The roots of Haemanthus kalbreyeri contain a new phenanthridone alkaloid, kalbretorine, and a new glucosyloxy alkaloid, kalbreclasine. Additionally, six known alkaloids, viz. haemanthamine, haemanthidine, hippadine, lycorine, narciclasine and pratorimine, previously reported from other Amaryllidaceous plants have now been isolated also from this species. Kalbretorine produced marked inhibition of growth and viability of S-180 tumour cells. Kalbreclasine caused significant mitogenic activation of splenic lymphocytes characteristic of immuno stimulants.  相似文献

Two rare indazole-type alkaloids, nigeglapine (1) and nigeglaquine (2) along with one known compound (3), were isolated from the seeds of Nigella glandulifera. Their structures were confirmed on the basis of chemical and spectroscopic methods, mainly NMR (1H, 13C, HSQC, HMBC and NOESY) and ESI-MS, and comparison with data in the literature. This is the first report of the natural occurrence of glycosylated indazole-type alkaloids.  相似文献

From an extract of the roots of Cadaba natalensis Sond. the novel macrocyclic dibenzo-diazacyclododecanedione 1 was isolated together with (S)-2-ethyl-2-methyloxazolidin-5-one (2a). In addition, the five known compounds were obtained. Of these, (R)-5-ethyl-5-methyloxazolidin-2-one (3) is reported as a natural product for the first time. The structures of the isolated compounds were elucidated by analysis of the spectral data, 1D NMR (1H, 13C, and DEPT), 2D NMR (COSY, HMQC, HMBC, and NOESY), and HRESIMS, as well as by the comparison with previously reported data.  相似文献

Three new C19-diterpenoid alkaloids, tianshanisine (1), tianshanine (2) and tianshanidine (3), together with six known C19-diterpenoid alkaloids (4–9) were isolated from the whole herb of Delphinium tianshanicum W. Twelve prenylated carbazole alkaloids, containing a novel prenylated carbazole alkaloid, named as clausevestine (1), and 11 known prenylated carbazole alkaloids (212), were isolated and identified from the stems and leaves of Clausena vestita, which is a Chinese endemic plant. The chemical structure of 1 was established by means of comprehensive spectroscopic data analyses and the known compounds were determined via comparing their NMR and MS data as well as optical rotation values with those reported in literature. Especially, clausevestine (1) is an unusual prenylated carbazole alkaloid possessing an unprecedented carbon skeleton holding 20 carbon atoms. The anti-inflammatory effects and antiproliferative activities of those isolated prenylated carbazole alkaloids were tested. Prenylated carbazole alkaloids 112 displayed remarkable inhibitory effects on NO (nitric oxide) production with IC50 values equivalent to that of the positive control (hydrocortisone). Meanwhile, prenylated carbazole alkaloids 112 exhibited remarkable antiproliferative activities against diverse human cancer cell lines in vitro holding the IC50 values ranging from 0.32 ± 0.04 to 18.76 ± 0.18 µM. These findings indicate that these prenylated carbazole alkaloids possessing remarkable anti-inflammatory effects and antiproliferative activities could be meaningful to the discovery of new anti-inflammatory and anti-tumor candidate drugs.  相似文献

Seventeen quinazoline alkaloids and derivatives, containing two pairs of new epimers, named as (S)- and (R)-1-(2-aminobenzyl)-3-hydroxypyrrolidin-2-one β-d-glucopyranosyl-(1?→?6)-β-d-glucopyranoside (1, 2), (S)- and (R)-vasicinone β-d-glucopyranosyl-(1?→?6)-β-d-glucopyranoside (3, 4), and a new enantiomer (12b), together with six known ones (58, 10, and 12a), and three pairs of known enantiomers (9, 11, and 13), were isolated from the ethanol extracts of the seeds of Peganum harmala L.. Their structures including the absolute configuration were elucidated by using 1D and 2D NMR, and ECD calculation approaches. The cytotoxic activities of all isolated compounds were evaluated. 11 showed moderate cytotoxicity against PC-3 cells with an IC50 value of 15.41?μM.  相似文献

Phytochemical investigation of the alkaloidal extract of the roots of Toddalia asiatica led to the isolation of a new seco-benzophenanthridine alkaloid (1) and twelve known alkaloids (213). The new structure was elucidated by means of spectroscopic analysis, and the known alkaloids were identified by comparison with the literature. Among these alkaloids, 1 and 57 were reported from the genus Toddalia for the first time. The distribution of the isolated alkaloids at genus/family level was presented via network analysis and their chemotaxonomic significance was also discussed.  相似文献

A new cytotoxic β-carboline alkaloid, 1-methyl-3-(2-hydroxypropan-2-yl)-2-(5-methoxy-9H-β-carbolin-1-yl)-cyclopentanol (1), was isolated from roots of Galianthe thalictroides, together with the alkaloid 1-(hydroxymethyl)-3-(2-hydroxypropan-2-yl)-2-(5-methoxy-9H-β-carbolin-1-yl)-cyclopentanol (2), the anthraquinones 1-methyl-alizarin and morindaparvin-A, the coumarin scopoletin, homovanillic alcohol, (−)-epicatechin, and the steroids stigmast-4-en-3-one, 4,22-stigmastadien-3-one, campest-4-en-3-one, stigmast-4-en-3,6-dione, 6-β-hydroxy-stigmast-4-en-3-one, stigmasterol, campesterol, β-sitosterol, and β-sitosterol-3-O-β-d-glucopyranoside. Among the previously known compounds, homovanillic alcohol is a novel finding in Rubiaceae, while 1-methyl-alizarin, morindaparvin-A, scopoletin, stigmast-4-en-3-one, 4,22-stigmastadien-3-one, campest-4-en-3-one, stigmast-4-en-3,6-dione, and 6-β-hydroxy-stigmast-4-en-3-one is reported for the first time in the genus Galianthe. The cytotoxic β-carboline alkaloids 1 and 2 exhibited potent antitopoisomerase I and IIα activities and strong evidence is provided for their action as topoisomerase IIα poisons and redox-independent inhibitors.  相似文献

The phytochemical investigation of the stems of Fissistigma maclurei Merr. led to the isolation of a new eudesmane-type sesquiterpene 6α,12-dihydroxy-4(15)-eudesmen- 1-one (1), together with seven known eudesmane-type sesquiterpenes (28) and four known aporphine-type alkaloids (912). The structures of these compounds were elucidated using comprehensive spectroscopic methods. All compounds were isolated from the F. maclurei for the first time. Chemical investigation on the whole plants of Stenosolenium saxatile led to the isolation of three naphthoquinones (13), five pyrrolizidine alkaloids (PAs) (4–8), and six phenolic acids (9–14). The structure of the isolated compounds was elucidated by different spectrometric methods (1H NMR, 13C NMR and ESI-MS). These results are the first chemical data on constituents of this genus. The chemotaxonomic relationships between the genera Stenosolenium, Onosma, Arnebia, Echium and Lithospermum are also discussed.  相似文献

Two new benzylisoquinoline alkaloids, namely, hexapetalines A (1) and B (2), along with 11 known alkaloids were isolated from the stems of Artabotrys hexapetalus. The structures of 1 and 2 with their absolute configurations were elucidated by extensive spectroscopic methods and the known compounds were identified by comparisons with data in the literature. All new compounds were evaluated for their cytotoxicities against five human cancer cell lines: HL-60, SMMC-7721, A-549, MCF-7 and SW480 in vitro. Alkaloids 1 and 2 exhibited inhibitory effects with IC50 values comparable to those of cisplatin.  相似文献

Three new compounds, 17β-cevanin-6-oxo-5α,20β-diol yibeinine (1), 2-(tetrahydro-5-(2-hydroxyphenyl)-2H-pyran-3-yl) phenol (2), 1,3-O-diferuloyl-2-methoxypropane diol (3), as well as four known compounds (47), have been isolated from the ethanol extract of dried bulbs of Fritillaria pallidiflora Schrenk. All structures were determined based on their spectroscopic data (1D and 2D NMR (including 1H NMR, 13C NMR, 1H-1H COSY, HMBC, HSQC, HSQC-TOCSY, and NOESY experiments), and MS). Biological evaluation showed that compounds 14 inhibited the production of nitric oxide (NO) in LPS-stimulated RAW 264.7 cells with IC50 values of 18.0, 38.7, 29.5, and 47.1 μM, respectively. These results indicated that compound 1 has potential anti-inflammatory activity.  相似文献

The phytochemical investigation of the leaves and branches of Annona coriacea Mart. (Annonaceae) led to the isolation and characterization of eight compounds: five isoquinoline-derived alkaloids, including pukateine (1), liriodenine (4), anonaine (5), obovanine (6), and norisocorydine (7); one terpene lactone known as loliolide (3); one benzoic acid derivative, 2-methoxybenzoic acid (2), and 3,4,5-trimethoxyphenol (8). All compounds, except liriodenine, are being described for the first time in the species A. coriacea, and their chemophenetics relationships were discussed. The structures were elucidated by extensive analyses of 1D and 2D NMR (COSY, HSQC, and HMBC) spectroscopy in combination with MS, and the data were compared with literature values. The NMR dataset of pukateine and obovanine was reviewed. Our results showed that A. coriacea is a typical species of the Annonaceae family and an important source of aporphine alkaloids with chemophenetic relationships with Xylopia, Duguetia, Guatteria, Artabotrys, and Goniothalamus genera.  相似文献

Investigation of the Red Sea marine tunicate Symplegma rubra Monniot, 1972 gave three new purine alkaloids namely 6-methoxy-7,9-dimethyl-8-oxoguanine (1), 6-methoxy-9-methyl-8-oxoguanine (2), and 2-methoxy-7-methyl-8-oxoadenine (4) together with seven known compounds: 6-methoxy-7-methyl-8-oxoguanine (3), 9-methyl-8-oxoadenine (5), 7-methyl-8-oxoadenine (6), 8-oxoadenine (7), 3-methylxanthine (8), inosine (9), and homarine (pyridinium-2-carboxylic acid-1-methyl) (10). Compound 6 was reported here for the first time from a natural source. The structure determination of the compounds was accomplished by extensive interpretation of their spectroscopic data including 1D (1H and 13C) and 2D (1H–1H COSY, HSQC, and HMBC) NMR and high-resolution mass spectral data. The isolated compounds were evaluated for their protein kinase inhibitory activity against different kinases (CDK5, CK1, DyrK1A, and GSK3) at 10 μg/mL. Two new monoterpenoid indole alkaloids, melohenryines A and B (1 and 2), along with six known indole alkaloids, were isolated from the twigs and leaves of Melodinus henryi. Structures of the new alkaloids were established by extensive spectroscopic techniques including NMR spectroscopy and mass spectrometry. Melohenryine A (1) represents the first example of monoterpenoid indole alkaloids characterized an ester carbonyl group at C-19 position. A phytochemical investigation on the aerial parts of Gardneria ovata Wall resulted in the isolation and identification of 14 compounds, including three gardneria glycoalkaloids (1–2 and 6), seven gardneria alkaloids (3–5 and 7–10), and four oxindole alkaloids (1114). The structure of compound 1 was elucidated by means of spectroscopic analysis, including HRESIMS together with 1D and 2D NMR experiments. Compounds 1–2 and 11–14 are the first time reported from of G. ovate, while the compounds 3–4, 6, and 8 are also the characteristic secondary metabolites of the title plant. The chemotaxonomic significance and distribution of these monoterpenoid indole alkaloids in Gardneria genus are discussed.  相似文献

Three new norditerpenoids alkaloids, 1β-hydroxy,14β-acetyl condelphine (1), jadwarine-A (2), jadwarine-B (3) along with two known alkaloids isotalatizidine hydrate (4) and dihydropentagynine (5) were isolated from medicinal plant Delphinium denudatum. The structures of natural products 15 were established on the basis of HR-EIMS, 1H and 13C NMR (1D & 2D) spectroscopic data as well as by comparison from literature data. The structures of compound 1 and 4 were also confirmed by single crystal X-ray diffraction studies. In-vitro AChE and BChE enzyme inhibitory activities of compounds 15 and molecular docking studies were performed to investigate the possible molecular inhibitory mechanism of the isolated natural products. Compound 2, 4 and 5 showed competitive inhibitory effects by inhibiting AChE and BChE, respectively, while 1 and 3 showed non-competitive inhibition. This work is the first report that provides a supporting evidence about the use of constituents of Delphinium denudatum in cerebral dementia and Alzheimer diseases.  相似文献

Clausena lansium (Lour.) Skeels is an evergreen small tree or shrub with great economic value, which belongs to the genus Clausena of the Rutaceae family. C. lansium is indigenous to Southern China, while currently widely cultivated in subtropical and tropical regions not only for the nutritional value and pharmacological uses of its fruits but also as a medicinal and ornamental plant. In this study, a systematic phytochemical study on the stems and leaves of C. lansium caused the separation and identification of two new geranylated carbazole alkaloids, clauselansiumines A (1) and B (2), as well as 10 known geranylated carbazole alkaloids (312). The chemical structures of these isolated geranylated carbazole alkaloids (112) were unambiguously determined based on comprehensive spectral data analyses. All these isolated geranylated carbazole alkaloids were tested for their neuroprotective effects against 6-hydroxydopamine induced cell death in human neuroblastoma SH-SY5Y cells in vitro. Compounds 112 displayed remarkable neuroprotective effects holding the EC50 values ranging from 0.48 ± 0.04 to 12.36 ± 0.16 μM. These research results disclosed that the separation and purification of these geranylated carbazole alkaloids possessing remarkable neuroprotective effects separated from C. lansium could be extremely important to the discovery of new agents for the treatment and prevention for Parkinson's disease.  相似文献

By feeding 1-methyl-l-tryptophan (1-MT) into cultures of the arthropod-associated fungus Chaetomium globosum TW1-1, three novel cytochalasan alkaloids, termed as armochaetoglosins A–C (13), together with five known analogues, namely prochaetoglobosin I (4), chaetoglobosin T (5), chaetoglobosin C (6), armochaetoglobin Y (7), and chaetoglobosin Vb (8), were isolated and characterized. Their structures including absolute configurations were elucidated by means of NMR spectroscopy, single-crystal X-ray crystallography, and comparison of the experimental electronic circular dichroism (ECD) spectra. Structurally, compounds 13 represented the first examples of 1′-N-methyl-chaetoglobosins, which were possibly biosynthesized from the additive 1-MT rather than tryptophan. Additionally, compound 3 showed the highest antibacterial activity against K. pneumoniae and ESBL-E. coli with MIC values of 4.0 μg/mL and 16.0 μg/mL, respectively, wherein the inhibitory effect of 3 against K. pneumoniae was stronger than that of the clinically used antibiotic meropenem, with an MIC value of 8 μg/mL. Our findings may provide new chemical templates for the development of new antibacterial agents against drug-resistant microbial pathogens.  相似文献

Two new acridone alkaloids, verdoocridone A (1) and B (4), together with fifteen known compounds were isolated from methanol extracts of the roots and leaves of Vepris verdoorniana. The structures of all compounds were determined by comprehensive spectroscopic analyses (1D and 2D NMR, EI- and ESI–MS). The 13C NMR values of 1,2,3,5-tetramethoxy-N-methylacridone (2) and 5-methoxyaborinine (3) are also reported. The crude extracts and compounds (1-6) were tested for their antimicrobial activity. The test delivered moderate activities for crude extracts and compounds 1, 5 and 6 against the bacterium Staphylococcus aureus and the fungi Mucor meihei and Candida albicans with MIC values between 115 and 180 μg/mL for extracts and between 21.3 and 29.4 μM for compounds, compared to gentamycin with 0.2 μM and nystatin with 5.2 μM against both fungi. The determination of the radical scanvenging activity using 1,1-dephenyl-2-picrylhydrazyl (DPPH) assay gave moderate antioxidant values for all tested compounds, with IC50 between 0.29 and 0.41 μM, compared to the standard 3-t-butyl-4-hydroxyanisole (BHA) displaying 0.03 μM.  相似文献
