A method is reported for the isolation of `vacuoplasts' from Poterioochromonas malhamensis. Vacuoplasts are separated mechanically by centrifugation on silica-sol gradients. They consist of the leucosin storage vacuole, a portion of the plasma membrane, and some cytoplasmic components. This method is suited to give a high yield of vacuoplasts.  相似文献

We isolated hybridomas that produced monoclonal antibodies specific for the UDP-galactose: sn -glycerol-3-phosphate α-D-galactosyltransferase (IFP synthase, EC 2.4.1.96), an enzyme involved in the volume regulation of Poterioochromonas malhamensis Peterfi. Western blotting of native gradient gels with the most reactive antibody S 162 revealed several immunoreactive proteins in crude homogenates suggesting the occurrence of multiple molecular mass species of the galactosyltransferase. The amount of the presumed enzyme monomer (64 kDa under native conditions) was strongly increased by a pH shift of crude homogenates from pH 8 to 6. During activation of the galactosyltransferase in the cell homogenate and also by shrinking the cells, the presumed enzyme monomer appeared to be proteolytically degraded generating stepwise products of 52 and 40 kDa. We assume that the proteolytically processed enzyme becomes highly active, but is very susceptible to further proteolytic degradation.  相似文献

Fixation of the chrysophycean flagellate Poterioochromonas malhamensis in the presence of potassium (pyro) antimonate have been carried out. Intense antimonate deposits are characteristic of the various elements of the contractile vacuole as well as a portion of the plasma membrane adjacent to the contractile vacuole. Neither pretreatment of the cells with EGTA or EDTA nor the treatment of sections with these chelators is successful in removing the antimonate precipitates. Despite this, X-ray microanalyses of these deposits indicate the presence of calcium.  相似文献

Changes in membrane topography in the flagellate Poterioochromonas malhamensis, as a result of colchicine and osmotic-stress treatments, have been studied using freeze-fracturing and thin sectioning. Ridges, but not rows of intramembrane particles, in the PF-face which denote the position of underlying cortical microtubules, together with the ridge associated with their point of origin (flagellar root fibre 1), dissappear after colchicine or short-term (5 min) osmotic treatments. Cortical microtubules are destroyed as a result of the former, but not the latter treatment. Longer periods in osmoticum allow a recovery of the microtubule — associated membrane ridges. Despite careful isosmotic fixations distinct cross-bridges between microtubules and the plasmalemma were not discernible in thin section.  相似文献

1. Previous studies of mixotrophy in the flagellate Poterioochromonas malhamensis (Chrysophyceae) were performed on strains that had been in culture for > 30 years. This study aims to compare mixotrophy in a cultured strain with one recently isolated from a mesotrophic lake (Lacawac) in Pennsylvania, U.S.A. 2. P. malhamensis from the lake exhibited a nutritional flexibility similar to that of the culture strain, growing phototrophically but inefficiently in comparison to other nutritional modes (growth rate (μ) = 0.015 h?1). Supplementing an inorganic salts medium with 1 mM glucose resulted in a doubling of μ to 0.035 h?1 and 0.033 h?1 in the light and the dark, respectively. Addition of an algal prey, Nannochloris, to the inorganic salts medium increased growth to rates similar to those observed with glucose. Maximum growth of the lake strain, 0.095 h?1, was achieved when bacteria was supplied as food. During growth on bacteria, cellular chlorophyll a (Chl a) decreased from 140 fg cell?1 to 10 fg cell?1 over 22 h when cultured either in the light or dark. In illuminated cultures, cell-specific Chl a concentration recovered to 185 fg cell?1 after bacteria became limiting. 3. In contrast to the cultured strain, however, the lake isolate exhibited an inverse relationship between light intensity and ingestion rate. Calculated grazing rates, based upon the ingestion of fluorescently labeled bacteria, were 3.2, 5.2 and 9.4 bacteria flagellate?1 h?1, for P. malhamensis incubated in high light, low light and darkness, respectively. Phagotrophy is thus influenced by a light regime in this predominately heterotrophic mixotroph.  相似文献

Algal blooms have become a worldwide issue recently, especially those comprised of toxic cyanobacteria. Grazers' predation of bloom-forming algae plays an important role in water clearing. In this study, a species of golden alga (strain ZX1), capable of feeding on the toxic cyanobacteria Microcystis aeruginosa, was isolated and identified as Poterioochromonas sp. (GenBank accession: EU586184) on the basis of morphological characteristics and 18s rRNA gene sequencing. Feeding experiments showed that ZX1 could clear high densities of M. aeruginosa (7.3 x 10(5)-4.3 x 10(6) cells mL(-1)) in a short time (40 h), with inhibition ratios higher than 99.9%. ZX1 grew during the feeding processes and achieved a maximum density of 10-20% of the initial M. aeruginosa density. Furthermore, this study is the first to report that ZX1 was able to degrade microcystin-LR (MC-LR) in cells of M. aeruginosa while digesting the whole cells, and that the degradation process was determined to be carried out inside the ZX1 cell. For a total MC-LR (intra- and extracellular) concentration of up to 114 microg L(-1), 82.7% was removed in 40 h. This study sheds light on the importance of golden alga in aquatic microbial ecosystems and in the natural transportation/transformation of MC-LR.  相似文献

We investigated the effect of pH on the competition of two closely related chrysomonad species, Poterioochromonas malhamensis originating from circumneutral Lake Constance, and Ochromonas sp. isolated from a highly acidic mining lake in Austria (pH ∼2.6). We performed pairwise growth experiments between these two species at four different pH ranging from 2.5 to 7.0. Heterotrophic bacteria served as food for both flagellates. Results were compared to growth rates measured earlier in single species experiments over the same pH range. We tested the hypothesis that the acidotolerant species benefits from competitive release under conditions of acid stress. The neutrophilic strain numerically dominated over the acidotolerant strain at pH 7.0, but was the inferior competitor at pH 2.5. At pH 3.5 and 5.0 both strains coexisted. Surprisingly, P. malhamensis prevailed over Ochromonas sp. under moderately acidic conditions, i.e. at the pH where growth rates of the latter peaked when grown alone. Since bacterial food was not limiting, resource competition is improbable. It appears more likely that P. malhamensis ingested cells of its slightly smaller competitor. Adverse effects mediated via allelopathy, either directly on the competing flagellate or indirectly by affecting its bacterial food, might also have affected the outcome of competition.  相似文献

Depletion of glucose from the culture medium by Poterioochromonas malhamensis results in cessation of growth and accumulation of cyclic adenosine 3′:5′-monophosphate (cAMP), followed by formation of chlorophyll and an increase in extracellular cAMP. Readdition of glucose to the culture medium causes P. malhamensis to release its intracellular cAMP into the medium. These results suggest that formation of the photosynthetic apparatus in P. malhamensis may be repressed by glucose, and that high cAMP conveys the regulatory information that the glucose supply is inadequate. This pattern is reminiscent of cAMP-mediated escape from catabolite repression in bacteria.  相似文献

Algal heterotrophy is a potentially important considerationin the flow of carbon through aquatic food webs. The physiologicalresponses to organic compound additions under various lightintensities were examined with Poterioochromonas malhamensis,a freshwater chrysophyte with an exceptionally high heterotrophiccapability. P. malhamensis demonstrated a much greater potentialfor heterotrophic growth than for photoautotrophic growth. Whenorganic substrates (glucose, glycerol, or ethanol) were addedto the culture medium, the growth rate of P. malhamensis significantlyincreased while the chlorophyll content cell –1 decreased,even at light intensities saturating for photoautotrophic growth.After an initial decline in chlorophyll production caused byorganic substrate uptake, chlorophyll cell1 increased and theuptake rate of organic substrates decreased, despite the persistenceof a relatively high substrate concentration in the medium.The results are consistent with the production of substance(s)by P. malhamensis that conditioned the culture medium, leadingto a relief of the inhibitory effect of organic substrates onchlorophyll production. Controlling glucose feeding rate, according to the change of ethanol concentration and respiratory quotient (RQ), has been studied in high-cell-density cultivation of Saccharomyces cerevisiae for glutathione (GSH). GSH yield and dry cell weight reached 1620 mg/L and 140 g/L, respectively, after 52 h of cultivation. In addition, optimized single shot addition of precursor amino acids (performed in both flask and fermentor experiments) at 32 h could result in the GSH yield reached 2020 mg/L after 38 h of cultivation. The yield and productivity of GSH increased 25% and 70%, respectively. Moreover, cultivation time was reduced to 38 h, compared with results without adding the precursor amino acids.  相似文献

Summary The viability of the chrysophycean flagellate,Poterioochromonas malhamensis, was examined after treatment of the cells with high temperatures. The fine structure of the cells was studied after heat-shock (42 °C, 16 minutes). Heat injury effects are visible at the nucleus, the chloroplast (distortion of the thylakoids), the mitochondria (increased occurrence of matrical crystalline inclusions) and, especially, at the dictyosome which is completely destroyed into some single vesicles and remnants of cisternae. Most damages are repaired within one hour; the reconstitution of the dictyosome takes 3–6 hours. It is inhibited severely by actinomycin D.The effect on the dictyosome reflects its labile position in the endomembrane system. The dependence of its reconstitution on protein synthesis indicates that membrane components are destroyed by the heat-shock.  相似文献

High-cell-density fed-batch cultures of Pseudomonas putida were carried out for the production of medium-chain-length polyhydroxyalkanoates (PHAs) using oleic acid as a carbon source. By employing an optimal feeding strategy without the limitation of any nutrient, a high cell concentration of 173 g/L was achieved, but the PHA concentration and PHA content were only 32.3 g/L and 18.7 wt%, respectively. To increase the PHA concentration and content, phosphorus limitation was applied during fed-bath culture by reducing the initial KH(2)PO(4) concentration. When the initial KH(2)PO(4) concentration was reduced to 4 g/L, cell concentration, PHA concentration, and PHA content obtained in 38 h were 141 g/L, 72. 6 g/L, and 51.4 wt%, respectively, resulting in a high productivity of 1.91 g PHA/L per hour.  相似文献

Four automatic substrate feeding strategies were developed and investigated in this study to obtain rapid, repeatable, and reliable high cell densities of Pseudomonas putida KT2440 from glucose. Growth yield data of the key nutrients, Y X/Glucose, Y X/NH4, Y X/PO4, Y X/Mg, and Y CO2/Glucose, were determined to be 0.41, 5.44, 13.70, 236, and 0.65 g g−1, respectively. Although standard exponential feeding strategy worked well when the predetermined μ was set at 0.25 h−1, an exponential glucose feeding strategy with online μ max estimation resulted in a higher average biomass productivity (3.4 vs 2.8 g l−1 h−1). A CO2 production rate based pulse glucose feeding strategy also resulted in good overall productivity (3.0 g l−1 h−1) and can be used as an alternative to pH-stat or DO-stat feeding. A cumulative CO2 production based continuous feed with real-time cumulative glucose consumption estimation resulted in much higher biomass productivity (4.3 g l−1 h−1) and appears to be an excellent and reliable approach to fully automating high-cell-density fed-batch cultivation of P. putida.  相似文献

Significant leakage of host-cell proteins into the culture medium occurred during high-cell-density cultivation of E. coli. Identification of these medium proteins revealed almost exclusively a periplasmic origin. Release of periplasmic proteins into the culture medium was observed throughout the entire cultivation of recombinant or non-recombinant cells. Leakage was intensified, however, in the final part of high-cell-density cultures (>100 g L(-)(1) dry cell mass) or when a temperature upshift was used for induction of recombinant protein synthesis. After temperature upshift, formation rates and residual cellular concentrations of periplasmic proteins declined with individual rates; e.g., the cellular content of the large periplasmic dipeptide binding protein DppA (57.4 kDa) started to decline about 4 h after the temperature upshift, whereas the smaller periplasmic d-galactose/d-glucose binding protein MglB (33.4 kDa) was already lost during the first hour after the upshift. In addition to periplasmic proteins, the osmotic-shock-sensitive heat-shock protein DnaK was found in significantly higher proportion in the cell-free medium of the temperature-challenged culture than other cytoplasmic proteins. Cell lysis was not observed even after prolonged cultivation. Thus, loss of a subset of cellular proteins of mainly periplasmic origin ordinarily occurs during cultivation and is intensified through stressful conditions in high-cell-density cultures. The selective release of cellular proteins of periplasmic origin offers the opportunity to simplify the downstream processing of recombinant proteins directed to the periplasm of E. coli.  相似文献

Dissociated cells separated from a natural colony of Nostoc flagelliforme were cultivated heterotrophically in the darkness on glucose under fed-batch culture conditions. The effects of carbon sources (glucose, fructose, xylose, and sucrose) and concentrations on cell growth and extracellular polysaccharide (EPS) production were investigated. At harvest, the culture contained 1.325 g L?1 of biomass and 117.2 mg L?1 of EPS, respectively. The gravimetric EPS production rate was 16.7 mg g?1 cell dry weight day?1, which was 2.1 times higher than previously reported. Using sigmoid model, batch fermentation of N. flagelliforme was kinetically simulated to obtain equations including substrate consumption, biomass growth, and EPS accumulation. Results from a simulation correlated well with the experimental ones, indicating that this method could be useful in studying EPS production from batch and fed-batch cultures.  相似文献

The newly screened Aeromonas hydrophila produces copolymer consisting of 3-hydroxybutyrate (3HB) and 3-hydroxyhexanoate (3HHx). The characteristics of cell growth and polymer accumulation were examined using various carbon sources. P(3HB-co-3HHx) was produced from lauric acid and oleic acid only. P(3HB-co-3HHx) content can be increased by limitation of phosphorus. A maximal P(3HB-co-3HHx) content of 28.8 wt% could be obtained in flask culture. By applying the optimally designed nutrient feeding strategy, cell dry weight, P(3HB-co-3HHx) content, and 3HHx fraction obtained over the course of 43 h were 95.7 g/L, 45.2 wt%, and 17 mol%, respectively, resulting in a productivity of 1.01 g polyhydroxyalkanoate (PHA)/L. h.  相似文献

Mangueira Lagoon, located in the extreme south of Brazil, has water with physicochemical characteristics such as alkaline pH and carbonate levels propitious for the growth of the cyanobacterium Spirulina platensis. Previously published studies have shown that Mangueira Lagoon water supplemented with small quantities of carbon and nitrogen is suitable for S. platensis cultivation and can significantly reduce production costs. We studied mixed cultures of Spirulina platensis and the toxic cyanobacterium Microcystis aeruginosa using a 2(3) factorial design in which the three factors were the initial biomass concentration of S. platensis and M. aeruginosa and the type of culture medium (100% Zarrouk's medium or 80% Mangueira Lagoon water plus 20% Zarrouk's medium). The highest S. platensis maximum specific growth rate (mu(max)) occurred in the culture with the highest M. aeruginosa biomass concentration and when undiluted culture medium was used (micro(max) = 0.283 d(-1)). The highest M. aeruginosa specific death rate (k) was obtained in the presence of S. platensis (k = 0.555 d(-1)) and was independent of the initial M. aeruginosa biomass concentration and culture medium, demonstrating that S. platensis cultures are not susceptible to contamination by M. aeruginosa. The culture medium had no significant influence (p > 0.05) on S. platensis micro(max) values, indicating that production costs could be reduced by using a medium consisting of 80% Mangueira Lagoon water plus 20% Zarrouk's medium.  相似文献

This study investigated the heterotrophic growth behavior of mung beans cultivated in an individual bed under water supply. The fresh weight of mung beans in the bed was estimated, and changes in temperature, and oxygen and carbon dioxide concentrations were recorded during the cultivation period. The specific growth rate, oxygen uptake rate, and carbon dioxide evolution rate, based on the fresh weight in the bed, were calculated. Growth under heterotrophic cultivation can be classified into the following three stages. Reductions in specific oxygen uptake rate, specific carbon dioxide evolution rate, and specific energy production rate corresponded to that of specific growth rate. Indicators of biological activity related to oxygen and carbon dioxide were evaluated quantitatively for beds under high-density heterotrophic cultivation. Moreover, the results obtained from this study successfully demonstrate that there is a relationship between the growth of mung beans and indicators of biological activity.  相似文献
