The problems encountered with a glass slide circulation are legion but timely circulation is a major problem and is an inherent deficiency of our Non-gynaecological EQA scheme. This applies not only to consultants but also to specialist registrars (SpRs) and technical staff that are not formally included in the circulation list. In 2005 only 7 technical staff and 4 out of 47 SpRs took part on a formal basis, their participation being dependant on access to slides during their cytology attachment. The results for the 2005 circulation have been analysed and despite the small numbers of participating technical staff and SpRs their answers concur with the consultant body. To address the issues of timeliness and circulation problems a pilot teaching set has been developed by SlidePath into a virtual microscope web based circulation and sent to all SpRs in our region. They have recorded their answers and been given immediate access to the consensus consultant opinion with illustrations of follow up histology. A questionnaire was completed to evaluate the scheme. The facility of immediate feedback of consultant consensus is particularly pertinent to the educational element of the scheme and use of virtual microscopy addresses the issue of timely circulation. If further funding was made available technical staff could also be given the opportunity to try this web-based circulation.  相似文献

The 8th circulation of this scheme was preceded by a CD ROM circulation of selected digital images and presented as individual JPEG file images in a single folder on the CD ROM. The number of images per case ranged from three to eight. The delegates will have an opportunity to assess these themselves prior to uncovering the consensus diagnosis provided by the participants. Of the 76, 29 participants of the scheme provided a completed answer sheet for the image‐based circulation. To date, 19 have now also supplied answers on the actual slide circulation. The same diagnosis on image and slide circulation varied from 5/19 to 19/19 with a mode of 14/19. The case that provided the poorest correlation was because of a generalized undercall of suspicious in a case which achieved almost complete consensus on the slide circulation. It had been the most difficult to photograph. The case that provided the next poorest correlation 11/19 was the case that had the poorest consensus diagnosis on the slide circulation. The two cases that provided the best correlation 18/19 and 19/19 were two cases that provided 100% consensus diagnosis on the slide‐based circulation. Comments were received from 20 participants about the image circulation ranging from ‘great’ to ‘awful’ with the majority of 12 participants not happy for their diagnostic capability to be assessed on such images alone. Two stated that the CD was easier to use on their home computer than their NHS one. In conclusion an image circulation overcomes many of the inherent problems with a slide circulation for EQA purposes and can provide an overall 70% correlation with a slide circulation but a significant number of pathologists do not find this an acceptable method for EQA.  相似文献

Geoprocessing of large gridded data according to overlap with irregular landscape features is common to many large‐scale ecological analyses. The geoknife R package was created to facilitate reproducible analyses of gridded datasets found on the U.S. Geological Survey Geo Data Portal web application or elsewhere, using a web‐enabled workflow that eliminates the need to download and store large datasets that are reliably hosted on the Internet. The package provides access to several data subset and summarization algorithms that are available on remote web processing servers. Outputs from geoknife include spatial and temporal data subsets, spatially‐averaged time series values filtered by user‐specified areas of interest, and categorical coverage fractions for various land‐use types.  相似文献

The question of how best to compare and classify the (three‐dimensional) structures of proteins is one of the most important unsolved problems in computational biology. To help tackle this problem, we have developed a novel shape‐density superposition algorithm called 3D‐Blast which represents and superposes the shapes of protein backbone folds using the spherical polar Fourier correlation technique originally developed by us for protein docking. The utility of this approach is compared with several well‐known protein structure alignment algorithms using receiver‐operator‐characteristic plots of queries against the “gold standard” CATH database. Despite being completely independent of protein sequences and using no information about the internal geometry of proteins, our results from searching the CATH database show that 3D‐Blast is highly competitive compared to current state‐of‐the‐art protein structure alignment algorithms. A novel and potentially very useful feature of our approach is that it allows an average or “consensus” fold to be calculated easily for a given group of protein structures. We find that using consensus shapes to represent entire fold families also gives very good database query performance. We propose that using the notion of consensus fold shapes could provide a powerful new way to index existing protein structure databases, and that it offers an objective way to cluster and classify all of the currently known folds in the protein universe. The foraging behaviour of species determines their diet and, therefore, also emergent food‐web structure. Optimal foraging theory (OFT) has previously been applied to understand the emergence of food‐web structure through a consumer‐centric consideration of diet choice. However, the resource‐centric viewpoint, where species adjust their behaviour to reduce the risk of predation, has not been considered. We develop a mechanistic model that merges metabolic theory with OFT to incorporate the effect of predation risk on diet choice to assemble food webs. This ‘predation‐risk‐compromise’ (PR) model better captures the nestedness and modularity of empirical food webs relative to the classical optimal foraging model. Specifically, compared with optimal foraging alone, risk‐mitigated foraging leads to more‐nested but less‐modular webs by broadening the diet of consumers at intermediate trophic levels. Thus, predation risk significantly affects food‐web structure by constraining species’ ability to forage optimally, and needs to be considered in future work.  相似文献

The rise of smartphones and web services made possible the large-scale collection of personal metadata. Information about individuals'' location, phone call logs, or web-searches, is collected and used intensively by organizations and big data researchers. Metadata has however yet to realize its full potential. Privacy and legal concerns, as well as the lack of technical solutions for personal metadata management is preventing metadata from being shared and reconciled under the control of the individual. This lack of access and control is furthermore fueling growing concerns, as it prevents individuals from understanding and managing the risks associated with the collection and use of their data. Our contribution is two-fold: (1) we describe openPDS, a personal metadata management framework that allows individuals to collect, store, and give fine-grained access to their metadata to third parties. It has been implemented in two field studies; (2) we introduce and analyze SafeAnswers, a new and practical way of protecting the privacy of metadata at an individual level. SafeAnswers turns a hard anonymization problem into a more tractable security one. It allows services to ask questions whose answers are calculated against the metadata instead of trying to anonymize individuals'' metadata. The dimensionality of the data shared with the services is reduced from high-dimensional metadata to low-dimensional answers that are less likely to be re-identifiable and to contain sensitive information. These answers can then be directly shared individually or in aggregate. openPDS and SafeAnswers provide a new way of dynamically protecting personal metadata, thereby supporting the creation of smart data-driven services and data science research.  相似文献

Although much effort has been put in the studies of weak in vivo microscale movements due to its importance, the real‐time, long‐time, and accurate monitoring is still a great challenge because of the complexity of the in vivo environment. Here, a new type of mechanically asymmetrical triboelectric nanogenerator with ultrashort working distance and high anti‐interference ability is developed to accurately and real‐timely monitor the microscopically weak movement of intestinal motility at low frequencies even around 0.3 Hz. The intestinal status after the glucose absorption, and physiological states in different times also have been monitored successfully in the complex in vivo environment with many kinds of interference and noises. This work gives a new self‐powered, long‐time and in vivo technical way for the real‐timely gastrointestinal motility monitoring, and contributes to the detection of every kind of gentle movements in various complex bio‐systems.  相似文献

Behavioral and hormonal data were compared for four giant pandas in two management conditions: (1) panda confined to exhibit area and (2) panda given choice to move freely between exhibit and off‐exhibit bedroom areas. Pandas displayed fewer signs of behavioral agitation and lower urinary cortisol in the free choice condition. Time active did not differ between the two conditions. These results suggest that simply offering pandas free access to alternative locations can improve behavioral and hormonal variables that may be related to well‐being. Circulation E of the North West non gynaecological cytology EQA scheme was split into two with one half receiving conventional glass slides and the other half receiving digital images supplied by the SlidePath company over the web. More of the participants eligible participated in the slide half (43/65) than the digital half (17/41).
Similar results were obtained for seven out of ten of the scoring cases and for both the educational cases. However for one case eight of the digital participants could not obtain the images over the web. In the remaining three scoring cases the digital participants were less good at obtaining the correct answer compared to the slide based participants. The use of this virtual microscopy has shortened the circulation length considerably. The digital participants complained about the speed the digital slides appeared on their computers and whilst most agreed that this is the way forward they are still uncomfortable with their performance being marked on their digital submissions. The increasing role played by liquid chromatography‐mass spectrometry (LC‐MS)‐based proteomics in biological discovery has led to a growing need for quality control (QC) on the LC‐MS systems. While numerous quality control tools have been developed to track the performance of LC‐MS systems based on a pre‐defined set of performance factors (e.g., mass error, retention time), the precise influence and contribution of the performance factors and their generalization property to different biological samples are not as well characterized. Here, a web‐based application (QCMAP) is developed for interactive diagnosis and prediction of the performance of LC‐MS systems across different biological sample types. Leveraging on a standardized HeLa cell sample run as QC within a multi‐user facility, predictive models are trained on a panel of commonly used performance factors to pinpoint the precise conditions to a (un)satisfactory performance in three LC‐MS systems. It is demonstrated that the learned model can be applied to predict LC‐MS system performance for brain samples generated from an independent study. By compiling these predictive models into our web‐application, QCMAP allows users to benchmark the performance of their LC‐MS systems using their own samples and identify key factors for instrument optimization. OBJECTIVE: To develop a local quality assurance program in cytopathology based on circulation of patient specimens on glass slides, with limited resources. STUDY DESIGN: A working group was set up for design and running of the program. Participation is on a laboratory basis. The scope and frequency of testing are defined. Well-documented cases (including gynecologic, nongynecologic and fine needle aspiration cytology) with commonly encountered diagnoses are collected. Consensus concerning the diagnosis, interpretive menu and scoring system is sought before the actual slide circulations using express mail. After returning their answers to the program organizer, the participating laboratories receive immediate feedback on their scores, with reference answers, explanatory notes, "whole-mount" images of glass slides and cumulative responses of peer laboratories for on-site checking. At the end of each year, an electronic file containing representative photomicrographs of all cases examined is provided to individual laboratories for their permanent records and training purposes. RESULTS: The program was launched in mid-2003. There were 24 and 27 participating laboratories from Hong Kong (and Macau) in 2003 and 2004, respectively. To date, >150 well-documented cytology cases are available in the slide pool and ready for circulation. As the revenue is mainly to cover the expenses of express mail, the program can be carried out at a relatively low cost. CONCLUSION: In order to have any cytology quality assurance program accepted by local laboratories, it has to be fair and practical. Strict confidentiality needs to be observed throughout the process. This program emphasizes both performance assessment and educational value. Adequate representation from experienced local cytology workers, detailed documentation support from authorities and assistance from dedicated staff are essential to the success of any external proficiency testing scheme. Regular review and evaluation are also necessary for continuous improvement. The Hong Kong experience can serve as an example of running a glass slide-based cytology quality assurance program in a small region with limited resources.  相似文献

A common paradigm in studies of human cooperation is that individuals mediate their investment in a public good according to the investments of others. However, most research on this topic has been conducted in artificial laboratory settings and has assumed that cooperation imposes relatively high monetary costs on players. As such, we do not yet know how humans behave when faced with non‐monetary costs of cooperating in more realistic settings. Here, we present results from a real‐world public good experiment where the monetary costs of cooperation were relaxed. We manipulated the cleanliness of a sink in a staff tea room (a public good) by adding or removing dirty kitchenware and then recorded the number of items subsequently added to the sink by staff members. We found that when the sink had no dirty items, people were less likely to dump their own used kitchenware and the public good was maintained. In contrast, people were more likely to dump items in the sink when it already contained unwashed kitchenware and increasing the number of dirty items in the sink led to an increase in the number of items that were subsequently dumped. This study shows that defection begets defection in real‐life situations, even when cooperation is relatively cheap.  相似文献

Synthesis Metacommunity theory aims to elucidate the relative influence of local and regional‐scale processes in generating diversity patterns across the landscape. Metacommunity research has focused largely on assemblages of competing organisms within a single trophic level. Here, we test the ability of metacommunity models to predict the network structure of the aquatic food web found in the leaves of the northern pitcher plant Sarracenia purpurea. The species‐sorting and patch‐dynamics models most accurately reproduced nine food web properties, suggesting that local‐scale interactions play an important role in structuring Sarracenia food webs. Our approach can be applied to any well‐resolved food web for which data are available from multiple locations. The metacommunity framework explores the relative influence of local and regional‐scale processes in generating diversity patterns across the landscape. Metacommunity models and empirical studies have focused mostly on assemblages of competing organisms within a single trophic level. Studies of multi‐trophic metacommunities are predominantly restricted to simplified trophic motifs and rarely consider entire food webs. We tested the ability of the patch‐dynamics, species‐sorting, mass‐effects, and neutral metacommunity models, as well as three hybrid models, to reproduce empirical patterns of food web structure and composition in the complex aquatic food web found in the northern pitcher plant Sarracenia purpurea. We used empirical data to determine regional species pools and estimate dispersal probabilities, simulated local food‐web dynamics, dispersed species from regional pools into local food webs at rates based on the assumptions of each metacommunity model, and tested their relative fits to empirical data on food‐web structure. The species‐sorting and patch‐dynamics models most accurately reproduced nine food web properties, suggesting that local‐scale interactions were important in structuring Sarracenia food webs. However, differences in dispersal abilities were also important in models that accurately reproduced empirical food web properties. Although the models were tested using pitcher‐plant food webs, the approach we have developed can be applied to any well‐resolved food web for which data are available from multiple locations.  相似文献

Li metal batteries are considered a promising candidate for next‐generation rechargeable batteries. However, the practical application of Li metal batteries has been hindered by many challenges, especially the cycling stability of Li anodes due to their uncontrollable dendrite growth, volume fluctuation, and side reactions. These problems are more severe under high‐rate charge/discharge process. Therefore, the realization of stable cycling of Li anodes under high current density is crucial for the practical application of Li metal batteries. In this Progress Report, the authors focus on the stability of metallic Li through interphase design or microstructure construction. The advantages and drawbacks of the first‐generation 3D scaffolds are summarized, and a review of recent research progress in this area is generated. As high‐rate cycling of metallic Li is a complex dynamic problem, a scaffold with high mixed ionic and electronic conductivity may be a promising approach. The different design strategies of mixed ion and electron‐conductive scaffolds working with liquid and solid electrolytes are discussed, along with their technical challenges. Further directions of mixed ion and electron‐conductive scaffolds are also proposed.  相似文献

Food webs can respond in surprising and complex ways to temporary alterations in their species composition. When such a perturbation is reversed, food webs have been shown to either return to the pre‐perturbation community state or remain in the food web configuration that established during the perturbation. Here we report findings from a replicated whole‐lake experiment investigating food web responses to a perturbation and its consecutive reversal. We could identify three distinct community states in the food web that corresponded to the periods before, during and after the perturbation. Most importantly, we demonstrate the establishment of a distinct post‐perturbation food web configuration that differed from both the pre‐ and during‐perturbation communities in phytoplankton biomass and micro‐ and mesozooplankton species composition. We suggest that the pre‐ and post‐perturbation food web configurations may represent two alternative stable community states. We provide explanations for how each of the contrasting communities may be maintained through altered species interactions. These findings add to the discussion of how natural food webs react to environmental change and imply that the range of potential ecosystem dynamics in response to perturbations can be wider and more complex than is often recognized.  相似文献
