1. Flagellates are positively chemotactic to a certain concentration of dissolved oxygen which is lower than that in water saturated with oxygen under atmospheric partial pressure. Consequently, when a small aquatic animal is held motionless between cover-slip and slide in a suspension of flagellates in water saturated with oxygen, the flagellates are attracted to those parts of the animal which are absorbing oxygen. The relative sizes of the flagellate aggregations then show the relative activities of the different surfaces of the animal in absorbing oxygen. 2. Applying this method to the red Chironomus larva it was found that the animal respires by the whole body surface except by the head and the "ventral gills" and that the relative intensity of oxygen intake by the different parts of the body varies in different individuals and in the same individual at different times. 3. The absence of oxygen intake by the "ventral gills" was confirmed with the microspectroscope. In oxygen-free water all the hemoglobin of the blood becomes reduced. When an air bubble is now introduced so that it touches the "ventral gills" oxyhemoglobin first appears in the nearest body segment to the bubble, not in the "gills." 4. When a small aquatic animal is held motionless between cover-slip and slide in a solution of an indicator which changes color about the neutral point of water the relative extent of color change at different surfaces of the animal''s body indicates the relative amounts of carbon dioxide given off by these surfaces. 5. Using this method with the red Chironomus larva similar conclusions were reached for carbon dioxide output as for oxygen intake.  相似文献

The axostyles of the flagellates Oxymonas, Saccinobaculus, and Notila are large ribbon-shaped structures which undulate actively in the cytoplasm. The form of their movements is described and illustrated. Axostyles consist of regular arrays of longitudinal fibres, the number of which varies between 100 and 5000 in different species. The fibres are about 240 A in diameter, apparently hollow, regularly cross-banded with a periodicity of about 150 A, and connected by delicate cross-links, also at regular intervals of about 150 A. They resemble very closely the central fibres of cilia and flagella. No other structural components are present, except at the anterior end, where the fibres are attached to one or more basal bodies, and at the posterior tip, where they are anchored to the plasma membrane. The relevance of the findings to an understanding of the mechanism of ciliary and flagellar movements is discussed.  相似文献

Oxygen consumption of luminous bacteria determined by the Thunberg micro respirometer and by the time which elapses before the luminescence of an emulsion of luminous bacteria in sea water begins to dim, when over 99 per cent of the dissolved oxygen has been consumed, agree exactly. Average values for oxygen consumption at an average temperature of 21.5°C. are 4.26 x 10–11 mg. O2 per bacterium; 2.5 x 104 mg. per kilo and 5.6 mg. O2 per sq. m. of bacterial surface. The only correct comparison of the oxygen consumption of different organisms or tissues is in terms of oxygen used per unit weight with a sufficient oxygen tension so that oxygen consumption is independent of oxygen tension. Measurement of the oxygen concentration which just allows full luminescence, compared with a calculation of the oxygen concentration at the surface of a bacterial cell just necessary to allow the observed respiration throughout all parts of the cell, indicates that oxygen must diffuse into the bacterium much more slowly than through gelatin or connective tissue but not as slowly as through chitin.  相似文献

应用双液相酶化学技术研究茶色素的形成与制取,并以单液(水)相系统进行茶多酚酶促氧化制取茶色素为对照,引入氧载体后,构成了双液相酶化学氧化系统,对其中溶解氧浓度,PPO活性,TFs,TRs含量的变化和HPLC图谱等分析研究,制品中TFs的HPLC图谱可知:TOP-1中TFs的峰高和峰面积均高于TOP-2制品中TFs,尤以TFG与TFDG较高,TOP-1制品中儿茶素类残留量较对照少,尤其是简单儿茶素的列留量(如DL-C(-)-EC)少得更多。  相似文献

The resistance to rapidly lethal concentrations of un-ionized ammonia ranging from about 2.0 to 8.8 p. p. m. N was determined for rainbow trout ( Salmo gairdnerii Richardson), perch ( Perca fluviatilis L.), roach ( Rutilus rutilus (L.)), and gudgeon ( Gobio gobio (L.)) in tensions of dissolved oxygen 53.4 and 96.7% of air-saturation value at 15.2°C.
The relation between concentration of un-ionized ammonia and period of survival from a few hours to 13 days was investigated for rainbow trout in two tensions of dissolved oxygen, 45.7 and 100.3% of air-saturation value, at 20.1°C.
Period of survival of all the species tested decreased with rise in concentration of un-ionized ammonia.
Decrease in tension of dissolved oxygen increased the toxicity of un-ionized ammonia to all species except gudgeon, for which there was no significant difference in period of survival at the two levels of oxygen.
The effect of oxygen tension on period of survival was greatest in the lowest concentrations of un-ionized ammonia.
Resistance of perch and roach to lack of oxygen was not significantly affected, but that of rainbow trout was reduced significantly, by the presence of a small concentration of ammonia (not toxic to trout within at least 13 days).  相似文献

The ecology of epipelic algae on the marginal sediments of five Welsh lakes was studied over an annual cycle. The lakes, Llydaw, Cwellyn, Padarn, Maelog and Coron ranged from very oligotrophic to nutrient-rich. Attention was focussed on chlamydomonad flagellates, diatoms, blue-green algae and euglenoids and the different proportions of these in algae in the epipelon of lakes of contrasting water quality. A total of 75 algal taxa was found in the five lakes, 25 were species of volvocalean flagellates. Mean annual population density of these flagellates differed by an order of magnitude between the lakes. The greatest population density was recorded for Chlamydomonas anticontata Schiller in nutrient-rich Llyn Maelog. Twenty species of pennate diatoms were recorded frequently in the epipelon. In the nutrient-rich lakes, Maelog and Coron, pennate diatoms were dominant on the sediments, where they exhibited population maxima in spring and autumn. Increase in numbers of epipelic diatoms was recorded when silica concentrations were minimum in the overlying lake waters. Navicula hungarica Grun. achieved the maximum population density, 260 000 cells · cm?2. Euglenoids formed large epipelic populations during late-summer and autumn in these nutrient-rich lakes. Blue-green algae were more important, proportionally, in the nutrient-poor mountain lakes, which had sediments of higher organic content. Chlamydomonads were the major algal component of the epipelon in the mountain lakes, Llydaw and Cwellyn, where the sediments were characterized by larger particle size, and higher organic content. In the nutrient-rich lakes, where the sediments had higher calcium content, chlamydomonads formed significant populations only during spring and summer, when nutrient levels were minimal in the overlying lakewaters.  相似文献

The reaction kinetics for phenol biodegradation at low substrate concentrations can be estimated based on the analysis of changes in the dissolved oxygen concentration in the bulk liquid during biodegradation. The measured oxygen concentration changes with an interesting behavior as biodegradation proceeds. The oxygen concentration in the bulk liquid decreases rapidly in the early stages of degradation and subsequently decreases linearly and then rapidly recovers to the initial saturated level. Taking into account the oxygen transfer rate between gas and liquid phases and oxygen consumption rate by microbes, the change in the dissolved oxygen concentration can be simulated with an unsteady state mass balance equation and three kinetic models for the rate of phenol metabolism: a substrate-inhibited model; a zero-order model; and a combined model. In the combined model, it is assumed that, at phenol concentrations above 10 mg/L, the degradation rate is expressed by a substrate-inhibited model; whereas at concentrations below 10 mg/L the zero-order model is applied. It was found that the characteristics of the change in the dissolved oxygen concentration, especially the rapid increase at the end of degradation, can only be described by the combined kinetic model. This result suggests that conventional Haldane-type kinetics would be unsuitable for estimating the phenol consumption rate at low phenol concentrations, in particular, at concentrations less than 10 mg/L. 根据1979—1984年对主养鲢、鳙、非鲫,主养青、草鱼,主养青鱼3种养殖结构类型高产鱼池溶氧变化周期的系统研究,揭示了高产养鱼池塘溶氧的昼夜、垂直、水平变化及季节变化周期,分析了光合作用、呼吸作用、扩散作用在高产养鱼池塘溶氧动力学上的地位和作用。对高产养鱼池塘的溶氧收入、支出及平衡情况进行了定量研究,在主要饲养季节,测得晴到多云天、晴天的溶氧来源分别为:浮游植物光合产氧占86.0和95.3%,大气扩散溶入占14.0和4.7%;氧的消耗分别为:“水呼吸”消耗氧占72.0和72.0%,鱼类呼吸消耗氧占22.0和13.1%,淤泥中生物呼吸消耗氧占2.9、5.5%,扩散逸出占3.1、8.8%。文中还对溶氧变化周期与养鱼池塘管理的环境控制、结构控制之间的关系进行了分析。    相似文献

Pelagic ecosystems can become depleted of dissolved oxygen as a result of both natural processes and anthropogenic effects. As dissolved oxygen concentration decreases, energy shifts from macrofauna to microorganisms, which persist in these hypoxic zones. Oxygen-limited regions are rapidly expanding globally; however, patterns of microbial communities associated with dissolved oxygen gradients are not yet well understood. To assess the effects of decreasing dissolved oxygen on bacteria, we examined shifts in bacterial community structure over space and time in Hood Canal, Washington, USA−a glacial fjord-like water body that experiences seasonal low dissolved oxygen levels known to be detrimental to fish and other marine organisms. We found a strong negative association between bacterial richness and dissolved oxygen. Bacterial community composition across all samples was also strongly associated with the dissolved oxygen gradient, and significant changes in bacterial community composition occurred at a dissolved oxygen concentration between 5.18 and 7.12 mg O2 L-1. This threshold value of dissolved oxygen is higher than classic definitions of hypoxia (<2.0 mg O2 L-1), suggesting that changes in bacterial communities may precede the detrimental effects on ecologically and economically important macrofauna. Furthermore, bacterial taxa responsible for driving whole community changes across the oxygen gradient are commonly detected in other oxygen-stressed ecosystems, suggesting that the patterns we uncovered in Hood Canal may be relevant in other low oxygen ecosystems.  相似文献

Photoresponsive behavior in phytoplanktonic flagellates potentially controls depth regulation, vertical migration, and the accumulation of cells in preferred conditions and hence has major implications for photosynthesis, growth, and species competition. This study used microscale laboratory chambers with cell track analysis and mesoscale lake studies to investigate the responses of five phylogenetically contrasting freshwater flagellates to gradients of light. Laboratory results demonstrated that these species differed in their light preferences despite being grown at the same photon irradiance. Preferred photon irradiances were 20–120, 20, 65–120, 4–20, and>200 μmol photons·m?2·s?1, respectively, for Ceratium furcoides (Levander) Langhans, Chlamydomonas moewusii Gerloff, Dinobryon sertularia Ehrenberg, Euglena gracilis Klebs, and Plagioselmis nannoplanctica (Skuja) Novarino. Analysis of the response of individual cells showed that in all species, photoresponsive preference was the result of positive and negative phototaxis combined with step‐up and step‐down photophobic reactions. There was no evidence for photokinesis or cell memory. Only in C. furcoides did the preferred photon irradiance alter with growth conditions. In C. furcoides, D. sertularia, and P. nannoplanctica, irradiance preference matched the optimal irradiance for growth, whereas in the two remaining species a lower than optimal irradiance was preferred, suggesting that light may be used as an indicator of other ecological conditions. Mesoscale experiments in a lake demonstrated that the laboratory microscale measurements provide information relevant to understanding ecological distributions. Behavioral responses to light contribute to the delineation of vertical niche separation and provide a method for predicting the spatial and temporal distribution of flagellates.  相似文献

In aerobic bioprocesses, oxygen is a key substrate; due to its low solubility in broths (aqueous solutions), a continuous supply is needed. The oxygen transfer rate (OTR) must be known, and if possible predicted to achieve an optimum design operation and scale-up of bioreactors. Many studies have been conducted to enhance the efficiency of oxygen transfer. The dissolved oxygen concentration in a suspension of aerobic microorganisms depends on the rate of oxygen transfer from the gas phase to the liquid, on the rate at which oxygen is transported into the cells (where it is consumed), and on the oxygen uptake rate (OUR) by the microorganism for growth, maintenance and production.  相似文献

本文采用离体鱼头灌注法,以两个造纸厂的制浆造纸废水对147个鲤鱼头进行了人工灌流。结果表明:废水在低浓度(如4%以下)体积稀释液时,鱼头呼吸速率基本未产生影响,而以较高浓度的废水(如5%以上)稀释液灌流与不含废水的鸭绿江水源地水灌流的对照组鱼头比较,几乎均发生了显著性变化,即对鱼鳃盖运动产生了抑制作用,出现了Biot氏呼吸。以往学者们认为,造纸废水主要是由于水中溶解氧(DO)减少而使鱼类致死的,但本工作灌注液中已人工给予足够的DO,提示:有毒物质本身似已直接危及延髓呼吸中枢。 实验同时表明,离体鱼头灌注法可供环境保护部门应用于含多种有机毒性物质的制浆造纸废水的现场监测。  相似文献

Diatoms, but not flagellates, have been shown to increase rates of nitrogen release after a shift from a low growth irradiance to a much higher experimental irradiance. We compared NO3 ? uptake kinetics, internal inorganic nitrogen storage, and the temperature dependence of the NO3 ? reduction enzymes, nitrate (NR) and nitrite reductase (NiR), in nitrogen‐replete cultures of 3 diatoms (Chaetoceros sp., Skeletonema costatum, Thalassiosira weissflogii) and 3 flagellates (Dunaliella tertiolecta, Pavlova lutheri, Prorocentrum minimum) to provide insight into the differences in nitrogen release patterns observed between these species. At NO3 ? concentrations <40 μmol‐N·L ? 1, all the diatom species and the dinoflagellate P. minimum exhibited saturating kinetics, whereas the other flagellates, D. tertiolecta and P. lutheri, did not saturate, leading to very high estimated K s values. Above ~60 μmol‐N·L ? 1, NO3 ? uptake rates of all species tested continued to increase in a linear fashion. Rates of NO3 ? uptake at 40 μmol‐N·L ? 1, normalized to cellular nitrogen, carbon, cell number, and surface area, were generally greater for diatoms than flagellates. Diatoms stored significant amounts of NO3 ? internally, whereas the flagellate species stored significant amounts of NH4 + . Half‐saturation concentrations for NR and NiR were similar between all species, but diatoms had significantly lower temperature optima for NR and NiR than did the flagellates tested in most cases. Relative to calculated biosynthetic demands, diatoms were found to have greater NO3 ? uptake and NO3 ? reduction rates than flagellates. This enhanced capacity for NO3 ? uptake and reduction along with the lower optimum temperature for enzyme activity could explain differences in nitrogen release patterns between diatoms and flagellates after an increase in irradiance.  相似文献

1. The oxygen consumption of cultural forms of Trypanosoma cruzi decreases in intensity with increasing age of the cultures; no correlation with any other factor studied could be established. 2. The respiratory quotient was high for the first 10 days, i.e. as long as the population increased; with the onset of a decline in numbers, the R.Q. began to drop. It is believed that the flagellates consume in the beginning predominantly sugar and later predominantly protein. Observations on the pH of the cultures bear out this view. 3. The oxygen consumption was independent of the oxygen tension over a wide range of tensions. 4. The oxygen consumption increased in the temperature range 13° to 40°C., while a temperature of 44°C. proved to be lethal. Upon application of Arrhenius'' equation, two straight lines, intersecting at about 28°C., resulted. The µ values were 23,980 and 5275 for the lower and higher temperature range respectively. 5. Of the oxidase inhibitors tested, strong inhibition of the oxygen consumption was achieved with azide, cyanide, and hydrogen sulfide. Pyrophosphate had no influence at all. There is some probability that cytochrome oxidase is the chief oxidase present. 6. The strongest inhibitory influence due to dehydrogenase inhibitors was observed with propyl carbamate and high concentrations of ethyl carbamate. 7. A small fraction of the oxygen consumption, about 10 per cent, may be due to substances with sulfhydryl groups, as indicated by a slight but distinct inhibition due to dilute iodoacetate and to arsenite.  相似文献

1. The ability of marine fishes to absorb oxygen at low tension from the sea water is more or less dependent upon the hydrogen ion concentration of the water. 2. The ability of fishes to withstand wide variations in the range of hydrogen ion concentration of the sea water can be correlated with their habitats. The fishes that are most resistant to a wide variation in the hydrogen ion concentration are most cosmopolitan in their habitat. Those that are least resistant to a variation in the hydrogen ion concentration are the most restricted in their range of habitat. 3. There is a close correlation between the optimum condition of the sea water for the absorption of oxygen at low tension by the herring (Clupea pallasii), the condition of the sea water to which they react positive and that in which they are found most abundantly. 4. It is suggested that the variation in the ability to absorb oxygen at low tension at a given pH of individuals of a species is dependent upon the alkaline reserve of the blood of the individual fish.  相似文献

1. The respiration of luminous bacteria has been studied by colorimetric and manometric methods. 2. Limulus oxyhaemocyanin has been used as a colorimetric indicator of oxygen consumption and indicator dyes were used for colorimetric determination of carbon dioxide production. 3. The Thunberg-Winterstein microrespirometer has been used for the measurement of the rate of oxygen consumption by luminous bacteria at different partial pressures of oxygen. 4. The effect of oxygen concentration upon oxygen consumption has been followed from equilibrium with air to low pressures of oxygen. 5. Luminous bacteria consume oxygen and produce carbon dioxide independent of oxygen pressures from equilibrium with air (152 mm.) to approximately 22.80 mm. oxygen or 0.03 atmosphere. 6. Dimming of a suspension of luminous bacteria occurs when oxygen tension is lowered to approximately 2 mm. Hg (0.0026 atmosphere) and when the rate of respiration becomes diminished one-half. 7. Pure nitrogen stops respiratory activity and pure oxygen irreversibly inhibits oxygen consumption. 8. The curve for rate of oxygen consumption with oxygen concentration is similar to curves for adsorption of gasses at catalytic surfaces, and agrees with the Langmuir equation for the expression of the amount of gas adsorbed in unimolecular layer at catalytic surfaces with gas pressure. 9. A constant and maximum rate of oxygen consumption occurs in small cells when oxygen concentration becomes sufficient to entirely saturate the surface of the oxidative catalyst of the cell.  相似文献

The relative importance of dissolved oxygen and dissolved carbon dioxide in determining the lethal effect of an environment for rainbow trout and perch has been investigated with an apparatus which controls the concentrations of these gases in a body of water. It is shown that concentrations of carbon dioxide which sometimes occur in polluted streams can more than double the minimum concentration of dissolved oxygen necessary for the survival of half a population of rainbow trout fingerlings for 24 hr. Increase in temperature between 12·5 and 19·5°C. shortens period of survival in solutions containing up to 67 p. p. m. CO2. Within the range of dissolved oxygen concentration which is lethal in the presence of 59 p. p. m. CO2 or more, perch are more resistant than rainbow trout in the lower, but less resistant in the higher, oxygen concentrations. The relation between carbon dioxide concentration and the oxygen tension at which rainbow trout blood is half saturated with oxygen is similar to the relation between carbon dioxide concentration and the oxygen tension at which the median period of survival of this species is I hr.  相似文献
