Following a series of continuous exposures to 14CO2 for different lengths of time, leaves from Neurachne munroi (C4), N. minor (C3-C4) and N. tenuifolia (C3| were estimated to assimilate 100%, 9% and 2–4%, respectively, of atmospheric CO2 by the C4 pathway. The percentage of 14C-label appearing in malate and aspartate in leaves of N. minor progressively increased with longer exposure times indicating that a significant proportion of its C4 acids are formed as secondary products. In 14CO2/12CO2 pulse/chase experiments, the 14C-label in leaves of N. munroi was rapidly transferred from C4 acids to sugar monophosphates plus sugar diphosphates, and finally to sucrose. In leaves of N. minor, the 14C-label was slowly metabolized from the C-4 carboxyl of malate and asparate (apparent half-time = 250 s), and the formation of C4 acids as secondary products was again evident. 14C-label in serine/glycine accumulated to comparable magnitudes in both N. minor and in N. tenuifolia, but there was an initial lag phase in the accumulation of label in N. minor. C4 photosynthesis is apparently of minimal importance in reducing photorespiration in N. minor, but leaf anatomical specializations and a possible compartmentation of photorespiratory metabolism may be of considerable importance.  相似文献

Panicum milioides, a naturally occurring species with C4-like Kranz leaf anatomy, is intermediate between C3 and C4 plants with respect to photo-respiration and the associated oxygen inhibition of photosynthesis. This paper presents direct evidence for a limited degree of C4 photosynthesis in this C3-C4 intermediate species based on: (a) the appearance of 24% of the total 14C fixed following 4 s photosynthesis in 14CO2-air by excised leaves in malate and aspartate and the complete transfer of label from the C4 acids to Calvin cycle intermediates within a 15 s chase in 12CO2-air; (b) pyruvate- or alanine-enhanced light-dependent CO2 fixation and pyruvate stimulation ote- or alanine-enhanced light-dependent CO2 fixation and pyruvate stimulation of oxaloacetate- or 3-phosphoglycerate-dependent O2 evolution by illuminated mesophyll protoplasts, but not bundle sheath strands; and (c) NAD-malic enzyme-dependent decarboxylation of C4 acids at the C-4 carboxyl position, C4 acid-dependent O2 evolution, and 14CO2 donation from (4-14C)C4 acids to Calvin cycle intermediates during photosynthesis by bundle sheath strands, but not mesophyll protoplasts. However, P. milloides differs from C4 plants in that the activity of the C4 cycle enzymes is only 15 to 30% of a C4 Panicum species and the Calvin cycle and phosphoenolpyruvate carboxylase are present in both cell types. From these and related studies (Rathnam, C.K.M. and Chollet, R. (1979) Arch. Biochem. Biophys. 193, 346-354; (1978) Biochem. Biophys. Res. Commun. 85, 801-808) we conclude that reduced photorespiration in P. milioides is due to a limited degree of NAD-malic enzyme-type C4 photosynthesis permitting an increase in pCO2 at the site of bundle sheath, but not mesophyll, ribulose-bisphosphate carboxylase-oxygenase.  相似文献

1. The agranal bundle sheath chloroplasts of Sorghum bicolor possess very low Photosystem II activity compared with the grana-containing mesophyll chloroplasts.

2. Sorghum mesophyll chloroplasts have a chlorophyll (chl) and carotenoid composition similar to that of spinach chloroplasts. In contrast, the sorghum bundle sheath chloroplasts have a higher chl a/chl b ratio; they are enriched in β-carotene and contain relatively less xanthophylls as compared to sorghum mesophyll or spinach chloroplasts.

3. Sorghum mesophyll chloroplasts with 1 cytochrome f, 2 cytochrome b6 and 2 cytochrome b-559 per 430 chlorophylls have a cytochrome composition similar to spinach chloroplasts. Sorghum bundle sheath chloroplasts contain cytochrome f and cytochrome b6 in the same molar ratios as for the mesophyll chloroplasts, but cytochrome b-559 is barely detectable.

4. The chl/P700 ratios of mesophyll chloroplasts of S. bicolor and mesophyll and bundle sheath chloroplasts of Atriplex spongiosa are similar to that of spinach chloroplasts suggesting that these chloroplasts possess an identical photosynthetic unit size to that of spinach. The agranal bundle sheath chloroplasts of S. bicolor possess a photosynthetic unit which contains only about half as many chlorophyll molecules per P700 as found in the grana-containing chloroplasts.

5. The similarity of the composition of the bundle sheath chloroplasts of S. bicolor with that of the Photosystem I subchloroplast fragments, together with their smaller photosynthetic unit and low Photosystem II activities suggests that these chloroplasts are highly deficient in the pigment assemblies of Photosystem II.  相似文献

We have integrated two cDNAs expressing Sorghum photosynthetic phosphoenolpyruvate carboxylase (C4-PEPC) and NADP-malate dehydrogenase (cpMDH), two key enzymes involved in the primary carbon fixation pathway of NADP-malic enzyme-type C4 plants, separately or together into a C3 plant (potato). Analysis of the transgenic plants showed a 1.5-fold increase in PEPC and cpMDH activities compared to untransformed plants. Immunolocalization confirmed an increase at the protein level of these two enzymes in the transgenic plants and indicated that the Sorghum cpMDH was specifically addressed to the chloroplasts of potato mesophyll cells. However, integration of either or both of the cDNAs into the potato genome did not appear to significantly modify either tuber starch grain content or the rate of photosynthetic O2 production compared to control untransformed plants. The low level of transgene expression probably explains the lack of influence on carbon metabolism and photosynthetic rates. This general observation suggests that some complex mechanism may regulate the level of production of foreign C4 metabolism enzymes in C3 plants.  相似文献

1. Evidence is presented for high rates of carrier-mediated uptake of pyruvate into the stroma of intact mesophyll chloroplasts of the C4 plant Digitaria sanguinalis, but not the chloroplasts of the C3 plant Spinacea oleracea. Uptake of pyruvate in the dark with the C4 mesophyll chloroplasts was followed using two techniques: uptake of [14C]pyruvate as determined by silicon oil centrifugal filtration and uptake as indicated by absorbance changes at 535 nm (shrinkage/swelling) after addition of 0.1 M pyruvate salts.

2. Uptake of the pyruvate anion by an electrogenic carrier is suggested to be the major mode of transport. Chloroplast swelling was observed in potassium pyruvate plus valinomycin and uptake of [14C]pyruvate was inhibited by membrane-permeant anions. Valinomycin reduced uptake in the absence of external potassium and the inhibition could be reversed by addition of external potassium.

3. Uptake of pyruvic acid (or a pyruvate /OH antiport) is ruled unlikely since [14C]pyruvate uptake was relatively independent of the pH gradient across the envelope and addition of pyruvate to chloroplasts did not result in an alkalization of the medium. The low rate of swelling observed in ammonium pyruvate may be due to non-mediated permeation of pyruvic acid, which is possible only at high pyruvate concentrations.

4. The concentration of pyruvate in the stroma increased with external concentration over the range tested (up to 40 mM) but the concentration ratio (internal/external) was always less than one. The steady-state concentration of [14C]pyruvate in the stroma was dependent on the ionic strength of the medium, with saturation at roughly I = 0.04 M, while accumulation of the membrane-permeant cation tetraphenylmethylphosphonium decreased with increasing ionic strength. This suggests that ionic strength modifies a membrane potential (inside negative) across the envelope and that pyruvate uptake responds to the magnitude and direction of that potential (−80 mV at low ionic strength).

5. Chloride and inorganic phosphate were potent inhibitors of [14C]pyruvate uptake. Of the sulfhydryl reagents tested, N-ethylmaleimide was not inhibitory while mersalyl completely blocked [14C]pyruvate uptake and swelling in potassium pyruvate plus valinomycin. Pyruvate uptake, as measured by valinomycin induced swelling in potassium pyruvate, was highly temperature sensitive, with an energy of activation of 39 kcal/mol above 9 °C.

6. Phenylpyruvate, -ketoisovalerate, -ketoisocaproate, -cyano-4-hydroxycinnamic acid and -cyanocinnamic acid inhibited [14C]pyruvate but not [14C]-acetate uptake in the dark and also reduced pyruvate metabolism by the chloroplasts in the light.  相似文献

The North American tallgrass prairie is composed of a diverse mix of C3 and C4 plant species that are subject to multiple resource limitations. C4 grasses dominate this ecosystem, purportedly due to greater photosynthetic capacity and resource-use efficiency associated with C4 photosynthesis. We tested the hypothesis that intrinsic physiological differences between C3 and C4 species are consistent with C4 grass dominance by comparing leaf gas exchange and chlorophyll fluorescence variables for seven C4 and C3 herbaceous species (legumes and non-legumes) in two different settings: experimental mesocosms and natural grassland sites. In the mesocosms, C4 grasses had higher photosynthetic rates, water potentials and water-use efficiency than the C3 species. These differences were absent in the field, where photosynthetic rates declined similarly among non-leguminous species. Thus, intrinsic photosynthetic advantages for C4 species measured in resource-rich mesocosms could not explain the dominance of C4 species in the field. Instead, C4 dominance in this ecosystem may depend more on the ability of the grasses to grow rapidly when resources are plentiful and to tolerate multiple limitations when resources are scarce.  相似文献

1. Cyclic photophosphorylation driven by white light, as followed by 14CO2 fixation by mesophyll chloroplast preparations of the C4 plant Digitaria sanguinalis, was specifically inhibited by disalicylidenepropanediamine (DSPD), antimycin A, 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone (DBMIb), 1-ethyl-3(3-dimethylaminopropyl)-carbodiimide (EDAC), and KCN suggesting that ferredoxin, cytochrome b563, plastoquinone, cytochrome f, and plastocyanin are obligatory intermediates of cyclic electron flow. It was found that 0.2 μM DCMU and 40 μM o-phenanthroline blocked noncyclic electron flow, stimulated cyclic photophosphorylation, and caused a partial reversal (40–100%) of the inhibition by DBMIB and antimycin A, but not DSPD.

2. Cyclic photophosphorylation could also be activated using only far-red illumination. Under this condition, however, cyclic photophosphorylation was much less sensitive to the inhibitors DBMIB, EDAC and antimycin A, but remained completely sensitive to DSPD and KCN. Inhibition in far-red light was not increased by preincubating the chloroplasts with the various inhibitors for several minutes in white light.

3. The striking correspondence between the effects of photosystem II inhibitors, DCMU and o-phenanthroline, on cyclic photophosphorylation under white light and cyclic photophosphorylation under far-red light (in the absence of photosystem II inhibitors) suggests that electrons flowing from photosystem II may regulate the pathway of cyclic electron flow.  相似文献

Cp#2Yb (Cp#=C5H4(CH2)2NMe2) has been obtained by reaction of YbI2(THF)2 with 2 equiv. of C5H4(CH2CH2NMe2)K in THF. The X-ray structure analysis shows a bent structure with intramolecular coordination of both nitrogen atoms to ytterbium. The reaction of C60-fullerene with Cp#2Yb leads to the formation of the fullerenide derivative [Cp#2Yb]2C60, which shows an ESR signal in the solid state and in THF solution at room temperature (solid: ΔH = 50 G, G = 1.9992; solution: ΔH = 10 G, G = 2.0001) and a magnetic moment of 3.6 BM. The lutetium fullerenides CpLu(C60)(DME) (3) and Cp*Lu(C60)(DME)(C6H5CH3) (4), (Cp = η5−C5H5, Cp* = η5−C5Me5), were obtained by reaction of C60 with CpLu(C10H8) (DME) and Cp*Lu(C10H8) (DME) in toluene. Both complexes are paramagnetic (μeff = 1.4 and 0.9 BM) and exhibit temperature-dependent ESR signals (293 K: g = 1.992 and 2.0002 respectively).  相似文献

1. 1. The effect of the Mg2+ concentration on the CO2 fixation activity in situ in isolated and intact spinach chloroplasts upon suspension in hypotonic medium was examined. CO2 fixation in the dark was activated 25–100 fold by 20 mM Mg2+ in the presence of added ATP plus either ribulose 5-phosphate or ribose 5-phosphate. 20 mM Mg2+-stimulated fixation only 2–3 fold in the presence of the substrate of fixation, ribulose 1,5-diphosphate. The highest Mg2+-stimulated rate of fixation in the dark observed with chloroplasts was 480 μmoles CO2 fixed per mg chlorophyll per h.

2. 2. The concentration of bicarbonate at half of the maximal velocity (apparent Km) during the Mg2+-stimulated fixation of CO2 was 0.4 mM in the presence of ATP plus ribose 5-phosphate and 0.6 mM with ribulose 1,5-diphosphate.

3. 3. Dithioerythritol or light enhanced Mg2+-stimulated CO2 fixation 1–3 fold in the presence of ATP plus ribose 5-phosphate but not ribulose 1,5-diphosphate.

4. 4. These results indicate that Mg2+ fluxes in the stroma of the chloroplast could control the activity of the phosphoribulokinase with a lesser effect on the ribulosediphosphate carboxylase. An increase in Mg2+ of 6–10 mM in the stroma region of the chloroplast would be enough to activate CO2 fixation during photosynthesis.

The hydrothermal reactions of (Ph4P)[VO2Cl2] and H2C2O4 at 150 and 125°C yield (Ph4P)2[V2O2(H2O)2(C2O4)3]·4H2O (1) and (Ph4P)[VOCl(C2O4)] (2), respectively. The structure of the molecular anion of 1 consists of a binuclear unit of oxovanadium(IV) octahedra bridged by a bisbidentate oxalate group. The VO6 coordination geometry at each vanadium site is defined by a terminal oxo group, an aquo ligand, and four oxygen donors — two from the bisbidentate bridging oxalate and two from the terminal bidentate oxalate. The structure of 2 consists of discrete Ph4P+ cations occupying regions between [VOCl(C2O4)] spiral chains. The structure of the one-dimensional anionic chain exhibits V(IV) octahedra bridged by bisbidentate oxalate groups. Warming responses of photosynthesis and its temperature dependence in two C3 grass (Agropyron cristatum, Stipa krylovii), one C4 grass (Pennisetum centrasiaticum), and two C3 forb (Artemisia capillaris, Potentilla acaulis) species in a temperate steppe of northern China were investigated in a field experiment. Experimental warming with infrared heater significantly increased daily mean assimilation rate (A) in P. centrasiaticum and A. capillaris by 30 and 43%, respectively, but had no effects on other three species. Seasonal mean A was 13, 15, and 19% higher in the warmed than control plants for P. centrasiaticum, A. capillaries, and S. krylovii, respectively. The mean assimilation rate in A. cristatum and P. acaulis was not impacted by experimental warming. All the five species showed photosynthetic acclimation to temperature. The optimum temperature for photosynthesis (Topt) and the assimilation rate at Topt in the five species increased by 0.33–0.78 °C and 4–27%, respectively, under experimental warming. Elevated temperature tended to increase the maximum rate of ribulose-1,5-bisphosphate (RuBP) carboxylation (Vcmax) and the RuBP regeneration capacity (Jmax) in the C3 plants and carboxylation efficiency and the CO2-saturated photosynthetic rate in the C4 plant at higher leaf temperature, as well as the optimum temperatures for the four parameters. Our results indicated that photosynthetic responses to warming were species-specific and that most of the species in the temperate steppe of northern China could acclimate to a warmer environment. The changes in the temperature dependence of Vcmax and Jmax, as well as the balance of these two processes altered the temperature dependence of photosynthesis under climatic warming.  相似文献

The amino-functionalized metallocene (C5H4CH(Me)NMe2)2ZrCl2, [(CpN)2ZrCl2] was synthesized by salt metathesis of ZrCl4 and 2 equiv. of C5H4CH(Me)NMe2Li. The metallocene was obtained in good yield as a mixture of rac and meso diastereomers as established by NMR spectroscopy. The addition of 2 equiv. of n-BuLi to the metallocene (CpN)2ZrCl2 produced a co-catalyst system which was active, at a 1.0 mol% loading, in the dehydropolymerization of PhSiH3 to poly(phenylsilane), PPSi. The PPSi was obtained as a 9:1 linear–cyclic mixture (Mw=3850, Mn=2300) as established by GPC analysis; 29Si{1H} NMR spectroscopy revealed an atactic polymer microstructure.  相似文献

Mesophyll protoplasts and bundle sheath cells were prepared by enzymatic digestion of leaves of Alternanthera tenella, a C3-C4 intermediate species. The intercellular distribution of selected photosynthetic, photorespiratory and respiratory (mitochondrial) enzymes in these meso-phyll and bundle sheath cells was studied. The activity levels of photosynthetic enzymes such as PEP carboxylase (EC 4.1.1.31) or NAD-malic enzyme (EC 1.1.1.39) and photorespiratory enzymes such as glycolate oxidase (EC 1.1.3.1) or NADH-hydroxypyruvate reductase (EC 1.1.1.29) were similar in the two cell types. The activity levels of mitochondrial TCA cycle enzymes such as citrate synthase (EC 4.1.3.7) or fumarase (EC 4.2.1.2) were 2- to 3-fold higher in bundle sheath cells. On the other hand, the activity levels of mitochondrial photorespiratory enzymes, namely glycine decarboxylase (EC 2.1.2.10) and serine hydroxymethyltransferase (EC 2.1.2.1), were 6-9-fold higher in bundle sheath cells than in mesophyll protoplasts. Such preferential localization of mitochondria enriched with the glycine-decarboxylating system in the inner bundle sheath cells would result in efficient refixa-tion of CO2 from not only photorespiration but also dark respiration before its exit from the leaf. We propose that predominant localization of mitochondria specialized in glycine decarboxylation in bundle sheath cells may form the basis of reduced photorespiration in this C3-C4 intermediate species.  相似文献

For one group of C4 species we have proposed that the C4 acid decarboxylation phase of C4 photosynthesis proceeds via a NAD ‘malic’ enzyme located in bundle sheath mitochondria. The present studies with Atriplex spongiosa demonstrate the capacity of isolated mitochondria and bundle sheath cell strands to decarboxylate malate at rates commensurate with an integral role in photosynthesis. With bundle sheath cells, rates of H14CO3? fixation into Calvin cycle intermediates and evolution of O2 when HCO3? was added, were above 2 μmoles/min/mg chlorophyll. Similar rates of O2 evolution resulted from the addition of C4 acids, and the C-4 carboxyl of malate was rapidly assimilated into photosynthetic intermediates and products.  相似文献

The characteristic surface lipid compositions of several C3 and C4 plants are discussed. C4 plants produce surface lipids (epicuticular waxes) made up of the ubiquitous classes of aliphatic compounds: free fatty acids, aldehydes, primary alcohols, alkanes and aliphatic linear esters. C3 plants synthesize surface lipids comprising the ubiquitous classes and either of the two following groups of compound: (i) lβ-diketones, hydroxy lβ-diketones, alkan-2-ol esters; (il) ketones and secondary alcohols with the functional group in the middle of the hydrocarbon chain. These features are suggested to represent physioIogical characteristics of the plant and to be related to ecological adaptations. Wax class compositions might also be an ancillary method for defining the C3 or C4 mechanism of CO2 assimilation in cases where uncertainty exists.  相似文献

Reactions of [Rh(COD)Cl]2 with the ligand RN(PX2)2 (1: R = C6H5; X = OC6H5) give mono- or disubstituted complexes of the type [Rh2(COD)Cl22−C6H5N(P(OC6H5)2)2}] or [RhCl{ν2−C6H5 N(P(OC6H5)2)2 }]2 depending on the reaction conditions. Reaction of 1 with [Rh(CO)2Cl]2 gives the symmetric binuclear complex, [Rh(CO)Cl{μ−C6H5N(P(OC6H5)2)2} 2, whereas the same reaction with 2 (R = CH3; X = OC6H5) leads to the formation of an asymmetric complex of the type [Rh(CO)(μ−CO)Cl{μ−CH3N(P(OC6H5)2)2}2 containing both terminal and bridging CO groups. Interestingly the reaction of 3 (R = C6H5, X = OC6H4Br−p with either [Rh(COD)Cl]2 or [Rh(CO)2Cl]2 leads only to the formation of the chlorine bridged binuclear complex, [RhCl{ν2−C6H5N(P(OC6H4Br−p)2)2}]2. The structural elucidation of the complexes was carried out by elemental analyses, IR and 31P NMR spectroscopic data.  相似文献

1. Circular dichroism spectra of the cytochromes in membrane fragments derived from sonicated beef heart mitochondria have been obtained in the wavelength region 400–480 nm in which the major absorbance maxima of the heme prosthetic groups are found.

2. 2. Cytochrome oxidase in the mitochondrial membrane fragments has a band of positive ellipticity at 426 nm in the oxidized form and a pronounced band of positive ellipticity at 445 nm in the reduced form. The reduced-minus-oxidized difference molar ellipticity at 445 nm, Δ[θ]445 is 3.0·105 degree·cm−2·dmole−1 heme a for membrane-bound oxidase compared to 1.6·105 degree·cm−2·dmole−1 heme a for the purified oxidase. The membrane-bound oxidase in the reduced form also appears to have a band of negative ellipticity at 426 nm not found in the purified oxidase.

3. 3. When reduced with succinate in the presence of cyanide and oxygen, cytochrome oxidase in the membrane fragments has a positive band at 442 nm very similar to that observed with the purified oxidase.

4. 4. Cytochrome c, which has a positive band at 426 nm in the purified form when reduced, appears to have a negative band at this wavelength in the mito-chondrial membrane fragments which contributes to the pronounced negative band at 426 nm observed in the membrane fragments reduced with succinate in anaerobiosis. There is no evidence for a contribution to the CD spectra of the membrane fragments from cytochrome c1 or from cytochrome b561 in either the oxidized or the reduced form.

Cytochrome b566 in the mitochondrial membrane fragments has no detectable CD spectrum in the oxidized form, but has a small positive band at 427 nm and a small negative band at 436 nm in the reduced form. The same CD spectrum is observed with cytochrome b566 reduced with succinate in the presence of antimycin A or 2-heptyl-4-hydroxyquinoline-N-oxide. The same increase in positive ellipticity is observed at 427 nm in the mitochondrial membrane fragments, treated with oligomycin to restore energy coupling, when cytochrome b566 is reduced with succinate in the energized membrane, as is observed in the inhibitor-treated membrane fragments. The absence of a pronounced conformational change in cytochrome b566 on energization, as revealed by its CD spectrum, favors the concept that its reduction by succinate in the energized state is due to reversed electron transport rather than an intrinsic shift in the cytochrome's midpoint redox potential.

New mixed metal complexes SrCu2(O2CR)3(bdmap)3 (R = CF3 (1a), CH3 (1b)) and a new dinuclear bismuth complex Bi2(O2CCH3)4(bdmap)2(H2O) (2) have been synthesized. Their crystal structures have been determined by single-crystal X-ray diffraction analyses. Thermal decomposition behaviors of these complexes have been examined by TGA and X-ray powder diffraction analyses. While compound 1a decomposes to SrF2 and CuO at about 380°C, compound 1b decomposes to the corresponding oxides above 800°C. Compound 2 decomposes cleanly to Bi2O3 at 330°C. The magnetism of 1a was examined by the measurement of susceptibility from 5–300 K. C3 photosynthesis is an inefficient process, because the enzyme that lies at the heart of the Benson–Calvin cycle, ribulose 1,5-bisphosphate carboxylase-oxygenase (Rubisco) is itself a very inefficient enzyme. The oxygenase activity of Rubisco is an unavoidable side reaction that is a consequence of its reaction mechanism. The product of oxygenation, glycollate 2-P, has to be retrieved by photorespiration, a process which results in the loss of a quarter of the carbon that was originally present in glycollate 2-P. Photorespiration therefore reduces carbon gain. Purely in terms of carbon economy, there is, therefore, a strong selection pressure on plants to reduce the rate of photorespiration so as to increase carbon gain, but it also improves water- and nitrogen-use efficiency. Possibilities for the manipulation of plants to decrease the amount of photorespiration include the introduction of improved Rubisco from other species, reconfiguring photorespiration, or introducing carbon-concentrating mechanisms, such as inorganic carbon transporters, carboxysomes or pyrenoids, or engineering a full C4 Kranz pathway using the existing evolutionary progression in C3–C4 intermediates as a blueprint. Possible routes and progress to suppressing photorespiration by introducing C4 photosynthesis in C3 crop plants will be discussed, including whether single cell C4 photosynthesis is feasible, how the evolution of C3–C4 intermediates can be used as a blueprint for engineering C4 photosynthesis, which pathway for the C4 cycle might be introduced and the extent to which processes and structures in C3 plant might require optimisation.  相似文献

1. 1. Cyanide inhibits the catalytic activity of cytochrome aa3 in both polarographic and spectrophotometric assay systems with an apparent velocity constant of 4·103 M−1·s−1 and a Ki that varies from 0.1 to 1.0 μM at 22 °C, pH 7·3.

2. 2. When cyanide is added to the ascorbate-cytochrome c-cytochromeaa3−O2 system a biphasic reduction of cytochrome c occurs corresponding to an initial Ki of 0.8 μM and a final Ki of about 0.1 μM for the cytochrome aa3−cyanide reaction.

3. 3. The inhibited species (a2+a33+HCN) is formed when a2+a33+ reacts with HCN, when a2+a32+HCN reacts with oxygen, or when a3+a33+HCN (cyano-cytochrome aa3) is reduced. Cyanide dissociates from a2+a33+HCN at a rate of 2·10−3 s−1 at 22 °C, pH 7.3.

4. 4. The results are interpreted in terms of a scheme in which one mole of cyanide binds more tightly and more rapidly to a2+a33+ than to a3+a33+.
