Cattle ticks are an important constraint on the livestock industry, particularly in tropical and subtropical areas, mainly because of the diseases they transmit and the costs of control. Conventional control is by means of acaricides; although there are still serious drawbacks, these can be minimized by a strategic approach. In this review Julio de Castro and Robin Newson look at alternatives. One is to make use of the host's innate abilities. In the simplest situation, where no control measures are applied, the animals develop their own protective immunity against ticks and tick-borne diseases (TBDs). Alternatively, breeds of cattle with naturally high resistance to ticks can be used, enhanced by selection. Methods of immunizing cattle against ticks are also under development. Ideally, ticks should be managed at an economically acceptable level by a combination of techniques, supported where necessary by vaccination against TBDs.  相似文献

Livestock industry is an essential part of Pakistan’s economy, and a variety of ruminants (including sheep and goats) are reared for the increasing demand of milk, meat and hide products. Haemoparasitic illnesses such as theileriosis, anaplasmosis, and babesiosis are a significant health risk for small ruminants in our country. Information regarding distribution patterns, the tick species involved and effective strategies to control tick-borne diseases (TBD) in goats and sheep of Pakistan is limited. To this end, it is required to assess the present rank of TBDs in small ruminants of Pakistan with a note on their vector ticks in order to control and identify the gaps in the knowledge of TBDs. This will recommend areas for future research and will add to the understanding of these diseases and will draw attention to the need for better-quality tools for the diagnosis and control of TBDs in small ruminants of Pakistan.  相似文献

Infectious diseases are common diseases all over the world. A recent World Health Organization report indicated that infectious diseases are now the world's biggest killer of children and young adults. Infectious diseases in non-industrialized countries caused 45% in all and 63% of death in early childhood. In developed countries, the emergence of new, rare or already-forgotten infectious diseases, such as HIV/AIDS, Lyme disease and tuberculosis, has stimulated public interest and inspired commitments to surveillance and control. Recently, it is reported that infectious diseases are responsible for more than 17 million deaths worldwide each year, most of which are associated with bacterial infections. Hence, the control of infectious diseases control is still an important task in the world. The ability to control such bacterial infections is largely dependent on the ability to detect these aetiological agents in the clinical microbiology laboratory. Diagnostic medical bacteriology consists of two main components namely identification and typing. Molecular biology has the potential to revolutionise the way in which diagnostic tests are delivered in order to optimise care of the infected patient, whether they occur in hospital or in the community. Since the discovery of PCR in the late 1980s, there has been an enormous amount of research performed which has enabled the introduction of molecular tests to several areas of routine clinical microbiology. Molecular biology techniques continue to evolve rapidly, so it has been problematic for many laboratories to decide upon which test to introduce before that technology becomes outdated. However the vast majority of diagnostic clinical bacteriology laboratories do not currently employ any form of molecular diagnostics but the use such technology is becoming more widespread in both specialized regional laboratories as well as in national reference laboratories. Presently molecular biology offers a wide repertoire of techniques and permutations of these analytical tools, hence this article wishes to explore the application of these in the diagnostic laboratory setting.  相似文献

In Sicily, as in other Mediterranean areas, livestock represents one of the most important resources for the island economy. This sector involves more than 16,000 farms of cattle and 10,000 farms of sheep and goats (respectively 6% and 15% of national production) which are actually increasing their number. Most livestock in Sicily is owned by small holders and pastoralists. Regional production of milk feeds some industries, which involve a large and increasing occupational area. Due to its peculiar geographic aspect Sicily is constituted by hill areas with sporadic grasses, therefore extensive grazing methods represent an ancient, traditional practice for using poor lands. For the control of infectious diseases Veterinary Services (VS) are based on the public regional network that is coordinated by the Ministry of Health in Rome. Even if Sicilian VS had to solve many constraints related to traditional "pastoral management system", to the lack of any sanitary background of this sector, to unknown data on the numbers of the whole livestock, the effort done in the last years has allowed to monitor for brucellosis, bovine leukemia virus (BLV), bovine TBC, swine vescicular disease (MVS) and other diseases for the majority of the farms. Tick-borne diseases (TBDs) still represent a serious impediment to the improvement of meat and milk production in the region. Every year, several outbreaks related to Theileria, Babesia or Anaplasma infections in cattle are recorded. Imported breeds pay the most expensive tribute often with a mortality rate of 100%. In the last five years more than 170 outbreaks of TBDs have been notified by our Institute although the pathogen prevalence and economical impact in the Sicilian livestock is still unknown. The outdoor grazing of the animals, far away from observation, a subclinical course of the disease, can in fact create difficulties in discovering infection and therefore the therapy is often too late. New diagnostic and control methods (PCR, vaccination) are being developed at the Istituto Zooprofilattico Sperimentale of Sicily in cooperation with other national and international laboratories (such as the University of Utrecht).  相似文献

Tick borne diseases (TBDs), though neglected, are possibly the major constraints for livestock and agricultural production in southern Italy. The diseases affect mainly the livestock imported from foreign Northern countries. But now, very timidly indeed, the awareness that the use of native breeds could contribute to reduce the ravages of these diseases is growing. Over a period of many centuries Apulian farm animals have naturally developed the ability to thrive in TBDs endemic areas. This trait, which can be defined as tolerance to TBDs is associated with the ability to control parasitaemia and to resist the development of anaemia in the face of infection. It is a matter of fact that the hematological features of Apulian native farm animals are rather peculiar as the authors highlighted by studying the blood of these animals over a period of more than ten years. In the light of their experience the authors considered that the phenomenon of tolerance is a broad-based one and possibly not unrelated to the erythropoietic system of the Apulian animals. Thus the ability to resist anaemia per se was tested in four Altamurana sheep compared to that of four Romanov sheep. This work summarises the results obtained from the experimental anaemization of the above two breed groups and discusses the findings on the basis of a review of the authors' experience in studying TBDs in sheep.  相似文献

Tick-borne protozoan diseases, babesiosis and theileriosis, are among the most important diseases affecting the productivity of livestock worldwide and resulting in high economic losses. A prerequisite for the control of these diseases is to study their epidemiology by mapping their distribution and seasonality. As clinical diagnostic and surveillance tools, serological tests such as the complement fixation test (CFT), the indirect fluorescent antibody test (IFAT) and the enzyme linked immunosorbent assay (ELISA) have been successfully used over decades. With the development in molecular biology, recombinantly expressed parasite molecules have emerged and substituted crude parasite antigen used in serology. A popular format of these tests is the antibody binding competitive inhibition and the indirect antibody detection ELISA. Under the precondition that these tests are correctly designed and validated, they provide a powerful tool for epidemiology, with greater advantages of affordability and amenability to standardization. This paper reviews the pathogenic tick-borne protozoan diseases and the respective diagnostic ELISA based serological tests currently available for serosurveillance.  相似文献

Tick-borne diseases (TBDs) are a major economic constraint to livestock production in sub-Saharan Africa. ILRI is focussing on developing a range of products, such as vaccines, diagnostics and decision support services to underpin improved control programmes against these diseases. We have developed three highly sensitive and specific enzyme linked immuno-assays (ELISAs), which allow precise diagnosis of Theileria parva, Babesia bigemina and Anaplasma marginale. These tests have been standardised and validated using defined experimental and field infection sera. Parasite specific recombinant antigens and monoclonal antibodies against bovine immunoglobulins as secondary antibodies have played an important role in in enhancing the sensitivity and specificity of the assays. They have been further evaluated in on-farm longitudinal sero-epidemiological studies to define infection dynamics and disease risks in various farming systems in Kenya and Uganda. In addition, DNA-based tests for differentiation of Theileria species and characterisation of Theileria parva stocks have been developed. These tests have been derived through physical mapping and sequencing of key elements of the T. parva genome, which include repetitive and telomeric regions, minisatellite sequences, antigen genes and a number of random DNA sequences. These tools are currently being deployed in conjunction with field immunisation programmes to determine the biological impact of introducing live vaccines of T. parva on population dynamics.  相似文献

Recent advances in the knowledge of the biology of the protozoan family Babesiidae is reviewed with special emphasis on those species which cause economically important diseases in cattle. The life cycle of Babesia of cattle is illustrated taking into account these recent advances. Areas of controversy in the life cycles still exist particularly over the presence or absence of a sexual cycle within the tick vector and of exoerythrocytic schizonts in the mammalian host. In the mammalian host, the major parasite division cycle occurs in erythrocytes and it is these stages which induce the pathogenic effects. The recent progress in the understanding of the biology of Babesia parasites may allow important developments in the control of babesiosis.  相似文献

For their biological and ecological characteristics, ticks are vectors of the widest variety of pathogens causing tick-borne diseases (TBDs). Little information is available about the ways in which time spent by ticks to feed on hosts, transmission times and TBD prevention are related and it is exclusively limited to laboratory reports on adult stages. In particular, the time required by immature stages to transmit pathogens has not been determined for most TBDs. Considering their importance for animal and human health, effective control of immature ticks is advisable to reduce the damage ticks cause. Recently, the efficacy of a combination of imidacloprid 10%/permethrin 50% was compared with that of fipronil 10%/S-methoprene 9% against ticks when administered to naturally infected dogs. A semi-quantitative method was used to assess the methodological parameters to calculate drug efficacy on immature stages. On adult ticks, the efficacy of both products was high and overall very similar, whereas for the immature stages the combination of imidacloprid 10% and permethrin 50% had a higher efficacy than fipronil 10% and methoprene 9% throughout the observation period (statistically significant difference on day + 28 only). The semi-quantitative method proposed for the evaluation of immature stages can be considered a useful tool for calculating the efficacy of a drug in the field. Due the important role of immatures in the spread of TBDs, the immature tick load should be calculated to assess the efficacy of acaricidal products both under laboratory and field conditions.  相似文献

Molecular biological tools currently available to us are revolutionizing the way in which we can address questions in evolutionary biology. The purpose of this article is to provide an overview of molecular techniques and applications available to biologists who are interested in evolutionary studies but who have little acquaintance with molecular biology. In evolutionary biology, techniques designed to determine degree of nucleic acid similarity are in common use and will be dealt with first. Another approach, namely gene expression studies, has strong implications for evolutionary biology but generally requires substantial familiarity with molecular biological tools. Expression studies provide powerful tools for discerning processes of speciation, as in the selection of genetic variants, as well as discerning lineages, e.g., expression of specific homeobox genes during segment formation. For investigations where either nucleic acid identity or gene expression are the ultimate goal, detailed information, protocols and appropriate controls are beyond the scope of this work but, where possible, recent review articles are cited.  相似文献

Tick-borne diseases (TBDs) remain to be a global animal health threat. Developing countries like the Philippines is not exempt to this. Despite the potential impact TBDs can give to these countries, local government initiatives and researches remain to be limited. In the Philippines, most epidemiological studies were confined only to specific areas, and predominantly in the Northern Area. Due to its unique geography and limited studies, the current nationwide status of most TBDs could not be clearly established. This review mainly covered published studies and presented challenges in the conduct of TBD research in the Philippines, which may be similar to other Southeast Asian or developing countries. To date, reported livestock TBD pathogens in the Philippines include Anaplasma, Babesia, Theileria, and Mycoplasma spp. With the ubiquitous presence of the Rhipicephalus microplus ticks in the country, it is highly probable that other pathogens transmitted by these vectors could be present. Despite studies on different TBDs in the livestock sector, the Philippine government has not yet heightened its efforts to implement tick control measures as part of the routine animal health program for local farmers. Further studies might be needed to determine the nationwide prevalence of TBDs and the presence of other possible tick species and TBD pathogens. The Philippine scenario may present situations that are similar to other developing countries.  相似文献

Trypanosomatid protozoans cause important diseases of humans and their domestic livestock. Various molecular genetic tools are now allowing rapid progress in understanding many of the unique aspects of the molecular and cell biology of these organisms. Diploidy and the lack or difficulty of sexual crossing has been a challenge for forward genetics, but powerful selections and functional complementation have helped to overcome it in Leishmania. RNA interference has been adapted for forward genetics in trypanosomes, in which it is also a powerful tool for reverse genetics. Interestingly, the efficacy of different genetic tools has steered research into different aspects of the biology of these parasites.  相似文献

Plants are important sources of food and plant products are essential for modern human life. Plants are increasingly gaining importance as drug and fuel resources, bioremediation tools and as tools for recombinant technology. Considering these applications, database infrastructure for plant model systems deserves much more attention. Study of plant biological pathways, the interconnection between these pathways and plant systems biology on the whole has in general lagged behind human systems biology. In this article we review plant pathway databases and the resources that are currently available. We lay out trends and challenges in the ongoing efforts to integrate plant pathway databases and the applications of database integration. We also discuss how progress in non-plant communities can serve as an example for the improvement of the plant pathway database landscape and thereby allow quantitative modeling of plant biosystems. We propose Good Database Practice as a possible model for collaboration and to ease future integration efforts.  相似文献

Background

Ontologies represent powerful tools in information technology because they enhance interoperability and facilitate, among other things, the construction of optimized search engines. To address the need to expand the toolbox available for the control and prevention of vector-borne diseases we embarked on the construction of specific ontologies. We present here IDODEN, an ontology that describes dengue fever, one of the globally most important diseases that are transmitted by mosquitoes.

Methodology/Principal Findings

We constructed IDODEN using open source software, and modeled it on IDOMAL, the malaria ontology developed previously. IDODEN covers all aspects of dengue fever, such as disease biology, epidemiology and clinical features. Moreover, it covers all facets of dengue entomology. IDODEN, which is freely available, can now be used for the annotation of dengue-related data and, in addition to its use for modeling, it can be utilized for the construction of other dedicated IT tools such as decision support systems.

Conclusions/Significance

The availability of the dengue ontology will enable databases hosting dengue-associated data and decision-support systems for that disease to perform most efficiently and to link their own data to those stored in other independent repositories, in an architecture- and software-independent manner.  相似文献

Diseases transmitted by mosquitoes impose enormous burden towards human morbidity and mortality. Over the last three decades, Brazil has suffered from severe Dengue epidemics. In September 2014, this situation is further complicated by the introduction of two other viruses, Zika and Chikungunya, placing Brazil in a triple epidemic. In this article, we discuss the biology of Aedes aegypti Linnaeus, and the principal initiatives currently used to control mosquito populations and the diseases they transmit. Aedes aegypti has broad global distribution and is involved in the transmission of various arboviral diseases such as Dengue, Zika, and Chikungunya. Several factors contribute to the success of the species, particularly behavioral plasticity, rapid development, desiccation-resistant eggs, resistance to the principle insecticide classes currently available on the market, preference for the urban environment, and proximity to humans. Vector control programs are the best way to reduce the burden of mosquito-borne diseases. Chemical control is most commonly used in recent times, and unfortunately, the results have not been satisfactory but instead, there is increased vector dispersal and, subsequently, the spread of disease epidemics. Investigations of alternative control methods such as release of Wolbachia-infected mosquitoes for blocking vector-borne pathogens, release of transgenic mosquitoes carrying a lethal gene for offspring, and the use of insecticide-dispersing mosquitoes are under way in Brazil, and some have shown promising results. Special emphasis should be placed on integrated management of all available tactics, so as to maximize efforts towards mosquito control. Finally, we emphasize that continuous actions and community participation control initiatives are critically important for success.  相似文献

Infectious hematopoietic necrosis (IHN) is one of the most important viral diseases of salmon and trout reared in culture. The disease remains untreatable with avoidance being the only control measure. Much has been learned about the chemical, physical, and serological characteristics of the rhabdovirus causing IHN, but critical gaps exist in our understanding of the biology of the virus in nature. The tools of molecular biology have provided improved methods for detection of pathogens and new strategies for control of viral diseases. This paper reviews several recent improvements in methods for detecting infectious hematopoietic necrosis virus including the application of enzyme-linked immunosorbent assays, development of monoclonal antibodies and DNA probes, and use of the polymerase chain reaction. New strategies for control of IHN through the use of better water treatment, more resistant fish, antiviral drugs or chemicals, and new generation vaccines are discussed.  相似文献
