Black spot disease is an important disease of roses with worldwide occurrence. It is caused by Diplocarpon rosae , an ascomycetous fungus and its control relies on fungicides. The effects of strobilurins and triazoles on D. rosae development within the host and on disease symptoms have not been well studied. Strobilurins completely inhibited germination of conidia when applied protectively 1 week before inoculation or on the same day with the inoculum. They were, however, not effective in eradicating the disease when applied after the fungus was established in the host. Triazoles reduced the germination rate of the conidia when applied protectively and they inhibited disease symptom development when applied after the fungus was established in the host but before symptom expression. Application of triazoles after symptom development suppressed further development of the disease, but in the case of treatment with myclobutanil yellowing and defoliation still occurred. Post-infection application of triazoles led to the apparent breakdown of subcuticular mycelia, intercellular mycelia, and hyphae in the epidermal cells, while the effects of strobilurins were limited to the subcuticular mycelia. Triazoles were more effective than strobilurins because they are more systemic. 相似文献

Black spot disease of roses caused by the ascomycetous fungus Diplocarpon rosae teleomorph (Marssonina rosae anamorph) is a widespread and devastating disease. Despite considerable progress in the management of black spot disease in the recent years, it is still unclear by which mechanisms this fungus colonises and invades the host system, and without a good knowledge of such infection machineries, it not possible to fully overcome the challenges of D. rosae infection. By exploring research contributions up to date, we highlight in this review the ultrastructure of D. rosae infection cycle in the host cell by emphasising on several aspects related to its in vitro and in vivo germinations, the infection mechanisms of the fungus, the different fungal structures formed in the host cells and the optimum storage conditions for D. rosae to retain its viability and pathogenicity over time. Here, attention is particularly focused on the asexual life cycle of D. rosae, with the sexual cycle being briefly discussed. In addition, a new dimension of research approaches to effectively control black spot disease of roses, that is, how to accurately use the advanced biotechnology tools to speed up the current state of the disease management, is proposed here. 相似文献

Abstract Fifteen pathogenic isolates of Helminthosporium oryzae were established from 15 different rice growing areas representing nine districts: Madurai, Theni, Thanjavur, Thiruvarur, Nagapatinum, Tirunelveli, Trichy, Ariyalur and Cuddalore district of Tamil Nadu. The symptoms initially appeared on the leaves as small, oval, dark brown to black spots. Under favourable conditions, the fungus attacks grain and caused grain discoloration. The isolate, collected from Ammapettai of Thanjavur district was the most virulent (Grade 7.47, PDI 82.96) on rice plants followed by Trichy isolate I2 (Grade 7.26; PDI 80.74) while I15 (Cumbum) was the least virulent (Grade 1.8, PDI 20.00). The isolates of H. oryzae varied in size (length and width) of the conidia, colour and the number of cells per conidium. The maximum (39.1 μm) length of the conidium was observed in I10 followed by I11 (37.3 μm). The maximum (17.1 μm) conidial width was observed in I8. The colour of the conidium varied from light brown (isolates I3, I6 and I14) to brown, while there was no appreciable difference in the conidial shape. The number of cells varied from 2 – 4 among the isolates. Six phylloplane microorganisms were tested against the virulent isolate of H. oryzae. Among them, Cladasporium spp was very effective in inhibiting the mycelial growth and spore germination of H. oryzae followed by Penicillum spp and Aspergillus flavus while Bacillus subtilis was least effective in inhibiting the mycelial growth and spore germination of H. oryzae. 相似文献

Rose black spot is one of the most severe diseases of field-grown roses. Though R-genes have been characterised, little information is known about the molecular details of the interaction between pathogen and host. Based on the recently published genome sequence of the black spot fungus, we analysed gene models with various bioinformatic tools utilising the expression data of infected host tissues, which led to the prediction of 827 secreted proteins. A significant proportion of the predicted secretome comprises enzymes for the degradation of cell wall components, several of which were highly expressed during the first infection stages. As the secretome comprises major factors determining the ability of the fungus to colonise its host, we focused our further analyses on predicted effector candidates. In total, 52 sequences of 251 effector candidates matched several bioinformatic criteria of effectors, contained a Y/F/WxC motif, and did not match annotated proteins from other fungi. Additional sequences were identified based on their high expression levels during the penetration/haustorium formation phase and/or by matching known effectors from other fungi. Several host genotypes that are resistant to the sequenced isolate but differ in the R-genes responsible for this resistance are available. The combination of these genotypes with functional studies of the identified candidate effectors will allow the mechanisms of the rose black spot interaction to be dissected. 相似文献

【背景】由互隔交链孢(Alternaria alternata)引起的黑斑病是库尔勒香梨采后贮期的主要病害之一,严重影响果实品质,造成巨大的经济损失。【目的】筛选对梨黑斑病菌具有高拮抗活性的菌株并探究其可能的抑菌机制,为香梨采后保鲜提供理论依据。【方法】从库尔勒香梨园健康植株根际采集土壤样品,采用稀释涂布法分离细菌并进行纯化,通过平板对峙法和琼脂打孔法筛选对梨黑斑病菌具有显著拮抗作用的菌株,结合形态学观察和16S rRNA基因序列分析进行初步鉴定;通过平板对峙法对拮抗菌抑菌谱进行测定;采用特异性平板定性检测拮抗菌产酶活性,采用3,5-二硝基水杨酸比色法法及福林-酚法定量酶活;通过孢子萌发抑制试验和扫描电镜观察,测定拮抗菌对梨黑斑病菌的拮抗作用。【结果】筛选得到2株对梨黑斑病菌有较强抑制活性的菌株JE53和JE56,经分析鉴定为多粘类芽孢杆菌(Paenibacillus polymyxa)及枯草芽孢杆菌(Bacillus subtilis),其发酵上清液对梨黑斑病菌的抑菌圈直径分别为22.49 mm和22.40 mm。抑菌谱测定结果表明,菌株JE53和JE56对马铃薯黑痣病菌等9种植物病原菌均有不同程度的抑制作用。胞外酶测定结果显示这2株菌可产生4种胞外酶;其发酵上清液能够显著抑制病原菌孢子萌发,抑制率分别为70.30%和88.87%;扫描电镜发现这2株菌可导致病原菌菌丝表面粗糙、扭曲变形。【结论】JE53和JE56对梨黑斑病菌有较好的抑制效果,具有进一步开发和应用的潜力。 相似文献

Aims: Assessment of biological control of Cercospora sojina, causal agent of frogeye leaf spot (FLS) of soya bean, using three indigenous bacterial strains, BNM297 (Pseudomonas fluorescens), BNM340 and BNM122 (Bacillus amyloliquefaciens). Methods and Results: From cultures of each bacterial strain, cell suspensions and cell‐free supernatants were obtained and assayed to determine their antifungal activity against C. sojina. Both mycelial growth and spore germination in vitro were more strongly inhibited by bacterial cell suspensions than by cell‐free supernatants. The Bacillus strains BNM122 and BNM340 inhibited the fungal growth to a similar degree (I~52–53%), while cells from P. fluorescens BNM297 caused a lesser reduction (I~32–34%) in the fungus colony diameter. The foliar application of the two Bacillus strains on soya bean seedlings, under greenhouse conditions, significantly reduced the disease severity with respect to control soya bean seedlings and those sprayed with BNM297. This last bacterial strain was not effective in controlling FLS in vivo. Conclusions: Our data demonstrate that the application of antagonistic bacteria may be a promising and environmentally friendly alternative to control the FLS of soya bean. Significance and Impact of the Study: To our knowledge, this is the first report of biological control of C. sojina by using native Bacillus strains. 相似文献

A previous culture-dependent survey of phylloplane yeasts from selected Mediterranean plants showed that a few species were present in high densities in almost all leaf samples, regardless of the plant type, location or sampling season. However, a few species appeared to be restricted to Cistus albidus leaves, namely Cryptococcus cistialbidi . Here, we describe a culture-independent FISH assay to detect and quantify whole yeast cells in leaf washings. After optimization, the technique was used to check the apparent association between C. albidus leaves and C. cistialbidi and the abundance and ubiquity of other basidiomycetous yeast species such as Erythrobasidium hasegawianum and Sporobolomyces spp. in leaf samples from this and other neighboring plants ( Acer monspessulanum and Quercus faginea ). No yeast cells were detected in Pistacia lentiscus leaf samples. We were also able to demonstrate that three phylloplane yeasts ( C. cistialbidi, E. hasegawianum and Sporobolomyces spp.) appeared to be log-normally distributed among individual C. albidus leaves. The log-normal distribution has important implications for the quantification of phylloplane yeasts based on the washing and plating of bulk leaf samples, which will tend to overestimate the size of the respective populations and become an error source in yeast surveys or related biocontrol studies. 相似文献

[背景] 马铃薯黑痣病是由立枯丝核菌(Rhizoctonia solani)引起的一种典型土传病害,目前该病害生物防治的菌种资源比较有限,相应菌株生防机制的研究更是缺乏。[目的] 明确马铃薯黑痣病病原菌立枯丝核菌(R. solani) JT18的拮抗菌QHZ11对马铃薯黑痣病的生防效果,揭示QHZ11对黑痣病的部分防治机理。[方法] 在灭菌土壤中分别接种R. solani JT18(CK),R. solani JT18和普通有机肥(Organic Fertilized,OF),R.solaniJT18和氨基酸有机肥(AA+OF)及R. solani JT18和QHZ11生物有机肥(BOF11),结合实时荧光定量PCR (Real-Time Fluorescence Quantitative PCR,RT-qPCR)等方法,研究马铃薯全生育期不同处理R.solaniJT18在马铃薯根际和植株不同部位的数量变化及拮抗菌QHZ11与R.solaniJT18的数量消长规律,同时比较不同处理黑痣病的病情指数及相应的防效。[结果] RT-qPCR结果表明,随马铃薯生育进程的推进,马铃薯根际、根系和匍匐茎R.solaniJT18的数量在各处理中均呈现先升高至块茎膨大期到达峰值后下降的趋势,而且各部位R.solaniJT18的数量为CK>OF>AA+OF>BOF11且根际>根系>匍匐茎;拮抗菌QHZ11的数量变化趋势与R.solaniJT18相同,但峰值在块茎形成期,并且同时期同一部位QHZ11的定殖数量均显著高于R.solaniJT18,甚至高出2个数量级,说明QHZ11占用了一定的营养资源和生态位点,严重抑制了R.solaniJT18的生长和繁殖。病情结果表明:CK病情指数最高,OF、AA+OF和BOF11处理均显著低于CK,其中BOF11处理发病最轻;生防结果则相反,为BOF11>AA+OF>OF处理,说明普通有机肥、氨基酸有机肥及生物有机肥均可不同程度地防治马铃薯黑痣病,其中以生物有机肥效果最显著。[结论] QHZ11以有机肥为载体施入土壤后,可以通过在马铃薯根际及植株不同部位竞争营养和生态位点,从而有效抑制黑痣病病原菌R.solaniJT18的生存和繁殖,起到显著的生防效果,这对QHZ11生物有机肥的应用和推广具有重要意义,并为进一步研究QHZ11的生防机制奠定了基础。 相似文献

Three fungal isolates, KF-1, HN-14 and K-12, from barnyardgrass leaf blight, which were collected from Hunan and Beijing, were studied morphologically and molecularly. The results showed that there were differences between the three isolates and the two Exserohilum monoceras strains G-9 and X-27 which were accurately identified and deposited in culture collections of China Agricultural University in colonial morphology, sporulation and spore morphology especially septa of spore. G-9, KF-1, X-27 and HN-14 grow quickly and produce abundant spores on PDA plate, but K-12 grows slowly and produced less spores. To further identify the relationship of 5 isolates, 5.8S-ITS sequence were compared. Results indicated that the similarity of ITS sequence of five isolates were over 98% and the phylogenetic tree based on ITS1 and ITS2 sequence also revealed that G-9, KF-1, K-12, X-27, and HN-14 were all clustered into one group and distinct from the other outgroup and suboutgroup. Based on the above data, these three isolates were proved to be E. monoceras. 相似文献

采用形态学及分子生物学的方法对采集自湖南和北京的3株尖角突脐孢菌分离物进行了鉴定。结果表明,3株分离物(KF-1、HN-14和K-12)和保存于中国农业大学已定名的尖角突脐孢菌菌株G-9和X-27之间在菌落形态、产孢量及孢子大小和分隔方面存在较大差异。其中,K-12在PDA培养基上生长缓慢、产孢量小;菌株G-9、KF-1、X-27和HN-14生长迅速,产孢丰富。对菌株进行分子鉴定结果表明,菌株间ITS区序列相似度达98%以上,聚类分析也表明,种内各菌株之间的遗传距离明显小于种间的遗传距离;基于ITS1及ITS2序列,能将尖角突脐孢菌和突脐蠕孢属中其它种分开。由此可确定分离自湖南水稻田及中国农业大学科学园温室中自然发病的稗草病样上的3株病原真菌均为尖角突脐孢菌。 相似文献

Fifty plant extracts, four oil cakes and eight antagonistic organisms were tested against Bipolaris oryzae (Cochliobolus miyabeanus), the causal agent of brown spot disease of rice. In vitro studies indicated that two leaf extracts, Nerium
oleander and Pithecolobium dulce exerted the higher percent inhibition to mycelial growth (77.4, 75.1%) and spore germination (80.3, 80.0%) of B. oryzae. Among the four oil cake extracts tested in vitro against B. oryzae, neem cake extract showed the maximum inhibition percent to mycelial growth (80.18%) and spore germination (81.13%) of the
pathogen followed by mahua cake extract, castor and gingelly cake extract. Trichoderma viride (Tv2) was significantly effective in inhibiting the mycelial growth (62.92%) and spore germination (77.03%) of the pathogen followed
by Trichoderma harzianum (Th5) and Trichoderma reesei (Tr3). The promising leaf extracts, oil cake extracts and antagonistic microorganisms were further evaluated for their efficacies
in disease management under glasshouse and field conditions. In glasshouse studies, post-infectional spraying of rice plants
with neem cake extract, N. oleander leaf extract and T. viride (Tv2) was significantly effective in reducing the incidence of brown spot of rice by 66, 52 and 45 percent respectively.
Two rounds of spraying of rice plants with neem cake extract, N. oleander leaf extract and T. viride (Tv2) in the field at initial appearance of disease and 15 days later reduced the incidence of brown spot (70, 53 and 48%

柑橘黑斑病是柑橘的重要病害之一。本文通过病原菌的形态特征和致病性,并结合其rDNA-ITS区域序列及柯赫法则,对从福建省福州市闽侯县福橘果上分离的黑斑病病原菌进行了鉴定分析。结果表明:PDA平板上的病原菌菌落生长缓慢,呈圆形凸起;分生孢子器近球形、黑色炭质;分生孢子单孢、无色,呈椭圆形,大小为(7~12)μm×(5~8)μm,分生孢子外包裹一层半透明凝胶状物质。结合序列比对分析结果,最终将分离的福橘果实黑斑病的病原菌鉴定为柑橘球座菌Guig-nardia citricarpa Kiely。本研究为柑橘黑斑病的早期诊断和防治奠定了基础。 相似文献

Plant growth-promoting bacteria-mediated biocontrol of plant pathogens is renowned to enhance the growth of the plants using different direct or indirect mechanisms. The goal of the present investigation was the evaluation of Pseudomonas aeruginosa Z5 isolated from cotton grown in Pakistani soils for the suppression of Fusarium oxysporum associated with cotton seedling disease. In dual culturing techniques, four bacterial strains inhibited fungal pathogens, i.e. F. oxysporum, Fusarium moniliforme, Fusarium solani and Rhizoctonia solani, significantly with percent inhibition ranging from 25% to 91.5%. P. aeruginosa Z5 showed maximum suppression of all the tested pathogens. Net-house experiments showed that the application of P. aeruginosa Z5 both separately and in combination with Bacillus fusiformis S10 significantly reduced the disease incidence by suppressing F. oxysporum (the causal agent of cotton seedling disease) up to 64–65% and improved the percent germination as compared to the infected control plants. The production of antibiotics, proteases and siderophores may be the contributing factors for its antagonistic properties. Highest bacterial population (8.9 CFU/g root) observed on roots of cotton plants inoculated with P. aeruginosa Z5 showed its good colonisation aptitudes even in the presence of high inoculation of soil with F. oxysporum. Confocal laser scanning microscopy supported the root colonisation of cotton plants with fluorescently labelled P. aeruginosa Z5. Because of innate fungicidal potential, growth promoting P. aeruginosa Z5 can be used as a bioinoculant and an antagonist to suppress the growth of cotton root-associated fungal pathogen. 相似文献

Experiments were conducted with Botrytis cinerea on strawberry leaves to investigate where combinations of commercially available biological control agents (BCAs) might control B. cinerea more effectively than individual BCAs. Specifically, we studied the persistence of biocontrol activities, spread of BCAs among leaves, and biocontrol efficacy in relation to application regimes: mixed versus single BCA, pre-versus post-inoculation application, and sequential versus simultaneous application. Three BCA products (Sentinel, Serenade and Trianum) were used for this study. Overall, Serenade did not significantly reduce sporulation of B. cinerea on strawberry leaf discs whereas Sentinel and Trianum gave a similar and significant biocontrol efficacy. Biocontrol efficacy remained almost unchanged 10 days after application at 20/20°C (day/night) or 24/16°C temperature regimes. In contrast, reduced biocontrol efficacy at 26/14°C suggests BCA survival was reduced under these conditions. Incidence of B. cinerea sporulation on leaf discs was ca. 60% higher on leaves that emerged after the BCA application than on leaves directly exposed to BCA, indicating insufficient amount of the BCA had managed to spread to new leaves. Combinations of BCAs, whether applied simultaneously or sequentially (48 h apart), did not improve disease control over the most effective BCA within the combination applied alone. This indicated possible antagonism or interference between the BCAs. Results suggested that there was significant antagonism for most combinations of the three BCAs tested and the degree of antagonism increased as the time from BCA application to pathogen introduction lengthened. 相似文献
