The first examples of binary palladium(II) derivatives of unsaturated carboxylic acids are reported. It was found that the interaction of Pd3(μ-OAc)6 with the ,β-unsaturated 1-methylcrotonic (tiglic) and crotonic acids leads to the corresponding carboxylates of composition Pd3[μ-O2CC(R′) = CHMe]6, where R′ = Me (1) or H (2). The new compounds have been characterized by elemental analysis, solid and solution IR, 1H and 13C NMR, and ESI mass spectrometry. The crystal structure of 1 has been determined. This molecule displays a central Pd3 cyclic core with Pd–Pd distances of 3.093–3.171 Å. Each Pd–Pd bond is bridged by a pair of carboxylate ligands, one above and the other below the Pd3 plane, providing a square planar coordination for each Pd atom in an approximate D3h overall symmetry arrangement. Solution spectroscopic data show that the bridging η112 interaction of the carboxylates of 1 and 2 is readily displaced, with a change of the ligand to the terminal (η1) coordination mode.  相似文献

The reaction of thiamine with K2PtIICl4 and with PtIVCl4 in the presence of excess NaSCN in aqueous solution gave thiamine salts, (H-thiamine)[Pt(SCN)4] · 3H2O (1) and (H-thiamine)[Pt(SCN)6] · H2O (2), respectively, structures of which have been determined by X-ray diffraction. The thiamine molecule adopts the usual F conformation in each salt. In 1, [Pt(SCN)4]2− ions act as large planar spacers in the crystal lattice and interact scarcely with thiamine, except for a hydrogen bonding with the terminal hydroxy O(5γ). Instead, water molecules form two types of host–guest-like interactions with the pyrimidine and the thiazolium moieties of a thiamine molecule, one being a C(2)–Hwaterpyrimidine bridge and the other being an N(4′)–Hwaterthiazolium bridge. In 2, despite the much larger ion size, octahedral [Pt(SCN)6]2− ions form a C(2)–Hanionpyrimidine bridge and an N(4′)–Hanionthiazolium bridge. An additional hydrogen bonding between the anion and the terminal O(5γ) of thiamine creates a hydrogen-bonded macrocyclic ring {thiaminium–[Pt(SCN)6]2−}2, a supramolecule.  相似文献

The structures of cell-wall mannans isolated from Aphamoascus mephitalus, A. Fulvescens, A. verrucosus, and A. reticulisporus have been investigated by chemical analyses and 1D and 2D 1H and 13C NMR techniques. It was found that all of them consists of a relatively simple comb-like structure of the disaccharide repeating block {→ 6)-[-Man p-(1 → 2)]--Man p-(1 →}. The conformations around the -(1 → 2) and -(1 → 6) linkages in thes kinds of polymers were also studied by using molecular mechanics and dynamics calculations, together with NOE data. The results are similar to those found within the oligosaccharide chains of glycoproteins, with a well-defined conformation for the -(1 → 2) linkage and a certain restriction around the -(1 → 6) bonding imposed by the 2-substitution.  相似文献

The 1H- and 13C-NMR spectra of dogoxin in solution in Me2So-d6 have been assigned completely. Measurement of the 3JC,H values has enabled estimation of the torsional angles involving the bonds linking the digitoxose residues, between the inner digitoxose and the genin unit, and for the unsaturated γ-lactone ring. These values have been supplemented by 1H---1H NOE data. In general, there is good agreement between the conformations in solution (NMR data) and the solid state (X-ray data), and that derived from theological modelling which shows evidence of conformational flexibility. The major difference occurs for the torsion between the genin and the innermost digitoxose residue where molecular dynamics predict the presence of two conformations, one similar to that seen by NMR and the other similar to the X-ray structure.  相似文献

The synthesis of the 3-heptyl, and the eleven isomeric 3-methylheptyl-Δ8-tetrahydrocannabinols (3–7, R and S methyl epimers, and 8) has been carried out. The synthetic approach entailed the synthesis of substituted resorcinols, which were subjected to acid catalyzed condensation with trans-para-menthadienol to provide the Δ8-THC analogue. The 1′-, 2′- and 3′-methylheptyl analogues (3–5) are considerably more potent than Δ8-THC. The 4′-, 5′- and 6′-methylheptyl isomers (6–8) are approximately equal in potency to Δ8-THC.  相似文献

Comparative measurements of bacterial total counts and volumes of flow cytometry (FCM), transmission electron (TEM), and epifluorescence microscopy (EFM), were undertaken during a four week mesocosm experiment. Total counts of bacteria measured by TEM, EFM, and FCM were in the range of 1 · 106−6 cells ml−1, 1 · 106−3 · 1016 cells ml−1, and 5 · 105 cells ml−1 respectively. The mean volume of the bacterial community, measured by means of EFM and TEM, increased from 0.12–0.15 μm3 at the start of the experiment to 0.39–0.53 μm3 at the end. Generally, there was good agreement between the two methods and regression analyses gave r = 0.87 (p < < 0.01) for cell volume and r = 0.97 (p < < 0.01) for cell number. DAPI stained bacteria with volumes less than 0.2 μm3 were not detected by flow cytometry and these were generally an order of magnitude lower than counts made by TEM and EFM. For samples where the mean bacterial cell volume was longer than 0.3 μm3, all three methods were in agreement both with respect to counts and volume estimates.  相似文献

The reaction of perrhenate with 2-hydrazinopyrimidine in MeOH–HCl yields [ReCl31-NNC4H3N2H)(η2-HNNC4H3N2)] (1). The analogous reaction with Na2MoO4 yields [MoCl31-NNC4H3N2H)(η2-HNNHC4H3N2)] (1a). The reaction of 1 with pyrimidine-2-thiol and triethylamine produces [Re(η1-C4H3N2S)(η2-C4H3N2S)(η1-NNC4H3N2)(η2-HNNC4H3N2)] (2), while reaction of 1 with the Schiff base HSC6H4N=C(H)C6H4OH provides [Re(η3-SC6H4N=C(H)C6H4O)(η1-NNC4H3N2)(η2-HNNC4H3N2)]·0.6CH2Cl2 (3·0.6CH2Cl2). The analogous hydrazinopyridine complex of the Schiff base, [Re(η3-SC6H4N=C(H)C6H4O)(η1-NNC5H4N)(η2-HNNC5H4N)] (4), was also synthesized by reacting [ReCl31-NNC5H4NH)(η2-HNNC5H4N)] with HSC6H4N=C(H)C6H4OH. The main features of the primary structure of the octasaccharide, - -Glcp-(1→2)-- -Glcp-(1→2)-[- -GalpNAc- (1→3)]-- -Galp-(1→3)-- -Glcp-(1→3)-[- -Hepp-(1→7)]-- -Hepp-(1→3)-- -Hep, have been determined in the ab initio manner by 1H-n.m.r. spectroscopy without resorting to biochemical methods of analysis. WAY–100635 is the first selective, silent 5–HT1A (5-hydroxytryptamine1A, serotonin-1A) receptor antagonist. We have investigated the use of [3H]WAY–100635 as a quantitative autoradiographic ligand in post-mortem human hippocampus, raphe and four cortical regions, and compared it with the 5–HT1A receptor agonist, [3H]8–OH–DPAT. Saturation studies showed an average Kd for [3H]WAY–100635 binding in hippocampus of 1.1 nM. The regional and laminar distributions of [3H]WAY–100635 binding and [3H]8–OH–DPAT binding were similar. The density of [3H]WAY–100635 binding sites was 60–70% more than that of [3H]8–OH–DPAT in all areas examined except the cingulate gyrus where it was 165% higher. [3H]WAY–100635 binding was robust and was not affected by the post-mortem interval, freezer storage time or brain pH (agonal state). Using [3H]WAY–100635, we confirmed an increase of 5–HT1A receptor binding sites in the frontal cortex in schizophrenia, previously demonstrated with [3H]8–OH–DPAT. Compared to [3H]8–OH–DPAT, [3H]WAY–100635 has two advantages: it has a higher selectivity and affinity for the 5–HT1A receptor, and it recognizes 5–HT1A receptors whether or not they are coupled to a G-protein, whereas [3H]8–OH–DPAT primarily detects coupled receptors. Given these considerations, the [3H]WAY–100635 binding data in schizophrenia clarify two points. First, they indicate that the elevated [3H]8–OH–DPAT binding seen in the same cases is attributable to an increase of 5–HT1A receptors rather than any other binding site. Second, the enhanced [3H]8–OH–DPAT binding in schizophrenia reflects an increased density of 5–HT1A receptors, not an increased percentage of 5–HT1A receptors which are G-protein-coupled. We conclude that [3H]WAY–100635 is a valuable autoradiographic ligand for the qualitative and quantitative study of 5–HT1A receptors in the human brain.  相似文献

σ-Methyl-(η5-indenyl) chromium tricarbonyl (III) rearranges quantitatively into η6-1-endo-methylindene) chromium tricarbonyl (IV) in C6D6 solution at 30–60°C. Methyl group attachment to the positions 2 or 3 of indenyl ligand in (III) has no influence on the activation parameters of this ricochet inter-ring haptotropic rearrangement (ΔG#=23.6 kcal mol−1; ΔH#=18.9±0.2 kcal mol−1; ΔS#=−18.6±0.2 cal K−1 mol−1). (IV) undergoes further irreversible isomerization at 60–120° into (ν6-3-methylindene) chromium tricarbonyl (V) with a higher activation barrier (ΔG#=28.5±0.1 kcal mol−1) via two consecutive [1,5]-sigmatropic hydrogen shifts. The mechanisms of both rearrangements have been studied in detail using density functional theory (DFT) calculations with extended basis sets. Calculations show that the rearrangement (III) → (IV) proceeds in two steps. Methyl group migration from chromium into position 1 of the indenyl ligand is the rate-determining step leading to the formation of the 16-electron intermediate (VII). The calculated activation barrier (Ea=19.6 kcal mol−1) is in good agreement with the experimental one. Further rearrangement (VII) → (V) proceeds via a trimethylenemethane-type transition state (XVIII) with an activation barrier 11.8 kcal mol−1. The coordination of the chromium tricarbonyl group at the six-membered ring has only minor influence on the kinetic parameters of the hydrogen [1,5]-sigmatropic shift in indene.  相似文献

The new organometallic cluster (η24-CO)2(CO)136-C6Me6) has been prepared by the thermolysis of Ru3(CO)12 with hexamethylbenzene in octane and characterised by a single crystal X-ray diffraction study. It is isostructural with the known cluster Ru624-CO)2(CO)136-C6H3Me3) and the metal core constitutnts the same tetrahedral Ru4 unit with two edge-bridging Ru atoms. The mesitylene derivative has been shown to undergo rearrangement to afford the octahedral carbido cluster Ru6C(CO)146-C6H3Me3), but this conversion is not observed for the new hexamethylbenzene derivative.  相似文献

The reversible equilibrium conversion under H2 of [RuCl(dppb) (μ-Cl)]2 (1) to generate (η2-H2) (dppb) (μ-Cl)3RuCl(dppb) in CH2Cl2 (dppb = Ph2P(CH2)4PPh2) has been studied at 0–25 °C by UV-Vis and 31P{1H} NMR spectroscopy, and by stoppe kinetics; the equilibrium constant and corresponding thermodynamic parameters, and the forward and reverse rate constants at 25 °C have been determined. A measured ΔH° value of 0 kJ mol−1 allows for an estimation of an exothermicity of 60 kJ mol−1 for binding an η2-H2 at an Ru(II) centre; a ΔS° value of 60 J mol−1 K−1 indicates that in solution 1 contain s coordinated CH2Cl2. The kinetic and thermodynamic data are compared to those obtained from a previously studied hydrogenation of styrene catalyzed by 1. Preliminary findings on related systems containing Ph2P(CH2)3PPh2 and (C6H11)2P(C6H11)2 are also noted.  相似文献

The reaction of [N(PPh3)2]2[Ni6(CO)12] with Cu(PPh3)xCl (x=1, 2), as well as the degradation of [N(PPh3)2]2[H2Ni12(CO)21] with PPh3, affords the new and unstable dark orange–brown [N(PPh3)2]2[Ni9(CO)16].THF salt in low yields. This salt has been characterized by a CCD X-ray diffraction determination, along with IR spectroscopy and elemental analysis. The close-packed two-layer metal core geometry of the [Ni9(CO)16]2− dianion is directly related to that of the bimetallic [Ni6Rh3(CO)17]3− trianion and may be envisioned to be formally derived from the hcp three-layer geometry of [Ni12(CO)21]4− by the substitution of one of the two outer [Ni3(CO)3(μ−CO)3]2− layers with a face-bridging carbonyl group.  相似文献

The present study was undertaken to characterize the binding activities of propiverine and its N-oxide metabolites (1-methyl-4-piperidyl diphenylpropoxyacetate N-oxide: P-4(N → O), 1-methyl-4-piperidyl benzilate N-oxide: DPr-P-4(N → O)) toward L-type calcium channel antagonist receptors in the rat bladder and brain. Propiverine and P-4(N → O) inhibited specific (+)-[3H]PN 200–110 binding in the rat bladder in a concentration-dependent manner. Compared with that for propiverine, the Ki value for P-4(N → O) in the bladder was significantly greater. Scatchard analysis has revealed that propiverine increased significantly Kd values for bladder (+)-[3H]PN 200–110 binding. DPr-P-4(N → O) had little inhibitory effects on the bladder (+)-[3H]PN 200–110 binding. Oxybutynin and N-desethyl-oxybutynin (DEOB) also inhibited specific (+)-[3H]PN 200–110 binding in the rat bladder. Propiverine, oxybutynin and their metabolites inhibited specific [N-methyl-3H]scopolamine methyl chloride ([3H]NMS) binding in the rat bladder. The ratios of Ki values for (+)-[3H]PN 200–110 to [3H]NMS were markedly smaller for propiverine and P-4(N → O) than oxybutynin and DEOB. Propiverine and P-4(N → O) inhibited specific binding of (+)-[3H]PN 200–110, [3H]diltiazem and [3H]verapamil in the rat cerebral cortex in a concentration-dependent manner. The Ki values of propiverine and P-4(N → O) for [3H]diltiazem were significantly smaller than those for (+)-[3H]PN 200–110 and [3H]verapamil. Further, their Ki values for [3H]verapamil were significantly smaller than those for (+)-[3H]PN 200–110. The Ki values of propiverine for each radioligand in the cerebral cortex were significantly (P < 0.05) smaller than those of P-4(N → O). In conclusion, the present study has shown that propiverine and P-4(N → O) exert a significant binding activity of L-type calcium channel antagonist receptors in the bladder and these effects may be pharmacologically relevant in the treatment of overactive bladder after oral administration of propiverine.  相似文献

The mixture of isomers of silylated cyclopentadiene derivative C5H5CH2CH2Si(OMe)3 (1) has been used for the syntheses of the mononuclear Rh(I) complexes [η5-C5H4(CH2)2Si(OMe)3]Rh(CO)2 (3). [η5-C5H4(CH2)2Si(OMe)3]Rh(COD) (4) and [η5-C5H4(CH2)2Si(OMe)3]Rh(CO)(PPh3) (5). Upon entrapment of 3–5 in silica sol-gel matrices, air stable, leach-proof and recyclable catalysts 6–8 resulted. Their catalytic activities in some hydrogenation processes were compared with those of the non-immobilized complexes 3–5, as well as with those of homogeneous and heterogenized non-silylated analogs, 9–14.  相似文献

The reactions of arene–metal complexes (arene = p-cymene, benzene or pentamethylcyclopentadienyl, metal = Ru, Rh or Os), including 1,3,5-triaza-7-phosphatricyclo-[3.3.1.1]decanephosphine (pta) and chloro co-ligands, with 9-methylguanine, adenine, and a series of nucleosides were studied in water to ascertain the binding modes. The products were characterized by NMR spectroscopy and electrospray ionization mass spectrometry (ESI-MS). Tandem mass spectrometry was found to provide excellent information on preferential binding sites. In general, the N7 position on guanine (the most basic site) was found to be the preferred donor atom for coordination to the metal complexes. The X-ray structures of the precursor complexes, [(η5-C10H15)RhCl(pta-Me)2]Cl2, [(η6-C10H14)OsCl(pta)2]Cl, and [(η6-C6H6)OsCl2(CH3CN)], are also reported.  相似文献

A series of cationic nickel complexes [(η3-methally)Ni(PP(O))]SbF6 (1–4) [PP(O) = Ph2P(CH2)P(O)Ph2 (dppmO) (1), Ph2P(CH2)2P(O)Ph2 (dppeO) (2), Ph2P(CH2)3P(O)Ph2 (dpppO) (3), pTol2P(CH2)P(O)pTol2 (dtolpmO) (4)] has been synthesized in good yields by treatment of [(η3-methally)NiBr]2 with biphosphine monoxides and AgSbF6. The ligands are coordinated in a bidentate way. Starting from [(η3-all)PdI]2 the cationic complexes [(η3-all)PP(O))]Y (8–14). [PP(O) = dppmO, dppeO, dpppO, dtolpmO;Y = BF4, SbF6, CF3SO3, pTolSO3] were synthesized in good yields. The coordination mode of the ligand is dependent on the backbone and the anion, revealing a monodentate coordination with dppmO for stronger coordinating anions. The intermediates [(η3-all)Pd(I)(PP(O)-κ1-P)] (5–7) [PP(O) = dppmO (5), dppeO (6), dtolpmO (7)] were isolated and characterized. Neutral methyl complexes [(Cl)(Me)Pd(PP(O))] (15–18). [PP(O) = dppmO (15), dppeO (16), dpppO (17), dtolpmO (18)] can easily be obtained in high yields starting from [(cod)PdCl2]. For dppmO two different routes are presented. The structure of [(Me)(Cl)Pd{;Ph2P(CH2-P(O)Ph22-P,O};] · CH2Cl2 (15) with the chlorine atom trans to phosphorus was determined by X-ray diffraction.  相似文献

Vasoactive intestinal peptide (VIP) bound with high affinity (Kd 0.13 nmol/l) to receptors on the human glioma cell line U-343 MG Cl 2:6. The receptors bound the related peptides helodermin. PHM and secretin with 10, 400 and 5000 times lower affinity, respectively. Deamidated VIP (VIP-COOH) and [des-His1]VIP bound with 10 and 100 times lower affinity. The fragment VIP(7–28) displaced 25% of the receptor-bound 125I-VIP whereas VIP(16–28) and VIP(1–22-NH2) were inactive. The binding of 125I-VIP could be completely inhibited by 10 μmol/l of the antagonists [N-Ac-Tyr1,D-Phe2]GRF(1–29)-NH2, [pCl-D-Phe6,Leu17]VIP and VIP(10–28); in contrast, the antagonist L-8-K was inactive. Affinity labeling showed that VIP bound to proteins with Mr's of 75 kDa, 66 kDa and 50 kDa, respectively. Following binding, the peptide was rapidly internalized, and at steady-state only 20% of cell-associated 125I-VIP was bound to receptors on the cell surface. The internalized 125I-VIP was completely degraded to 125I-tyrosine which was released from the cells. Degradation of internalized 125I-VIP was significantly reduced by chloroquine phenantroline and pepstatin-A. Surface binding and internalization of 125I-VIP was increased 3 times by phenantroline, and pepstatin-A caused a 5 times increase in surface binding. Chloroquine reduced surface-bound 125I-VIP, but caused retention of internalized 125I-VIP.  相似文献

Prostaglandin (PG) and thromboxane B2 (TXB2) biosynthesis was studied in cultured astrocytes from neonatal rat brain hemispheres. After two weeks of cultivation, prostanoids were formed with the spectrum: PGD2 > TXB2 > PGF2 > PGE2, as measured by specific radioimmunoassays. Under basal conditions PGD2 biosynthesis (9.55 ng/mg protein/15 min) was in the same order of magnitude as the sum of the other prostanoids. The formation of prostanoids was stimulated in a concentration dependent manner (up to 6–10 fold) by the calcium ionophore A 23187 (0.01–10 μM) as well as by melittin (0.01–5 μg/ml), phospholipase A2 (10–40 U/ml) and phospholipase C (0.01–1 U/ml). Basal and evoked PG and TXB2 biosynthesis depended on the availability of Ca2+, as demonstrated in Ca2+ free incubation medium containing Na2EDTA (1 μM), or with verapamil (100 μM) and 3,4,5-trimethoxybenzoic acid-8-(diethylamino)-octylester-HCl (TMB-8, 1–100 μM). Indomethacin (10 μM), mepacrine (100 μM) and p-bromophenacylbromide (50 μ M) inhibited basal and evoked PG formation. Thin-layer chromatography (TLC) detection after incubation of the cells with [3H]arachidonic acid (1 μCi/ml, for 60 min) confirmed the results obtained by radioimmunoassay. Incubation of [3H]arachidonic acid labelled cells with inonophore or phospholipases, followed by lipid extraction and TLC, showed that A 23187 liberated [3H]arachidonic acid predominantly from phosphatidylethanolamine, whereas phospholipase A2 and C reduced mainly the labelling of the phosphatidyl-inositol/-choline fraction. Potassium depolarization of the cells did not enhance prostanoid formation. Similarly, drugs with affinity to - or β-adrenoceptors, or to dopamine-, 5-hydroxytryptamine-, muscarine-, histamine-, glutamate-, aspartate-, GABA, adenosine- and opioid-receptors failed to stimulate prostanoid biosynthesis. Also compounds like angiotensin, bradykinin and thrombin were ineffective in this respect.

In conclusion, our results confirm that cultured astrocytes possess the complete pattern of enzymes necessary for prostanoid formation and hence might play a crucial role in brain prostanoid biosynthesis. Stimulation of prostanoid biosynthesis involves Ca2+-dependent activation of phospholipase A2, cyclooxygenase reaction and further PG metabolism. However, the endogenous stimulus for enhanced prostanoid synthesis in the brain still has to be established.  相似文献

To probe the importance of a proposed β-turn within residues S9-R12 of PACAP for recognition by VIP/PACAP receptors, compounds 1 and 2, two conformationally restricted analogues of PACAP27 incorporating respectively (S)- or (R)-IBTM as type II or II′ β-turn dipeptide mimetic at the Y10-S11 position, were synthesized. According to 1H NMR conformational analyses in aqueous solution and 30% TFE, both PACAP27 and the [S-IBTM10,11]PACAP27 analogue 1 adopt similar ordered structures. PACAP27 shows an N-terminal disordered region (residues H1-F6) and an -helical conformation within segment T7–L27. For residues S9–R12, our data seem more compatible with a segment of the -helix than with the β-turn previously proposed for this fragment. In compound 1 the -helix, also spanning T7–L27 residues, appears slightly distorted at the N-terminus relative to the native peptide. Although this distortion could lead to the marked decrease in binding affinity of this compound at the VIP/PACAP receptors, the lack of the Y10 side chain in analogues 1 and 2 could also significantly affect the binding of these compounds.  相似文献
