Tradescantia pallida cv. purpurea, a popular garden plant in Brazil, was used for the Tradescantia micronucleus (Trad-MCN) assay. In situ monitoring of the genotoxicity of air pollutants was carried out by sentinel approach, using the plant grown in the field or using the plants in pots which were carried to the monitoring sites. Two highly polluted sites, in S?o Paulo city (Cerqueira Cesar and Congonhas) and two rural sites (the cities of Pirassununga, 200 km and Caucaia do Alto, 50 km from S?o Paulo, respectively) were chosen for this study, in order to determine the gradient difference of the air pollution levels. Sentinel plants in Congonhas site presented the highest frequency of micronuclei (4.4%), in comparison with 2.2 and 2.3% found in plants from Pirassununga and Cerqueira Cesar sites, respectively (Kruskal-Wallis; P<0.020). Significant increases (F test; P<0.0001) in the frequency of micronuclei were observed in plants exposed in the polluted urban sites (Cerqueira Cesar: 5.7%; Congonhas: 7.1% and Caucaia do Alto: 2.3%). The increase in the frequency of micronuclei observed indicates the potential risk of mutagenicity in presence of high concentrations of pollutants.  相似文献

Despite a long tradition of research in applied genetics, particularly in agricultural research, in Italy the transition to the new knowledges and techniques of molecular biology was long and difficult. Political and financial constraints made academic institutions very slow to grasp the importance of molecular approaches to biology and medicine. In fact, the main studies concerning problems of molecular biology took place inside non-academic institutions. We reconstruct the complex paths leading to the birth of the International Laboratory of Genetics and Biophysics (LIGB) in Naples, and describe its work and activities in the period in which it was directed by its creator, Adriano Buzzati-Traverso, between 1962 and 1969. The origins of the LIGB are inextricably bound to the growth of biophysical research at the University of Pavia and at the Higher Health Institute in Rome. For a short period, with the aid provided by Italian and European physicists to biological research on the effects of nuclear radiation, LIGB became a focal point in European molecular biology. In 1968 the scientific and educational activities of the LIGB dramatically fell victim to the contradictions of the Italian academic and scientific system, and to the political climate which emerged in Italy at the time.  相似文献

Monitoring of fish and crustaceans in the Wadden Sea (WS) must cope with rapid changes in distribution patterns, access to certain areas and gear efficiency. Application and limitations of a variety of fishing devices (fyke nets, gill nets, enclosures, stow nets, purse seines, beam trawls, push nets, beach seines, bottom trawls, pelagic trawls) are discussed with regard to different objectives of monitoring. Furthermore, the validity of data from three current monitoring programmes is also discussed. The emergence and spread of carbapenemase-producing Enterobacteriaceae (CPE) represent a major public health concern because these bacteria are usually extensively resistant to most antibiotics. In order to evaluate their dissemination in Quebec, a surveillance program was introduced in 2010. We report the molecular and epidemiological profiles of CPE isolates collected. Between August 2010 and December 2012, a total of 742 non-duplicate isolates non-susceptible to carbapenems were analysed. AmpC β-lactamase and metallo-β-lactamase production were detected by Etest and carbapenemase production by the modified Hodge test (MHT). Antibiotic susceptibility profiles were determined using broth microdilution or Etest. Clonality of Klebsiella pneumoniae carbapenemase (KPC) strains was analyzed by pulsed-field gel electrophoresis (PFGE). The presence of genes encoding carbapenemases as well as other β-lactamases was detected using PCR. Of the 742 isolates tested, 169 (22.8%) were CPE. Of these 169 isolates, 151 (89.3%) harboured a bla KPC gene while the remaining isolates carried bla SME (n = 9), bla OXA-48 (n = 5), bla NDM (n = 3), and bla NMC (n = 1) genes. Among the 93 KPC strains presenting with a unique pattern (unique PFGE pattern and/or unique antibiotics susceptibility profile), 99% were resistant to ertapenem, 95% to imipenem, 87% to meropenem, 97% to aztreonam, 31% to colistin and 2% to tigecycline. In 19 patients, 2 to 5 KPC strains from different species or with a different PFGE pattern were isolated. CPE strains were present in the province of Quebec with the majority of strains harbouring KPC. Alternately, SME, OXA-48 and NMC containing strains were rarely found.  相似文献

Since 1979 the flora and fauna of the artificial rocky shores of the Oosterschelde estuary have been investigated by diving biologists.Spatial and temporal patterns of species diversity and biomass were studied and several groups of species (communities) were distinguished. A west-east gradient in species composition was found. Species distribution proved to be mainly determined by current velocities, silt and plankton concentrations in the water, underwater irradiance, and the nature of the substratum. Some indications of migration between the basin and the North Sea are shown. Biomass of the hard substratum fauna was compared with that of the soft bottom fauna; the ratio appeared to be about 1 : 2, which is surprisingly high considering the small area available for hard substratum fauna.The key to the interactions with the North Sea is found in the water movements and the volume of water passing through the mouth of the basin during each tide. The abiotic and subsequent biotic changes in the basin caused by the building of the storm surge barrier are discussed, and used to illustrate the measure of dependence of the Oosterschelde on the North Sea.  相似文献

Due to the similarity between Pd and Pt, the complexes of palladium(II) can be considered as potential anticancer agents. Activity of six PdCl2(X2Py)2 complexes (Py = pyridine, and X = CH3 or Cl) was measured by MTT test using MCF7, CCRF-SB, PC3 and human B-lymphoblastoid cell lines. We found that the effect of PdCl2(XnPy)2 was cell-specific and time-dependent. Obtained results were discussed and compared with the activation parameters calculated for the hydrolysis of PdCl2(XnPy)2, indicating a correlation between viability of MCF7 and CCRF-SB cells and rates of hydrolysis of the Pd(II) complexes.  相似文献

The debate on the relative benefits of conventional and organic farming systems has in recent time gained significant interest. So far, global agricultural development has focused on increased productivity rather than on a holistic natural resource management for food security. Thus, developing more sustainable farming practices on a large scale is of utmost importance. However, information concerning the performance of farming systems under organic and conventional management in tropical and subtropical regions is scarce. This study presents agronomic and economic data from the conversion phase (2007–2010) of a farming systems comparison trial on a Vertisol soil in Madhya Pradesh, central India. A cotton-soybean-wheat crop rotation under biodynamic, organic and conventional (with and without Bt cotton) management was investigated. We observed a significant yield gap between organic and conventional farming systems in the 1st crop cycle (cycle 1: 2007–2008) for cotton (−29%) and wheat (−27%), whereas in the 2nd crop cycle (cycle 2: 2009–2010) cotton and wheat yields were similar in all farming systems due to lower yields in the conventional systems. In contrast, organic soybean (a nitrogen fixing leguminous plant) yields were marginally lower than conventional yields (−1% in cycle 1, −11% in cycle 2). Averaged across all crops, conventional farming systems achieved significantly higher gross margins in cycle 1 (+29%), whereas in cycle 2 gross margins in organic farming systems were significantly higher (+25%) due to lower variable production costs but similar yields. Soybean gross margin was significantly higher in the organic system (+11%) across the four harvest years compared to the conventional systems. Our results suggest that organic soybean production is a viable option for smallholder farmers under the prevailing semi-arid conditions in India. Future research needs to elucidate the long-term productivity and profitability, particularly of cotton and wheat, and the ecological impact of the different farming systems.  相似文献

13C-NMR analysis of the glutathione conjugates formed from (±)-benzo(a)-pyrene 4,5-oxide-4,5-13C by a purified glytathione transferase from little skate (Raja erinacea) liver demonstrated that equivalent amounts of the positional isomers (4,5-dihydro-4-hydroxy-5-glutathionylbenzo(a)pyrene and 4,5-dihydro-4-glutathionyl-5-hydroxybenzo(a)pyrene) were formed. Separation of these conjugates by HPLC and subsequent 13C-NME studies showed that only one diastereoisomer of each positional isomer was formed by the skate enzyme, each enantiomer of the arene oxide having produced only one of the two possible positional isomers. The non-enzymic reaction of (±)-benzo(a)pyrene 4,5-oxide with glutathione produced the four possible stereoisomers resulting from trans addition to the epoxide ring. This was also true when rat liver cytosol was used as the source of transferase activity. The data demonstrate that skate liver glutathione transferase 4 has high substrate regiospecificity and stereospecificity for (±)-benzo(a)pyrene 4,5-oxide.  相似文献

Monthly collections were made using two traps 5 m apart exposed for 48h, containing sardines and installed at points: A at the edge (500 m from the entrance of the Reserve); B 1200 m from the entrance and 1000 m inside the forest; and C 1700 m from the entrance and 500 m inside the forest. The purpose was to evaluate the abundance and richness of calliphorid species as a function of the environmental conditions using Pearson's correlation, compare the richness of the areas using ANOVA and Tukey's test, compare the abundances of the areas by the Kruskal-Wallis test, and also assess the possible influence of the anthropic presence. Rare, intermediary and common species were identified. The collection totalized 8515 Calliphoridae belonging to 26 species, with a predominance of females. None of the 13 species considered common presented a correlation between abundance and temperature: only Cochliomyia hominivorax (Coquerel) and Chrysomya megacephala (Fabricius) were correlated with humidity and only Mesembrinella semihyalina Mello with precipitation. This parameter was the only climatic variable correlated with richness. The greatest abundance and richness of calliphorids occurred in September 2006. From the 13 common species, seven were considered synanthropic, indicating the effect of anthropic action in this site.  相似文献

Low molecular weight zinc(II) and copper(II) binding ligands were detected in ultrafiltered human, bovine, and goat milk by the application of the method of modified gel chromatography. Human milk contains at least three detectable low molecular weight copper binders, whereas bovine and goat milk contain at least two. All three milks show two copper binding peaks with the same elution volumes. Zinc chromatograms were less specific than copper. Zinc showed only a single detectable low molecular weight binding ligand common to all three milks. Elution volumes for both zinc(II) and copper(II) citrate and picolinate systems were measured. Elution volumes of both copper(II) and zinc(II) citrate complexes are identical to elution volumes of an intense peak observed with all three milks; it is reasonable to assume that at least part of this peak corresponds to citrate. Human milk alone has a relatively intense binding peak for copper(II) at the same elution volume as the glutamate complex. Human and goat milk have another low intensity copper(II) binding ligand peak at the same elution volume; a number of amino acid complexes have binding peaks at this position. No peak characteristic of the zinc(II) or copper(II) picolinate systems could be found with any of the milks.  相似文献

Summary 2-Macrpglobulin (A2M) is a major human plasma protease inhibitor capable of inhibiting most endopeptidases tested so far. In the case of the other major plasma protease inhibitor, 1-antitrypsin, genetically determined deficiency states are known to increase the risk of chronic obstructive pulmonary disease (COPD) 20- to 30-fold in affected individuals. No defects of the A2M gene have been described as yet, but A2M may play a role in the regulation of protease activity in the lung, especially with respect to those proteases not inhibited by 1-antitrypsin. We report here the molecular genetic detection of an alteration of the A2M gene in a patient with serum A2M deficiency and chronic lung disease since childhood. The alteration involves restriction sites detected with 10 different enzymes and is most probably caused by a major deletion or rearrangement of the gene. Nine of the restriction enzymes used detected no polymorphisms in 40 healthy control subjects and 39 COPD patients. The polymorphism detected in this patient with the enzyme PvuII was different from another described previously, and was found in this patient only. The patient is heterozygous for an alteration in the A2M gene; this may be responsible for his serum A2M deficiency and may be relevant to the early onset of pulmonary disease in his case.  相似文献

Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

Radiation sterilization eliminates microbiological infections but causes the degradation of the cell factor. The negative result of microbiological examination for tissue transplants is one of the conditions for approval for distribution in patients. The study attempts to verify impact of the presence of microbes onto material for transplant loss. In the 2011–2015 period, we analyzed 293 donors of skin and amnion. Microbiological sampling was performed. The total of 21 strains of bacteria, molds and fungi was identified in collected tissue. The widest spectrum of strains was found in skin (17), followed by amnia (8). The total number of positive findings was 147 and was again highest in skin (129), while the number of positive findings in amnia was 18 only. The general percentage of fungal infections was very low. The presence of fungal strains was only observed in allogeneic skin (2%). Large number of microorganisms isolated from the skin before sterilization was observed, so it seems impossible to use allogeneic intravital skin. However, the intravital application of allogeneic amnion obtained from cesarean section remains to be considered.  相似文献

The aim of this study was to evaluate the Temperament and Character Inventory (TCI) scores of a sample of Korean patients with remitted depression who had attempted suicide and reported suicidal ideation and to compare their scores with those of remitted depressed patients without suicidal ideation. Adult depression patients who had completed 12 weeks of follow-up (N = 138) were divided into three groups: patients with a history of suicide attempts (N = 23); patients with current suicidal ideation (N = 59); and patients without current suicidal ideation (N = 56). After controlling for covariates, no significant differences were found among the three groups on any measure of temperament or character except self-directedness and self-transcendence. The self-transcendence scores of the lifetime suicide-attempt group were significantly higher compared with those of the suicidal-ideation group; post hoc analysis revealed that self-directedness was significantly lower in the suicide-attempt group compared with the non-suicidal group. The results from the present study suggest that remitted depression patients with a history of suicide attempts do not differ from non-attempters in temperament, but do differ in certain character traits.  相似文献

Rabbit antisera and monoclonal antibodies were raised against factor C, a regulatory protein of Streptomyces griseus. ELISA and immunoblotting techniques suitable to determine and characterize factor C antigen in bacterial specimens were developed. Factor C antigen was detected in all the 23 Streptomyces strains and variants examined thus far and in one Bacillus subtilis too. Depending on the strain analysed it has a molecular mass of 34 000 or 70 000 in mycelial homogenates. Most of factor C was found excreted into the cultivation medium. The quantity of factor C antigen in different Streptomyces strains showed great variation. Amy + strains were usually good producers of factor C while Amy were not. To study the characteristics of the structure and the functional role of chloroplast membranes of halophytes with different salt tolerance strategy, raft structures were isolated. Lipid analysis data provided evidence for the presence of rafts in the chloroplast membranes. The discovered significant differences in the content of raft-forming lipids in different halophytes suggest a relationship between the functions of rafts and the mechanisms of salt tolerance in plants.  相似文献

Tardigrades have two forms of dormancy, namely cryptobiosis and encystment. The encystment is a form of diapause known for a limited number of species of tardigrades and still little studied. To increase the knowledge on encystment, two species of eutardigrades from Italy have been considered: the moss-dwelling Amphibolus volubilis (Eohypsibiidae), and the limnic Dactylobiotus parthenogeneticus (Murrayidae). Cysts have been collected in nature, or induced under laboratory conditions. In the latter case, it was possible to follow the several steps of encystment processes. Two different types of cyst (“type 1” and “type 2”) have been found in A. volubilis, while in D. parthenogeneticus only one type has been found. In general, the ovoid-shaped cysts are constituted by a series of cuticles surrounding the animals and resemble an onion or a Matrioshka Russian doll. In all three types of cyst, the encystment processes show both common and peculiar traits. Encystment begins with the discharging of the sclerified parts of the buccal-pharyngeal apparatus, as in the molting process, but without the loss of the old cuticle. Then, two or three new cuticles are serially synthesized, according to cyst type. In A. volubilis, the ultrastructure of these new cuticular involucra is similar to that of non-encysted animal cuticles, while in D. parthenogeneticus the ultrastructure of the new cuticular involucra differs from that of non-encysted animal cuticle. A modified buccal-pharyngeal apparatus has been observed both in “type 2” cyst of A. volubilis and in the D. parthenogeneticus cyst.  相似文献
