Molecular cloning and characterization of human papilloma virus DNA derived from a laryngeal papilloma 总被引:22,自引:17,他引:22 下载免费PDF全文
Papilloma virus DNA from a laryngeal papilloma was cloned in phage lambda L 47 and characterized after cleavage with different restriction enzymes. Hybridization with the DNAs of human papilloma virus types 1, 2, 3, 4, 5, and 8 showed no homology under stringent hybridization conditions. Human papilloma virus type 6 DNA, however, was partially identical to laryngeal papilloma virus DNA; different restriction enzyme fragments hybridizing with the other DNA were identified on each genome. The degree of homology was determined by reassociation kinetics to be 25%. According to the present nomenclature, laryngeal papilloma virus therefore represents a different type of human papilloma virus and is tentatively designated as human papilloma virus type 11. Sequences homologous to laryngeal papilloma virus DNA were also found in four of nine additional laryngeal papillomas. Attempt to detect homologous DNA in 12 carcinomas of the larynx were negative. 相似文献

In 1997, cutaneous papillomatosis caused by Florida manatee (Trichechus manatus latirostris [Tm]) papillomavirus 1 (TmPV1) was detected in seven captive manatees at the Homosassa Springs Wildlife State Park, Florida, USA, and, subsequently, in two wild manatees from the adjacent Homosassa River. Since then, papillomatosis has been reported in captive manatees housed in other locations, but not in wild animals. To determine TmPV1 antibody prevalence in captive and wild manatees sampled at various locations throughout Florida coastal regions, virus-like particles, composed of the L1 capsid protein of TmPV1, were generated with a baculovirus expression system and used to measure anti-TmPV1 antibodies in an enzyme-linked immunosorbent assay. Serologic analysis of 156 manatees revealed a TmPV1 antibody prevalence of 26.3%, with no significant difference between captive (n=39) and wild (n=117) manatees (28.2% and 25.6%, respectively). No antibody-positive wild animal showed PV-induced cutaneous lesions, whereas papillomatosis was observed in 72.7% of antibody-positive captive manatees. Our data indicate that Florida manatees living in the wild are naturally infected by TmPV1 but rarely show TmPV1-induced papillomatosis. Hence, it appears that the wild population would not be harmed in a case of contact with captive animals without visible lesions and productive infections, which could be thus released into the wild. 相似文献

The paper addresses the effect of captivity on body size in Macaca mulatta, comparing a captive with the free-ranging population from which it was derived. The captive population, which was housed socially, was smaller in most linear measures and in body mass, particularly in young up to 13 month old animals. Nutritional differences in the diet may be at least partially responsible; the captive population was fed a lower level of total protein. Nasal secretions, leukocytes and preputial or vaginal swabs from a group of 15 captive wild ruminants, comprising six pronghorn antelope (Antilocapra americana), seven fallow deer (Dama dama) and two mule deer (Odocoileus hemionus), and from 50 free-ranging pronghorns in southern Alberta, were examined for viral agents. Captive animals were given injections of dexamethasone daily for 6 days in attempts to reactivate latent infections. Specimens were collected at 2-3 day intervals from days 0 to 18. Free-ranging pronghorns were sampled only once, at the time of capture. Fifteen viral isolates were obtained from the animals: six isolates of parainfluenza 3 (PI3) from nasal swabs from one fallow deer and one mule deer; five isolates of herpesvirus from leukocytes, vaginal, preputial and nasal swabs from three fallow deer; and four isolates of PI3 from nasal secretions of the 50 free-ranging pronghorns. 相似文献

Records from 42 zoos and from long-term studies of wild populations were analysed to describe the reproductive biology of spider monkeys (Ateles spp.). Both data sets suggested that spider monkey females typically have their first infant between 7 and 8 years of age with an interbirth interval of approximately 32–36 months. Infant sex ratio for zoo populations was approximately 1 male to 1 female; infant sex ratios from wild populations were variable. Zoo records provided adequate sample size to suggest that interbirth interval was not influenced by the sex of the infant produced, and that the sex ratio and the probability of infant survival did not change with the number of infants the mother had produced. The findings of this study have implications with respect to the conservation of New World primate species. Since spider monkeys take a long time to reach sexual maturity and their interbirth interval is longer than that expected based on their body size, their populations may be slow to recover following disturbances. Thus, particular care should be taken for the protection of these species. 相似文献

The mesowear method evaluates the wear patterns of herbivore cheek teeth by visually evaluating the facet development of the occlusal surfaces. It thus allows classification of most herbivorous ungulates into browsers, grazers or intermediate feeders, due to the fact that in grazers, tooth wear is characterized by a comparatively high degree of abrasion, most probably due to the presence of silicacious phytoliths in grasses, a higher amount of dust and grit adhering to their forage, or both. It has been suggested that excessive tooth wear could be a particularly limiting factor in the husbandry of captive large browsing species, and major tooth wear was demonstrated in captive as compared to free-ranging giraffe. If this increased tooth wear in captivity was an effect of feeding type and diets fed, then it would be expected that other browsing species are affected in a similar manner. In order to test this hypothesis, we investigated the dental mesowear pattern in captive individuals of 19 ruminant species and compared the results to data on free-ranging animals. Compared to free-ranging populations, captive browsers show a significantly more abrasion-dominated tooth wear signal. The reverse applies to captive grazers, which tend to show a less abrasion-dominated wear in captivity. Captive ruminants were generally more homogenous in their wear signature than free-ranging ruminants. If grit contamination in the natural habitat is a major cause of dental wear in grazers, then diets in captivity, although similar in botanical composition, most likely contain less abrasives due to feeding hygiene. If dental wear is one of the major factors limiting longevity, then captive grazers should achieve longer lifespans than both captive browsers and free-ranging grazers. In particular with respect to browsers, the results suggest that captive feeding regimes could be improved. 相似文献

Toxoplasma gondii is a significant worldwide parasitic protozoan. In the present study, prevalence of antibodies of T. gondii was examined from 29 free-ranging black bears ( Ursus americanus ) from south-central Florida where the host species was listed as state threatened during this project. Overall T. gondii prevalence was found to be 44.8%, specifically 46.2% in male and 43.8% in female U. americanus , using a modified agglutination test (1:25 titer). Seroprevalence differences between sexes were not significant (P > 0.05). Results of the present study add supportive data to the growing body of evidence suggesting that U. americanus has one of the highest T. gondii seroprevalences among all known intermediate hosts. In addition, our data emphasize the importance of understanding parasitic disease dynamics from a conservation perspective. 相似文献

Given the expense and time required to monitor marine mammal populations effectively, approaches that fully exploit the resulting data certainly are warranted. We employed a two‐step modeling approach to estimate key demographic parameters, including immigration, from aerial surveys of manatees (Trichechus manatus latirostris) in the Northwest management unit of Florida. Abundances of adults and calves were predicted by multivariate adaptive regression spline (MARS) models, after accounting for heterogeneous detection rates caused by variable environmental conditions. The resulting predictions were incorporated into a stage‐structured, deterministic model that used an inverse method to estimate parameters with and without immigration. The model without immigration estimated mean survival probabilities of 0.966, 0.923, and 0.794 for adults, subadults and calves, respectively, with a per capita reproductive rate of 0.135. These parameter estimates yielded an overall mean population growth rate of approximately 1.037, which is comparable to rates from mark‐recapture studies. When we added an immigration term that accounted for the greater slope in adult counts since 1999, as identified by the MARS model, the estimated per capita reproductive rate was 0.122, with survival probabilities for adults, subadults and calves of 0.926, 0.920, and 0.833, respectively. These rates were coupled with an estimated mean winter immigration rate corresponding to roughly 5.2% of the adult and subadult population. In this latter scenario, the number of manatees in the core population of the Northwest management unit was predicted to remain constant, with a population growth rate near one, and additional manatees counted during aerial surveys were deemed to be immigrants. While further studies could certainly expound on the potential effects of migrants on population indices, we present this first published immigration estimate for wintering manatees in northwest Florida. 相似文献

Five free-ranging mink, Neovison vison mink, from in or near the Fakahatchee Strand Preserve State Park, Collier County, Florida (26 degrees 00'N, 81 degrees 25'W), were examined for parasitic helminths. Nine species of helminths were identified (2 trematodes, 5 nematodes, and 2 acanthocephalans). The most prevalent parasites were Molineus patens (4 of 5 mink), mean intensity 173 (range, 12-342); Strongyloides sp. (4 of 5), mean intensity 48 (range, 1-170); Macracanthorhynchus ingens (immature) (4 of 5), mean intensity 2.5 (range, 1-4); and Alaria mustelae (3 of 5), mean intensity 59.3 (range, 11-127). Polymorphus brevis and Dirofilaria lutrae are reported from the mink for the first time. 相似文献

Several disease syndromes in captive rhinoceroses have been linked to low vitamin status. Blood samples from captive and free-ranging black (Diceros bicornis) and white rhinoceros (Ceratotherium simum) and tissue samples of captive individuals from four rhinoceros species were analysed for vitamins A and E. Circulating vitamin A levels measured as retinol for free-ranging versus captive black and white rhinoceros were 0.04 (+/- 0.03 SD) vs. 0.08 (+/- 0.08) and 0.07 (+/- 0.04) vs. 0.06 (+/- 0.02) microgram/ml, respectively. Circulating vitamin E levels measured as alpha-tocopherol were 0.58 (+/- 0.30) vs. 0.84 (+/- 0.96) and 0.62 (+/- 0.48) vs. 0.77 (+/- 0.32) microgram/ml, respectively. In contrast to earlier findings, there was no significant difference in vitamin E concentration between captive and free-ranging black rhinoceros. When the samples of captive black rhinoceros were grouped into those taken before 1990 and after 1990, however, those collected before 1990 had significantly lower (P < 0.001) vitamin E levels (0.46 +/- 0.83 microgram/ml) and those collected in 1990 or later significantly higher (P < 0.001) vitamin E levels (1.03 +/- 1.04 micrograms/ml) than the captive population as a whole. This is probably due to increased dietary supplementation. There were significant differences in circulating vitamin concentrations in black rhinoceroses from different regions in the wild. Serum 25-hydroxy (OH) vitamin D3 averaged 55.7 ng/ml in free-ranging rhinoceroses; no carotenoids were detected in any blood samples. Captive black and white rhinoceroses appear to be adequately supplemented in vitamin A and E. Captive Indian rhinoceroses (Rhinoceros unicornis) had significantly lower vitamin A concentrations in blood (P < 0.001) and higher vitamin A concentrations in liver tissue samples (P < 0.001) than other rhinoceros species. The present study demonstrated that pathogenic Naegleria, the causative agent, is relatively widespread. Twelve of 26 lakes sampled only once yielded the amoeba. As a means to compare and contrast to free‐ranging Florida manatees, we examined the behavior and movement patterns of nine adult‐female captive manatees at Homosassa Springs Wildlife State Park (HSWSP). HSWSP has a unique manatee exhibit that consists of a natural river and a man‐made pool. Because the manatees are exposed to a natural environment including natural food resources, the study of various aspects of behavioral and physiological parameters of the captive manatees is especially valuable with this group. We determined activity pattern and spatial use of the facility by manatees over three allotted periods of a day (noon, mid‐afternoon, and late‐afternoon) and three seasons (winter, spring, and summer). Behavioral strategies of the manatees were: 1) to remain “inactive,” probably to conserve energy, during the day while the park provisioned food; 2) to alter locations during the course of the afternoon, apparently with the direction of the sun, possibly to assist with thermoregulation; and 3) to change location seasonally where and when food resources became abundant, showing a strong interest in natural vegetation as it became available. Spatial use and activity pattern of the HSWSP captive manatees apparently were influenced by energy constraints and nutrient intake by provisioned food availability over the day and natural vegetation over the study period. The behavior of the female group of manatees may have been affected by the single‐sex living arrangement. In addition, these manatees are subject to a largely invariable temperature (ca. 23°C) that may have created thermal stress and immunological suppression over time. This study demonstrated that for these captive manatees the activity patterns and spatial use were comparable to that of free‐ranging manatees in relation to the availability of food resources. We obtained blood smears from 114 Florida sandhill crane (Grus canadensis pratensis) chicks in Osceola and Lake Counties, Florida, USA, during 1998-2000. Leucocytozoon grusi was observed in 11 (10%) chicks; Haemoproteus antigonis was observed in eight (7%) chicks; and three (3%) chicks were infected with Haemoproteus balearicae. One chick infected with H. balearicae suffered from severe anemia (packed cell volume = 13%) and was later found moribund. At necropsy this bird also had severe anemia and damage to the heart possibly due to hypoxia. This is the first report of H. balearicae in free-ranging North American cranes. 相似文献

The discovery of Meloidogyne mayaguensis is confirmed in Florida; this is the first report for the continental United States. Meloidogyne mayaguensis is a virulent species that can reproduce on host cultivars bred for nematode resistance. The perineal patterns of M. mayaguensis isolates from Florida show morphological variability and often are similar to M. incognita. Useful morphological characters for the separation of M. mayaguensis from M. incognita from Florida are the male stylet length values (smaller for M. mayaguensis than M. incognita) and J2 tail length values (greater for M. mayaguensis than M. incognita). Meloidogyne mayaguensis values for these characters overlap with those of M. arenaria and M. javanica from Florida. Enzyme analyses of Florida M. mayaguensis isolates show two major bands (VS1-S1 phenotype) of esterase activity, and one strong malate dehydrogenase band (Rm 1.4) plus two additional weak bands that migrated close together. Their detection requires larger amounts of homogenates from several females. Amplification of two separate regions of mitochondrial DNA resulted in products of a unique size. PCR primers embedded in the COII and 16S genes produced a product size of 705 bp, and amplification of the 63-bp repeat region resulted in a single product of 322 bp. Nucleotide sequence comparison of these mitochondrial products together with sequence from 18S rDNA and ITS1 from the nuclear genome were nearly identical with the corresponding regions from a M. mayaguensis isolate from Mayaguez, Puerto Rico, the type locality of the species. Meloidogyne mayaguensis reproduced on cotton, pepper, tobacco, and watermelon but not on peanut. Preliminary results indicate the M. mayaguensis isolates from Florida can reproduce on tomato containing the Mi gene. Molecular techniques for the identification of M. mayaguensis will be particularly useful in cases of M. mayaguensis populations mixed with M. arenaria, M. incognita, and M. javanica, which are the most economically important root-knot nematode species in Florida, and especially when low (<25) numbers of specimens of these species are recovered from the soil. 相似文献

The entire genome of human papilloma virus type 1a was cloned in Escherichia coli using the plasmid pBR322 as vector. The integrity and the homogeneity of the viral DNA thus obtained was confirmed by restriction endonucleases analysis. Viral DNA isolated from a single wart was partially methylated at only one out of the four HpaII sites, d(C-C-G-G). Recognition sites for Bg/I, Bg/II, PstI and PvuII restriction endonucleases were located on the cloned genome. 相似文献

Cutaneous nodular lesions caused by parapoxvirus infections are commonly observed in stranded pinnipeds following their arrival at rehabilitation facilities. An indirect enzyme-linked immunosorbent assay (ELISA) was developed and validated to determine exposure to parapoxviruses in California sea lions Zalophus californianus in captivity and in the wild. The diagnostic performance of this assay was evaluated using receiver-operating characteristic analysis. At a selected cut-off value, the calculated sensitivity was 100% (95% CI = 86 to 100%) and the specificity was 100% (95% CI = 87 to 100%). Analysis of sera collected from 26 affected sea lions during various stages of the disease revealed anti-parapoxvirus antibodies in all affected sea lions prior to the development of cutaneous pox lesions. This indicated that previous exposure to a parapoxvirus does not confer protection against clinical disease. In at least 7 cases, exposure to the virus occurred during hospitalization. Analysis of paired sera from 74 unaffected sea lions indicated subclinical infections in at least 3 animals. Finally, the prevalence of anti-parapoxviral antibodies in 761 free-ranging California sea lions captured and tested was 91% (95% CI = 89 to 93%). This indicated that infection with a parapoxvirus is a common occurrence in the wild and that the release of captive sea lions infected with parapoxvirus into the wild should not increase the risk of a parapoxvirus outbreak in free-ranging sea lions. 相似文献

BACKGROUND AND METHODS: To investigate the seroprevalence of polyomavirus infections in macaques, we analyzed 1579 sera from nine different species for antibodies cross-reactive with simian virus 40 (SV40) in an enzyme-linked immunosorbent assay. Most samples were collected from captive animals, but we also investigated a colony of free-ranging Barbary macaques (Macaca sylvanus). RESULTS: High seropositive rates were found in rhesus macaques (Macaca mulatta; 74.7%), cynomolgus macaques (Macaca fascicularis; 44.8%) and Tonkean macaques (Macaca tonkeana; 41.7%), especially in animals imported from China. Low rates were measured in cynomolgus macaques from Mauritius (8.8%), and in Barbary macaques (1.4%). Seropositivity was age-dependent increasing to >70% in animals of 5 years and older. CONCLUSIONS: High seroprevalence rates were found in different species of macaques, dependent on their origin. Very low infection rates found in Barbary macaques and cynomolgus macaques from Mauritius suggest that these animals in the wild are not commonly infected by SV40-like viruses. 相似文献

Iridoviruses of the genus Ranavirus are well known for causing mass mortality events of fish and amphibians with sporadic reports of infection in reptiles. This article describes five instances of Ranavirus infection in chelonians between 2003 and 2005 in Georgia, Florida, New York, and Pennsylvania, USA. Affected species included captive Burmese star tortoises (Geochelone platynota), a free-ranging gopher tortoise (Gopherus polyphemus), free-ranging eastern box turtles (Terrapene carolina carolina), and a Florida box turtle (Terrepene carolina bauri). Evidence for Ranavirus infection was also found in archived material from previously unexplained mass mortality events of eastern box turtles from Georgia in 1991 and from Texas in 1998. Consistent lesions in affected animals included necrotizing stomatitis and/or esophagitis, fibrinous and necrotizing splenitis, and multicentric fibrinoid vasculitis. Intracytoplasmic inclusion bodies were rarely observed in affected tissues. A portion of the major capsid protein (MCP) gene was sequenced from each case in 2003-2005 and found to be identical to each other and to Frog virus 3 (FV3) across 420 base pairs. Ranavirus infections were also documented in sympatric species of amphibians at two locations with infected chelonians. The fragment profiles of HindIII-digested whole genomic DNA of Ranavirus, isolated from a dead Burmese star tortoise and a southern leopard frog (Rana utricularia) found nearby, were similar. The box turtle isolate had a low molecular weight fragment that was not seen in the digestion profiles for the other isolates. These results suggest that certain amphibians and chelonians are infected with a similar virus and that different viruses exist among different chelonians. Amphibians may serve as a reservoir host for susceptible chelonians. This report also demonstrated that significant disease associated with Ranavirus infections are likely more widespread in chelonians than previously suspected. 相似文献
