To investigate the changes in the morphology and polysaccharide content of Microcystis aeruginosa (Kütz.) Kütz. during flagellate grazing, cultures of M. aeruginosa were exposed to grazing Ochromonas sp. for a period of 9 d under controlled laboratory conditions. M. aeruginosa responded actively to flagellate grazing and formed colonies, most of which were made up of several or dozens of cells, suggesting that flagellate grazing may be one of the biotic factors responsible for colony formation in M. aeruginosa. When colonies were formed, the cell surface ultrastructure changed, and the polysaccharide layer on the surface of the cell wall became thicker. This change indicated that synthesis and secretion of extracellular polysaccharide (EPS) of M. aeruginosa cells increased under flagellate grazing pressure. The contents of soluble extracellular polysaccharide (sEPS), bound extracellular polysaccharide (bEPS), and total polysaccharide (TPS) in colonial cells of M. aeruginosa increased significantly compared with those in single cells. This finding suggested that the increased amount of EPS on the cell surface may play a role in keeping M. aeruginosa cells together to form colonies.  相似文献

This study assessed the implication of oxidative stress in the mortality of cells of Microcystis aeruginosa Kütz. Cultures grown at 25°C were exposed to 32°C, darkness, and hydrogen peroxide (0.5 mM) for 96 h. The cellular abundance, chl a concentration and content, maximum photochemical efficiency of PSII (Fv/Fm ratio), intracellular oxidative stress (determined with dihydrorhodamine 123 [DHR]), cell mortality (revealed by SYTOX‐labeling of DNA), and activation of caspase 3–like proteins were assessed every 24 h. The presence of DNA degradation in cells of M. aeruginosa was also assessed using a terminal deoxynucletidyl transferase‐mediated dUTP nick end labeling (TUNEL) assay at 96 h. Transferring cultures from 25°C to 32°C was generally beneficial to the cells. The cellular abundance and chl a concentration increased, and the mortality remained low (except for a transient burst at 72 h) as did the oxidative stress. In darkness, cells did not divide, and the Fv/Fm continuously decreased with time. The slow increase in intracellular oxidative stress coincided with the activation of caspase 3–like proteins and a 15% and 17% increase in mortality and TUNEL‐positive cells, respectively. Exposure to hydrogen peroxide had the most detrimental effect on cells as growth ceased and the Fv/Fm declined to near zero in less than 24 h. The 2‐fold increase in oxidative stress matched the activation of caspase 3–like proteins and a 40% and 37% increase in mortality and TUNEL‐positive cells, respectively. These results demonstrate the implication of oxidative stress in the stress response and mortality of M. aeruginosa.  相似文献

The overall appearance of the flagellar apparatus in the isogametes of Batophora oerstedii. J. Ag. is most like that which occurs in motile cells of the Ulvophyceae. Like other Ulvophyceae, the basal bodies overlap and are arranged in the 11/5 configuration, microtubular roots are arranged in a cruciate pattern and system II striated fibers are present. The basal body connective which generally lacks striation in the Ulvophyceae is clearly different in Batophora, being composed of two large non-striated halves which connect to the anterior surface of each basal body and are then connected to one another by a distinctly fibrous centrally striated region. This variation in the basal body connective and the presence of two posteriorly directed system II striated fibers is clearly different from homologous structures reported in siphonous green algae of the Caulerpales. Based upon these variations and similarities among flagellar apparatus components in siphonous green algae, it is suggested that the Dasycladales and Siphonodadales are more closely related to one another than to the Caulerpales.  相似文献

Over the course of one year the volatile organic biogenic substances (VOBS) found in the water of a eutrophied shallow lake were determined by gas chromatographic and mass spectrometric methods. The substances detected belonged to nor-carotenoids, terpenoids, unsaturated hydrocarbons, ketones, and aldehydes. The occurrence of particular VOBS correlated with the frequency with which certain phytoplankton species were recorded. The latter showed a well developed succession in that year. The following correspondences were found: β-cyclocitral and Microcystis; heptadec-cis 5-ene and Oscillatoria redekei; geosmin and argosmin and Aphanizomenon gracile; hepta-trans, cis 2,4-dienal, deca-trans, cis 2,4-dienal and Dinobryon; octa-trans, cis 1,3,5-triene and Asterionella formosa. Dictyopterenes and ectocarpene were only detected in high amounts in one sample obtained in August. The VOBS exhibited marked dynamics in the lake and usually were rapidly eliminated from the water body.  相似文献

Progress in the study of stable isotope discrimination in carbon assimilation by aquatic macrophytes has been slower than for other groups of primary producers, such as phytoplankton and terrestrial plants. A probable reason has been the methodologies employed for such a study: field collections or long‐term incubations, both relying on the observation of changes in carbon isotope composition of plant tissue. Here, we present a short‐term incubation method based on the change in carbon stable isotope composition in water. Its fundamental advantage over the other approaches is that the change in stable isotope composition in water in a closed system is much faster than in the plant tissue. We applied the method to investigate the relationship between carbon assimilation intensity and isotope discrimination. The results included a relatively small discrimination in respiration, a significant influence of carbon assimilation rate on discrimination, and the suggestion of HCO3? or CO2 uptake in photosynthesis. The information gathered using this method would be difficult to obtain in other ways, and so we believe that it should contribute to a better understanding of the physiology and ecology of aquatic macrophytes.  相似文献

As assayed by fluorescent reporter dyes, nitric oxide (NO) and H2O2, two downstream signaling agents induced by wounding in the alga Dasycladus vermicularis (Scop.) Krasser, can also be induced in unwounded Dasycladus cells by μM Adenosine 5′[γ‐thio]triphosphate (ATPγS) and Adenosine 5′‐[β‐thio]diphosphate (ADPβS), but not by Adenosine 5′‐O‐thiomonophosphate (AMPS). These nucleotide‐induced responses are blocked by pyridoxalphosphate‐6‐azophenyl‐2′,4′‐disulphonic acid (PPADS), an antagonist of animal purinoceptors, and by adenosine, a feed‐back inhibitor of extracellular nucleotide responses in animals. Similar nucleotide‐ and nucleotide‐antagonist responses were observed in Acetabularia acetabulum (L.) P. C. Silva. Significant levels of ATP released from Dasycladus cells were measured at wound sites by a sensitive luciferin‐luciferase assay. Additionally, the normal wound‐induced production of NO and H2O2 in Dasycladus can be blocked by pretreating the cells with PPADS. Our results indicate that nucleotides released from wounds can serve as a signal to trigger wound responses in algae, and that coordinated signaling between extracellular nucleotides and the NO pathway may have been established early during the evolution of plants.  相似文献

An electrophoretic survey of 12 new isolates of Thalassiosira pseudonana Hasle & Heimdal and 25 new isolates of Skeletonema costatum (Grev.) Cleve revealed several heterozygote genotypes at malate dehydrogenase (MDH) and phosphohexose isomerase (PHI) loci. The new clones were maintained in culture for 6 mo and then reasayed at these two loci. All MDH heterozygotes and halj of the PHI heterozygotes had become homozygous. This resulted in a collection of clones that are largely homozygous but that are samples of polymorphic species. The physlogical implications of this loss of heterozygosity in clonal cultures has not been analyzed. Hawever, any change in a clone that is the result of culturing conditions reduces the usefulness of that clone as a laboratory test organism for ecological correlations.  相似文献

The leaffooted bug, Leptoglossus zonatus (Hemiptera: Coreidae) is an emerging pest of several crops around the World and up to now very little is known of its digestive system. In this article, glycoside hydrolase (carbohydrase) activities in the adult midgut cells and in the luminal contents of L. zonatus adult females were studied. The results showed the distribution of digestive carbohydrases in adults of this heteropteran species in the different intestinal compartments. Determination of the spatial distribution of α‐glucosidase activity in L. zonatus midgut showed only one major molecular form, which was not equally distributed between soluble and membrane‐bound isoforms, being more abundant as a membrane‐bound enzyme. The majority of digestive carbohydrases were found in the soluble fractions. Activities against starch, maltose and the synthetic substrate NPαGlu were found to show the highest levels of activity, followed by enzymes active against galactosyl oligosaccharides. Based on ion‐exchange chromatography elution profiles and banding patterns in mildly denaturing electrophoresis, both midgut α‐amylases and α‐galactosidases showed at least two isoforms. The data suggested that the majority of carbohydrases involved in initial digestion were present in the midgut lumen, whereas final digestion of starch and of galactosyl oligosaccharides takes place partially within the lumen and partially at the cell surface. The complex of carbohydrases here described was qualitatively appropriate for the digestion of free oligosaccharides and oligomaltodextrins released by α‐amylases acting on maize seed starch granules.  相似文献
