The molecular biology of self-incompatibility systems in flowering plants   总被引:5,自引:0,他引:5  
Self-incompatibility is a common mechanism by which flowering plants can exert some control over the process of fertilization. Typically, the self-incompatibility response involves the recognition and rejection of self-incompatible pollen which leads to a block in self-fertilization and, as a consequence, promotes outcrossing. In recent years, considerable progress has been made in the molecular understanding of several self-incompatibility systems. Interestingly, a common mechanism for self-incompatibility is not employed by all flowering plants, but in fact quite diverse mechanisms have been recruited for the rejection of self-incompatible pollen. In this review, the recent advances in the self-incompatibility systems of the Solanaceae, Papaveraceae, and Brassicaceae will be described as well as some of the molecular work that is emerging for the Poaceae and the heteromorphic self-incompatibility systems.  相似文献

Solanaceous plants with gametophytic self-incompatibility produce ribonucleases in the transmitting tract of the style that interact with self-pollen and inhibit its growth. These ribonucleases are a series of allelic products of the S-locus, which controls self-incompatibility. Little is known about the pollen components involved in this interaction or whether a signal transduction pathway is activated during the self-incompatibility response. We have partially purified a soluble protein kinase from pollen tubes of Nicotiana alata that phosphorylates the self-incompatibility RNases (S-RNases) from N. alata but not Lycopersicon peruvianum. The soluble protein kinase (Nak-1) has several features shared by the calcium-dependent protein kinase (CDPK) class of plant protein kinases, including substrate specificity, calcium dependence, inhibition by the calmodulin antagonist calmidazolium, and cross-reaction with monoclonal antibodies raised to a CDPK from soybean. Phosphorylation of S 2-RNase by Nak-1 is restricted to serine residues, but the site(s) of phosphorylation has not been determined and there is no evidence for allele-specific phosphorylation. The microsomal fraction from pollen tubes also phosphorylates S-RNases and this activity may be associated with proteins of Mr60 K and 69 K that cross-react with the monoclonal antibody to the soybean CDPK. These results are discussed in the context of the involvement of phosphorylation in other self-incompatibility systems.  相似文献

The structural events accompanying the maturation of the pistil of Petunia hybrida have been studied in detail, together with the changes in the protein spectrum of the transmitting tissue that occur over this period. These events have been considered in terms of the acquisition of the self-incompatibility response, which occurs while the pistil is enclosed in the bud. Apart from several minor differences, the young pistils differ only from the mature in that their transmitting tissue cells fail to respond to pollination by undergoing characteristic ultrastructural changes. Data from electrofocusing indicates that several proteins, mobilised in the mature transmitting tissue some three hours after pollination, are absent from bud pistils. It is proposed that the pollination-stimulated release of certain polypeptides, believed to be involved in the self-incompatibility response, does not take place in young pistils. These observations are considered with reference to currently-accepted models of the operation of the self-incompatibility mechanism in Petunia.  相似文献

In this study, the tissue printing technique has been used to rapidly localize in female tissues the presence of specific mRNA representing the products (or some of the products) of the self-incompatibility S-locus gene(s). The methodology, initially developed for Brassica oleracea (sporophytic self-incompatibility) has been successfully employed on Solanum chacoense (gametophytic self-incompatibility). In the Brassica system tissue printing has allowed rapid discrimination between S alleles belonging to class 1 (dominant types) vs. class 2 (recessive types), and thus parallels findings obtained by restriction analyses. In the Solanum system the level of the S-RNase messages was analysed by scanning laser densitometry, and it was found that the message levels of the allele S14 declined faster than those coming from S13 in mature flowers.  相似文献

The self-incompatibility reaction of cruciferous plants prevents self-fertilization and has been shown to be controlled by at least two genes situated at a single multiallelic locus, theS locus. One of these two genes, theS locus glycoprotein (SLG) gene, encodes an abundant glycoprotein secreted to the cell wall of stigma papillae. Identification of thoseS alleles present at theS locus is of prime interest when studying the self-incompatibility response and can be achieved by identifying the SLG of the stigma. Here, we show that using anti-SLG antibodies in an immunochemical analysis, combined with a SSCP (single-strand conformation polymorphism) approach to characterize the corresponding stigma-specific, SLG mRNA, allowed the identification of plants heterogeneous at theS locus among populations of plants that were thought to be homozygous for known SLG alleles. This analysis stresses the importance of testing the homozygosity at theS locus of lines considered inbred for a knownS allele as mix-up of seeds may occur during the breeding programme.  相似文献

Summary It has been claimed that Borage (Borago officInalis L.) has a multifactorial self-incompatibility system. Such systems may have a high level of ineffective pollination, and we show that this is the case in borage. The ranking of seed set from highest to lowest is as follows: bee-pollination; natural pollination in the absence of bees; artificial cross-pollination between unrelated plants; artificial cross-pollination between related plants; artificial self-pollination. In diallel crosses, significant parental effects were detected but no consistent patterns of seed set, which suggest a simple self-incompatibility system, were detected. The level of outcrossing with natural pollination was very variable but greater than 50%. Thus, there appears to be no straightforward self-incompatibility system in borage.  相似文献

Summary Genetic and molecular analysis of the self-incompatibility locus (S-locus) of the crucifer Brassica has led to the characterization of a multigene family involved in pollen-stigma interactions. While the crucifer Arabidopsis thaliana does not have a self-incompatibility system, S-related sequences were detected in this species by cross-hybridization with Brassica DNA probes. In this paper, we show that an A. thaliana S-related sequence, designated AtS1, is expressed specifically in flower buds. Sequence analysis suggests that AtS1 encodes a secreted glycoprotein that is most similar to the Brassica S-locus related protein SLR1. As has been proposed for SLR1, this gene may be involved in determining some fundamental aspect of pollen-stigma interactions during pollination. The molecular and genetic advantages of the Arabidopsis system will provide many avenues for testing this hypothesis.  相似文献

De-novo synthesis of the S-allele-specific glycoproteins of Brassica oleracea is demonstrated in stigmas at different developmental stages. Excised stigmas incorporate 14C-labeled amino acids into their S-glycoproteins early in development and before the self-incompatibility response is acquired, but the rate of synthesis accelerates prior to anthesis, resulting in the accumulation of high levels of the S-glycoproteins in the stigma and coinciding with the acquisition of the pollen-stigma incompatibility response. Since the self-compatible and self-incompatible zones of developing inflorescences are very sharply delineated, a threshold quantity of S-glycoproteins appears to be critical for the onset of self-incompatibility. Cross-compatibility behaviour of doubled haploid (DH) and hybrid genotypes of Coffea camphora was established using both phenotypic bioassay and in situ seed-set examination. The availability of DHs provided the opportunity of working with genetically homogenous pollen and female parents. The aniline blue fluorescence (ABF) method was applied to detect callose accumulation in pollen and pistil. Clear cross-compatibility/incompatibility situations were observed and confirmed by in situ seed-set analysis. Cross-compatibility analysis of hybrid combinations involving different DHs corroborated the crossing behaviour observed at the DH level. Expression of the self-incompatibility system did not appear to be affected by the low vigour of the DH. The crossing-behaviour distribution observed within DHs derived from clone IF200 confirmed that self-incompatibility in C. canephora is a gametophytic self-incompatibility system controlled by a single locus (S-locus). Reduced seed-set developments following incompatible crosses may indicate the occurrence of pseudo-incompatibility. Molecular marker linkage analysis showed that the S-locus is associated with an RFLP marker on linkage group 9. The availability of a linked DNA marker should facilitate the genetic analysis of self-incompatibility in relation to coffee breeding programmes.  相似文献

Quantitative studies of the adhesion of pollen grains to the stigma in Brassica oleracea revealed that self-pollen is initially less firmly bound than cross-pollen. The pollen grain tryphine, believed to be important in the adhesion process, has been shown to differ in mobility following self- and cross-pollination when observed using fluorescent probes. The hydration of the pollen grains has been investigated in vitro by measuring the changes in shape, volume and fresh weight of the imbibing grains. Whilst little change in volume could be detected there was a considerable increase in fresh weight together with a change of shape. The relative success of fruit from paired self- and cross-pollinations was examined in Phormium tenax when the contrasted pollinations were separated by different distances on the same and different inflorescences. We determined whether the retention of selfed fruits differed from that of crossed fruits and whether it depended on the level of competition with crossed fruit, the number of seeds per fruit, and/or the presence of earlier developing fruit. We found that the success of selfed fruits is determined by the degree of competition with crossed fruits and may be an expression of self-incompatibility. Competition-dependence of the abscission of selfed flowers has not been documented previously. It is parallel to cryptic self-incompatibility in which individual self-pollen grains are not as successful as cross-pollen when competing on the same pistil. The competition-dependent abscission of self-pollinations considered here, however, operates at the level of whole flowers. The phenomenon of competition-dependent abscission of selfed flowers in P. tenax also has implications for the measurement and interpretation of self-incompatibility in other species. Self-incompatibility is a quantitative phenomenon. The facultative success of selfing shows that the effective strength of self-incompatibility can be highly susceptible to the conditions of competition under which it is measured. The competition-dependent abscission of selfed flowers allows a high level of outcrossing to be achieved while it assures seed set when pollinations are scarce. Several other causes of intermediate selfing frequencies can also be explained by this “best-of-both-worlds” hypothesis.  相似文献

Summary We have identified three alleles of the S-locus controlling self-incompatibility and their associated pistil proteins in Petunia inflata, a species that displays monofactorial gametophytic self-incompatibility. These S-allele-associated proteins (S-proteins) are pistil specific, and their levels are developmentally regulated. The amino-terminal sequences determined for the three S-proteins are highly conserved and show considerable homology to those of S-proteins from Petunia hybrida, Nicotiana alata and Lycopersicon peruvianum, three other species of the Solanaceae that also exhibit gametophytic self-incompatibility. cDNA clones encoding the three S-proteins were isolated and sequenced. Comparison of their deduced amino acid sequences reveals an average homology of 75.6%, with conserved and variable residue interspersed throughout the protein. Of the 137 conserved residues, 53 are also conserved in the N. alata S-proteins studies so far; of the 64 variable residues, 29 were identified as hypervariable based on calculation of the Similarity Index. There is only one hypervariable region of significant length, and it consists of eight consecutive hypervariable residues. This region correspond approximately to the hypervariable region HV2 identified in N. alata S-proteins. Of the two classes of N. alata S-proteins previously identified, one class exhibits greater homology to the three P. inflata S-proteins reported here than to the other class of N. alata S-proteins.  相似文献

Phylogenetic reconstruction based on sequence variation in the internal transcribed spacer (ITS) region of rDNA was used to investigate the evolutionary dynamics of homomorphic self-incompatibility in Linanthus section Leptosiphon (Polemoniaceae), a group of annual plant species. Hand-pollination experiments revealed that five species were self-incompatible and four were self-compatible. Optimization of breeding systems onto the tree resulting from maximum-likelihood analysis, with no assumptions made about the ancestral condition, indicated that self-incompatibility has been lost four times in this section. An alternative tree rearrangement conforming to the hypothesis of three losses of self-incompatibility did not have a significantly lower likelihood than the maximum-likelihood tree as determined by a paired-sites test, but all rearrangements resulting in fewer than three losses were statistically rejected. Linanthus bicolor, a selfing species, was found to be polyphyletic, with populations from different geographic regions occurring in three well-supported clades. Morphological similarity in these distinct lineages is likely to have resulted from convergent evolution of traits associated with self-fertilization. Selection for reproductive assurance is hypothesized to have played an important role in the recurrent transformations from self-incompatibility to selfing in this group of annual species.  相似文献

Self-incompatibility is a genetic mechanism enforcing cross-pollination in plants. Hazelnut (Corylus avellana L.) expresses the sporophytic type of self-incompatibility, for which the molecular genetic basis is characterized only in Brassica. The hypothesis that the hazelnut genome contains homologs of Brassica self-incompatibility genes was tested. The S-locus glycoprotein gene (SLG) and the kinase-encoding domain of the S-receptor kinase (SRK) gene of B. oleracea L. were used to probe blots of genomic DNA from six genotypes of hazelnut. Weak hybridization with the SLG probe was detected for all hazelnut genotypes tested; however, no hybridization was detected with PCR-generated probes corresponding to two conserved regions of the SLG gene. One of these PCR probes included the region of SLG encoding the 11 invariant cysteine residues that are an important structural feature of all S-family genes. The present evidence suggests that hazelnut DNA hybridizing to SLG differs significantly from the Brassica gene, and that the S-genes cloned from Brassica will not be useful for exploring self-incompatibility in hazelnut.  相似文献

Summary A cDNA sequence homologous to the Brassica self-incompatibility locus specific glycoprotein (SLSG) sequence was isolated from stigmas of B. oleracea plants homozygous for the S5 allele. The nucleotide sequence of this cDNA was obtained and compared with the S6 allelic form of the SLSG. A transformation and regeneration system has been developed for Nicotiana alata, a plant which is being intensively studied as a model of gametophytic self-incompatibility. Plantlets can be regenerated efficiently from seedling hypocotyls. Kanamycin-resistant, transformed plants have been obtained by cocultivation of regenerating hypocotyls with Agrobacterium tumefaciens strain LBA4404 containing a binary vector. The transformation frequency was low with <1% of tissue explants regenerating transformed plants. The transformed plants contained from one to three copies of the introduced DNA. In most cases, the kanamycin resistance phenotype was transmitted to the offspring as a normal Mendelian factor. In one unusual case, none of the offspring inherited the kanamycin resistance of the transformed maternal parent. This plant may have been chimeric or the kanamycin resistance gene may have been inactivated.  相似文献

Ovule fertilization and seed set were investigated inCrocus thomasii andCrocus vernus subsp.vernus, after different matings. The results gave evidence for a strict ovarian barrier inC. thomasii, which prevents self-fertilization either in self- or mixed- and double-pollination. InC. vernus the ovarian barrier to selfing is not so effective, but a further incompatibility reaction is expressed after self-fertilization resulting in a complete failure of embryogenesis. The implications of the present observations in respect to the mechanisms of pregamic- and postgamic rejection, indicate that a gametophytic system of self-incompatibility controls the ovule fertilization, and a not yet defined system of self-incompatibility controls embryogenesis.  相似文献

In many flowering plants, self-fertilization is prevented by an intraspecific reproductive barrier known as self-incompatibility (SI), that, in most cases, is controlled by a single multiallelic S locus. So far, the only known S locus product in self-incompatible species from the Solanaceae, Scrophulariaceae and Rosaceae is a class of ribonucleases called S RNases. Molecular and transgenic analyses have shown that S RNases are responsible for pollen rejection by the pistil but have no role in pollen expression of SI, which appears to be mediated by a gene called the pollen self-incompatibility or Sp gene. To identify possible candidates for this gene, we investigated the genomic structure of the S locus in Antirrhinum, a member of the Scrophulariaceae. A novel F-box gene, AhSLF-S 2, encoded by the S 2 allele, with the expected features of the Sp gene was identified. AhSLF-S 2 is located 9 kb downstream of S 2 RNase gene and encodes a polypeptide of 376 amino acids with a conserved F-box domain in its amino-terminal part. Hypothetical genes homologous to AhSLF-S 2 are apparent in the sequenced genomic DNA of Arabidopsis and rice. Together, they define a large gene family, named SLF (S locus F-box) family. AhSLF-S 2 is highly polymorphic and is specifically expressed in tapetum, microspores and pollen grains in an allele-specific manner. The possibility that Sp encodes an F-box protein and the implications of this for the operation of self-incompatibility are discussed.  相似文献

Identification of a S-ribonuclease-binding protein in Petunia hybrida   总被引:6,自引:0,他引:6  
To investigate protein-protein interactions in gametophytic self-incompatibility, we used a yeast two-hybrid assay to identify proteins that could interact with the S-ribonuclease protein. These assays identified a pollen-expressed protein, which we have named PhSBP1, that appears to bind with a high degree of specificity to the Petunia hybrida S-ribonuclease. Although PhSBP1 activates reporter gene expression only when expressed in tandem with a S-RNAse bait protein, binding is not allele-specific. Sequence analysis demonstrated that PhSBP1 contained a C-terminal cysteine-rich region that includes a RING-HC domain. Because many RING-finger domain proteins appear to function as E3 ubiquitin ligases, our results suggest that ubiquitination and protein degradation may play a role in regulating self-incompatibility interactions. Together, these results suggest that PhSBP1 may be a candidate for the recently proposed general inhibitor (RI) of self-incompatibility ribonucleases.  相似文献

The male component of the self-incompatibility response in Brassica has recently been shown to be encoded by the S locus cysteine-rich gene (SCR). SCR is related, at the sequence level, to the pollen coat protein (PCP) gene family whose members encode small, cysteine-rich proteins located in the proteo-lipidic surface layer (tryphine) of Brassica pollen grains. Here we show that the Arabidopsis genome includes two large gene families with homology to SCR and to the PCP gene family, respectively. These genes are poorly predicted by gene-identification algorithms and, with few exceptions, have been missed in previous annotations. Based on sequence comparison and an analysis of the expression patterns of several members of each family, we discuss the possible functions of these genes. In particular, we consider the possibility that SCR-related genes in Arabidopsis may encode ligands for the S gene family of receptor-like kinases in this species.  相似文献
