Chromosome behaviour during male meioses has been studied in two bisexual amphimictic populations of two tardigrade species, namely Richtersius coronifer and Macrobiotus richtersi (Eutardigrada, Macrobiotidae). Both bisexual populations exhibit a diploid chromosome number 2n=12 and no sex chromosomes were identified. DAPI staining and C-banding data indicate that all chromosomes of the bisexual population of R. coronifer are acrocentric. In both species, at male meiotic prophase, all six bivalent homologous chromosomes are aligned side by side along their length and show no evidence of chiasmata. However, in the oocytes of both species a chiasma is generally present in each bivalent at diplotene stage. Lack of recombination is previously unknown in tardigrades, but is a well known phenomenon in many other metazoans where it is always restricted to the heterogametic sex. In tardigrades there is no evidence of heterochromosomes, but it does not mean that in tardigrades, the heterogametic sex does not exist. The adaptive and evolutionary significance of achiasmatic meiosis is discussed.  相似文献

Coexisting bisexual and unisexual populations of individuals belonging to the genus Heterocypris are found in ephemeral freshwater ponds on the island of Lampedusa (Pelagie Islands, Italy). Different reproductive modes were associated with a peculiar morphological trait: a lamella hyalina on the posterior margin of the left valve was observed in amphimictic females, a feature missing in apomictic females. In order to clarify the phylogenetic relationships among taxa with different morphological traits and reproductive modes, we used four polymorphic enzyme loci (GPI, ICD2, MPI and PGM) and mitochondrial DNA 16S ribosomal sequences. We identified three main evolutionary units that showed a combination of morphological and reproductive characteristics: (1) amphimictic females of H. barbara with a lamella hyalina, according to the typical feature of the species, and apomictic females of H. barbara without lamella that are sympatric in one temporary pond; (2) apomictic females of H. incongruens without a lamella, as typical of the species; (3) apomictic females without a lamella, living in sympatry with H. barbara, but characterised by a high genetic diversity from both H. incongruens and H. barbara. We discuss the possible origin of apomictic lineages as a result of independent transition episodes to apomixis from different sexual ancestors. Time of divergence reflected the genetic differentiation within and among multiple ancestors and different possible routes to parthenogenesis.  相似文献

The little fire ant, Wasmannia auropunctata, displays a peculiar breeding system polymorphism. Classical haplo-diploid sexual reproduction between reproductive individuals occurs in some populations, whereas, in others, queens and males reproduce clonally. Workers are produced sexually and are sterile in both clonal and sexual populations. The evolutionary fate of the clonal lineages depends strongly on the underlying mechanisms allowing reproductive individuals to transmit their genomes to subsequent generations. We used several queen-offspring data sets to estimate the rate of transition from heterozygosity to homozygosity associated with recombination events at 33 microsatellite loci in thelytokous parthenogenetic queen lineages and compared these rates with theoretical expectations under various parthenogenesis mechanisms. We then used sexually produced worker families to define linkage groups for these 33 loci and to compare meiotic recombination rates in sexual and parthenogenetic queens. Our results demonstrate that queens from clonal populations reproduce by automictic parthenogenesis with central fusion. These same parthenogenetic queens produce normally segregating meiotic oocytes for workers, which display much lower rates of recombination (by a factor of 45) than workers produced by sexual queens. These low recombination rates also concern the parthenogenetic production of queen offspring, as indicated by the very low rates of transition from heterozygosity to homozygosity observed (from 0% to 2.8%). We suggest that the combination of automixis with central fusion and a major decrease in recombination rates allows clonal queens to benefit from thelytoky while avoiding the potential inbreeding depression resulting from the loss of heterozygosity during automixis. In sterile workers, the strong decrease of recombination rates may also facilitate the conservation over time of some coadapted allelic interactions within chromosomes that might confer an adaptive advantage in habitats disturbed by human activity, where clonal populations of W. auropunctata are mostly found.  相似文献

Cyclical parthenogens, including aphids, are attractive models for comparing the genetic outcomes of sexual and asexual reproduction, which determine their respective evolutionary advantages. In this study, we examined how reproductive mode shapes genetic structure of sexual (cyclically parthenogenetic) and asexual (obligately parthenogenetic) populations of the aphid Rhopalosiphum padi by comparing microsatellite and allozyme data sets. Allozymes showed little polymorphism, confirming earlier studies with these markers. In contrast, microsatellite loci were highly polymorphic and showed patterns very discordant from allozyme loci. In particular, microsatellites revealed strong heterozygote excess in asexual populations, whereas allozymes showed heterozygote deficits. Various hypotheses are explored that could account for the conflicting results of these two types of genetic markers. A strong differentiation between reproductive modes was found with both types of markers. Microsatellites indicated that sexual populations have high allelic polymorphism and heterozygote deficits (possibly because of population subdivision, inbreeding or selection). Little geographical differentiation was found among sexual populations confirming the large dispersal ability of this aphid. In contrast, asexual populations showed less allelic polymorphism but high heterozygosity at most loci. Two alternative hypotheses are proposed to explain this heterozygosity excess: allele sequence divergence during long-term asexuality or hybrid origin of asexual lineages. Clonal diversity of asexual lineages of R. padi was substantial suggesting that they could have frozen genetic diversity from the pool of sexual lineages. Several widespread asexual genotypes were found to persist through time, as already seen in other aphid species, a feature seemingly consistent with the general-purpose genotype hypothesis.  相似文献

A Kamuela, Hawaii, population of Drosophila mercatorum was surveyed for enzyme variability. The mean heterozygosity and the proportion of polymorphic loci were estimated as 0.1255 and 0.37, respectively. Neither deviates more than one standard error from their respective means for 43 Drosophila species (Nevo 1978). Heterozygosity was distributed across enzyme categories in much the same manner as observed in other species (Gillespie and Kojima 1968; Johnson 1974), and enzymes associated with glycolysis were about as variable as other enzymes of central metabolism.--The levels of heterozygosity and polymorphism in this population do not seem to have been affected by a low-level capacity for parthenogenesis. The observed parthenogenetic reproduction is not strongly associated with particular allelic variants among viable parthenogenetic adults; however, the capacity to establish a self-sustaining parthenogenetic clone is strongly associated with the phenotype with the most frequent allele at every locus studied. We interpret these results to mean that isozyme variants do not strongly influence viability under total homozygosity (the genetic condition imposed by parthenogenesis), but they do have an impact upon the reproductive biology of parthenogenetic adults.  相似文献

Several hundred starlings (Sturnus vulgaris) were introduced to New Zealand from Great Britain during1860–1880. Allozymic variation at 24 loci was analysed in winter populations sampled at six localities in each country. New Zealand samples had fewer alleles per locus but the same mean heterozygosity (3% per locus) and proportion of polymorphic loci as did British samples. Winter populations in Britain contain European migrants and were genetically homogeneous. Paradoxically, genetic distances among derived New Zealand populations, and between New Zealand and Great Britain were much greater, similar in magnitude to those observed among allopatric populations in other avian species. The geographical pattern of genetic variation in New Zealand suggests that reproductive isolation of populations and random drift have contributed to the development of population differentiation.  相似文献

This study reports on two parthenogenetic strains of the migratory locust Locusta migratoria. The offspring of thelytokou females had a single fragment per microsatellite loci. All offspring of the parthenogenetic F1 females were genetically identical. These results further confirmed that restitution of the sister products of early cleavage mitoses and cell fusion might be the most likely diploidization mechanisms in the thelytokous locusts. Polymerase chain reaction amplification results demonstrated that thelytoky in the locust was not induced by Wolbachia bacteria. Apart from the low fitness gained in thelytokous females, large populations with migration and losing heterozygosity may be other reasons why regular parthenogenesis has not evolved in the locust.  相似文献

Northeastern Turkey is recognized as one of the most important germplasm centers for the grape in the world. In the present study, simple sequence repeat markers were used to investigate the genetic diversity between four Vitis vinifera cv. Kabarcik populations sampled from the Coruh Valley in Turkey, at altitudes of 800-1,150 m. The mean observed number of alleles per locus varied from 2 (loci VVMD7 and VVMD24) to 6 (VVS2) among populations. The population from the highest altitude showed the greatest average number of alleles, 4.5. With regard to the six loci examined in all populations, the mean observed heterozygosity was higher than the expected heterozygosity. Among the loci, VVS2 (probability of identity = 0.137) was found to be the most informative among populations. Genetic distances between populations ranged from 0.072 to 0.216. Genetic differentiation among populations was strongly related to geographic distances in all populations.  相似文献

Drosophila mercatorum is a sexual species that can reproduce parthenogenetically. Previous studies revealed that parthenogenetic strains had "coadapted genomes" with high fitness under parthenogenesis and total homozygosity due to nonadditive and nonmultiplicative fitness interactions between chromosomal segments scattered throughout the genome. To study the evolutionary origins of such coadapted genomes, females from sexual matings in nature were isolated as virgins and challenged to reproduce parthenogenetically. Fitness studies were performed on genomes derived from these sexual females and upon their successful parthenogenetic progeny. By straddling the reproductive transition from sex to parthenogenesis, these fitness studies demonstrated that coadapted genomes arise immediately, apparently due to an intense selective bottleneck accompanying the reproductive transition, and are not due to the slow accumulation of epistatic complexes via mutation after parthenogenesis has already been established. The reproductive transition may also serve as an experimental model of the "genetic revolution" theory of speciation because the transition involves (1) the ultimate founder effect (one genome), (2) maximal genetic drift and fixation, (3) a drastic change in genetic environment characterized by total homozygosity, and (4) an intense selective bottleneck that interacts with the change in genetic environment and the need to adapt to a laboratory environment and a novel system of reproduction. Thus, all the elements theorized to underlie genetic revolution are present, albeit in extreme form. This study indicates that genetic revolutions are real phenomena that can quickly alter morphology, development, life history parameters and behavior. Indeed, the alterations can be so drastic that a new "species" evolves, complete with pre- and post-mating isolating mechanisms. However, isozyme loci do not appear to be the target of this genetic revolution, but rather loci regulating fundamental developmental processes. However, isozyme loci may be useful in predicting the a priori chance of a successful revolution since they can indicate how the population structure of the parent population influences levels of individual heterozygosity, the prime source of the genetic variability in the founder population that must pass through the selective bottleneck.  相似文献

1. In the heterogonic life cycle of monogonont rotifers, amictic (female‐producing) females develop from two types of eggs: fertilised resting (diapausing) eggs and parthenogenetic subitaneous eggs. Females hatched from resting eggs initiate clonal populations by female parthenogenesis and are called stem females. This study compares females from resting and parthenogenetic eggs that were produced under identical culture conditions and were of similar birth order. 2. Newborn stem females had many more lipid droplets in their tissues than similar‐sized, newborn females from parthenogenetic eggs. When neonates were stained with Nile Red and viewed under epifluorescent illumination, these droplets were shown to be sites of neutral‐lipid storage products. 3. Stem females had no posterolateral spines and short anterior spines, while their mothers and offspring in subsequent, parthenogenetic generations typically had long posterolateral spines and elongated anterior spines. 4. Newborn stem females survived starvation significantly longer than newborn females from parthenogenetic eggs. 5. When females from resting and parthenogenetic eggs were cultured from birth to death at a high food concentration, the reproductive potential (r day?1) of the stem females was significantly higher (0.82–0.88 versus 0.70), primarily because of egg production at an earlier age. The mean lifetime fecundity (Ro) of stem females was significantly greater than that of females from parthenogenetic eggs. 6. Extensive lipid reserves should increase the ability of stem females to colonise new habitats. Firstly, compared with females from parthenogenetic eggs, stem females are more likely to experience starvation or food limitation. Resting eggs hatch in response to physical and chemical factors that are not directly related to food availability, and from sediments that may be far from food‐rich surface waters. Secondly, when food is abundant, stem females have a greater reproductive potential.  相似文献

Nine polymorphic out of 13 isoenzymes (Gdh-A, Lap-A, Mdh-B, Mdh-C, Ndh-A, 6Pgd-B, Pgi-B, Pgm-A and Pgm-B) were used to assess genetic diversity in seven natural Cupressus sempervirens populations in South-western Anatolia. Except for Pgi-B, all loci were in Hardy–Weinberg equilibrium. A high level of genetic diversity was observed within populations. The mean number of alleles was 2.3±0.08, the effective number of alleles was 1.53±0.10, the percentage of polymorphic loci was 82.6±8.7%, expected heterozygosity was 0.350±0.039, observed heterozygosity was 0.326±0.039, and the fixation index was 0.049. Interpopulation differentiation was moderate, with differentiation indices of GST=0.073 and δ=0.104. A UPGMA tree and differentiation (Dj) showed that Kumluca, Kemer and Kas were the most differentiated populations. Pgm-B was one of the loci contributing most to interpopulation differentiation (GST=0.119; Di=0.201), clearly distinguishing Eastern populations, as indicated by a highly significant correlation coefficient between Dj and longitude (R2=0.88**). No other significant correlation was found between genetic measures and geographical traits for the Turkish cypress populations. A metanalysis of these and other cypress populations from Crete and the Aegean islands, that had been characterised previously using the same isoenzymes, showed no significant relationship between altitude and genetic measures (within-population differentiation or δT, observed heterozygosity, fixation index and Dj). Observed heterozygosity and latitude were negatively correlated. Longitude was found to be correlated with heterozygosity, fixation index and differentiation. Genetic and geographic distances were positively correlated over loci. Deviations from selective neutrality by Ewens–Watterson test were observed for Pgm-B, Ndh-A and 6Pgd-B.  相似文献

The citrine forktail, Ischnura hastata, is an American damselfly species, widely distributed, with only‐female populations also found at the Azores islands. Here we report the development of nine microsatellite loci for this species. The number of alleles per locus ranged from six to 11, with an observed heterozygosity ranging from 0.245 to 0.737. Eight of the nine loci successfully amplified in a sample of parthenogenetic females from the Azores. The developed microsatellite system will be a useful tool to investigate population structure, as well as the number of clones, the type of parthenogenesis and the origin of the parthenogenetic populations of this species.  相似文献

Heterozygosity at nine genetic loci (PI, TF, PGM1, ACP1, HP, GC, GLO1, C3, and ESD) was analyzed in pulmonary tuberculosis patients with good (group 1, N = 71) and poor (group 2, N = 35) response to treatment. The observed heterozygosities were compared with the expected values, which were calculated from allele frequencies in a control sample of healthy individuals (N = 328 with all but one locus and 78 with ESD) according to Hardy-Weinberg expectations. The analysis showed that the observed heterozygosities gl of patients significantly differed from the expected values hl in the case of four loci (GC, PI, C3, and ACP1). The observed heterozygosity was higher than expected in three cases (PI, C3, and ACP1) and lower then expected (GC) in one case. When data on each individual locus were compared using Fisher's exact test, both groups of patients proved to significantly differ (PF < 0.05) from the control group in the same four loci. No difference in observed heterozygosity was detected between the two groups of patients. The mean expected heterozygosity was h = 0.386 +/- 0.00674; the mean observed heterozygosity was g = 0.415 +/- 0.02 in group 1, g = 0.402 +/- 0.026 in group 2, and g = 0.371 +/- 0.00955 in the control group. The t test did not reveal a significant difference between the mean values of expected observed heterozygosities. Heterozygosity at individual loci, rather than mean heterozygosity, was proposed as an integral nonspecific indicator of the genetic control of a disease, because the former directly implicates individual marker loci in the development of a disorder, whereas effects of individual loci may eliminate each other when mean heterozygosity is computed. Based on the results obtained, a genetic control was assumed for the development of the tuberculosis process in the lungs.  相似文献

Twenty enzyme loci were studied in 44 Illinois populations of Oenothera biennis; four were polymorphic. Most of the variation was distributed between populations. Fifty-nine percent of the populations had one genotype, 27% two genotypes and the remaining 16% from three to five genotypes; the average was 1.50. The proportion of genetic diversity present in single populations is.38 of that present in the state. Members of single populations were uniformly heterozygous for 1 to 4 loci. The mean heterozygosity per population ranged from 0 to 20%. For Illinois populations collectively, heterozygosity averaged 4.5%. There was much gene frequency heterogeneity between populations. The true standardized genetic variance among populations for alleles at polymorphic loci varied from.40 to.78. Populations from Cook County were much more similar inter se than those downstate, had fewer genotypes and polymorphic loci, and had less heterozygosity than downstate populations. The mean normalized genetic identity among Cook County populations was.987 versus.947 for downstate populations. The mean number of genotypes per population in Cook County was 1.06 versus 2.40 in downstate populations. There was only one polymorphic locus in Cook County, VLP. The genetic structure of Oe. biennis suggests that single populations are colonized by one, or at best a few individuals. Cook County populations are judged to be less variable than downstate populations because the mean age of the populations probably is less than that of those downstate.  相似文献

The genetic population structure of Polyommatus coridon (Poda 1761) over large regions of France, Italy and Germany was studied by allozyme electrophoresis. The genetic diversity within populations was high for all parameters analysed (number of alleles 2.72; observed and expected heterozygosity 19.6% and 20.3%, respectively; percentage of polymorphic loci: total: 76.4% and, with polymorphism if the frequency of the commonest allele is below 95%: 53.1%), whereas genetic differentiation between populations was comparatively low (FST = 0.021 +/- 0.002). The mean number of alleles declined significantly from southern to northern populations (r = -0.53, P = 0.0005). Similar effects were found also for other parameters of genetic diversity. This is interpreted as a loss of genetic diversity during postglacial expansion. However, samples from France and Italy had similar patterns of genetic diversity indicating no significant loss in this region. Populations from southern Germany were genetically uniform, well differentiated from French populations and showed a significant loss of genetic diversity. Probably, this is due to a bottleneck during passing through the Burgundian Gap, which is a migration corridor from north-eastern France to southern Germany. In contrast to southern German populations, western German populations were not well differentiated from French populations. Nevertheless, they were genetically impoverished, probably as a result from local bottlenecks and post-expansion phenomena.  相似文献

Six polymorphic microsatellites (eight loci) were used to study the genetic diversity and population structure of common carp from Dongting Lake (DTC), Poyang Lake (PYC), and the Yangtze River (YZC) in China. The gene diversity was high among populations with values close to 1. The number of alleles per locus ranged from 2 to 11, and the average number of alleles among 3 populations ranged from 6.5 to 7.9. The mean observed (H O) and expected (H E) heterozygosity ranged from 0.4888 to 0.5162 and from 0.7679 to 0.7708, respectively. Significant deviations from Hardy–Weinberg Equilibrium expectation were found at majority of the loci and in all three populations in which heterozygote deficits were apparent. The analysis of molecular variance (AMOVA) indicated that the percent of variance among populations and within populations were 3.03 and 96.97, respectively. The Fst values between populations indicated that there were significant genetic differentiations for the common carp populations from the Yangtze River and two largest Chinese freshwater lakes. The factors that may result in genetic divergence and significant reduction of the observed heterozygosity were discussed.  相似文献

In four isolated populations of parthenogenetic Caucasian rock lizard Lacerta unisexualis, variability of (TCC)n loci was examined using multilocus DNA fingerprinting. Unexpectedly high variability of (TCC)n microsatellites was found in all four populations. The mean similarity index was 0.825, which is higher than similarity estimates obtained for other mini- and microsatellite loci in L. unisexualis and parthenogenetic species L. dahli and L. armeniaca studied earlier. The high variation level of (TCC)n loci was shown to be at least partially associated with the presence of a diverged (TCC)n sequence fraction in the L. unisexualis genome. Mutations at some other genetically unstable (TCC)n loci may cause their structural diversity in populations of L. unisexualis.  相似文献

New Zealand populations of Gracilaria chilensis are uniform in anatomical reproductive characteristics but vary morphologically and have been found to separate into two distinct groups with respect to agar methylation level, namely low (24–30%) and high (43–47%). To investigate the genetic variation within New Zealand populations of this species, 14 isozyme loci detected by starch-gel electrophoresis were examined in 17 wild populations from a wide range of localities, and in cultures derived from these populations. Five of these loci were polymorphic, but the genetic variation within populations was low: of the 17 populations examined, 15 were fixed at all loci (heterozygosity 0.000) and in the remaining two populations the observed heterozygosity was still low (0.004 and 0.011). The genetic distances between the populations ranged from 0.00 to 0.43. UPGMA cluster analysis separated the populations into two groups, a northern group and a group found throughout the country. Although these two groups do not correlate with the two groups based on agar methylation level at every locality, the correlation is sufficiently striking to merit further investigation.  相似文献

Changium smyrnioides Wolff is an endangered species of the monotypic genus Changium (Umbelliferae). For a better understanding of its genetic diversity, four populations of this species were cytologically and allozymically investigated. The karyotypic analysis of C. smyrnioides indicated that the chromosomal number was stable (2n = 20) in all populations. Karyotypes of the populations were all “2A” type. Karyotypical polymorphism was shown as the chromosomal heterozygosity, and position shift of the satellite. Genetic diversity of 4 populations was analysed with starch gel electrophoresis. A low level of allozymic diversity was estimated based on 19 loci of 10 enzyme systems. Genetic diversity within 4 populations was relatively low with the proportion of polymorphic loci (P) ranging from 19% to 42.9%, average number of alleles per locus (A) from 1.2 to l. 7, mean expected heterozygosity ( He ) 0. 073 and mean observed heterozygosity ( Ho ) of O.030. In general, about 58% of genetic variation were attributed to the differentiation among populations, while the remaining 42% resided within populations. The genetic diversity of the eastern populations was higher than that of the southwestern populations. On the basis of this studies, we thought that the habitat of C. smyrnioides should be protected and most of the populations should be sampled soas to retain as much genetic diversity as possible in ex-situ conversation.  相似文献
