Effects of stimulation of the claustrum and caudate nucleus in the neocortex and various deep brain structures were studied in acute experiments on unanesthetized cats immobilized with tubocurarine. A rhythmic after discharge appeared in neocortical areas 4–7 and 18 (according to Reinoso-Suarez' atlas), and also in the caudate nucleus and various parts of the thalamus. A similar discharge also was observed in the claustrum itself. Diencephalic brain section at the level of the ventral anterior nucleus weakened but did not completely abolish the cortical rhythmic after-discharge in the anterior regions of the neocortex evoked by stimulation of the claustrum. This discharge was completely blocked after sagittal brain section between the claustrum and the rest of the thalamus.I. S. Somatosensory evoked potentials following lesions of the claustrum   总被引:1,自引:0,他引:1  
Ipsi- and contralateral cortical somatosensory evoked potentials (SEP) were recorded following median nerve stimulation in 12 patients with unilateral brain lesions and in 5 healthy subjects. Computed tomographic scans of brain were performed on admission. In all patients with lesions of the claustrum there was absence of SEP contralateral to the side of the lesion and ipsilateral to the stimulated nerve. This phenomenon did not appear in our material following lesions involving other structures e.g. thalamus or somatosensory cortex. Our observations suggest that the claustrum may influence deeply the contralateral somatosensory cortex. This may be due to the fact that a large part of the claustrum is involved in transmission of the sensory information from receptors to the somatosensory cortex.  相似文献

To investigate the role of vestibular afferent input in systemic interactions between cortical areas of the human brain, the dynamics of interregional cortical interactions has been studied in patients with cervical dystonia during their therapy by the removal of the transtympanic chemical vestibular receptor. It has been found that even unilateral vestibular dereception leads to a profound reorganization of systemic interactions between remote cortical areas, particularly, the anterior and posterior associative areas in both hemispheres. Relationships indicating the role of vestibular input in the organization of integrative brain activity have been found, which confirms its systemic role.  相似文献

The dynamics of spontaneous electrical activity of the rostral, dorsal, ventral, and caudal parts of the claustrum were studied in chronic experiments on 12 dogs with implanted electrodes during extinction of orienting reactions and during biologically meaningful changes in the animals' functional state. In the animal at rest, electrical activity of the claustrum was similar to that of subcortical structures, but in a state of attention, its activity began to resemble that of the electrocorticogram, in agreement with the view that this nucleus occupies an intermediate position between cortical and subcortical structures. Enhancement of fast activity in the claustrum with the appearance of orienting reactions was expressed more intensively, and during extinction it was preserved for a longer time, in the dorsal and ventral parts of the claustrum than in the rostral and caudal parts, evidence of structural heterogeneity of the nucleus. Under conditions of increased food excitability the amplitude and frequency of the fast waves recorded from the rostral and caudal parts of the claustrum were selectively increased, whereas during reflexes to a rejected stimulus (acids) and in defensive reflexes they were increased in recordings from the dorsal and ventral parts, on which basis the existence of specialized efferent zones participating in biologically different types of activity can be postulated. Enhancement of fast activity in the claustrum during reflexes to food and acid differed in its genesis: The switch from the first to the second took place through a phase of suppression of the first. Differentiation of a food-conditioned stimulus was accompanied by the development of active inhibition in the "food zones" of the claustrum and by enhancement of activity in the "zones of orienting reactions."A. The number of different cortical structures in mammalian brains and the number of extrinsic fibres linking these regions are both large. As with any complex system, systematic analysis is required to draw reliable conclusions about the organization of the complex neural networks comprising these numerous elements. One aspect of organization that has long been suspected is that cortical networks are organized into 'streams' or 'systems'. Here we report computational analyses capable of showing whether clusters of strongly interconnected areas are aspects of the global organization of cortical systems in macaque and cat. We used two different approaches to analyse compilations of corticocortical connection data from the macaque and the cat. The first approach, optimal set analysis, employed an explicit definition of a neural 'system' or 'stream', which was based on differential connectivity. We defined a two-component cost function that described the cost of the global cluster arrangement of areas in terms of the areas' connectivity within and between candidate clusters. Optimal cluster arrangements of cortical areas were then selected computationally from the very many possible arrangements, using an evolutionary optimization algorithm. The second approach, non-parametric cluster analysis (NPCA), grouped cortical areas on the basis of their proximity in multidimensional scaling representations. We used non-metric multidimensional scaling to represent the cortical connectivity structures metrically in two and five dimensions. NPCA then analysed these representations to determine the nature of the clusters for a wide range of different cluster shape parameters. The results from both approaches largely agreed. They showed that macaque and cat cortices are organized into densely intra-connected clusters of areas, and identified the constituent members of the clusters. These clusters reflected functionally specialized sets of cortical areas, suggesting that structure and function are closely linked at this gross, systems level.  相似文献

In this paper, we review evidence from comparative studies of primate cortical organization, highlighting recent findings and hypotheses that may help us to understand the rules governing evolutionary changes of the cortical map and the process of formation of areas during development. We argue that clear unequivocal views of cortical areas and their homologies are more likely to emerge for "core" fields, including the primary sensory areas, which are specified early in development by precise molecular identification steps. In primates, the middle temporal area is probably one of these primordial cortical fields. Areas that form at progressively later stages of development correspond to progressively more recent evolutionary events, their development being less firmly anchored in molecular specification. The certainty with which areal boundaries can be delimited, and likely homologies can be assigned, becomes increasingly blurred in parallel with this evolutionary/developmental sequence. For example, while current concepts for the definition of cortical areas have been vindicated in allowing a clarification of the organization of the New World monkey "third tier" visual cortex (the third and dorsomedial areas, V3 and DM), our analyses suggest that more flexible mapping criteria may be needed to unravel the organization of higher-order visual association and polysensory areas.  相似文献

Reelin binds alpha3beta1 integrin and inhibits neuronal migration   总被引:23,自引:0,他引:23  
Mice that are mutant for Reelin or Dab1, or doubly mutant for the VLDL receptor (VLDLR) and ApoE receptor 2 (ApoER2), show disorders of cerebral cortical lamination. How Reelin and its receptors regulate laminar organization of cerebral cortex is unknown. We show that Reelin inhibits migration of cortical neurons and enables detachment of neurons from radial glia. Recombinant and native Reelin associate with alpha3beta1 integrin, which regulates neuron-glia interactions and is required to achieve proper laminar organization. The effect of Reelin on cortical neuronal migration in vitro and in vivo depends on interactions between Reelin and alpha3beta1 integrin. Absence of alpha3beta1 leads to a reduction of Dab1, a signaling protein acting downstream of Reelin. Thus, Reelin may arrest neuronal migration and promote normal cortical lamination by binding alpha3beta1 integrin and modulating integrin-mediated cellular adhesion.  相似文献

The claustrum is a complex telencephalic structure owing to its reciprocal connectivity with most—if not all—cortical areas. However, there is a paucity of data in the literature concerning its histochemical components, including opioid peptide neurotransmitters. The aim of the present study was to examine the morphology, distribution and ultrastructure of leucine-enkephalin-immunoreactive (Leu-enk-ir) neurons and fibers in the dorsal claustrum (DC) of the cat. Seven healthy, adult male and female cats were used in our study. All animals received humane care. They were irreversibly anesthetized and transcardially perfused with fixative. Brains were removed, postfixed, blocked and sectioned. Sections were incubated with polyclonal anti-Leu-enk antibodies using the Avidin–Biotin–Peroxidase Complex method. Leu-enk-ir neurons and fibers were distributed throughout the DC. Some of the neurons were lightly-stained, while others were darkly-stained. Light-microscopically, they varied in shape: oval, fusiform, multipolar and irregular. With regard to size, they were categorized as small (15?μm or less in diameter), medium (16–20?μm in diameter) and large (21?μm or more in diameter). No specific pattern of regional distribution was found. On the electron microscope level, immunoproduct was observed in neurons, dendrites and terminal boutons. Different types of Leu-enk-ir neurons differ in their ultrastructural features, including two types of synaptic boutons. No gender-specific features were observed. In conclusion, it is our hope that our study will serve to contribute to a better understanding of the functional neuroanatomy of the DC in the cat, and that it can be extrapolated and applied to other mammals, including humans.  相似文献

Proteins of the ERM family (ezrin, moesin, radixin) play a fundamental role in tethering the membrane to the cellular actin cortex as well as regulating cortical organization and mechanics. Overexpression of dominant inactive forms of ezrin leads to fragilization of the membrane-cortex link and depletion of moesin results in softer cortices that disrupt spindle orientation during cytokinesis. Therefore, the kinetics of association of ERM proteins with the cortex likely influence the timescale of cortical signaling events and the dynamics of membrane interfacing to the cortex. However, little is known about ERM protein turnover at the membrane-cortex interface. Here, we examined cortical ezrin dynamics using fluorescence recovery after photobleaching experiments and single-molecule imaging. Using multiexponential fitting of fluorescence recovery curves, we showed that ezrin turnover resulted from three molecular mechanisms acting on very different timescales. The fastest turnover process was due to association/dissociation from the F-actin cortex, suggesting that ezrin acts as a link that leads to low friction between the membrane and the cortex. The second turnover process resulted from association/dissociation of ezrin from the membrane and the slowest turnover process resulted from the slow diffusion of ezrin in the plane of the membrane. In summary, ezrin-mediated membrane-cortex tethering resulted from long-lived interactions with the membrane via the FERM domain coupled with shorter-lived interactions with the cortex. The slow diffusion of membranous ezrin and its interaction partners relative to the cortex signified that signals emanating from membrane-associated ezrin may locally act to modulate cortical organization and contractility.  相似文献

The pattern of myelination over the cerebral cortex, termed myeloarchitecture, is an established and often-used feature to visualize cortical organization with histology in a variety of primate species. In this paper, we use in vivo magnetic resonance imaging (MRI) and advanced image processing using surface rendering to visualize and characterize myeloarchitecture in a small nonhuman primate, the common marmoset (Callithrix jacchus). Through images made in four female adult marmosets, we produce a representative 3D map of marmoset myeloarchitecture and flatten and annotate this map to show the location and extent of a variety of major areas of the cortex, including the primary visual, auditory, and somatosensory areas. By treating our MRI data as a surface, we can measure the surface area of cortical areas, and we present these measurements here to summarize cortical organization in the marmoset.  相似文献

The tracts between cortical areas are conceived as playing a central role in cortical information processing, but their actual numbers have never been determined in humans. Here, we estimate the absolute number of axons linking cortical areas from a whole-cortex diffusion MRI (dMRI) connectome, calibrated using the histologically measured callosal fiber density. Median connectivity is estimated as approximately 6,200 axons between cortical areas within hemisphere and approximately 1,300 axons interhemispherically, with axons connecting functionally related areas surprisingly sparse. For example, we estimate that <5% of the axons in the trunk of the arcuate and superior longitudinal fasciculi connect Wernicke’s and Broca’s areas. These results suggest that detailed information is transmitted between cortical areas either via linkage of the dense local connections or via rare, extraordinarily privileged long-range connections.

Using data from Human Connectome Project to estimate the absolute number of axons linking cortical areas yields surprisingly sparse connectivity; reconciling large-scale functional synchronization with sparse anatomical connectivity presents a challenge for our present understanding of human brain organization.  相似文献

The event-related potentials (ERPs) in visual discrimination task in parietal and temporal cortical areas were recorded in 11 young adults during passive observation (involuntary attention) and target selection (voluntary attention). The voluntary selective attention resulted in: 1) increased ERP correlation between the parietal; and temporal cortical areas; 2) increased correlation of sequential ERPs in monopolar leads (P3, P4, T3, T4, T5, T6); and 3) increased correlation of sequential ERPs in bipolar leads (P3-T3, P3-T5, P4-T4, P4-T6). The findings suggest that voluntary attention maintains a concordant activity of the parietal and temporal cortical areas in execution of visual selection tasks.  相似文献

Using digital images of stained sections, we conducted a comparative survey of the visible gross morphology of the insular cortex and the claustrum in 26 mammalian species, representing most of the major mammalian radiations. We observed several features that are conserved in the mammals that we examined, including the absence of expansion of the insular cortex into out-branching lobes, the adherence of the claustrum to the fundus of the rhinal sulcus and to endopiriform cellular regions, and several features that varied, including the colocation of the insular and claustrocortex, the cortical origin of the operculations that cover the insula, the locations of large accumulations of claustral cells, and the presence of thin extensions of groups of claustral cells. We suggest that the final shape of the claustrum is highly influenced by the expansion of two medially adjacent subcortical structures, the striatum and amygdala. We further suggest that the neighboring locations of the claustrum and insula may be due to a dependence of each of these structures upon proximity to olfactory cortex, rather than to any connectional relationships between them.  相似文献

Aging is accompanied by substantial changes in brain function, including functional reorganization of large-scale brain networks. Such differences in network architecture have been reported both at rest and during cognitive task performance, but an open question is whether these age-related differences show task-dependent effects or represent only task-independent changes attributable to a common factor (i.e., underlying physiological decline). To address this question, we used graph theoretic analysis to construct weighted cortical functional networks from hemodynamic (functional MRI) responses in 12 younger and 12 older adults during a speech perception task performed in both quiet and noisy listening conditions. Functional networks were constructed for each subject and listening condition based on inter-regional correlations of the fMRI signal among 66 cortical regions, and network measures of global and local efficiency were computed. Across listening conditions, older adult networks showed significantly decreased global (but not local) efficiency relative to younger adults after normalizing measures to surrogate random networks. Although listening condition produced no main effects on whole-cortex network organization, a significant age group x listening condition interaction was observed. Additionally, an exploratory analysis of regional effects uncovered age-related declines in both global and local efficiency concentrated exclusively in auditory areas (bilateral superior and middle temporal cortex), further suggestive of specificity to the speech perception tasks. Global efficiency also correlated positively with mean cortical thickness across all subjects, establishing gross cortical atrophy as a task-independent contributor to age-related differences in functional organization. Together, our findings provide evidence of age-related disruptions in cortical functional network organization during speech perception tasks, and suggest that although task-independent effects such as cortical atrophy clearly underlie age-related changes in cortical functional organization, age-related differences also demonstrate sensitivity to task domains.  相似文献

Instrumental food-procuring movements were studied in cats before and after unilateral or bilateral ablation of the motor or premotor cortical area. It is shown that unilateral impairment of the motor area affects the strength and accuracy of movements of the contralateral fore-leg, whereas the ablation of the premotor area leads to a slowing down of movements and breaking of a goal-directed movement into separate components. Bilataral ablation of the motor area irreversibly abolished the instrumental reflex. The ablation of the premotor cortex destroyed the animal's reaction to the sound signal, but food-procuring movements of the fore-legs were disturbed only temporarily. The obtained data are discussed on the basis of the concept that in cats the above cortical areas play different roles in the organization of goal directed behaviour.  相似文献

The organization of neocortex in the short-tailed opossum (Monodelphis domestica) was explored with multiunit microelectrode recordings from middle layers of cortex. Microelectrode maps were subsequently related to the chemoarchitecture of flattened cortical preparations, sectioned parallel to the cortical surface and processed for either cytochrome oxidase (CO) or NADPH-diaphorase (NADPHd) histochemistry. The recordings revealed the presence of at least two systematic representations of the contralateral body surface located in a continuous strip of cortex running from the rhinal sulcus to the medial wall. The primary somatosensory area (S1) was located medially while secondary somatosensory cortex (S2) formed a laterally located mirror image of S1. Auditory cortex was located in lateral cortex at the caudal border of S2, and some electrode penetrations in this area responded to both auditory and somatosensory stimulation. Auditory cortex was outlined by a dark oval visible in flattened brain sections. A large primary visual cortex (V1) was located at the caudal pole of cortex, and also consistently corresponded to a large chemoarchitecturally visible oval. Cortex just rostral and lateral to V1 responded to visual stimulation, while bimodal auditory/visual responses were obtained in an area between V1 and somatosensory cortex. The results are compared with brain organization in other marsupials and with placentals and the evolution of cortical areas in mammals is discussed.  相似文献
