The carbon metabolism of the cryptoendolithic microbiota of sandstones from the Ross Desert of Antarctica was studied in situ and in vitro. Organic and inorganic carbon compounds were metabolized by the microbiota, with bicarbonate incorporation into community lipids occurring primarily in the light. Light intensity affected the photometabolism of carbon with a photosynthesis-intensity response optimum at about 200 to 300 micromoles of photons per m2 per s. Photosynthesis was also affected by temperature, with a minimum activity at -5 degrees C, an optimum activity at 15 degrees C, and complete inhibition at 35 degrees C, indicating that the cryptoendolithic community was psychrophilic. The primary source of CO2 for photosynthesis in situ was the atmosphere. CO2 may also be photometabolized by using the carbon produced from respiration within the endolithic community. Photosynthesis occurred maximally when the microbiota was wet with liquid water and to a lesser extent in a humid atmosphere. This simple microbial community, therefore, exists under extremes of water, light, and temperature stress which affect and control its metabolism.  相似文献

To help understand carbon balance between shoots and developing roots, 41 bare-root crassulacean acid metabolism (CAM) plants native to the Sonoran Desert were studied in a glass-panelled sealable room at day/night air temperatures of 25/15 degrees C. Net CO(2) uptake by the community of Agave schottii, Carnegia gigantea, Cylindropuntia versicolor, Ferocactus wislizenii and Opuntia engelmannii occurred 3 weeks after watering. At 4 weeks, the net CO(2) uptake rate measured for south-east-facing younger parts of the shoots averaged 1.94 micro mol m(-2) s(-1) at night, considerably higher than the community-level nocturnal net CO(2) uptake averaged over the total shoot surface, primarily reflecting the influences of surface orientation on radiation interception (predicted net CO(2) uptake is twice as high for south-east-facing surfaces compared with all compass directions). Estimated growth plus maintenance respiration of the roots averaged 0.10 micro mol m(-2) s(-1) over the 13-week period, when the community had a net carbon gain from the atmosphere of 4 mol C while the structural C incorporated into the roots was 23 mol. Thus, these five CAM species diverted all net C uptake over the 13-week period plus some existing shoot C to newly developing roots. Only after sufficient roots develop to support shoot water and nutrient requirements will the plant community have net above-ground biomass gains.  相似文献

植物叶片水分利用效率的高低取决于气孔控制的光合作用和蒸腾作用两个相互耦合的过程,模拟水分利用效率对环境变化的响应特征和机制是理解生态系统碳循环和水循环及其耦合关系的基础。研究通过人工控制光强和CO2浓度,对叶片水分利用效率进行了研究。提出了植物水分利用效率在光强和CO2浓度共同作用下的估算模型。数据分析表明,该模型在包括C3和C4植物、草本和木本植物在内的9种植物上能很好地模拟水分利用效率对光强和CO2浓度共同作用的响应。该模型可以用于估算CO2浓度升高条件下光合速率的提高和蒸腾速率的降低对水分利用效率提高的贡献量。CO2浓度变化条件下,水分利用效率在不同植物之间有巨大差异,研究区域尺度植物的水分利用效率时至少需要将植物区分为C4植物和C3植物,其中C3植物区分为草本和木本植物3种生态功能型才能较为准确地估算植物的整体水分利用效率。应用本研究提出的水分利用效率估算模型和植物水分利用效率生态功能型分类标准,可以为建立以植物的水分利用效率为基本参数的陆地生态系统水循环模型和陆地生态系统生产力模型提供重要依据。  相似文献

Angiosperm plants were grown under either the present day 21 kPa O(2) atmosphere or 28 kPa, as estimated for the end-Cretaceous (100-65 MyBP). CO(2) was held at different levels, within the 24-60 Pa range, as also estimated for the same period. In C(3) Xanthium strumarium and Atriplex prostrata, leaf area and net photosynthesis per unit leaf area, were reduced by the high O(2), while the whole-plant respiration/photosynthesis ratio increased. The high O(2) effects were strongest under 24 Pa, but still significant under 60 Pa CO(2). Growth was reduced by high O(2) in these C(3) species, but not in Flaveria sp., whether C(3), C(4), or intermediary grown under light intensities <350 micromol m(-2) s(-1) PPF. Photosynthesis of C(3) Flaveria sp. was reduced by high O(2), but only at light intensities >350 micromol m(-2) s(-1) PPF. It is concluded that the high O(2) atmosphere at the end-Cretaceous would have reduced growth of at least some of the vegetation, thus adversely affecting dependent fauna. The weakened biota would have been predisposed to the consequences of volcanism and the K/T boundary bolide impact. Conversely, photosynthesis and growth of C(4) Zea mays and Atriplex halimus were little affected by high, 28 kPa, O(2). This suggests an environmental driver for the evolution of C(4) physiology.  相似文献

Photosynthesis, the conversion of light energy into chemical energy, is a critical biological process, whereby plants synthesize carbohydrates from light, carbon dioxide (CO2) and water. The influence of gravity on this biological process, however, is not well understood. Thus, centrifugation was used to alter the gravity environment of Euglena gracilis grown on nutritive agar plates illuminated with red and blue light emitting diodes. The results showed that hypergravity (up to 10xg) had an inhibitory effect on photosynthetic CO2 fixation. Chlorophyll accumulation per cell was essentially unaffected by treatment; however, Chl a/Chl b ratios decreased in hypergravity when compared to 1xg controls. Photosynthesis in Euglena appears to have limited tolerance for even moderate changes in gravitational acceleration.  相似文献

C(4) plants are rare in the cool climates characteristic of high latitudes and elevations, but the reasons for this are unclear. We tested the hypothesis that CO(2) fixation by Rubisco is the rate-limiting step during C(4) photosynthesis at cool temperatures. We measured photosynthesis and chlorophyll fluorescence from 6 degrees C to 40 degrees C, and in vitro Rubisco and phosphoenolpyruvate carboxylase activity from 0 degrees C to 42 degrees C, in Flaveria bidentis modified by an antisense construct (targeted to the nuclear-encoded small subunit of Rubisco, anti-RbcS) to have 49% and 32% of the wild-type Rubisco content. Photosynthesis was reduced at all temperatures in the anti-Rbcs plants, but the thermal optimum for photosynthesis (35 degrees C) did not differ. The in vitro turnover rate (kcat) of fully carbamylated Rubisco was 3.8 mol mol(-)(1) s(-)(1) at 24 degrees C, regardless of genotype. The in vitro kcat (Rubisco Vcmax per catalytic site) and in vivo kcat (gross photosynthesis per Rubisco catalytic site) were the same below 20 degrees C, but at warmer temperatures, the in vitro capacity of the enzyme exceeded the realized rate of photosynthesis. The quantum requirement of CO(2) assimilation increased below 25 degrees C in all genotypes, suggesting greater leakage of CO(2) from the bundle sheath. The Rubisco flux control coefficient was 0.68 at the thermal optimum and increased to 0.99 at 6 degrees C. Our results thus demonstrate that Rubisco capacity is a principle control over the rate of C(4) photosynthesis at low temperatures. On the basis of these results, we propose that the lack of C(4) success in cool climates reflects a constraint imposed by having less Rubisco than their C(3) competitors.  相似文献

Supercritical carbon dioxide (SC CO(2)) has been evaluated as a new sterilization technology. Results are presented on killing of B. pumilus spores using SC CO(2) containing trace levels of additives. Complete killing was achieved with 200 part per million (ppm) hydrogen peroxide in SC CO(2) at 60 degrees C, 27.5 MPa. Addition of water to SC CO(2) resulted in greater than three-log killing, but this is insufficient to claim sterilization. Neither ethanol nor isopropanol when added to SC CO(2) affected killing.  相似文献

The main forms of terrestrial life in the cold, desolate Ross Desert of Antarctica are lichen-dominated or cyanobacterium-dominated cryptoendolithic (hidden in rock) microbial communities. Though microbial community biomass (as measured by extractable lipid phosphate) was well within the range of values determined for other microbial communities, community lipid carbon turnover times (calculated from community lipid biomass, rates of community photosynthetic carbon incorporation into lipids versus temperature, and the in situ temperature record) were among the longest on Earth (ca. 20,000 years). When the temperature is above freezing and moisture is present, moderate rates of photosynthesis can be measured. Lichen communities had a psychrophilic temperature response (maximal rate of 4.5 ng of C h-1 m-2 at 10°C) while cyanobacteria communities had maximal rates at 20 to 30°C (3 ng of C h-1 m-2). These extraordinarily slowly growing communities were not nutrient limited. No significant changes in photosynthetic metabolism were observed upon additions of 100 nM to 1 mM nitrate, ammonium, phosphate, and manganese. These simple, tenacious microbial communities demonstrate strategies of survival under conditions normally considered too extreme for life.  相似文献

High carbon dioxide atmospheres combined with high temperature were effective for controlling Indian meal moth, Plodia interpunctella (Hübner) pupae. Pupae were exposed to atmospheres of 60, 80, or 98% carbon dioxide (CO2) in nitrogen (N2), or 60 or 80% CO2 in air at temperatures of 26.7 degrees C or 32.2 degrees C and 60% RH. Controlled atmosphere treatments at 32.2 degrees C controlled pupae faster than the same treatments at the lower temperature. At both temperatures high CO2 concentration treatments combined with nitrogen killed pupae faster than high CO2 concentration treatments combined with air. Exposure to 80% carbon dioxide mixed with nitrogen was the most effective treatment causing 100% mortality in 12 h at 32.2 degrees C and 93.3% mortality in 18 h at 26.6 degrees C. High-temperature controlled atmosphere treatments had no adverse effects on quality of two preserved floral products, Limonium sinuatum (L.) and Gypsophila elegans (Bieb.), tested for 12, 18, and 24 h according to industry standards.  相似文献

Microbial communities growing in the bed of the alkaline, sulfide hot spring Bol'sherechenskii (the Baikal rift area) were studied over many years (1986-2001). The effluent water temperature ranged from 72 to 74 degrees C, pH was from 9.25 to 9.8, and sulfide content was from 12 to 13.4 mg/ml. Simultaneous effects of several extreme factors restrict the spread of phototrophic microorganisms. Visible microbial fouling appears with a decrease in the temperature to 62 degrees C and in the sulfide content to 5.9 mg/l. Cyanobacteria predominated in all biological zones of the microbial mat. The filamentous cyanobacteria of the genus Phormidium are the major mat-forming organisms, whereas unicellular cyanobacteria and the filamentous green bacterium Chloroflexus aurantiacus are minor components of the phototrophic communities. No cyanobacteria of the species Mastigocladus laminosus, typical of neutral and subacid springs, were identified. Seventeen species of both anoxygenic phototrophic bacteria and cyanobacteria were isolated from the microbial mats, most of which exhibited optimum growth at 20 to 45 degrees C. The anoxygenic phototrophs were neutrophiles with pH optimum at about 7. The cyanobacteria were the most adapted to the alkaline conditions in the spring. Their optimum growth was observed at pH 8.5-9.0. As determined by the in situ radioisotope method, the optimal growth and decomposition rates were observed at 40-32 degrees C, which is 10 to 15 degrees C lower than the same parameter in the sulfide-deficient Octopus Spring (Yellowstone, United States). The maximum chlorophyll a concentration was 555 mg/m2 at 40 degrees C. Total rate of photosynthesis in the mats reached 1.3 g C/m2 per day. The maximum rate of dark fixation of carbon dioxide in the microbial mats was 0.806 g C/m2 per day. The maximum rate of sulfate reduction comprised 0.367 g S/m2 per day at 40 degrees C. The rate of methanogenesis did not exceed 1.188 micrograms C/m2 per day. The role of methanogenesis in the terminal decomposition of the organic matter was insignificant. Methane formation consumed 100 times less organic matter than sulfate reduction.  相似文献

Inorganic carbon can be in short supply in freshwater relative to that needed by freshwater plants for photosynthesis because of a large external transport limitation coupled with frequent depleted concentrations of CO(2) and elevated concentrations of O(2). Freshwater plants have evolved a host of avoidance, exploitation and amelioration strategies to cope with the low and variable supply of inorganic carbon in water. Avoidance strategies rely on the spatial variation in CO(2) concentrations within and among lakes. Exploitation strategies involve anatomical and morphological features that take advantage of sources of CO(2) outside of the water column such as the atmosphere or sediment. Amelioration strategies involve carbon-concentrating mechanisms based on uptake of bicarbonate, which is widespread, C(4)-fixation, which is infrequent, and crassulacean acid metabolism (CAM), which is of intermediate frequency. CAM enables aquatic plants to take up inorganic carbon in the night. Furthermore, daytime inorganic carbon uptake is generally not inhibited and therefore CAM is considered to be a carbon-conserving mechanism. CAM in aquatic plants is a plastic mechanism regulated by environmental variables and is generally downregulated when inorganic carbon does not limit photosynthesis. CAM is regulated in the long term (acclimation during growth), but is also affected by environmental conditions in the short term (response on a daily basis). In aquatic plants, CAM appears to be an ecologically important mechanism for increasing inorganic carbon uptake, because the in situ contribution from CAM to the C-budget generally is high (18-55%).  相似文献

We studied the carbon dioxide fixation activity in a stratified hypereutrophic karstic lagoon using a combination of fingerprinting techniques targeting bacterial and archaeal 16S rRNA genes, functional gene cloning [the acetyl-CoA carboxylase (accC)], and isotopic labelling ((14)C-bicarbonate) coupled to single-cell analyses [microautoradiography combined with catalyzed reported deposition-FISH (MAR-CARD-FISH)]. The microbial planktonic community was dominated by bacteria with maximal abundances of archaea just below the oxic/anoxic transition zone (7% of total cells). In situ incubations with radiolabelled bicarbonate showed maximal photoassimilation activity in the oxic epilimnion, whereas dark CO(2) fixation was consistently observed throughout the water column, with a maximum at the oxic/anoxic interface (8.6 mg C m(-3) h(-1)). The contributions of light and dark carbon fixation activities in the whole water column were 69% and 31% of the total C incorporated, respectively. MAR-CARD-FISH incubations corroborated these results and revealed that the highest fraction of bacterial and archaeal cells actively uptaking bicarbonate in the light was found at the surface. The bacterial community was mainly composed of green sulfur bacteria (Chlorobi) and members of the Betaproteobacteria and the Bacteroidetes. The archaeal assemblage was composed of phylotypes of the Miscellaneous Crenarchaeotic Group and a few methanogens. Clone libraries of the accC gene showed an absolute dominance of bacterial carboxylases. Our results suggest that the dark carbon fixation activity measured was mainly related to CO(2) incorporation by heterotrophs rather than to the activity of true chemoautotrophs.  相似文献

Sites in the West Siberian peat bog 'Bakchar' were acidic (pH 4.2-4.8), low in nutrients, and emitted CH4 at rates of 0.2-1.5 mmol m(-2) h(-1). The vertical profile of delta13CH4 and delta13CO2 dissolved in the porewater indicated increasing isotope fractionation and thus increasing contribution of H2/CO2-dependent methanogenesis with depth. The anaerobic microbial community at 30-50 cm below the water table produced CH4 with optimum activity at 20-25 degrees C and pH 5.0-5.5 respectively. Inhibition of methanogenesis with 2-bromo-ethane sulphonate showed that acetate, phenyl acetate, phenyl propionate and caproate were important intermediates in the degradation pathway of organic matter to CH4. Further degradation of these intermediates indicated that 62-72% of the CH4 was ultimately derived from acetate, the remainder from H2/CO2. Turnover times of [2-14C]acetate were on the order of 2 days (15, 25 degrees C) and accounted for 60-65% of total CH4 production. Conversion of 14CO2 to 14CH4 accounted for 35-43% of total CH4 production. These results showed that acetoclastic and hydrogenotrophic methanogenesis operated closely at a ratio of approximately 2 : 1 irrespective of the incubation temperature (4, 15 and 25 degrees C). The composition of the archaeal community was determined in the peat samples by terminal restriction fragment length polymorphism (T-RFLP) analysis and sequencing of amplified SSU rRNA gene fragments, and showed that members of Methanomicrobiaceae, Methanosarcinaceae and Rice cluster II (RC-II) were present. Other, presumably non-methanogenic archaeal clusters (group III, RC-IV, RC-V, RC-VI) were also detected. Fluorescent in situ hybridization (FISH) showed that the number of Bacteria decreased (from 24 x 10(7) to 4 x 10(7) cells per gram peat) with depth (from 5 to 55 cm below the water table), whereas the numbers of Archaea slightly increased (from 1 x 10(7) to 2 x 10(7) cells per gram peat). Methanosarcina spp. accounted for about half of the archaeal cells. Our results show that both hydrogenotrophic and acetoclastic methanogenesis are an integral part of the CH4-producing pathway in acidic peat and were represented by appropriate methanogenic populations.  相似文献

The winter photosynthetic activity (quantified by net CO(2) assimilation rates and chlorophyll (Chl) a fluorescence parameters) of 20 plant species (including two lichens and two mosses) of a Hungarian temperate semi-desert sand grassland was determined on one occasion per year in 1984, 1989 and 1994. Throughout winter, the overwintering green shoots, leaves or thalli were regularly exposed to below zero temperatures at night and daytime temperatures of 0-5 degrees C. In situ tissue temperature varied between -2.1 and +6.9 degrees C and the photosynthetic photon flux density (PPFD) between 137 and 351 micromol m(-2)s(-1). Under these conditions 18 of the grassland species exhibited photosynthetic CO(2) uptake (range: vascular plants ca. 0.2-3.8 micromol m(-2)s(-1), cryptogams 0.3-2.79 micromol kg(-1)s(-1)) and values of 0.9-5.1 of the Chl fluorescence decrease ratio R(Fd). In 1984, Festuca vaginata and Sedum sexangulare had net CO(2) assimilation at leaf temperatures of -0.85 to -1.2 degrees C. In 1989, all species except Cladonia furcata showed net CO(2) assimilation at tissue temperatures of 0 to +3.3 degrees C, with the highest rates observed in Poa bulbosa and F. vaginata. The latter showed a net CO(2) assimilation saturation at a PPFD of 600 micromol m(-2)s(-1) and a temperature optimum between +5 and +18 degrees C. At the 1994 measurements, the photosynthetic rates were higher at higher tissue water contents. The two mosses and lichens had a net photosynthesis (range: 1.1-2.79 micromol CO(2)kg(-1)s(-1)) at 2 degrees C tissue temperature and at 4-5 degrees C air temperature. Ca. 80% of the vascular grassland plant species maintained a positive C-balance during the coldest periods of winter, with photosynthetic rates of 1.5-3.8 micromol CO(2)m(-2)s(-1). In an extremely warm beginning March of the relatively warm winter of 2006/2007, the dicotyledonous plants had much higher CO(2) assimilation rates on a Chl (range 6-14.9 micromol g(-1)Chl s(-1)) and on a dry weight basis (9-48 micromol kg(-1)dw s(-1)) than in the cold winter of 1994. However, the assimilation rates of the three investigated cryptogams (Tortula and two Cladonia) and the two grasses Festuca and Poa were not affected by this increase. The results indicate that the photosynthetic activity of temperate semi-desert sand grassland species can help somewhat in slowing the general CO(2) rise in winter and function as a potential carbon sink of the investigated semi-desert Hungarian grassland species.  相似文献

CO(2) transfer conductance from the intercellular airspaces of the leaf into the chloroplast, defined as mesophyll conductance (g(m)), is finite. Therefore, it will limit photosynthesis when CO(2) is not saturating, as in C3 leaves in the present atmosphere. Little is known about the processes that determine the magnitude of g(m). The process dominating g(m) is uncertain, though carbonic anhydrase, aquaporins, and the diffusivity of CO(2) in water have all been suggested. The response of g(m) to temperature (10 degrees C-40 degrees C) in mature leaves of tobacco (Nicotiana tabacum L. cv W38) was determined using measurements of leaf carbon dioxide and water vapor exchange, coupled with modulated chlorophyll fluorescence. These measurements revealed a temperature coefficient (Q(10)) of approximately 2.2 for g(m), suggesting control by a protein-facilitated process because the Q(10) for diffusion of CO(2) in water is about 1.25. Further, g(m) values are maximal at 35 degrees C to 37.5 degrees C, again suggesting a protein-facilitated process, but with a lower energy of deactivation than Rubisco. Using the temperature response of g(m) to calculate CO(2) at Rubisco, the kinetic parameters of Rubisco were calculated in vivo from 10 degrees C to 40 degrees C. Using these parameters, we determined the limitation imposed on photosynthesis by g(m). Despite an exponential rise with temperature, g(m) does not keep pace with increased capacity for CO(2) uptake at the site of Rubisco. The fraction of the total limitations to CO(2) uptake within the leaf attributable to g(m) rose from 0.10 at 10 degrees C to 0.22 at 40 degrees C. This shows that transfer of CO(2) from the intercellular air space to Rubisco is a very substantial limitation on photosynthesis, especially at high temperature.  相似文献

In Antarctica there exists a cryptoendolithic microbiota which survives extremely low temperatures. Fatty acid analysis of the membrane phospholipids of this microbial community showed a predominance of polyunsaturated fatty acids. Artificial membranes made from the purified community phospholipids remained fluid to below −20°C and had unusual hydration properties.  相似文献

植物稳定碳同位素技术是近年兴起的一项快速、可靠的技术。利用稳定碳同位素技术可以揭示碳同化的过程的许多方面的信息。1 3C和1 2 C同位素效应 ,使它们在进行碳循环时发生稳定碳同位素的分馏。植物光合作用过程中CO2 经气孔扩散分差和RUBPCase及PEPCase羧化分馏是造成植物稳定碳同位素比率 (R =1 3C/ 1 2 C)不同于源CO2 中碳同位素比率的主要原因。遗传因素和环境因子同时决定植物碳同位素组成。植物稳定碳同位素技术同时还是古气候重建和预测未来环境变化的理论基础。本文综述了光照、温度、水分、二氧化碳、矿质营养、盐分和大气污染物等环境因素对植物稳定碳同位素组成影响方面的研究进展。  相似文献

植物稳定碳同位素技术是近年兴起的一项快速、可靠的技术。利用稳定碳同位素技术可以揭示碳同化的过程的许多方面的信息。13C和12C同位素效应,使它们在进行碳循环时发生稳定碳同位素的分馏。植物光合作用过程中CO2经气孔扩散分差和RUBPCase及PEPCase羧化分馏是造成植物稳定碳同位素比率(R=13C/12C)不同于源CO2中碳同位素比率的主要原因。遗传因素和环境因子同时决定植物碳同位素组成。植物稳定碳同位素技术同时还是古气候重建和预测未来环境变化的理论基础。本文综述了光照、温度、水分、二氧化碳、矿质营养、盐分和大气污染物等环境因素对植物稳定碳同位素组成影响方面的研究进展。  相似文献
