The nucleotide composition, relative concentration of pyrimidine clusters, and the degree of methylation of the mitochondrial and nuclear DNA's of various vertebrates and the protozoan Crithidia oncopelti have been studied. With respect to the relative concentration of GC pairs, the mtDNA of animals (bull, rat) does not differ from the corresponding nDNA. The relative concentration of GC pairs in the mtDNA of certain fish and birds is 1.5-2.5 mole% higher than in the respective nDNA. The kinetoplast DNA of the protozoan C. oncopelti (where the relative concentration of the GC pairs is 42.9 mole %) differs very sharply in composition from the nDNA (where the relative concentration of GC pairs is 51.3 mole %). The mtDNA's and kDNA's studied are distinguished from the respective nDNA'S by a lower degree of clustering of pyrimidine nucleotides. The proportion of mono- and dipyrimidine fragments in the mtDNA and kDNA is 30 mole %, while in the nDNA it does not exceed 23 mole %. The relative concentration of long pyrimidine clusters (hexapyrimidine clusters of larger) in the mtDNA is smaller than in the nDNA by a factor of 2-5. The low degree of clustering of the pyrimidine nucleotides is apparently characteristic of all the known mtDNA's and may support the fact that they have a single type of organization and are of a single origin. All the vertebrate mtDNA's studied contain 5-methylcytosine as a minor base (1.5-3.15 mole %), and their level of methylation is 1.5-2 times greater than that in the respective nDNA's. It has been shown that animals display species specificity with respect to the 5-methylcytosine content in the mtDNA. Its distribution among the pyrimidine clusters in the bovine heart mtDNA differs substantially from that in the nDNA. This suggests that the methylation specificities of nuclear and mitochondrial DNA are different. A DNA methylase, which effects the in vitro methylation of cytosine residues both in the homologous mtDNA and in different heterologous DNA's, has been found in rat liver and bovine heart mitochondria. The specificity of the in vitro methylation of the cytosine residues in the same heterologous Escherichia coli B DNA by the nuclear and mitochondrial enzymes is different: The mitochondrial enzyme methylates predominantly in monopyrimidine fragments, and the nuclear enzyme methylates mostly in di- and tripyrimidine fragments. In recent years, it has become increasingly evident that there is a genetic component to alcoholism. Attempts to isolate alcoholism genes have met with modest success, in part because alcoholism is a multigenic trait. Recently, experimental animal models and novel genetic manipulations have provided several clues as to the specific genes involved in alcoholism, and extensive research has identified many genes that might influence responses to alcohol. Although not all of these might be proven to influence drug sensitivity, research has provided evidence for the involvement of a few genes. Ultimately, findings from animal models that investigate the function of specific genes could aid the development of pharmacotherapies to treat alcohol dependence.  相似文献

In the past decade, the development of new DNA, RNA, and protein technologies has greatly incremented the knowledge about the organization and expression of mitochondrial DNA. The complete base sequence of mitochondrial DNA of several animals is known and many data are rapidly accumulating on the mitochondrial genomes of other systems. Here we discuss the results so far obtained that disclosed unexpected features of mitochondrial genetics. Furthermore, mitochondrial DNA has become established as a powerful tool for evolutionary studies in animals. Evidences are presented demonstrating that the evolution of mitochondrial DNA has proceeded in different ways in the various taxonomic groups. Data on heteroplasmic animals, which demonstrate the rapid evolution of mitochondrial DNA, are also presented.  相似文献

This paper presents an asymmetric game-theoretical model to the following type of animal conflicts: a member of a group is at risk and needs the help of another member to be saved. As long as assistance is not provided, this individual has a positive, time-dependent rate of dying. Assisting the individual which is at risk accrues a cost, but losing it decreases each member's inclusive fitness. A potential helper's interval between the moment a group member gets into trouble and the moment it assists is a random variable, hence its strategy is to choose the distribution of this random variable. In the asymmetric conflict all the potential helpers have identical strategy sets, but each plays a different role. For example, male or female and young or old. We consider both payoff-irrelevant asymmetry and payoff-relevant asymmetry and characterize each role's stable replies. The evolutionarily stable strategies (ESS) are computed, and the model is applied to the n brothers' problem. According to our results immediate assistance and no assistance are possible ESS both under payoff-relevant asymmetry and under payoff-relevant asymmetry.  相似文献

A morphometric study of the common peroneal nerve in early experimental diabetes in rats showed that fibre size was diminished. The reduction in the size of the axon was twice that of the myelin sheath. This may contribute to the understanding of the impaired motor conduction velocity found in diabetics shortly after the onset of their disease.  相似文献

Animal conflicts are often characterized by time-dependent strategy sets. This paper considers the following type of animal conflicts: a member of a group is at risk and needs the assistance of another member to be saved. As long as assistance is not provided, the individual which is at risk has a positive, time-dependent rate of dying. Each of the other group members is a potential helper. Assisting this individual accrues a cost, but losing him decreases the inclusive fitness of each group member. A potential helper's interval between the moment an individual finds itself at risk and the moment it assists is a random variable, hence its strategy is to choose the probability distribution for this random variable. Assuming that each of the potential helpers knows the others' strategies, we show that the ability to observe their realizations influences the evolutionarily stable strategies (ESS) of the game. According to our results, where the realizations can be observed ESS always exist: immediate assistance, no assistance and delayed assistance. Where the realizations cannot be observed ESS do not always exist, immediate assistance and no assistance are possible ESS, while delayed assistance cannot be an ESS. We apply our model to the n brothers' problem and to the parental investment conflict.  相似文献

Replacing animal procedures with methods such as cells and tissues in vitro, volunteer studies, physicochemical techniques and computer modelling, is driven by legislative, scientific and moral imperatives. Non-animal approaches are now considered as advanced methods that can overcome many of the limitations of animal experiments. In testing medicines and chemicals, in vitro assays have spared hundreds of thousands of animals. In contrast, academic animal use continues to rise and the concept of replacement seems less well accepted in university research. Even so, some animal procedures have been replaced in neurological, reproductive and dentistry research and progress is being made in fields such as respiratory illnesses, pain and sepsis. Systematic reviews of the transferability of animal data to the clinical setting may encourage a fresh look for novel non-animal methods and, as mainstream funding becomes available, more advances in replacement are expected.  相似文献
