The cell cycling characteristics of the regions of the apicalmeristems of underground shoots and buds of Solanum tuberosumL. were investigated by stathmokinesis and labelling. The apicaldomes of orthotropic shoots produce cells at twice the elementalrate of those of stolons, and their youngest leaf primordiaat twelve times the rate. Changing the fate of stolons so thatthey will become orthotropic by decapitating the tuber sproutsthat bear them results within 24 h in a general increase incell production especially in the leaf primordia. Axillary buds on tuber sprouts induced to become precursorsof orthotropic shoots instead of stolons undergo a spectacularincrease in cell division within 24 h in all regions, especiallyin the primordia and bud anlagen where the rate increases 20-foldor more. The summit is slower to react than other regions, but,by 24 h, its rate of cell division increases 11-fold and itis contributing 14 cells per day to the flanks from its 80 cells. In all the axillary buds the rate of mitosis in the summit ishalf that of the flanks of the apical dome, but, in both stolonsand orthotropic underground shoots, the rate is higher in thesummit than in the flanks or rib meristem. Relation between currents and the growth of Palaeocene reef-mounds   总被引:1,自引:0,他引:1  
The Lower Palaeocene (Danian) reef-mounds of southern Scandinavia are asymmetrical with steep SE flanks and more gently sloping NW flanks. Fragments of mainly arborescent bryozoans make up between 23 and 45% by weight of the sediment. The asymmetrical development of the mounds is a result of differences in the accumulation rate of bryozoans which was controlled by the growth and density of the fauna. Density was relatively high on the SE flank and low on the NW flank. The morphology of individual bryozoan species also changes across the mound. Colonics living on the summit and the SE flank were more robust than colonics living on the NW flank and in the basins. The patterns of morphological and distributional changes are compared with the current regime of the mound reconstructed on the basis of model experiments in a flume. The experiments indicate that the current velocity was highest on the summit and lowest in the basins. Velocities on the flanks were intermediate but considerably higher on the upcurrent SE flank than on the downcurrent NW flank. The robustness of the colonics is positively correlated with the estimated current velocities, whereas the density of the fauna was highest in areas of relatively moderate velocities. The cell-doubling times in the shoot apical regions of 4-dayold rice plants have been measured using colchicine-inducedmetaphase accumulation. Plants were grown submerged at a constantillumination of 10 000 lx and constant temperature of 30°C.Colchicine was applied by adding the drug to the surroundingwater which therefore involved no other disturbance of the apexor the plant. Measurement of summit metabolism (the maximum rate of heat production) in lambs aged 1 or 4h revealed considerable between animal variation. Summit metabolism per unit body weight decreased as body weight increased whereas summit metabolism per unit body surface area was independent of body weight. Severe pre-partum hypoxia was apparently associated with a low summit metabolism at 1 or 4h of age which made such lambs very susceptible to hypothermia. This deficiency in heat production capacity did not appear to be a permanent featuresince most lambs so affected recovered full thermoregulatory ability by 12h of age. Feeding of colostrum conferred an immediate 18% increase in summit metabolism. The gravistimulation of primary roots of seedlings of Phaseolus vulgaris for 0.5 h did not cause any conspicuous difference in the microtubule arrays between the upper and the lower flanks. Exposure to gravistimulation of 1 h resulted in the transversely-orientated microtubules being reduced significantly in the lower flank of the actively elongating region, especially in the epidermis and the outer first and second layers of the cortex. This reduction was compensated for by an increase of randomly-orientated microtubules. In the region where the elongation rate of cells began to decrease, the microtubule arrays did not show a distinct difference between the two opposite flanks. When gravistimulation was prolonged to 2 h, the microtubule arrangement was about the same as in the control. The present results are not compatible with the concept that longitudinal microtubule arrays are induced in the lower flank under gravistimulation and thereby hinder the longitudinal expansion of cells in the lower flanks of roots.  相似文献

Barometric pressures (PB) near the summit of Mt. Everest (altitude 8, 848 m) are of great physiological interest because the partial pressure of oxygen is very near the limit for human survival. Until recently, the only direct measurement on the summit was 253 Torr, which was obtained in October 1981, but, despite being only one data point, this value has been used by several investigators. Recently, two new studies were carried out. In May 1997, another direct measurement on the summit was within approximately 1 Torr of 253 Torr, and meteorologic data recorded at the same time from weather balloons also agreed closely. In the summer of 1998, over 2,000 measurements were transmitted from a barometer placed on the South Col (altitude 7,986 m). The mean PB values during May, June, July, and August were 284, 285, 286, and 287 Torr, respectively, and there was close agreement with the PB-altitude (h) relationship determined from the 1981 data. The PB values are well predicted from the equation PB = exp (6.63268 - 0.1112 h - 0.00149 h2), where h is in kilometers. The conclusion is that on days when the mountain is usually climbed, during May and October, the summit pressure is 251-253 Torr.  相似文献

Seamounts and their influence on the surrounding environment are currently being extensively debated but, surprisingly, scant information is available for the Mediterranean area. Furthermore, although the deep Tyrrhenian Sea is characterised by a complex bottom morphology and peculiar hydrodynamic features, which would suggest a variable influence on the benthic domain, few studies have been carried out there, especially for soft-bottom macrofaunal assemblages. In order to fill this gap, the structure of the meio-and macrofaunal assemblages of the Vercelli Seamount and the surrounding deep area (northern Tyrrhenian Sea – western Mediterranean) were studied in relation to environmental features. Sediment was collected with a box-corer from the seamount summit and flanks and at two far-field sites in spring 2009, in order to analyse the metazoan communities, the sediment texture and the sedimentary organic matter. At the summit station, the heterogeneity of the habitat, the shallowness of the site and the higher trophic supply (water column phytopigments and macroalgal detritus, for instance) supported a very rich macrofaunal community, with high abundance, biomass and diversity. In fact, its trophic features resembled those observed in coastal environments next to seagrass meadows. At the flank and far-field stations, sediment heterogeneity and depth especially influenced the meiofaunal distribution. From a trophic point of view, the low content of the valuable sedimentary proteins that was found confirmed the general oligotrophy of the Tyrrhenian Sea, and exerted a limiting influence on the abundance and biomass of the assemblages. In this scenario, the rather refractory sedimentary carbohydrates became a food source for metazoans, which increased their abundance and biomass at the stations where the hydrolytic-enzyme-mediated turnover of carbohydrates was faster, highlighting high lability.  相似文献

We used live cell imaging to compare the fate of human nontransformed (RPE-1) and cancer (HeLa, U2OS) cells as they entered mitosis in nocodazole or taxol. In the same field, and in either drug, a cell in all lines could die in mitosis, exit mitosis and die within 10 h, or exit mitosis and survive > or =10 h. Relative to RPE-1 cells, significantly fewer HeLa or U2OS cells survived mitosis or remained viable after mitosis: in nocodazole concentrations that inhibit spindle microtubule assembly, or in 500 nM taxol, 30% and 27% of RPE-1 cells, respectively, died in or within 10 h of exiting mitosis while 90% and 49% of U2OS and 78% and 81% of HeLa died. This was even true for clinically relevant taxol concentrations (5 nM) which killed 93% and 46%, respectively, of HeLa and U2OS cells in mitosis or within 10 h of escaping mitosis, compared to 1% of RPE-1 cells. Together these data imply that studies using HeLa or U2OS cells, harvested after a prolonged block in mitosis with nocodazole or taxol, are significantly contaminated with dead or dying cells. We also found that the relationship between the duration of mitosis and survival is drug and cell type specific and that lethality is related to the cell type and drug used to prevent satisfaction of the kinetochore attachment checkpoint. Finally, work with a pan-caspase inhibitor suggests that the primary apoptotic pathway triggered by nocodazole during mitosis in RPE-1 cells is not active in U2OS cells. Cell Motil. The intronic sequences flanking exon-intron junctions (i.e., exon flanks) are important for splice site recognition and pre-mRNA splicing. Recent studies show a higher degree of sequence conservation at flanks of alternative exons, compared to flanks of constitutive exons. In this article we performed a detailed analysis on the evolutionary divergence of exon flanks between human and chimpanzee, aiming to dissect the impact of mutability and selection on their evolution. Inside exon flanks, sites that might reside in ancestral CpG dinucleotides evolved significantly faster than sites outside of ancestral CpG dinucleotides. This result reflects a systematic variation of mutation rates (mutability) at exon flanks, depending on the local CpG contexts. Remarkably, we observed a significant reduction of the nucleotide substitution rate in flanks of alternatively spliced exons, independent of the site-by-site variation in mutability due to different CpG contexts. Our data provide concrete evidence for increased purifying selection at exon flanks associated with regulation of alternative splicing.  相似文献

ABSTRACT. Mitosis and cytokinesis in Katablepharis ovalis , a colorless flagellate, was investigated. Two new flagella are produced prior to prophase, resulting in a motile quadriflagellate cell during mitosis. the inner array of microtubules of the feeding apparatus disappears before prophase begins. the nuclear envelope disperses during prophase, apparently being converted into rough endoplasmic reticulum. the chromatin condenses and the nucleolus disperses with spindle microtubules appearing oriented perpendicular to the longitudinal axis of the cell. At metaphase, the chromatin is condensed as a single disc-shaped mass and rough endoplasmic reticulum flanks the chromatin mass on each side. Groups of spindle microtubules pass through tunnels in the rough endoplasmic reticulum and through electron-translucent areas of the chromatin. the spindle microtubules end at a number of minipoles in the cytoplasm. Vesicles, ribosomes, mitochondria and endoplasmic reticulum migrate among the spindle microtubules. There is no polar body or any electrondense area associated with the spindle poles. the basal bodies of the flagella remain attached to the axonemes and do not participate in mitosis. In anaphase, the chromatin separates and migrates to the poles. During telophase, the nuclear envelope reforms from the rough endoplasmic reticulum and the nucleoli reappear. the spindle microtubules are persistent during telophase. Cytokinesis occurs by longitudinal fission, starting at the anterior end and progressing posteriorly. Cytokinesis may be driven by elongation of the spindle microtubules since there is no visible structure associated with the furrowing.  相似文献

The reconstruction of the nucleolus after mitosis was analyzed by electron microscopy in cultured mammalian (L929) cells in which nucleolar RNA synthesis was inhibited for a 3 h period either after or before mitosis. When synchronized mitotic cells were plated into a concentration of actinomycin D sufficient to block nucleolar RNA synthesis preferentially, nucleoli were formed at telophase as usual. 3 h after mitosis, these nucleoli had fibrillar and particulate components and possessed the segregated appearance characteristic of nucleoli of actinomycin D-treated cells. Cells in which actinomycin D was present for the last 3 h preceding mitosis did not form nucleoli by 3 h after mitosis though small fibrillar prenucleolar bodies were detectable at this time. These bodies subsequently grew in size and eventually acquired a particulate component. It took about a full cell cycle before nucleoli of these cells were completely normal in appearance. Thus, nucleolar RNA synthesis after mitosis is not necessary for organization of nucleoli after mitosis. However, inhibition of nucleolar RNA synthesis before mitosis renders the cell incapable of forming nucleoli immediately after mitosis. If cells are permitted to resume RNA synthesis after mitosis, they eventually regain nucleoli of normal morphology.  相似文献

Background  

Climbers on the summit of Mt. Everest are exposed to extreme hypoxia, and the physiological implications are of great interest. Inferences have been made from alveolar gas samples collected on the summit, but arterial blood samples would give critical information. We propose a plan to insert an arterial catheter at an altitude of 8000 m, take blood samples above this using an automatic sampler, store the samples in glass syringes in an ice-water slurry, and analyze them lower on the mountain 4 to 6 hours later.  相似文献

Cycloheximide pulses applied to Physarum polycephalum surface plasmodia delay mitosis. Pulses applied in G2 cause a delay of mitosis which is linearly dependent on the phase in the cell cycle at which the pulse is applied. A 30 min pulse of 10 micrograms/ml cycloheximide starting in G2 at time t after mitosis induces an excess delay (delay in excess of pulse duration) of the next mitosis of (0.55) t-1.3 h. The excess delays induced by 7 h pulses during G2 are at most 1 h larger. Pulses applied less than 30 min before mitosis induce only small delays.  相似文献

The ontogeny of the translator is analysed inFockea edulis, Pachycarpus dealbatus, andCalotropis procera. The three species show a similar basic pattern in the construction of the corpusculum. Each side wall consists of two originally separate flanks. In the primitive state (Fockea), these flanks remain separate throughout their development, only the two inner flanks fuse along their base. In the derived state, the two flanks on each side fuse to form solid side walls, and the corpusculum develops a floor by additional secretion. The primitive state of divided flanks is recapitulated in early ontogeny. InFockea, the pollinia are attached apically and dorsally to the outer flanks by means of unmoulded adhesive material; in more advanced translators, caudicles are inserted as new elements between the outer flanks and the pollinia. The caudicles are attached to the lower part of the corpusculum. Within the four tribes of the family, many species have retained parts of the primitive structure in their corpuscula. Flanks divided in the lower part of the corpusculum and caudicles attached dorsally to the outer flanks seem to be common inMarsdenieae andStapelieae. InAsclepiadeae andGonolobeae the side walls are solid throughout the length of the corpusculum. — The question of cellular mechanisms underlying the formation of the translator is discussed. In all probability, the main factor responsible for the complex structure of the corpusculum is a differential secretory activity of the glandular field. This, in turn, points to differential genetic activity as a regulating factor of the ontogeny of the translator.  相似文献

In S. cerevisiae, the mitotic exit network (MEN) proteins, including the Polo-like protein kinase Cdc5 and the protein phosphatase Cdc14, are required for exit from mitosis. In pre-anaphase cells, Cdc14 is sequestered to the nucleolus by Net1 as a part of the RENT complex. We examined the effects of thermal injury on intestinal epithelial cell proliferation and death. We recorded histologically identifiable mitotic and apoptotic crypt cells in relation to cell position after a 60% full thickness cutaneous thermal injury in the rat. The injury significantly reduced mitosis (0.53 +/- 0.11 vs. 1. 50 +/- 0.70, P < 0.05) at cell positions 4-6, stem cells, 6 h after injury. A similar reduction in mitosis (1.13 +/- 0.59 vs. 3.50 +/- 0. 80, P < 0.05) was observed at higher cell positions 7-9 12 h after injury, indicating a positional cell shift. In addition, a significant increase in the number of apoptotic bodies occurred at cell positions 7-9 (2.32 +/- 0.87 vs. 0.13 +/- 0.22, P < 0.05) and 10-12 (2.2 +/- 0.12 vs. 0.00, P < 0.05) 6 h after injury. Thermal injury-induced alterations in mitotic and apoptotic activities were transient since crypts recovered with a moderate increase in mitotic activity 24 h after injury. In control and thermal-injury rats 24 h after injury, crypt cell mitosis and apoptosis did not differ significantly. This demonstrates that cutaneous thermal injury causes a transient suppression of mitosis as well as induction of apoptosis in a cell position-dependent manner in the small intestinal crypt.  相似文献

Summary. Division and partitioning of microbodies (peroxisomes) of the green alga Klebsormidium flaccidum, whose cells contain a single microbody, were investigated by electron microscopy. In interphase, the rod-shaped microbody is present between the nucleus and the single chloroplast, oriented perpendicular to the pole-to-pole direction of the future spindle. A centriole pair associates with one distal end of the microbody. In prophase, the microbody changes not only in shape, from a rodlike to a branched form, but also in orientation, from perpendicular to parallel to the future pole-to-pole direction. Duplicated centriole pairs are localized in close proximity to both distal ends of the microbody. In metaphase, the elongated microbody flanks the open spindle, with both distal ends close to the centriole pair at either spindle pole. The microbody further elongates in telophase and divides after septum formation (cytokinesis) has started. The association between the centrioles and both distal ends of the microbody is maintained throughout mitosis, resulting in the distal ends of the elongated microbody being fixed at the cellular poles. This configuration of the microbody may be favorable for faithful transmission of the organelle during cell division. After cytokinesis is completed, the microbody reverts to the perpendicular orientation by changing its shape. Microtubules radiating from the centrosomes flank the side of the microbody throughout mitosis. The close association of centrosomes and microtubules with the microbody is discussed in respect to the partitioning of the microbody in this alga. Correspondence: H. KORN, R., 1993. Heterogeneous growth of plant tissues. Heterogeneous growth is defined as different rates or patterns of growth in adjacent tissue regions, in contrast to homogeneous growth where a region expresses a uniform rate or pattern of growth. Heterogeneous growth is inspected in a variety of plant tissues and the pattern of expansion is characterized for each. In the case of epidermal cell proliferation, different growth rates for cell plates and old walls lead to the feature of coordinated growth in which slow growth of the former is compensated for by a faster rate of the latter. Examples include leaf epidermal cells above veins growing differently from those above areole regions, and pairs of guard cells of stomata ceasing to expand before other epidermal cells. In the alga Coleochaete only marginal walls grow, and at different rates around the colony, to generate a fractal, stochastic type of coordinated growth. In the fern gametophyte there are complex gradients of differential growth rates. Epidermal cells of apices are often of mixed growth, as cells at the summit undergo two dimensional expansion while cells along the flanks express one dimensional expansion. Coordinated growth requires matched rates where the constraining effect of the slower growing region is compensated for by a faster rate in an encircling region compared to the average rate of the overall tissue. Mixed and differential growth patterns do not necessarily create constraints and so lead to smooth tissue expansion. Emergence of some constraints leads to breaking of symmetry and disruptive growth as in the appearance of new axes found in organs and epidermal derivatives. In planar development heterogeneous growth appears to be the rule, and homogeneous growth the exception.  相似文献

Numerous areas of granulomatous inflammation develop in the lungs of rabbits following the intravenous injection of Freund's complete adjuvant (FCA). Within a few days after FCA injection, hyperplasia of type B (type I) alveolar cells is present on the surface of the septa in which an inflammatory reaction is developing. Mitosis of type B cells is detected 12 h after FCA injection and is common over the next 120 h. In addition, there are morphologic changes that are consistent with migration of these cells. The type B cells in mitosis extend across alveolar septa as well as along the alveolar surface. The extension of type B cells through alveolar septa is not limited to cells in mitosis, but is also observed in non-mitotic type B cells. Stimulation of mitosis and hyperplasia of type B cells is discussed in relation to the focal tissue injury and inflammatory response.  相似文献
