Sperm cells within pollen grains and pollen tubes of alfalfa (Medicago sativa L.) were observed at the ultrastructural level, and their plastid DNA was detected by DAPI (4,6-diamidino-2-phenylindole) staining. One sperm pair within the pollen grain and three sperm pairs within pollen tubes were reconstructed in three-dimensions from serial ultrathin sections. The two sperm cells are linked by cytoplasmic bridges in both pollen grains and tubes, and the vegetative nucleus is closely associated with the sperm cells within the pollen tube. The number of plastids and plastid nucleoids (DNA aggregates) in the sperm cell pair, collectively, is not significantly different from that in the generative cell; however, over 60% of the sperm cell plastids contain no DNA detectable with DAPI. The mean number of mitochondria in sperm cells is reduced from that in the generative cell (from 54 to 17), which suggests that paternal mitochondrial inheritance probably does not occur in the genotype investigated. Sperm cells of a pair may vary in their shape within the pollen grain and tube, but the number of plastids and mitochondria is not significantly different between the sperm cells. Therefore, heterospermy is not a factor determining cytoplasmic inheritance patterns in this species.  相似文献

Electron microscopic investigation has demonstrated that plastids and mitochondria are conserved in the generative cell, sperm cells and egg cell of Pelargonium hortorum Bailey. The plastids in the generative cell which contain starch for a short period, gradually changed to proplastids during the maturation of generative cell. The plastids in the sperm cells are large and numerous the characteristics of dense matrix and an abundant endomembrane systems. These plastids always appear ringlike in cross section. In the generative cell and sperms, the spherical or rod-shaped mitochondria are smaller than the.plastids and remain unchanged during the development process from generative cell to sperm cells. DNA filaments are visualized in the transparent central zone of the mitochondria. In the egg cell, plastids are more abundant than mitochondria. The structures of the plastids and mitochondria are obviously different from those in the sperm cell. Most of the plastids are irregularly rod-shaped and contain starch, the mitochondria are about 3 times larger than those in the sperm cells. Most of them are cup-shaped as proved by successive sections. DNA epifluorescence study demonstrated that DNA nucleoids are present in both plastids and mitochondria of the egg, generative cell and sperm cells. In the sperm cells, there is no ringlike DNA nucleoid as is existed in the egg cell. This study has defined the characteristics of the plastids and mitochondria in both male and female gemates of P. hortorurn. The results are essential contributions for further investigation of the biparental organelle transmission in the zygote and proembryo.  相似文献

The three-dimensional configuration of the nuclei of the trinucleate pollen grain of Spinacia oleracea L. has been examined by means of confocal laser scanning fluorescence microscopy (CLSM). It shows the presence of a male germ unit (MGU) in which all three nuclei are usually positioned in the periphery of the pollen grain. After freeze fixation and freeze substitution, the ultrastructure is better preserved than with standard chemical fixation. It shows the presence inside the pollen grain of mitochondria, dictyosomes, large starch-containing plastids, endoplasmic reticulum (ER), vacuoles and the MGU. In the sperm cells mitochondria, vesicles, dictyosomes and ER are also found. No microtubules were found in the grain and only very few inside the sperm cells. This is in contrast with earlier published results where fluorescent-labeled antibodies were used.  相似文献

It is known that extranuclear organelle DNA is inherited maternally in the majority of angiosperms. The mechanisms for maternal inheritance have been well studied in plastids but not in mitochondria. In the present study we examined the mitochondrial DNA in the male reproductive cells of Hordeum vulgare L. by immunoelectron microscopy. Our results show that the number of anti-DNA gold particles on sections of sperm cell mitochondria decreased by 97% during pollen development. The reduction occurred rapidly in the generative cells and subsequently in the sperm cells, concomitant with a remarkable reduction in mitochondrial volume. It seems that the copy numbers of mitochondrial DNA were reduced in the male reproductive cells, which may be a possible mechanism by which paternal transmission is inhibited. Unlike mitochondria, plastids are excluded from the generative cells during the first pollen mitosis. These data suggest a mechanism for maternal inheritance of mitochondria in angiosperms and for independent control of inheritance of mitochondria and plastids in H. vulgare.  相似文献

Sperm competition studies typically identify copulation duration as an important predictor of paternity as it may determine the quantity of sperm transferred and thus paternity success. This study explores the relationship between copulation duration, male body size, male age and sperm transfer in the golden orb‐weaving spider, Nephila edulis. Paternity in this species is strongly associated with the relative frequency and duration of copulation, which is also influenced by male size. We determined the number of sperm transferred during copulation, by performing sperm counts in both the male copulatory organs (palps) and female sperm receptacle (spermatheca) of recently mated pairs. The total number of sperm recorded (the sum in the male palps and female spermathecae) was greater for younger males than older males, but did not vary with male body size. In general, younger males transferred more sperm and a greater proportion of their sperm supplies than older males and, among these younger males, larger individuals transferred more sperm. However, there were no significant size effects for older males. More sperm was transferred with longer copulations, but in contrast with previous studies, we found that larger males copulated for longer. The rate of sperm transferred was negatively correlated with the duration of copulation, suggesting that the variation in copulation duration in N. edulis may represent strategic investment by males to alter patterns of paternity, in addition to transferring additional sperm.  相似文献

The mature pollen grains of Rhododendron mucronulatum Turcz. conform to the 2-celled type. Sperm cells differentiated within the pollen tube about 24 hours after germination in vitro and paired together, one of which being linked with the vegetative nucleus, forming a male germ unit (MGU). Abundance of plastids, mitochondria, microtubules and single-membrane-bounded vesicles could be visualized in each sperm cell, however, endoplasmic reticulum and Golgi apparatus were scarce. The electron-dense plastids with normal structure gave ring-like or dumbbell appearance in sections. Mitochondria were smaller and less electron-dense' in contrast to the plastids. DNA epifluorescence technique revealed that the generative and sperm cells contained numerous organelle nuclei (nucleoids). There was no difference in nucleoid number between the two sperm cells in a pollen tube. The results confirmed the possible existance of cytoplasmic inheritance potential of the male gametes of Rhododendron.  相似文献

Pollen grains of Plumbago zeylanica L. were serially sectioned and examined using transmission electron microscopy to determine the three-dimensional organization of sperm cells within the microgametophyte and the quantity of membrane-bound organelles occurring within each cell. Sperm cells occur in pairs within each pollen grain, but are dimorphic, differing in size, morphology and organelle content. The larger of the two sperm cells (Svn) is distinguished by the presence of a long (approx. 30 m) projection, which wraps around and lies within embayments of the vegetative nucleus. This cell contains numerous mitochondria, up to two plastids and, infrequently, microbodies. It is characterized by a larger volume and surface area and contains a larger nucleus than the other sperm cell. The second sperm cell (Sua) is linked by plasmodesmata with the Svn, but is unassociated with the vegetative nucleus. It is smaller and lacks a cellular projection. The Sua contains relatively few mitochondria, but numerous (up to 46) plastids and more microbodies than the other sperm. Summary The cytoplasmic content and the distribution of intramembrane particles (IMPs) of the plasma membrane of isolated sperm cells of perennial ryegrass (Lolium perenne L.) have been characterized using flow cytometry, transmission electron microscopy, confocal scanning laser microscopy and freeze-fracture studies. The isolated haploid sperm cells contain a variety of cell organelles with the exception of microtubules. Proplastids and plastids with starch were observed, although only rarely. Vacuoles containing remnants of organelles and stacked lamellae of endoplasmic reticulum with cytoplasmic inclusions were observed frequently, indicating that autophagy takes place. The number of mitochondria varies from 11 to 26 with an average of 17. Generally, the nucleus has a lobed shape and displays various interphasic stages of chromatin condensation. The analysis of the number of mitochondria and the nuclear state did not show evidence of sperm cell dimorphism. The cytological variability observed, could be explained by differences in developmental stages already present in vivo at the moment of isolation. No correlation between the number of mitochondria and the nuclear cross-sectioned area and/or the condensation state of the chromatin could be found. The density of intramembrane particles of the plasma membrane on the exoplasmic fracture face is more than twice that on the protoplasmic fracture face. That is the opposite of what was found for sporophytic cells of perennial ryegrass. These results are discussed in relation to the potential use of these cells for biotechnology and developmental studies.  相似文献

The subcellular compartmentation of β -glucosidase was studied in rye, maize and wheat seedlings by immunocytochemical methods. For detection, we used a 10 nm gold-labeled secondary antibody, and results were observed using transmission electron microscopy. In all three species, β -glucosidase was found in plastids, cytoplasm and cell walls. In rye, gold particles were seen on cell walls and cytoplasm in epidermal cells of the root tip and shoot, in bundle sheath cells of the shoot and in all cells, except the vascular bundle cells of the coleoptile. Gold labeling was also observed in plastids of the bundle sheath cells of rye shoot tips and in cortical cells of root tips. In wheat, gold labeling was observed on cell walls and cytoplasm of epidermal cells in the shoot base and coleoptile, and on cell walls and plastids in epidermal cells of the root tip. In maize, gold labeling was mainly found in plastids or proplastids in vascular bundle cells and bundle sheath cells of the shoot, in bundle sheath cells of the coleoptile and in epidermal cells of the root. Some gold particles were also found in cell walls and cytoplasm of stomatal guard cells of the shoot base and vascular bundle cells of the shoot tip and in the cell walls of bundle sheath cells of the shoot tip and root tip epidermal cells. Results are discussed in relation to the role of β -glucosidase in hydroxamic acid release and overall defense mechanism of monocotyledons.  相似文献

Sperm competition theory predicts that males should modulate sperm investment according to their social status. Sperm speed, one proxy of sperm quality, also influences the outcome of sperm competition because fast sperm cells may fertilize eggs before slow sperm cells. We evaluated whether the social status of males predicted their sperm speed in a wild population of dunnocks (Prunella modularis). In addition to the traditional analysis of the average speed of sperm cells per sample, we also analysed subsamples of the fastest sperm cells per sample. In other words, we systematically evaluated the effects of including different numbers of the fastest sperm in our analyses, ranging from the 5‐fastest sperm cells to the 100‐fastest sperm cells in a sample. We further evaluated whether fitness, defined here as the number of chicks sired per male per breeding season, relates to the sperm speed in the same population. We found that males in monogamous pairings (i.e. low levels of sperm competition), produced the slowest sperm cells, whereas subordinate males in polyandrous male–male coalitions (i.e. high levels of sperm competition) produced the fastest sperm cells. This result was consistent regardless of the number of fastest sperm included in our analyses, but statistical support was conditional on the number of sperm cells included in the analysis. Interestingly, we found no significant relationship between fitness and sperm speed, which suggests that it is possible that the differential mating opportunities across social status levelled out any possible difference. Our study also suggests that it is important to identify biologically meaningful subsets of fastest sperm and cut‐offs for inclusions for assessing sperm competition via sperm speed.  相似文献

 Mature Jasminum officinale and J. nudiflorum pollen grains were stained with 4′,6-diamidino-2-phenylindole (DAPI) and examined by epifluorescence microscopy. The pollen grains were found to be trinucleate, and the sperm cells in both species contained a large number of epifluorescent spots that corresponded to cytoplasmic DNA aggregates (nucleoids). The nucleoids of J. nudiflorum were observed to be dimorphic under the epifluorescence microscope, indicating that the sperm cells might contain both plastid and mitochondrial DNA. The nucleoids of J. officinale presented a similar appearance when stained with DAPI, but electron microscopic examination of the sperm cells revealed that they contained both plastids and mitochondria. When analyzed by DNA immunogold electron microscopy, gold particles were detected on both plastids and mitochondria. These findings demonstrated the preservation of plastid and mitochondrial DNA in mature sperm cells and thus the potential for biparental cytoplasmic inheritance in J. officinale and J. nudiflorum. There are 2 models of male adjustment of sperm investment inthe ejaculate in relation to sperm competition. The "risk model"predicts that as "risk" of sperm competition increases, sperminvestment also increases. This prediction has been supportedin many species, including mammals. The "intensity model" involvesthe number of competing males copulating with the same femaleand predicts that males will allocate the highest sperm investmentat low sperm competition intensity (SCI) and then decreasingsperm investments as SCI increases. Two alternative outcomesare that sperm investment is unaffected by SCI and that sperminvestment increases as SCI increases. There are studies supportingall 3 possible outcomes in relation to SCI but no data on mammals.The present paper presents the first study of SCI in a mammalspecies, the meadow vole, Microtus pennsylvanicus. We used odorsof conspecific males to simulate low and high intensities ofsperm competition. We found that males allocate the highestsperm investment at low SCI and decrease significantly theirsperm investment at high SCI. We also found that males allocatethe lowest sperm investment at low sperm competition risk (SCR)and the highest sperm investment at high SCR. Lepidopteran males produce two sperm types: nucleated eupyrene sperm and non‐nucleated apyrene sperm. Although apyrene sperm are infertile, both sperm types migrate from the spermatophore to the spermathecal after copulation. As a dominant adaptive explanation for migration of apyrene sperm in polyandrous species, the cheap filler hypothesis suggests that the presence of a large number of motile apyrene sperm in the spermatheca reduces female receptivity to re‐mating. However, apyrene sperm are also produced in males of the monandrous swallowtail butterfly Byasa alcinous Klug. To identify the role of apyrene sperm in these males, the present study examines the number of spermatozoa produced and transferred and the dynamics and motility of spermatozoa in the spermatheca for each type of sperm. Apyrene sperm represents approximatey 89% of the sperm produced and transferred, which is comparable to polyandrous species. Two‐day‐old males transfer approximately 17 000 eupyrene and 230 000 apyrene spermatozoa to a spermatophore; approximately 5000 eupyrene and 47 000 apyrene spermatozoa arrive at the spermatheca. Eight days after copulation, most eupyrene spermatozoa remain in the spermatheca and a quarter of them are still active. However, the number of apyrene spermatozoa decreases and those remaining lose their motility after the arriving at the spermatheca. Consequently, 8 days after copulation, no motile apyrene sperm are found. The high proportion of apyrene sperm in the spermatophore, as well as in sperm migration, suggests that the production and migration of apyrene sperm is not simply an evolutionary vestigial trait. The possible functions of apyrene sperm in monandrous species are discussed.  相似文献

迎红杜鹃 ( Rhododendron mucronulatum Turcz.)的成熟花粉为二细胞型 ,精细胞在花粉管中形成。花粉管中的两个精细胞及与营养核之间互相联结 ,形成雄性生殖单位。两个精细胞的细胞质中均含有丰富的细胞器 ,包括质体、线粒体、小泡及微管 ,内质网和高尔基体稀少。具正常结构的精细胞质体在切面上多呈环形或哑铃形 ,内膜不发达 ,基质电子密度高。线粒体为球形或棒状 ,基质电子密度较低。 DNA特异性荧光染色显示 ,生殖细胞及精细胞中均含有大量类核 ( nucleoid) ,两个精细胞中的类核数量无明显差异。结果证明了杜鹃精细胞中存在大量具 DNA的可遗传细胞器 ,为杜鹃属植物的双亲细胞质遗传方式提供了细胞学证据。  相似文献

Summary For a given cell type and genotype a close positive correlation exists between the number of plastids in a cell and the amount of DNA in the nucleus. Comprehensive evidence is presented. The duplication of the DNA amount entails an increase of the plastid number in differentiating cells by about 70%. Exceptions reported in the literature are critically examined. The odds are in favour of the assumption that exceptions to the rule which are not due to special circumstances do not exist. In meristematic cells even a duplication of the plastid number will occur, for cells without plastids are not to be found. The plastids are always ready to divide, the interpretation goes, but the size of their populations is limited by the amount of nuclear DNA. Thus meristematic cells manage to control their plastid populations by releasing once in a cell cycle the brakes imposed upon plastid division, whereupon the plastids make use of their newly won freedom, dividing until the old ratio between plastid number and nuclear DNA amount is established again. As a shorter time is needed for plastid division than for mitosis, there is no danger of cells arising without plastids; no distributing mechanism is required if at least three to four plastids are present in a cell. The findings are consistent with and would appear to be best explained by the theory of the symbiotic origin of the plastids.  相似文献

The structure of rabbit, fowl, and Xenopus laevis sperm chromatin was explored by study of the reaction of their decondensed nuclei with DNase 1 and micrococcal nuclease. Those of rabbit and fowl were readily digested by DNase 1, and the polyacrylamide gel electrophoresis profiles of DNAs extracted from the digests were similar, each being polydisperse with a single discrete band of DNA smaller than 72 base pairs. There were differences, however, between the sperm chromatins in the course of their digestion by micrococcal nuclease. A limit digest at about 45% acid solubility was obtained with Xenopus sperm chromatin, while 90% of fowl sperm DNA was rendered acidsoluble by the enzyme. The gel profiles of the limit digests were polydisperse, but only those of rabbit and fowl sperm chromatins possessed a discrete band of DNA smaller than 72 base pairs. Bleomycin did not react with DNA of rabbit, fowl, or Xenopus spermatozoa. Since bleomycin reacts with somatic cell chromatin, and the course of DNase 1 or micrococcal nuclease digestion of sperm chromatin was different from that found for somatic cell chromatin, it would appear that sperm chromatin does not have the repeating nucleosometype structure of somatic cell chromatin. The nuclease digestion studies further suggest that the organization of rabbit and fowl sperm chromatins is similar, and is different from that of Xenopus sperm chromatin. The dependence of the structure of sperm chromatin on the composition of its basic proteins, and a possible structure for a protamine-type sperm chromatin, are discussed.  相似文献

Recent attention has focused on the role that sperm competitionmay play in the evolution of sperm morphology. Theoretical analysespredict increased sperm size, decreased sperm size, and no changein sperm size in response to sperm competition, depending onthe assumptions made concerning the life history and functionof sperm. However, although there is good evidence that spermmorphology varies widely within and between species, the adaptivesignificance of this variation has not been examined. Here wedocument significant intraspecific variation in sperm lengthin the field cricket, Teleogryllus oceanicus. Sperm length didnot influence the rate of migration of sperm from the spermatophoreto the female's spermatheca. We performed sperm competitiontrials in which we varied the numbers of sperm transferred byeach of two males that differed in the length of sperm theyproduced. Neither sperm length nor the number of sperm transferredinfluenced paternity. The same results were obtained using twodifferent methods for assigning paternity. The distributionof paternity across a female's mates was highly variable, withfrequently one, or more in the case of females mated to fourmales, principal sire. There were no mating order effects onpaternity. These data show that sperm do not mix randomly inthe female's spermatheca. We discuss several alternative explanationsfor the patterns of paternity observed.  相似文献
