Greenhouse experiments were conducted to assess the effect of salinisation of soil on emergence, growth, water content, proline content and mineral accumulation of seedlings of Delonix regia (Hook.) Raf. (Fabaceae). Sodium chloride (NaCl) was added to the soil and salinity was maintained at 0.3, 1.9, 3.9, 6.0 and 7.9 dS m−1. A negative relationship between seedling emergence and salt concentration was obtained. Salinity caused reduction in water content and water potential of tissues (leaves, stems, tap roots and lateral roots) that resulted in internal water deficit to plants. Consequently, shoot and root elongation, leaf expansion and dry matter accumulation in leaves, stems, tap roots and lateral root tissues of seedlings significantly decreased in response to increasing concentration of salt. Proline content in tissues was very low. There were no effective mechanisms to control net uptake of Na on root plasma membrane and subsequently its transport to shoot tissues. Potassium content significantly decreased in tissues in response to salinisation of soil. This tree species is a moderate salt-tolerant glycophytic plant. Nitrogen and calcium content in tissues significantly decreased as soil salinity increased. Phosphors content in tissues exhibited a declining trend with increase in soil salinity. Changes in tissues and whole-plant accumulation pattern of other elements tested, as well as possible mechanisms for avoidance of Na toxicity in this tree species in response to salinisation, are discussed.

Depending on salt concentrations, different mechanisms are involved in the tolerance of pistachio and an acclimation to salinity conditions occurs in the leaves that develop in the presence of salt.

Abstract

Pistachio (Pistacia vera L.) is a salt tolerant species that is considered an alternative crop for cultivation in salinzied orchard soils. In this work, 12-week-old pistachio seedlings cultivated in soil under greenhouse conditions were treated with five levels of salinity including control (0.63 dSm?1), low (2 and 4 dSm?1) and high (8 and 10 dSm?1) salt concentrations for further 12 weeks. Plant growth parameters were not affected by mild salinity; a significant reduction was only observed from 8 dSm?1. Considerable differences were observed between the young and mature leaves regarding osmotic and ionic stress effects of salt. Main compatible solutes were proline in mature leaves, proline and soluble sugars in young leaves, and soluble sugars and amino acids, other than proline, in roots. Concentration and content of Na in the leaves were not significantly increased at low levels of salinity and the K:Na and Ca:Na ratio of leaves were affected only by higher salt concentrations. Using the sequential extraction procedure for cell wall isolation, we observed that both absolute and relative amounts of Na in the cell wall fraction increased under low salinity, while decreased under higher levels of salt supply. Stable water relations, photochemistry and CO2 assimilation rates particularly of young leaves, as well as ion homeostasis were mechanisms for maintenance of plants growth under mild salinity. Under severe saline conditions, the impaired ability of mature leaves for synthesis of assimilates, preferent allocation of carbohydrates to roots for maintenance of osmotic homeostasis and finally, reduction of protein synthesis caused growth inhibition in pistachio.

The purpose of this study was to investigate the mechanisms underlying alleviation of salt stress by mycorrhization. Solanum lycopersicum L. cultivars Behta and Piazar with different salinity tolerance were cultivated in soil without salt (EC?=?0.63 dSm?1), with low (EC?=?5 dSm?1), or high (EC?=?10 dSm?1) salinity. Plants inoculated with the arbuscular mycorrhizal fungi Glomus intraradices (+AMF) were compared to non-inoculated plants (?AMF). Under salinity, AMF-mediated growth stimulation was higher in more salt tolerant Piazar than in sensitive Behta. Mycorrhization alleviated salt-induced reduction of P, Ca, and K uptake. Ca/Na and K/Na ratios were also better in +AMF. However, growth improvement by AMF was independent from plant P nutrition under high salinity. Mycorrhization improved the net assimilation rates through both elevating stomatal conductance and protecting photochemical processes of PSII against salinity. Higher activity of ROS scavenging enzymes was concomitant with lowering of H2O2, less lipid peroxidation, and higher proline in +AMF. Cultivar differences in growth responses to salinity and mycorrhization could be well explained by differences in ion balance, photochemistry, and gas exchange of leaves. Function of antioxidant defenses seemed responsible for different AMF-responsiveness of cultivars under salinity. In conclusion, AMF may protect plants against salinity by alleviating the salt-induced oxidative stress.

In the era of climate change, decreased precipitation and increased evapo-transpiration hampers the yield of several cereal crops along with the soil salinity and poor ground water resource. Wheat being the moderately tolerant crop face many challenges in the arid and semi-arid regions under irrigated agriculture. In view of this, the study was planned to explore the potential of durum wheat genotypes under salinity on the basis of physiological traits. Experiment was designed as RBD in three replications to evaluate 15 wheat genotypes with moderate saline irrigation (ECiw – 6 dS m−1) and extreme saline irrigation (ECiw – 10 dS m−1) along with one set of control (Best available water). Different physiological traits such as water potential (ψp), osmotic potential (ψs), relative water content (RWC), Na+ and K+ content were recorded in roots as well as shoots at the reproductive stage whereas photosynthetic rate and chlorophyll content were measured in the flag leaves. A significant variability (p < 0.001) was noted among the genotypes under different stress environments and it was observed that durum genotype HI 8728 and HI 8737 showed less reduction in plant water traits (RWC, ψp and ψs) than the salinity tolerant checks of bread wheat KRL 99 and KRL 3–4. HD 4728 and HI 8708 maintained higher photosynthetic rate as well as higher chlorophyll content under the extreme salinity level of ECiw – 10 dSm−1. No significant differences were found in root Na+ in genotypes KRL 99 (3.17g), KRL 3–4 (3.34g) and HI 8737 (3.41g) while in shoots, lowest accumulation was seen in KRL 99, MACS 3949 and KRL 3–4 at ECiw – 10 dSm−1. The mean range of K+ content was 7.60–9.74% in roots and 4.21–6.61% in shoots under control environment which decreased to 50.77% in roots and 46.05% in shoots under extreme salinity condition of ECiw – 10 dSm−1. At ECiw – 10 dSm−1, KRL 99 maintained highest K+/Na+ in both root and shoot followed by KRL 3–4, HI 8737, MACS 3949, HD 4728 in roots and MACS 3949, KRL 3–4, MACS 4020, HD 4758, MACS 3972 and HI 8713 in shoots. The differential response of durum wheat genotypes under salinity particularly for physiological traits, confer their adaptability towards stress environments and exhibit their potential as genetic sources in breeding programs for improving salt stress tolerance.

The relationship between Na+ accumulation and salt tolerance was tested by comparing subspecies of the halophyte, Atriplex canescens (fourwing saltbush), that differed markedly in Na+ content and Na:K ratios. Above ground tissues of one low-sodium and two high-sodium subspecies were compared with respect to cation accumulation, osmotic adjustment and growth along a salinity gradient in greenhouse trials. Plants of each subspecies were grown for 80 d on 2.2, 180, 540 and 720 mol m?3 NaCl. At harvest, A. canescens ssp. canescens had significantly lower Na+ levels, higher K+ levels and lower Na:K ratios in leaf and stem tissues than A. canescens ssp. macropoda and linearis over the salinity range (P < 0.05 or 0.01). Na:K ratios in leaves of the latter two, high-sodium, subspecies were approximately 2 on the lowest salinity treatment and ranged from 5 to 10 on the more saline solutions. By contrast, Na:K ratios in leaves of the low-sodium subspecies canescens, were only 0.4 on the lowest salinity and ranged narrowly from 1.7 to 2.3 at higher salinities. However, despite different patterns of Na+ and K+ accumulation, all three subspecies exhibited equally high salt tolerance and had similar osmotic pressures in their leaves or stems over the salinity range. Contrary to expectations, high salt tolerance was not necessarily dependent on high levels of Na+ accumulation in this species.

This study establishes relationships between salt resistance and solute accumulation in roots and leaves of two contrasting cashew species. The sensitive (Anacardium microcarpum) and resistant (A. occidentale) species showed maximum root LD50 values (the external NaCl concentration required for a 50% reduction in dry weight) of 63 and 128?mM NaCl, whereas the shoot LD50 values were 90 and 132?mM, respectively. The salt sensitivity was directly associated with Na+ accumulation and especially with the Cl? content in leaves and to a minor extent in roots. The accumulation of saline ions was associated with higher net uptake rates by roots and transport rates from root to shoot in the sensitive cashew species. The K+/Na+ ratios were not associated with salt resistance either in roots or leaves. Proline and free amino acid concentrations were strongly increased by salinity, especially in the leaves of the resistant species. The soluble sugar concentrations were not influenced by NaCl treatments in leaves of both species. In contrast, the root soluble sugar content was significantly decreased by salinity in the sensitive species only. In conclusion, the higher salt sensitivity of A. microcarpum is associated to an inefficient salt exclusion system of the leaves, especially for Cl?. On the other hand, the resistant species displays higher concentrations of organic solutes especially a salt-induced accumulation of proline and free amino acids in leaves.

The salt tolerance of the commercial F1 tomato hybrid (Lycopersicon esculentum Mill) Radja (GC-793) has been agronomically and physiologically evaluated under greenhouse conditions, using a control (nutrient solution), a moderate (70 mM NaCl added to the nutrient solution) and a high salt level (140 mM NaCl), applied for 130 days. The results show that Radja is a Na+-excluder genotype, tolerant to moderate salinity. Fruit yield was reduced by 16% and 60% and the shoot biomass by 30% and more than 75% under moderate and high salinities, respectively. At 90 days of salt treatment (DST), the mature leaves feeding the 4th truss at fruiting accumulated little Na+ (178 mmol kg-1 DW). At this time, the sucrose concentration in these leaves even increased with moderate salinity and the amino acid proline was not accumulated under salt conditions as compared to control. At 130 DST, Na+ was accumulated mainly by the roots in proportion to the salt level applied, while in leaves appreciable amounts were found only at high salinity (452 mmol kg-1 DW). In the leaves, Cl- was always accumulated in proportion to the salt level and in a very much greater amounts than Na+ (until 1640 mmol kg-1 DW). The sucrose content was reduced in all plants by salinity, and was distributed preferentially toward the distal stem and peduncle of a truss at fruiting under moderate salinity, and toward the basal stem and root at high salinity. Moreover, proline was accumulated in different organs of the plant only at high salinity, coinciding with Na+ accumulation in leaves. Attempts are made to find a clear relationship between physiological behaviour triggered by stress and the agronomical behaviour, in order to assess the validity of physiological traits used for salt-tolerance selection and breeding in tomato.

Control of sodium transport in durum wheat
In many species, salt sensitivity is associated with the accumulation of sodium (Na(+)) in photosynthetic tissues. Na(+) uptake to leaves involves a series of transport steps and so far very few candidate genes have been implicated in the control of these processes. In this study, Na(+) transport was compared in two varieties of durum wheat (Triticum turgidum) L. subsp. durum known to differ in salt tolerance and Na(+) accumulation; the relatively salt tolerant landrace line 149 and the salt sensitive cultivar Tamaroi. Genetic studies indicated that these genotypes differed at two major loci controlling leaf blade Na(+) accumulation (R. Munns, G.J. Rebetzke, S. Husain, R.A. James, R.A. Hare [2003] Aust J Agric Res 54: 627-635). The physiological traits determined by these genetic differences were investigated using measurements of unidirectional (22)Na(+) transport and net Na(+) accumulation. The major differences in Na(+) transport between the genotypes were (1) the rate of transfer from the root to the shoot (xylem loading), which was much lower in the salt tolerant genotype, and (2) the capacity of the leaf sheath to extract and sequester Na(+) as it entered the leaf. The genotypes did not differ significantly in unidirectional root uptake of Na(+) and there was no evidence for recirculation of Na(+) from shoots to roots. It is likely that xylem loading and leaf sheath sequestration are separate genetic traits that interact to control leaf blade Na(+).

Identification and characterization of proteins involved in salt tolerance are imperative for revealing its genetic mechanisms. In this study, ionic and proteomic responses of a Tibetan wild barley XZ16 and a well‐known salt‐tolerant barley cv. CM72 were analyzed using inductively coupled plasma‐optical emission spectrometer, 2DE, and MALDI‐TOF/TOF MS techniques to determine salt‐induced differences in element and protein profiles between the two genotypes. In total, 41 differentially expressed proteins were identified in roots and leaves, and they were associated with ion homeostasis, cell redox homeostasis, metabolic process, and photosynthesis. Under salinity stress, calmodulin, Na/K transporters, and H+‐ATPases were involved in establishment of ion homeostasis for barley plants. Moreover, ribulose‐1,5‐bisphosphate carboxylase/oxygenase activase and oxygen‐evolving enhancer proteins were significantly upregulated under salinity stress, indicating the great impact of salinity on photosynthesis. In comparison with CM72, XZ16 had greater relative dry weight and lower Na accumulation in the shoots under salinity stress. A higher expression of HvNHX1 in the roots, and some specific proteins responsible for ion homeostasis and cell redox homeostasis, was also found in XZ16 exposed to salt stress. The current results showed that Tibetan wild barley XZ16 and cultivated barley cultivar CM72 differ in the mechanism of salt tolerance.

Salt affected soil inhibits plant growth, development and productivity, especially in case of rice crop. Ion homeostasis is a candidate defense mechanism in the salt tolerant plants or halophyte species, where the salt toxic ions are stored in the vacuoles. The aim of this investigation was to determine the OsNHX1 (a vacuolar Na+/H+ exchanger) and OsHKT2;1 (Na+/K+ transporter) regulation by salt stress (200 mM NaCl) in two rice cultivars, i.e. Pokkali (salt tolerant) and IR29 (salt susceptible), the accumulation of Na+ in the root and leaf tissues using CoroNa Green® staining dye and the associated physiological changes in test plants. Na+ content was largely increased in the root tissues of rice seedlings cv. Pokkali (15 min after salt stress) due to the higher expression of OsHKT2;1 gene (by 2.5 folds) in the root tissues. The expression of OsNHX1 gene in the leaf tissues was evidently increased in salt stressed seedlings of Pokkali, whereas it was unchanged in salt stressed seedlings of IR29. Na+ in the root tissues of both Pokkali and IR29 was enriched, when subjected to 200 mM NaCl for 12 h and easily detected in the leaf tissues of salt stressed plants exposed for 24 h, especially in cv. Pokkali. Moreover, the overexpression of OsNHX1 gene regulated the translocation of Na+ from root to leaf tissues, and compartmentation of Na+ into vacuoles, thereby maintaining the photosynthetic abilities in cv. Pokkali. Overall growth performance, maximum quantum yield (Fv/Fm), photon yield of PSII (ΦPSII) and net photosynthetic rate (Pn) was improved in salt stressed leaves of Pokkali than those in salt stressed IR29.

The effects of three concentrations of sodium chloride (NaCl) on seven citrus rootstocks were studied under greenhouse conditions. Leaf and root mineral concentrations and seedling growth were measured. Sodium chloride was added to the nutrient solution to achieve final osmotic potentials of –0.10, –0.20, and –0.35 MPa. Increasing the concentration of NaCl in the nutrition solution reduced growth proportionally and altered leaf and root mineral concentrations of all rootstocks. Significant differences in leaf and root mineral concentration among rootstocks were also found under stressed and non-stressed conditions. Salinity caused the greatest growth reduction in Milam lemon and trifoliate orange and the least reduction in sour orange and Cleopatra mandarin. No specific nutrient deficiency was the sole factor reducing growth and causing injury to citrus rootstocks. Sodium chloride sensitivity of citrus rootstocks in terms of leaf burn symptoms and growth reduction could be attributed more to Cl than to Na. Sodium and Cl concentrations were greater in the leaves than in the roots, particularly at the medium and high salinity levels. Root Cl was not useful for assessing injury because no differences were found in root Cl concentrations among rootstocks. Increasing salinity level did not affect the level of N and Ca in the roots but did reduce N and Ca levels in the leaves. No relationship in mineral concentration or accumulation seemed to exist between citrus leaves and roots. At the –0.10 MPa salinity level, sour orange, rough lemon, and Milam were not able to exclude either Na or Cl from their leaves. Trifoliate orange and its two hybrids (Swingle citrumelo and Carrizo citrange) excluded Na at the lowest salt level used, but were unable to exclude Na at the higher salinity levels. Similarly, Cleopatra mandarin excluded Cl at the lowest salt level, but was not able to exclude Cl at higher salt concentrations. Hence, the ability of citrus rootstocks to exclude Na or Cl breaks down at higher salt concentrations.Florida Agricultural Experiment Station Journal Series No. R-02276.

The effects of ionic stress on the physiology and gene expression of two rice genotypes (IR4630 and IR15324) that differ in salt tolerance, were investigated by evaluating changes in the biomass, Na+ and K+ concentrations and applying the cDNA-AFLP technique to highlight changes in gene expression. Over 8 days of salinisation, the effect of NaCl on the reduction of biomass (dry weight) was apparent from 24 h after salinisation (the first time point), indicating that the consequences of the build up of Na+ (and Cl-) in the leaves of both lines was rapid. Furthermore, root growth of IR15324 was much more sensitive to salt than that of IR4630 (the reduction in root dry weight compared to non-salinised plants was three times greater in IR15324 than IR4630). The two rice lines also differed in their Na+ accumulation in saline conditions, a difference that was more marked in the shoots, particularly at the final harvest, than in the roots. Under salt stress, the K+ content (µmol/shoot) increased over four successive harvests (24, 48, 96, 192 h) in both lines, but was always greater in IR4630 than in IR15324: differences in Na+/K+ ratio appear to be an important determinant of salt tolerance in rice. To separate osmotic from ionic effects of salt, mannitol was applied as a non-ionic osmoticum at an osmotic potential estimated to be equivalent to 50 mM NaCl. Messenger RNA was sampled at 0.5, 6, 24, 48 and 192 hours after salinisation. Several products (AFLP-bands) were detected, which were upregulated in the response to ionic effects of salt in the tolerant line (IR4630) and not expressed in the sensitive line (IR15324). Bioinformatic analysis indicated three of these AFLP-bands have a high-degree of sequence similarity with the genes encoding a proline rich protein, senescence associated protein and heat-shock protein. The data are novel in that they differentially highlight changes induced by the ionic rather than osmotic effects of salt and in a tolerant rather than a sensitive genotype. The possible roles of the products of these genes are discussed.

Two soybean (Glycine max (L.) Merr.) cultivars, Tachiyutaka and Dare, were grown in pots at 30 and 70 % relative humidity (RH) and treated with 0 (control), 40 (moderate), 80 and 120 (severe) mM NaCl for 3 weeks. Increasing RH enhanced growth of salt sensitive cultivar, Tachiyutaka, but had no effect on salt tolerant cultivar, Dare, under control and moderate saline conditions. Both cultivars benefited from elevated humidity under severe saline conditions. Cultivar Tachiyutaka had poorer ability for controlling translocation of Na+ to the leaves, lower Na+ exclusion ability in the roots, and lower root activity under NaCl treatment, compared with cv. Dare. The increased growth of cv. Tachiyutaka at high RH was consistent with decreased Na+ accumulation in the leaves, increased stomatal conductance and root activity, while the unchanged growth of cv. Dare was consistent with similar Na+ accumulation in the leaves, and the decreased root activity.

Impacts of salinity become severe when the soil is deficient in oxygen. Oxygation (using aerated water for subsurface drip irrigation of crop) could minimize the impact of salinity on plants under oxygen-limiting soil environments. Pot experiments were conducted to evaluate the effects of oxygation (12% air volume/volume of water) on vegetable soybean (moderately salt tolerant) and cotton (salt tolerant) in a salinized vertisol at 2, 8, 14, 20 dS/m ECe. In vegetable soybean, oxygation increased above ground biomass yield and water use efficiency (WUE) by 13% and 22%, respectively, compared with the control. Higher yield with oxygation was accompanied by greater plant height and stem diameter and reduced specific leaf area and leaf Na+ and Cl-concentrations. In cotton, oxygation increased lint yield and WUE by 18% and 16%, respectively, compared with the control, and was accompanied by greater canopy light interception, plant height and stem diameter. Oxygation also led to a greater rate of photosynthesis, higher relative water content in the leaf, reduced crop water stress index and lower leaf water potential. It did not, however, affect leaf Na+ or Cl- concentration. Oxygation invariably increased, whereas salinity reduced the K+ : Na+ ratio in the leaves of both species. Oxygation improved yield and WUE performance of salt tolerant and moderately tolerant crops under saline soil environments, and this may have a significant impact for irrigated agriculture where saline soils pose constraints to crop production.

Summary Oryza coarctata, a highly salt-resistant wild rice species, is commonly found on the banks of coastal rivers in India. This species can also withstand saline water (20 to 40 dSm−1 E.C) submergence for quite a long period. It was revealed thatO. coarctata has some special unicellular salt hairs (trichomes) on the adaxial surface of the leaves, by which they efficiently maintain a low concentration of toxic salts in the plant tissue. Sodium and chloride were the dominant ions in the excreted material but they also excrete potassium, magnesium and calcium. With the increase in soil salinity sodium, magnesium and chloride excretion increased.O. coarctata maintained the optimum mineral concentration in its tissues. Maximum accumulation of potassium was observed in the leaves. With the increase in salt stress total biomass production and osmotic potential increased over control but there was no change in the moisture percentage of leaves.

This paper describes experiments on Cl transport into the roots, stem and leaves of bean plants, the roots of which have been exposed to lipids in the root solution. Monoand digalactose diglyceride strongly increased Cl transport into all plant parts, probably by transport of the glycolipids further into the plant. Phosphatidyl choline increased Cl absorption by the roots, but transport into the stem and leaves was not affected. This phospholipid was only absorbed by the root tissue. 32P-glycerophosphoryl choline added to the root solution was readily transported and esterified as phospholipid in all plant parts. This chemical did increase Cl uptake by the roots but Cl accumulation in the leaves was reduced by as much as 40%. Phosphatidyl glycerol, phosphatidyl inositol, and sulfolipid increased Cl transport into roots, stem, and leaves, and a high mobility of 32P-phosphatidyl glycerol was demonstrated. Generally no significant effect of the above lipids on Na transport in beans and cotton was noted except that monogalactose diglyceride did increase Na transport in cotton.
