Low back pain is amongst the top ten risk factors that contribute to disability, ranking higher than diabetes and mental health disease globally as a contributor to years lost to disability (YLD), and escalating as Western societies age. Abundant evidence suggests that intervertebral disc (IVD) damage is central to the origin of pain in the spine. IVD degeneration involves the progressive deterioration of the highly organized disc tissue extracellular matrix, losing its elasticity and hence its' cushioning ability for the spine.Cartilage derived morphogenetic protein-2 (CDMP2) is a small peptide morphogen. Naturally occurring mutations segregate with skeletal defects in IVD development. CDMP2 signalling influences chondrogenic tissue determination, retards osteogenic tissue development and is crucial to early dorso–ventral axis defining events in zebrafish and Xenopus laevis.The potential of biological treatments to offer cutting edge early intervention, tissue regeneration and to preserve spinal motion segments shows great promise. The unique qualities of CDMP2 in IVD tissue formation, delineating discal matrix from vertebral bone, may prove adaptable in therapeutic applications to early discal degeneration.Here we explore the prevalence and origin of backache, the biology of CDMP2 and its potential application as an early intervention to arrest the disc degeneration sequelae.  相似文献

Glyphosate is the active ingredient in a wide range of herbicides used for weed control, including weed control in genetically modified, glyphosate-insensitive crops. In addition, glyphosate herbicides are used for pre-harvest desiccation of glyphosate-sensitive crops. Together, the use of glyphosate leads to residues in livestock feed. In addition to its herbicidal property, glyphosate has documented antimicrobial and mineral-chelating properties. The aim of the present paper is to address, based on the published literature and own observations, whether dietary glyphosate residues may affect livestock gut microbiota and/or mineral status potentially with derived unfavourable effects on animal health and productivity. However, and as reported, literature on the potential effects of glyphosate on livestock is very scarce and mainly reporting in vitro studies; hence, a solid basis of in vivo studies with livestock in physiological and productive phases, particularly sensitive to disorders in mineral status and in the gut microbiota, is needed for drawing final conclusions.  相似文献

BackgroundThe application of mushrooms for health purposes has a long tradition and is very common in Asian countries. This trend is also becoming increasingly popular in the western hemisphere. However, mushrooms from European tradition are being treated in a restrained manner despite having significant potential as drugs or as sources of pure bioactive substances.Aim: The present review provides an overview of the most important mushrooms used in European ethnomedical traditions and explores their pharmacological potential and the challenges for the development of new drugs from these sources of natural products.Method: Mushroom species were selected based on information in old herbal books and dispensaries, uninterrupted use and scientific literature in the PubMed database up to June 2019.Results: Traditional experiences and modern studies have demonstrated that medical mushrooms used in European traditions have promising distinct pharmacological potential mediated through defined mechanisms (anti-tumour, anti-inflammatory, anti-oxidative and anti-bacterial). However, the number of modern chemical, biological and pharmacological studies remains relatively small, and some mushroom species have not been studied at all. Unfortunately, no valid clinical studies can be found. Unlike the case with herbal and fungal drugs from traditional Chinese medicine, we are far from comprehensively exploring this potential.Conclusions: Mushrooms from traditional European medicine have the potential to be used in modern medicine. Considerable research, interdisciplinary collaboration, involvement of the pharmaceutical industry, time and money are necessary to explore this potential not only in the form of dietary supplements but also in the form of approved drugs.  相似文献

Nose-to-brain (N-to-B) delivery offers to protein and peptide drugs the possibility to reach the brain in a non-invasive way. This article is a comprehensive review of the state-of-the-art of this emerging peptide delivery route, as well as of the challenges associated to it. Emphasis is given on the potential of nanosized drug delivery carriers to enhance the direct N-to-B transport of protein or peptide drugs. In particular, polymer- and lipid- based nanocarriers are comparatively analyzed in terms of the influence of their physicochemical characteristics and composition on their in vivo fate and efficacy. The use of biorecognitive ligands and permeation enhancers in order to enhance their brain targeting efficiency is also discussed. The article concludes highlighting the early stage of this research field and its still unveiled potential. The final message is that more explicatory PK/PD studies are required in order to achieve the translation from preclinical to the clinical development phase.  相似文献

The plastic crisis requires drastic measures, especially for the plastics’ end-of-life. Mixed plastic fractions are currently difficult to recycle, but microbial metabolism might open new pathways. With new technologies for degradation of plastics to oligo- and monomers, these carbon sources can be used in biotechnology for the upcycling of plastic waste to valuable products, such as bioplastics and biosurfactants. We briefly summarize well-known monomer degradation pathways and computed their theoretical yields for industrially interesting products. With this information in hand, we calculated replacement scenarios of existing fossil-based synthesis routes for the same products. Thereby, we highlight fossil-based products for which plastic monomers might be attractive alternative carbon sources. Notably, not the highest yield of product on substrate of the biochemical route, but rather the (in-)efficiency of the petrochemical routes (i.e., carbon, energy use) determines the potential of biochemical plastic upcycling. Our results might serve as a guide for future metabolic engineering efforts towards a sustainable plastic economy.  相似文献

Aims

Phosphorus (P) is frequently limiting crop production in agroecosystems. Large progress was achieved in understanding root traits associated with P acquisition efficiency (PAE, i.e. P uptake achieved under low P conditions). Most former studies were performed in controlled environments, and avoided the complexity of soil-root interactions. This may lead to an oversimplification of the root-soil relations. The aim of the present study was, therefore, to identify the dominant root and rhizosphere-related traits determining PAE, in contrasting soil conditions in the field.

Methods

Twenty-three maize hybrids were grown at two contrasting P levels of a long-term P-fertilizer trial in two adjacent soil types: alkaline and neutral. Bulk soil, rhizosphere and root parameters were studied in relation to plant P acquisition.

Results

Soil type had robust effect on PAE. Hybrids’ performance in one soil type was not related to that in the other soil type. In the neutral soil, roots exhibited higher specific root length, higher root/shoot ratio but lower PAE. Best performing hybrids in the neutral soil were characterized by top soil exploration, i.e., greater root surface and topsoil foraging. In contrast, in the alkaline soil, PAE and foraging traits were not correlated, P availability in the rhizosphere was greater than the bulk soil and phosphatase activity was higher, suggesting a ‘mining strategy’ in that case (i.e. traits that facilitate elevated P availability).

Conclusions

These results indicate the key role of environmental factors for roots traits determining high PAE. The study highlights the need to consider soil properties when breeding for high PAE, as various soil types are likely to require different crop ideotypes.

We rely on the helicoidal Peyrard-Bishop model for DNA dynamics. Interaction between nucleotides at a same site belonging to different strands is modelled by a Morse potential energy. This potential depends on two parameters that are different for AT and CG pairs, which is a possible source for inhomogeneity. It was shown recently (Zdravkovic and Sataric 2011) that certain values of these parameters bring about a negligible influence of inhomogeneity on the solitonic dynamics. We propose an experiment that should be carried out in order to determine the values of both of these parameters.  相似文献

Truffle harvests are declining in Europe, due to droughts, and this offers an opportunity for production to be developed in countries such as the UK and Poland, where rainfall tends to be higher. Drier Mediterranean summers seem to be associated with a decrease in the harvest of the Périgord truffle (Tuber melanosporum) in Spain, France and Italy. However, other species, for example the Burgundy truffle (Tuber aestivum) offer opportunities for production in the more temperate environments north of the Alps. Truffles across Europe can be infested by insect larvae, seriously reducing their economic and culinary quality. Here, using a combination of literature sources and a field survey, we present a commentary on insects attacking truffles, aiming to highlight those species that could be potential pests in the British and Polish emergent industries. There is a remarkable disparity in coincidence of records of insects and truffles in these countries, yet a survey in Poland confirms that insects can be abundant. We discuss reasons for this disparity and suggest that biochemical methods could easily be developed for detection of the truffles and their attackers.  相似文献

Head and neck squamous cell carcinomas (HNSCC) are in a group of cancers that are the most resistant to treatment. The survival rate of HNSCC patients has been still very low since last 20 years. The existence of relationship between oncogenic and surrounding cells is probably the reason for a poor response to treatment. Fibroblasts are an important element of tumor stroma which increases tumor cells ability to proliferate. Another highly resistance, tumorigenic and metastatic cell population in tumor microenvironment are cancer initiating cells (CICs). The population of cancer initiating cells can be found regardless of differentiation status of cancer and they seem to be crucial for HNSCC development.In this review, we describe the current state of knowledge about HNSCC biological and physiological tumor microenvironment.  相似文献

Understanding the underlying signalling pathways that enable fouling algae to sense and respond to surfaces is essential in the design of environmentally friendly coatings. Both the green alga Ulva and diverse diatoms are important ecologically and economically as they are persistent biofoulers. Ulva spores exhibit rapid secretion, allowing them to adhere quickly and permanently to a ship, whilst diatoms secrete an abundance of extracellular polymeric substances (EPS), which are highly adaptable to different environmental conditions. There is evidence, now supported by molecular data, for complex calcium and nitric oxide (NO) signalling pathways in both Ulva and diatoms being involved in surface sensing and/or adhesion. Moreover, adaptation to stress has profound effects on the biofouling capability of both types of organism. Targets for future antifouling coatings based on surface sensing are discussed, with an emphasis on pursuing NO-releasing coatings as a potentially universal antifouling strategy.  相似文献

Biosurfactants containing rhamnose and β-hydroxydecanoic acid and called rhamnolipids are reviewed with respect to microbial producers, their physiological role, biosynthesis and genetics, and especially their microbial overproduction, physicochemical properties and potential applications. With Pseudomonas species, more than 100 g l−1 rhamnolipids were produced from 160 g l−1 soybean oil at a volumetric productivity of 0.4 g l−1 h−1. The individual rhamnolipids are able to lower the surface tension of water from 72 mN m−1 to 25–30 mN m−1 at concentrations of 10–200 mg l−1. After initial testing, rhamnolipids seem to have potential applications in combating marine oil pollution, removing oil from sand and in combating zoosporic phytopathogens. Rhamnolipids are also a source of l-rhamnose, which is already used for the industrial production of high-quality flavor components. The application potential of microfungal strains (Cadophora malorum, Mucor circinelloides, Trichoderma viride, nonsporulating culture Mycelia sterilia) as promising lipid producers is investigated. The C. malorum strain is found to be optimal for oil sludge recycling into biofuel. Its palmitic acid content is 52.9%, and it ensures a cetane number of the obtained biodiesel. The ability of the C. malorum strain to degrade n-alkanes and polyaromatic hydrocarbons allows the effective bioremediation of oil sludge to be performed.  相似文献

Laccases belong to multicopper oxidases and are widespread in nature. Currently, mainly fungal laccases are applied in biotechnological processes. One reason for this is that fungal laccases are much better studied. Compared to fungal laccases, bacterial laccases possess some advantageous characteristics like high stability at elevated temperatures and alkaline pH values. Intracellular recombinant expression of bacterial laccases in E. coli makes however downstream processing more complex and time-consuming compared to extracellular expression of fungal enzymes. Here, we demonstrate that cell disruption by cell thermolysis is an efficient and simple method for the isolation and partial purification of recombinant bacterial laccases. Three different laccases, Tth from Thermus thermophilus, CotA from Bacillus subtilis and Ssl1 from Streptomyces sviceus, were used to compare cell disruption by cell thermolysis with sonication and high-pressure homogenization, with and without subsequent heat treatment. Cell thermolysis resulted in high laccase activities per gram of cell wet weight and in the highest specific activities of the laccases. For example, specific activity of Tth laccase after cell thermolysis was 469-fold higher than after sonication. Furthermore, high decolorization activity towards indigo carmine and alizarin red S of these laccases, isolated via cell thermolysis, demonstrate their potential for technical applications.  相似文献
