Twenty stomachs of Bactrian camels (Camelus bactrianus) were studied by gross dissection. Based on the configuration of the stomach and the structure of the mucous membrane, the stomach was divided into three ventricles that differ from the arrangement described for ox and sheep. The first and second ventricles of the proventriculus of camel form one stomach rather than two different stomachs. These ventricles of the proventriculus do not correspond to the rumen and reticulum of ox and sheep. The third ventricle appears to be the abomasum. One part of the abomasum has reticular mucosal folds that indicate it is not the reticulum. A second part of the abomasum has longitudinal mucosal folds suggesting it is not the omasum. Three glandular sac areas associated with the preventriculus and abomasum are also described.  相似文献

1. The activities of rhodanese and beta-mercaptopyruvate sulfurtransferase (MST) in different organs of sheep and cattle were measured. 2. Liver, kidney, omasum, and rumen were the richest sources of both enzymes. The activities of both enzymes in other organs of the sheep and the cattle decreased in the order of lung, brain, heart, abomasum, lymph node, urinary bladder, spleen, and the skeletal muscle. 3. The activities of both enzymes in most organs of the sheep were higher than the cattle. 4. Both enzymes showed higher activities in the epithelial layers than the muscular layers of rumen, omasum and reticulum. 5. In most of the tissues of both species the level of rhodanese activity was greater than MST.  相似文献

Pure ochratoxin A (OA) was added to buffered rumen fluid collected from fistulated cows and incubated under anaerobic conditions. The kinetic pattern of the disappearance of OA and the appearance of ochratoxin alpha (Oalpha) was principally the same with four diets fed to donor animals and with three donor animals. The concentration of OA declined to a very low or non-detectable level with half-lives at 0.17-1.84 h; its rate of disappearance was first order throughout. The concentration of Oalpha increased to a constant level under all conditions examined. The average amounts of Oalpha formed relative to the disappearance of OA were not different (p > 0.05) and ranged between 94 +/- 8 and 98 +/- 8 %. The rate of disappearance of OA differed (p < 0.001) between diets and animals. It was accelerated by increasing the content of concentrate in the diet. Ochratoxin alpha was not metabolized when added in pure form to buffered rumen fluid and incubated under the same conditions as OA. Pure OA was also added to buffered fluids from the forestomach and abomasum of a slaughtered cow. It disappeared exponentially in a mixture of fluids from the rumen and reticulum and in fluid from the omasum, with the average amounts of Oalpha formed relative to OA disappearance being 107 and 109 %, respectively. Ochratoxin A also was not metabolized in fluid from the abomasum. These studies demonstrate that OA is hydrolysed in the rumen via first order kinetics, diet and animal affect the rate of hydrolysis, OA is quantitatively converted to Oalpha and Oalpha is not degraded.  相似文献

In tropical regions, protein supplementation is a common practice in dairy and beef farming. However, the effect of highly degradable protein in ruminal fermentation and microbial community composition has not yet been investigated in a systematic manner. In this work, we aimed to investigate the impact of casein supplementation on volatile fatty acids (VFA) production, specific activity of deamination (SAD), ammonia concentration and bacterial and archaeal community composition. The experimental design was a 4×4 Latin square balanced for residual effects, with four animals (average initial weight of 280±10 kg) and four experimental periods, each with duration of 29 days. The diet comprised Tifton 85 (Cynodon sp.) hay with an average CP content of 9.8%, on a dry matter basis. Animals received basal forage (control) or infusions of pure casein (230 g) administered direct into the rumen, abomasum or divided (50 : 50 ratio) in the rumen/abomasum. There was no differences (P>0.05) in ruminal pH and microbial protein concentration between supplemented v. non-supplemented animals. However, in steers receiving ruminal infusion of casein the SAD and ruminal ammonia concentration increased 33% and 76%, respectively, compared with the control. The total concentration of VFA increased (P<0.05) in steers receiving rumen infusion of casein. SAD and the microbial protein concentration did not vary significantly among treatments during the feeding cycle, but mean SAD values were greater in steers supplemented in the rumen and rumen/abomasum. Ruminal ammonia concentration was positively correlated with SAD in animals receiving ruminal infusion of casein. Polymerase chain reaction–denaturing gradient gel electrophoresis (PCR-DGGE) analysis revealed low similarity between treatments, animals and time of sample collection. Richness analysis and determination of the Shannon–Wiener index indicated no differences (P>0.05) in species richness and diversity of γ-proteobacteria, firmicutes and archaea between non-supplemented Nellore steers and steers receiving casein supplementation in the rumen. However, species richness and the Shannon–Wiener index were lower (P<0.05) for the phylum bacteroidetes in steers supplemented with casein in the rumen compared with non-supplemented animals. Venn diagrams indicated that the number of unique bands varied considerably among individual animals and was usually higher in number for non-supplemented steers compared with supplemented animals. These results add new knowledge about the effects of ruminal and postruminal protein supplementation on metabolic activities of rumen microbes and the composition of bacterial and archaeal communities in the rumen of steers.  相似文献

为分析不同饲料喂养对黄牛胃中微生物群落结构的影响,分别用芒草单一喂养和农家混合饲料喂养两年的黄牛为实验组和对照组。以瘤胃、蜂巢胃、重瓣胃和皱胃四个胃中的微生物为研究对象,采用原位包埋裂解法和脉冲场电泳(pulsed field gel electrophoresis,PFGE)提取微生物总 DNA,脉冲场电泳(pulsed field gel elec-trophoresis,PFGE)。使用细菌16S rRNA 引物341F /534R 及真菌18S rDNA 引物 NS1/GCFungi 进行降落 PCR 扩增。变性梯度凝胶电泳(denaturing gradient gel electrophoresis,DGGE)对扩增产物进行区分,使用硝酸银染色。电泳扫描结果通过 Quantity one 软件进行分析,SPSS 软件进行数据统计。针对实验组和对照组瘤胃样品共性条带和特性条带测序并比对。结果表明:实验组与对照组细菌群落结构变化较大,UPGAM聚类图上被分为两支,相似性只有0.35。且香农多样性指数及条带丰度都明显少于对照组。真菌 DGGE 图谱条带差别不大,聚类图上显示实验组四个样品与对照组四个样品相似性在0.43~0.68之间。多样性及条带丰度在实验组与对照组之间差异0.0027~0.5999。测序结果,细菌与数据库中未培养细菌种类较为接近,而真菌中部分与已知菌种接近。芒草单一饲养对牛胃中的微生物群落结构具有极大的影响,对细菌的影响尤为明显。  相似文献

The effects of mild heat exposure (30 degrees C; 30% relative humidity), and of the suppression of prolactin secretion under such conditions, were studied in anoestrous ewes given daily 791 g dry matter of a pelleted mixture of lucerne hay and oat grain (3:2). 51Cr-EDTA, 103Ru-phenanthroline and lignin were used to determine mean retention times (MRT) in the gastro-intestinal (GI) tract by a continuous infusion-total sampling procedure. Mild heat exposure reduced the digesta-free tissue weight of all GI tract segments distal to the omasum. Increases in the amounts of digesta in the stomach compartments were largely due to increases in water content, although the solids content of the abomasum (P less than 0.10) and omasum also increased. The treatment caused an increase in water intake (P less than 0.10), increased the MRT of 51Cr-EDTA in the reticulo-rumen (P less than 0.05), and tended to increase the MRT of all three markers in the omasum and abomasum. However, MRT in the whole GI tract was unaffected because of a compensating decrease (P less than 0.01) in digesta MRT in the distal large intestine. Suppression of prolactin secretion with 2-bromo-alpha-ergocryptine impaired the ewes' ability to maintain their body temperature under the warm conditions imposed. This was associated with a consistent reduction in the degree to which water intake and its transactions in the GI tract changed in response to mild heat exposure. The treatment also caused decreases in omasal MRT and increases in abomasal MRT. Increased plasma concentrations of somatostatin and gastrin were associated with reduced plasma concentrations of prolactin, and it was postulated that some of the effects of prolactin on the GI tract may be mediated via somatostatin.  相似文献

Urease activity, expressed as mg N-NH3/g dry weight per 30 min at 25 degrees C, was determined in the various parts of the sheep, chicken and pig digestive apparatus. The results were as follows. Sheep: contents--rumen 1.25"/-0.09, reticulum 0.78+/-0.02, omasum 0.44+/-0.02, abomasum 0.002+/-0.001, duodenum 0.003+/-0.001, jejunum 0.18+/-0.03, ileum 0.42+/-0.03, caecum 1.34+/-0.11, colon 0.76+/-0.08, walls-rumen 0.88+/-0.16, reticulum 0.38+/-0.04, omasum 0.11+/-0.02, abomasum 0.01+/-0.002, ileum 0.092+/-0.01, caecum 0.14+/-0.03, colon 0.16+/-0.02. Chicken: contents--jejunum 0.028+/-0.009, ileum 0.043+/-0.013, caecum 0.17+/-0.03, colon and cloaca 0.04+/-0.013. Pigs: contents--jejunum 0.02+/-0.01, ileum 0.14+/-0.08, caecum 0.62+-0.12, colon 0.43+/-0.06. No urease activity was found in the walls of the digestive apparatus or the contents of the duodenum in chickens, or in the walls of the stomach and intestine and the contents of the duodenum in pigs. The results show that urease activity in the digestive apparatus of pigs and poultry is lower than in sheep. Inadequate urease activity in the digestive apparatus explains why chickens and pigs are significantly less capable than ruminants of utilizing urea nitrogen as a substitute for some of the protein in the diet.  相似文献

Male Kazak sheep and Xinjiang fine wool sheep, six for each different age group (days 2, 30, 60, 90 and 120), were used in the present study to investigate the tissue distribution and developmental changes of ghrelin mRNA expression in abomasum; however, there was no 120-day-old Kazak sheep. After measurement of body weight, the tissues such as hypothalamus, pituitary, heart, liver, rumen, reticulum, omasum, abomasum, duodenum, and longissimus dorsi muscle were sampled. And the total RNA of different tissues was extracted to determine the abundance of ghrelin mRNA by RT-PCR and real-time PCR. The results showed that (1) for both breeds, body weight among different ages was significantly different (P<0.05). And from day 30 to 90, the body weight of Kazak was significantly higher than that of Xinjiang (P<0.01); (2) Ghrelin mRNA existed in all the above tissues and was significantly higher in the abomasum than in other tissues (P<0.05); (3) the temporal patterns of abomasum ghrelin mRNA expression in Kazak and Xinjiang were similar. From day 2 to 60 in Kazak and 2 to 90 in Xinjiang, there was a steady increase in the ghrelin mRNA level. By day 60 in Kazak and day 90 in Xinjiang, the level reached a plateau and remained steady. These results also demonstrated that from birth to day 90, ghrelin mRNA level was significantly higher in Kazak than in Xinjiang (P<0.01).  相似文献

选取2、30、60、90和120日龄的雄性哈萨克羊和新疆细毛羊各6只(无120日龄的哈萨克羊),测体重后屠宰,采下丘脑、垂体、心脏、肝脏、瘤胃、网胃、瓣胃、皱胃、十二指肠、背最长肌,用RT-PCR和荧光实时定量PCR法检测ghrelin基因表达的组织分布,及其在皱胃中的发育性变化。研究结果表明:(1)品种内各生长时期的体重差异显著(P〈0.05)。雄性哈萨克羊和新疆细毛羊的体重在2日龄时无显著差异(P〉0.05),30~90日龄间,前者的体重极显著高于后者(P〈0.01);(2)所检测的各组织中都有ghrelin mRNA分布,但主要在皱胃中表达,其表达量远高于其他组织(P<0.05);(3)两品种绵羊皱胃ghrelin基因表达的发育性变化模式基本相似,都随着日龄的增加而呈上升趋势,其中雄性哈萨克羊的表达量在2~60日龄间持续上升,60日龄后趋于水平;雄性新疆细毛羊的表达量在2~90日龄间持续上升,90日龄后趋于水平。研究还发现雄性哈萨克羊皱胃胂relin基因的表达量在2~90日龄间极显著高于新疆细毛羊(P<0.01)。  相似文献

Rhodanese (thiosulfate: cyanide sulfurtransferase, EC. 2.8.1.1) is a ubiquitous enzyme present in all living organisms, from bacteria to humans and plays a central role in cyanide detoxification. The purpose of this investigation is to determine and compare rhodanese activity in different tissues of adult male and female goats (Capra hircus). The results showed that the specific activity of rhodanese in different tissues was significantly different (P<0.05). The highest activity of rhodanese was in epithelium of rumen, followed by epithelia of reticulum and omasum and liver. No significant difference was observed when tissues of male and female goats were compared. The lowest specific activity of rhodanese was observed in spleen, urinary bladder, lymph node, ovary, skeletal muscle and pyloric muscle of abomasum. The results of this study may indicate the involvement of rhodanese in cyanide detoxification in goat tissues that have greater potential to be exposed to higher levels of cyanide.  相似文献

Glutamate dehydrogenase (GLDH, EC 1.4.1.3) and glutamine synthetase (GS, EC 6.3.1.2) activity were determined in the contents and tissues of the various parts of the sheep and chicken digestive tract, GLDH activity in the tissues of the sheep omasum, duodenum, rumen, reticulum, colon, caecum, jejunum and ileum ranged from 3.25+/-0.7 U (mumol/g dry weight . min) to 5.94+/-2.28 U; in the abomasum it was 9.67+/-1.27 U. GLDH activity in the contents of the ileum, abomasum, jejunum and duodenum varied from 0.85+/-0.19 U to 3.29+/-0.53 U and in the colon, caecum, reticulum, omasum and rumen from 6.34+/-2.64 U to 16.96+/-3.83 U. GS activity in the tissues of these parts of the digestive tract varied from 2.8+/-0.59 U to 8.6+/-1.4 U and their contents from 2.49+/-0.85 U to 10.76+/-2 U. GS activity in the contents of the colon was very low (0.26+/-0.07 U). In the tissues of the chicken duodenum, caecum, jejunum and ileum we found GLDH activity of 4.68+/-1.64 U to 7.96+/-1.73 U; in their contents it was 3.31+/-1.06 U to 3.8+/-0.73, but in the caecum it attained up to 66.7+/-24.3 U. GS activity was high from 57.6+/-2.0 U to 231+/-84 U in the tissues and 357+/-53 U to 383+/-76 U in the contents (in the caecum up to 2,500+/-233 U). The results show that conditions for the utilization of ammonia are present in the tissues and the contents in the whole of the sheep and chicken digestive apparatus. The hypothesis is confirmed that the different ability of ruminants and fowls to utilize ammonia formed from urea added to their feed, including ammonia formed by hydrolysis of blood urea, is due to the different GLDH and GS activity in their digestive tract as well as in their liver.  相似文献

Non-keratinized, stratified squamous epithelium lined the mucosa of the fore-stomach. The mucosal appendages in the form of ruminal papillae did not develop in the rumen during this study, however, omasal laminae and reticular ribs were evident in groups I and II, respectively. Cytoplasmic accumulations were evident in the middle layer of the epithelium, which is characteristic of this region. The thickness of the epithelium increased constantly in the rumen and reticulum, but in the omasum it remained constant with the increase in the size of the fetus. The corial papillae in the rumen, reticulum and omasum appeared at the 12.7-, 11.5-and 14.6-cm CVR stage, respectively. The muscularis mucosae appeared in the omasum at the 14.6-cm CVR stage in the first-order laminae. The tunica muscularis consisted of spirally arranged muscle fibre bundles. Cross-cut muscle fibre bundles below the tip of the omasal laminae descended downward with the increase in the size of the fetus and continued with the inner circular layer. Acid and alkaline phosphatases and fat could not be observed. The concentration of glycogen in the epithelium was maximum in group III and minimum in group II.  相似文献

Isovalerate supplements could stimulate rumen development by improving morphology and function of rumen mucosa, and then promote the growth of calves. This study was done to evaluate the effects of isovalerate supplements on morphology and functional gene expression of rumen mucosa in dairy calves. In total, 48 Chinese Holstein male calves with 15 days of age and 45.1±0.36 kg of BW were randomly assigned to four groups. The treatments were: control, low-isovalerate, moderate-isovalerate and high-isovalerate with 0, 3, 6 and 9 g isovalerate per calf per day, respectively. Supplementary isovalerate was hand-mixed into milk in pre-weaning calves and into concentrate portion in post-weaning calves. The study consisted of a 15-day-adaptation period and a 60-day-sampling period. Calves were weaned at 60 days of age. Three calves were slaughtered from each of the four treatments at 30, 60 and 90 days of age. The weight of body and stomach were measured, samples of ruminal tissues and blood were analyzed. Total stomach weight, total stomach to BW ratio, rumen wall and keratinized layer thickness, serum growth hormone and IGF-1 for both pre- and post-weaning calves increased linearly with increasing isovalerate supplements. Rumen to total stomach weight ratio, the length and width of rumen papillae, and serum β-hydroxybutyrate increased linearly for post-weaning calves. However, abomasum weight to total stomach weight ratio decreased linearly for both pre- and post-weaning calves. The relative messenger RNA expression for growth hormone receptor, IGF-1 receptor and 3-hydroxy-3-methylglutaryl-CoA synthase 1 in rumen mucosa increased linearly for post-weaning calves. Our results suggested that isovalerate supplements promoted rumen development in a dose-dependent manner. The optimum dose was 6.0 g isovalerate per calf per day.  相似文献

We assessed sexual variation in food quality and gut macrostructure in adult male and pregnant female sika deer, Cervus nippon (Temminck, 1838), in Japan during winter. These variations might have important implications relative to sexual differences in habitat use, forage acquisition, and digestive strategy. According to the sexual dimorphism-body size hypothesis the larger males would feed on poorer forage and have heavier stomach contents and heavier intestine contents and longer intestines than smaller females. However, the food quality in rumen contents of males was higher than, or at least similar with, that of pregnant females. In correspondence to food quality, the relative weights of stomach contents and intestines with contents, the relative lengths of intestines to the lengths of body and total intestines in pregnant females were similar to adult males. The relative weights of omasum and abomasum tissues in pregnant females were greater than in males. Our findings suggest sexual differences in feeding strategy in sika deer in Japan during winter. To meet greater nutritional demands of high metabolic rate and gestation, pregnant females seemed to maintain a greater volume of digesta in guts and had more stomach tissues than expected by the sexual dimorphism-body size hypothesis to compensate for poorer forage quality.  相似文献

Twelve bulls of approximately 140 kg bodyweight were each fitted with simple cannulae in the rumen and abomasum and a re-entrant cannula in the terminal ileum. They were kept in individual pens and given daily 1.8 kg of a basal diet containing dried maize meal (whole plant), dried sugarbeet pulp and a urea/mineral preparation in the proportions 0.80, 0.18 and 0.02. Four animals received this diet alone, four received in addition 198 g daily (11 g N) of rapeseed meal by infusion into the abomasum and the remaining four the basal diet plus the same amount of formaldehyde-treated rapeseed also by infusion.The concentration of ammonia-N in the rumen fluid of animals given the untreated rapeseed meal was higher (P < 0.05) than for those on the other two treatments. The pH of the rumen and abomasal fluid was similar on all treatments. Coefficients for the apparent disappearance in the small intestine of the dry matter and nitrogen of untreated rapeseed meal infused into the abomasum were 0.76 and 0.75, respectively. In contrast formaldehyde-treated rapeseed meal was poorly digested. It is suggested that procedures for the treatment of rapeseed meal that will minimise the degradability of its protein in the rumen, yet prevent depression of its digestion and absorption in the abomasum and small intestine have still to be defined.  相似文献

Isobutyrate supplements could improve rumen development by increasing ruminal fermentation products, especially butyrate, and then promote the growth performance of calves. The objective of this study was to evaluate the effects of isobutyrate supplementation on growth performance, rumen development, blood metabolites and hormone secretion in pre- and post-weaned dairy calves. In total, 56 Chinese Holstein male calves with 30 days of age and 72.9±1.43 kg of BW, blocked by days of age and BW, were assigned to four groups in a randomized block design. The treatments were as follows: control, low-isobutyrate, moderate-isobutyrate and high-isobutyrate with 0, 0.03, 0.06 and 0.09 g isobutyrate/kg BW per calf per day, respectively. Supplemental isobutyrate was hand-mixed into milk of pre-weaned calves and the concentrate portion of post-weaned calves. The study consisted of 10 days of an adaptation period and a 50-day sampling period. Calves were weaned at 60 days of age. Seven calves were chosen from each treatment at random and slaughtered at 45 and 90 days of age. BW, dry matter (DM) intake and stomach weight were measured, samples of ruminal tissues and blood were determined. For pre- and post-weaned calves, DM intake and average daily gain increased linearly (P<0.05), but feed conversion ratio decreased linearly (P<0.05) with increasing isobutyrate supplementation. Total stomach weight and the ratio of rumen weight to total stomach weight tended to increase (P=0.073) for pre-weaned calves and increased linearly (P=0.021) for post-weaned calves, whereas the ratio of abomasum weight to total stomach weight was not affected for pre-weaned calves and decreased linearly (P<0.05) for post-weaned calves with increasing isobutyrate supplementation. Both length and width of rumen papillae tended to increase linearly for pre-weaned calves, but increased linearly (P<0.05) for post-weaned calves with increasing isobutyrate supplementation. The relative expression of messenger RNA for growth hormone (GH) receptor and 3-hydroxy-3-methylglutaryl-CoA synthase 1 in rumen mucosa increased linearly (P<0.05) for pre- and post-weaned calves with increasing isobutyrate supplementation. Blood concentrations of glucose, acetoacetate, β-hydroxybutyrate, GH and IGF-1 increased linearly (P<0.05) for pre- and post-weaned calves, whereas blood concentration of insulin decreased linearly with increasing isobutyrate supplementation. The present results indicated that isobutyrate promoted growth of calves by improving rumen development and its ketogenesis in a dose-dependent manner.  相似文献

The effect of a 5-day continuous intravenous infusion of somatostatin (4.6 ng min-1 kg-1) was studied, using anoestrous ewes given 791 g dry matter per day of a 60:40 lucerne hay:oat grain pelleted diet from a continuously moving belt. 51Cr-EDTA, 103Ru-phenanthroline and lignin were used as markers to determine digesta mean retention times (MRT) by a continuous infusion-total sampling procedure. The somatostatin infusion increased the concentration of somatostatin in venous plasma within the physiological range from 10 to 76 ng/l, decreased plasma concentrations of prolactin and thyroxine, but had no effect upon plasma concentrations of insulin and glucagon. It had no effect upon digesta-free weight of the rumen and omasum but consistently decreased the weight of all post-ruminal segments of the gastrointestinal (GI) tract. The infusion increased the accumulation of digesta in the abomasum and caecum. Total MRT of all three markers in the entire GI tract was unaffected by somatostatin infusion, but the proportion of total MRT spent in the abomasum + small intestine + caecum increased and the proportion spent in the large intestine and rumen decreased. Somatostatin infusion decreased apparent endogenous abomasal secretion, increased water flow from the rumen and into the abomasum and decreased voluntary water consumption. It is proposed that the prime site of somatostatin action was in the abomasal to caecal region, where somatostatin-secreting D cells are found in greatest concentration, that effects observed in the large intestine and rumen may represent secondary compensatory mechanisms and that the effects observed were due to direct action of somatostatin and were not mediated by other GI hormones.  相似文献

In 3 sheep with subcutaneously exposed vagus nerves and with chronic large fistula of the rumen pharmacological analysis of the motor activity of omasum was carried out after blockade of vagus nerves. The contractions of omasal canal, reticulum and rumen were recorded by the baloon method. After blockade of the vagus nerves with anesthetics the proper pharmacological agents were administered in intravenous injections. In the light of the obtained results it was found that contractions of the omsal canal, persisting after vagal blockade, are independent of the influence of the autonomic system and they are probably due to myogenic automatism. Pharmacological studies of abomasum motor activity were carried out in 3 sheep with chronic fistulae in the rumen and the fundus and pyloric part of the abomasum. The contractions of these parts of the stomach were recorded by the balloon method. The obtained results showed that in the abomasum stimulation of alpha-adrenergic receptors raises the degree of motor activity while stimulation of beta-adrenergic receptors inhibits it. It has been shown also that the sympathetic system has a greater influence through the beta-adrenergic receptors on the motor activity of the pyloric part than on that of the fundus of the abomasum. The analysis of the records obtained after bilateral vagotomy demonstrated that the automatism of the pyloric part was greater than that of the fundus of the abomasum.  相似文献
