The cell has been represented as a charged liquid drop. Contrary to the DLVO-theory, the effect of the surface potential upon the value of the interfacial tension of the cell membrane has also been taken into consideration. The cell membrane has visco-elastic properties and its constituents may move against each other. Cell movement is caused by the appearance of a small number of the electrically charged constituents of the cell membrane on the leading edge of the cell. This produces a local decrease in the surface tension and the cell membrane expansion. At the moment of contact between two cells proton transfers occur between the strongly negatively charged microvilli of one cell and the body of the other, analogous to a condenser breakdown. This, through the effect on the surface tension, causes contact inhibition of movement. The distribution of the proton dissociable groups modifies the interaction between the cells (differentiation) and between the cell and the substratum (adhesion). Adsorption of the charged compounds at the surface of the cell membrane, decreasing the surface potential and increasing the surface tension, causes the phenomena of chemotaxis, phagocytosis and pinocytosis. Cell division, considered in the terms of the surface energy, requires an adequate supply of considerable quantities of energy inversely proportional to the surface potential value. In case of a reduction of the distance between the cells, their surface potential and the energetic barrier of the cell division processes increases, and causes contact inhibition of cell division. Due to their high charge, division of neoplastic cells is inhibited much later than division of normal cells, or is completely ininhibited due to geometric conditions. Fusion of the cell membrane in the intra-cellular and intercellular processes is a reverse process in relation to the cell division.  相似文献

Leishmania major Friedlin (LmjF) is a protozoan parasite whose genomic sequence has been recently elucidated. Here we have cloned, overexpressed, purified, and characterized the product of the gene from LmjF chromosome 16: LmjF16.0530, which encodes a protein with putative dihydroorotate dehydrogenase activity. Dihydroorotate dehydrogenase (DHODH) is a flavoprotein that catalyses the oxidation of L-dihydroorotate to orotate, the fourth sequential step in the de novo pyrimidine nucleotide synthesis pathway. The predicted enzyme from L. major was cloned and expressed in Escherichia coli strain BL21(DE3) as a histidine-tag fusion protein and purified to homogeneity using affinity chromatography. The final product was homogeneous in SDS-PAGE gel electrophoresis. The dihydroorotate oxidase activity has been assayed and the steady-state kinetic mechanism has been determined using fumarate as the oxidizing substrate. The catalysis by LmDHODH enzyme proceeds by a Ping-Pong Bi-Bi mechanism and the kinetic parameters Km were calculated to be 90 and 418 microM for dihydroorotate and fumarate, respectively, and Vmax was calculated to be 11 micromol min-1 mg-1. Our results confirmed that the product of the gene LmjF16.0530, whose function has previously been predicted based on homology to known proteins, can therefore be positively assigned as L. major DHODH.  相似文献

Ewaso Narok Swamp, formed along the Eng'are Narok river, is located in the semi-arid part of the Laikipia District, Kenya. The area, of bushy grassland, is characterised by low rainfall (less than 750 mm annually) and episodic rivers. Before the 1970s, the dominant land use was large scale ranching and nomadic pastoralism. Since 1970, this has slowly been transformed into high density small-scale farming. There has been a strong trend towards settlement along riverine and wetland areas due to their suitability for farming and easy availability of water for cultivation. Ewaso Narok swamp has a rich species diversity of over 170 bird species, resident and migrant, over 100 plant species and it also provides an important dryland refuge for both domestic and wild animals. The swamp also provides socio-economic products such as plant matter for building. The result of its land use transformation has been ecosystem alteration, habitat modification and destruction both for wetland and rangeland species. This change has also been accompanied by escalating human–wildlife conflict. However, although this process is self-destructive, the lack of economic returns from wildlife to some extent justify the land use transformation since the communities settling here have to satisfy the basic requirements of food and shelter. This poses the challenge of the developing appropriate ways to conserve the dryland wetlands whilst attaining maximum returns for the local community.  相似文献

Slapton Ley, a coastal lake in SW England, has been shown by a variety of paleolimnological studies, to have become increasingly eutrophic in the period since 1950 AD. Since that time, intensification of agriculture has resulted in increased erosion of topsoil from fields in the catchment of the Ley. Sediment accumulation rates, as estimated by 210Pb-dating and multiple core correlation of peaks in whole core volume magnetic susceptibility, are equivalent to a catchment erosion rate of 13.4 t km–2 a–1, which figure agrees well with directly monitored data. Diatom and chlorophyll a analysis of the uppermost sediments shows that the Ley has recently experienced a major shift in its trophic status, changing from a clear water, macrophyte lake to one dominated by plankton in a hypertrophic system. This last point is further amplified in the paper by Heathwaite & O'Sullivan (1991).  相似文献

In the United States alone, the National Heart, Lung, and Blood Institute (NHLBI) has invested several hundred million dollars in pursuit of myocardial infarct-sparing therapies. However, due largely to methodological limitations, this investment has not produced any notable clinical application or cardioprotective therapy. Among the major methodological limitations is the reliance on animal models that do not mimic the clinical situation. In this context, the limited use of conscious animal models is of major concern. In fact, whenever possible, studies of cardiovascular physiology and pathophysiology should be conducted in conscious, complex models to avoid the complications associated with the use of anesthesia and surgical trauma. The mouse has significant advantages over other experimental models for the investigation of infarct-sparing therapies. The mouse is inexpensive, has a high throughput, and presents the ability of one to create genetically modified models. However, successful infarct-sparing therapies in anesthetized mice or isolated mouse hearts may not be successful in more complex models, including conscious mice. Accordingly, a conscious mouse model of myocardial ischemia and reperfusion has the potential to be of major importance for advancing the concepts and methods that drive the development of infarct-sparing therapies. Therefore, we describe, for the first time, the use of an intact, conscious, and unrestrained mouse model of myocardial ischemia-reperfusion and infarction. The conscious mouse model permits occlusion and reperfusion of the left anterior descending coronary artery in an intact, complex model free of the confounding influences of anesthetics and surgical trauma. This methodology may be adopted for advancing the concepts and ideas that drive cardiovascular research.  相似文献

The rat liver prolactin receptor has been purified to homogeneity, and partial amino acid sequences have been obtained. The structure of the receptor has been deduced from a single complementary DNA clone. The mature protein of 291 amino acids has a relatively long extracellular region, a single transmembrane segment, and a short (57 amino acids) cytoplasmic domain. With the rat cDNA used as a probe, the prolactin receptor in rabbit mammary gland and human hepatoma cells has also been isolated. These tissues contain a second, longer form of the receptor (592 and 598 amino acids, respectively). Both the short and long forms of the prolactin receptor show regions of strong sequence identity with the human and rabbit growth hormone receptors, suggesting that the prolactin and growth hormone receptors originate from a common ancestor.  相似文献

The stability between helical conformations of homopeptides of alanine, glycine, and aminoisobutyric acid has been studied by means of quantum-mechanical methods. The influence of peptide length on the relative stability between helical conformations has also been analyzed by means of systematic studies for peptides of size up to 11 residues. Finally, the influence of the solvent has been examined by using self-consistent reaction field methods. A vast amount of literature has accumulated on the characterization of DNA methyltransferases. The HhaI DNA methyltransferase, a C5-cytosine methyltransferase, has been the subject of investigation for the last 2 decades. Biochemical and kinetic characterization have led to an understanding of the catalytic and kinetic mechanism of the methyltransfer reaction. The HhaI methyltransferase has also been subjected to extensive structural analysis, with the availability of 12 structures with or without a cofactor and a variety of DNA substrates. The mechanism of base flipping, first described for the HhaI methyltransferase, is conserved among all DNA methyltransferases and is also found to occur in numerous DNA repair enzymes. Studies with other methyltransferase reveal a significant structural and functional similarity among different types of methyltransferases. This review aims to summarize the available information on the HhaI DNA methyltransferase.  相似文献

An isolated population of red lechwe living in the Busanga Plain, Zambia, has been protected from human hunting since 1948. Probably as a result the population has increased, as indicated by reliable census data. From 1956 to 1972 this increase has very closely fitted an exponential rate; the calculated value of the infinitesimal rate of population increase (r) is 0–131, and the finite rate of increase is 14-0% per year. This exponential phase is considered to represent a segment of a logistic curve.  相似文献

Holohalaelurus regani was caught in 38% of the 3314 bottom trawls conducted during routine demersal surveys o. the South African west and south coasts from 1986 to 1999. An index of biomass for H. regani has increased on the west coast, from 1606 t in 1986–1993 to 3012 t in 1994–1999, despite c. 130 t being taken annually as by–catch in the demersal fishery. On the south coast, there has also been an increase over the same period, from 793 to 1350 t. Females and juveniles were generally found in shallower water (<300 m) than males, suggesting an inshore nursery area. Male H. regani become mature at 450–500 mm LT, whereas females become mature at 400–450 mm LT. There is reproductive activity throughout the year and fecundity appears to be high. This species is a generalist feeder, with the diet comprising teleosts, crustaceans and cephalopods. H. regani also scavenges offal opportunistically. Its high fecundity, the relative protection of females and juveniles in shallow water that is rarely trawled, its opportunistic diet and its robust nature that may allow it to survive after it has been discarded, have enabled H. regani to increase in numbers, despite indirect fishing pressure.  相似文献

Human alpha 1-antichymotrypsin has been cloned, sequenced and expressed in Escherichia coli and recombinant protein as well as point-specific mutants have been purified and characterized. The corrected gene-deduced amino acid sequence has 45% overall identity with alpha 1-protease inhibitor, which is higher than the 42% previously reported (Chandra, T., Stackhouse, R., Kidd, V. J., Robson, J. H., and Woo, S. L. C. (1983) Biochemistry 22, 5055-5060). Recombinant antichymotrypsin (rACT) is similar to natural antichymotrypsin with respect to the specificity of its interactions with proteases. Its second-order rate constant for association with bovine chymotrypsin is 6-8 x 10(5) M-1 s-1, which is identical to that of the serum-derived inhibitor. Site-specific mutagenesis has been used to produce two variants of rACT in which the P1 position has been changed from leucine to either methionine (L358M-rACT) or arginine (L358R-rACT). L358M-rACT has a specificity of inhibitory activity toward serine proteases closely similar to that of native rACT. By contrast, the specificity of L358R-rACT is quite different from that of native rACT, most notably in efficiently inhibiting trypsin and human thrombin while showing a decreased ability to inhibit chymotrypsin.  相似文献

A gene coding for lipase-solubilized bovine liver microsomal cytochrome b5 has been synthesized, expressed in Escherichia coli, and mutated at functionally critical residues. Characterization of the recombinant protein revealed that it has a reduction potential that is approximately 17 mV lower than that of authentic wild-type protein at pH 7 (25 degrees C). Structural studies determined that the recombinant protein differed in sequence from authentic wild-type cytochrome b5 owing to three errors in amidation status in the published sequence for the protein on which the gene synthesis was based. The structural origin of the lower reduction potential exhibited by the triple mutant has been investigated through X-ray crystallographic determination of the three-dimensional structure of this protein and is attributed to the presence of Asp-57 within 3.3 A of heme vinyl-4 in the mutant. In addition, the model developed by Argos and Mathews [Argos, P., & Mathews, F.S. (1975) J. Biol. Chem. 250, 747] for the change in cytochrome b5 oxidation state has been studied through mutation of Ser-64 to Ala. In this model, Ser-64 is postulated to stabilize the oxidized protein through H-bonding interactions with heme propionate-7 that orients this propionate group 6.2 A from the heme iron. Spectroelectrochemical studies of a mutant in which Ser-64 has been changed to an alanyl residue demonstrate that this protein has a reduction potential that is 7 mV lower than that of the wild-type protein; moreover, conversion of the heme propionate groups to the corresponding methyl esters increases the potential by 67 mV.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Abstract

The validity of the monotypic ichnogenus Thatchtelithichnus has been called into question due to an inference that it is a bioclaustration. Close analysis of the Thatchtelithichnus holmani holotype reveals that no part of this ichnotaxon involves a bioclaustration, and thus it remains a valid ichnotaxon. Thatchtelithichnus holmani has been observed on the bottom shell (plastron) of cryptodiran and pleurodiran turtles in North America and Egypt where it is interpreted to represent an attachment trace of aquatic ectoparasites. It has also been observed on dinosaur long bones where it may represent the activity of beetle larvae.  相似文献

The neutral mononuclear vanadyl complex with the quinolone antibacterial drug enrofloxacin has been prepared and characterized with physicochemical and spectroscopic techniques and molecular mechanics calculations.
