The bacterial pathogen Salmonella typhimurium resides within phagosomes in host cells and is able to deflect the host immune response. In this issue of Cell, Bader et al. (2005) decipher an elegant mechanism by which the PhoQ sensor kinase of Salmonella is switched on by host cationic antimicrobial peptides, leading to changes in gene expression that enable Salmonella to combat the host immune response.  相似文献

昆虫内共生菌与宿主之间的互作关系已逐渐成为昆虫学的研究热点之一。昆虫内共生菌具有协助宿主营养代谢、 逃避天敌攻击和增强抗药性等功能: 通过协助宿主营养代谢, 提供食物中缺乏的营养物质来弥补食物中营养物质的不足; 分泌抗菌肽、 毒素等物质以增强对外源寄生物等的防御能力, 抑制对宿主的不利影响; 同时, 也可以增强宿主抗逆性, 调控植物生理反应, 抑制植物对宿主的不利影响; 利用对抗逆性基因精确的表达调控来增强宿主抗药性等。因此, 内共生菌介导的宿主生物学性状的改变, 扩大了宿主昆虫的生态位, 成为昆虫生长发育过程中的重要调控因子。目前, 昆虫内共生菌的功能往往是通过研究宿主感染共生菌前后性状的变化而证实。近几年, 转录组学、 蛋白质组学、 基因组学等技术的进步, 促进了内共生菌与宿主昆虫共生机制研究的发展。通过研究内共生菌及其功能基因在昆虫种群动态中的作用, 特别是内共生菌感染对宿主生殖、 存活、 适应环境能力的影响, 将有利于揭示内共生菌与宿主的共生机制, 并最终为开发新的防控技术提供理论依据。本文针对昆虫内共生菌的功能进行了综述, 并对日后的研究方向进行了展望, 提供了研究昆虫内共生菌与宿主互作关系的方法及建议。  相似文献

Microarray analysis of large-scale temporal and tissue-specific plant gene expression changes occurring during a susceptible plant-pathogen interaction revealed different gene expression profile changes in cotton root and hypocotyl tissues. In hypocotyl tissues infected with Fusarium oxysporum f. sp. vasinfectum, increased expression of defense-related genes was observed, whereas few changes in the expression levels of defense-related genes were found in infected root tissues. In infected roots, more plant genes were repressed than were induced, especially at the earlier stages of infection. Although many known cotton defense responses were identified, including induction of pathogenesis-related genes and gossypol biosynthesis genes, potential new defense responses also were identified, such as the biosynthesis of lignans. Many of the stress-related gene responses were common to both tissues. The repression of drought-responsive proteins such as aquaporins in both roots and hypocotyls represents a previously unreported response of a host to pathogen attack that may be specific to vascular wilt diseases. Gene expression results implicated the phytohormones ethylene and auxin in the disease process. Biochemical analysis of hormone level changes supported this observation.  相似文献

Host-pathogen interactions result in protein expression changes within both the host and the pathogen. Here, results from proteomic characterization of host response following exposure to Yersinia pestis, the causative agent of plague, and to two near neighbors, Yersinia pseudotuberculosis and Yersinia enterocolitica, are reported. Human monocyte-like cells were chosen as a model for macrophage immune response to pathogen exposure. Two-dimensional electrophoresis followed by mass spectrometry was used to identify host proteins with differential expression following exposure to these three closely related Yersinia species. This comparative proteomic characterization of host response clearly shows that host protein expression patterns are distinct for the different pathogen exposures, and contributes to further understanding of Y. pestis virulence and host defense mechanisms. This work also lays the foundation for future studies aimed at defining biomarkers for presymptomatic detection of plague.  相似文献

The Gram-negative bacterium Escherichia coli and its close relative Salmonella enterica have made important contributions historically to our understanding of how bacteria control DNA supercoiling and of how supercoiling influences gene expression and vice versa. Now they are contributing again by providing examples where changes in DNA supercoiling affect the expression of virulence traits that are important for infectious disease. Available examples encompass both the earliest stages of pathogen–host interactions and the more intimate relationships in which the bacteria invade and proliferate within host cells. A key insight concerns the link between the physiological state of the bacterium and the activity of DNA gyrase, with downstream effects on the expression of genes with promoters that sense changes in DNA supercoiling. Thus the expression of virulence traits by a pathogen can be interpreted partly as a response to its own changing physiology. Knowledge of the molecular connections between physiology, DNA topology and gene expression offers new opportunities to fight infection.  相似文献

The potential role of a GSK3 protein kinase homolog, cggsk, was examined from Colletotrichum gloeosporioides f.sp. malvae, a fungal pathogen of Malva pusilla. A peak in cggsk expression relative to a constitutively expressed fungal actin gene occurred during host penetration and was followed by much lower expression levels during subsequent biotrophic and necrotrophic growth in host tissue. The peak level of cggsk expression observed during penetration was 21-fold greater than that during necrotrophic growth. Expression of cggsk showed small but reproducible changes during growth in culture; however, the levels were always similar to that during necrotrophic growth in the host. One possible role for cggsk could be to coordinate fungal development during host penetration.  相似文献

Many bacteria respond in a coordinate manner to environmental changes. External stimuli, sensed by receptors, are transduced to regulatory proteins which participate in well defined pathways of gene expression by varying their structure and mode of action. The network of environmental signal transduction is responsible for a fine and continuous communication between the host and the pathogenic bacteria. As a result, the gene expression machinery of the pathogen is modified continuously, in order to establish the optimal conditions for bacterial survival and multiplication.  相似文献
