An extraction procedure was developed for determining vitamin B12, thiamine, and biotin contents of marine phytoplankton. Phytoplankters were collected either by centrifugation or by retention on a glass fiber filter, then heated at 100 C for I hr in 100 ml of vitamin-free seawater acidified to pH 3.5 with HCl. The extract, after debris removal, was filter-sterilized and analyzed, for vitamin B12, thiamine, and biotin with standard vitamin assay procedures. The vitamin contents of haeodactylum tricornutum, Skeletonema costatum, Stephanopyxis turris, and occolithus liuxleyi were determined during growth in batch cultures. P. tricornutum (non-vitamin requirer) growing in aerated cultures contained 0.29–0.96 ng B12, 5–15 ng thiamine, and 0.45–1.70 ng biotin/mg C. Under similar conditions S. costatum (B12-requirer) contained about 0.06 ng B12, 5–36 ng thiamine, and 0.16–2.10 ng biotin/mg C. The concentrations of vitamin were generally similar during some portion of the growth curve, eg, logarithmic growth. The vitamin B12, content of S. costatum growing under nonaerated conditions decreased when medium B12, was reduced. The biotin content did not change when medium B12 was decreased. The thiamine content per unit weight of C. huxleyi (thiamine-requirer) growing with either 10 or 120 ng/liter thiamine decreased under both medium concentrations, indicating no net synthesis of the vitamin.  相似文献

Bud formation in the moss Pylaisiella selwynii is greatly enhanced by cytokinins at concentrations as low as 10−12m , yet these buds usually fail to develop into normal gametophores. Various ratios at different concentrations of the cytokinin N-6-γ,γ-dimethylallylaminopurine to indoleacetic acid failed to enhance bud initiation over that obtained with cytokinin alone or to permit normal gametophore development. Deletion of the cobaltous ions from the culture medium prevented the appearance of the few gametophores usually formed in the complete medium, but different amounts of cobaltous ion did not significantly enhance initiation of gametophore development. Bud initiation was enhanced 3- to 20-fold by vitamin B12 at 10−5m or by B12 coenzyme at 10−4m , and the time of appearance of these buds was advanced by 6–12 days compared to control plants. At these concentrations of the B12 compounds the buds formed normal gametophores, but at 10−4m vitamin B12 they grew into callus-like masses similar to those obtained with cytokinins. Although the effects of B12 on bud initiation and development mimicked those of cytokinins, except in permitting normal development, no additive or synergistic effects were observed when they were tested together. It is suggested that B12 may play a regulatory role in the control of gametophore initiation and development in mosses.  相似文献

The influence of standing dead biomass on available solar radiation, leaf temperature (Tleaf) and leaf water potential (ѱleaf) of Andropogon gerardii in unburned tallgrass prairie was compared to burned prairie in eastern Kansas. The standing dead reduced photosynthetically active radiation incident on emerging shoots by 58.8% in unburned compared to burned prairie during the initial 30 days of the growing season. Aboveground production in unburned prairie was similarly reduced during this period (55.4%) compared to burned prairie. Leaf temperatures in A. gerardii were greater in unburned prairie than in burned early in the season, but were nearly equal by the end of the growing season. The maximum elevation of Tleaf in unburned prairie above burned was 9.5 C. The maximum unburned Tleaf measured was 41.5 C compared to 39.4 C in burned prairie. Lower windspeed adjacent to leaves in unburned prairie resulting in reduced convective cooling may have caused higher Tleaf in unburned prairie. Leaf water potential was significantly lower in unburned prairie than in burned prairie early in the season but was higher in unburned prairie by late season. The seasonal minimum ѱleaf in burned prairie was — 1.60 MPa compared to —1.45 MPa in unburned prairie. The combined effect of these post-burn differences in solar radiation, Tleaf and ѱleaf may be significant in contributing to the lower production in unburned compared to burned tallgrass prairie.  相似文献

The influence of chronic exposure to UV-B and UV-A radiation on growth and photosynthesis of two polar marine diatoms (Pseudonitzschia seriata and Nitzschia sp.) was investigated in cultures exposed to moderate photon fluences for 3–7 days. Population growth rates were diminished 50% by UV-B. Fluorescence induction kinetics of photo-system II (PSII) revealed that UV-B caused lower Fv/Fm ratios and half-rise times, indicating damage to the reaction center of PSII and to related elements of the photosynthetic electron transport chain. Carbon assimilation rates per cell and per chlorophyll a were nonetheless highest for UV-B—exposed populations, which also had the highest chlorophyll a content per cell. The UV-B—exposed cells were, however, more vulnerable to visible light-induced photoinhibition. Exposure to UV-A in the absence of UV-B had little effect on growth, fluorescence induction of PSII, or chlorophyll a contents but did have some inhibitory effects on carbon assimilation per chlorophyll a and per cell. The increased photosynthetic capacity of UV-B-exposed cells suggested some ability to compensate for damage to the photosynthetic apparatus.  相似文献

Euglena gracilis Klebs, Poterioochromonas malhamensis (Pringsheim) Peterfi, Monochrysis lutheri Droop, Isochrysis galbana Parke and Phaeodactylum tricornutum Bohlin are known to release into the medium a substance which binds free vitamin B12. The binder, apparently a glycoprotein, makes vitamin B12 unavailable and inhibits growth of vitamin B12-requiring microorganisms. Culture filtrates of selected marine diatoms, chrysophytes, cryptophytes, dinoflagellates and green algae contained the binder, indicating that binder release was not restricted to any algal group. However the three prokaryotic bluegreens tested do not produce B12-binder. Production is also independent of the nutritional requirements of the donor, being produced by autotrophs and auxotrophs. The binders produced by these marine species have similar properties: they are heat labile; inhibition is not species-specific; it is competitive, being reversed by adding B12. Production increases with density of the culture and is not restricted to stationary or scenescent cells. The marine species tested produced much less B12 than the freshwater Euglena and Poterioochromonas; the inhibition is reversed by 20–50 ng · 1?1 of B12 for marine species which reach moderate densities and by 150–300 ng · 1?1 B12 for the densely growing species. Consequently the binder may affect the growth of B12-requiring species only in environments like the open ocean poor in B12. By contrast, the marine algae produce other inhibitors which are often heat stable and very inhibitory. These inhibitors, being species-specific and less labile, may be more important in affecting the succession of algal species in natural waters.  相似文献

Department of Botany, Oklahoma State University, Stillwater, OK 74078-3013 USA Preliminary data suggest Nannochloris sp., isolated from the Great Salt Plains National Wildlife Refuge, is a true extremophile. This alga is able to withstand salinities ranging from 0 to 150 ç and temperatures up to 45°C. To test the hypothesis that acclimation to high salinity confers tolerance to high temperature, experimental cultures were acclimated to salinities of 25 and 100 ç and/or temperatures of 23 and 38°C; irradiance (500 mol photons m-2 s-1) was saturating for both growth and photosynthesis. Cells acclimated to low salt and low temperature exhibited high photosynthetic performance in terms of both light-saturated photosynthesis (Pmax; 45.0 fmol O2 cell-1 h-1) and light-harvesting efficiency (0.103 fmol O2 cell-1 h-1/mol photons m-2 s-1). However, high-salinity cells exhibited values for net Pmax (18.1 fmol O2 cell-1 h-1), (0.107 fmol O2 cell-1 h-1/mol photons m-2 s-1) and growth rates (ca. 0.4 d-1) that were equal to, or higher than, those of low-salinity cells when acclimated to high temperature. Both the amount of light required to achieve net photosynthesis (Ic) and that required to achieve light-saturated photosynthesis (Ik) were lower in high-salinity cells than those exhibited by low-salinity cells grown at high temperature; reductions in Ic and Ik were primarily due to increases in light-harvesting efficiency. We propose that an increase in growth temperature might release Nannochloris sp. from energy constraints associated with osmolyte production and low-temperature effects on enzyme activity. These data are consistent with effects of short-term temperature stress on Chl a fluorescence kinetics in this alga.  相似文献

The vitamin B12 requirement of several marine diatoms can be satisfied in B12?limited laboratory cultures by heterotrophic marine bacteria isolated from the same waters and from sediments. The bacteria can utilize diatom excretory products, or the remains of dead diatom cells, in the production of the vitamin. The growth of 12 B121? requiring diatoms (7 genera) in mixed cultures with 14 different bacteria (without added B12) was compared to the growth of those same diatoms in axenic cultures with excess added B12. Diatom growth was generally rapid in the first few days, followed by sustained, slower growth. The diatom yields in mixed cultures ranged from 0.8 to 84% of the yields in axenic cultures with added B12. In a detailed study of one mixed culture, increases in diatom densities were paralleled by increases in cell densities of the bacterium during the first few days of exponential diatom growth. During the period of slow diatom growth, when diatom densities oscillated but steadily increased, the decreases in diatom densities were associated with increased bacterial growth. This suggests that death of a fraction of the B12-limited diatom population releases sufficient organic matter to stimulate growth of the bacteria and their subsequent excretion of B12; this B12 in turn stimulates further growth of the diatoms. Diatom-bacteria interactions leading to the production of B12 may be important in maintaining viable populations of B12-requiring diatoms in nutrient-poor waters during periods between blooms, when conditions are unfavorable for rapid growth.  相似文献

The degree of leaf pubescence development in the arid land shrub Encelia farinosa Gray is affected by air temperature, leaf water potential, and previous history of the apical meristem during the current growing season. Changes in leaf pubescence levels change leaf spectral characteristics and affect both leaf temperature and photosynthesis. Decreasing leaf water potentials and increasing air temperatures both independently increase pubescence development as measured by decreased leaf absorptances. During any one growing season leaf absorptance may change reversibly coincident with air temperature changes, but with respect to water stress leaf absorptance only decreases as the season progresses. The ecological significance of regulation of the leaf spectral characteristics is discussed.  相似文献

目的探讨不同抗原修复方法对肝癌组织中内源性生物素的影响以及消除办法.方法采用pH6.0、pH8.0及pH9.0不同pH值的抗原修复液进行修复,对30例经甲醛固定石蜡包埋的肝细胞癌组织中内源性生物素活性进行检测,并通过不同检测方法对30例肝细胞癌中AFP(Alpha Fetoprotein)染色结果进行对比.结果加热抗原修复暴露肝细胞癌组织中内源性生物素的活性.不同pH值抗原修复液对内源性生物素的活性影响各不相同,阳性强度随着pH值增高而增强.生物素阻断剂能有效阻断内源性生物素的活性.采用pH9.0Tris-EDTA进行修复加Envision检测系统可提高肝癌组织中AFP的阳性率及阳性强度.结论加热抗原修复可使被甲醛封闭的内源性生物素的活性重新暴露,对免疫组化的染色结果造成影响,生物素阻断系统是消除内源性生物素的有效方法,采用pH9.0的抗原修复液进行修复加Envision非生物素检测系统是免疫组化染色中较为理想的检测方法.  相似文献

The purpose of the present study was to examine the nutritional status of vitamin B1, B2, and B6 in respect to dietary intake of these vitamins and activity coefficients of the erythrocyte enzymes transketolase, glutathione reductase, and aspartic aminotransferase in young men and women with different physical activity levels. The participants of this study were 20 women and 20 men with high physical activity (groups HAW and HAM, respectively), and 20 women and 20 men with low physical activity (groups LAW and LAM, respectively). The intake of vitamins B1, B2, B6, proteins, and calorie content of the diet was based on the average of the 4-day dietary recalls. To assess nutritional status of vitamin B1, B2, and B6, the activity coefficients (α) of erythrocyte transketolase (ETK), erythrocyte glutathione reductase (EGR), and erythrocyte aspartic aminotransferase (EAST) were estimated in blood hemolysates. The intake of the studied vitamins in the diet was statistically significantly lower in the female groups compared with the respective male groups. Deficiency of vitamin B6 in the diet was present more often in women than in men (in terms of the recommended dietary allowances [RDA]). Values of the activity coefficient αETK indicated that none of the groups in this study suffered the risk of vitamin B1 deficiency. The value of the activity coefficient αEGR indicated that the groups of women and men with low physical activity were more prone to vitamin B2 deficiency compared with the high physical activity groups. The risk of vitamin B6 deficiency (αEAST) in both male groups was higher than in both female groups. The obtained results do not allow for unequivocal determination of the impact of sex and the level of physical activity on intake and nutritional status of vitamin B1, B2, and B6. Independently of sex and the level of physical activity, the women and men consumed insufficient quantities of vitamins B1 and B6, although this was not always related to increased values of corresponding activity coefficients.  相似文献

对不同海拔地区的太阳 UV- B辐射和植物叶片的光学特性进行了比较研究。结果表明 :位于高海拔地区的海北高寒草甸生态系统定位站 ,太阳 UV- B辐射明显高于相近纬度的西宁、兰州和南京地区。UV- B辐射与总辐射和 PAR的日变化规律相似 ,都受太阳高度角的直接影响 ,在当地太阳正午时最高。UV- B/Q的日变化也为单峰曲线 ,海北站地区的 UV- B/Q高于西宁的同期测定结果。对珠芽蓼等植物的研究表明 ,生长于海北站地区的珠芽蓼 ,其叶片中紫外线吸收物质的含量明显高于西宁的同种植物 ,也略高于海拔较高的达坂山和小达坂山山顶的同种植物。叶绿素含量以海北站珠芽蓼最低 ,达坂山和小达坂山的同种植物最高。珠芽蓼叶片中类胡萝卜素的含量以西宁最低 ,海北站、达坂山和小达坂山依次升高。海北站矮嵩草与从海北站移植到西宁生长 4年的同种植物相比 ,叶片中紫外线吸收物质、叶绿素、类胡萝卜素的变化与生长于两地区的珠芽蓼相同  相似文献
