Bel-W3 Tobacco, which is highly sensitive to ozone, was grown in two glass chambers and exposed to the ambient air at the periphery of Tel-Aviv, during winter, spring, summer and autumn 1978. During the exposure time, atmospheric ozone was continuously measured by a chemiluminescent monitor. Throughout the experiments, plants' height was measured and the number of leaves was determined three times weekly. The extent of injury to the tobacco plants was measured by the percentage of injured plants, the percentage of injured leaves and the percentage of leaves' area injured. Necrotic lesions, typical for ozone injury, appeared on the mature leaves of the exposed tobacco plants in three out of four exposures. Appearance of incipient injury differed among the experiments and depended not only on exposure duration and on ozone concentrations, but also on the exposure conditions (like light intensity, temperature and humidity), which considerably influenced the appearance of the injury. The percentage of injured leaves and the percentage of leaves' area injured, increased with the duration of exposure and with rising cumulative ozone concentrations.Presented at the Eighth International Congress of Biometeorology 9–14 September 1979, Shefayim, Israel.  相似文献

烟草的花蜜腺位于子房基部 ,围绕子房 ,属于子房蜜腺。蜜腺由分泌表皮和泌蜜组织组成 ,分泌表皮角质膜厚薄不匀 ,表皮上分布少量气孔器 ,气孔凹陷 ,孔下室不明显 ,泌蜜组织细胞多层。蜜腺邻接子房壁维管束 ,本身没有维管组织。蜜腺由子房基部的外壁表皮及其相邻的内侧细胞经分裂、生长、分化而来 ,在发育过程中 ,细胞中的液泡和淀粉粒都呈现一定的消长规律。原蜜汁由子房壁维管束提供 ,经过泌蜜组织细胞加工后 ,蜜汁通过气孔和薄的角质膜处泌出。  相似文献

D-type cyclins (CycD) are thought to control the onset of cell division and responses to extracellular signals during G1 phase. In this study a cyclin D3 (CycD3) gene was isolated from Nicotiana tabacum. Southern blot analysis indicated that it was present in a single-copy in the N. tabacum genome, and Northern analysis showed that it was only expressed in organs and tissues containing dividing cells. Exposure to cold and salt stresses resulted in decreased CycD3 expression supporting the view that its expression is closely related to cell growth.  相似文献

Salicylic acid content and benzoic acid 2-hydroxylase (BA2H) activity were investigated in tobacco Bel-W3 and Bel-B leaves after exposure to tropospheric ozone in the conditions of ambient air. Plants were exposed in accordance with a standard methodology for ozone biomonitoring, in a three-year experiment. Free salicylic acid (SA), conjugated with glucose (SAG), and as a product of the BA2H activity was quantified with HPLC. In order to evaluate ozone injuries of leaves, an open source image analysis software was employed. Plants exposure to ambient ozone resulted in enhanced BA2H activity and intensified salicylic acid biosynthesis in leaves of Bel-W3 cultivar showing visible ozone injuries. The BA2H activity significantly correlated with SAG for ozone-exposed Bel-W3 plants. Both injuries and salicylic acid biosynthesis rate depended on the growth phase of leaves and nearly linear correlation between SA content and injuries was found for particular leaves of Bel-W3.  相似文献

Immunoglobulin light and heavy chains are synthesized in mammalian cells as precursors containing a signal peptide. Processing and assembling result in formation of active antibodies. Chimeric genes have been made containing the coding sequence of the barley -amylase signal peptide which has been fused to cDNAs coding for either the mature light or the mature heavy chain of a monoclonal antibody. A plasmid was constructed linking both chimeric genes under the control of plant active promoters in an expression cassette. This DNA fragment was stably integrated into the genome of Nicotiana tabacum by Agrobacterium tumefaciens mediated gene transfer. Synthesis of light and heavy chains and assembly to antibodies was detected in transgenic tobacco tissue using specific secondary antibodies. By electron microscopic immunogold labeling, the presence of assembled antibody could be detected within the endoplasmic reticulum. Affinity chromatography indicated biological activity of the assembled immunoglobulin produced in plant cells. Unexpectedly, a significant amount of assembled antibodies was found within chloroplasts.  相似文献

Organogenesis in thin cell layers of Nicotiana tabacum L. was studied in relation to the effects of natural and synthetic auxins in combination with various cytokinins. All cytokinins tested, benzyladenine (BA), kinetin, zeatin (Z), zeatin riboside (ZR), N62-isopentenyl) adenine (IPA), dihydrozeatin [(diH)Z] and dihydrozeatin riboside [(diH)ZR], seem to be active in flower bud formation. In addition to the initiation of flower buds, vegetative buds or roots were also formed on the explants in the presence of BA, Z or IPA as exogenous cytokinins. Only dihydrozeatin and its riboside stimulated the initation of flower buds alone (as is known for kinetin), especially if supplemented with indole-3-acetic acid (IAA) as exogenous auxin. A high number of explants with flower buds was also found with high cytokinin/2,4-D ratios. In these conditions the presence of (diH)Z yielded the higest number of flower buds per explant.  相似文献

Tobacco (Nicotiana tabacum) serve as the top leading commercial, non-food, and model crop worldwide. Cyclic nucleotide-gated channels (CNGCs) are ligand-gated, calcium-permeable, divalent, cation-selective channels, involved in important biological functions. Here, we systematically characterized thirty-five CNGC genes in the genome of Nicotiana tabacum, and classified into four phylogenetic groups. Evolutionary analysis showed that NtabCNGC family of N. tabacum originated from the parental genome of N. sylvestris and N. tomentosiformis, and further expanded via tandem and segmental duplication events. Tissue-specific expression analysis showed that twenty-three NtabCNGC genes are involved in the development of various tobacco tissues. Subsequent RT-qPCR analyses indicated that these genes are sensitive towards external abiotic and biotic stresses. Notable performances were exhibited by group-I and IV CNGC genes against black shank, Cucumber mosaic virus, Potato virus Y, cold, drought, and cadmium stresses. Our analyses also suggested that NtabCNGCs can be regulated by phosphorylation and miRNAs, and multiple light, temperature, and pathogen-responsive cis-acting regulatory elements present in promotors. These results will be useful for elaborating the biological roles of NtabCNGCs in tobacco growth and development.  相似文献

We compared stomatal ammonia compensation point (χs) and its metabolic regulation in tobacco (Nicotiana tabacum) leaves of a quick-leaf-senescence phenotype ZY90 and a slow-leaf-senescence phenotype NC89. Compared with NC89, ZY90 had significantly higher χs values between 40 and 60 d after leaf sprouting in spite of its lower nitrogen content. During the same time, a steeper decline in glutamine synthetase activity was detected in ZY90 leaves, simultaneously with a steep increase in χs. These results suggested that the quick leaf senescence phenotype exhibited high NH3 emission potential due to efficient nitrogen recycling and remobilization, and glutamine synthetase played a key role in regulating χs in ZY90.  相似文献

Origin and rearrangement of ribosomal DNA repeats in natural allotetraploid Nicotiana tabacum are described. Comparative sequence analysis of the intergenic spacer (IGS) regions of Nicotiana tomentosiformis (the paternal diploid progenitor) and Nicotiana sylvestris (the maternal diploid progenitor) showed species-specific molecular features. These markers allowed us to trace the molecular evolution of parental rDNA in the allopolyploid genome of N. tabacum; at least the majority of tobacco rDNA repeats originated from N. tomentosiformis, which endured reconstruction of subrepeated regions in the IGS. We infer that after hybridization of the parental diploid species, rDNA with a longer IGS, donated by N. tomentosiformis, dominated over the rDNA with a shorter IGS from N. sylvestris; the latter was then eliminated from the allopolyploid genome. Thus, repeated sequences in allopolyploid genomes are targets for molecular rearrangement, demonstrating the dynamic nature of allopolyploid genomes.  相似文献

环丁烷嘧啶二聚体(CPD)和6-4光产物(6-4PP)是两种主要的UV-B诱导的DNA光损伤产物。利用单克隆抗体酶联免疫吸附分析法(ELISA),研究了温度对UV-B诱导的烟草叶圆片DNA损伤的影响。室温(24℃)条件下,UV-B处理引起了烟草叶圆片DNA中CPD和6-4PP的积累。0℃条件下,UV-B处理的烟草叶圆片DNA中CPD和6-4PP的积累比室温下分别降低了9.8%和12%。UV-B诱导的DNA损伤曾被认为是纯粹的光化学过程而与不受温度影响,而本实验结果表明,UV-B诱导的烟草叶圆片DNA形成CPD和6-4PP的过程具有温度依赖性。这一特性有利于植物对全球变化的适应,因而具有重要的生态学意义。  相似文献

The terminal and axillary buds of the day-neutral plant, Nicotiana tabacum cv. Wisconsin 38, become determined for floral development during the growth of the plant. This state of determination can be demonstrated with a simple experiment: buds determined for floral development produce the same number of nodes in situ and if rooted. After several months of growth and the production of many leaves, the terminal bud became determined for floral development within a period of about 2 days. After the terminal bud became florally determined, it produced four nodes and a terminal flower. The buds located in the axils of leaves borne just below the floral branches became florally determined 5 to 9 days after the terminal bud became florally determined. Since florally-determined axillary buds were not clonally derived from a florally-determined terminal meristem, axillary buds and the terminal bud acquired the state of floral determination independently. These data indicate that a pervasive signal induced a state of floral determination in competent terminal and axillary buds.  相似文献

We have examined the structure and chromatin organization of telomeres in Nicotiana tabacum. In tobacco the blocks of simple telomeric repeats (TTTAGGG)n are many times larger than in other plants, e.g., Arabidopsis thatiana or tomato. They are resolved as multiple fragments 60–160 kb in size (in most cases 90–130 kb) on pulsed-field gel electrophoresis (PFGE) of restriction endonuclease-digested DNA. The major subtelomeric repeat of the HRS60 family forms large homogeneous blocks of a basic 180 by motif having comparable lengths. Micrococcal nuclease (MNase) cleaves tobacco telomeric chromatin into subunits with a short repeat length of 157±5 bp; the subtelomeric heterochromatin characterized by tandemly repeated sequences of the HRS60 family is cut by MNase with a 180 by periodicity. The monomeric and dimeric particles of telomeric and subtelomeric chromatin differ in sensitivity to MNase treatment: the telomeric particles are readily digested, producing ladders with a periodicity of 7 bp, while the subtelomeric particles appear to be rather resistant to intranucleosomal cleavage. The results presented show apparent similarities in the organization of telomeric chromatin in higher plants and mammals.  相似文献
