The effects of different gaseous atmospheres were determined on the maximum specific growth rate (mumax) and end-product formation by Brochothrix thermosphacta ATCC 11509T, Lactobacillus viridescens SMRICC 174 and Lactobacillus sp. SMRICC 173 (homofermentative). The highest mumax-values for Lact. viridescens (0.47/h) and Broc. thermosphacta (0.49/h) were obtained in air. Under anaerobic conditions mumax was reduced, an atmosphere containing CO2 alone giving the greatest reduction. Lactobacillus sp. 173 did not grow in air or N2. Aerobic growth was obtained by adding peroxidase while anaerobic growth occurred in the presence of 5-20% CO2. Carbon dioxide alone reduced the growth rate. All test organisms produced mainly lactic acid anaerobically. Lactobacillus viridescens also produced ethanol while Broc. thermosphacta produced small amounts of ethanol and formic acid. With O2 present, the number of end-products increased for all organisms. Lactobacillus sp. 173 produced small amounts of acetic acid and acetoin together with lactic acid. Oxygen induced acetic acid production in Lact. viridescens and Broc. thermosphacta. Aerobically, Broc. thermosphacta also produced a large amount of acetoin and smaller amounts of 2,3-butanediol, iso-valeric acid and iso-butyric acid. The production of lactic acid by Broc. thermosphacta was completely prevented under strictly aerobic conditions. All test organisms consumed O2 during aerobic growth. Hydrogen peroxide was produced by Lact. viridescens and Lactobacillus sp. 173.  相似文献

Lactobacillus viridescens, Lactobacillus sp. strain 173 (homofermentative), and Brochothrix thermosphacta ATCC 11509T were studied at different pH values and temperatures in aerobic and anaerobic batch cultures. The growth rates were higher in aerobic than in anaerobic cultures. L. viridescens grew faster at pH 5.8 than at pH 6.3, whereas the opposite was true for B. thermosphacta. Lactobacillus sp. strain 173 was inhibited in air or at 8 degrees C in anaerobic culture. B. thermosphacta did not grow in anaerobic culture at pH 5.3. The following variations in growth yields were found in the different environments studied: Lactobacillus sp. strain 173, 23 to 25 g (dry weight) per mol of glucose consumed; L. viridescens, 11 to 23 g/mol; B. thermosphacta, 16 to 38 g/mol. In air, L. viridescens produced D-lactic acid, ethanol, and acetic acid, whereas no acetic acid was produced anaerobically. Acetic acid and ethanol together constituted 41 to 48% of the total product yield irrespective of pH and temperature. Lactobacillus sp. strain 173 produced a racemic mixture of D- and L-lactic acid at pH 6.3, whereas the proportion of L-lactic acid was higher than that of D-lactic acid at pH 5.3. In air, product formation of B. thermosphacta varied from a domination of L-lactic acid to increasing yields of acetoin, acetic acid, 2,3-butanediol and isovaleric acid. The effect of pH and temperature on product formation was difficult to separate from the effect of O2 availability in aerobic cultures. Summary The effect of water activity (aw) on the growth and end-product formation of Lactobacillus viridescens SMRICC 174, Lactobacillus SMRICC 173 (homofermentative) and Brochothrix thermosphacta ATCC 11509T was studied. All strains orginated from meat or meat products. The aw was adjusted in the range 0.94–0.99 with NaCl or glycerol. A greater reduction in growth rates was found for L. viridescens and B. thermosphacta when aw was regulated with NaCl rather than with glycerol, the opposite was true for Lactobacillus 173. L. viridescens grew at aw >-0.94. At 0.94 aw B. thermosphacta was totally inhibited when NaCl was the solute and Lactobacillus 173 when glycerol was the solute. Only minor variations in the end-product formation of the Lactobacillus spp. were found at different aw values. In aerobic culture B. thermosphacta produced less l-lactic acid and more acetic acid as the aw was decreased with NaCl, while the yields were unaffected when glycerol was used.  相似文献

An electroporation procedure was developed for the genetic transformation of intact cells of Lactobacillus manihotivorans , a new Lactobacillus species isolated from cassava sour starch fermentation in Colombia. Transformation efficiency of Lact. manihotivorans strains LMG 18010T and LMG 18011 was measured and compared with electroporability of Lact. plantarum strains NCIMB 8299 and LMG 9211, by investigating the effect of changes in various parameters. For Lact. manihotivorans strain LMG 18010T, the composition of the culture medium, such as the type of peptone and the presence of Tween-80, was found to be the most critical parameter, as well as the aeration conditions of the culture; better electroporation was obtained without air. The presence of MgCl2 in the recovery medium was favourable to regeneration of electroporated cells. Plasmid-curing of the cells did not improve their electroporability. Transformants were obtained with Lact. manihotivorans strain LMG 18010T and the plasmids pLZ12 and pGK13, whereas Lact. manihotivorans strain LMG 18011 was transformable with plasmids pLP825 and pLZ12, with different electroporation conditions.  相似文献

Fifteen strains of Brochothrix thermosphacta , isolated from various meat sources, produced the same end-products during aerobic growth in a tryptone-based medium containing 0.2% (w/v) glucose. Growth of one strain at different glucose concentrations showed that 2-methylpropanol and 2,3-butanediol were also produced. Formation of the branched chain acids was favoured by low glucose concentrations, that of the alcohols and the diol by higher concentrations. The traces of the diol, but not those of the alcohols, produced during the early stages of growth at low, limiting concentrations of glucose, gradually disappeared following glucose depletion. These findings provide adequate explanations for the observed differences in end-product formation during growth of Broc. thermosphacta on non-processed and corned beef.  相似文献

Aims:  To determine the effects of carbohydrates on Bacillus cereus ATCC14579T anaerobic metabolism and enterotoxin production in amino acids rich medium.
Methods and Results:  Bacillus cereus anaerobic growth on different carbohydrates (glucose, fructose, sucrose or glucose–fructose mixture) was examined in synthetic mMOD medium under continuous cultures (μ = 0·2 h−1). Fermentation end-products, flux partitioning at each key branch points of the mixed acid pathway and consumption or production of amino acids were determined. On both fructose and sucrose, ATP production was favoured via acetate production from acetyl-CoA. In addition, amino acids present in the growth medium showed significant variations with high consumption of serine and net production of glutamate and alanine on some or all sugars. Enterotoxins Hbl and Nhe production was high during growth on fructose (or mixtures involving a fructose moiety).
Conclusions:  Fructose was identified as a key sugar influencing anaerobic metabolism and toxin production of B. cereus .
Significance and Impact of the Study:  The physiological differences associated with the fermentation of the various carbohydrates clearly modify toxinogenesis indicating that the risk of foodborne pathogens is to some extent dependent upon the prevailing nutritional environment.  相似文献

End-product synthesis and enzyme activities involved in pyruvate catabolism, H2 synthesis, and ethanol production in mid-log (OD600  0.25), early stationary (OD600  0.5), and stationary phase (OD600  0.7) cell extracts were determined in Clostridium thermocellum ATCC 27405 grown in batch cultures on cellobiose. Carbon dioxide, hydrogen, ethanol, acetate and formate were major end-products and their production paralleled growth and cellobiose consumption. Lactate dehydrogenase, pyruvate:formate lyase, pyruvate:ferredoxin oxidoreductase, methyl viologen-dependant hydrogenase, ferredoxin-dependant hydrogenase, NADH-dependant hydrogenase, NADPH-dependant hydrogenase, NADH-dependant acetaldehyde dehydrogenase, NADH-dependant alcohol dehydogenase, and NADPH-dependant alcohol dehydrogenase activities were detected in all extracts, while pyruate dehydrogenase and formate dehydrogenase activities were not detected. All hydrogenase activities decreased (2–12-fold) as growth progressed from early exponential to stationary phase. Alcohol dehydrogenase activities fluctuated only marginally (<45%), while lactate dehydrogenase, pyruvate:formate lyase, and pyruvate:ferredoxin oxidoreductase remained constant in all cell extracts. We have proposed a pathway involved in pyruvate catabolism and end-product formation based on enzyme activity profiles in conjunction with bioinformatics analysis.  相似文献

Summary The aerobic growth and metabolism of eleven homofermentative and three heterofermentative Lactobacillus strains, three Leuconostoc strains, two Brochothrix thermosphacta strains and two Carnobacterium strains were studied in batch cultures at pH 6.0 and 25°C on a complex substrate containing 10.0 g glucose per litre. All strains, except Carnobacterium divergens 69, grew well aerobically. An oxygen consumption was registered for 18 of the strains—the exceptions being Lactobacillus alimentarius DSM 20249T, Lactobacillus farciminis DSM 20284T and Lactobacillus sharpeae DSM 20505T. The homofermentative lactobacilli showed a maximal oxygen consumption during the stationary growth phase and this was coupled with a low final viable count. Leuconostoc strains, heterofermentative lactobacilli, Brochothrix thermosphacta and Carnobacterium strains showed a maximal oxygen consumption during the exponential growth phase together with a high final viable count. The maximum specific growth rate varied from 0.19 to 0.54 h-1 while the growth yield varied from 19 to 86 g dry weight per mol glucose consumed. In general, homofermentative lactobacilli produced dl-lactic acid, acetic acid and acetoin. The three heterofermentative lactobacilli produced dl-lactic acid and acetic acid, two strains also produced ethanol Leuconostoc spp. formed d-lactic acid, acetic acid, and ethanol. B. thermosphacta produced acetoin, acetic acid, formic acid, isobutyric acid and isovaleric acid but no lactic acid. Carnobacterium produced l-lactic acid, acetic acid and acetoin. tRNA containing N6-(Δ2-isopentenyl)adenosine may be precursors for the plant hormone cytokinin. To discriminate between tRNA containing and not containing cytokinin nucleotides, double labelling experiments were made by the use of [214C]-mevalonic acid and [3H-methyl]-methionine. At a generation cycle of 2 h for Lactobacillus acidophilus ATCC 4963, the half-lives of tRNA labelled with [3H-methyl]-methionine and [2-14C]-mevalonic acid are similar, namely 3 h. Plants of barley ( Hordeum vulgare L. cv. Salve) were grown with 6.5–35% relative increase of K+ supply per day (RKR) using a special computer-controlled culture unit. After a few days on the culture solution the plants adapted their relative growth rate (RGR) to the rate of nutrient supply. The roots of the plants remained in a low salt status irrespective of the rate of nutrient supply, whereas the concentration of K+ in shoots increased with RKR. Both Vmax and Km for K+(86Rb) influx increased with RKR. It is concluded that with a continuous and stable K+ stress, the K+ uptake system is adjusted to provide an effective K+ uptake at each given RKR. Allosteric regulation of K+ influx does not occur and efflux of K+ is very small.  相似文献

The influx of Rb+ into the roots of two barley varieties (Hordeum vulgare L. cv. Salve and cv. Ingrid) from a K+-free 86Rb-labelled nutrient solution with 2.0 mM Rb+, was checked at intervals from day 6 to day 18. The control plants were continuously grown in complete nutrient solution containing 5.0 mM K+, while two other groups of plants were grown in K+-free nutrient solution starting on day 6 and between day 6 and day 9, respectively. The pattern of Rb+ influx was similar for both varieties, although their efficiencies in absorbing Rb+ were different. The relationship between Rb+ influx and K+ concentration of the root could be interpreted in terms of negative feedback through allosteric control of uptake across the plasmalemma of the root cells. Hill plots were bimodal, but in the opposite direction. The Hill coefficients, reflecting the minimum number of interacting allosteric binding sites for K+ (Rb+), were low (≤–3.0). It is discussed whether the threshold value, that is the breaking point in the Hill plot, is indicative of a changed efficiency of transporting units for K+ (Rb+) transport to the xylem. Moreover, feedback regulation might be involved in transport of K+ between root and shoot. The variation in K+ concentrations in the roots and shoots of control plants were cyclic but in phase opposition despite an exponential growth. Encystation in Azospirillum brasilense ATCC 29145 was observed by using routine laboratory staining and phase-contrast and electron microscopy. Encystment occurred in liquid and in solid or semisolid media containing fructose (8 mM) and KNO3 (0.5 mM). The encysted forms consisted of a central body filled with poly-beta-hydroxybutyric acid granules, an electron-transparent intinelike region, and a thick outer layer. Enlarged giant encysted forms with multiple central bodies were also observed during the germination of a desiccated brown colony. Morphogenetically different forms in an aging culture could be resolved by sucrose density gradient centrifugation. The dense encysted forms along with numerous granules in a fibrillar network pelleted at 70% sucrose, while empty saclike envelopes along with vegetative cells and coccoid bodies pelleted at 55% sucrose. Different media induced various degrees of pigmentation in A. brasilense ATCC 29145 after aging. The pigment possessed several of the properties reported for microbial melanins, including insolubility in water and organic solvents, solubility in cold and hot alkali, and bleaching in hydrogen peroxide. The UV absorption maxima of the alkali extract were at 280 and 310 nm. Electron micrographs of the brown pigment showed that it occurred as aggregated granules surrounding the encysting cells as well as being excreted into the medium in an aging culture. It is concluded that A. brasilense ATCC 29145 produces compounds that form a brown pigment similar to melanin and are expressed under the influence of certain cultural conditions conducive for encystment.  相似文献

NH4+ and K+ uptake experiments have been conducted with 3 ectomycorrhizal fungi, originating from Douglas fir (Pseudotsuga menziesii (Mirb.] Franco) stands. At concentrations up to 250 μM, uptake of both NH4+ and K+ follow Michaelis-Menten kinetics. Laccaria bicolor (Maire) P. D. Orton, Lactarius rufus (Scop.) Fr. and Lactarius hepaticus Plowr. ap. Boud. exhibit Km values for NH4+ uptake of 6, 35, and 55 μM, respectively, and Km values for K+ uptake of 24, 18, and 96 μM, respectively. Addition of 100 μM NH4+ raises the Km of K+ uptake by L. bicolor to 35 μM, while the Vmax remains unchanged. It is argued that the increase of Km is possibly caused by depolarization of the plasma membrane. It is not due to a competitive inhibition of K+ by NH4+ since the apparent inhibitor constant is much higher than the Km, for NH4+ uptake. The possibility that NH4+ and K+ are taken up by the same carrier can be excluded. The Km, values for K+ uptake in the two other fungi are not significantly affected by 100 μM NH4+. Except for a direct effect of NH4+ on influx of K+ into the cells, there may also be an indirect effect after prolonged incubation of the cells in the presence of 100 μM NH4+.  相似文献

Six cultivars of spring barley ( Hordeum vulgare L. cvs Salve, Nümberg II, Bomi, Risø 1508, Mona and Sv 73 608) were grown in water culture for three weeks with various combinations of mineral supply and differential roots/shoot temperatures during the growth period. Most important for growth and accumulation of N, K+, Ca2+ and Mg2+ was the mineral supply, followed by the root temperature and the choice of cultivar. Treatments with low mineral supply or low root temperature induced a uniform reduction in growth and accumulation of the ions studied. The effects of low mineral supply and low root temperature on growth and N accumulation was additive, which indicates that these factors exert their influence independently of each other.
Roots grown at 10°C were smaller and Rb+(86Rb) influx was higher than in roots grown at 20°C. It is suggested that the control of Rb+(86Rb) influx is affected by the root temperature and the age of the plants. The higher 86Rb+ (86Rb) influx into the low temperature roots could not compensate for the smaller root size. However, the lower total mineral accumulation made up for the needs of the smaller plants and cannot explain the reduction in growth.  相似文献

Plasmid profiles of 30 strains of Lactobacillus plantarum isolated from 3-year-old tissue cultures and surface sterilized stem sections of glasshouse grown plants of three different varieties of Hemerocallis were compared to pinpoint the source of Lact. plantarum contamination in plant tissue cultures. Since the profiles of strains isolated from in vitro and in vivo plant material were generally identical, it is thought that the source of Lact. plantarum contamination is the plant material used to initiate plant tissue cultures.  相似文献

Uptake of Rb+ from a complete nutrient solution with 2.0 mM Rb+ was studied in roots of spring wheat seedlings ( Triticum aestivum L. cv. Svenno) with different K+ levels. The relationship between Rb+ uptake and concentration of K+ in the roots indicated a negative feedback mechanism operating through allosteric control. The Rb+ uptake process in root cells was divided into two steps: (1) binding of the ion in the free space, and (ii) transmembrane transport into the cytoplasm. Metabolic and non-metabolic components of uptake were separated by addition of the metabolic inhibitor 2,4-dinitrophenol (DNP) to the nutrient solution. It is suggested that metabolic Rb+ uptake requires energy in two uptake steps (for binding to the carrier entity in the free space and for transmembrane transport) or in one step only (for transmembrane transport), dependent on the K+ status of the roots. The change from metabolic to non-metabolic binding in the free space is accomplished by changing the conformational state of the carrier (slow/fast transitions). There may be a hysteretic effect on metabolic Rb+ uptake through a slow transition between carrier states. This is superimposed on the negative cooperativity, strengthening further cooperativity at intermediate K+ levels in the roots. Non-metabolic Rb+ uptake probably consists of two components, a carrier-mediated (facilitated diffusion) and a parallel diffusive component.  相似文献

The potentially low infective dose of Escherichia coli O157 makes it necessary to be able to detect low numbers in food, and the lack of sensitivitiy of direct plating has led to the development of various enrichment and detection methods. Until now, the most selective procedure for detection of E. coli O157 isolates was the immunomagnetic separation (IMS) method. The number of sorbitol non-fermenting micro-organisms other than E. coli O157 that adhere non-specifically to the magnetic beads hampers the application of IMS. The use of IMS in conjunction with 3 M Petrifilm-HECTM yielded EHEC O157 in 21 of 165 samples of minced meat (12·7%). Without advance application of IMS, Petrifilm plates often yield confluent growth and colonies too numerous to count. The Vitek Immunodiagnostic Assay SystemTM (VIDAS-ECO) showed good sensitivity when testing artificially contaminated beef samples, but only four of 21 naturally contaminated samples were recognized. The addition of 3 M Petrifilm to IMS resulted in less growth of contaminants and eliminated much of the need to test presumed colonies for confirmation. The combination of IMS with 3 M Petrifilm-HECTM is a fast and efficient screening procedure for E. coli O157 in minced meat.  相似文献
