光照、湿度和培养基对苎麻疫霉卵孢子产生量的影响   总被引:1,自引:0,他引:1  
采用3因素随机区组设计研究了光照、湿度和培养基对苎麻疫霉(hytophthoraboehmeriae)卵孢子产生量的影响。结果表明,各因子对卵孢子产生量的影响效应大小次序为培养基>光照>湿度,其中培养基和光照两因子影响均在0.01水平上显着。在供试的4种常用培养基上,卵孢子产生量的大小次序为:SLA培养基(SLA)>利马豆培养基(LBA)>V6汁培养基(V6A)>澄清的V6汁培养基(V6B).在设置的3种光照条件中,卵孢子产生量以连续黑暗处理最高,连续光照最低,光照与黑暗交替处理居中。3个试验因子间的所有互作对卵孢子产生量均有极显着的影响。在不同光照、湿度、培养基组合中,卵孢子产生量以低湿+连续黑暗+SLA组合最高,低湿+连续光照+V6B组合最低。  相似文献

以2个雄器大多围生、少数侧生的苎麻疫霉菌株与1个雄器侧生、偶有围生的恶疫霉菌株为材料,采用真菌核糖体基因转录间隔区(ITS)通用引物,PCR扩增3个供试菌株核糖体基因的ITS1和ITS2,并对PCR产物进行了克隆和序列分析。结果是苎麻疫霉的ITS1和ITS2分别由206和453个碱基组成,而恶疫霉则分别由218和415个碱基组成。2个供试苎麻疫霉菌株的ITS1和ITS2的碱基序列同源性均分别为10  相似文献

利用从126个RAPD(Random Amplifled Polymorphic DNAs)随机引物中筛选到的可扩增出清晰条带、主带明显、稳定的8条引物,对采集自江苏、安徽和江西不同寄主的45个Phytophthora boehmeriae菌株进行全基因组DNA RAPD标记遗传多样性分析。选用引物共标出DNA指纹图带68条,其中多态性条带20条,多态性检测率为29.4%,表明该种内不同地区和寄主来源的菌株间变异较小。利用Popgene软件计算供试菌株间的遗传距离并绘制聚类树状图,供试菌株被划分为2个遗传聚类组。菌株间的遗传相似性与菌株的寄主来源有一定的相关性,来自江苏、江西和安徽棉花上的27个菌株被划分在同一遗传聚类组内,而分离自构树、枫杨和苎麻的18个菌株被划分在另一个遗传聚类组。结果还表明菌株间遗传相似性与其地区来源无直接相关性。  相似文献

在实验室条件下研究了不同地区和不同寄主来源的苎麻疫霉的菌落形态,菌丝线性生长速率(以下简称为生长速率)及同宗配合性状的遗传与变异,结果指出苎麻疫霉菌落形态和生长速率的遗传存在3种类型:(1)在单游动孢子后代和自交后代均可稳定遗传;(2)在单游动孢子后代稳定遗传而在自交后代发生变异;(3)在单游动孢子后代和自交后代均发生变异。结果表明,该菌菌落形态和生长速率的遗传具有多样性,上述两性状可以由细胞核杂  相似文献

苎麻疫霉可分泌具有诱抗作用的激发蛋白(α-elicitin),根据α-clicitin第24 ̄30和56 ̄63位保守区氨基酸推导的寡核苷酸引物序列,对苎麻疫霉基因组DNA进行特异PCR扩增反应,发现其扩增的DNA片段大于预计的片段。回收纯化的特异扩增DNA,并进行克隆和测序分析,结果表明特异扩增的clicitin基因亚克隆DNA为570bp,大于预计的117bp。在特异片段中,存在3个内含子将基因  相似文献

本文第一作者于1979年发表乌头霜霉 Peronospora aconiti Yu新种时缺乏对卵孢子的描述,现在四川雅安的乌头 Aconitum cramichaeli Dexb.上发现了该菌的卵孢子,特予补充描述:藏卵器亚球形或卵形,平滑,直径25.8—51.6(平均39.42±4.47)μm;卵孢子球形,浅黄褐色,平滑,单生,不充满藏卵器,直径22.4—37.8(平均28.76±2.96)μm;卵孢子壁厚,其厚度为 2.84—5.04(平均3.77±0.61)μm。  相似文献

研究了苎麻疫霉菌株JS-5自交S1代单卵孢株的菌落形态和生长速率在其单游动孢子无性系后代的遗传与变异。结果表明,亲本菌株JS-5的菌落形态和生长速率在连续4代单游动孢子无性系后代稳定遗传,而该菌株的自交S1代的约2/3单卵孢株的上述性状在其单孢无性后代中发生分离,且上述性状的分离在无性单孢后代中至少可连续保持3 ̄4代。本研究结果提示,苎麻疫霉有性生殖导致上述性状的变异,除有性生殖过程中的基因重组外  相似文献

Cotton blight, caused by the oomycete Phytophthora boehmeriae, is a serious disease of cotton in China. In wet weather conditions, P. boehmeriae is usually the primary pathogen, followed by many saprophytic fungi and pathogens such as Pythium spp., Fusarium spp., Rhizoctonia and others. As P. boehmeriae grows much slower than other pathogens, it is difficult to isolate and identify. A rapid and accurate method for its specific identification is necessary for the detection of blight in infected cotton tissue. The internal transcribed spacer (ITS) regions of ribosomal DNA (rDNA) from three isolates of P. boehmeriae were amplified using the polymerase chain reaction (PCR) with the universal primers DC6 and ITS4. PCR products were cloned and sequenced. The sequences were aligned with those published of 50 other Phytophthora species, and a region specific to P. boehmeriae was used to construct the specific PCR primers PB1 and PB2. Over 106 isolates of 14 Phytophthora species and at least 20 other fungal species were used to check the specificity of the primers. PCR amplification with primers PB1 and PB2 resulted in the amplification of a product of approximately 750 bp only from isolates of P. boehmeriae. Using primers PB1 and PB2, detection sensitivity was approximately 10 fg DNA/μl. In inoculated plant material, P. boehmeriae could be detected in tissue 1 day after inoculation, prior to the appearance of symptoms. The PB primer‐based PCR assay provides an accurate and sensitive method for detecting P. boehmeriae in cotton tissue.  相似文献

Limited information is known on the basic biology of the recently described Phytophthora kernoviae that produces homothallic oospores. In this study, different P. kernoviae isolates were used to investigate oospore maturity, germination, and infection. All isolates produced oospores in V8 broth at 20°C in the dark by 6d. Oospores also formed at 10 and 15°C, but did not form at 25 and 28°C. Continuous light inhibited oospore production of some isolates but had no negative effect on others. Maturation time of the oospores, as noted by germination and staining with tetrazolium bromide, was not much different among the isolates between 2 and 14 weeks. Oospore germination was optimal at 18 and 20°C, and did not occur at 5, 25, and 30°C. Oospore germination under continuous light was higher than in the dark, but individual isolates showed variable results. Rhododendron leaf disks inoculated with oospores and maintained in the dark at 20°C were necrotic after 1 week, while those kept under continuous light did not develop necrosis. The percentage of leaf disks infected with P. kernoviae was lower in the leaves exposed to continuous light (40%) compared to those kept in the dark (100%).  相似文献

Gametangial development and oospore formation were studied, with emphasis on cell wall morphogenesis, on mated cultures (A1xA2) of Phytophthora capsici. In this species, the oogonial and antheridial hyphae interact to produce a typical amphigynous antheridium. The following developmental steps were recognized: 1) contact between oogonial and antheridial initials; 2) penetration of the antheridial initial by the oogonial initial; 3) reemergence of the oogonial initial; 4) oogonial expansion; 5) gametangial delimitation and oogonial wall thickening; 6) penetration of the oogonium by the antheridial fertilization tube; 7) oosphere formation; 8) periplasm degeneration and outer oospore wall formation; and 9) inner oospore wall formation. Electron micrographs were obtained of steps 3–9. Steps 1 and 2 were reconstructed from subsequent events. Steps 3–6 are stages of active wall formation with clear indication of intensive dictyosome activity leading to the formation of numerous wall-destined vesicles of two different sizes and electron densities. No vesicles were seen associated with the development of the inner oospore wall; however, by this stage of development the oosphere cytoplasm exhibited an overall intense electron density that obscured fine detail. Cytoplasmic appearance changed enormously during differentiation, from a developing oogonium rich in mitochondria, ribosomes, rough endoplasmic reticulum, dictyosomes and their vesicles, through an oosphere filled with large finger-print vacuoles and lipid-like bodies, to a mature oospore with a large central vacuole (ooplast) surrounded by a cortex of numerous lipid-like bodies; other organelles are confined to the interstitial space between these storage bodies.  相似文献

在富氧和缺氧环境下室内模拟研究光照对清洁湖区沉积物吸收磷酸盐和对上覆水质变化的影响.结果表明,光照使缺氧环境上覆水体中的pH值显著提高,并增加其溶解氧的含量.上覆水体中磷酸盐浓度的变化在实验初期主要受水-沉积物界面溶解氧含量的影响,表现为富氧组磷酸盐的浓度下降迅速,但随实验时间的推移,光照逐渐起主要作用,表现为实验结束时有光条件下的磷酸盐浓度明显低于无光条件,磷酸盐的减少量和减少速度明显高于无光条件.无论是富氧、缺氧,还是有光、无光,当上覆水体中磷酸盐的浓度很高时,沉积物都能够吸收磷酸盐,但吸收量各不相同.光照对沉积物吸收上覆水中溶解性无机磷酸盐(DIP)的影响受缺氧和富氧环境的限制.  相似文献

Oomycete pathogens that belong to the genus Phytophthora cause devastating diseases in solanaceous crops such as pepper, potato, and tobacco, resulting in crop production losses worldwide. Although the application of fungicides efficiently controls these diseases, it has been shown to trigger negative side effects such as environmental pollution, phytotoxicity, and fungicide resistance in plant pathogens. Therefore, biological control of Phytophthora-induced diseases was proposed as an environmentally sound alternative to conventional chemical control. In this review, progress on biological control of the soilborne oomycete plant pathogens, Phytophthora capsici, Phytophthora infestans, and Phytophthora nicotianae, infecting pepper, potato, and tobacco is described. Bacterial (e.g., Acinetobacter, Bacillus, Chryseobacterium, Paenibacillus, Pseudomonas, and Streptomyces) and fungal (e.g., Trichoderma and arbuscular mycorrhizal fungi) agents, and yeasts (e.g., Aureobasidium, Curvibasidium, and Metschnikowia) have been reported as successful biocontrol agents of Phytophthora pathogens. These microorganisms antagonize Phytophthora spp. via antimicrobial compounds with inhibitory activities against mycelial growth, sporulation, and zoospore germination. They also trigger plant immunity-inducing systemic resistance via several pathways, resulting in enhanced defense responses in their hosts. Along with plant protection, some of the microorganisms promote plant growth, thereby enhancing their beneficial relations with host plants. Although the beneficial effects of the biocontrol microorganisms are acceptable, single applications of antagonistic microorganisms tend to lack consistent efficacy compared with chemical analogues. Therefore, strategies to improve the biocontrol performance of these prominent antagonists are also discussed in this review.  相似文献

应用盆栽试验 ,通过调节不同光照强度并控制其它条件相互一致的条件下 ,研究了光照条件对土壤 植物系统N素状况以及作物 (莴笋 )产量的影响 .结果表明 ,光照强度的改变会引起作物生长状况的相应变化 ,同时也导致土壤N素 (NH 4 N、NO-3 N)状况、作物吸收N量以及作物对N素吸收速度等的改变 .在试验所处的光照强度范围内 ,光照较强时 ,则作物吸收N素的速度较快、吸收N量增加 ,且产量高 ,但土壤中相应的N素含量(NH 4 N、NO-3 N)则只能维持在相对较低的水平 ;光照较弱时 ,则出现与此相反的情况 .  相似文献

作者研究了紫外光对苎麻疫霉的抗甲霜灵诱变效应及对苎麻疫霉生物学特性的影响。结果表明,供试6个苎麻疫霉野生型菌株经菌丝块紫外光药剂诱变和菌丝块药剂驯化3周后,均获得抗甲霜灵突变体,且紫外光药剂诱变处理角变区出现频率明显高于药剂驯化处理,说明紫外光对苎麻疫霉抗甲霜灵突变有一定促进效应。紫外光显著地抑制苎麻疫霉游动孢子的萌发和菌丝的生长,在一定范围内,处理时间愈长,抑制率愈高。苎麻疫霉耐紫外光菌株(经亚致死剂量的紫外光照射处理后存活的游动孢子所形成的菌株)与野生型亲本相比,对温度和pH的敏感性大致相同,但菌落形态有一定变异,菌丝生长速率、卵孢子产生量均显著下降,表明紫外光对苎麻疫霉的菌丝生长和卵孢子产生量有明显的抑制作用。苎麻疫霉耐紫外光菌株EC50值比野生型亲本菌株EC50值提高了23.21%-56.70%,即耐紫外光菌株对甲霜灵敏感性比野生亲本菌株显著下降,这与紫外光诱变试验结果是相一致的。  相似文献
