Multiple virus infections in the honey bee and genome divergence of honey bee viruses 总被引:1,自引:0,他引:1

Using uniplex RT-PCR we screened honey bee colonies for the presence of several bee viruses, including black queen cell virus (BQCV), deformed wing virus (DWV), Kashmir bee virus (KBV), and sacbrood virus (SBV), and described the detection of mixed virus infections in bees from these colonies. We report for the first time that individual bees can harbor four viruses simultaneously. We also developed a multiplex RT-PCR assay for the simultaneous detection of multiple bee viruses. The feasibility and specificity of the multiplex RT-PCR assay suggests that this assay is an effective tool for simultaneous examination of mixed virus infections in bee colonies and would be useful for the diagnosis and surveillance of honey bee viral diseases in the field and laboratory. Phylogenetic analysis of putative helicase and RNA-dependent RNA polymerase (RdRp) encoded by viruses reveal that DWV and SBV fall into a same clade, whereas KBV and BQCV belong to a distinct lineage with other picorna-like viruses that infect plants, insects and vertebrates. Results from field surveys of these viruses indicate that mixed infections of BQCV, DWV, KBV, and SBV in the honey bee probably arise due to broad geographic distribution of viruses. 相似文献

Because bees fly around, visit flowers and chase mates, we conclude intuitively that they see things as we do. But their vision is unexpectedly different, so we say it is anti-intuitive. Detailed tests have demonstrated separate detectors for modulation of blue and green receptors, edge orientation (green only), and areas of black. The edge detectors are about 3° across, independent, and not re-assembled to make lines, shapes or textures. Instead, the detectors of each type are summed quantitatively to form cues in each local region with an order of preference for learning the cues. Trained bees remember the positions of the total modulation (preferred), the average edge orientation, areas of black or colour, and positions of hubs of radial and circular edges in each local region, but not the original responses, so the pattern is lost. When presented with a yellow spot on a blue background with no UV reflected, the preferred cue is not the colour, but a measure of the modulation detected by the green and separately by the blue receptors. 相似文献

Colony condition and differences in individual preferences influence forage type collected by bees. Physiological bases for the changing preferences of individual foragers are just beginning to be examined. Recently, for honey bees octopamine is shown to influence age at onset of foraging and probability of dance for rewards. However, octopamine has not been causally linked with foraging preference in the field. We tested the hypothesis that changes in octopamine may alter forage type (preference hypothesis). We treated identified foragers orally with octopamine or its immediate precursor, tyramine, or sucrose syrup (control). Octopamine-treated foragers switched type of material collected; control bees did not. Tyramine group results were not different from the control group. In addition, sugar concentrations of nectar collected by foragers after octopamine treatment were lower than before treatment, indicating change in preference. In contrast, before and after nectar concentrations for bees in the control group were similar. These results, taken together, support the preference hypothesis. 相似文献

The allergenic activities of four purified components of honeybee venom were studied by using histamine release from leukocytes of bee sting-allergic patients. The components studied were hyaluronidase, phospholipase A2, melittin and apamin with molecular weights, respectively, of about 50,000, 15,800, 2840 and 2038 d. In six of the seven patients studied, hyaluronidase and phospholipase were, respectively, on the average about two and eight times more active by weight than the venom. The situation was reversed in one patient in that hyaluronidase and phospholipase A2 were, respectively, 90 and 0.5 times more active than the venom. With this single exception, hyaluronidase and phospholipase were about equally active on a molar basis as allergens. Melittin was on the average about one-tenth as active as the venom, and apamin was inactive as an allergen.Chemical modifications of phospholipase A2 were carried out. Succinylation of eight of its eleven amino groups yielded a derivative that retained 4% of the enzymic activity of the native enzyme. Reduction and carboxymethylation of its four disulfide bonds or cyanogen bromide cleavage of its three methionyl bonds yielded enzymatically inactive derivatives. These derivatives showed varying decreases of allergenic activities when compared to the native enzyme. The results indicate that the antigenic determinants of phospholipase depend on the charge, the amino acid sequence and the conformation of the molecule. 相似文献

Chronic bee-paralysis virus and sacbrood virus occur commonly in apparently normal honey-bee colonies in Britain. Most sick adult bees not affected by Nosema apis, Malpighamoeba mellificae or Acarapis woodi have chronic paralysis and most dead larvae not affected by micro-organisms have sacbrood. Both virus diseases are probably limited by hereditary factors, but unknown environmental factors also seem to influence disease. Paralysed bees from Australia, Canada, Eire, Germany and Mexico were found to be infected with chronic paralysis virus. 相似文献

A general mathematical model for population dispersal featuring long range taxis is presented and exemplified by the dispersal
episode of the Africanized honey bees (Apis mellifera adansonii) throughout the American Continent. The mathematical model is a discrete-time and nonlocal model represented by an integrodifference
recursion. A newtaxis concept is defined and introduced into the mathematical model by an appropriate modification of the
redistribution kernel. The model is capable of predicting the natural barrier for the expansion of the Africanized honey bees
in the southern part of the Continent due to low winter temperatures. It also describes a sensitive expansion velocity with
respect to the quality of resources, which can explain the AHB’s astounding spread rate, by using two different kinds of population
dynamics strategies, one for a resourceful environment and the other for poor regions.

ABSTRACT. The fine structure and function of a honey bee's ( Apis mellifera Linn.) proventriculus were studied by scanning electron microscopy and video-recording. Our observations revealed that the proventriculus is used to engulf pollen and other particles which contaminate the nectar carried into the crop. The four lips are closed and opened, pulled backwards and straightened by the external circular muscles and internal longitudinal muscles. Combs of filiform-hairs (70 μm in length) located on the margins of the lips 'catch' and filter particles from the fluid. By repeated filtering, opening and closing actions of the hairs and lips, particles are filtered and collected in pouches between the ventricular folds to form boluses and are eventually passed into the midgut. In the present experiment, particle sizes ranging from 0.5 to 100 μm in diameter, including dandelion pollen ( Taraxacum officinale Web.), Torula yeast ( Candida utilis Lodder et Kreger-Van Rij), bee disease spores of Nosema apis Zander and Bacillus larvae White, and man-made particles can be filtered by the hairs. Small particles (0.23 μm in diameter) filter through the hair and return back to the fluid. Large particles (100–200 μm in diameter) are caught between the stylets of the mouthparts and are not ingested. These observations suggest that the particle size plays an important role in determining what can be taken by the mouthparts and the proventriculus and what can later be utilized as a food source by the bee. The role of the proventriculus in disease transmission is also discussed. 相似文献

A protocol has been developed for the purification of vitellogenin from the honey bee, Apis mellifera. Purification allows for the first characterization of a vitellogenin from the large order Hymenoptera. Hymenopteran vitellogenins are unusual among insect vitellogenins in that they contain only one type of apoprotein. The honey bee vitellogenin was isolated from hemolymph of honey bee queens by a combination of density gradient ultracentrifugation, ion-exchange chromatography, and affinity chromatography. The native vitellogenin particle is a very high density glycolipoprotein containing approximately 91% protein, 7% lipid, and 2% carbohydrate. Phospholipid and diacylglycerol are the major lipid components. The equilibrium density (1.28 g/ml) is the same as that for Manduca sexta vitellogenin, which contains a much higher proportion of lipid. The covalently bound carbohydrate moiety of the particle is high in mannose. The amino acid composition of vitellogenin is similar to those of vitellogenins from other insect species. The N-terminal amino acid sequence of the apoprotein was determined, the first such sequence for any insect vitellogenin. When analyzed by sodium dodecyl sulfate (SDS)-gel electrophoresis, A. mellifera vitellogenin resolved into a single band with an apparent Mr of 180,000. Recent losses in honey bee colonies are unusual in their severity, geographical distribution, and, in some cases, failure to present recognized characteristics of known disease. Domesticated honey bees face numerous pests and pathogens, tempting hypotheses that colony collapses arise from exposure to new or resurgent pathogens. Here we explore the incidence and abundance of currently known honey bee pathogens in colonies suffering from Colony Collapse Disorder (CCD), otherwise weak colonies, and strong colonies from across the United States. Although pathogen identities differed between the eastern and western United States, there was a greater incidence and abundance of pathogens in CCD colonies. Pathogen loads were highly covariant in CCD but not control hives, suggesting that CCD colonies rapidly become susceptible to a diverse set of pathogens, or that co-infections can act synergistically to produce the rapid depletion of workers that characterizes the disorder. We also tested workers from a CCD-free apiary to confirm that significant positive correlations among pathogen loads can develop at the level of individual bees and not merely as a secondary effect of CCD. This observation and other recent data highlight pathogen interactions as important components of bee disease. Finally, we used deep RNA sequencing to further characterize microbial diversity in CCD and non-CCD hives. We identified novel strains of the recently described Lake Sinai viruses (LSV) and found evidence of a shift in gut bacterial composition that may be a biomarker of CCD. A common and widespread disease of honey bees, Apis mellifera, is caused by an unoccluded, Feulgen-positive, filamentous nuclear virus. Ovoid viral particles seen in diseased bee hemolymph consisted of a folded nucleocapsid within a viral envelope and were 0.40 by 0.10 μm. Virions with unfolded nucleocapsids were about 3060 by 60 nm. The disease was transmissible to bees both per os and by injection, but efforts to infect oriental cockroaches, Blatta orientalis, and the greater wax moth, Galleria mellonella, failed. The disease is apparently the same as that described as a rickettsial disease of European bees. 相似文献

First, we wished to obtain an improved set of gene models with increased coverage of known genes, while maintaining gene model quality. Across the Northern hemisphere, managed honey bee colonies, Apis mellifera, are currently affected by abrupt depopulation during winter and many factors are suspected to be involved, either alone or in combination. Parasites and pathogens are considered as principal actors, in particular the ectoparasitic mite Varroa destructor, associated viruses and the microsporidian Nosema ceranae. Here we used long term monitoring of colonies and screening for eleven disease agents and genes involved in bee immunity and physiology to identify predictive markers of honeybee colony losses during winter. The data show that DWV, Nosema ceranae, Varroa destructor and Vitellogenin can be predictive markers for winter colony losses, but their predictive power strongly depends on the season. In particular, the data support that V. destructor is a key player for losses, arguably in line with its specific impact on the health of individual bees and colonies. 相似文献
