Thirty one selected bean lines were evaluated in the field for ability to support N2 fixation when intercropped with maize which received 0, 30 and 60 kg N ha–1 as ammonium sulphate. The amount of fixed N2 was estimated using the natural variation of 15N and wheat as the standard non-fixing crop. Nitrogen as low as 15 kg N ha–1 at sowing suppressed nodule weight and activity (acetylene reduction activity) but not nodule number, suggesting that the main effect of mineral N was on nodule development and function. 15N data revealed a high potential of the bean genotypes to fix N2, with the most promising ones averaging between 50–60% of seed N coming from fixation. Bean lines CNF-480, Puebla-152, Mexico-309, Negro Argel, CNF-178, Venezuela-350 and WBR22-3, WBR22-50 and WBR22-55 were ranked as good fixers.  相似文献

Two field experiments were performed to evaluate the nitrogen fixation potential of twenty common bean cultivars and breeding lines during summer and winter seasons of 1986 and 1988, respectively. The 15N isotope dilution method was used to quantify N2 fixation. The cultivars and breeding lines were variable in terms of their N2 fixation. The cv. Caballero was very efficient, with more than 50% N derived from the atmosphere and 60–80 kg N ha–1 fixed in both seasons. Other cultivars were less efficient, since the poorest ones derived less than 30% of their nitrogen from the atmosphere and fixed less than 20 kg N ha–1. After additional testing the best cultivars may be used directly by the farmers for cultivation. The experiments have provided information about which genotypes may be used to breed for enhanced fixation in common bean.  相似文献

An experiment was conducted under greenhouse conditions to evaluate the effect of mineral nitrogen on N2 fixation of two cultivars of Phaseolus vulgaris L., Puebla 152 and Negro Argel. Nitrogen application was 0, 2.5, 12.5 and 25 mg N Kg–1 of a vermiculite-sand-mixture at planting time. Shoot and root growth were elevated by nitrogen application at all growth stages. During vegetative growth (V 5) nodule dry weight and nitrogenase activity (acetylene reducing activity) per plant were reduced by nitrogen supply in both cultivars, but less in Negro Argel than in Puebla 152. At later stages nodulation in nitrogen-treated Puebla 152 did not differ from that in non-treated plants, whereas increased nodule number was found in Negro Argel at high nitrogen levels. The influence of mineral N on the total amount of nitrogen fixed in the two bean cultivars was only slightly different.  相似文献

Common bean,Phaseolus vulgaris L., is known to be ‘inefficient’ in nodulation and N2 fixation although it responds to applied nitrogen. An experiment was conducted to identify and to characterize bean cultivars nodulating in the presence of a high level of nitrogen. Sixteen cultivars and a check for inefficient nodulation, OAC Seaforth, were inoculated and grown for 40 days in replicated pots supplied with zero, 3.5 and 10.5 mM combined nitrogen as NO 3 and NH 4 + . Seven traits relating to nodulation and N2 fixation were all significantly affected by N level (N), cultivar (Cv) and N × Cv interactions (except for root dry weight), indicating that cultivars responded differently to the N treatments. Total dry weight (W) and shoot to root ratio (S/R) increased with the increased N levels. Nodule dry weight (Wn), visual nodulation score (Nv) and nodulation index (Nx) decreased as the N increased. Percent N and N content per plant increased with the increased N level. Plant weight (W) was positively correlated with Wn, Nv and N content and negatively correlated with %N. Nodulation score was positively associated with Wn and plant N content. Genotypes superior in nodulation and N2 fixation in the presence of N were identified. Cultivars Italian Barlotti, California Light Red Kidney, Kentucky Wonder A and Pueblo 152 were selected for further testing and use in improving the nitrate tolerant nodulating characteristic of bean.  相似文献

Nitrogen fixation in nine common bean (Phaseolus vulgaris L.) lines was estimated using the 15N isotope dilution method at two locations in two seasons. In the first season at one location no N2 fixation was detected while in the second season up to 51 kg N ha–1 were estimated. There were significant differences between lines and correlations between trials were significant for the amounts of N2 fixed, but not for total shoot nitrogen. The plants that fixed the most nitrogen nodulated rapidly after germination. Differences in maximum nodule mass, but not specific nodule activity, were detected also.  相似文献

Seed of white bean (Phaseolus vulgaris L. cv. OAC Rico) was treated with 0.04 M ethyl-methane sulphonate. Screening of the M2 progeny from 175 M1 lines in the presence of 1 mM nitrate revealed two nodulation mutants. One line was essentially non-nodulating in several tests, but small white nodules were observed occasionally in other tests with 16 separate Rhizobium strains. The other line formed tiny, creamy white, non-functional (ineffective) and tumor-like pseudo-nodules. The M3 and M4 progenies were fertile and bred true.  相似文献

Summary The uptake and distribution of15NH 4 + ,15NO 3 and15N2 was studied in greenhouse-grown beans (Phaseolus vulgaris L.) with a commercial cultivar and 2 recombinant inbred backcross lines;15N was supplied in the nutrient solution at the R3 (50% bloom) stage. Plants were harvested 1, 5 and 10 days after treatment, and were separated into nodules, roots, stems, mature leaflets, immature leaflets, and flowers/fruits. All 3 lines showed rapid increases in the N content of flowers/fruits after the R3 stage. However, the percentage N in these tissues decreased after the R3 stage. One of the recombinant lines showed a greater uptake of NH 4 + than the other 2 lines. Rates of15N2 fixation and NO 3 uptake were similar for all 3 lines, N2 fixation estimated from total N content showed the 2 recombinant lines with 24 and 34 percent greater activity than the commercial cultivar. Distribution of15N at the whole plant level was similar for all 3 lines for a similar N source.15NO 3 was transported first to leaflets and the label then moved into flowers/fruits. Transport of fixed N2 was from the nodules to roots, stems and into flowers/fruits; usually less than 10 percent entered the leaflets. This indicates that N2 fixation furnishes N directly to flowers/fruits with over 50 percent of the fixed N being deposited into flowers/fruits within 5 days after treatment.  相似文献

The improvement of N2 fixation in legumes may lead to increased yields and reduced fertilizer requirement. Levels of N2 fixation were determined for three cultivars and nine progeny lines from two inbred backcross common bean (Phaseolus vulgaris L.) populations that were grown at Hancock, Wissconsin in 1984 and 1985 using 15N-depleted (NH4)2SO4. The high N2-fixing line Puebla 152 was the donor parent for both inbred backcross populations and the cultivars Porrillo Sintetico and Sanilac were the recurrent parents for populations 21 and 24, respectively. Total N yield, fixed N2 and % N derived from the atmosphere were determined for whole plants and plant parts at the R3 (50% bloom) and R9 (maturity) growth stages. Significant year-by-line interactions were found for N2 fixation traits among the population 21 lines and parents, but not for population 24 lines and their parents. Measures of N2 fixation at R3 were inadequate to predict N2 fixation at R9. Population 24 lines and parents differed for N2 fixation ability at R9, and fixed N2 was correlated with maturity. The recovery of an inbred backcross progeny line, 24-21, which matured earlier and fixed more N2 than the recurrent parent Sanilac indicated that N2 fixation was heritable and that favorable alleles, independent of maturity, were recovered from a late-maturing, high N2-fixing donor parent by utilizing the inbred backcross breeding method. Since most fixed N2 and non-fixed N (>80%) was found in the seeds at maturity, and most lines did not vary for the distribution of nitrogen throughout the plant, selection for improved remobilization of nitrogen to the seed to increase yield is impractical in this genetic material. A field experiment under rainfed conditions was conducted in Durango, México, to assess N2-fixation of three cultivars of common bean (Phaseolus vulgaris L.) using 15N-methodology. In addition, diversity of rhizobial isolates obtained from nodules of the different plant genotypes was evaluated by intrinsic antibiotic resistance (IAR), PCR using enterobacterial repetitive intergenic consensus (ERIC) primers, PCR-RFLP analysis of the 16S rRNA gene and multilocus enzyme electrophoresis (MLEE). Selected isolates were used to determine acetylene reduction and competitive ability under greenhouse conditions. The three cultivars tested did not show high variation in N2-fixation, the %Ndfa values ranged from 19 to 26%. Variability in N2-fixation efficiency among various native rhizobial isolates was very high and our results indicate that differences in competitive abilitiy exist also. PCR-RFLP of the 16S rRNA gene and MLEE revealed that most of the isolates belong to the species Rhizobium etli. Intrinsic antibiotic resistance analysis and ERIC-PCR showed high diversity among isolates. In contrast, our results using MLEE show low genetic diversity (H = 0.105).  相似文献

Common bean (Phaseolus vulgaris L.) is able to fix 20–60 kg N ha–1 under tropical environments in Brazil, but these amounts are inadequate to meet the N requirement for economically attractive seed yields. When the plant is supplemented with N fertilizer, N2 fixation by Rhizobium can be suppressed even at low rates of N. Using the 15N enriched method, two field experiments were conducted to compare the effect of foliar and soil applications of N-urea on N2 fixation traits and seed yield. All treatments received a similar fertilization including 10 kg N ha–1 at sowing. Increasing rates of N (10, 30 and 50 kg N ha–1) were applied for both methods. Foliar application significantly enhanced nodulation, N2 fixation (acetylene reduction activity) and yield at low N level (10 kg N ha–1). Foliar nitrogen was less suppressive to nodulation, even at higher N levels, than soil N treatments. In the site where established Rhizobium was in low numbers, inoculation contributed substantially to increased N2 fixation traits and yield. Both foliar and soil methods inhibited nodulation at high N rates and did not significantly increase bean yield, when comparing low (10 kg N ha–1) and high (50 kg N ha–1) rates applied after emergence. In both experiments, up to 30 kg N ha–1 of biologically fixed N2 were obtained when low rates of N were applied onto the leaves.  相似文献

N2 fixation in lines of Phaseolus vulgaris was measured by 15N-isotope dilution to determine whether a programme of crossing and recurrent selection had resulted in enhanced nitrogen fixation. In field experiments on an isohyperthermic Aquic Hapludoll soil the amount of N2 fixed by the different lines ranged from 18 to 36 kg ha–1 (32 to 47% of plant N) in 56 days. The quantity of N2 fixed and the proportion of plant N derived from fixation was not significantly greater in the lines selected for N2 fixation (RIZ lines) than parental lines. Total shoot N ranged from 53 to 77 kg ha–1 and partitioning of N to pods differed from 28 to 52% among the lines which all had similar growth habit and duration. Nodulation patterns were also distinct. Nodules formed early (10 to 15 plant–1 at 13 days) in many lines, and smallest amounts of fixation were observed in those lines which nodulated slowly and did not form substantial nodule mass until after 40 days. The screening criteria used in the selection of the RIZ lines had been largely indirect with other factors such as disease resistance also being included. Progress for increasing N2 fixation over good-fixing parental lines such as BAT76 was not significant and it is recommended that more attention be paid to early nodulation, to the use of soils with lower available N and to inter-crossing of lines having different good N2 fixation traits in order to further enhance the potential for N2 fixation in beans.  相似文献

Common beans usually achieve grain yields less than the genotypic potential of the cultivar under Romanian field conditions. To understand better the contribution of nitrogen fixation to the yield formation, I made a long-term evaluation (1977–1994) of inoculation effects of 19 Rhizobium leguminosarum bv. phaseoli strains on common bean cultivated in several locations. Grain yields were significantly influenced by all selected strains, years and locations in all experimental cycles, and only partially by the interactions between strains and years or strains and locations. The average yield increases induced by these strains during the four cycles ranged from 6 to 20%. Four bacterial strains proved to be more stable in their field performances, taking into account the yield increases greater than 10% over controls observed in all individual trials. Mean yields and variation limits recorded during the long-term evaluation of strain efficacy in locations with different soil pH values showed similar patterns of yield increases from soils with acidic to neutral pH values. Linear regression between mean grain yields and average temperatures demonstrated the limiting effect of temperature on yield. The interaction between bacterial strain and nitrogen fertiliser rate demonstrated the ability of dinitrogen fixation to satisfy the crop requirements for this element. An evaluation of the amounts of nitrogen fixed in three common bean cultivars inoculated with two bacterial strains showed different N2-fixing capacities among plant genotypes.  相似文献

Field and greenhouse experiments were conducted to assess the nitrogen fixation rates of four cultivars of common bean (Phaseolus vulgaris L.) at different growth stages. The 15N isotope dilution technique was used to quantify biological nitrogen fixation. In the greenhouse, cultivars M4403 and Kallmet accumulated 301 and 189 mg N plant–1, respectively, up to 63 days after planting (DAP) of which 57 and 43% was derived from atmosphere. Under field conditions, cultivars Bayocel and Flor de Mayo RMC accumulated in 77 DAP, 147 and 135 kg N ha–1, respectively, of which approximately one-half was derived from the atmosphere. The rates of N2 fixation determined at different growth stages increased as the plants developed, and reached a maximum during the reproductive stage both under field and greenhouse conditions. Differences in translocation of N were observed between the cultivars tested, particularly under field conditions. Thus, the fixed N harvest index was 93 and 60 for cultivars Flor de Mayo and Bayocel, respectively. In early stages of growth, the total content of ureides in the plants correlated with the N fixation rates. The findings reported in the present paper can be used to build a strategy for enhancing biological N2 fixation in common bean.  相似文献

The efficacy of the alumina system for differentiating between bean (Phaseolus vulgaris L.) genotypes for growth at different levels of phosphorus availability was determinated. In addition to response to P levels, comparisons were made between plants receiving N either from fertilizer or nitrogen fixation. When the cv. Carioca was provided with either 100 ppm of N or inoculated withRhizobium leguminosarum biovarphaseoli, differences in shoot dry weight and nodule number were related to P level. There was a greater proportion of green, ineffectivevs. red, active nodules at the low P concentration than at the higher P concentration. In a second experiment, two cvs., Puebla 152 and Carioca and the breeding line UW 24-21, either were inoculated with rhizobia or provided with 150ppm of N. Each genotype-nitrogen combination was grown at 8 levels of P. There was a positive effect of P level on shoot dry weight, nodule number and nodule mass. Root mass was affected less than nodule or shoot mass by the P level of the growth medium. Nodule mass, but not P concentration in the nodules, was affected by P level, whereas in the other plant tissues, P concentrations were lower at lower P levels in the media.  相似文献

The 15N isotope dilution technique and the N difference method were used to estimate N2 fixation by clover growing in a mixture with ryegrass, in a field experiment and a controlled environment experiment. Values obtained using N difference were approximately 25% lower than those estimated using 15N isotope dilution. In the field experiment there was a measured N benefit to grass growing with clover, equivalent to 42.7 kgN ha-1. The grass in the mixture had a lower atom %15N content and a higher N content than grass in a monoculture; therefore values for N2 fixation were different depending on choice of control plant i.e. monoculture or mixture grass. In the controlled environment experiment there were no significant differences between either the atom %15N contents or the N contents of monoculture grass and grass growing in a mixture with clover. It is concluded that there is a long term indirect transfer of N from clover to associated grass which can lead to errors in estimates of N2 fixation.  相似文献

Before starting a breeding program aimed at improving the nitrogen nutrition ofVicia faba, the authors tried an alternative technique to the acetylene reduction assay, to measure some genetic variability in the plant material. The quantity of dinitrogen fixed by several cultivars ofVicia faba was estimated using a low enrichment15N tracer method and high precision15N mass spectrometry. The fababeans were cultivated for two years in two different soils. The percentage of fixed dinitrogen in the seed varied between genotypes from 40 to 83% of the total nitrogen and was positively correlated with the total seed nitrogen (r=0.64 to 0.86). A highly significant positive correlation was also found between the total seed nitrogen and the quantity of fixed dinitrogen in the seed (r=0.95 to 0.99). The technique used to measure dinitrogen fixation proved to be useful and reliable enough to discriminate between various genotypes, grown over a period of two years in two different soils. However, several non-fixing control plants showed significant differences in their15N enrichment and the problem of choosing a good reference plant was raised and discussed.  相似文献

Summary Homozygous mutant lines of field bean selected for (a) improved yielding potential and (b) altered plant architecture and/or physiological response were tested for symbiotic nitrogen fixing ability under field conditions in comparison with their parent cultivar.15N-tracer techniques were applied to determine %N derived from atmosphere. Data were collected on assimilate and nitrogen accumulation and distribution among various plant parts during two stages of reproductive growth.Symbiotic nitrogen fixation was closely correlated with total plant top biomass and nitrogen yield. A similar close association was found between crop yield and nitrogen harvest index. Both harvest indices tended to be negatively correlated with stage of maturity and with the amount of N derived from air per unit of area. The generally high %N derived from symbiotic N2 fixation and its comparatively small variability implies that this parameter may be difficult to improve inVicia faba under field conditions.It is concluded, that the main genetic potential for improving the amount of biological nitrogen fixation in this crop depends upon factors that promote high photosynthetic productivity and efficient N-use under appropriate agronomic conditions and with effective rhizobial associations. The establishment of rational ideotypes with a possitive impact on yield appears to be of practical significance for increasing the amount of symbiotically fixed nitrogen.  相似文献

The survival of Rhizobium leguminosarum biovar phaseoli on seeds of bean was tested, using the cultivar Carioca. The seeds were treated seven days before inoculation with Benlate, Vitavax, Banrot, Difolatan or Ridomil fungicides. The rhizobial strains used were: CIAT 899, CPAC 1135 and CIAT 652. Strain CIAT 899 showed greater survival on the seed with fungicide than the other strains. Two hours after the contact with fungicides strains CIAT 652 and CPAC 1135 had significantly lower numbers of rhizobia than the treatment without fungicide. The Benlate and Banrot fungicides had the greatest effect on survival of rhizobial strains. There was a drastic mortality of the two strains, CIAT 652 and CPAC 1135, on seeds treated with Benlate and Ridomil. Under field conditions, granular inoculation produced fewer nodules, but a similar total nodule weight as seed inoculation. Serological tests (ELISA) showed that seed treatment with Benlate in connection with seed inoculation reduced drastically the occurrence of inoculated strains in nodules, while the same fungicide treatment and inoculation applied in the seed furrow did not affect the survival of the inoculated strain.  相似文献

Summary The kinetic of15N2 diffusion has been measured in a system similar to that for the estimation of N2 fixation in plant microorganism associations cultivated in soil. The15N2 enrichment of the soil atmosphere reached an homogenous value one hour after injection of15N2 and is identical to that obtained by calculation, indicating that no adsorption occurs in the soil particles.

The combined effect of Vesicular Arbuscular Mycorrhizae (VAM) and Rhizobium on the cold season legumes, lentil and faba bean, as well as on summer legume, soybean, were studied in soils with low indeginous VA mycorrhizal spores. Inoculation of the plant with VA mycorrhizal fungi increased the level of mycorrhizal root infection of lentil, faba bean and soybean. The inoculation with Rhizobium had no significant effect on VA mycorrhizal infection percent, but VA mycorrhizal inoculation increased nodulation of the three legumes. The inoculation with Rhizobium alone significantly increased plant dry weight and N content of lentil and faba bean as well as seed yield of soybean. VA mycorrhizal inoculation also significantly increased plant dry weight and phosphorus content of the plants as did fertilization with superphosphate. Rock phosphate fertilization, however, had no significant effect on plant growth or phosphorus uptake. The addition of rock phosphate in combination with VA mycorrhizal inoculation significantly increased plant dry weight and P uptake of the plants. The dual inoculation with both rhizobia and mycorrhizae induced more significant increases in plant dry weight, N and P content of lentil and faba bean as well as seed yield of soybean than inoculation with either VA mycorrhizae or Rhizobium alone.  相似文献
