The spacing of lateral root primordia in the primary root of Pisum sativum (cv. Alaska) seedlings is influenced by both predetermined lateral root initiation sites in the embryonic radicle and by factors present during seedling growth. When pea seeds were germinated in the presence of the mitotic inhibitor, colchicine, the triarch radicle produced three ranks of primordiomorphs indicating sites of embryonic lateral root primordia. The number of primordiomorphs was not the same along the three xylem strands in the radicle. Normally germinated seedling roots (5 days old) also showed a different number of lateral root primordia associated with the three strands. In both cases, the strand with the greatest number of primordia (or primordiomorphs) was associated with a cotyledonary trace. This indicated a possible role for the cotyledons in setting the pattern of lateral root distribution during radicle development. The spacing of lateral root primordia could be altered by the application of growth regulators. Seedling root tips (2 mm) were removed (? rt) and replaced with indoleacetic acid (+IAA), and in some instances seedlings were also treated with the auxin transport inhibitor, 3,3a-dihydro-2-(p-methoxyphenyl)-8H-pyrazolo[5, 1-α]isoindol-8-one (+DPX). In the growth regulator treatments, primary root elongation was inhibited, a greater number of lateral root primordia were initiated compared to controls, and the spacing intervals between primordia were greatly reduced. The — rt, +IAA, +DPX-treatment resulted in the closest possible spacing intervals (av. 0.4 ? 0.6 mm), but resulted in fused or fasciated laterals. The — rt, + IAA-treatment produced the shortest spacing intervals which resulted in “normal” lateral roots (0.8 ? 1.1 mm).  相似文献

Anatomical changes in the radicle and shoot meristems of embryos of germinating seeds of the obligate root parasites, Alectra vogelii and Striga gesnerioides were studied. When germination of seeds was stimulated by cowpea (Vigna unguiculata) root exudate, growth occurred mainly in the radicular pole of embryos and minimally in the plumular pole, resulting in seedlings with elongated radicles. Maximum radicle elongations of about 3 mm in A. vogelii and 2 mm in S. gesnerioides were recorded during a period of 8 and 11 days, respectively. Analysis of the radicular tip during the course of seed germination revealed that the activity of the meristematic tissue progressively decreased until it completely disappeared. When germinated seeds were cultured on nutrient agar media, the radicle meristem of A. vogelii continued to grow producing a normal root with a root cap. On the other hand, the radicles of cultured S. gesnerioides seeds elongated only slightly before meristematic activity ceased. During continued growth of seedlings of both species on agar media, lateral roots whose tips had typical angiosperm root topography, were initiated from the radicle.  相似文献

Progressive development in cotton root morphology of resistant A623 and susceptible M-8 cotton (Gossypium hirsutum L.) lines following infection by the root-knot nematode Meloidogyne incognita was studied in glass front boxes. Symptom development and radicle growth were observed; degree of galling, gall and egg mass diameter, and number of eggs per egg mass were recorded; and root segments were examined histologically. Small cracks caused by M. incognita appeared in the root epidermis and cortex soon after the cotyledons expanded on day 4. The cracks were longer and wider and extended through the cortex when the first true leaf became visible at day 8. Galls had formed on taproots by this time. When exposed to M. incognita, A623 had faster radicle growth (22%), fewer and smaller cracks in the root epidermis and cortex, fewer and smaller root galls, one-twelfth as many egg masses, and one-fourth as many eggs per egg mass as M-8. Root cracking, galling, and giant cell formation are major effects of M. incognita that may predispose cotton roots to pathogens resulting in synergistic interactions and diseases.  相似文献

Soil sickness is often observed in cucurbit crops such as Citrullus lanatus, Cucumis melo and Cucumis sativus, but not in cucurbit crops such as Cucurbita moschata, Lagenaria leucantha and Luffa cylindrica. Results showed that root aqueous extracts of Citrullus lanatus, Cucumis melo and Cucumis sativus were autotoxic, but those of Cucurbita moschata, Momordica charantia and Luffa cylindrica were less autotoxic to the radicle elongation of respective species. Plant growth of Citrullus lanatus, Cucumis melo and Cucumis sativus were greatly inhibited by autotoxic substances released from powered root tissue at a rate of 1 g per seedling. Root exudates of Citrullus lanatus, Cucumis melo and Cucumis sativus were autotoxic to radicle elongation and seedling growth of respective species. However, root exudates of Citrullus lanatus did not inhibit radicle elongation of Cucurbita ficifolia, which is commonly used as rootstock for the grafting of Citrullus lanatus, Cucumis melo and Cucumis sativus to decrease soil-borne diseases in commercial production. It seems possible to overcome autotoxicity in cucurbit crops by grafting on Cucurbita ficifolia. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

The developmental anatomy of the primary root of Ipomoea purpurea was studied at several growth stages, beginning with the radicle. The radicle is generally composed of three superimposed tiers of initials, which produce the vascular cylinder, cortex, and columella; and a peripheral band of lateral rootcap-epidermal initials. The radicular cortex contains 16–19 immature laticifers; none of the tissue regions in the radicle contains mature cells. Following germination and during the first 2–3 cm growth of the primary root the apical meristem and its derivative tissues undergo a series of modifications. Root apical diameter decreases as cells in lateral portions of the rootcap elongate; meanwhile, the columella enlarges vertically. The relationship between cortical and columellar initials changes as fewer mitoses occur in the former while the latter remain active. In longer roots the columellar initials are directly in contact with the vascular initials. Cortical size diminishes during early root growth as cortical laticifers and their associated cells cease to be produced by the outer cortical initials and ground meristem. Early procambium, at the level of vascular pattern initiation, decreases in diameter by cellular reorientation, and the vascular cylinder decreases in overall diameter although the tetrarch pattern remains unchanged.  相似文献

Wild oat (Avena fatua L.) caryopses were germinated on moist filter paper and under water in the presence and absence of hydrogen peroxide (H2O2). The sequential growth and development of embryo parts were studied. Germination, as indicated by radicle emergence, was least and slowest in caryopses submerged in deoxygenated water. The coleorhiza in such caryopses elongated much earlier than the root, in contrast to the other treatments where the coleorhiza and the root emerged at about the same time. In caryopses incubated on moist filter paper all embryo parts showed considerable growth. In H2O2 treated caryopses only the epicotyl showed substantial growth over the experimental period. In all treatments the first mitotic peaks were noticed at the same period. The occurrence of these early nuclear divisions may be due to release of 4 C nuclei from inhibition by the uptake of water during caryopsis imbibition. The mitosis continued in the radicle of the embryo in those caryopses germinating on moist filter paper, indicating occurrence of DNA synthesis. In the other two treatments, however, few divisions were detected. Here the early growth of the root, causing caryopsis germination, was due to cell elongation, especially in the proximal part of the root.  相似文献

Laboratory, greenhouse and field studies were conducted to determine the allelopathic potential ofMikania micrantha H.B.K. on the germination and growth of three weeds, namelyAsystasia intrusa Bl.,Chrysopogon aciculatus (Retz.) Trin. andPaspalum conjugatum Berg. The plant height of the test species decreased with increasing amounts of debris when either leaf or root debris of Mikania was present on the soil surface or incorporated into the soil. In general, incorporated debris caused greater reduction in height and seedling fresh weight than debris placed on the soil surface. Paspalum was more sensitive to either root or leaf of Mikania when incorporated into soil than on the soil surface. Leachate of Mikania leaf caused a significant reduction in radicle length and fresh weight of the seedlings of the test species. However, only Paspalum seeds showed significant decrease in germination when exposed to leaf leachate. Full-strength extract of either leaf or root caused a significant decrease in both germination and fresh weight of the test species. At the highest concentration, root extract of Mikania caused a marked reduction of radicle elongation of Paspalum.  相似文献

Summary We investigated several ecological correlates of seed mass variation in the hemiparasitic, xylemtapping mistletoe, Phoradendron juniperinum. Mean seed mass varied two-fold among plants between the ages of 4 and 14 years old and was positively correlated with parental plant age. Both the standard deviation and the coefficient of variation in mean seed mass decreased with increasing plant age demonstrating that, on average, younger plants produced seed with more variable mass. Nitrogen concentrations (mg nitrogen per gram of seed) of both the seed and fruit (pericarp) were not correlated with mass or the age of the parent plant from which the seed was taken. However, the nitrogen content per seed (mg nitrogen per seed) was positively correlated with the mean seed dry mass and the age of the seed parent, suggesting that the carbon to nitrogen ratio of individual seeds remained relatively constant as seed mass increased and plants grew older. Seed germination ranged between 20% and 86% and was positively correlated with mass and parental plant age. Heavier seeds (seeds from older plants) also had the highest root radicle growth rates. Furthermore, the final root radicle length after 76 d of growth was positively correlated with seed dry mass. When grown on a medium containing an extract prepared from the host plant foliage, all seeds showed lower germination, grew more slowly and had shorter overall root radicles, but had significantly greater development of the haustorial disks (the holdfast which forms the host-parasite junction in Phoradendron) than seeds grown on a control medium. Our results suggest that, on average, seeds of greater mass produced by older plants have a greater total resource pool per propagule (fruit + seed). This resource pool may be important in conferring a greater potential for dispersal (fruit), survival, colonization, and establishment.  相似文献

DNA synthetic activity in the radicle meristem of embryos of germinating seeds of the obligate root parasites, Alectra vogelii and Striga gesnerioides was followed by autoradiography of 3H-thymidine incorporation. Incorporation of 3H-thymidine occurred in the nuclei of cells destined to form the vascular tissues, ground meristem and epidermis. An analysis of the distribution of labeled nuclei demonstrated the presence of a quiescent center of 2-4 cells in the radicle at the beginning of seed germination, becoming more prominent at later stages of germination. During continued growth of the radicle which resulted in a reduction in size of the meristem, cells of the original quiescent center were activated to undergo DNA synthesis.  相似文献

The seedling development of an undescribed Malaccotristicha species was observed by using seedling culture and microtomy to infer the evolution of body plan with a focus on the root, which is a developmentally leading organ of most Podostemaceae. The young seedling has a small primary shoot apical meristem and a primary root apical meristem. The shoot meristem develops into a plumular ramulus, and the root meristem, into a cylindrical radicle with no root cap. The radicle transforms to a dorsiventral, flattened, capped primary root. An adventitious root develops endogenously on the lateral side of the hypocotyl and is similar to the primary root. This is a new pattern in Podostemaceae. Comparison of this and described patterns of Podostemaceae (and the sister-group Hypericaceae) suggests that the radicle was lost in the early evolution of Podostemaceae and instead adventitious roots replaced it as a leading organ.  相似文献

Aims: Previously, we selected a bacterial strain (GSE09) antagonistic to Phytophthora capsici on pepper, which produced a volatile compound (2,4‐di‐tert‐butylphenol), inhibiting the pathogen. In this study, we identified strain GSE09 and characterized some of the biological traits of this strain in relation to its antagonistic properties against P. capsici. In addition, we examined bacterial colonization on the root surface or in rhizosphere soil and the effect of various concentrations of the volatile compound and strain GSE09 on pathogen development and radicle infection as well as radicle growth. Methods and Results: Strain GSE09 was identified as Flavobacterium johnsoniae, which forms biofilms and produces indolic compounds and biosurfactant but not hydrogen cyanide (HCN) with little or low levels of antifungal activity and swimming and swarming activities. Fl. johnsoniae GSE09 effectively colonized on pepper root, rhizosphere, and bulk (pot) soil, which reduced the pathogen colonization in the roots and disease severity in the plants. Various concentrations of 2,4‐di‐tert‐butylphenol or strain GSE09 inhibited pathogen development (mycelial growth, sporulation, and zoospore germination) in I‐plate (a plastic plate containing a center partition). In addition, germinated seeds treated with the compound (1–100 μg ml?1) or the strain (102–1010 cells ml?1) significantly reduced radicle infection by P. capsici without radicle growth inhibition. Conclusions: These results indicate that colonization of pepper root and rhizosphere by the Fl. johnsoniae strain GSE09, which can form biofilms and produce indolic compounds, biosurfactant, and 2,4‐di‐tert‐butylphenol, might provide effective biocontrol activity against P. capsici. Significance and Impact of the Study: To our knowledge, this is the first study demonstrating that the Fl. johnsoniae strain GSE09, as a potential biocontrol agent, can effectively protect pepper plants against P. capsici infection by colonizing the roots.  相似文献

The aim of this work was to examine whether seed ecophysiological traits in three closely related Crocus species were associated with ecological niche differentiation and species divergence. Seeds of the temperate tetraploid cytotype of Crocus neapolitanus, the sub‐Mediterranean C. etruscus and the Mediterranean C. ilvensis were placed either on agar in the laboratory under different periods of simulated seasonal conditions or in nylon mesh bags buried outdoors to examine embryo growth, radicle and shoot emergence. In agreement with the phenology observed outdoors, in the laboratory embryos required a cool temperature (ca. 10 °C) to grow to full size (embryo length:seed length, E:S ratio ca. 0.75) but only after seeds received a warm stratification; radicle emergence then followed immediately (November). Shoot emergence is a temporally separated phase (March) that was promoted by cold stratification in C. neapolitanus while in the other two species this time lag was attributed to a slow continuous developmental process. These species have similar embryo growth and radicle phenology but differ in their degree of epicotyl dormancy, which is related to the length of local winter. Conclusions from laboratory experiments that only consider root emergence could be misleading; evaluating the phenology of both root and shoot emergence should be considered in order to demonstrate ecologically meaningful differences in germination behaviour and to develop effective propagation protocols. Although these taxa resulted from recent speciation processes, the outcomes suggest an early onset of adaptation to local ecological factors and that phylogeny may represent a significant constraint in the evolution and expression of seed traits in Crocus.  相似文献

Deschampsia flexuosa (L.) Trin. is an abundant grass species in the ground flora of acidic beech forests in southern Sweden. Generally, the species is restricted to a limited soil pH range (pH 4–5). The main objective was to study the influence of different soil acidities on germination, initial root development and on the growth of the species. The experiments were carried out under controlled conditions and designed to simulate the physico-chemical conditions present in the field. By using forest soils within the pH range 4.0 to 8.3 and artificial variation in pH (3.2 to 7.6) of soil-water extracts, it was possible to evaluate the influence of soil reaction and the H+ per se. In all experiments seeds have been used. Germination was significantly delayed in the very acid soil (pH 4.0) in comparison to the germination in soils within the pH range (4.4 to 6.4). Soil substances, other than the H+, might be responsible for this delay in germination, whereas development of the radicle was markedly affected by increasing H+ concentrations. Especially the development of root hairs was sensitive to H+ and was significantly reduced at a pH<-3.8. By increasing soil acidity the injury symptoms, including curling and discolouring, became more intense and at the highest acidity (pH 3.2) the radicles appeared brown, stunted and the root hairs were lacking. Most favourable growth was obtained at pH 4.4 and 5.0. Soil pH levels above and below this range limited both shoot and root growth. The results showed very good correspondence with observations made in Beech forest soils in southern Sweden, where the species was growing in soils within the pH range 3.9 to 5.1 with a peak growth at pH 4.3. This study shows that in soils at pH≤3.8, the poor development of the radicle may be crucial in the establishment ofDeschampsia flexuosa. Root hair development was more sensitive to soil acidity than radicle elongation. Germination was delayed in very acid Beech forest soils but other factors than the H-ion per se may be responsible for this delay.  相似文献

The allelopathic competence of tamarind root was evaluated using several weed and edible crop species under both laboratory and greenhouse conditions. Bio-assay guided studies using agar and soil medium revealed that the growth of both radicle and hypocotyl were strongly inhibited under both conditions. Accelerated root exudation observed with an increase in the age of tamarind seedlings caused a high magnitude of growth inhibition of the plant species tested by the plant-box method. Tamarind seedlings at 21-DAG (days after germination) exerted the strongest inhibitory effect (85.0–95.1%) on the growth of the plant species tested. Root dry weight of tamarind seedlings in the plant-box method experiment was highly correlated (R 2 values more than 0.92) with the percentage of growth inhibition. The growth of species grown in the soil under the tamarind tree was inhibited by 85.3–97.1% in the greenhouse. The percentage of growth inhibition declined by 18.4–22.0% (as compared to the natural soil condition) when autoclaved soil of the same trees was used for bio-assay of plant species by the soil-agar sandwich method. This indicates that ca. a 20% increase in response was associated with the allelopathic activity of tamarind root exuded into the natural soil and was due to the effects of soil microbes and soil texture. In terms of growth inhibition of the plant species tested, the root zone soil of the tamarind tree showed stronger inhibitory effects (80.1–94.2%) than the rhizosphere soil, as determined by the soil-agar sandwich method. In all cases, growth inhibition especially in the radicle was higher in the weed species than the edible crop species. Our observations clearly indicate that tamarind root exudate has allelochemical competence and this contributes to a weed free environment around the tamarind tree.  相似文献

Summary In order to determine the effects of concentration on plant growth, aluminium (Al) was extracted (10–3 M CaCl2) from 4 acid brown hill soils which had been treated with superphosphate at rates equivalent to 0 to 300 kg P ha–1. The soils ranged in pH (CaCl2) from 3.5 to 4.9, and Al concentration from 0 to 0.6 mM. The effects of Al on ryegrass growth in the 4 soils in a glasshouse was compared with its effect on radicle elongation of seeds germinated in contact with CaCl2 extracts from the same soils.Ryegrass root growth in the glasshouse, and radicle elongation in the bioassay test were both unaffected by Al concentrations below 0.1 mM. Root growth was substantially reduced when Al concentration exceeded 0.1 mM and above 0.2 mM growth was almost completely inhibited. Radicle elongation rate was also reduced when the concentration of Al was greater than 0.2 mM agreeing well with the observation from the pot experiment.It is concluded that because of its speed and convenience the bioassay method offers a useful method of establishing critical levels of Al for crop plants.  相似文献

Mature seeds of strawberry (Fragaria x ananassa) were placed on Murashige and Skoog medium supplemented with 2.22 μM 6-benzyladenine. After four weeks of culture, and without an intervening callus phase, approximately 36% of the resulting seedling radicles had formed numerous adventitious buds near their tips. A few buds on each radicle developed into shoots, while others formed disorganized calli. Consequently, the seedlings exhibited shoot apices at both ends of the axis of polarity. Our overall results suggest that a considerable level of plasticity in organ determination occurs even in higher plants, and that exogenous growth regulators can cause a root primordium in the radicle to be converted to a shoot primordium.  相似文献

采用形态观察和GMA切片相结合的方法,对龙舌草果实、种子及种苗发育的形态结构进行解剖学观察研究,探讨龙舌草个体发育中表现出的与水生环境相适应的结构特征.结果显示:(1)龙舌草由佛焰苞包被的果实通常由6心皮构成,心皮边缘不完全愈合,层片状胎座,水面形成的幼嫩果实被果柄拉入水中发育成熟;种子具毛,萌发时苗端先于根端发育,在胚根分化之前先出现单细胞的下胚轴毛,随后胚根发育为初生根,并在真叶的节处发育出不定根;果皮和叶肉内的维管束仅由少数细胞构成,且细胞分化不明显,有发达的通气道;叶表皮无角质层,细胞外凸,叶肉细胞数量少;表明龙舌草的结构特征与水生环境相适应.(2)研究还发现,龙舌草的种子无胚乳,下胚轴贮存种苗发育需要的大量淀粉粒,以及初生根和不定根均有根毛的结构,且与泽泻科的慈菇和泽泻的结构一致,证明水鳖科与泽泻科具有较近的亲缘关系.  相似文献

As an initial step in the study of the molecular events surrounding the establishment of a mature root apex, analyses of the early germinative growth of the Allium radicle were undertaken. Changes in the level and pattern of the soluble proteins were investigated by polyacrylamide gel disc electrophoresis and Joyce-Loebl densitometry. Histochemical studies revealed the presence of protein bodies in the terminal mm of the Allium radicle. These bodies underwent depletion and breakdown during germination. The results of the electrophoretic fractionation of the soluble proteins during germination showed that there was not a proliferation of protein components but, rather, a considerable change in the concentration of existing protein fractions. Several fractions, assumed to be components of the reserve or storage proteins, showed a quantitative depletion correlated in time with the loss of the protein bodies. Quantification of the soluble proteins during germination showed a 32% decrease per radicle tip over a 67-hour period, but when expressed on a per cell basis a 32% increase was noted. A dramatic increase in protein synthesis, as measured by 3H-leucine incorporation, was initiated after 36 hours' exposure to germinative conditions, that is, at the beginning of radicle protrusion. A time-ordered sequence of events was noted for the depletion of the protein bodies and their assumed reserve components, suggesting their ultimate utilization in new protein synthesis.  相似文献

Abstract

Allelopathic potential of Thymus pulegioides L. chemical polymorphism was investigated under natural and laboratory conditions. A field analysis of 127 natural habitats hosting chemotypes of T. pulegioides with different ratios of phenolics, geraniol, and ɑ-terpinyl acetate was conducted. Effects of chemotypes, and their main compounds on seed germination and radicle growth of Trifolium pratense L. and Poa pratensis L. were conducted under laboratory conditions. Field analysis showed that Poa species were more plentiful in comparison with Trifolium species, independent of the chemotypical composition of T. pulegioides habitats. Laboratory tests with plant-acceptors showed a stronger inhibitory effect of essential oils on the germination and radicle growth of P. pratensis but in some instances germination was stimulated. Dissimilar effects were observed for the same allelochemical through air and water on the same plant-acceptor. Significantly, different effects of essential oils on radicle growth occurred in T. pratense and P. pratensis: with sensitivity to the phenolic chemotype via air and the ɑ-terpinyl acetate chemotype through water. This demonstrates that chemical polymorphism can expand communication opportunities of T. pulegioides with associated plant species. Combining investigations in natural habitats with laboratory experiments can help understand the effect of chemical polymorphism on plant-plant ecological interactions.  相似文献
