Fusarium head blight (FHB) is a serious disease in wheat and barley affecting both yield and quality. To identify genes for resistance to infection, the RIL population derived from ‘Nanda2419’ × ‘Wangshuibai’ and the parents were evaluated for percentage of infected spikes (PIS) in four different environments. Using a 2,960 cM marker framework map constructed for this population, ten chromosome regions were detected for their association with type I resistance through interval mapping with Mapmaker/QTL, among which QTLs mapped in the intervals of Xwmc349~Xgwm149 on chromosome 4B, of Xwmc96~Xgwm304 on chromosome 5A and of Xgwm408~Xbarc140 on chromosome 5B were revealed in at least three environments and have Wangshuibai as the source of resistance alleles. Qfhi.nau-4B and Qfhi.nau-5A had larger effects and explained up to 17.5 and 27.0% of the phenotypic variance, respectively. To detect epistasis QTLs, two-locus interactions were examined by whole genome scan. Interactions of five locus pairs were found to have significant effects on type I resistance with the LOD score ranging 3.8–6.5 and four of them conferred resistance in parental phase. The one with the most significant effect was Xcfd42~Xgwm469 (6D)/Xwmc390-2~Xbd04 (2A) pair. No QTL × E interaction was detected for PIS. It was found that flowering time did not have significant effects on PIS in this population. Our studies indicated that Wangshuibai is useful for breeding for both type I and type II scab resistance and the markers associated with the QTLs could be used in marker-assisted selection and isolation of scab-resistance QTLs. Morinda royoc L. (Rubiaceae) root cultures were established for the production of anthraquinones (AQs). Three independent experiments were performed to evaluate the effects of different levels of indole-3-acetic acid (0–22.8 μM), culture duration (15–75 days) and subculture number (0–4). The following indicators were recorded: root fresh weight per Erlenmeyer and intracellular and extracellular AQ production. The experiments performed in this study allowed an increase of intracellular AQ content up to a maximum of 4.5 mg g−1 of fresh mass, after 30 days of culture in a medium 5.7 μM of IAA. In addition, isolation and identification of seven AQs from M. royoc L. roots is described, one of them being reported for the first time for this species. The structures of isolated compounds were determined from 1H-NMR data. To the best of our knowledge, this is the first report on AQ production from root culture of this plant.  相似文献

Poplar, a fast-growing species widely used outside its original area of distribution, was evaluated in the present study to verify its tolerance and hardening to waterlogging in anticipation of its implementation in rehabilitation practices for marginal lands subjected to flooding. Three water regimes were applied to the plants, grown in pots, with two clones (I-488 and D-64) with different sensitivities to flooding. These plants included a lot consisting of control plants (C), which were irrigated with good drainage, a non-preconditioned lot (NPr), and a third lot that was preconditioned to flooding (Pr). Furthermore, flooding was imposed on NPr and Pr plants by submerging pots up to 5 cm above the collar for 60 days followed by a 40-day recovery period. At the end of these two periods, shoot dry mass, foliar concentrations of certain ions, soluble sugars, starch and proline, and the relative electrolyte leakage were evaluated. The preconditioning treatment conferred different degrees of hardness on the treated plants compared with NPr plants. Our results revealed that high carbohydrate availability (soluble sugars and starch) is suggested to participate in sustaining membrane integrity which may affect the recuperative potential of Pr plants, notably those of clone I-488, from flooding damage.  相似文献

Randomly Amplified Polymorphic DNA markers (RAPD) were used to assess the hybrid identity of individuals sampled as Phlomis × termessi Davis. Out of 95 primers screened, 11 primers produced reproducible amplification patterns used for discrimination of P. × termessi and their parents. Eleven primers produced 81 bands. Forty two percent of the RAPD bands existed in parents. Of the 54 bands found in P. lycia, 19 were found only in this species and 7 of these were monomorphic. Similarly, of 57 RAPD bands observed in P. bourgaei, 18 were found only in P. bourgaei and 6 of these were monomorphic. Among hybrid individuals, 35 of the 73 markers were monomorphic. Fifteen of these existed in individual parents showing that parents were homozygous for these markers. Of the 35 monomorphic bands observed among hybrid individuals, 5 were present in the samples of one of the parents and completely absent from the samples of the other; therefore, additive inheritance is indicated. Of the 5 additive bands, 1 was inherited from P. bourgaei and 4 were inherited from P. lycia. Among 38 polymorhic markers observed in hybrid individuals, 9 were new and hybrid-specific. Pollen fertility was also investigated. Mean pollen fertility for P. lycia and P. bourgaei was 93% and 97% respectively. However, mean pollen fertility for hybrids was 65% (±10.5).  相似文献

Cultures of seven Daphne species: Daphne caucasica, D. cneorum, D. giraldii, D. retusa, D. jasminea, D. laureola and D. tangutica were established in vitro on MS/WPM based media. Five of the species responded best on MS-based media (D. tangutica, D. laureola, D. caucasica, D. retusa and D. giraldii), while the remaining two species performed best on WPM-based media (D. cneorum, and D. jasminea). Shoot proliferation was achieved from both apical and nodal explants. Shoots were sub-cultured from stock cultures, cut into nodal explants 3–5 cm long and place vertically on basal media supplemented with different concentrations and combinations of cytokinins and auxins. Individual species displayed different responses to the various cytokinins and auxins. Among species, D. jasminea produced the greatest proliferation rate with an average of 7.84 + 0.6 shoots per explant on WPM supplemented with 2.32 μM BA + 0.0045 μM TDZ + 0.054 μM NAA, while the best multiplication rate for the same species grown on the same media supplemented with a single cytokinin (BA) and no auxin was 2.60 + 1.3 shoots per explant. Following multiplication, new shoots transferred to the elongation trails and then 50–100 mm Shoots used for rooting experiments. Increased rooting efficiencies were observed on in vitro-generated shoots with the two-layer medium or dipping methods over when PGRs were uniformly incorporated into the medium. Maximum rooting frequencies (average) ranged from 59% in D. tangutica to 85% in D. jasminea. Following in vitro rooting, rooted shoots immersed in 0.01% solution of humates and planted into a standard horticultural substrate composed and watered weekly with a solution containing half-strength MS salts.  相似文献

In this work, embryos of selected Vicia species were examined to reveal the micromorphological characters that could have taxonomic significance. Experimental results show that morphological characters such as cotyledons shape and colour, radicle shape, colour and position, plumule colour and stipular parts attachments are variable and could be used for taxa identification in the genus Vicia. Numerical analysis based on 38 micromorphological characters was used to build a phenogram that indicated the relationships among the studied taxa. In addition, an identification key using embryo characters was prepared for Vicia species. The variability of embryo characters were discussed in the taxonomic context. Although the embryo characters have only limited phylogenetic signal for Vicia species, they might be used for their morphological delimitation.  相似文献

The taxonomic position of an unknown actinomycete isolated from a sand dune soil sample collected at Borg El-Arab in Egypt was established using a combination of genotypic and phenotypic data. Isolate S136(T) had chemotaxonomic and morphological properties consistent with its classification in the genus Nonomuraea and formed a distinct phyletic line in the Nonomuraea 16S rRNA gene tree. It was most closely related to the type strains of Nonomuraea helvata, Nonomuraea kuesteri and Nonomuraea turkmeniaca, sharing 16S rRNA gene similarities with these species of 97.1, 97.2 and 97.3%, respectively. The organism was distinguished from representatives of validly described Nonomuraea species using a range of phenotypic properties. It is apparent that the isolate belongs to a novel Nonomuraea species. The name proposed for this taxon is Nonomuraea aegyptia sp. nov., the type strain is S136(T) (=CGMCC 4.2054(T) = DSM 45082(T)).  相似文献

The physiological role of the Norway spruce [ Picea abies (L.) Karst.] spi 2 gene, encoding a defense-related cationic peroxidase was examined in transgenic tobacco (Nicotiana tabacum L.). Expression of spi 2, under control of the 35S promoter, in tobacco plants resulted in higher total peroxidase activities. The phenotype of the spi 2-transformed lines was normal. The spi 2-transformed lines displayed lignin levels similar to levels in the control line, but with some alteration in lignin histochemistry and structure. These changes were associated with reduced flexibility of the tobacco stems. The defense against pathogenic microorganisms was altered in the transgenic tobacco plants compared with control plants. High peroxidase activities increased the susceptibility to the pathogenic oomycete Phytophthora parasitica var. nicotianae, but increased the ability of the tobacco plants to suppress growth of the pathogenic bacterium Erwinia carotovora.  相似文献

To elucidate the role of amh and foxl2 in sex differentiation of the teleost fish Schizothorax kozlovi, the full-length cDNAs were cloned from the mature testis and ovary by rapid amplification of cDNA ends (RACE), and their relative mRNA expression levels were determined by quantitative real-time polymerase chain reaction among tissues and temperature groups. The complete amh and foxl2 cDNAs of S. kozlovi were 2060 bp and 1750 bp, which encoded 568 and 306 amino acids, respectively. The amh were expressed only in gonads, while foxl2 was expressed in the gills, brain and gonads, both exhibiting relatively high tissue specificity. The amh exhibited sex-specific expression pattern in the gonads. No sex differences in the foxl2 expression were observed in the brain and gonads, but significant sex differences were found in the gills. No significant differences were found in the foxl2 expression, from the larval to the juvenile stage, and also between different temperature groups. However, significant differences were found in the expression levels of amh from the larval (12–63 days posthatching (dph)) to the juvenile stage (190 dph), and also among the \(18{^{\circ }}\hbox {C}\) and \(10{^{\circ }}\hbox {C}\) groups at 31 dph. This result suggested that amh plays an important role in male sex differentiation of S. kozlovi during the early developmental stage, but no similar effect was observed in foxl2.  相似文献

The advanced backcross QTL (AB-QTL) strategy was utilised to locate quantitative trait loci (QTLs) for baking quality traits in two BC2F3 populations of winter wheat. The backcrosses are derived from two German winter wheat cultivars, Batis and Zentos, and two synthetic, hexaploid wheat accessions, Syn022 and Syn086. The synthetics originate from hybridisations of wild emmer (T. turgidum spp. dicoccoides) and T. tauschii, rather than from durum wheat and T. tauschii and thus allowed for the first time to test for exotic QTL effects on wheat genomes A and B in addition to genome D. The investigated quality traits comprised hectolitre weight, grain hardness, flour yield Type 550, falling number, grain protein content, sedimentation volume and baking volume. One hundred and forty-nine SSR markers were applied to genotype a total of 400 BC2F3 lines. For QTL detection, a mixed-model ANOVA was conducted, including the effects DNA marker, BC2F3 line, environment and marker × environment interaction. Overall 38 QTLs significant for a marker main effect were detected. The exotic allele improved trait performance at 14 QTLs (36.8%), while the elite genotype contributed the favourable effect at 24 QTLs (63.2%). The favourable exotic alleles were mainly associated with grain protein content, though the greatest improvement of trait performance due to the exotic alleles was achieved for the traits falling number and sedimentation volume. At the QTL on chromosome 4B the exotic allele increased the falling number by 19.6% and at the QTL on chromosome 6D the exotic allele led to an increase of the sedimentation volume by 21.7%. The results indicate that synthetic wheat derived from wild emmer × T. tauschii carries favourable QTL alleles for baking quality traits, which might be useful for breeding improved wheat varieties by marker-assisted selection.  相似文献

Cacao (Theobroma cacao L.) is one of the most important perennial crops in the world. In Brazil, the decrease in production in the last 25 years was caused by the entry and dissemination of witches’ broom (WB) (Moniliophthora perniciosa Aime & Phillips-Mora) in the cacao region of Bahia, the main producing region of the country. This disease increases costs and reduces crop yields. The main objective of this study was to evaluate interactions between scion and rootstocks for WB resistance through gas exchange and chlorophyll fluorescence measurements, determinations of the activity of enzymes involved in the antioxidant metabolism and of macro and micronutrient concentration at the leaf level. The experiment was conducted under greenhouse conditions using two cacao clones as rootstock, a tolerant (SCA-6) and a susceptible (SIC-876) to WB and CCN-51 as scion, a reference material due to tolerance to the disease, good productivity and high beans butter concentration. No grafted plants of SCA-6, SIC-876 and CCN-51 were used as controls. Were observed either in the grafted and non-grafted genotypes that M. perniciosa infection provoked alterations in the photochemical and biochemical phases of photosynthesis, in activity of enzymes involved in the antioxidant metabolism and in macro and micronutrients concentrations. It was concluded that interactions between scion and rootstock in T. cacao made the scion more tolerant to M. perniciosa infection. SIC-876 rootstock, considered susceptible to WB, had a positive effect on the performance of CCN-51 graft with respect to tolerance to the disease. The higher tolerance of CCN-51 graft to WB, provided by the SIC-876 rootstock, was helped by the increased activity of ascorbate and guaiacol peroxidases, enzymes involved in the antioxidant metabolism.  相似文献

Interspecific somatic hybrids between a diploid potato clone DG 81-68 susceptible to Phytophthora infestans (Mont.) de Bary and a resistant diploid tuber-bearing species Solanum × michoacanum were generated and analyzed. About 30 regenerants displaying an intermediate morphology were obtained as a result of three separate PEG-mediated fusion experiments. The RAPD analysis confirmed the hybridity of all the regenerants. About 50% of the hybrid plants exhibited vigorous growth and were stable in culture, while the rest of them rooted poorly and grew slowly in vitro. Most of the hybrid clones were at the tetraploid level (70%), while 30% of the clones examined were at the hexaploid level. The S. × michoacanum (+) DG 81-68 hybrids with growth anomalies were aneuploid. The variation in late blight resistance of the hybrid clones was found in detached leaflet tests, with enhanced resistance characteristic for three tetraploid hybrids.  相似文献

The ethanolic and aqueous extracts from in vitro shoots of Quillaja saponaria Mol. (Quillay) were studied for their antifungal activity against the phytopathogenic fungus Botrytis cinerea Pers. These extracts reduced conidial germination and mycelial growth of B. cinerea, ethanolic extracts being more active than aqueous extracts. In addition, the damage areas produced by this fungus on tomato leaves and strawberry fruits pre-treated with quillay extracts were diminished. The fungitoxic effect of in vitro-grown quillay extract was similar to those obtained with commercial fungicides of both natural (BC-1000) and synthetic (iprodione–dicarboximide) origin. On the other hand, the antifungal action of quillay extracts obtained from adult trees naturally grown was only slightly superior to the fungitoxic activity of the extract from in vitro plants. HPLC analysis of the extract showed that it contained saponins and some phenolic compounds such as chlorogenic, caffeic, vanillic, and salicylic acids, and scopoletin, which have been identified as antifungal agents on phytopathogenic fungi. The results obtained in this work, suggests that extracts of in vitro-grown quillay have an important protective effect against B. cinerea and support the use of an in vitro culture system as a biotechnological alternative to obtain environmental safe antifungal quillay extracts to control B. cinerea, contributing to the preservation of this indigenous Chilean species.  相似文献

While many reports on genetic analysis of Fusarium head blight (FHB) resistance in bread wheat have been published during the past decade, only limited information is available on FHB resistance derived from wheat relatives. In this contribution, we report on the genetic analysis of FHB resistance derived from Triticum macha (Georgian spelt wheat). As the origin of T. macha is in the Caucasian region, it is supposed that its FHB resistance differs from other well-investigated resistance sources. To introduce valuable alleles from the landrace T. macha into a modern genetic background, we adopted an advanced backcross QTL mapping scheme. A backcross-derived recombinant-inbred line population of 321 BC2F3 lines was developed from a cross of T. macha with the Austrian winter wheat cultivar Furore. The population was evaluated for Fusarium resistance in seven field experiments during four seasons using artificial inoculations. A total of 300 lines of the population were genetically fingerprinted using SSR and AFLP markers. The resulting linkage map covered 33 linkage groups with 560 markers. Five novel FHB-resistance QTL, all descending from T. macha, were found on four chromosomes (2A, 2B, 5A, 5B). Several QTL for morphological and developmental traits were mapped in the same population, which partly overlapped with FHB-resistance QTL. Only the 2BL FHB-resistance QTL co-located with a plant height QTL. The largest-effect FHB-resistance QTL in this population mapped at the spelt-type locus on chromosome 5A and was associated with the wild-type allele q, but it is unclear whether q has a pleiotropic effect on FHB resistance or is closely linked to a nearby resistance QTL.  相似文献

Theobroma cacao, is a tropical understorey tree that is a major economic resource to several tropical countries. However, the crop is under increased threat from several diseases that are responsible for 30% loss of harvest globally. Although QTL data related to the genetic determinism of disease resistance exist in cocoa, QTL mapping experiments are heterogeneous, thus making comparative QTL mapping essential for marker assisted selection (MAS). Sixteen QTL experiments were analysed, and the 76 QTLs detected were projected on a progressively established consensus map. Several hot spots, with QTLs related to different Phytophthora species and other diseases, were observed. The likely number of “real” QTLs was estimated by using a meta-analysis implemented in BioMercator software. There was a twofold reduction in average confidence interval observed when compared to the confidence interval of individual QTLs. This alternative approach confirms the existence of several sources of resistance to different diseases of cocoa which could be cumulated in new varieties to increase the sustainability of cocoa resistance using MAS strategies.  相似文献

The efficiency of two pollinators, Apis mellifera L. (Hymenoptera: Apidae) and the mason bee Osmia cornuta (Latreille) (Hymenoptera: Megachilidae), as carriers of biocontrol agents (BCA) from flower to flower (secondary colonisation) was investigated on apple cv ‘Golden Delicious’. The BCA tested was Bacillus subtilis, strain BD170 (Biopro®) developed for the control of the ‘fire blight’ caused by Erwinia amylovora (Burril) Winslow et al. The two insect species were studied as secondary BCA carriers on apple plants in pots under net screened tunnels. Their behaviour and capacity to deposit the BCA in the most receptive flower parts were compared both by washing, diluting and plating the flower organs on a recovery medium and by means of PCR analyses based on a molecular marker. O. cornuta showed better performances with respect to A. mellifera. For the field trials, pollinators were introduced in four apple orchards. During apple’s flowering, the BD170 (100 g hl?l) was sprayed once in two fields, and twice in the others. The pollinators’ efficacy in carrying the BCA from sprayed flowers to the stigmas of newly opened ones at different times after the spray treatment was evaluated. The detection of the BCA was performed by PCR analysis. The percentages of positive PCR flower samples were higher in the internal treated areas of the fields with respect to the external untreated ones, but the high colonisation level found in the latter and in the flowers opened in both areas several days after the treatment(s) demonstrated that pollinators can play an important role as secondary carriers.  相似文献

Two strains of Agrobacterium rhizogenes (15834, LBA 9402) and one Agrobacterium tumefaciens strain [GV 3101 (PMP90RK, p35SGUS-2)] and four culture media were tested and compared for their ability to induce hairy root formation on wounded Papaver somniferum L. hypocotyls. Five weeks after the infection with A. rhizogenes LBA 9402, hairy roots appeared on 80% of the hypocotyls maintained in the hormone-free liquid medium. Six hairy-root cultures were established. Transformation was confirmed by polymerase chain reaction analysis. One clone was analysed for its alkaloid production. The total alkaloid content was higher in the transformed roots (0.46±0.06% DW) than in the untransformed roots (0.32±0.05% DW). The transformed roots accumulated three times more codeine (0.18±0.02% DW) than intact roots (0.05±0% DW). A high-resolution physical map targeting a cluster of yield-related QTLs on the long arm of rice chromosome 9 has been constructed across a 37.4 kb region containing seven predicted genes. Using a series of BC3F4 nearly isogenic lines (NILs) derived from a cross between the Korean japonica cultivar Hwaseongbyeo and Oryza rufipogon (IRGC 105491), a total of seven QTLs for 1,000-grain weight, spikelets per panicle, grains per panicle, panicle length, spikelet density, heading date and plant height were identified in the cluster (P ≤ 0.0001). All seven QTLs were additive, and alleles from the low-yielding O. rufipogon parent were beneficial in the Hwaseongbyeo background. Yield trials with BC3F4 NILs showed that lines containing a homozygous O. rufipogon introgression in the target region out-yielded sibling NILs containing Hwaseongbyeo DNA by 14.2–17.7%, and out-yielded the Hwaseongbyeo parent by 16.2–23.7%. While higher yielding plants containing the O. rufipogon introgression were also taller and later than controls, the fact that all seven of the QTLs were co-localized in the same 37.4 kb interval suggests the possibility that a single, pleiotropic gene acting as a major regulator of plant development may control this suite of agronomically important plant phenotypes. This is the first report on the development of interspecific hybrids between T. alexandrinum and T. resupinatum, using embryo rescue. T. resupinatum is an important donor species for traits such as resistance to root and stem rot diseases, and tolerance to alkalinity for the potential improvement of T. alexandrinum. T. alexandrinum, as female parent, was crossed with T. resupinatum and embryos were excised 10–12 days after pollination and cultured in vitro on EC3 medium. Of the recovered embryos, 23.08% showed plumule emergence. For subculturing, shoot-induction medium LSP3 was used. Roots were induced on RL1 medium. Regenerants were hardened and inoculated with Rhizobium before transfer to the field. Morphological appearance of the hybrids was intermediate to both the parents. The hybrids were erect, leafy with short internodes and possessed distinct dwarf bushy compact appearance and were profusely branched. The plants were late flowering with pollen fertility more than 90%. Three out of five hybrid plants set seed following selfing. Hybridity of the plants was also confirmed through Est, PRX, SOD, GOT isozymes and protein banding patterns.  相似文献
