目的:探讨细胞凋亡在星形细胞瘤中的作用及其与p53、Fas和Fas配体(Fas ligand,FasL)的关系。方法:对43例星形细胞瘤的标本分别进行HE染色,TUNEL及免疫组化分别标记p53,Fas和FasL。结果:高级别肿瘤和低级别肿瘤间的凋亡无显著差异(P〉0.05)。高级别星形细胞瘤的p53,Fas和FasL的表达均显著高于低级别肿瘤(P均〈0.05)。结论:突变型p53可作为评价星形细胞瘤生物学行为的参考指标。与低级别星形细胞瘤相比,高级别肿瘤中的细胞凋亡受到了抑制,且Fas与FasL的过表达对细胞凋亡可产生明显影响。  相似文献

目的:研究p27kip1蛋白和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)在星形细胞瘤中的表达与肿瘤病理分级的关系,探讨p27kip1蛋白在星形细胞瘤演变过程中的意义。方法:SP免疫组化法对64例星形细胞瘤的p27kip1蛋白和PCNA表达进行观察。结果:随着病理级别的升高,p27kip1阳性细胞百分率降低,而PCNA则相反,两者的表达成显著负相关。结论:p27kip1表达的缺失可能与星形细胞瘤的发生发展密切相关,PCNA能较客观地反映肿瘤的恶性程度。  相似文献

Fas,FasL均是细胞表面受体。正常情况下,Fas分子主要分布于人体各个组织器官及病变细胞表面,FasL广泛分布于活化的T细胞等免疫细胞表面。Fas与FasL的结合可导致表达Fas的细胞凋亡,平衡机体免疫反应。当机体发生肿瘤时,肿瘤细胞表面Fas表达下调,有时甚至出现FasL的表达,不但削弱了Fas阳性淋巴细胞的攻击能力,而且赋予了肿瘤细胞杀伤免疫细胞的能力,即肿瘤免疫逃逸的新机制—Fas反击。就这一新机制与消化道肿瘤尤其胃部肿瘤的相关性做一综述。  相似文献

目的:研究凋亡相关基因Bcl-2、Fas和FasL在前列腺增生组织中表达,并探讨其与前列腺增生症的关系.方法:收集哈尔滨医科大学附属第二医院2004年5月--2005年8月前列腺增生症(BPH)组织标本36例,前列腺癌(PCa)组织标本12例和正常前列腺(NP)组织标本7例.应用免疫组织化学SABC方法,检测BPH、PCa和NP组织标本中凋亡相关基因Bcl-2、Fas和FasL的表达,并探讨Bcl-2、Fas和FasL的表达与前列腺增生症的关系.结果:①BPH与PCa组Bcl-2蛋白阳性表达明显高于NP组(P<0.05),而BPH组与PCa组阳性表达无显著差异;PCa组Fas表达显著低于BPH和NP(P<0.05、0.01),而BPH和NP组之间差异也有显著性(P<0.05);FasL在三组之间表达差异均无显著性(P>0.05).②在BPH组织中Bcl-2的表达与Fas表达呈负相关(P(0.05),与FasL表达呈正相关(P<0.05),余均未见相关性.结论:Bcl-2、Fas、FasL均参与了BPH的发生发展过程,Bcl-2、Fas、FasL表达的综合结果可能是引起BPH的原因之一.  相似文献

目的通过研究辛伐他汀对动脉粥样硬化大鼠血管壁中细胞凋亡相关基因Fas及FasL蛋白表达产物的影响,探讨其在预防动脉粥样硬化发生中的可能机制。方法复制动脉粥样硬化大鼠模型,以辛伐他汀干预,取胸主动脉,观察其斑块变化,采用免疫组化Elivision法测定动脉粥样硬化血管壁中Fas、FasL蛋白表达。结果Fas蛋白表达在实验组明显高于对照组及干预组(P<0.01,P<0.05),实验组FasL蛋白表达也明显高于对照组及干预组(P<0.05)。结论Fas及FasL基因通过促进细胞凋亡作用而诱发动脉粥样硬化过程,辛伐他汀可通过调节细胞凋亡过程发挥抗动脉粥样硬化作用。  相似文献

[目的]探讨嗜铬细胞瘤抵抗细胞凋亡的潜在机制。[方法]顺铂处理或不处理嗜铬细胞瘤细胞PC-12后,通过蛋白质组学检测两者蛋白表达差异。通过siRNA敲低上述差异基因,检测嗜铬细胞瘤细胞PC-12的凋亡水平。[结果]顺铂处理后,BIM和Egln3蛋白表达量增高,Egln3的mRNA水平显著上升(P<0.05)。过表达Egln3后,BIM的表达显著上升。敲低Egln3后,BIM的表达显著下降。Egln3能够降低BIM的泛素化水平。敲低BIM和Egln3后嗜铬细胞瘤的凋亡水平显著上升(P<0.05)。过表达BIM和Egln3后,嗜铬细胞瘤的凋亡水平显著下降(P<0.05)。过表达rno-miR-9a-5p后,Egln3的表达水平下降。敲低rno-miR-9a-5p后,Egln3的表达水平上升。过表达rno-miR-9a-5p后,嗜铬细胞瘤凋亡水平上升(P<0.05);敲低rno-miR-9a-5p后,嗜铬细胞瘤凋亡水平下降(P<0.05)。[结论]rno-miR-9a-5p通过靶向Egln3及Egln3的mRNA,降低了Egln3的蛋白表达,抑制了蛋白酶体途径对...  相似文献

去铁酮可诱导p53的表达和引发肿瘤细胞凋亡。RRM2B基因在铁螯合剂抑制肿瘤细胞生长中同样起重要作用,且其表达受p53调节。为了探讨去铁酮对宫颈癌细胞的影响,以及p53和RRM2B基因二者的关系,通过MTT法检测不同浓度及不同时间去铁酮处理对HeLa细胞活性的影响,采用流式细胞仪检测细胞凋亡水平。同时,利用Western-blot和荧光定量PCR技术分别在蛋白和mRNA水平检测p53和RRM2B表达变化,并进一步利用p53抑制剂检测p53对RRM2B表达的影响。结果表明,HeLa细胞活性随着去铁酮作用浓度及作用时间的增加而下降,在500μmol/L去铁酮处理48h后出现明显凋亡。进一步研究发现,去铁酮可显著上调p53表达水平(P0.05),而p53可显著下调RRM2B表达水平(P0.05)。综上所述,去铁酮可通过p53下调RRM2B表达水平而参与抑制宫颈癌细胞增殖。  相似文献

目的:研究雷公藤甲素对柯萨奇病毒B3病毒(CVB3)感染的病毒性心肌炎小鼠心肌细胞凋亡和Fas/FasL蛋白表达的抑制作用,探讨TP治疗病毒性心肌炎的作用机制。方法:将Balb/c小鼠随机分成4组作为动物模型,分别为对照组、模型组、利巴韦林组和TP组。对照组腹腔注射生理盐水,其余三组腹腔注射CVB3,利巴韦林组和TP组小鼠分别予以相应的药物治疗后,测定各组小鼠存活率及心肌病变积分,采用末端转移酶标记技术(TUNEL法)检测小鼠心肌细胞凋亡,免疫组化法检测Fas/FasL蛋白阳性表达。结果:空白对照组心肌无病变,利巴韦林组、TP组与模型组相比有显著性差异(P<0.01)。正常组鲜见心肌细胞凋亡,模型组细胞凋亡率较正常组显著增加(P<0.01),治疗组利巴韦林组和TP组凋亡率比模型组明显降低(P<0.05,P<0.01)。模型组Fas/FasL表达比正常组显著增多(P<0.01),治疗组利巴韦林组和TP组较模型组显著降低(P<0.01)。结论:雷公藤甲素具有通过抑制Fas/FasL蛋白的表达,减缓心肌细胞凋亡,达到抑制病毒性心肌炎从而保护心肌细胞的作用。  相似文献

凋亡相关因子(Fas)、凋亡相关因子配体(FasL)和磷蛋白53(P53)是细胞发生凋亡的重要调控因素。本文研究了天府肉鸭(Anas platyrhynchos domestica)胸腺发育过程中Fas、FasL和P53蛋白的动态表达,旨在更深入地认识胸腺细胞自然凋亡的调控机制。将65羽天府肉鸭分为13组,即22、24、26天胚龄,胚后0(新生雏)、3、5、8、14、17、20、26、29、32周龄。分别采取各组鸭的胸腺组织,4%多聚甲醛固定,常规石蜡切片,Fas、Fas L及P53蛋白免疫组织化学染色,镜检并进行阳性细胞计量分析。结果显示,Fas阳性反应见于胸腺淋巴细胞,其阳性率在22~26天胚龄无明显变化;新生雏显著升高,随后趋于恒定,直到17周龄,20周龄以后呈上升趋势。Fas L阳性反应见于胸腺淋巴细胞及上皮细胞,在胚胎及胚后发育过程中,其淋巴细胞阳性率无明显变化。P53阳性反应见于胸腺皮质淋巴细胞和胸腺上皮细胞,其淋巴细胞阳性率在各组中无显著性差异。结果说明Fas,Fas L和P53蛋白在天府肉鸭胸腺发育中呈现出不同表达规律。  相似文献

Apoptosis induced in male germ cells following ionizing radiation is dependent on functional p53 (Trp53) being present. We sought to determine whether Fas (Tnfrsf6/CD95/APO-1), an apoptotic factor, is involved in this p53-dependent germ cell death. In p53 knock-out mice exposed to 5 Gy of x-radiation, germ cells were protected from cell death, as assessed by counting apoptotic seminiferous tubules 12 h following radiation. Similarly, spermatid head counts in p53 knock-out mice remained near normal 29 days after exposure to 0.5 Gy of radiation, whereas wild-type animals had a more than twofold reduction in spermatid head counts. Fas mRNA expression remained at pretreatment levels in p53 knock-out mice; however, Fas increased in a time-dependent manner in wild-type mice following exposure to 5 Gy of radiation, indicating that radiation-induced Fas expression is p53-dependent. The functional significance of Fas involvement was demonstrated when lpr(cg) mice, having a nonfunctional Fas receptor, were exposed to 5 Gy of radiation; the number of apoptotic seminiferous tubules 12 h following radiation was significantly reduced compared to that of wild-type mice. Additionally, lpr(cg) mice exposed to 0.5 Gy of radiation had increased spermatid head counts 29 days following radiation compared to wild-type mice. Interestingly, gld mice with a non-functional Fas ligand (Tnfsf6/FasL/CD95L) were as sensitive to radiation as wild-type animals, and levels of FasL mRNA were not affected by radiation treatment. These results indicate that apoptosis and up-regulation of Fas following radiation are both p53-dependent events. Although Fas is necessary, in part, for radiation-induced p53-dependent apoptosis, FasL is not.  相似文献

The effects of tetrandrine in the human hepatoblastoma G2 (Hep G2) cell line were investigated in this study. The results showed that tetrandrine not only inhibited Hep G2 growth but also induced apoptosis and blocked cell cycle progression in the G1 phase. ELISA assay demonstrated that tetrandrine significantly increased the expression of p53 and p21/WAF1 protein, which caused cell cycle arrest. An enhancement in Fas/APO-1 and its two form ligands, membrane-bound Fas ligand (mFasL) and soluble Fas ligand (sFasL), might be responsible for the apoptotic effect induced by tetrandrine. Taken together, p53 and Fas/FasL apoptotic system possibly participated in the antiproliferative activity of tetrandrine in Hep G2 cells.  相似文献

Engagement of the cell death surface receptor Fas by Fas ligand (FasL) results in apoptotic cell death, mediated by caspase activation. Cell death mediated via Fas/FasL interaction is important for homeostasis of cells in the immune system and for maintaining immune-privileged sites in the body. Killing via the Fas/FasL pathway also constitutes an important pathway of killing for cytotoxic T cells. Fas ligand is induced in activated T cells, resulting in activation-induced cell death by the Fas/FasL pathway. Recently it has been shown that the Fas receptor can also be up-regulated following a lesion to the cell, particularly that induced by DNA-damaging agents. This can then result in killing of the cell by a Fas/FasL-dependent pathway. Up-regulation of Fas receptor following DNA damage appears to be p53 dependent.  相似文献

A p53-derived C-terminal peptide induced rapid apoptosis in breast cancer cell lines carrying endogenous p53 mutations or overexpressed wild-type (wt) p53 but was not toxic to nonmalignant human cell lines containing wt p53. Apoptosis occurred through a Fas/APO-1 signaling pathway involving increased extracellular levels of Fas/FasL in the absence of protein synthesis, as well as activation of a Fas/APO-1-specific protease, FLICE. The peptide activity was p53-dependent, and it had no effect in three tumor cell lines with null p53. Furthermore, the C-terminal peptide bound to p53 protein in cell extracts. Thus, p53-dependent, Fas/APO-1 mediated apoptosis can be induced in breast cancer cells with mutant p53 similar to the recently described Fas/APO-1 induced apoptosis by wt p53. However, mutant p53 without p53 peptide does not induce a Fas/APO-1 activation or apoptosis. Docking of the computed low energy conformations for the C-terminal peptide with those for a recently defined proline-rich regulatory region from the N-terminal domain of p53 suggests a unique low energy complex between the two peptide domains. The selective and rapid induction of apoptosis in cancer cells carrying p53 abnormalities may lead to a novel therapeutic modality.  相似文献

Cigarette smoking has been shown to affect human immune responses. We have studied Fas/Fas ligand (FasL) expression, which is involved in the cytotoxic activity, immune privilege, and self-tolerance, and other apoptosis-associated molecule expression of peripheral blood lymphocytes (PBL) in healthy subjects with/without cigarette smoking. We found that expression of FasL protein was detected marginally in the fresh PBL and was induced upon mitogen activation in normal individuals without smoking. In contrast, fresh PBL from those with chronic cigarette smoking exhibited enhanced expression of FasL protein without in vitro mitogen stimulation. Moreover, mitogen stimulation failed to augment FasL protein expression of their lymphocytes, suggesting dysregulation of FasL expression of PBL in individuals with cigarette smoking. In contrast, Fas, Bcl-2, and p53 expression were not significantly different between normal individuals with chronic cigarette smoking and those without smoking. In addition, we found that in vitro brief treatment with nicotine induces and/or enhances FasL mRNA and protein expression of lymphocytes from normal donors without smoking. These results suggest that aberrant FasL expression of lymphocytes is, at least in part, involved in the immune impairment in individuals with chronic cigarette smoking.  相似文献

OBJECTIVE: To understand the role of apoptosis through Fas/Fas ligand (FasL) interaction in the pathogenesis of silicosis, we examined the expression of Fas antigen, FasL and apoptosis in bronchoalveolar lavage fluid lymphocytes obtained from patients with silicosis. MATERIALS AND METHODS: Ten patients with silicosis, and 10 healthy controls were studied. Non-adherent cells were separated and analysed by cytometry for the expression of Fas antigen, FasL, and the co-expression of Fas/FasL. By double staining, we studied the FasL expression on CD4, CD8, CD56 and CD45RO-positive cells. DNA fragmentation was investigated by the terminal deoxy(d) UTP nick end labelling (TUNEL) method. RESULTS: We have found Fas and FasL expression in silicosis patients to be significantly higher than those in healthy controls. Interestingly, 6-18% of lymphocytes from silicosis patients co-expressed Fas and FasL. In silicosis patients, FasL was highly expressed on CD4+, CD56+ and CD45RO+ bronchoalveolar lavage cells. Fas antigen expressing cells showed DNA fragmentation characteristic for apoptosis. CONCLUSION: FasL was significantly expressed on cytotoxic effector and memory cells. The Fas/FasL system is implicated in the inflammatory process observed in silicosis patients.  相似文献

Saikosaponin D is a saponin extract derived from several species of Bupleurum (Umbelliferae), which is used for the treatment of various liver diseases in traditional Chinese medicine. In this study, we report that Saikosaponin D inhibits the cell growth of human lung cancer cell line A549 and provide a molecular understanding of this effect. The results showed that Saikosaponin D inhibited the proliferation of A549 by inducing apoptosis and blocking cell cycle progression in the G1 phase. ELISA assay showed that Saikosaponin D significantly increased the expression of p53 and p21/WAF1 protein, contributing to cell cycle arrest. An enhancement in Fas/APO-1 and its two form ligands, membrane-bound Fas ligand (mFasL) and soluble Fas ligand (sFasL), as well as Bax protein, was responsible for the apoptotic effect induced by Saikosaponin D. Taken together, our study suggests that the induction of p53 and activity of the Fas/FasL apoptotic system may participate in the antiproliferative activity of Saikosaponin D in A549 cells.  相似文献
