Many comparative physiological studies aim to determine if a particular species differs from a prediction based on a linear allometric regression for other species. However, the judgment as to whether the species in question conforms to this allometric relationship is often not based on any formal statistical analysis. An appropriate statistical method is to compare the new species’ value with the 95% confidence limits for predicting an additional datum from the relationship for the other species. We examine the basal metabolic rate (BMR) of the termitivorous numbat (Myrmecobius fasciatus) and aardwolf (Proteles cristatus) to demonstrate the use of the 95% prediction limits to determine statistically if they have a lower-than-expected BMR compared to related species. The numbat’s BMR was 83.6% of expected from mass, but fell inside the 95% prediction limits for a further datum; a BMR < 72.5% of predicted was required to fall below the one-tail 95% prediction limits. The aardwolf had a BMR that was only 74.2% of predicted from the allometric equation, but it also fell well within the 95% prediction limits; a BMR of only 41.8% of predicted was necessary to fall below the one-tail 95% prediction limits. We conclude that a formal statistical approach is essential, although it is difficult to demonstrate that a single species statistically differs from a regression relationship for other species.  相似文献

DNA variability was investigated in the last intron of the Y-chromosome-specific zinc finger gene, ZFY, and its X homolog on Xp21.3, ZFX. No polymorphisms were found in the 676-bp ZFY segment in a sample of 205 world-wide-distributed Y chromosomes, other than a solitary nucleotide variant in one individual (nucleotide diversity pi = 0.0014%). In contrast, 10 segregating sites (pi = 0.082%) were identified within 1,089 bp of the ZFX sequence in a sample of 336 X chromosomes. Four of these polymorphisms, which contributed most of the diversity, were located within an Alu insert disrupting the ZFY-ZFX homology (pi Alu = 0.24%). The diversity in the homologous portion of the ZFX intron, although higher than that in ZFY, was lower than that found in genomic segments believed to evolve neutrally; interspecies divergence in both segments was also reduced. Although this suggests that the evolution of both ZFY and ZFX homologs may not be entirely neutral, both Tajima and HKA tests did not reject neutrality. The lack of statistical significance may be attributed to a lack of power in these tests (the low divergence and variability values reduce the power of the HKA and Tajima tests, respectively); furthermore, Homo sapiens has recently undergone a rapid population growth, and selection is more difficult to detect in an expanding population. Therefore, the failure to reject neutrality does not necessarily indicate the absence of selection. In this context, the phylogenetic argument was given more weight in out interpretations. The high level of sequence identity in ZFY and ZFX segments, in spite of their separation 80-130 MYA, reflects a lower mutation rate as compared with other segments believed to undergo unconstrained evolution. Thus, the possibility of weak selection contributing to the low level of nucleotide diversity in the last ZFY intron cannot be excluded and should be kept in mind in the population genetics studies based on Y chromosome variability.  相似文献

Peripheral blood and tissue eosinophilia characterize trichinellosis in humans, and present in addition to the increased total IgE levels that occur in many helminth infections. Both processes are the consequence of T-helper 2 activation. Blood and tissue eosinophilia begins with eosinophilopoiesis in the bone marrow, which is followed by the migration of eosinophils through the circulatory system, the eosinophil infiltration of tissues at the inflammatory foci and, finally, degranulation and cell death. Recently, some aspects of eosinophilia caused by Trichinella spiralis infection have been elucidated; however, the protective role of this population of cells against Trichinella parasites remains controversial. Furthermore, when eosinophils are numerous, they can be toxic for host tissues. This review discusses these issues in both human and rodent infection models.  相似文献

Agriculture originated independently in many distinct regions at approximately the same time in human history. This synchrony in agricultural origins indicates that a global factor may have controlled the timing of the transition from foraging to food-producing economies. The global factor may have been a rise in atmospheric CO2 from below 200 to near 270 μol mol?1 which occurred between 15,000 and 12,000 years ago. Atmospheric CO2 directly affects photosynthesis and plant productivity, with the largest proportional responses occurring below the current level of 350 μol mol?1. In the late Pleistocene, CO2 levels near 200 μol mol?1 may have been too low to support the level of productivity required for successful establishment of agriculture. Recent studies demonstrate that atmospheric CO2 increase from 200 to 270 μol mol?1 stimulates photosynthesis and biomass productivity of C3 plants by 25% to 50%, and greatly increases the performance of C3 plants relative to weedy C4 competitors. Rising CO2 also stimulates biological nitrogen fixation and enhances the capacity of plants to obtain limiting resources such as water and mineral nutrients. These results indicate that increases in productivity following the late Pleistocene rise in CO2 may have been substantial enough to have affected human subsistence patterns in ways that promoted the development of agriculture. Increasing CO2 may have simply removed a productivity barrier to successful domestication and cultivation of plants. Through effects on ecosystem productivity, rising CO2 may also have been a catalyst for agricultural origins by promoting population growth, sedentism, and novel social relationships that in turn led to domestication and cultivation of preferred plant resources.  相似文献

Isolated heart preparations of frog and rat were used to test the validity of the adrenoceptor interconversion hypothesis. This hypothesis claims that low temperature converts the inotropic beta-adrenoceptors in isolated frog and rat heart to alpha-adrenoceptors. The present results do not support the adrenoceptor interconversion hypothesis. In the isolated frog ventricle, lowering the temperature from 24 C to 14 C did not significantly alter the inotropic potency of the sympathomimetic drugs isoprenaline, epinephrine, and phenylephrine and did not reduce the potency of the beta-adrenoceptor blocking drug propranolol as an epinephrine antagonist. In the isolated rat left atrium, lowering the temperature from 31 C to 17-19 C did not significantly change the inotropic potency of isoprenaline, norepinephrine and phenylephrine, did not diminish the potency of propranolol, and did not increase the potency of the alpha-adrenoceptor blocking drug phentolamine.--Benfey, B. A new model for the catabolism of very low density lipoprotein will be proposed following a brief discussion of the relevant information about its metabolism. This model is based on the assumption that each very low density lipoprotein derivative has a distinct structural feature which determines its biological fate. Since this unique determinant may reside in only a small portion of the particles, such as apoprotein, the different very low density lipoprotein derivatives may possess similar or even identical electrophoretic mobility or hydrated density. For this reason, we propose that very low density lipoprotein derivatives be defined according to their putative position in the metabolic pathways rather than according to their hydrated density or their electrophoretic behavior. Thus it is recommended that biochemical separation methods based on principles other than electrophoretic mobility or hydrated density be used to isolate, purify and define very low density lipoprotein derivatives. The position that a lipoprotein particle occupies in the catabolic pathways should be determined by its ability to interact with liver cells or its ability to become converted to low density lipoprotein. This new nomenclature would eliminate unnecessary confusion and stimulate more research toward elucidating the unique structural feature of each very low density lipoprotein derivative.  相似文献

According to the 'mental time travel hypothesis' animals, unlike humans, cannot mentally travel backwards in time to recollect specific past events (episodic memory) or forwards to anticipate future needs (future planning). Until recently, there was little evidence in animals for either ability. Experiments on memory in food-caching birds, however, question this assumption by showing that western scrub-jays form integrated, flexible, trial-unique memories of what they hid, where and when. Moreover, these birds can adjust their caching behaviour in anticipation of future needs. We suggest that some animals have elements of both episodic-like memory and future planning.  相似文献

Natural variation and changing climate in coastal oceans subject meroplanktonic organisms to broad ranges of pH and oxygen ([O2]) levels. In controlled‐laboratory experiments we explored the interactive effects of pH, [O2], and semidiurnal pH fluctuations on the survivorship, development, and size of early life stages of two mytilid mussels, Mytilus californianus and M. galloprovincialis. Survivorship of larvae was unaffected by low pH, low [O2], or semidiurnal fluctuations for both mytilid species. Low pH (<7.6) resulted in delayed transition from the trochophore to veliger stage, but this effect of low pH was absent when incorporating semidiurnal fluctuations in both species. Also at low pH, larval shells were smaller and had greater variance; this effect was absent when semidiurnal fluctuations of 0.3 units were incorporated at low pH for M. galloprovincialis but not for M. californianus. Low [O2] in combination with low pH had no effect on larval development and size, indicating that early life stages of mytilid mussels are largely tolerant to a broad range of [O2] reflective of their environment (80–260 μmol kg?1). The role of pH variability should be recognized as an important feature in coastal oceans that has the capacity to modulate the effects of ocean acidification on biological responses.  相似文献

Radiation-induced bystander mutagenesis at a relatively low dose range was investigated using low LET β-particles in a three-dimensional cell culture model. CHO cells were labeled with 0, 0.5, 1.0 or 5.0 μCi tritiated thymidine (3HdTTP) for 12 h and subsequently incubated with AL cells for 24 h at 11°C. The cell mixture was centrifuged to produce a spheroid of 4 × 106 cells of which there was five times more AL than CHO cells. The short-range β-particles emitted by 3HdTTP result in self-irradiation of labeled CHO cells, thus biological effects on neighboring AL cells can be attributed to the bystander response. To evaluate such response, non-labeled bystander AL cells were isolated from among labeled CHO cells and studied independently for survival and mutagenesis. Treatment of CHO cells with 3HdTTP resulted in a dose-dependent increase in bystander mutation incidence among neighboring AL cells compared to controls. In addition, multiplex PCR analysis revealed the types of mutants to be significantly different from those of spontaneous origin. These data provide evidence that low dose low LET radiation can induce bystander mutagenesis in a three-dimensional model. The results of this study will address the relevant issues of actual target size and radiation quality, and are likely to have a significant impact on our current understanding of radiation risk assessment.  相似文献

Abstract

The adenylate cyclase-stimulatory β2-adrenergic receptor has been purified to apparent homogeneity from hamster lung. Partial amino acid sequence obtained from isolated CNBr peptides was used to clone the gene and cDNA for this receptor. The predicted amino acid sequence for the hamster β2-adrenergic receptor revealed that the protein consists of a single polypeptide chain of 418 aa with consensus N-glycosylation and phosphorylation sites predicted by previous in vitro data. The most striking feature of the receptor protein however, is that it contains seven stretches of hydrophobic residues similar to the proposed seven transmembrane segments of the light receptor rhodopsin. Significant amino acid homology (30-35%) can be found between the hamster β2-adrenergic receptor and rhodopsin within these putative membrane spanning regions. Using a hamster β2-adrenergic receptor probe, the gene and cDNA for the human β2-adrenergic receptor were isolated, revealing a high degree of homology (87%) between the two proteins from different species. Unlike the genes encoding the family of opsin pigments, of which rhodopsin is a member, the genes encoding both hamster and human β2-adrenergic receptors are devoid of introns in their coding as well as 5′ and 3′ untranslated nucleotide sequences. The cloning of the genes and the elucidation of the aa sequences for these G-protein coupled receptors should help to determine the structure-function as well as the evolutionary relationship of these proteins.  相似文献

This review summarizes and discusses methodological approaches for studies on the impact of plant roots on the surrounding rhizosphere and for elucidation of the related mechanisms, covering a range from simple model experiments up to the field scale. A section on rhizosphere sampling describes tools and culture systems employed for analysis of root growth, root morphology, vitality testing and for monitoring of root activity with respect to nutrient uptake, water, ion and carbon flows in the rhizosphere. The second section on rhizosphere probing covers techniques to detect physicochemical changes in the rhizosphere as a consequence of root activity. This comprises compartment systems to obtain rhizosphere samples, visualisation techniques, reporter gene approaches and remote sensing technologies for monitoring the conditions in the rhizosphere. Approaches for the experimental manipulation of the rhizosphere by use of molecular and genetic methods as tools to study rhizosphere processes are discussed in a third section. Finally it is concluded that in spite of a wide array of methodological approaches developed in the recent past for studying processes and interactions in the rhizosphere mainly under simplified conditions in model experiments, there is still an obvious lack of methods to test the relevance of these findings under real field conditions or even on the scale of ecosystems. This also limits reliable data input and validation in current rhizosphere modelling approaches. Possible interactions between different environmental factors or plant-microbial interactions (e.g. mycorrhizae) are frequently not considered in model experiments. Moreover, most of the available knowledge arises from investigations with a very limited number of plant species, mainly crops and studies considering also intraspecific genotypic differences or the variability within wild plant species are just emerging.  相似文献

ABSTRACT

Animal welfare concerns have plagued the professional zoo and aquarium field for decades. Societal differences remain concerning the well-being of animals, but it appears a shift is emerging. Scientific studies of animal welfare have dramatically increased, establishing that many previous concerns were not misguided public empathy or anthropomorphism. As a result, both zoo and aquarium animal welfare policy and science are now at the center of attention within the world’s professional zoos and aquariums. It is now possible to view a future that embraces the well-being of individual captive exotic animals, as well as that of their species, and one in which professional zoos and aquariums are dedicated equally to advancing both. Though the ethics of keeping exotic animals and animals from the wild in captivity are still a contentious subject both outside and even within the profession, this study argues. We argue that this path forward will substantially improve most zoo and aquarium animals' welfare and could significantly reduce societal concerns. If animal welfare science and policy are strongly rooted in compassion and embedded in robust accreditation systems, the basic zoo/aquarium paradigm will move toward a more thoughtful approach to the interface between visitors and animals. It starts with a fundamental commitment to the welfare of individual animals.  相似文献

The first health effect of cadmium (Cd) was lung damage, reported in workers already in the 1930's, while bone effects and proteinuria were reported in the 1940's. After World War II, a bone disease with fractures and severe pain, the itai-itai disease, a form of Cd-induced renal osteomalacia, was identified in Japan. Subsequently, the toxicokinetics and toxicodynamics of Cd were described including its binding to the protein metallothionein. International warnings of health risks from cadmium pollution were issued in the 1970's. WHO, 1992, identified renal dysfunction as the critical effect and a crude quantitative evaluation was presented. In the 1990's population groups in China exposed to Cd via rice were studied and new information on skeletal, renal and reproductive toxicity of Cd was obtained in the ChinaCad project. There was a decrease in Bone Mineral Density (BMD), an increased prevalence of fractures and an increased urinary content of marker proteins of renal dysfunction among persons with long term exposure to Cd. The development of such biomarkers can be seen as a result of applied 'proteomics' research. Variation in metallothionein gene expression was related to development of renal dysfunction, supporting the usefulness of this 'genomic' approach. The ongoing rapid development of 'genomics' and 'proteomics' technologies will improve possibilities for molecular epidemiology studies in the future, providing an even better basis for preventive action. In many countries, Cd exposures are now under better control than in the past. The target for the 21st century is to achieve a totally acceptable exposure situation without adverse health effects from Cd.  相似文献
