apparently strongly associated with growth. Under the best operating conditions (1.30 m s−1 impeller tip speed, 4 vvm aeration rate), the yield of l-phenylalanine on glycerol was 0.58 g g−1, or more than twice the best yield attainable on sucrose (0.25 g g−1). In the best case, the peak concentration of l-phenylalanine was 5.6 g L−1, or comparable to values attained in batch fermentations that use glucose or sucrose. The use of glycerol for the commercial production of l-phenylalanine with E. coli BL21(DE3) has the potential to substantially reduce the cost of production compared to sucrose- and glucose-based fermentations.  相似文献

Schizothorax chongi (locally known as Xilian Yu), a fish species commonly found in Yalong River, has been declining quickly in recent years. One of the important factors, among many, is the interruption of the free flowing river by dams. To obtain data that can be applied to the design of a fishway for S. chongi and other species in the community, a laboratory study of juvenile S. chongi’s swimming energetics and kinematics was conducted in a flume-type respirometer equipped with a high speed video camera system to record swimming behavior. The aerobic metabolic rate, tail beat frequency (TBF) and tail beat amplitude (TBA) were measured during steady swimming at varying flow rates for fish of similar mass. A power function accurately describes the relationship between oxygen consumption rate (MO2) and swimming speed (U). The estimated standard metabolic rate (SMR) calculated from the power function was 445.34 mg O2 kg−1 h−1, similar to the experimental result of 431.5 mg O2 kg−1 h−1. The relationship between cost of transport (COT) and U was, characteristically, inverse bell-shaped, with COTmin = 44.6 J kg−1 m−1 at U opt = 5.5 body lengths per second (bl s−1). There was a significant positive linear correlation between TBF and U. The slope of the correlation (0.33) was lower than for many other species, implying that S. chongi swim efficiently. The TBA, ranging from 0.15 to 0.2 bl, was found to be independent of U. Kinematic analyses indicates that S. chongi primarily depends on the caudal fin to generate forward thrust and employs three velocity-dependent swimming gaits. This investigation provides data on the swimming ability of S. chongi that will add to the basic science required for fishway design.  相似文献

Fast cyclic electron transport (CET) around photosystem I (PS I) was observed in sunflower (Helianthus annuus L.) leaves under intense far-red light (FRL) of up to 200 μmol quanta m−2 s−1. The electron transport rate (ETR) through PS I was found from the FRL-dark transmittance change at 810 and 950 nm, which was deconvoluted into redox states and pool sizes of P700, plastocyanin (PC) and cytochrome f (Cyt f). PC and P700 were in redox equilibrium with K e = 35 (ΔE m = 90 mV). PS II ETR was based on O2 evolution. CET [(PS I ETR) − (PS II ETR)] increased to 50–70 μmol e m−2 s−1 when linear electron transport (LET) under FRL was limited to 5 μmol e m−2 s−1 in a gas phase containing 20–40 μmol CO2 mol−1 and 20 μmol O2 mol−1. Under these conditions, pulse-saturated fluorescence yield F m was non-photochemically quenched; however, F m was similarly quenched when LET was driven by low green or white light, which energetically precluded the possibility for active CET. We suggest that under FRL, CET is rather not coupled to transmembrane proton translocation than the CET-coupled protons are short-circuited via proton channels regulated to open at high ΔpH. A kinetic analysis of CET electron donors and acceptors suggests the CET pathway is that of the reversed Q-cycle: Fd → (FNR) → Cyt cn → Cyt bh → Cyt bl → Rieske FeS → Cyt f → PC → P700 →→ Fd. CET is activated when PQH2 oxidation is opposed by high ΔpH, and ferredoxin (Fd) is reduced due to low availability of e acceptors. The physiological significance of CET may be photoprotective, as CET may be regarded as a mechanism of energy dissipation under stress conditions.  相似文献

An isolated, perfused salmon tail preparation showed oxyconformance at low oxygen delivery rates. Addition of pig red blood cells to the perfusing solution at a haematocrit of 5 or 10% allowed the tail tissues to oxyregulate. Below ca. 60 ml O2 kg−1 h−1 of oxygen delivery (DO2), VO2 was delivery dependent. Above this value additional oxygen delivery did not increase VO2 of resting muscle above ca. 35 ml O2 kg−1 h−1. Following electrical stimulation, VO2 increased to ca. 65 ml O2 kg−1 h−1, with a critical DO2 of ca. 150 ml O2 kg−1 h−1. Dorsal aortic pressure fell to 69% of the pre-stimulation value after 5 min of stimulation and to 54% after 10 min. Microspheres were used to determine blood flow distribution (BFD) to red (RM) and white muscle (WM) within the perfused myotome. Mass specific BFD ratio at rest was found to be 4.03 ± 0.49 (RM:WM). After 5 min of electrical stimulation the ratio did not change. Perfusion with saline containing the tetrazolium salt 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) revealed significantly more mitochondrial activity in RM. Formazan production from MTT was directly proportional to time of perfusion in both red and WM. The mitochondrial activity ratio (RM:WM) did not change over 90 min of perfusion.  相似文献

Stratified eutrophic lakes often suffer from hypolimnetic oxygen depletion during summer. This may lead to low redox conditions and accumulation of phosphate and ammonia in the hypolimnion. Hypolimnetic oxygenation has been used as a lake management strategy to improve the water quality in five eutrophic dimictic Danish lakes where oxygenation was conducted for 4–20 years. In one lake, the hypolimnetic oxygen concentration clearly improved by oxygenation, whereas the other four lakes still exhibited low mean summer levels (<2.2 mg O2 l−1). Oxygenation generally increased the hypolimnetic water temperature by 0.5–2°C, but in one lake it increased by 4–6°C. In all lakes, oxygenation significantly reduced the hypolimnetic concentrations of phosphorus and ammonia during stratification. The accumulation of phosphorus and ammonia typically decreased by 40–88%. In two lakes oxygenation was stopped for 1–2 years and here hypolimnion concentrations of both phosphorus and ammonia increased again. Surface water quality only improved in one lake, but was likely also influenced by simultaneously occurring changes in external nutrient loading. Overall, it is concluded that hypolimnetic oxygenation reduces the hypolimnetic accumulation of phosphorus and ammonia and may prevent anoxia in the deeper parts of the lake. However, long-term oxygenation is required and it is uncertain whether the overall lake water quality can be improved by oxygenation.
