对日本血吸虫中国大陆株湖南、湖北、江西、安徽、四川、云南隔离群以及一个实验室传代品系从基因水平进行了多态性研究。首先,在用PCR-SSWCP技术分析了28S rDNA-D2高变区基础上,测定了该区安徽和云南隔离群的DNA序列;其次,用PCR获得了含有ITS的rDNA片段,并对其进行了酶切点重复序列的多态性分析;最后,用RAPD技术分析了全基因组DNA的多态性。结果表明,安徽与云南隔离群的28S r  相似文献

DNA双链断裂(DSBs)是电离辐射诱导的最重要的原发损伤,研究DSBs有利于揭示细胞辐射敏感性的机理。用倒转脉冲场凝胶电泳结合荧光扫描进行DNA定量研究75MeV/u12C6+对小鼠B16黑色素瘤细胞DSBs的诱导,结果表明:DSBs产额约为0.74DSBs/100Mbp/Gy;DNA片段分布在两个区域。大片段区分子量约为1.4Mbp~3.2Mbp,分子量小于1.2Mbp的为小片段区;并且随着剂量的增加,大片段区DNA含量逐渐下降,而小片段区的DNA含量显著增加。表明B16DNA分子上可能存在对重离子较为敏感的位点。  相似文献

利用ESR技术研究了博莱霉素A5(BLMA5)、Fe^2+,O2体系中.OH的产生。并用TBA反应检测了该系对DNA的断链作用。以Fe^3+,Cu^+或Cu^2+替换Fe^2+,则体系中无.OH产生,同时也失去对DNA的断链作用;一定浓匠.OH清除剂可完全清除.OH,地对DNA断链影响甚微;还原剂可淬灭体系中的.OH,却能促进DNA的断链反应;SOD可清除体系中的.OH,失活的SOD夫此功能,但两  相似文献

比较了凝胶电泳示检测质粒DNA时不同激发波长对DNA-EB荧光强度的影响,发现短波长激发光可增加DNA的探测灵敏度。采用260nm作为激光光时可探测到少至0.7ng的线性DNA。且在很广的DNA的质量范围内,DNA-EB荧光强度 与DNA量或正比。以此改进方法检测电离辐射诱导的DNA单、双链断裂岢得到与其它研究结果相一致的G(SSB)和G(DSB)值。  相似文献

用Ca2+ 和胰酶处理大叶藻(Zostera m arina)叶绿体膜研究了其类囊体膜多肽成分与Mg2+ 诱导其Chla荧光和类囊体膜表面电荷变化之间的相互关系,观察到:1.在正常的叶绿体膜中,Mg2+ 诱导PSⅡ荧光强度的增高与其诱导类囊体膜表面电荷密度的降低密切相关;2.用Ca2+ 处理这种叶绿体膜,除去类囊体膜表面的32~34 kD多肽对Mg2+ 诱导的上述现象无影响;3.如果用胰酶消化Ca2+ 处理过的叶绿体膜,进一步除去膜表面的26 kD多肽,Mg2+诱导的这些现象则全部消失。这些实验结果清楚地表明,在大叶藻的叶绿体膜中,类囊体膜表面的26 kD 多肽是阳离子诱导这两种相关现象的特异性作用部位。对阳离子调节激发能在PSⅡ和PSⅠ之间分配的机理进行了讨论  相似文献

观察了亚硒酸钠(Na2SeO3)在体外作用于大鼠晶状体上皮细胞(RLE cells)而造成的DNA单链断裂(SSB),并对其DNA损伤、修复动力学做了初步研究。发现SSB严重程度与亚硒酸钠的浓度呈线性相关,其SSB重接修复约在30 ̄60min内完成,还作了有关非程序DNA合居(UDS)的检测,发现与SSB相比,UDS发生迟且持续时间更长,提示Na2SeO3可能在体外对大鼠晶状体上皮细胞除造成SSB  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
