An endogenous inhibitor of γ-aminobutyric acid (GABA) receptors was partially purified from bovine brain striatum. It was obtained as a low molecular weight fraction by gel filtration on Biogel P-2 and was adsorbed to Dowex AG 50W-X8, but not to Dowex AG 1-X8. It was ninhydrin-negative, basic, heat-stable substance. It caused dose-dependent inhibition of Na+-independent [3H]GABA bindings. Scatchard plot analysis of the [3H]GABA binding to GABA “B” receptor recognition site showed this inhibitor increased the Kd value (24.1 nM to 3.6 nM) without changing the Bmax. On the other hand, Scatchard plot analysis of the [3H]GABA binding to GABA “A” receptor recognition site showed that the inhibitor decreased number of binding sites (706 fmol/mg protein to 494 fmol/mg protein) without affecting the Kd value. These results suggest that the endogenous inhibitor functions as a modulator for GABAB and GABAA receptors.  相似文献

The neurotransmitter gamma-aminobutyric acid (GABA) mediates inhibitory signaling in the brain via stimulation of both GABA(A) receptors (GABA(A)R), which are chloride-permeant ion channels, and GABA(B) receptors (GABA(B)R), which signal through coupling to G proteins. Here we report physical interactions between these two different classes of GABA receptor. Association of the GABA(B) receptor 1 (GABA(B)R1) with the GABA(A) receptor gamma2S subunit robustly promotes cell surface expression of GABA(B)R1 in the absence of GABA(B)R2, a closely related GABA(B) receptor that is usually required for efficient trafficking of GABA(B)R1 to the cell surface. The GABA(B)R1/gamma2S complex is not detectably functional when expressed alone, as assessed in both ERK activation assays and physiological analyses in oocytes. However, the gamma2S subunit associates not only with GABA(B)R1 alone but also with the functional GABA(B)R1/GABA(B)R2 heterodimer to markedly enhance GABA(B) receptor internalization in response to agonist stimulation. These findings reveal that the GABA(B)R1/gamma2S interaction results in the regulation of multiple aspects of GABA(B) receptor trafficking, allowing for cross-talk between these two distinct classes of GABA receptor.  相似文献

Interactions between GABAA and GABAB receptors were studied using muscimol-stimulated uptake of 36Cl- by membrane vesicles from mouse cerebellum. Baclofen inhibited muscimol-stimulated 36Cl- uptake and this action was more pronounced with longer flux times (30 vs. 3 s) and after predesensitization of GABAA receptors. Baclofen also inhibited 36Cl- flux by cortical membranes but was more effective with cerebellar preparations. The action of baclofen was stereoselective, calcium-dependent, and blocked by the GABAB receptor antagonist 2-OH-saclofen. It was mimicked by GTP-gamma-S but not by GDP-beta-S, which suggests that baclofen may be acting via a G protein. The action of baclofen was inhibited by U73122, an inhibitor of phospholipase C. However, the potassium channel blockers tetraethylammonium or Ba2+ did not affect the action of baclofen. The results show that activation of GABAB receptors can inhibit the function of GABAA receptors and suggest that this action involves either a nondesensitizing subtype of GABAA receptor or the rate or recycling of desensitized to nondesensitized receptors. We speculate that this action of baclofen results from activation of phospholipase C and phosphorylation of a subtype of GABAA receptor by protein kinase C.  相似文献

Evoked focal potentials which were induced in vitro in a slice of olfactory tract by stimulation of the lateral olfactory tract (LOT) have been studied. The potential consisted of an initial biphasic wave, the compound action potential of LOT, population synaptic responses, and population spike. Investigation into the influence of motor training on the functional activity of the rat sensorimotor cortex in ontogenesis has shown that three to four-month training, starting at the age of four weeks, leads to a statistically significant enhancement of sensorimotor cortex activity both by latencies and recovery cycles durations. A similar six to seven-month locomotor training produces the same statistically significant results. The differences in the shifts of functional activity after motor training observed between two age groups are not statistically significant. The probability of changes in the average definitive electrophysiological parameters of functional activity after motor training observed between two age groups are not statistically significant. The probability of changes in the average definitive electrophysiological parameters of functional activity of the sensorimotor cortex is suggested in rats aged more than a month, as a result of individual experience.  相似文献

Evoked potentials (EP) of the cerebellar cortex in response to stimulation of peripheral nerves are characterized by a two-phase positive-negative oscillation of the potential having a latent period of 10–25 msec. The electropositive phase can contain up to three components. The latent period of component I comprises 3–9 msec. The latent period and amplitude of this component are distinguished by considerable stability, which indicates the predominant significance of presynaptic processes in its formation. The sign of component II changes at a depth of 500 µ (and more), which corresponds to the position of the granular cell layer. At this level there arises in the neurons a response with a latent period of 4–10 msec in the form of a group (3–10) of impulses with a frequency of up to 200 per sec. It is concluded that the granular cells participate in the formation of component II and partially participate in the formation of components I and III of the EP. Responses to stimulation of the nerves appear synchronously with the EP in 24% of responding Purkinje cells; they fall on the maximum electropositive deviation or component III of the EP. Microinjections of 1% strychnine into the cerebellar cortex cause an increase of EP amplitude; impulse activity of the neurons is intensified. This indicates participation of postsynaptic processes in the formation of EP. No shifts in the EP of the cerebellar cortex were observed after intracortical injection of 0.1% atropine.N. I. In acute experiments on cats we investigated evoked potentials from the cingulate gyrus developed in response to stimulation of somatic and visceral nerves; also potentials from various parts of the hypothalamus, and midbrain reticular formation. We showed that the nonspecific afferent system influences electrical activity in the limbic cortex through hypothalamic pathways. We consider the limbic cortex to be part of the association area of the neocortex and that the associative responses of the cortex are more complex in nature than is usually thought to be the case, and that they are formed under the influence of impulses arriving at the cortex along many specific and nonspecific pathways. The hypothalamo—cingulate system is one of the main systems of cortico—subcortical integration. It plays an important part in regulation of autonomic, somatic, and emotional responses.L. A. We investigated evoked responses of the cerebellar cortex of rabbits under Nembutal or chloralose anesthesia to stimulation of the sciatic, brachial, and vagus nerves. The parameters of evoked potentials (E Ps), together with features of their distribution throughout the cerebellar cortex, enabled us to divide them provisionally into three types. Evoked potentials of the first type have a latent period of 5–10 msec and a two-phase or more complex shape. Evoked potentials of the second type have a latent period of 10–23 msec and include from one to four components. Evoked potentials of the third type are discharges with long latent periods (20–50 msec) and consist of a series of slow sinusoidal oscillations. Appearance of an initial electronegative component is characteristic of EPs of the cerebellar cortex of rabbits, especially those of the second and third types. Presynaptic GABA(B) receptors (GABA(B)R) control glutamate and GABA release at many synapses in the nervous system. In the present study we used whole-cell patch-clamp recordings of spontaneous excitatory and inhibitory synaptic currents in the presence of TTX to monitor glutamate and GABA release from synapses in layer II and V of the rat entorhinal cortex (EC)in vitro. In both layers the release of both transmitters was reduced by application of GABA(B)R agonists. Quantitatively, the depression of GABA release in layer II and layer V, and of glutamate release in layer V was similar, but glutamate release in layer II was depressed to a greater extent. The data suggest that the same GABA(B)R may be present on both GABA and glutamate terminals in the EC, but that the heteroreceptor may show a greater level of expression in layer II. Studies with GABA(B)R antagonists suggested that neither the auto- nor the heteroreceptor was consistently tonically activated by ambient GABA in the presence of TTX. Studies in EC slices from rats made chronically epileptic using a pilocarpine model of temporal lobe epilepsy revealed a reduced effectiveness of both auto- and heteroreceptor function in both layers. This could suggest that enhanced glutamate and GABA release in the EC may be associated with the development of the epileptic condition.  相似文献

Besides the intensity and frequency of an auditory stimulus, the length of time that precedes the stimulation is an important factor that determines the magnitude of early evoked neural responses in the auditory cortex. Here we used chinchillas to demonstrate that the length of the silent period before the presentation of an auditory stimulus is a critical factor that modifies late oscillatory responses in the auditory cortex. We used tetrodes to record local-field potential (LFP) signals from the left auditory cortex of ten animals while they were stimulated with clicks, tones or noise bursts delivered at different rates and intensity levels. We found that the incidence of oscillatory activity in the auditory cortex of anesthetized chinchillas is dependent on the period of silence before stimulation and on the intensity of the auditory stimulus. In 62.5% of the recordings sites we found stimulus-related oscillations at around 8-20 Hz. Stimulus-induced oscillations were largest and consistent when stimuli were preceded by 5 s of silence and they were absent when preceded by less than 500 ms of silence. These results demonstrate that the period of silence preceding the stimulus presentation and the stimulus intensity are critical factors for the presence of these oscillations.  相似文献

Potentials evoked in nuclei of the reticular formation by electrodermal stimulation of the limbs were investigated in acute experiments on unanesthetized, immobilized rats during cooling of the somatosensory cortex in the area of representation of one forelimb. Evoked potentials in the reticular formation were found to depend on the degree of cold inhibition of the cortical primary response to the same stimulation. The peak time of the main negative wave increased from 40–50 to 60–80 msec with a simultaneous decrease in its amplitude or its total disappearance in the case of deep cooling of the cortex. Cooling of the cortex had a similar although weaker effect on the earlier wave of the evoked potential with a peak time of 14 msec, recorded in the ventral reticular nucleus. In parallel recordings of potentials evoked by stimulation of other limbs they remained unchanged at these same points of the reticular formation or were reduced in amplitude while preserving the same temporal parameters. Cooling of the cortex thus selectively delays the development and reduces the amplitude of the response to stimulation of the limb in whose area of representation transformation of the afferent signal into a corticofugal volley is blocked. Consequently the normal development of both late and early components of the potential evoked in the reticular formation by somatic stimulation requires an additional volley, descending from the cortex, and formed as a result of transformation of the same afferent signal in the corresponding point of the somatosensory cortex.I. The tactile somatosensory pathway from whisker to cortex in rodents provides a well-defined system for exploring the link between molecular mechanisms, synaptic circuits, and behavior. The primary somatosensory cortex has an exquisite somatotopic map where each individual whisker is represented in a discrete anatomical unit, the "barrel," allowing precise delineation of functional organization, development, and plasticity. Sensory information is actively acquired in awake behaving rodents and processed differently within the barrel map depending upon whisker-related behavior. The prominence of state-dependent cortical sensory processing is likely to be crucial in our understanding of active sensory perception, experience-dependent plasticity and learning.  相似文献

Intracellular records were made from neurones of the somatosensory cortex of cats, during stimulation of the ventrobasal complex of the thalamus. The aim of the experiments was to detect correlations between frequency dependence of surface evoked potentials and that of unit discharges. The amplitude of the surface evoked potential showed a strong diminution when the frequency of the thalamic stimulation was raised from 1 cps to 15 cps. In spite of this, frequency dependence in amplitude of unit discharges was never seen. As regards their frequency of occurrence the unit responses (full spikes, dendritic, postsynaptic potentials) behaved differently: a part of them showed increasing, another part gave decreasing occurrence, and the remaining portion did not change it. The authors conclude that temporal dispersion fails to give account for the frequency dependence, therefore further possibilities have to be examined.  相似文献
