Abstract: Relocations of gopher tortoises (Gopherus polyphemus) in Florida, USA, are frequently employed as mitigation tools when tortoises occupy land desired for development. Here we present information about retention and health of a relocated population of gopher tortoises 17 years after relocation. We combine our 17-year postrelocation data with earlier surveys 1 year and 2 years postrelocation to examine whether retention rates change over time. We also evaluate whether retention rates vary by age and gender. Of 74 gopher tortoises relocated in 1985, 31 were present in 2002. We found a 1-year retention rate of 42%, with retention rates of 100% each year thereafter, when we used the percentage of relocated individuals captured at each survey. We found a 1-year retention rate of 73%, a retention rate of 92% from year 1 to year 2, and an overall retention rate of 98.5% from year 2 through year 17, based on the assumption that all individuals present in later surveys were present in earlier surveys. We found no significant difference in retention rate over the 17-year period for adults and juveniles and for adult males and females. Relocated gopher tortoises showed natural growth patterns, indicating good health, but 35% of these gopher tortoises had ≥1 symptom of upper respiratory tract disease, a disease associated elsewhere with population declines of tortoises. Thus, retention rates of relocated gopher tortoises change over time, with relatively low retention during the first year postrelocation but nearly 100% retention in subsequent years. In general, our study shows that relocations can successfully lead to long-term retention of gopher tortoises, but we predict that without management (e.g., fire management and predator control) this relocated population is not viable.  相似文献

The effect of composition and origin of maltodextrins on the retention of 38 components in a mixture of volatile organic substances (odorants) during 6-month storage was studied by means of capillary gas-liquid chromatography. The retention of esters increased with an increase in their molecular weight. The retention of lactones, phenols, linalool, menthone, and damascone was 75–85%. Storage of aldehydes was accompanied by oxidation, and the retention of these substances did not exceed 55%. The retention of odorants increased with a decrease in the molecular weight of maltodextrins. The long-term viability and plasmid retention of recombinant Escherichia coli strains were investigated by real-time testing of master cell banks (MCBs) stored at the Roche Molecular Systems Culture Collection (RMSCC). MCBs at the RMSCC were cryogenically frozen and stored at -80 degrees C for long-term preservation. At regular intervals during a period of 5 to more than 10 years, representative cryovials of each MCB were tested for viability and plasmid retention. Plasmid retention and viability for all 30 MCBs were stable over time. Twenty-seven MCBs maintained high levels of plasmid retention (at or near 100%), while three MCBs showed lower plasmid retention rates (ranging from 13.9 to 96.5%) that were consistent over time. New MCBs with high plasmid retention were created from two of the MCBs with lower plasmid retention by selective pressure with high levels of antibiotics. These new MCBs have shown stable viability and high plasmid retention over the first 5 months of storage. In conclusion, this study shows that properly selected, frozen and stored MCBs retain viability and maintain plasmid retention over time. Moreover, it is possible to recover cultures with high plasmid retention from MCBs with low plasmid retention by selecting clones grown in the presence of high levels of antibiotics.  相似文献

Intravascular LPS rapidly induces neutrophil sequestration in pulmonary capillaries by mechanisms that, although currently unknown, must take into account the size difference between the neutrophil and capillary diameter. To determine whether LPS alters neutrophil stiffness, and hence the ability of neutrophils to traverse capillaries, neutrophil passage through pulmonary capillaries was modeled by passage through filters with 6.5-microns pores. LPS increased retention in the pores in a concentration-dependent fashion that required the presence of heat-inactivated platelet-poor plasma, and was evident as early as 10 min after stimulation. The effect of LPS on the structural properties of the neutrophil was then studied. LPS induced f-actin reorganization in neutrophils in the presence of plasma. Disruption of actin organization and assembly with cytochalasin D completely inhibited early LPS-induced retention and attenuated retention at later timepoints, indicating that LPS-stimulated retention depends on filament organization. LPS-induced actin assembly and retention were abrogated by an antibody directed against CD14, a putative LPS receptor. CD18-dependent adherence of neutrophils contributed significantly to retention only at later timepoints with no significant contribution to retention at 20 min as determined by inhibition of adherence with the mAb 60.3. Morphometric assessment of neutrophil accumulation in the lungs of rabbits given 1 microgram LPS showed a marked increase in apparent neutrophil number, which was unaltered by antibodies to CD18, suggesting that mechanisms other than adhesion may account for accumulation in vivo. Direct measurements showed that neutrophil stiffness increased with exposure to LPS in a fashion similar to LPS-induced retention and actin organization. Pretreatment of neutrophils with cytochalasin D attenuated the increased stiffness. These data suggest that reorganization of filamentous-actin induced by LPS leads to cell stiffening and retention in capillary-sized pores. Although the organization of f-actin continues to be important in retention at later time points, adherence of cells also contributes significantly to cell retention. The changes in mechanical properties of the neutrophil may be important in the sequestration of neutrophils in pulmonary capillaries noted in endotoxemia.  相似文献

We examined retention of butt-end aluminum leg bands on Rio Grande wild turkeys (Meleagris gallopavo intermedia) captured in Texas and Kansas, USA, 2000–2009. We examined 187 recaptured or harvested radiotagged wild turkeys to determine band retention and modeled band retention with Program MARK. We did not detect differences in band retention among age and gender classes or that band retention probability was time dependent. We estimated monthly probability of band retention was 0.990 (SE = 0.002). Band retention probability was 0.971 (SE = 0.006) at 3 months post-banding and 0.864 (SE = 0.028) at 15 months post-banding. Butt-end aluminum leg band retention was not 100% for wild turkeys marked during our work; however, our band retention rates were 3.7–5.7 times greater than described previously. © 2011 The Wildlife Society.  相似文献

水库对氮营养盐的滞留效应对水体富营养化防控有重要影响,为揭示不同时间尺度下水库氮营养盐的滞留特征及影响因素的差异,通过构建山美水库流域生态动力学模型,在统计水库氮滞留量和滞留率的基础上,从年、月、日不同时间尺度对氮营养盐的滞留效应进行分析;并通过多元线性相关分析,对入库流量、出入库流量比、水力滞留时间、流速、温度5种因素的影响进行研究.结果表明: 不同时间尺度下山美水库的氮滞留过程均呈现波动特征.在年尺度上,水库主要呈现正滞留效应,是流域重要的氮汇;在月尺度上,氮滞留呈现丰枯期变化特征,水库在源和汇的角色上发生转变;在日尺度上,枯水年氮滞留过程波动剧烈,滞留率在-300%~100%之间变化.不同时间尺度下,出入库流量比、流速因子均是影响水库氮滞留的重要因素;随着时间尺度的降低,入库流量和温度因子对氮滞留的影响程度有所增强;水力滞留时间对氮滞留的影响程度则随时间尺度变化而有正负差异.  相似文献

A comparative study of digestion in North Atlantic seabirds   总被引:2,自引:0,他引:2  
We present data on digestive efficiencies and gut retention times of eight North Atlantic seabird species, fed on two fish species – lesser sandeel Ammodytes marinus and whiting Merlangius merlangus – which commonly occur in the diet of wild seabirds. In an interspecific comparison, there was a positive relationship between retention time and digestive efficiency, which we suggest represents a trade-off between conflicting benefits of efficient digestion and rapid digestion. Analysis of excretion curves revealed that retention time of digesta in the stomach was more important than passage time of digesta through the intestine in determining whole gut retention time. Differences in stomach retention time of lesser sandeel and whiting explained the longer overall retention time of the latter diet. Stomach retention time and whole gut retention time were greater in species with relatively large stomachs, while intestine passage time was correlated with relative intestine length. Species which typically eat a wide range of food types, including low quality items, tended to have slow and efficient digestion and heavy stomachs, whereas species which specialise on readily digestible and energy dense food types had the opposite digestion strategy.  相似文献

Aggregated LDL (AgLDL) accumulates within the subendothelial space of developing atherosclerotic lesions. We were interested to learn whether endothelial cells can interact with AgLDL. Incubation of endothelial cells with AgLDL resulted in apparent cholesterol retention. Microscopic examination revealed that cholesterol retention resulted mainly from endothelial cell surface attachment of AgLDL. Little AgLDL entered endothelial cells consistent with the small amount of endothelial cell degradation of AgLDL. Although endothelial cell retention of AgLDL was inhibited by LDL, AgLDL retention was not blocked by lactoferrin, C7 anti-LDL receptor monoclonal antibody, or receptor-associated protein, suggesting that LDL receptor family members did not mediate this retention. Surface retention of AgLDL depended on microtubule function and could be regulated by the protein kinase C activator, PMA. Treatment of endothelial cells with PMA either before or during, but not after incubation with AgLDL, inhibited retention of AgLDL. Our findings show that endothelial cells can retain AgLDL but internalize and metabolize little of this AgLDL. Thus, it is unlikely that endothelial cells can transport AgLDL out of atherosclerotic lesions, but it is likely that retention of AgLDL affects endothelial function.  相似文献

We explored modulation of retention time in cedar waxwings (Bombycilla cedrorum) by feeding them diets varying in hexose concentration. Our goals were to (1) test three predictions of a chemical reactor-based model of how guts might respond optimally to diet shifts; (2) determine whether modulation of retention time can occur quickly, thereby facilitating rapid changes in diet; (3) tease apart the relative influence of ingestion rate and nutrient concentration on retention time; and (4) examine the degree of axial mixing in the intestine and its relationship with retention time. The model's predictions were rejected: mean retention time did not decrease, ingestion rate did not increase, and glucose assimilation efficiency did not decrease with increased hexose concentration of the diet. Instead, birds displayed maximal intake rate at intermediate sugar concentration, and mouth to cloaca mean retention times increased with hexose concentration. Significant modulation of retention time occurred quickly, within 3 h of exposure to a different diet. Birds did equally well in terms of total energy assimilated on diets differing 3.3-fold in hexose concentration (from 500 mmol/L to 1660 mmol/L) but showed reduced intake when fed food with low hexose concentration (110 mmol/L). Far more variation in retention time was explained by direct effects of ingestion rate than by direct effects of hexose concentration. Finally, a gut dispersion index that measured degree of axial mixing was positively correlated with mean retention time, indicating that higher retention times are accompanied by increased axial mixing. We propose a modification of the assumptions of the original model. The resulting "osmotic constraint" model better captures the interaction between feeding rate and digestive function in fruit-eating birds.  相似文献

Two models were used to examine the relationship between hydraulic retention time, nutrient source and total chlorophyll in a shallow lake (Bassenthwaite Lake, UK). The first model was a derivation of the Vollenweider model and the second was the phytoplankton community model, PROTECH. The adapted Vollenweider model produced two different responses to changing the retention time that were phosphorus source dependent. If the phosphorus was totally from a point source, then annual mean chlorophyll steadily declined with increasing flushing rate. However, when a diffuse source was used, the chlorophyll changed little and even increased with short retention times (retention time <40 days). The PROTECH model produced some similar responses but they were more season dependent. Winter mean chlorophyll always declined with decreasing retention time, regardless of nutrient source, but the summer mean curves were source dependent and similar to those produced by the adapted Vollenweider model. Further simulations with PROTECH using a standardized flow regime provided strong evidence as to the mechanisms behind these responses. Analysis showed that the decline in chlorophyll with decreasing retention time was the prevalent response of the PROTECH simulations due to flushing loss of both nutrients and algae. Furthermore, the curve formed an asymptote at long retention times because other factors (e.g. light) limited growth; retention times >100 days had little effect on chlorophyll. Rat exposure to 60% oxygen (O(2)) for 7 days (hyper-60) or to >95% O(2) for 2 days followed by 24 h in room air (hyper-95R) confers susceptibility or tolerance, respectively, of the otherwise lethal effects of subsequent exposure to 100% O(2). The objective of this study was to determine if lung retention of the radiopharmaceutical agent technetium-labeled-hexamethylpropyleneamine oxime (HMPAO) is differentially altered in hyper-60 and hyper-95R rats. Tissue retention of HMPAO is dependent on intracellular content of the antioxidant GSH and mitochondrial function. HMPAO was injected intravenously in anesthetized rats, and planar images were acquired. We investigated the role of GSH in the lung retention of HMPAO by pretreating rats with the GSH-depleting agent diethyl maleate (DEM) prior to imaging. We also measured GSH content and activities of mitochondrial complexes I and IV in lung homogenate. The lung retention of HMPAO increased by ~50% and ~250% in hyper-60 and hyper-95R rats, respectively, compared with retention in rats exposed to room air (normoxic). DEM decreased retention in normoxic (~26%) and hyper-95R (~56%) rats compared with retention in the absence of DEM. GSH content increased by 19% and 40% in hyper-60 and hyper-95R lung homogenate compared with normoxic lung homogenate. Complex I activity decreased by ~50% in hyper-60 and hyper-95R lung homogenate compared with activity in normoxic lung homogenate. However, complex IV activity was increased by 32% in hyper-95R lung homogenate only. Furthermore, we identified correlations between the GSH content in lung homogenate and the DEM-sensitive fraction of HMPAO retention and between the complex IV/complex I activity ratio and the DEM-insensitive fraction of HMPAO retention. These results suggest that an increase in the GSH-dependent component of the lung retention of HMPAO may be a marker of tolerance to sustained exposure to hyperoxia.  相似文献

MOTIVATION: Liquid chromatography-tandem mass spectrometry (LC-MS/MS) is a powerful tool in proteomics studies, but when peptide retention information is used for identification purposes, it remains challenging to compare multiple LC-MS/MS runs or to match observed and predicted retention times, because small changes of LC conditions unavoidably lead to variability in retention times. In addition, non-contiguous retention data obtained with different LC-MS instruments or in different laboratories must be aligned to confirm and utilize rapidly accumulating published proteomics data. RESULTS: We have developed a new alignment method for peptide retention times based on linear solvent strength (LSS) theory. We found that log k(0) (logarithm of retention factor for a given organic solvent) in the LSS theory can be utilized as a 'universal' retention index of peptides (RIP) that is independent of LC gradients, and depends solely on the constituents of the mobile phase and the stationary phases. We introduced a machine learning-based scheme to optimize the conversion function of gradient retention times (t(g)) to log k(0). Using the optimized function, t(g) values obtained with different LC-MS systems can be directly compared with each other on the RIP scale. In an examination of Arabidopsis proteomic data, the vast majority of retention time variability was removed, and five datasets obtained with various LC-MS systems were successfully aligned on the RIP scale.  相似文献

The influence of diet or its ingredients on 141Ce absorption and retention was investigated in six-day-old rats. Animals were fed over 8h with cow's milk, rat diet or a mixture of rat diet ingredients (fish meal, sunflower meal, alfalfa, cane molasses and premix) labelled with 141Ce. Whole-body radioactivity was determined in a double crystal scintillation counter every 24 h over a six-day period. Gut, liver, kidney and femur retention and cerium distribution in the gut was determined at the end of the experiment. Compared to milk diet, administration of rat diet or ingredients caused respectively 3 and 7.5 times lower whole body retention. Carcass retention was reduced by rat diet or ingredients 2-3 times and intestinal retention 3 and 8 times respectively. Irrespective of the dietary treatment the main site of cerium intestinal retention was the ileum. Our present results indicate that some compounds of rat diet might be considered as a means of reducing cerium absorption and intestinal retention in the very young.  相似文献

东江流域森林水源涵养功能对于保障流域可持续发展具有重要作用。应用In VEST模型,结合东江流域土地覆盖分类数据、气候数据、土壤数据评估了东江流域森林水源涵养功能的空间分布。结果表明:东江流域森林生态系统水源涵养总量为47.29×10~8m~3,水源涵养功能最高为572.6 mm,平均水源涵养功能为204.15 mm。东江流域森林水源涵养功能在空间上呈现出不同的分布特征,流域内森林水源涵养功能随海拔升高呈现先上升后降低的趋势,在海拔900—1200 m范围,水源涵养功能平均值达到最大值270 mm;流域内森林水源涵养功能随坡度升高呈增加趋势,在坡度大于50°的区域,森林水源涵养功能平均值增加到327.2 mm,高于流域平均水平60%。流域水源涵养功能呈现中游下游上游的空间格局,流域上、中、下游地区的水源涵养总量占流域的比例分别为11%、72%、17%。  相似文献

Summary A study was made of the retention times of N and P in the leaf biomass and their relationship with the retranslocation percentages and the leaf longevities in some woody species in Central Spain. The retention times of both nutrients were strongly related to the nutrient status of each species. These results suggest that a prolonged retention time is a way of increasing nutrient use efficiency in conditions of low nutrient availability. Plants can increase the retention time of nutrients in their leaf biomass by means of an increase in leaf longevity and/or by means of an increase in retranslocation efficiency. However, the effect of the retranslocation efficiency on retention times was almost negligible compared with the effect of leaf longevity. This suggests that an increase in leaf longevity is probably the best adaptation for increasing efficiency in the use of nutrients.  相似文献

水源涵养是生态系统的重要服务功能之一.识别水源涵养重要区对于制定水源保护规划,落实陆域-水域综合保护,防治水体污染,保障区域用水安全具有重要意义.以2008年四川都江堰市为研究范围,采用生态系统服务功能综合权衡工具-Integrated Valuation of Ecosystem Services and Tradeoffs(InVEST)中的水源涵养模型对县域生态系统的水源涵养功能进行了空间制图,然后采用综合指数法将水源涵养功能与植被减洪能力和水源地保护区进行叠加计算水源涵养重要性指数.结果表明:尽管汶川地震导致都江堰市域部分地区水源涵养功能降低,但都江堰市水源涵养功能整体仍然较高,部分地区多年平均水源涵养量超过200mm/a.都江堰市的水源涵养极重要区面积为421km2,占全市总面积的34.9%,远大于目前县域的水源保护区面积,未来应进一步加强对这些极重要区的保护,以保证本市以及成都平原的供水安全.水域涵养功能及其重要性都存在较明显的空间分异,大致呈现由西北向东南减少的趋势,水源涵养重要性与水源功能的分布密切相关,但又不完全一致.因此,在县域尺度上采用服务功能空间化评估结果进行重要性评价能够比较精细地反映生态系统水源涵养重要性的空间差异.但该方法还需要将人类用水需求进行空间表达,与本文的方法进行结合,以更准确反映水源涵养重要性的空间分异.  相似文献

These experiments examined the effects of hypophysectomy on retention of avoidance training. In addition, the experiments examined the effects, on retention, of post-training ACTH injections administered to hypophysectomized rats. Rats were trained in a visual discriminated avoidance Y maze. Each rat received six training trials followed by six retraining trials the next day. Retention was measured by the number of correct choices during the retraining trials. When trained with a low-footshock intensity (0.8 mA), hypophysectomized rats showed retention performance which was significantly poorer than that of intact animals. There was no significant difference in performance when the animals were trained with a higher footshock intensity (1.4 mA), in part because of poorer retention performance of intact animals under these training conditions. Under both footshock conditions, a single post-training injection of ACTH enhanced later retention performance of hypophysectomized rats. This effect on memory was timedependent; injections delayed 2 or 6 hr after training did not significantly enhance retention. These findings are consistent with the view that hormonal responses to training may modulate later retention of the training experience.  相似文献

Classically, endoplasmic reticulum (ER) retention signals in secreted integral membrane proteins impose the requirement to assemble with other cognate subunits to form functional assemblies before they can exit the ER. We report that GluK5 has two ER retention signals in its cytoplasmic C-terminus: an arginine-based signal and a di-leucine motif previously thought to be an endocytic motif. GluK5 assembles with GluK2, but surprisingly GluK2 association does little to block the ER retention signals. We find instead that the ER retention signals are blocked by two proteins involved in intracellular trafficking, SAP97 and CASK. We show that SAP97, in the presence of CASK and the receptor complex, assumes an extended conformation. In the extended conformation, SAP97 makes its SH3 and GuK domains available to bind and sterically mask the ER retention signals in the GluK5 C-terminus. SAP97 and CASK are also necessary for sorting receptor cargoes into the local dendritic secretory pathway in neurons. We show that the ER retention signals of GluK5 play a vital role in sorting the receptor complex in the local dendritic secretory pathway in neurons. These data suggest a new role for ER retention signals in trafficking integral membrane proteins in neurons.SignificanceWe present evidence that the ER retention signals in the kainate receptors containing GluK5 impose a requirement for sorting into local dendritic secretory pathways in neurons, as opposed to traversing the somatic Golgi apparatus. There are two ER retention signals in the C-terminus of GluK5. We show that both are blocked by physical association with SAP97 and CASK. The SH3 and GuK domains of SAP97, in the presence of CASK, bind directly to each ER retention signal and form a complex. These results support an entirely new function for ER retention signals in the C-termini of neuronal receptors, such as NMDA and kainate receptors, and define a mechanism for selective entry of receptors into local secretory pathways.  相似文献
