Induction of high-frequency shoot regeneration using nodal segments containing axillary buds from a 1-yr-old mother plants of Cannabis sativa was achieved on Murashige and Skoog (MS) medium containing 0.05–5.0 μM thidiazuron. The quality and quantity of regenerants were better with thidiazuron (0.5 μM thidiazuron) than with benzyladenine or kinetin. Adding 7.0 μM of gibberellic acid into a medium containing 0.5 μM thidiazuron slightly increased shoot growth. Elongated shoots when transferred to half-strength MS medium supplemented with 500 mg l−1 activated charcoal and 2.5 μM indole-3-butyric acid resulted in 95% rooting. The rooted plants were successfully acclimatized in soil. Following acclimatization, growth performance of 4-mo-old in vitro propagated plants was compared with ex vitro vegetatively grown plants of the same age. The photosynthesis and transpiration characteristics were studied under different light levels (0, 500, 1,000, 1,500, or 2,000 μmol m−2 s−1). An increase in photosynthesis was observed with increase in the light intensity up to 1,500 μmol m−2 s−1 and then decreased subsequently at higher light levels in both types of plants. However, the increase was more pronounced at lower light intensities below 500 μmol m−2 s−1. Stomatal conductance and transpiration increased with light intensity up to highest level (2000 μmol m−2 s−1) tested. Intercellular CO2 concentration (C i) and the ratio of intercellular CO2 concentration to ambient CO2 (C i/C a) decreased with the increase in light intensity in both in vitro as well as ex vitro raised plants. The results show that in vitro propagated and hardened plants were functionally comparable to ex vitro plants of same age in terms of gas and water vapor exchange characteristics, within the limits of this study.  相似文献

Summary In vitro banana (Musa spp.) shoots were cultured under photomixotrophic (30 gl−1 sucrose and 0.2 h−1 number of air exchanges of culture vessels) and photoautotrophic (0 gl−1 sucrose and 3.9 h−1 number of air exchanges) conditions for 28 d in 370 cm3 Magenta boxes (GA7-type) containing 70 ml of half-strength Murashige and Skoog (MS) medium with 22.2 μM N6-benzyladenine (BA). The effects of varying CO2 concentration (475 or 1340 μmol mol−1) and light intensity (photosynthetic photon flux (PPF) of 100 or 200 μmol m−2 s−1) were investigated. Fresh and dry weights of banana shoots grown photomixotrophically were significantly greater on day 28 than those grown photoautotrophically. Photoautorophic shoots had a larger number of unfolded leaves and greater leaf area than photomixotrophic plants by days 14 and 28, regardless of CO2 concentration. The shoot fresh and dry weights on day 14 in photoautotrophic conditions were significantly greater at PPF of 200 μmol m−2 s−1 than at 100 μmol m−2 s−1. The increase in net photosynthetic rate of photoautotrophic banana shoots was significant compared with photomixotrophic shoots. The multiplication ratio of in vitro banana shoots grown photoautotrophically in a 28-d culture period was the greatest at 100 μmol m−2 s−1 PPF and 475 μmol mol−1 CO2.  相似文献

Response to irradiance of leaf net photosynthetic rates (P N) of four carrot cultivars: Cascade, Caro Choice (CC), Oranza, and Red Core Chantenay (RCC) were examined in a controlled environment. Gas exchange measurements were conducted at photosynthetic active radiation (PAR) from 100 to 1 000 μmol m−2 s−1 at 20 °C and 350 μmol (CO2) mol−1(air). The values of P N were fitted to a rectangular hyperbolic nonlinear regression model. P N for all cultivars increased similarly with increasing PAR but Cascade and Oranza generally had higher P N than CC. None of the cultivars reached saturation at 1 000 μmol m−2 s−1. The predicted P N at saturation (P Nmax) for Cascade, CC, Oranza, and RCC were 19.78, 16.40, 19.79, and 18.11 μmol (CO2) m−2 s−1, respectively. The compensation irradiance (I c) occurred at 54 μmol m−2 s−1 for Cascade, 36 μmol m−2 s−1 for CC, 45 μmol m−2 s−1 for Oranza, and 25 μmol m−2 s−1 for RCC. The quantum yield among the cultivars ranged between 0.057–0.033 mol(CO2) mol−1(PAR) and did not differ. Dark respiration varied from 2.66 μmol m−2 s−1 for Cascade to 0.85 μmol m−2 s−1 for RCC. As P N increased with PAR, intercellular CO2 decreased in a non-linear manner. Increasing PAR increased stomatal conductance and transpiration rate to a peak between 600 and 800 μmol m−2 s−1 followed by a steep decline resulting in sharp increases in water use efficiency. Highly productive papyrus (Cyperus papyrus L.) wetlands dominate many permanently flooded areas of tropical East Africa; however, the cycling of carbon and water within these ecosystems is poorly understood. The objective of this study was to utilise Eddy Covariance (EC) techniques to measure the fluxes of carbon dioxide and water vapour between papyrus vegetation and the atmosphere in a wetland located near Jinja, Uganda on the Northern shore of Lake Victoria. Peak, midday rates of photosynthetic CO2 net assimilation were approximately 40 μmol CO2 m−2 s−1, while night time losses through respiration ranged between 10 and 20 μmol COm−2 s−1. Numerical integration of the flux data suggests that papyrus wetlands have the potential to sequester approximately 0.48 kg C m−2 y−1. The average daily water vapour flux from the papyrus vegetation through canopy evapotranspiration was approximately 4.75 kg H2O m−2 d−1, which is approximately 25% higher than water loss through evaporation from open water.  相似文献

The effects of temperature, irradiance, and daylength on Sargassum horneri growth were examined at the germling and adult stages to discern their physiological differences. Temperature–irradiance (10, 15, 20, 25, 30°C × 20, 40, 80 μmol photons m−2s−1) and daylength (8, 12, 16, 24 h) experiments were carried out. The germlings and blades of S. horneri grew over a wide range of temperatures (10–25°C), irradiances (20–80 μmol photons m−2s−1), and daylengths (8–24 h). At the optimal growth conditions, the relative growth rates (RGR) of the germlings were 21% day−1 (25°C, 20 μmol photons m−2s−1) and 13% day−1 (8 h daylength). In contrast, the RGRs of the blade weights were 4% day−1 (15°C, 20 μmol photons m−2s−1) and 5% day−1 (12 h daylength). Negative growth rates were found at 20 μmol photons m−2s−1 of 20°C and 25°C treatments after 12 days. This phenomenon coincides with the necrosis of S. horneri blades in field populations. In conclusion, we found physiological differences between S. horneri germlings and adults with respect to daylength and temperature optima. The growth of S. horneri germlings could be enhanced at 25°C, 20 μmol photons m−2s−1, and 8 h daylength for construction of Sargassum beds and restoration of barren areas.  相似文献

CO2 exchange of the endolithic lichen Verrucaria baldensis was measured in the laboratory under different conditions of water content, temperature, light, and CO2 concentration. The species had low CO2 exchange rates (maximum net photosynthesis: c. 0.45 μmol CO2 m−2 s−1; maximum dark respiration: c. 0.3 μmol CO2 m−2 s−1) and a very low light compensation point (7 μmol photons m−2 s−1 at 8°C). The net photosynthesis/respiration quotient reached a maximum at 9–15°C. Photosynthetic activity was affected only after very severe desiccation, when high resaturation respiratory rates were measured. Microclimatic data were recorded under different weather conditions in an abyss of the Trieste Karst (northeast Italy), where the species was particularly abundant. Low photosynthetically active radiation (normally below 40 μmol photons m−2 s−1), very high humidities (over 80%), and low, constant temperatures were measured. The microalga Haematococcus pluvialis Flotow has been the subject of a number of studies concerned with maximizing astaxanthin production for use in animal feeds and for human consumption. Several of these studies have specifically attempted to ascertain the optimal temperature and irradiance combination for growth of H. pluvialis, but there has been a great deal of disagreement between laboratories. “Ideal” levels of temperature and irradiance have been reported to range from 14 to 28°C and 30 to 200 μmol photons m−2 s−1. The objective of the present study was to simultaneously explore temperature and irradiance effects for a single strain of H. pluvialis (UTEX 2505) across an experimental region that encompassed the reported “optimal” combinations of these factors for multiple strains. To this end, a two-dimensional experimental design based on response surface methodology (RSM) was created. Maximum growth rates for UTEX 2505 were achieved at 27°C and 260 μmol photons m−2 s−1, while maximum quantum yield for stable charge separation at PSII (Fv/Fm) was achieved at 27°C and 80 μmol photons m−2 s−1. Maximum pigment concentrations correlated closely with maximum Fv/Fm. Numeric optimization of growth rate and Fv/Fm produced an optimal combination of 27°C and 250 μmol photons m−2 s−1. Polynomial models of the various response surfaces were validated with multiple points and were found to be very useful for predicting several H. pluvialis UTEX 2505 responses across the entire two-dimensional experimental design space.  相似文献

Rates of soil respiration (CO2 effluxes), subsurface pore gas CO2/O2 concentrations, soil temperature and soil water content were measured for 15 months in two temperate and contrasting Danish forest ecosystems: beech (Fagus sylvatica L.) and Norway spruce (Picea abies [L.] Karst.). Soil CO2 effluxes showed a distinct seasonal trend in the range of 0.48–3.3 μmol CO2 m−2 s−1 for beech and 0.50–2.92 μmol CO2 m−2 s−1 for spruce and were well-correlated with near-surface soil temperatures. The soil organic C-stock (upper 1 m including the O-horizon) was higher in the spruce stand (184±23 Mg C ha−1) compared to the beech stand (93±19 Mg C ha−1) and resulted in a faster turnover time as calculated by mass/flux in soil beneath the beech stand (28 years) compared to spruce stand (60 years). Observed soil CO2 concentrations and effluxes were simulated using a Fickian diffusion-reaction model based on vertical CO2 production rates and soil diffusivity. Temporal trends were simulated on the basis of observed trends in the distribution of soil water, temperature, and live roots as well as temperature and water content sensitivity functions. These functions were established based on controlled laboratory incubation experiments. The model was successfully validated against observed soil CO2 effluxes and concentrations and revealed that temporal trends generally could be linked to variations in subsurface CO2 production rates and diffusion over time and with depths. However, periods with exceptionally high CO2 effluxes (> 20 μmol CO2 m−2 s−1) were noted in March 2000 in relation to drying after heavy rain and after the removal of snow from collars. In an experimental site for reforestation of degraded area, three-year-old plants of Bertholletia excelsa Humb. & Bonpl. were subjected to different fertilization treatments: T0 = unfertilized control, T1 = green fertilization (branches and leaves) and T2 = chemical fertilization. Higher net photosynthetic rates (P N) were observed in T1 [13.2±1.0 μmol(CO2) m−2 s−1] compared to T2 [8.0±1.8 μmol(CO2) m−2 s−1] and T0 [4.8±1.3 μmol(CO2) m−2 s−1]. Stomatal conductance (g s), transpiration rate (E) and water use efficiency (WUE) of individuals of T1 and T2 did not differ significantly, however, they were by 88, 55 and 63%, respectively, higher in T1 than in the control. The mean values of variable fluorescence (Fv), performance index (P.I.) and total chlorophyll [Chl (a+b)] were higher in T1. Our results indicate that green fertilization improves photosynthetic structure and function in plants of B. excelsa in young phase.  相似文献

Symbiotic dinoflagellates of the species Amphidinium are expected to be pharmaceutically useful microalgae because they produce antitumor macrolides. A microalgae production system with a large number of cells at a high density has been developed for the efficient production of macrolide compounds. In the present study, the effects of culture conditions on the cellular growth rate of dinoflagellates were investigated to determine the optimum culture conditions for obtaining high yields of microalgae. Amphidinium species was cultured under conditions with six temperature levels (21–35°C), six levels of photosynthetic photon flux density (15–70 μmol photons m−2 s−1), three levels of CO2 concentration (0.02–0.1%), and three levels of O2 concentration (0.2–21%). The number of cells cultured in a certain volume of solution was monitored microscopically and the cellular growth rate was expressed as the specific growth rate. The maximum specific growth rate was 0.022 h−1 at a temperature of 26°C and O2 concentration of 5%, and the specific growth rate was saturated at a CO2 concentration of 0.05%, a photosynthetic photon flux density of 35 μmol photons m−2 s−1 and a photoperiod of 12 h day−1 upon increasing each environmental parameter. The results demonstrate that Amphidinium species can multiply efficiently under conditions of relatively low light intensity and low O2 concentration.  相似文献

The concept of using higher plants to maintain a sustainable life support system for humans during long-duration space missions is dependent upon photosynthesis. The effects of extended exposure to microgravity on the development and functioning of photosynthesis at the leaf and stand levels were examined onboard the International Space Station (ISS). The PESTO (Photosynthesis Experiment Systems Testing and Operations) experiment was the first long-term replicated test to obtain direct measurements of canopy photosynthesis from space under well-controlled conditions. The PESTO experiment consisted of a series of 21–24 day growth cycles of Triticum aestivum L. cv. USU Apogee onboard ISS. Single leaf measurements showed no differences in photosynthetic activity at the moderate (up to 600 μmol m−2 s−1) light levels, but reductions in whole chain electron transport, PSII, and PSI activities were measured under saturating light (>2,000 μmol m−2 s−1) and CO2 (4000 μmol mol−1) conditions in the microgravity-grown plants. Canopy level photosynthetic rates of plants developing in microgravity at ∼280 μmol m−2 s−1 were not different from ground controls. The wheat canopy had apparently adapted to the microgravity environment since the CO2 compensation (121 vs. 118 μmol mol−1) and PPF compensation (85 vs. 81 μmol m−2 s−1) of the flight and ground treatments were similar. The reduction in whole chain electron transport (13%), PSII (13%), and PSI (16%) activities observed under saturating light conditions suggests that microgravity-induced responses at the canopy level may occur at higher PPF intensity.  相似文献

The ability of spring barley (Hordeum vulgare cv. Akcent) to adjust the composition and function of the photosynthetic apparatus to growth irradiances of 25–1200 μmol m−2 s−1 was studied by gas exchange and chlorophyll a fluorescence measurements and high-performance liquid chromatography. The increased growth irradiance stimulated light- and CO2-saturated rates of CO2 assimilation expressed on a leaf area basis up to 730 μmol m−2 s−1 (HL730), whereas at an irradiance of 1200 μmol m−2 s−1 (EHL1200) both rates decreased significantly. Further, the acclimation to EHL1200 was associated with an extremely high chlorophyll a/b ratio (3.97), a more than doubled xanthophyll cycle pool (VAZ) and a six-fold higher de-epoxidation state of the xanthophyll cycle pigments as compared to barley grown under 25 μmol m−2 s−1 (LL25). EHL1200 plants also exhibited a long-term inhibition of Photosystem II (PS II) photochemical efficiency (F v/F m). Photosynthetic capacity, chlorophyll a/b and VAZ revealed a linear trend of dependence on PS II excitation pressure in a certain range of growth irradiances (100–730 μmol m−2 s−1). The deviation from linearity of these relationships for EHL1200 barley is discussed. In addition, the role of increased VAZ and/or accumulation of zeaxanthin and antheraxanthin in acclimation of barley to high irradiance is studied with respect to regulation of non-radiative dissipation and/or photochemical efficiency within PS II. Physiological measurements were used to investigate the dependence of photosynthesis on light, temperature, and intercellular carbon dioxide (CO2) levels in the C4 marsh grass Spartina alterniflora. Functional relationships between these environmental variables and S. alterniflora physiological responses were then used to improve C4-leaf photosynthesis models. Field studies were conducted in monocultures of S. alterniflora in Virginia, USA. On average, S. alterniflora exhibited lower light saturation values (~1000 μmol m−2 s−1) than observed in other C4 plants. Maximum carbon assimilation rates and stomatal conductance to water vapor diffusion were 36 μmol (CO2) m−2 s−1 and 200 mmol (H2O) m−2 s−1, respectively. Analysis of assimilation-intercellular CO2 and light response relationships were used to determine Arrhenius-type temperature functions for maximum rate of carboxylation (V cmax), phosphoenolpyruvate carboxylase activity (V pmax), and maximum electron transport rate (J max). Maximum V cmax values of 105 μmol m−2 s−1 were observed at the leaf temperature of 311 K. Optimum V pmax values (80.6 μmol m−2 s−1) were observed at the foliage temperature of 308 K. The observed V pmax values were lower than those in other C4 plants, whereas V cmax values were higher, and more representative of C3 plants. Optimum J max values reached 138 μmol (electrons) m−2 s−1 at the foliage temperature of 305 K. In addition, the estimated CO2 compensation points were in the range of C3 or C3–C4 intermediate plants, not those typical of C4 plants. The present results indicate the possibility of a C3–C4 intermediate or C4-like photosynthetic mechanism rather than the expected C4-biochemical pathway in S. alterniflora under field conditions. In a scenario of atmospheric warming and increased atmospheric CO2 concentrations, S. alterniflora will likely respond positively to both changes. Such responses will result in increased S. alterniflora productivity, which is uncharacteristic of C4 plants.  相似文献

Gross photosynthesis and respiration rates of leaves at different canopy heights in a Rhizophora stylosa Griff. stand were measured monthly over 1 year at Manko Wetland, Okinawa Island, Japan, which is the northern limit of its distribution. The light-saturated net photosynthesis rate for the leaves at the top of the canopy showed a maximum value of 17 μmol CO2 m−2 s−1 in warm season and a minimum value of 6 μmol CO2 m−2 s−1 in cold season. The light-saturated gross photosynthesis and dark respiration rates of the leaves existing at the top of the canopy were 2−7 times and 3–16 times, respectively, those of leaves at the bottom of the canopy throughout the year. The light compensation point of leaves showed maximum and minimum peaks in warm season and cold season, respectively. The annual canopy gross photosynthesis, foliage respiration, and surplus production were estimated as 117, 49, and 68 t CO2 ha−1 year−1, respectively. The energy efficiency of the annual canopy gross photosynthesis was 2.5%. The gross primary production GPP fell near the regression curve of GPP on the product of leaf area index and warmth index, the regression curve which was established for forests in the Western Pacific with humid climates.  相似文献

The effects of light and nitrogen deficiency on biomass, fatty acid content and composition were studied in Parietochloris incisa, the unicellular freshwater chlorophyte accumulating very high amounts of arachidonic-acid-rich triacylglycerols. P. incisa cultures grown on complete nutrient medium and under high light (400 μmol photons m− 2 s−1) showed the highest rate of growth in comparison to medium (200 μmol photons m−2 s−1) and low (35 μmol photons m−2 s−1) light intensity. Cultures grown under high light (on complete BG-11 medium) attained higher volumetric contents of total fatty acids and arachidonic acid due to greater increase in biomass. Nitrogen starvation brought about a strong increase in the arachidonic acid proportion of total fatty acids. Thus, adjustments to cultivation conditions could serve as an efficient tool for manipulation of yield and relative content of arachidonic acid in P. incisa. The significance of the changes in lipid metabolism for adaptation of P. incisa to high-light stress and nitrogen deficiency is also discussed.  相似文献

Although suckers and seedlings can be used for the propagation of bromeliads, the low number of propagules and cross-variation limit their uniformity and mass cultivation. In this study, high-efficiency shoot organogenesis and plant regeneration were achieved on callus derived from petal and ovary explants of Aechmea fasciata (Bromeliaceae). Calluses were induced on half-strength Murashige and Skoog inorganic salts (1/2MS) supplemented with 1.0–1.5 mg l−1 2,4-dichlorophenoxyacetic acid in combination with 1.0 or 0.5 mg l−1 α-naphthaleneacetic acid (NAA), and shoots regenerated after transfer to 1/2MS basal medium containing the combination of 1.0 mg l−1 NAA + 0.5 mg l−1 1-phenyl-3-(1,2,3-thiadiazol-5-yl) urea. Those plantlets grown under a middle light intensity (50 μmol m−2 s−1) showed a dramatic increase in survival percentage (up to 95%) and the maximum number of newly developing roots. The plantlets that were transplanted onto pots were successfully grown in the greenhouse.  相似文献

Morphology and culture studies on germlings of Sargassum thunbergii (Mertens et Roth) Kuntze were carried out under controlled laboratory conditions. Growth characteristics of these germlings grown under different temperatures (from 10 to 25°C), irradiances (from 9 to 88 μmol photons m−2 s−1), and under blue and white light conditions are described. The development of embryonic germlings follows the classic “8 nuclei 1 egg” type described for Sargassaceae. Fertilized eggs spent 5–6 h developing into multicellular germlings with abundant rhizoids after fertilization. Under conditions of 20°C, 44 μmol photons m−2 s−1 and photoperiod of 12 h, young germlings with one or two leaflets reached 2–3 mm in length after 8 weeks. Temperature variations (10, 15, 20, 25°C) under 88 μmol photons m−2 s−1 significantly influenced the growth rate within the first week, although this effect became less obvious after 8 weeks, especially at 15 and 20°C. Variation in germling growth was highly significant under different irradiances (9, 18, 44, 88 μmol photons m−2 s−1) at 25°C. Low temperature (10°C) reduced germling growth. Growth of germlings cultured under blue light was lower than in white light. Optimal growth of these germlings occurred at 25°C and 44 μmol photons m−2 s−1.  相似文献

The branching zooxanthellate soft coral Sinularia flexibillis releases antimicrobial and toxic compounds with potential pharmaceutical importance. As photosynthesis by the symbiotic algae is vital to the host, the light-dependency of the coral, including its specific growth rate (μ day−1) and the physiological response to a range of light intensities (10–1,000 μmol quanta m−2 s−1) was studied for 12 weeks. Although a range of irradiances from 100 to 400 μmol quanta m−2 s−1 was favorable for S. flexibilis, based on chlorophyll content, a light intensity around 100 μmol quanta m−2 s−1 was found to be optimal. The contents of both zooxanthellae and chlorophyll a were highest at 100 μmol quanta m−2 s−1. The specific budding rate showed almost the same pattern as the specific growth rate. The concentration of the terpene flexibilide, produced by this species, increased at high light intensities (200–600 μmol quanta m−2 s−1).  相似文献

Characterizing the spatial variation in the CO2 flux at both large and small scales is essential for precise estimation of an ecosystem’s CO2 sink strength. However, little is known about small-scale CO2 flux variations in an ecosystem. We explored these variations in a Kobresia meadow ecosystem on the Qinghai-Tibetan plateau in relation to spatial variability in species composition and biomass. We established 14 points and measured net ecosystem production (NEP), gross primary production (GPP), and ecosystem respiration (Re) in relation to vegetation biomass, species richness, and environmental variables at each point, using an automated chamber system during the 2005 growing season. Mean light-saturated NEP and GPP were 30.3 and 40.5 μmol CO2 m−2 s−1 [coefficient of variation (CV), 42.7 and 29.4], respectively. Mean Re at 20°C soil temperature, Re20, was −10.9 μmol CO2 m−2 s−1 (CV, 27.3). Re20 was positively correlated with vegetation biomass. GPPmax was positively correlated with species richness, but 2 of the 14 points were outliers. Vegetation biomass was the main determinant of spatial variation of Re, whereas species richness mainly affected that of GPP, probably reflecting the complexity of canopy structure and light partitioning in this small grassland patch.  相似文献

This work describes the long-term acclimation of the halotolerant microalga Dunaliella viridis to different photon irradiance, ranging from darkness to 1500 μmol m−2 s−1. In order to assess the effects of long-term photoinhibition, changes in oxygen production rate, pigment composition, xanthophyll cycle and in vivo chlorophyll fluorescence using the saturating pulse method were measured. Growth rate was maximal at intermediate irradiance (250 and 700 μmol m−2 s−1). The increase in growth irradiance from 700 to 1500 μmol m−2 s−1 did not lead to further significant changes in pigment composition or EPS, indicating saturation in the pigment response to high light. Changes in Photosystem II optimum quantum yield (Fv/Fm) evidenced photoinhibition at 700 and especially at 1500 μmol m−2 s−1. The relation between photosynthetic electron flow rate and photosyntetic O2 evolution was linear for cultures in darkness shifting to curvilinear as growth irradiance increased, suggesting the interference of the energy dissipation processes in oxygen evolution. Carbon assimilation efficiencies were studied in relation to changes in growth rate, internal carbon and nitrogen composition, and organic carbon released to the external medium. All illuminated cultures showed a high capability to maintain a C:N ratio between 6 and 7. The percentage of organic carbon released to the external medium increased to its maximum under high irradiance (1500 μmol m−2 s−1). These results suggest that the release of organic carbon could act as a secondary dissipation process when the xanthophyll cycle is saturated.
