Metarhizium anisopliae is a well-characterized biocontrol agent of a wide range of insects including cane grubs. In this study, a two-dimensional (2D) electrophoresis was used to display secreted proteins of M. anisopliae strain FI-1045 growing on the whole greyback cane grubs and their isolated cuticles. Hydrolytic enzymes secreted by M. anisopliae play a key role in insect cuticle-degradation and initiation of the infection process. We have identified all the 101 protein spots displayed by cross-species identification (CSI) from the fungal kingdom. Among the identified proteins were 64-kDa serine carboxypeptidase, 1,3 beta-exoglucanase, Dynamin GTPase, THZ kinase, calcineurin like phosphoesterase, and phosphatidylinositol kinase secreted by M. ansiopliae (FI-1045) in response to exposure to the greyback cane grubs and their isolated cuticles. These proteins have not been previously identified from the culture supernatant of M. anisopliae during infection. To our knowledge, this the first proteomic map established to study the extracellular proteins secreted by M. ansiopliae (FI-1045) during infection of greyback cane grubs and its cuticles.  相似文献

Frankia strains symbiotic with Ceanothus present an interesting opportunity to study the patterns and causes of Frankia diversity and distribution within a particular host infectivity group. We intensively sampled Frankia from nodules on Ceanothus plants along an elevational gradient in the southern Sierra Nevada of California, and we also collected nodules from a wider host taxonomic and geographic range throughout California. The two sampling scales comprised 36 samples from eight species of Ceanothus representing six of the seven major biogeographic regions in and around California. The primary objective of this study was to use a quantitative model to test the relative importance of geographic separation, host specificity, and environment in influencing the identity of Ceanothus Frankia symbionts as determined by ribosomal DNA sequence data. At both sampling scales, Frankia strains symbiotic with Ceanothus exhibited a high degree of genetic similarity. Frankia strains symbiotic with Chamaebatia (Rosaceae) were within the same clade as several Ceanothus symbionts. Results from a classification and regression tree model used to quantitatively explain Frankia phylogenetic groupings demonstrated that the only significant variable in distinguishing between phylogenetic groups at the more local sampling scale was host species. At the regional scale, Frankia phylogenetic groupings were explained by host species and the biogeographic province of sample collection. We did not find any significant correspondence between Frankia and Ceanothus phylogenies indicative of coevolution, but we concluded that the identity of Frankia strains inhabiting Ceanothus nodules may involve interactions between host species specificity and geographic isolation.  相似文献

Fifty-six human and 24 adult dairy cattle isolates of Salmonella enterica serovar Typhimurium from a single county in California were compared using ribotyping, insertion sequence typing (IS200), pulsed-field gel electrophoresis, plasmid typing, phage typing, and antimicrobial resistance testing. The majority of the isolates fell into one of two groups which were phage types DT104 and DT193. Combining the information from all typing methods, a total of 45 different “clusters” were defined, with 35 of those including only a single isolate. The library of isolates had a high degree of variability, but antibiotic resistance and plasmid typing each defined single clusters in which human or bovine isolates predominated (χ2, P < 0.05).  相似文献

Territorial species, such as the spotted owl (Strix occidentalis),are predicted to follow an ideal despotic distribution. However,debate exists on whether wild populations actually meet theassumptions of an ideal distribution, such as perfect perceptualabilities (i.e., the ability to recognize high- and low-qualitysites without error). Because this hypothesis has importantlife history ramifications for spotted owls, we investigatedwhether occupancy rates of California spotted owl (S. o. occidentalis)territories in the San Bernardino Mountains of southern Californiapositively correlated with a qualitative "potential fitness"(denoted by pf) estimated from survival and reproduction ofterritorial owls. Spotted owls in our study tended to occupyterritories with the highest pf, supporting the assumption ofideal perceptual abilities within this population. However,this relationship was noisy, and we suggest that some individualsdo not assess site quality accurately because of perceptuallimitations, prey dynamics, and large territory sizes. Furthermore,dispersal processes, high survival rates, and long life spansof spotted owls may be other key factors preventing some individualsfrom selecting sites of the highest quality and, consequently,our ability to precisely estimate pf.  相似文献

No remedies for prion disease have been established, and the conversion of normal to abnormal prion protein, a key event in prion disease, is still unclear. Here we found that substances in beetle grub hemolymph, after they were browned by aging for a month or heating for hours, reduced abnormal prion protein (PrP) levels in RML prion-infected cells. Active anti-prion components in the hemolymph were resistant to protease treatment and had molecular weights larger than 100 kDa. Aminoguanidine treatment of the hemolymph abolished its anti-prion activity, suggesting that Maillard reaction products are enrolled in the activity against the RML prion. However, levels of abnormal PrP in RML prion-infected cells were not decreased by incubation with the Maillard reaction products formed by amino acids or bovine serum albumin. The anti-prion components in the hemolymph modified neither cellular or cell-surface PrP levels nor lipid raft or autophagosome levels. The anti-prion activity was not observed in cells infected with 22 L prion or Fukuoka-1 prion, suggesting the anti-prion action is prion strain-dependent. Although the active components of the hemolymph need to be further evaluated, the present findings imply that certain specific chemical structures in the hemolymph, but not chemical structures common to all Maillard reaction products, are involved in RML prion formation or turnover, without modifying normal PrP expression. The anti-prion components in the hemolymph are a new tool for elucidating strain-dependent prion biology.  相似文献

The global distribution of bluetongue virus (BTV) has been changing recently, perhaps as a result of climate change. To evaluate the risk of BTV infection and transmission in a BTV-endemic region of California, sentinel dairy cows were evaluated for BTV infection, and populations of Culicoides vectors were collected at different sites using carbon dioxide. A deterministic model was developed to quantify risk and guide future mitigation strategies to reduce BTV infection in California dairy cattle. The greatest risk of BTV transmission was predicted within the warm Central Valley of California that contains the highest density of dairy cattle in the United States. Temperature and parameters associated with Culicoides vectors (transmission probabilities, carrying capacity, and survivorship) had the greatest effect on BTV’s basic reproduction number, R0. Based on these analyses, optimal control strategies for reducing BTV infection risk in dairy cattle will be highly reliant upon early efforts to reduce vector abundance during the months prior to peak transmission.  相似文献

The mass distribution along the fronds of Macrocystis is examined for plants collected from California and New Zealand. Analysis of fronds classified according to length and condition yields polynomial curves for cumulative mass as a function of distance above the holdfast. Models for this functional relationship are discussed. Similarities and differences between the deep-water California plant and the shallow-water New Zealand plant are highlighted.  相似文献

Pharmacokinetics of oxytetracycline (OTC) were studied in 4 cows after administration of either a conventional (OTC-C) or a long-acting (OTC-LA) preparation. After intravenous administration of OTG-G the elimination half-life for OTG was found to be 6 h. Intramuscular injection of OTC-C and OTC-LA resulted in almost identical plasma concentrations of OTC with peak values after 6–8 h. For both preparations the bioavailability after i.m. administration was 100 % and about 60 % of the dose was excreted in the urine during the first week. Plasma concentrations above 0.5 μg/ml were with both preparations maintained for approximately 60 h, indicating no retard effect of OTC-LA as compared to OTC-C.  相似文献

研究了亚热带高尔夫球场不同功能区优势种蛴螬种群空间分布及时间和空间的生态位.结果表明,球场4种优势种草地蛴螬时间生态位宽度几乎均在0.9以上(除果岭Ⅲ区及Ⅳ区个别蛴螬种外),其中短草高地和果岭Ⅰ、Ⅱ区中4种蛴螬时间生态位宽度均最大,长草高地和果岭Ⅳ区的时间生态位宽度则最低.除浅棕鳃金龟垂直空间生态位宽度在球道4区均达0.8以上外,其它种蛴螬在球场各区普遍偏低(0.2~0.7).在球道和果岭4区中,4种蛴螬种对时间生态位相似性指数均高达0.8以上,垂直空间生态位相似性指数大多数蛴螬种对在0.1~0.6之间,球道以铜绿丽金龟与大绿金龟种对为最高(0.8以上),果岭则是铜绿丽金龟与大绿金龟、华南大黑鳃金龟与浅棕鳃金龟种对为最高(0.8以上).垂直空间分布上,球道和果岭各区中4种蛴螬主要活动于食源最为丰富的上、中层土壤空间(0~5和5~10cm),少数出现于球道部分区10~15cm土层中活动和取食,4种蛴螬几乎不在果岭10~15cm土层中活动和取食。  相似文献

Plate tectonics can have profound effects on organismal distribution and is often the driving force in speciation. Through geologic processes, the Baja California Peninsula depicts two faunal patterns: one through southern vicariance with Cape separation, and the other through dispersal onto the northern peninsula, referred to as a ‘dual-peninsular effect.’ Here we apply a hierarchical sampling strategy that combines population-level sequence data (800 bp, nad4 region) with complete mt-genome data (aligned 15,549 bp) and 5 nuclear protein encoding loci (3315 bp), to test whether both patterns have occurred in one group of nightsnakes (Hypsiglena). The geologic formation of the peninsula is thought to have occurred in three stages: (1) Cape separation from mainland Mexico; (2) the northern peninsula separated, forming the northern Gulf of California; and (3) the peninsula was united through volcanic activity, while moving northward causing collision with southern California. However, the timing of events is debated. We explore phylogenetic relationships and estimate dates of divergence for nightsnakes using our hierarchical sampling strategy. Our data support both ‘southern-vicariance’ and ‘northern-dispersal’ have occurred in nightsnakes, forming a ring distribution around the Gulf of California. Two divergent forms are sympatric on the southern half of the peninsula with no indication of hybridization. Nightsnakes represent the first group to depict the ‘dual-peninsular effect’ with extensive overlap on the Baja California Peninsula.  相似文献

Abstract. Generalized additive, generalized linear, and classification tree models were developed to predict the distribution of 20 species of chaparral and coastal sage shrubs within the southwest ecoregion of California. Mapped explanatory variables included bioclimatic attributes related to primary environmental regimes: averages of annual precipitation, minimum temperature of the coldest month, maximum temperature of the warmest month, and topographically-distributed potential solar insolation of the wettest quarter (winter) and of the growing season (spring). Also tested for significance were slope angle (related to soil depth) and the geographic coordinates of each observation. Models were parameterized and evaluated based on species presence/absence data from 906 plots surveyed on National Forest lands. Although all variables were significant in at least one of the species’ models, those models based only on the bioclimatic variables predicted species presence with 3–26% error. While error would undoubtedly be greater if the models were evaluated using independent data, results indicate that these models are useful for predictive mapping – for interpolating species distribution data within the ecoregion. All three methods produced models with similar accuracy for a given species; GAMs were useful for exploring the shape of the response functions, GLMs allowed those response functions to be parameterized and their significance tested, and classification trees, while some-times difficult to interpret, yielded the lowest prediction errors (lower by 3–5%).  相似文献

Questions: To what extent do plant species traits, including life history, life form, and disturbance response characteristics, affect the degree to which species distributions are determined by physical environmental factors? Is the strength of the relationship between species distribution and environment stronger in some disturbance‐response types than in others? Location: California southwest ecoregion, USA. Methods: We developed species distribution models (SDMs) for 45 plant species using three primary modeling methods (GLMs, GAMs, and Random Forests). Using AUC as a performance measure of prediction accuracy, and measure of the strength of species–environment correlations, we used regression analyses to compare the effects of fire disturbance response type, longevity, dispersal mechanism, range size, cover, species prevalence, and model type. Results: Fire disturbance response type explained more variation in model performance than any other variable, but other species and range characteristics were also significant. Differences in prediction accuracy reflected variation in species life history, disturbance response, and rarity. AUC was significantly higher for longer‐lived species, found at intermediate levels of abundance, and smaller range sizes. Models performed better for shrubs than sub‐shrubs and perennial herbs. The disturbance response type with the highest SDM accuracy was obligate‐seeding shrubs with ballistic dispersal that regenerate via fire‐cued germination from a dormant seed bank. Conclusions: The effect of species characteristics on predictability of species distributions overrides any differences in modeling technique. Prediction accuracy may be related to how a suite of species characteristics co‐varies along environmental gradients. Including disturbance response was important because SDMs predict the realized niche. Classification of plant species into disturbance response types may provide a strong framework for evaluating performance of SDMs.  相似文献

We analyzed the variation with depth in the composition of members of the domain Bacteria in samples from alkaline, hypersaline, and currently meromictic Mono Lake in California. DNA samples were collected from the mixolimnion (2 m), the base of the oxycline (17.5 m), the upper chemocline (23 m), and the monimolimnion (35 m). Composition was assessed by sequencing randomly selected cloned fragments of 16S rRNA genes retrieved from the DNA samples. Most of the 212 sequences retrieved from the samples fell into five major lineages of the domain BACTERIA: alpha- and gamma-Proteobacteria (6 and 10%, respectively), Cytophaga-Flexibacter-Bacteroides (19%), high-G+C-content gram-positive organisms (Actinobacteria; 25%), and low-G+C-content gram-positive organisms (Bacillus and Clostridium; 19%). Twelve percent were identified as chloroplasts. The remaining 9% represented beta- and delta-Proteobacteria, Verrucomicrobiales, and candidate divisions. Mixolimnion and oxycline samples had low microbial diversity, with only 9 and 12 distinct phylotypes, respectively, whereas chemocline and monimolimnion samples were more diverse, containing 27 and 25 phylotypes, respectively. The compositions of microbial assemblages from the mixolimnion and oxycline were not significantly different from each other (P = 0.314 and 0.877), but they were significantly different from those of chemocline and monimolimnion assemblages (P < 0.001), and the compositions of chemocline and monimolimnion assemblages were not significantly different from each other (P = 0.006 and 0.124). The populations of sequences retrieved from the mixolimnion and oxycline samples were dominated by sequences related to high-G+C-content gram-positive bacteria (49 and 63%, respectively) distributed in only three distinct phylotypes, while the population of sequences retrieved from the monimolimnion sample was dominated (52%) by sequences related to low-G+C-content gram-positive bacteria distributed in 12 distinct phylotypes. Twelve and 28% of the sequences retrieved from the chemocline sample were also found in the mixolimnion and monimolimnion samples, respectively. None of the sequences retrieved from the monimolimnion sample were found in the mixolimnion or oxycline samples. Elevated diversity in anoxic bottom water samples relative to oxic surface water samples suggests a greater opportunity for niche differentiation in bottom versus surface waters of this lake.  相似文献

To better understand the differences in the efficacy of entomopathogenic nematode species against white grub species, we are studying the various steps of the infection process of entomopathogenic nematodes into different white grub species using nematode species/strains with particular promise as white grub control agents. In this study we compared the attraction of the entomopathogenic nematodes Steinernema scarabaei (AMK001 strain), Steinernema glaseri (NC1 strain), Heterorhabditis zealandica (X1 strain), and Heterorhabditis bacteriophora (GPS11 strain) to third-instars of the scarabs Popillia japonica, Anomala orientalis, Cyclocephala borealis, and Rhizotrogus majalis, and late-instar greater wax moth, Galleria mellonella, larvae. Individual larvae were confined at the bottom of 5.5 cm vertical sand columns, nematodes added to the sand surface after 24 h, and nematodes extracted after another 24 h. Nematode attraction to hosts was strongly affected by nematode species but the effect of insect species varied with nematode species. S. glaseri had a high innate dispersal rate (i.e., in absence of insects) and was strongly attracted to insects without significant differences among insect species. S. scarabaei had a very low innate dispersal rate so that even a strong relative response to insects resulted in low absolute dispersal rates toward insects. S. scarabaei tended to be most attracted to G. mellonella and least attracted to C. borealis. H. zealandica had a high innate dispersal rate but only responded weakly to insects without significant differences among species. H. bacteriophora had limited innate dispersal and only weakly responded to insects with G. mellonella tending to be the most attractive and C. borealis the least attractive insect. It has to be noted that we cannot exclude that the use of different rearing hosts (A. orientalis and P. japonica larvae for S. scarabaei, G. mellonella larvae for the other nematodes) might have had an impact on the nematodes dispersal and relative attraction behavior. This study indicates that host attractiveness and nematode dispersal rates may contribute but do not play a major role in the variability in white grub susceptibility and/or nematode virulence.  相似文献
