The qualitative and quantitative compositions of leaf cuticular waxes from potato (Solanum tuberosum) varieties were studied. The principal components of the waxes were very long chain n-alkanes, 2-methylalkanes and 3-methylalkanes (3.1-4.6 microg cm(-2)), primary alcohols (0.3-0.7 microg cm(-2)), fatty acids (0.3-0.6 microg cm(-2)), and wax esters (0.1-0.4 microg cm(-2)). Methyl ketones, sterols, beta-amyrin, benzoic acid esters and fatty acid methyl, ethyl, isopropyl and phenylethyl esters were found for the first time in potato waxes. The qualitative composition of the waxes was quite similar but there were quantitative differences between the varieties studied. A new group of cuticular wax constituents consisting of free 2-alkanols with odd and even numbers of carbon atoms ranging from C25 to C30 was identified.  相似文献

Blaesoxipha atlanis (Aldrich) is a common parasitic fly of agriculturally important grasshoppers in Canada. The suitability of Camnula pellucida (Scudder), Melanoplus bivittatus (Say), Melanoplus packardii Scudder, and Melanoplus sanguinipes (Fabricius) as hosts was studied in the laboratory. Grasshoppers were singly-parasitized or left unparasitized and reared for 9 days. Melanoplus bivittatus and M. packardii did not support parasite development, i.e. were non-permissive hosts. In both species, parasite larvae were melanized and encapsulated, but development proceeded further in M. packardii. Melanoplus sanguinipes and C. pellucida were permissive host species with, respectively, 70% and 35% of the implanted larvae emerging from their hosts of which 86% and 50% developed into adults. Parasite development time was longer in C. pellucida. Adult B. atlanis dry mass varied with host species and host mass at parasitism, but not with host sex. Parasites developing in M. sanguinipes were larger in terms of dry mass than counterparts developing in C. pellucida. In permissive species, unparasitized grasshoppers gained in body mass while parasitized insects lost mass during the 9-day observation period. In non-permissive species, all insects gained in body mass, but parasitized females gained less mass than unparasitized conspecifics. All unparasitized grasshoppers survived while 75-95% of permissive and 30-40% of non-permissive hosts died. Variation in the intensity of field parasitism among grasshopper species may be explained, at least in part, by qualitative differences in suitability between potential host species. Novel pest management strategies emphasize preservation of a small proportion of the pest population for natural enemies. Consideration of the outcome of specific host-parasite interactions should improve the understanding of grasshopper population dynamics and increase the predictive value of models that assess potential crop losses.  相似文献

The cuticular surface lipids of the red harvester ant, Pogonomyrmex barbatus, were found to contain minor amounts of novel wax esters, in addition to the major components, hydrocarbons. The wax esters ranged in carbon number from C19 to C31 and consisted of esters of both odd- and even-numbered alcohols and acids. Each wax ester with a given carbon number eluted at several different retention times indicating possible methyl branching in either the fatty acid or alcohol moiety, or in both moieties. Each eluting peak of wax esters consisted of a mixture of wax esters of the same carbon number in which the fatty acid moiety ranged from C8 to C18, and the alcohol moiety ranged from C8 to C17. Some wax esters were largely found on the head indicating they may be of a glandular origin. The hydrocarbons consisted of: n-alkanes, C23 to C33; odd-numbered n-alkenes, C27 to C35; and the major components, methyl-branched alkanes, C26 to over C49. Notable components of the methyl-branched alkanes were 2-methyltriacontane, and the novel trimethylalkanes with a single methylene between the first and second branch points, 13,15,19-trimethylhentriacontane and 13,15,21-trimethyltritriacontane.  相似文献

About 15% of the epidermal wax on Hordeum vulgare cv. Bonus barley spikes is n-alkanes. Longer homologues are greatly reduced in the eceriferum mutants, cer-a(6), cer-e(8), cer-n(26), cer-n(53), cer-n(985), cer-x(60), cer-yc(135) and cer-yl(187). Simultaneously hydrocarbons accounting for only traces in the wild-type become prominent in the mutants, although their chain-length distributions remain unchanged. Accordingly several new hydrocarbon series were identified. The two major ones were C(23)-C(35)cis monoenoic alkenes (the major 9-ene isomer was part of a homologous series including 11, 13 and 15-enes), and the novel C(27)-C(31) cyclopropanes (the ring carbons of major isomers were 9,10 and 11,12 with lesser amounts of 13,14). Three minor series included 2- and 3-methylalkanes plus C(25)-C(33) internally branched alkanes (methyls on carbons 9, 11, 13, 15 or 17; shorter homologues dominated by the 9 isomer, longer homologues by 11, 13 or 15 isomers). Acyl chains destined for spike waxes are synthesized via acyl and polyketide elongase systems plus associated reductive and decarbonylative/decarboxylative enzyme systems. Both elongation systems are defective in synthesizing C(32) acyl chains in all nine mutants. The similarities in the position of the chemical groups (primarily on carbon 9, secondarily on carbon 11) of the alkenes, cyclopropanes and internally branched methyl alkanes imply an origin from a common, hitherto unrecognized associated pathway in barley, designated the enoic pathway. The elongation system leading to the enoic derived hydrocarbons differs from the known elongation systems by inclusion of a mechanism for introducing a double bond.  相似文献

Hydrocarbon and wax ester components of cuticular lipids of the braconid parasitoid Habrobracon hebetor Say reared at 25 degrees C on larvae of a pyralid moth have been identified by GC-MS and analyzed with respect to adult age, mating status, and diet. The hydrocarbons range in carbon number from C(21) to C(45) and consist of a homologous series of n-alkanes, 11-, 13-, and 15-methyl alkanes, 13,17-dimethyl alkanes, and Z-5, Z-7, and Z-9-alkenes. The wax esters found in the cuticular lipid fraction are a series of homologous compounds with the acid portion being short chain, unbranched, even carbon number acids from C(8) to C(20) (predominately C(8) to C(16)). The alcohol portions of the esters are secondary alcohols with carbon number from C(22) to C(25) (predominately C(23) and C(25)) with the hydroxyl function located at C(6), C(7), C(8), and C(9). Gender, age, and nutritional states were significant factors for variation in several of the individual esters, but mating status did not affect wax ester composition. Ontogenetic examinations indicated that prepupal, and early pupal cuticular lipids contain only hydrocarbons. Low levels of wax esters are detectable in late stage pupae, and somewhat greater quantities of wax esters are present on newly eclosed adults. When pharate adults emerge from the cocoon, however, their cuticular lipids consist of approximately equal amounts of hydrocarbons and wax esters, and 6d post emergence from the cocoon, wax esters are the predominant lipid component.  相似文献

The Western Flower Thrips Frankliniella occidentalis effectively resists many insecticides, but it can be controlled by the use of bioinsecticides such as entomopathogenic fungi. The epicuticular chemistry of these insects is therefore of great interest, and accordingly, the cuticular lipid composition of F. occidentalis was analysed. It was found that the cuticular lipids of both the adult and larval stages of F. occidentalis consist of two groups of compounds--hydrocarbons and free fatty acids. The same hydrocarbon pattern was found in both adults and larvae, with the exception of n-hentriacontane, which was detected only in adult insects. The following homologous series were identified: n-alkanes from C-25 to C-29 (31) with the marked dominance of odd numbers of carbon atoms, 3-methylalkanes with 26 and 28 carbon atoms, and branched monomethylalkanes (branched at C-9, -11, -13 and -15) with 26, 28 and 30 carbon atoms. The chemical composition of the free fatty acids consists of two homologous series: saturated (C(14:0), C(16:0), C(18:0)) and unsaturated fatty acids (C(16:1) and C(18:1)). This analysis confirmed the lack of potential inhibitors of entomopathogenic fungi in the cuticular lipids of this insect species.  相似文献

Cuticular hydrocarbon mixtures can be used to discriminate insect taxa. They have utility for determining phylogenetic relationships where they are independent characters with discrete states and represent a hierarchical distribution of shared, derived characters. We report inferred degrees of relatedness among the chemical phenotypes of Reticulitermes from PAUP (phylogenetic analysis using parsimony) analyses of cuticular hydrocarbon characters. One hundred and forty-one Reticulitermes colonies collected from California, Georgia, New Mexico, Arizona and Nevada were used. Initial maximum parsimony analyses sorted the 141 colonies into 26 chemical phenotypes. Subsequent analyses, using the ancestral species Coptotermes formosanus and Heterotermes sp. as outgroups, sorted Reticulitermes taxa into three major lineages, each characterized by a different set of dominant methyl-branched or unsaturated hydrocarbon components. Reticulitermes in lineage I have cuticular hydrocarbon mixtures with a preponderance of internally branched monomethylalkanes and 11,15-dimethylalkanes. Those in lineage II are defined by a preponderance of 5-methylalkanes and 5,17-dimethylalkanes. Taxa in lineage III are characterized by the predominance of olefins and a relative paucity of n-alkanes and methyl-branched alkanes. Bootstrap analyses and decay indices provided statistical support and robustness for these chemical-based relationships.  相似文献

Labeled n-alkanes administered to the grasshopper Melanoplussanguinipes are hydroxylated at or near the middle of the carbon chain. The secondary alcohols formed are then esterified. Chain length specificity is evident in both the hydroxylation of n-alkanes and the esterification of secondary alcohols, with the shorter chain C23, C21, C19, and C25 compounds converted to secondary alcohol wax esters more readily than the longer chain C27, C29, and C31 compounds. Secondary alcohols and ketones are not reduced to alkanes.  相似文献

Sodium [1-14C]acetate, sodium [1-14C]propionate, sodium [2-14C]propionate, sodium [3-14C]propionate and sodium [methyl-14C]methylmalonate were readily incorporated into the cuticular hydrocarbons of nymphal stages of the cockroach Periplaneta fuliginosa both in vivo and in vitro, whereas no incorporation of [methyl-14C]methionine was observed. The alkanes of the nymphal stages of this insect are 25+% n-alkanes, 14% 3-methylalkanes, and 59+% internally branched monomethylalkanes, principally 13-methylpentacosane. Sodium [1-14C]acetate was incorporated into each class of alkane at about its percentage composition. In contrast, labeled sodium propionate and sodium methylmalonate were preferentially incorporated into the branched fractions. Radio-gas-liquid chromatography showed that sodium [1-14C]propionate was incorporated almost exclusively into 3-methyltricosane and 13-methylpentacosane, whereas sodium [1-14C]acetate was incorporated into each glc peak at about its percentage composition. These data suggest that propionate, incorporated during chain elongation, serves as the branching methyl group donor for both the 3-methyl and the internally branched monomethylalkanes in insects. The location of hydrocarbon synthesis in P. fuliginosa was studied using an in vitro tissue slice system. Excised cuticle slices, with adhering fat body tissue removed, gave good incorporation of labeled substrates into the hydrocarbon fraction. No hydrocarbon synthesis was observed in fat body preparations.  相似文献

Dietary hydrocarbons are incorporated into cuticular lipids of the grasshopper Melanoplus sanguinipes. Dietary secondary alcohols and ketones, however, are not incorporated into the cuticular lipids. In typical experiments from 8 to 28 per cent of the fed labeled n-alkanes are recovered in the cuticular lipids. Most of the radioactivity recovered from feeding the C23 n-alkane and a significant amount from the C25 was found as a secondary alcohol in the form of a wax ester. The C29 and C31 n-alkanes were recovered primarily unchanged as the n-alkane. Eighty-five per cent of injected acetate incorporated into the hydrocarbon fraction is in the branched hydrocarbons. These results show that the insect synthesizes its branched hydrocarbons, whereas a large part of the normal hydrocarbons can be dietary.  相似文献

The adult beetles Aphthona lacertosa and Aphthona nigriscutis, used as biocontrol agents for leafy spurge, had a complex mixture of hydrocarbons on their cuticular surface consisting of alkanes, methylalkanes, alkenes and alkadienes as determined by gas chromatography-mass spectrometry. A trace amount of wax esters were present. In both species, the hydrocarbons were the major cuticular lipid class and the gas chromatographic profiles of the total hydrocarbons were similar. However, the profiles for the saturated hydrocarbon fraction were distinct for each species. Alkanes (n-alkanes and methyl-branched alkanes), alkenes and alkadienes comprised 26, 44 and 30%, respectively, for A. lacertosa, and 48, 26 and 26%, respectively, for A. nigriscutis, of the total hydrocarbons. The major methyl-branched hydrocarbons were 2-methylalkanes: 2-methyloctacosane and 2-methyltriacontane. The major monoene was hentriacontene and the major diene was tritriacontadiene. The species were unique in that a number of di- and trimethyl-branched alkanes were present in minor quantities in which the first methyl branch was on carbon 2 or 3. Examples of structures were 2,10-, 2,12-, 2,6-, 2,4- and 3,7-dimethylalkanes. 2,10,12-Trimethylalkanes and a 2,10,12,24-tetramethylalkane with one methylene between adjacent methyl branch points also were identified. The adjacent methyl branch points of the 2,4- and 2,10,12- and 2,10,12,24-methyl-branched alkanes appeared to cause additional fragmentations in the mass spectra. Dimethylalkanes with an odd number of carbons in the backbone of the molecule were identified as 2,23-dimethylnonacosane and 2,25-dimethylhentriacontane; their mass spectra also corresponded to mass spectra expected for a 2,6 branching sequence. However, a 2,6 branching sequence is not biosynthetically feasible because such a structure has a straight-chain tail with an odd number of carbon atoms beyond the last methyl branch point. The 2,23 and 2,25 branching sequences could be synthesized starting with a primer derived from the amino acid leucine which would account for both the even number of carbons between the branch points and an even number of carbons beyond the last methyl branch point.  相似文献

Conidiobolus coronatus is an entomopathogenic fungus which has a potential as a biological control agent of insects. The cuticular and internal lipid composition of infected and noninfected Tettigonia viridissima males were analyzed by GC/MS. A total of 49 compounds were identified in the infected and noninfected males, including fatty acids, fatty acid methyl esters (FAMEs), n‐alkanes, alcohols, sterols, and other organic compounds. The most abundant components of the cuticular and internal lipids of the insects were fatty acids. After exposure to C. coronatus, the cuticular lipids of the T. viridissima males contained 17 free fatty acids from C(8) to C(22), while the cuticular lipids of the noninfected insects contained only 15 fatty acids from C(12) to C(24). The cuticular and internal lipids of both the infected and the noninfected males also contained five FAMEs from C(15) to C(19), seven n‐alkanes from C(25) to C(34), five alcohols from C(16) to C(25), five sterols, and the following six other organic compounds: azelaic acid, phenylacetic acid, glutaric acid, benzoic acid, sebacic acid, and glycerol. The compounds which were present only in the cuticular lipids of the infected males could be due to fungal infection.  相似文献

Skin surface lipids from mink (Mustela vison) were collected in acetone and analyzed by thin-layer chromatography and gas chromatography. The principal components were wax monoesters (92%), cholesteryl esters (5%), free fatty acids (1%), fatty alcohols (1%) and cholesterol (1%). The fatty acids and alcohols contained in these lipids were composed principally of homologous series of straight chained omega 7-unsaturated structures (C16-C24), accompanied by lesser proportions of homologous series of saturated (C14-C22) and omega 9-unsaturated (C18-C22) structures.  相似文献

Honey bees (Apis mellifera) are social insects which have remarkable complexity in communication pheromones. These chemical signals comprise a mixture of hydrocarbons, wax esters, fatty acids, aldehydes and alcohols. In this study, we detected several long chain aliphatic alcohols ranging from C18-C32 in honey bees and the level of these alcohols varied in each body segment. C18:0Alc and C20:0Alc are more pronounced in the head, whereas C22:0Alc to C32Alc are abundant in the abdomen. One of the cDNAs coding for a fatty acyl-CoA reductase (AmFAR1) involved in the synthesis of fatty alcohols was isolated and characterized. AmFAR1 was ubiquitously expressed in all body segments with the predominance in the head of honey bees. Heterologous expression of AmFAR1 in yeast revealed that AmFAR1 could convert a wide range of fatty acids (14:0–22:0) to their corresponding alcohols, with stearic acid 18:0 as the most preferred substrate. The substrate preference and the expression pattern of AmFAR1 were correlated with the level of total fatty alcohols in bees. Reconstitution of the wax biosynthetic pathway by heterologous expression of AmFAR1, together with Euglena wax synthase led to the high level production of medium to long chain wax monoesters in yeast.  相似文献

Triatomine insects (Hemiptera) are the vectors of Chagas disease. Their cuticular surface is covered by a thin layer of lipids, mainly hydrocarbons, wax esters, fatty alcohols, and free or esterified fatty acids. These lipids play a major role in preventing a lethal desiccation, altering the absorption of chemicals and microorganism penetration, they also participate in chemical communication events. Lipid components are biosynthetically related, the synthesis of long chain and very long chain fatty acids was first shown in the integument of Triatoma infestans through the concerted action of fatty acid synthases (FAS's) and fatty acyl-CoA elongases. A final decarboxylation step produces the corresponding hydrocarbon. Capillary gas chromatography coupled to mass spectrometry analyses showed that cuticular hydrocarbons of Triatominae comprise saturated straight and methyl-branched chains, from 18 to more than 43 carbon atoms. Odd-chain hydrocarbons, mostly from 27 to 33 carbons, are the major straight chains. Different isomers of mono, di, tri, and tetramethylcomponents, mostly from 29 to 39 atoms in the carbon skeleton, account for the major methyl-branched hydrocarbons. The presence, absence, and relative quantities of these hydrocarbons represent characters for their chemical phenotype, and are useful for differentiating genera, species and populations. In this review, we will discuss the metabolic pathways involved in hydrocarbon formation, and their structure, together with their role in insect survival. We will also review the utility of cuticular hydrocarbon fingerprints in chemotaxonomy.  相似文献

为明确小麦不同器官表皮蜡质晶体结构和蜡质组分的差异,该研究以小麦品系CP98(11)为材料,在小麦扬花期分别取小麦的旗叶、叶鞘、穗下茎、花药和颖壳,利用气相色谱技术对各器官表皮蜡质组分进行鉴定,并通过扫描电镜观察其蜡质晶体结构。结果表明:(1)小麦不同器官的蜡质成分共鉴定出30种,主要为初级醇、二酮、烷烃、脂肪醛、脂肪酸、酯。(2)叶鞘、穗下茎、颖壳的蜡质中二酮含量最高,分别占蜡质总量的78.96%、67.03%和68.6%;花药的蜡质中烷烃含量最高(75.82%);旗叶的蜡质中初级醇含量最高(45.91%),其次为烷烃33.19%。(3)扫描电镜观察显示,旗叶正面的蜡质晶体呈片状结构,旗叶反面和颖壳的蜡质晶体结构呈片状与柱状混合的结构,花药的蜡质呈明显的波浪状结构,穗下茎和叶鞘的蜡质晶体呈柱状结构。  相似文献

Whether the rumen microbes are able to synthesize and/or degrade long-chain alkanes in anaerobic conditions remains a question to be answered before these hydrocarbons can be confidently used as duodenal flow or rumen transit markers. In this context, an experiment in vitro was carried out to establish whether within a rumen liquor fermentation system, n-alkanes can be derived from de-waxed structures of the plant or from non-alkane wax components (long-chain fatty alcohols, long-chain fatty acids and esters), or may be metabolized by bacteria to other components or to shorter-chain hydrocarbons. Ryegrass was labelled with 14C in growth chambers under controlled conditions in order to use it as a substrate. The labelled material obtained was separated in three fractions: labelled alkanes, labelled de-waxed plant and labelled wax components without the alkanes. These fractions were used for three different incubations in vitro, which objectives were as follows: 1. To check whether rumen bacteria can synthesize alkanes from carbon structures other than waxes (e.g. sugars). 2. To verify whether rumen bacteria can metabolize the n-alkanes to other compounds. 3. To check whether rumen bacteria can synthesize n-alkanes from other carbon compounds from waxes. The results showed that there was neither bacterial synthesis nor metabolism of the n-alkanes in in vitro conditions.  相似文献

Alkylresorcinols (ARs) are bioactive compounds occurring in many members of the Poaceae, likely at or near the surface of various organs. Here, we investigated AR localization within the cuticular wax layers of rye (Secale cereale) leaves. The total wax mixture from both sides of the leaves was found to contain primary alcohols (71%), alkyl esters (11%), aldehydes (5%), and small amounts (<3%) of alkanes, steroids, secondary alcohols, fatty acids and unknowns. A homologous series of ARs (3%) was identified by GC-MS and comparison with a synthetic standard of nonadecylresorcinol. The alkyl side chains of the wax ARs contained odd numbers of carbons ranging from C19 to C27, with a prevalence of C21, C23 and C25. Waxes from both sides of the leaf, analyzed separately in a second experiment, comprised the same compound classes in similar relative amounts and with similar homolog patterns. Finally, the epicuticular and intracuticular wax layers were sampled separately from the abaxial side of the leaf. While ARs accounted for 2% of the intracuticular wax, they were not detectable in the epicuticular wax. The intracuticular wax was also slightly enriched in steroids, whereas the epicuticular layer contained more primary alcohols. All other wax constituents were distributed evenly between both wax layers.  相似文献

Wax esters extracted from human hair have been examined by capillary GC-MS and by nano electrospray ionization (ESI) mass spectrometry using a tandem quadrupole mass spectrometer. Initially, the wax esters were examined by capillary GC-MS using conventional means, thus revealing an incomplete chromatographic resolution of the complex array of >200 wax esters ranging from 28 to 40 carbons in length, including saturated/straight-chained, unsaturated/straight-chained, saturated/branched, and unsaturated/branched molecular species. ESI of wax esters produced ammonium adduct ions [M+NH4]+, and collisional activation of these ions formed abundant [RCO2H2]+ product ions. Wax esters containing a double bond in the fatty acyl or fatty alcohol portion of the molecule revealed identical behavior, suggesting little influence of the double bond on the ionization process or subsequent decomposition. The wax ester mixture was analyzed by ESI and tandem mass spectrometry using multiple reaction monitoring and neutral loss scanning. The neutral loss experiment [loss of NH3 and CH2=CH-(CH2)nCH3] was particularly effective at rapidly surveying the complex biological mixture, identifying>160 different wax esters that range from 24 to 42 total carbons.  相似文献

Long-chain aldehydes, alcohols, hydrocarbons and wax esters were major components of the external lipids of adult Aleyrodes singularis. In exuviae, acetate esters replaced the hydrocarbons as a major component. The major long-chain alcohol and aldehyde from adults were C32 and were essentially the exclusive components of the wax particles. The major alcohol from exuviae was C26 and the aldehydes were C26, C28, C30 and C32. The major acetate esters were C28 and C30 in both adults and exuviae. There were wax esters of similar carbon number in adults and exuviae although the exuviae had a greater amount of wax esters with unsaturated fatty acids. The fatty acid and alcohol composition of the wax esters differed markedly between adults and exuviae. Wax esters of adults had similar amounts of C16, C18, C20, C22 and C24 fatty acids while those from exuviae contained largely C16 and C18. The major alcohol in the wax esters of adults was C22 and those of exuviae were C26 and C28. The distribution of fatty acids and alcohols among wax esters of varying chain length also differed between adults and exuviae: in adults C22 was the major fatty acid found in the dominant wax ester, C44 and the C22 alcohol was the major alcohol and found in wax esters C42 and C44. In exuviae C16 and C18 were the major fatty acids found in most wax esters and a C28 alcohol was the major alcohol found in wax esters C44 and C46, the two dominant wax esters in exuviae. It was clear that the difference in chemistry of the wax esters between the adults and exuviae is not evident unless the acid and alcohol moieties are characterized.  相似文献
