Summary Attempts were made, through mentor pollen techniques, to overcome self-incompatibility in species belonging to the genera Nicotiana and Oenothera and in a hybrid of Lycopersicum, which are characterized by a gametophytic system of incompatibility. While radiation-killed incompatible pollen did not generate mentor effects in any of the material tested, radiation-killed compatible pollen was found to promote a high level of illegitimate fertilizations by incompatible pollen in N. alata. No evidence was obtained that radiation-killed compatible pollen could induce mentor effects in strictly self-incompatible clones of O. organensis and of the interspecific hybrid L. esculentum x L. peruvianum.This publication is contribution no 1563 from the Biology Radio-protection and Medical Research programme of the Directorate General XII of the Commission of the European Communities  相似文献

Summary Comparative studies on self-pollination after the application of high concentrations of CO2 gas, which is known to overcome the self-incompatibility reaction in Cruciferous species, have revealed significant changes in the pollen tubes of germinating grains prior to their penetration into the papilla cells. A remarkable increase in the width of the pollen tube was induced by treating the stigmatic papillae with high CO2 concentrations. The width of the pollen tube appeared to be greatest with CO2 concentrations ranging from 3% to 5%; these concentrations were also optimal for tube penetration. Callose accumulation was extensively induced in the stigmatic papilla with 10%–20% of CO2, although a typical callosic reaction remained through the ranges appropriate for blocking self-incompatibility. Observations using the scanning electron microscope (SEM) after pollination revealed that compatible pollen tubes in cross-pollinations fused completely to the papular surface during tube penetration, while in self-pollination, pollen tubes remained on the papilla with some additional diffusate. In the case of CO2 treatment for self-pollination, some pollen tubes behaved very similarly to the incompatible or compatible ones already described, while others were different from both of them: they showed a complete fusion, similar to compatible ones, with additional diffusate, similar to incompatible ones. These responses of the pollen and stigma to high CO2 concentrations are discussed with respect to their effect upon the expression of self-incompatibility.  相似文献

The cross B. campestris ssp. japonica x B. oleracea var. botrytis fails due to incompatibility barrier at the stigma. To realize this cross, irradiated compatible pollen (mentor pollen) was used before the incompatible pollination. The presence of mentor pollen stimulated the incompatible pollen to germinate and effect fertilization and seed set. One hybrid was thus obtained. Most of the seeds were inviable. Of the 5 plants raised one was a hybrid and 4 resembled the female parent.  相似文献

Self-incompatibility (SI) is an effective method for limiting self-fertilization in flowering plant species, but there are circumstances in which an otherwise functional SI system may fail. One of the most intriguing of these is the induction of selfing by mixed loads of self and heterospecific pollen (the mentor effect) because it is likely to occur under natural conditions, such as in hybrid zones. Here we conducted a series of controlled crosses to determine whether mentor effects operate in two SI annual species, Helianthus annuus and H. petiolaris, and whether the failure of SI results in a decrease in the frequency of hybridization between these two species. Of the 1396 achenes examined from pollen mixtures that included varying ratios of self, intraspecific compatible, and interspecific pollen, 71 (5.1%) were selfed. Selfing frequencies were significantly less than expected based on pollen ratios, except when the proportion of intraspecific compatible pollen was low. Hybridization frequencies from these same pollen ratios averaged 41.8% with H. annuus as the maternal species and 13.3% with H. petiolaris as the mother. Analysis of 1404 achenes from pollen mixtures that excluded self pollen resulted in hybridization frequencies for H. annuus (42.2%) and H. petiolaris (18.2%) that do not differ significantly from those including self pollen. Thus, mentor effects do not appear to play an important role in reproductive isolation between these species. On the other hand, even a modest increase in self-fertilization in hybrid populations, such as that due to mentor effects, could enhance the probability of hybrid species establishment.  相似文献

Recent studies have shown that certain normally incompatible, interspecific combinations in the genus Populus, and intraspecific combination in Cosmos bipinnatus can be made compatible by mixing irradiation- or chemically-killed compatible pollen (mentor) with normal incompatible pollen at the time of pollination. Knox and associates have proposed that in the sporophytic system of incompatibility the compatible mentor pollen provides the so-called specific recognition substances to the incompatible pollen thereby making the latter compatible. In the present experiments mentor pollen was used in attempts to overcome both intra- and interspecific incompatibility in the genus Nicotiana which has a gametophytic intraspecific incompatibility system. Although not denying that recognition substances specific to pollenpistil incompatibility reactions are present in pollen walls, the results suggest that a role of mentor pollen in overcoming incompatibility may be to provide extra free pollen growth promoting substances (PGS). These substances are apparently able to promote just sufficient further growth to allow some fertilizations by certain pollen tubes with inherently weak expression of incompatibility.The present proposed PGS-Hypothesis also elucidates the physiological basis of the widely-held relationship found between genetic systems of self-incompatibility and sites of pollen inhibition.  相似文献

Common buckwheat (Fagopyrum esculentum Moench) is an agriculturally and pharmaceutically valuable crop due to its wellbalanced essential amino acids and rutin content. However, global mass production of buckwheat is limited because its genetic self-incompatibility results in low seed sets and poor grain yield. Therefore, this study was conducted to classify the modes of pistil-pollen interaction between species belonging to the genusFagopyrum and to determine the optimal combination of outcrosses for the most successful pollinations. Based on the interaction between pistils and pollen, we classified the modes of pollen tube growth during interspecific crosses ofFagopyrum species into five categories: (i) Highly compatible: normal pollen tube elongation and style penetration within 6~24 hours of pollination, (ii) Slightly compatible: delayed (for 1~6 hours) pollen tube elongation and normal style penetration, (iii) Incompatible type I: pollen tube inhibition at the stigma, (iv) Incompatible type II: pollen tube inhibition at the style, and (v) Incompatible type III: pollen tube inhibition at the stylodium. Based on the observed pollen tube elongation and the following embryo development, highly compatible pollinations were found to be crosses betweenF. esculentum x F. cymosum and betweenF. esculentum (thrum)x F. homotropicum.  相似文献

Pollen tube dynamics following different competition regimes were studied in sweet cherry (Prunus avium L.). In the process from pollination to fertilization, a constant reduction in the number of pollen tubes that travel along the style is observed. There could be two main causes of this reduction. One is a physical or physiological constraint consisting of the progressive decrease in the reserves and space available for pollen tube growth along the transmitting tissue of the style, and the other is genetic interaction both among the male gametophytes and between the male gametophytes and the female tissues of the flower. To evaluate the roles that these two forces play in reducing the number of pollen tubes that travel along the style, pistils were subjected to various pollen competition regimes by applying different mixtures of live and dead pollen onto the stigmata. The results obtained were similar when the experiment was repeated with different genotypes over 2 years, both in the laboratory and in the field. The role of stylar constriction is important, but it is not the only cause of pollen tube attrition because with low pollen loads fewer pollen tubes reach the different parts of the style than could fit therein. The fact that under different pollen competition regimes the number of pollen tubes is reduced by the same proportion in each stylar level indicates that genetic interactions play an important role in the control of pollen tube attrition.  相似文献

利用磷酸铅淀淀技术对荞麦(Fagopyrum esculentum Monch.)pin型植株分别进行亲和授粉和不亲和授粉后的柱头、花粉粒、花粉管进行了ATPase的超微细胞化学定位。结果表明(1)亲和授粉和不亲和授粉后0.5h,柱头细胞的ATPase活性反应水平较低或基于无酶活性;柱头表面、柱头上附着的花粉粒内ATPase活性在不亲和授粉时较低,亲和授粉时较高,花粉粒内ATPase主要定位于线粒体和精子细胞;(2)授粉后1.5h,不亲和授粉的柱头细胞及花粉管的ATPase活性均较低,花粉管停止生长,细胞质开始解体;而亲和授粉的柱头细胞及花粉管的ATPase活性均较高,ATPase主要定位于柱头细胞的质膜、胞基质以及花粉管的壁、质体的膜、高尔基体、线粒体上。根据不同时期不同部位ATPase活性的差异,我们认为荞麦发生自交不亲和时,花粉管在花柱中停止生长不仅是因此花粉管得不到花柱中的营养物质而引起的,可能也与花粉管自身物质代谢发生障碍有关。  相似文献

异型花柱是一种受遗传因素控制的花型多态性现象,其适应意义在于提高不同花型间传粉精确性,从而促进异交。为检验二型花柱植物滇丁香(Luculia pinceana)是如何促进异交并确定有效传粉者的类型,实验统计自然种群中长、短柱型的植株数量,测量花部特征,统计自然种群植株柱头上的型间花粉落置的数量和比例,比较型内、型间花粉的萌发率以及花粉管长度,比较不同人工授粉处理结实情况,观察传粉昆虫类型并测量虫体特征。结果表明(1)滇丁香自然居群中长、短柱型植株的数量没有显著性差异。长柱型的筒深、筒直径、开口直径、雄蕊长和柱头长均显著小于短柱型,而长柱型的叶片长和宽、雌蕊长、花药体积、花粉粒体积、柱头厚均显著大于短柱型。(2)自然种群植株柱头上型间花粉所占比例显著低于型内花粉的比例。(3)型间授粉的花粉萌发率以及花粉管长度均大于型内授粉和混合授粉。(4)型间授粉的结实率显著高于型内授粉的结实率,型内授粉具有一定的结实。(5)夜行性长喙条背天蛾(Cechenena lineosa)是滇丁香的有效传粉者,其吻长和滇丁香的花筒深相适应。本研究表明二型花柱植物滇丁香主要是通过与其花部特征相匹配的长喙天蛾实现有效的传粉,通过促进型间花粉的萌发和花粉管的伸长而有利于型间授粉结实,提高传粉精确性。  相似文献

Summary In Primula obconica, a species with a heteromorphic self-incompatibility system, the distinction between compatible and incompatible pollen tubes takes place on the stigma surface in thrum flowers, self tubes growing randomly over the papillar cells. No differences were seen between self and cross tube behaviour on the pin stigma surface, but self tubes were inhibited within the stigmatic tissue with differences in tube length evident after 24 h. The stigma surface bears a proteinaceous pellicle and binds the lectin Concanavalin A. Removal of the stigma removes the incompatibility barrier in mature gynoecia. Bud pollination shows that pollen tubes cannot grow in a normal manner on immature stigmas; the random growth of tubes over the stigma surface resembles that of mature thrum selfs. Fewer compatible tubes reach the style base of young gynoecia and smaller numbers of seeds are set than in mature flowers. Pin and thrum pollen grains germinate and grow in aqueous media, thrum tubes growing longer than pin. The presence of H3BO4 and CaCl2 in the growth medium promotes tube elongation and lengths equivalent to compatible styles can be obtained. The pollen grains have proteinaceous materials in their walls which diffuse out on moistening. Prolonged washing in aqueous media removes these materials but the incompatibility reaction remains unchanged. Thus the incompatibility reaction is between pollen tubes and stigmatic tissue and differs from the homomorphic, sporophytic system where pollen wall proteins elicit the incompatibility response.  相似文献

Compatible pollination of Brassica napus necessitates pollen hydration, pollen germination and growth of the pollen tube through the loosened walls of stigmatic papillar cells, whereas self-incompatible (SI) pollinations fail at one of these stages. Analyses of the early stages of pollination show that at high (but not low) relative humidities, both compatible and SI pollen hydrates, but SI germination is reduced and the rare pollen tubes generally fail to penetrate the papillar walls, although there is some wall loosening. Inside the papillae, both compatible and SI interactions may induce the formation of callose, but there is no evidence for a major accumulation of cytoplasm or secretory vesicles in the vicinity of the pollen tubes and neither microtubule nor F-actin patterns re-arrange in this zone. These observations indicate that the source of the wall-loosening enzymes is probably the pollen tube or pollen coat, and that the common cellular responses of plants to attempted invasions have become suppressed in the papilla–pollen tube interaction.  相似文献

Four independent pollen-stigma binding forces are differentiated after pollen contacts stigmas of Brassica oleracea var. capitata. Identification is based on their different rates and sequence of appearance during gametophyte development; on their differential occurrence after compatible and incompatible pollinations; and on their different stabilities to NaOH and hexane. The first binding force develops most rapidly, begins seconds after pollen-stigma contact, is complete within minutes, occurs on compatible or incompatible papillae, and dissociates in methanol. A second slower binding reaction begins about 15 min after contact, continues for at least 90 min (when pollen tubes emerge), results in a binding structure termed the ocreatine, develops only on compatible papillae, and is dissociated by NaOH. A third attractive mechanism binds the tip of the emerging tube to a cuticle, is detected only on incompatible papillae, and is not dissociated by NaOH or methanol. Ocreatine formation and tube development beyond the emergence stage are prevented by the incompatibility response. A fourth attraction mechanism occurs between the surfaces of papilla and elongating tubes. Reviews of physical and biochemical evidence indicate that van der Waals forces and enzymatically mediated lipid polymerization are alternatives to agglutination as mechanisms for binding male gametophyte to papilla.  相似文献

In Petunia inflata, a species with gametophytic self-incompatibility, pollination triggers two phases of ethylene production by the pistil, the first of which peaks 3 hours after pollination with compatible or incompatible pollen. To investigate the physiological significance of the first phase of ethylene production, pollinated flowers were treated with 2,5-norbornadiene (NBD), an inhibitor of ethylene action. Treatment with NBD reduced pollen tube growth in a dose-dependent manner during the first six hours after pollination; however, pollen tube growth was insensitive to NBD if the treatment was applied 6 hours or more after pollination. Simultaneous application of exogenous ethylene substantially offset the inhibitory effects of NBD in flowers pollinated for 4 hours. Another inhibitor of ethylene action, 1-methylcyclopropene (1-MCP), also produced a strong inhibition of pollen tube growth during the first six hours of pollination. The experiments with 1-MCP pretreatment indicate that pistil tissues are the primary target of the pollination-induced ethylene.  相似文献

Summary Pollen size and pistil length data have been collected for 93 species of Rhododendron (Ericaceae) belonging to a number of different subgeneric taxa. For a sample of eight species in section Vireya, pollen tube growth in the style after selfor interspecific pollination has been quantified. Pollen volume and the time taken for pollen tubes to reach the ovary were both related to pistil length. Pollen-tube growth rates were generally greater for species with longer pistils and larger pollen. Increasing temperature increased the rate of pollen-tube growth. There was no detectable effect of pollen tube density on tube growth rate in the style. After interspecific pollinations tube growth rates in foreign styles could be faster or slower than in self styles. A semisterile individual with two viable pollen grains per tetrad and a plant grafted as scion to a longer-styled stock both showed more rapid pollen-tube growth than expected on the basis of pistil size. Data collected for 26 species in section Vireya showed that where extreme disparity of pollen/pistil size causes failure of interspecific crosses, one or more bridging species with intermediate pollen/pistil size can generally be selected.  相似文献

The reproductive system was studied inCrocus genus (Iridaceae) following intra- and interspecific pollination, by using light and scanning electron microscopy. The results suggest that the stigma-style tract of theCrocus pistil is a mere promoter of pollen tube growth, while intra- and interspecific discrimination of compatible and incompatible pollen occurs in the ovarian tract. Here, the transmitting tissue consists of special epidermal cells, whose granular or floccular secretions provide the selective medium for the growth of pollen tubes. The ovarian self-incompatibility (SI) is widespread within the genus, resulting in a partial or complete suppression of self-fertilization. Moreover, postzygotic SI mechanisms, as well as postzygotic mechanisms of unknown nature, seem to be recurrent and both are responsible for seed abortion. The interspecific ovarian incompatibility concerns only unrelated crosses; crosses between related fertile species succeed both in fertilization and seed-set.  相似文献

Summary. Phenylarsine oxide (PAO) and genistein are two well-known specific inhibitors of tyrosine phosphatases and kinases, respectively, that have been used in the functional analysis of the status of protein phosphotyrosine in different cell types. Our experiments showed that both PAO and genistein arrested pollen germination and pollen tube growth and led to the malformation of the pollen tubes, although genistein had a lesser effect. The malformations of the pollen tubes caused by PAO and genistein were, however, quite different. In addition, it was found that the rate of pollen germination and tube growth recovered to a certain extent when phalloidin was present during PAO treatment, but not when it was present during genistein treatment. Furthermore, PAO treatment also had a great effect on the dynamic organization of filamentous actin in the pollen grain and pollen tube, while genistein only caused reorganization of actin at the turning point of the pollen tube. Our results suggest that reversible protein tyrosine phosphorylation is a crucial step in pollen germination and pollen tube growth, but that tyrosine kinases and phosphatases may have different effects which may function through the reorganization of the actin cytoskeleton. The stylar component to gametophytic self-incompatibility in Solanaceae is an S-RNase. Its primary structure has a characteristic pattern of two hypervariable regions, involved in pollen recognition, and five constant regions. Two of the latter (C2 and C3) constitute the active site, while the highly hydrophobic C1 and C5 are believed to be involved in protein stability. We analyzed the role of the C4 region by site-directed mutagenesis. A GGGG mutant, in which the four charged residues in the C4 region were replaced with glycine, did not accumulate the protein to detectable levels in styles, suggestive of a role in protein stability. A R115G mutant, in which a charged amino acid was eliminated to reduce the potential binding affinity, had no effect on the pollen rejection phenotype. This suggests the C4 does not interact with partners such as potential pollen tube receptors facilitating S-RNase uptake. Finally, a K113R mutant replaced a potential ubiquitination target with arginine. However, this RNase acted as the wild type in both incompatible and compatible crosses. The latter crosses rule out the role of the conserved C4 lysine in ubiquitination.  相似文献

Outcross, self- and mixed pollinations were performed inCrocus vernus subsp.vernus, a species with bicellular pollen, dry stigmas and hollow style. No differences were noted among the above pollinations concerning the germination of pollen and the growth of pollen tubes until the top of ovary. Within 45 min after pollinations 62% of pollen grains germinated. Pollen tubes penetrated the papilla cuticle extending along the papilla wall; on entry into stigmatic lobes they continued growth in the stylar secretion to ovarian locules. Here, however, self-pollen tubes failed to reach or to enter the ovule micropyle; while pollen tubes from either outcross- or mixed pollinations grew until fertilizing ovules. These observations gave evidence of a self-incompatibility system inCrocus, which appeared to be neutralized by mentor effect. The ovary as site of incompatibility response is discussed.  相似文献

采用免疫胶体金定位和电子显微技术研究了离体条件下梨花柱S-RNase对自花、异花花粉管中钙调素(CaM)分布的影响.结果显示:(1)花粉管中CaM主要分布在质膜附近,在细胞壁上也有少量分布.(2)不亲和(自花)花柱S-RNase处理后花粉管远离质膜的位置发现CaM,处理24h后在花粉管中部出现CaM,且CaM位置向花粉管内部移动;而亲和(异花)S-RNase处理后花粉管的CaM分布没有明显变化.研究表明,CaM参与了自交不亲和反应过程.  相似文献
