Genetic variation in six Hungarian common carp (Cyprinus carpio L.) strains was evaluated using 12 microsatellite loci. The domesticated (Tatai, Biharugrai and Szarvasi) strains were derived from fish farms. Two of wild strains (Tiszai and Dunai) were sampled from brood stocks maintained at fish farms for breeding, and Kis-Balatoni wild carp were sampled from the Small Balaton Lake. Pairwise Fst-values (0.013–0.161) were highly significant (p<0.0001), demonstrating differentiation among strains. The mean number of alleles ranged between 3.9 and 8.2. Overall mean observed heterozygosity was lower (0.557) than the mean expected heterozygosity (0.700). By strain, the only exception to this trend was the Dunai (Danubian), which showed higher mean observed heterozygosity (0.764) than expected (0.602). For five loci the Dunai strain showed extremely high levels of heterozygosity (1.00). Two wild strains exhibited a number of loci (Tiszai, 4; Dunai, 6) that were not in Hardy–Weinberg equilibrium. A relatively high number of private alleles overall (n=26), as well as differences in allele frequencies supported our ability to assign most individual fish (over 90%) to each strain.  相似文献

Rhizopus is a zygomycetous genus. Several species of this taxon may infect humans and lower animals. Seventeen isolates ofRhizopus species in three distinct morphological groups were studied: the stolonifer group (sporangiophores greater than 1 mm in height, sporangial diameters of 100–275 µm, branched rhizoids); the arrhizus group (sporangiophores greater than 1 mm in height, branched rhizoids, sporangial diameters of 100–240 µm); and the microsporus group (sporangiophores less than 0.8 mm in height, sporangial diameters less than 100 µm, simple rhizoids). Maximal growth temperatures were characteristic: the stolonifer group grew at 30°C, the arrhizus group grew at 36°C, and the microsporus group grew at 45°C. The DNA mol% G + C base composition of all isolates ranged from 34.9 to 40.2% Species within the three groups were grouped by DNA differences. The arrhizus group was most distinctive with a value of 34.9–36.3%; the stolonifer and microsporus groups had G + C values of 37.0–39.3% and 37.8–40.2%, respectively. Our research clarifies and defines the G-C values of the three important groups ofRhizopus species.  相似文献

DNA polymorphism patterns linked to the A-globin gene were analyzed in healthy Japanese using four different restriction endonucleases. The chromosomes with the A-globin gene were mapped through an evaluation of the presence of seven different restriction sites (HincII 5 to ; HindIII in G and A; HincII in, and 3 to, 1; AvaII in ; Bam-HI 3 to ). Among 36 chromosomes analyzed, 20 chromosomes had a haplotype of [+–––––+]. Among 55 individuals examined, 7 possessed a homozygous haplotye of [+–––––+]. All Japanese with the AT-globin gene had a subhaplotype of [–++–+] 5 to the -globin gene. Their major haplotypes were [–++–+–+] and [–++–++–]. It was expected that the presence of the AT-globin gene in Japanese may be deduced from subhaplotypes 5 to the -globin gene.  相似文献

The activity of four different sialyltransferases acting on N- or O-linked chains of glycoproteins was studied in brains of 19 days-old embryos, 1 day-old newborns and adult rats. By using asialofetuin, fetuin andN-acetyllactosamine as acceptors, it has been possible to measure independently the following enzyme activities: CMP-NeuAc:Gal1-3GalNac (2–3)-sialyltransferase (EC 2.4.99.4), CMP-NeuAc:Gal1-4GlcNAc (2–3)-sialyltransferase (EC 2.4.99.6), CMP-NeuAc:Gal1-4GlcNAc (2–6)-sialyltransferase (EC 2.4.99.1) and CMP-NeuAc:NeuAc2-3Gal1-3GalNac (2–6)-sialyltransferase (EC 2.4.99.7). The specific activity of the first three enzymes which act on asialylated acceptors showed a 2.6-fold decrease in a parallel manner after ontogenic development, while the activity of NeuAc2-3Gal1-3GalNac (2–6)-sialyltransferase was four times lower in adult than in embryonic brain, showing a stronger dependence on ontogenic development. Summary Trichloro-, dichloro- and monochloroacetic and -dichloro-, -monochloro-and -monochloropropionic acids as sodium salts in 0.10–1.0mm concentration accelerated the growth of a strain of Rhizobium leguminosarum in a liquid synthetic medium with biotin as sole accessory growth factor. The strongest effect was shown by -monochloropropionate. Concentrations of 10mm and above were inhibitory. -Monochlorobutyrate did not accelerate growth.Ca-pantothenate and its precursor -alanine strongly improved growth in concentrations of 0.10 to 1.0 p.p.m. (0.001–0.011mm -alanine) and masked the effect of the chloro-substituted organic acids which seemed to function as more or less inferior substitutes for -alanine.The effect of -alanine was approximately ten times greater than that of -monochloropropionate which possibly owed its relatively strong activity to its close structure analogy with -alanine.Two other strains of rhizobia showed a moderate response to trichloroacetate and dichloropropionate, while others failed to do so and grew abundantly with biotin alone.  相似文献

Pseudomonas exotoxin A (PE) is one of the most potent cytotoxic agents produced byPseudomonas aeruginosa. In this study, we examined the possibility of using PE with a deletion of 38 carboxyl-terminal amino acid residues, designated PE(576–613), for active immunization against PE-mediated disease. We first examined the toxic effects of PE and PE(576–613) on 5- and 9-week-old ICR mice. The results show that the subcutaneous administration of PE(576–613) at a dose of 250 µg was still nontoxic to 5- and 9-week-old ICR mice, while native PE was lethal at a dose of 0.5 and 1 µg, respectively. PE(576–613) was then used to immunize ICR mice. The minimum dose of PE(576–613) that could effectively induce anti-PE antibodies in 5- and 9-week-old ICR mice was found to be 250 ng. However, immunization with 250 ng PE(576–613) failed to protect the immunized mice from a lethal dose of PE. The effective immunization dose of PE(576–613) that could protect mice against a 2 µg PE challenge was found to be 15 µg. In addition, sera obtained from PE(576–613)-immunized ICR mice were able to neutralize PE intoxication and effectively protect mice from PE. Thus, PE(576–613) may be used as an alternative route to new PE vaccine development.  相似文献

TNF alpha is required for hypoxia-mediated right ventricular hypertrophy   总被引:1,自引:0,他引:1  
Hypoxia has been shown to activate the pleiotropic cytokine TNF in the lung. TNF in turn, is known to induce pulmonary vasoconstriction. Additional effects of this cytokine in hypoxia mediated cardiopulmonary remodeling are poorly understood. To further evaluate the role of TNF in chronic hypoxia we exposed TNF null (TNF–/–) and wild-type mice to three weeks of hypobaric hypoxia (10% O2). Equivalent erythocytosis (Hematocrit increased by 40%) developed in both genetic backgrounds. In contrast, right ventricular systolic pressure increased in response to three weeks of hypoxia in the wild-type mice ( 75%), yet was unaltered in the TNF–/– mice. Concomitantly right ventricular hypertrophy was attenuated in the TNF–/– mice (35 ± 5% increase) when compared to wild-type mice (124 ± 6% increase p < 0.001, n 20). Interestingly in both strains the lung wet weights increased to a similar degree in response to hypoxia. In conclusion, our data demonstrate that TNF is an integral autocoid in chronic hypoxia mediated right ventricular hypertrophy. Moreover, additional components of cardiopulmonary remodeling may be regulated by TNF signaling as suggested by the negligible right ventricular systolic pressure response to hypoxia in the absence of TNF.  相似文献

An RNA polymerase chain reaction strategy was used to amplify and clone a cDNA segment encoding for the complete constant part of the axolotl IgY heavy (C) chain. C is 433 amino acids long and organized into four domains (C1–C4); each has the typical internal disulfide bond and invariant tryptophane residues. Axolotl C is most closely related to Xenopus C (40% identical amino acid residues) and C1 shares 46.4% amino acid residues among these species. The presence of additional cysteines in C1 and C2 domains is consistent with an additional intra-domain S-S bond similar to that suggested for Xenopus C and C, and for the avian C and the human C. C4 ends with the Gly-Lys dipeptide characteristic of secreted mammalian C3, human C4, and avian and anuran C4, and contains the consensus [G/GT(AA)] nucleotide splice signal sequence for joining C4 to the transmembrane region. These results are consistent with the hypothesis of an ancestral structural relationship between amphibian, avian chains, and mammalian chains. However, these molecules have different biological properties: axolotl IgY is secretory Ig, anuran and avian IgY behave like mammalian IgG, and mammalian IgE is implicated in anaphylactic reactions.The nucleotide sequence data reported in this paper have been submitted to the EMBL/GenBank nucleotide sequence database and have been assigned the accession number X69492. Polysaccharide composition of neutral, acid- and alkali-soluble fractions of the diatoms Stephanodiscus meyerii Genkal et Popovsk and Aulacoseira baicalensis (K. Meyer) Simonsen of Lake Baikal has been studied. Neutral polysaccharides were represented by chrysolaminarans (13;16--D-glucans). The chrysolaminaran from S. meyerii consists of the high- and low-molecular-weight fractions (40 and 2–5 kDa, respectively) and contains a large number of -16-bound glucose residues. The chrysolaminaran from A. baicalensis is a low-molecular-weight 13;16--D-glucan containing a small number of -16-bonds, with mannitol being attached to the reducing unit of its chain. Acid- and alkali-soluble polysaccharide fractions are practically absent in S. meyerii. In confirmation of earlier findings, surface-spread early diminution stages of Cyclops furcifer and C. divulsus yield numerous chromatin rings formed by the 25-to 30-nm type of fiber. Their contour lengths have a range of 0.6 16 m in C. divulsus and 0.4–40 m in C. furcifer. Employing the Miller spreading technique nucleosomal chromatin rings were detected in the critical stages of diminution in a size range of 0.6–100 m, though in lower frequencies. Instead, linear fragments of nucleosomal chromatin were found in numbers equal to or surpassing that of the rings.  相似文献

The organic-matter carbon isotope discrimination () of lichens with a wide range of photobiont and/or cyanobiont associations was used to determine the presence or absence of a carbon-concentrating mechanism (CCM). Two groups were identified within the lichens with green algal photobionts. One group was characterised by low, more C4-like values ( < 15), the other by higher, more C3-like values ( > 18). Tri-partite lichens (lichens with a green alga as the primary photobiont and cyanobacteria within internal or external cephalodia) occurred in both groups. All lichens with cyanobacterial photobionts had low values ( < 15). The activity of the CCM, organic-matter values, on-line values and gas-exchange characteristics correlated with the presence of a pyrenoid in the algal chloroplast. Consistent with previous findings, lichens with Trebouxia as the primary photobiont possessed an active CCM while those containing Coccomyxa did not. Organic values for lichens with Stichococcus as the photobiont varied between 11 and 28. The lichen genera Endocarpon and Dermatocarpon (Stichococcus + pyrenoid) had C4-like organic values ( = 11 to 16.5) whereas the genus Chaenotheca (Stichococcus — pyrenoid) was characterised by high C3-like values ( = 22 to 28), unless it associated with Trebouxia ( = 16). Gas-exchange measurements demonstrated that Dermatocarpon had an affinity for CO2 comparable to those species which possessed the CCM, with K0.5 = 200–215 1 · 1–1, compensation point () = 45–48 l · l–1, compared with K0.5 = 195 1 · 1–1, = 441 · 1–1 for Trebouxioid lichens. Furthermore, lichens with Stichococcus as their photobiont released a small pool (24.2 ± 1.9 to 34.2 ± 2.5 nmol · mg–1 Chl) of inorganic carbon similar to that released by Trebouxioid lichens [CCM present, dissolved inorganic carbon (DIC) pool size = 51.0 ± 2.8 nmol · mg–1 Chl]. Lichens with Trentepohlia as photobiont did not possess an active CCM, with high C3-like organic values ( = 18 to 23). In particular, Roccella phycopsis had very high on-line values ( = 30 to 33), a low affinity for CO2 (K0.5 = 400 1 · 1–1, = 120 1 · –1) and a negligible DIC pool. These responses were comparable to those from lichens with Coccomyxa as the primary photobiont with Nostoc in cephalodia (organic = 17 to 25, on-line = 16 to 21, k0.5 = 388 1 · 1–1, = 85 1 · 1–1, DIC pool size = 8.5 ± 2.4 nmol · mg–1 Chl). The relative importance of refixation of respiratory CO2 and variations in source isotope signature were considered to account for any variation between on-line and organic . Organic was also measured for species of Anthocerotae and Hepaticae which contain pyrenoids and/or Nostoc enclosed within the thallus. The resting state of wild-type nitrogenase MoFe protein exhibits an S=3/2 electron paramagnetic resonance (EPR) signal originating from the FeMo cofactor, the enzymes active site. When nitrogenase turns over under CO, this signal disappears and one (sometimes two) of three new EPR signals, which also arise from the FeMo cofactor, appears, depending on the CO concentration. The appearance and properties of these CO-inducible EPR signals, which were also generated with variant MoFe proteins (R96Q, R96K, Q191K, R359K, R96K/R359K, R277C, R277H, and nifV) that are impacted around the FeMo cofactor, have been investigated. No new CO-induced EPR signals arise from any variant, suggesting that no new CO-binding sites are produced by the substitutions. All variant proteins, except R277H, produce the lo-CO signal; all, except Q191K, produce the hi(5)-CO signal; but only two (R96Q and nifV) exhibit the hi-CO signal. FeMo cofactors environment clearly dictates which CO-induced EPR signals are generated; however, none of these EPR signals correlate with CO inhibition of H2 evolution observed with some of these variants. CO inhibition of H2 evolution is, therefore, due to CO binding to a different site(s) from those responsible for the CO-induced EPR signals. Some resting-state variants have overlapping S=3/2 EPR signals, whose intensities simultaneously decrease under turnover conditions, indicating that all FeMo cofactor conformations are catalytically active. Moreover, these variants produce a similar number of hi(5)-CO signals after turnover under CO to the number of resting-state S=3/2 signals. The FeMo cofactor associated with the hi(5)-CO signal likely contains two bridging CO molecules.  相似文献

Summary Growth responses of Ashmouni and Karnak cotton variety seedlings toRhizoctonia solani, the damping-off fungus, or toBacillus subtilis (two different strains),Aspergillus terreus andAspergillus flavus isolated from the rhizosphere of cotton, and all antagonistic to the pathogen, were expressed in terms of growth-vigour criteria.The presence ofR. solani in the soil inhibited the growth vigour of both cotton variety seedlings. However, Karnak seedlings were more sensitive to the pathogen than Ashmouni seedlings. One of the strains ofB. subtilis andA. terreus generally increased the vigour of both cotton variety seedlings.A. flavus lowered most of the growth criteria of Karnak or Ashmouni cotton seedlings.

Diese Organismen wurden als antagonistisch gegenRhizoctonia solani erkannt. Die Wirkung dieser Organismen auf das Wachstum von Keimlingen der Baumwolle sorten Ashmouni und Karnak wurde untersucht. R. solani hemmt das Wachstum der Keimlinge beider Baumwolle-Sorten. Es wurde festgestellt, dass Karnak empfindlicher ist als Ashmouni. Einer der Stämme vonB. subtilis undA. terreus erwiesen sich als Wachstum förderend bei beiden Baumwolle-Sorten.A. flavus dagegen vermindert das Wachstum von Karnak und Ashmouni.

The gene encoding 5 3 exonuclease domain-deleted Tfi DNA polymerase, named 5 3 Exo Tfi fragment, from Thermus filiformis was expressed in Escherichia coli under the control of the tac promoter on a high-copy plasmid, pJR. The expressed enzyme was purified 27-fold with a 19% yield and a specific activity of 2621 U mg–1 protein. The 5 3 exonuclease domain of Tfi DNA polymerase was removed without significant effect on enzyme activity and stability. PCR conditions for the 5 3 Exo Tfi fragment were more tolerant to the buffer composition as compared to the full-length enzyme.  相似文献

Summary Tabac anergié tissue has been used by many investigators as a habituated tobacco cell line. In this study, we have found sequences which are homologous to approximately 9 megadaltons of an Agrobacterium octopine-type plasmid in DNA isolated from Tabac anergié tissue. These T-DNA sequences were shown to be the same core DNA sequences found to be present in several other unorganized tumor lines which have been studied in our laboratory. The T-DNA was also shown to be integrated into plant DNA. Thus, the Tabac anergié tissue is a crown gall tumor induced by a strain of Agrobacterium tumefaciens containing an octopinetype plasmid. These results indicated that the plants regenerated from 40 clones of the Tabac anergié line by Sacristán and Melchers (1977) were actually reverted plants obtained from an unorganized crown gall tumor, kept in culture for more than 25 years. It is necessary to re-evaluate observations and conclusions which have been based upon Tabac anergié being a habituated tissue.  相似文献

As initial step in the transfer of dwarf bunt resistance from barley into wheat, the two cereal crops were hybridized. Using the wheat cultivars Fukuhokomugi and Chinese Spring (AABBDD genomes) as female parents and barley cultivar Luther (II genome) as male, we synthesized 9 euploid hybrids (2n = 4x = 28; ABDI genomes). The hybrids were vigorous, but highly sterile. Meiotic analyses of seven hybrids showed considerable variation in chromosome pairing. Of the hybrids involving Fukuhokomugi 3 had high pairing with a mean of 5.08–6.72 chiasmata per cell, while others had 2.16–3.52 chiasmata per cell. As many as 12 bivalents in some pollen mother cells would suggest at least some pairing between wheat and barley chromosomes. This level of homoeologous pairing, coupled with some, albeit low, female fertility of the F1 hybrids, could offer an opportunity for intergeneric gene transfers from barley into wheat and vice versa.  相似文献

Crude extracts or supernatants of broken cells of Clostridium formicoaceticum reduce unbranched, branched, saturated and unsaturated carboxylates at the expense of carbon monoxide to the corresponding alcohols. The presence of viologens with redox potentials varying from E 0=-295 to-650 mV decreased the rate of propionate reduction. The more the propionate reduction was diminished the more formate was formed from carbon monoxide. The lowest propionate reduction and highest formate formation was observed with methylviologen. The carbon-carbon double bond of E-2-methyl-butenoate was only hydrogenated when a viologen was present. Formate as electron donor led only in the presence of viologens to the formation of propanol from propionate. The reduction of propionate at the expense of a reduced viologen can be followed in cuvettes. With respect to propionate Michaelis Menten behavior was observed. Experiments are described which lead to the assumption that the carboxylates are reduced in a non-activated form. The N-terminal domain (1–318 amino acids) of mouse NFB (p65) has been purified to homogeneity from the soluble fraction of Escherichia coli cells expressing this protein. Its complex with a full-length iB- (MAD3, 1–317 amino acids) molecule was generated by binding the E. coli-derived iB- to the purified NFB and purifying the complex by sequential chromatography. The stoichiometry of NFB to iB in the complex was determined to be 2 to 1 by light scattering and SDS–polyacrylamide gel electrophoresis. The secondary structure of the NFB (p65) determined by Fourier-transform infrared (FTIR) spectroscopy is in good agreement with that of the p50 in the crystal structure of the p50/DNA complex, indicating that no significant structural change in NFB occurs upon binding of DNA. The FTIR spectrum of the NFB/iB complex indicates that its secondary structure is composed of 17% -helix, 39% -strand, 18% irregular structures, and 26% -turns and loops. By comparing these data to the FTIR data for NFB alone, it is concluded that the iB (MAD3) in the complex contains 35% -helix, 27% -strand, 22% irregular structures, and 16% -turns and loops. Circular dichroism (CD) analysis of a shorter form of iB (pp40) indicates that it contains at least 20% -helix and that the iB subunit accounts for nearly all of the -helix present in the NFB/iB complex, consistent with the FTIR results. The stabilities of NFB, iB, and their complex against heat-induced denaturation were investigated by following changes in CD signal. The results indicate that the thermal stability of iB is enhanced upon the formation of the NFB/iB complex.  相似文献
