Zeatin and zeatin riboside were identified by full-scan gas chromatography-mass spectrometry (GC-MS) in xylem sap of clonal apple rootstocks (M.27, M.9 and MM.106). These rootstocks exhibit a wide range of control over tree size when grafted to a common scion. The concentrations of zeatin and zeatin riboside were measured by GC-MS selected ion monitoring (SIM) in shoot xylem sap and root pressure exudate obtained from these rootstocks and from trees of Fiesta scion grafted onto the rootstocks. Zeatin was the predominant cytokinin in xylem sap from the dwarfing rootstocks, M.27 and M.9, while zeatin riboside was the predominant cytokinin in xylem sap from the more invigorating rootstock MM.106. Cytokinin concentrations (ng ml–1) in root pressure exudate and shoot xylem sap, (i.e. from above the graft union in composite trees), increased with increasing vigour of the rootstock, irrespective of whether the plants were non-grafted rootstocks, or were composite plants of Fiesta scion grafted onto the rootstocks. Cytokinin content (ng shoot–1) of shoot sap differed with rootstock; the more invigorating (MM.106) had greater amounts of cytokinins than the more dwarfing (M.9 and M.27) rootstocks. These results are discussed in relation to possible influences of roots on the growth of shoots via cytokinin supplies in the xylem sap.  相似文献

Summary Zeatin and its derivatives are major consituents of higher plant cytokinins. Metabolic steps modifying the isoprenoid side chain, such as O-glycosylation, are expected to have a direct bearing on cytokinin-mediated processes. To examine this possibility, transgenic tobacco plants were generated harboring a gene (ZOG1) encoding a zeatin O-glucosyltransferase from Phaseolus lunatus under the control of a constitutive (35S) and an inducible (Tet) promoter. The presence of the transgene resulted in elevated enzyme production and conversion of exogenous zeatin to its O-glucoside, confirming the expression of the ZOG1 gene in transgenic plants. Endogenous O-glucosylzeatin was increased from less than 1 pmol per g fresh weight in leaves and roots of controls to 26 and 68 pmol per g fresh weight in leaves and roots of 35S-ZOG1 transformants, respectively. In cytokinin/auxin interaction experiments, Tet-ZOG1 leaf discs, in the presence of tetracycline, required 10-fold higher zeatin concentrations for the formation of shoots and callus than the controls. In 35S-ZOG1 plants, developmental changes included adventitious root formation on the lower stems, shorter stature, and axillary shoot growth. Thus, increased zeatin O-glucosylation in detached, cytokinin-dependent tissues leads to a shift in the response to exogenous zeatin indicative of cytokinin sequestering. In whole plants the effect can simulate a reduction or a rise in cytokinin activity depending on the tissue and stage of development. The use of tissue- and stagespecific promoters in the future will allow more precise analyses and targeted growth alterations.  相似文献

Dihydrozeatin, at 4×10–5 M, delayed the senescence of carnation flowers while tZ, at the same concentration, accelerated it. cis-Zeatin was ineffective. The DHZ derivatives as well as the Z derivatives gave responses very similar to those observed for the parent free bases. The cytokinin content of stem tissues, primary genetic tumours (excised from 2-month-old plants) and 3-week-old in vitro cultured genetic tumour tissues derived from Nicotiana glauca (Grah.) × langsdorffii (Weinm.) and N. suaveolens (Lehm.) × langsdorffii (Weinm.) hybrids and stem tissues derived from 2-month-old N. suaveolens and N. langsdorffii plants has been analysed by radioimmunoassay. Stem tissues of tumour-prone hybrids contain high cytokinin levels (3–3.7 nmol g−1). This increase is caused mainly by increased levels of cytokinin nucleotides, particularly those of zeatin nucleotide (0.5 nmol g−1) in stem tissues of parent plants and 2.4 nmol g−1 in stem tissues of hybrids). All other tissues contain lower cytokinin levels (0.7–1.7 nmol g−1). Cytokinin bases and ribosides are major compounds in cultured tumour tissues while the nucleotides are dominant cytokinins in all freshly excised tissues from parent plants and their hybrids. In a separate study, the metabolic fate of supplied [3Hj-zeatin riboside. which is inactivated mainly by sidechain cleavage, has been studied. The results collectively suggest that cytokinins may be involved in tumourigenesis.  相似文献

Anthocyanin formation in a suspension culture of Daucus carota is induced by transfer from medium containing 2,4-dichlorophenoxyacetic acid (2,4-D) to one lacking 2,4-D. The specific yields were strongly influenced by the inoculum density. Inoculum density altered the effect of zeatin concentration on anthocyanin accumulation. The in part by increasing the sucrose levels. It was inferred from the results that sucrose was exhausted at a low concentration of sucrose and at a high cell density, resulting in the decrease of yield of anthocyanin.  相似文献

Elongation, indolyl-3-acetic acid (IAA) and abscisic acid (ABA) levels, – gas chromatography-mass spectrometry quantification –, in the elongating zone were analysed for maize ( Zea mays L., Cv. LG11) roots immersed in buffer solution with or without zeatin (Z). The effect of Z depends on the initial extension rate of roots. The slower growing roots are more strongly inhibited by Z (10−7−10−5 M ) and they show a greater increase in IAA and ABA content. When compared to the rapidly growing roots, the larger reactivity of the 'slow'ones cannot be attributed to a higher Z uptake as shown when using [14C]-Z. It is suggested that Z could regulate root elongation by acting on the IAA and/or ABA level. The comparative action of these two hormones is discussed.  相似文献

Irrespective of their age, leaves of Ginkgo biloba metabolised applied 8 (14C) zeatin to compounds of similar chromatographic properties. Glucosylation is apparently not a normal feature of cytokinin metabolism in immature leaves. However, the application of zeatin to these leaves did result in the formation of metabolites which co-chromatographed with glucosylated cytokinins. As far as cytokinin metabolism is concerned therefore, this application of excess zeatin allowed immature leaves to behave as mature or senescing leaves. Overall metabolism was fastest in immature leaves. From the metabolites formed it would appear as if oxidation, which resulted in the formation of a metabolite which co-eluted with N-(purin-6-yl)glycine, was also important in immature leaves. In senescing leaves glucosylation was the major form of metabolism. Extraction and re-application of the polar metabolites (formed from zeatin) to mature leaves resulted in the formation of compounds which co-chromatographed with zeatin. This suggests that these compounds could serve as precursors for zeatin or could be hydrolysed to form zeatin.Very little of the applied radioactivity was exported from the leaves irrespective of their physiological age. When the metabolites, obtained after zeatin application to mature leaves, were extracted and reapplied to the leaves, export of radioactive material was much improved. The results suggest that should cytokinins such as zeatin be translocated to mature leaves of this deciduous gymnosperm their export from the leaves would be unlikely unless first metabolised. In an effort to understand the causes of arrest of somatic embryo development, generally observed in grapevine (Vitis sp.), histological studies were undertaken, using two cultivars (CH76 and 41B) which differ in their ability to develop into plants. Embryos with a high conversion rate (70%; CH76) formed a well-structured and functional shoot apex between two thread-like cotyledons. In contrast, embryos with a low conversion rate (10%; 41B) formed a normal root apex but lacked a well-structured shoot apex and developed a wide range of aberrant forms in the intercotyledonary area: uncontrolled cellular proliferation, formation of adventitious buds, over-growth of cotyledonary or leaf meristems. ABA increased the conversion rate of 41B embryos from 10% to 20%, but failed to improve embryo morphology. Zeatin and BAP promoted growth of 41B somatic embryos, but generated a high level of abnormalities and failed to improve conversion rate. Applied in combination with ABA, these PGRs increased the frequency of cotyledonary embryos, but decreased the conversion rate.  相似文献

As a ubiquitous reaction, glucosylation controls the bioactivity of cytokinins in plant growth and development.Here we show that genetic manipulation of zeatin-Oglucosylation regulates the formation of important agronomic traits in rice by manipulating the expression of OscZOG1 gene,encoding a putative zeatin O-glucosyltransferase. We found that OscZOG1 was preferentially expressed in shoot and root meristematic tissues and nascent organs. The growth of lateral roots was stimulated in the overexpression lines, but inhibited in RNA interference lines. In shoots, knockdown of OscZOG1 expression by RNA interference significantly improved tillering, panicle branching, grain number per panicle and seed size, which are important agronomic traits for grain yield. In contrast, constitutive expression of OscZOG1 leads to negative effects on the formation of the grain-yielding traits with a marked increase in the accumulation levels of cis-zeatin O-glucoside(c ZOG) in the transgenic rice plants. In this study,our findings demonstrate the feasibility of improving the critical yield-determinant agronomic traits, including tiller number, panicle branches, total grain number per panicle and grain weight by downregulating the expression level of OscZOG1. Our results suggest that modulating the levels of cytokinin glucosylation can function as a fine-tuning switch in regulating the formation of agronomic traits in rice.  相似文献

应用酶联免疫吸附法(enzyme-linked immunosorbent assay, ELISA)研究了在褐飞虱胁迫下镇稻2号(粳稻)和协优63(籼稻)抽穗期和灌浆期水稻叶片和根中玉米素核苷(zeatin riboside, ZR)含量变化情况。结果表明, 镇稻2号抽穗期ZR含量变化比灌浆期对褐飞虱侵害更为敏感,密度分别为15、30、60、120头/株的褐飞虱侵害水稻抽穗期3、6、9天后,叶片和根中ZR含量显著下降;灌浆期除各个褐飞虱密度侵害9天叶片中ZR含量和120头/株褐飞虱密度侵害9天根部ZR含量显著下降外,其他处理根和叶片中ZR含量下降不明显。协优63抽穗期受褐飞虱侵害后体内ZR的变化不同于镇稻2号,密度分别为15、30、60头/株的褐飞虱侵害3天后,叶片中ZR含量明显升高;在灌浆期,除30、60、120头/株褐飞虱密度侵害6天和120头/株褐飞虱密度侵害9天叶片中ZR含量有显著增加外,其他处理变化不明显。表明不同品种水稻在不同密度褐飞虱侵害下对根和叶片中ZR含量有不同的影响。  相似文献

A new method is proposed for differential quantitative assay of two major endogenous cytokinin forms. It is based on determination of two effective parameters-concentrations of zeatin and zeatin riboside--with the use of appropriate antigens as standards. The method can be used for determining cytokinins in small samples of plant tissues without extract fractionation. This study pioneers in quantitation of changes in the hormonal status of ovules and ovaries of Triticum aestivum L. at early stages of embryogeny. A gradual increase in the content of the active and storage forms of the hormones from the ovary to the ovule was revealed.  相似文献

High temperatures during reproductive development alter kernel development and reduce yield and quality in wheat ( Triticum aestivum L.). Understanding how temperature alters kernel formation will help the development of genetic approaches to enhance heat tolerance in this cereal. A relationship between kernel development and postanthesis cytokinin accumulation is well documented, but the effect of temperature on this relationship is not known. This study quantified the effects of a postanthesis temperature treatment (7 d at 35/25 °C day/night) on kernel development and cytokinin accumulation in a soft white winter wheat (c.v. Stephens). Kernels from control plants maintained at 25/15 °C accumulated zeatin, dihydrozeatin and their corresponding 9-ribosides from 1 to 4 d postanthesis. Postanthesis kernel cytokinin accumulation was reduced by 50–80% by the temperature treatment. Kernel cytokinin content in control plants declined to baseline by 5 d postanthesis and remained at that level. Kernels from treated plants had a secondary peak of cytokinin accumulation 6–8 d after anthesis. This treatment significantly reduced kernel weight. The magnitude of the effect on kernel number was smaller than on kernel weight, but was statistically significant. Reduced kernel weight was accompanied by reduced cytokinin accumulation. Exogenous cytokinins did not mitigate the temperature effects on kernel weight, but at the highest concentrations, did reduce the effect on kernel number.  相似文献

Zeatin indole-3-acetate, 6-[4-(indole-3-acetoxy)-3-methyl-trans-2-butenylamino]purine, is at least as effective as zeatin on a molar basis in satisfying the cytokinin requirement for growth and bud formation in tobacco bioassays. It is less effective than indole-3-acetic acid and is needed as a variable function of the cytokinin concentration for satisfying the optimal requirement of an auxin. Comparisons of the types of growth and yield of tissue obtained with serial concentration of the ester and with equimolar mixtures of its free base and acid indicate that the relative requirement for auxin changes with the concentration of cytokinin and is related to the types of callus growth and differentiation which occur. The results also suggest that the ester serves as a source of auxin only after modification, presumably by hydrolysis to indoleacetic acid.  相似文献

Roots are recognised as the major sites of cytokinin synthesis and shoots receive a continuous supply of cytokinins from the roots. Although reports are available on the xylem mobility of putative free bases and their ribosides, relatively few studies on the phloem mobility of cytokinins have been reported. The origin of phloem-mobile cytokinins is uncertain but there is evidence which implicates a recirculation from the root source. This study is the first report in which zeatin and zeatin riboside from the root pressure exudate and phloem sap of Ricinus have been identified by full-scan GC-MS and quantified by GC-MS selective-ion-monitoring. In this study, the concentration of cytokinins in root pressure exudate was similar, but lower, and in the phloem sap higher than that reported previously. The concentration of cytokinins quantified in the phloem sap confirms their transport in the sieve tubes. The relatively high concentration of zeatin riboside detected in the root pressure exudate and of zeatin detected in the phloem sap indicate a possible vascular recirculation of these hormones.  相似文献

Cytokinin-like substances in both healthy and infected ( Plasmodiophora brassicae ) Wor. strain S) roots of Brassica campestris L. ssp. pekinensis cv. Granat have been tentatively identified and quantified by HPLC. The isomers of the cytokinins could be seperated on a reversed phase column using a gradient elution with increasing amounts of methanol. Secondary plasmodia were isolated mechanically from Plasmodiophora brassicae infected roots. The time course of adenine uptake and its conversion to cytokinins were investigated. Evidence is presented for the incorporation of [U-14C]-adenine into trans -zeatin by secondary plasmodia.  相似文献
