The effects of different applied NO3 concentrations onextension growth and final length and area of leaves 1–4of five cereals and six pasture grasses of temperate originwere examined. Increased applied NO3 in the range 0.1–0.5.0mol m–3 caused decreased duration of growth but increasedgrowth rate and final length of leaves 2–4 of the cerealsAvena saliva, Hordeum vulgare, Secale cereale, x Triticosecaleand Triticum aestivum. For all cereals, increased NO3resulted in increased area of leaves 1–4. Pasture grasseswere supplied either 0.5 or 50 mol m–3 NO3. Increasedapplied NO3 (0.5–5.0 mol m–3) resulted indecreased duration of growth and increased growth rate and finalarea of leaves 1–4 of Bromus wiltdenowii, leaves 2–4ofFestuca arundinaceae and leaves 3 and 4 of Lolium muitiflorum.In addition, length of leaves 3 and 4 of B. witidenowii increasedwith increased NO3. Increased NO3 resulted inincreased area of leaves 2–4 of Dactylis gtomerata andLolium perenne and leaves 3 and 4 of Phalaris aquaiica but hadno effect on extension growth of all three species. Lolium temulentum plants were grown at 20 °C, under an 8-hdaylength, in a controlled-environment chamber, and the kineticsof leaf expansion were observed by measuring the movement ofan optical grid attached to the fourth leaf. The leaf emerged23–24 d after sowing and was fully expanded 9–10d later. Extension rate was maximal between the second and fifthdays after emergence and declined markedly thereafter. Duringthe rapid growth phase the rate of elongation exhibited a distinctdiurnal rhythm, fluctuating between 1.9 to 2.3 mm h–1in the light period, and 1.3 to 1.7 mm h–1 in the dark.A circadian oscillation with a period of about 27 h was observedin leaves elongating in continuous darkness. When plants weretransferred to 5 °C soon after emergence of the fourth leafthere was an immediate reduction in rate of growth to about22 per cent of the rate at 20 °C: the Q10 for the mean elongationrate in the range 20–5 °C was 3.7. When plants weretransferred from 20 to 2 °C at fourth leaf emergence, meanextension rate declined to less than 5 per cent, correspondingto a Q10 in the range 5–2 °C of more than 300. Furthermore,growth at 2 °C was confined almost entirely to the darkphase of the photoperiod cycle. The responsive tissue was shownto be a small area of expanding leafless than 1.5 cm above theshoot apex and the possible mechanisms underlying low temperatureeffects in this region are discussed. The plastochron index (PI) has been compared with leaf growthand biomass accumulation in young soya bean plants of severalcultivars that were grown in controlled environments with differentirradiance levels and durations, temperatures, and nitrogen(N) regimes. Increasing the photoperiod from 10 to 16 h day–1 increasedthe plastochron rate (PR) and the proportion of axillary growth.Doubling the photosynthetic photon flux density (PPFD) to 1000µmol m–2S–1, increased PR and the proportionof roots to total plant weight, but decreased the proportionof stems plus petioles to total. In a series of experiments,the plants were grown in an 8 h photoperiod at constant temperaturesof 17, 20, 26 or 32 °C. As temperature increased, PR increased,but the duration of leaf expansion decreased. Leaves were largestat 20 and progressively smaller at 26, 32 and 17 °C. Biomasswas greatest for a given PI at 20 °C and decreased in theorder of 26, 32, and 17 °C. The proportion of axillary growthalso was greatest at 20 °C. When plants were grown in a15 h photoperiod at temperatures from 17.1 to 26.6 °C, leafsize continued to increase up to the highest temperature. At17 °C, the PR in the 15 h photoperiod (PPFD 390 µmol;m–2S–1) was about threefold greater than in 8 h(500 µmol m–2 S–1); biomass accumulation perday was about fivefold greater. Increasing N from 3 to 36 mMincreased PR about 10 per cent, altered biomass partitioningamong plant parts, and increased the biomass of the plants.The NO2 form of N markedly stimulated axillary growth as comparedwith the NH4+ form. Environment or cultivar had little influenceon the duration of leaf expansion in terms of PI. Cultivarsdid not differ consistently in biomass production and allocationin the different environments. Acclimation of single plants of Lolium temulentum to changing[CO2] was studied on plants grown in controlled environmentsat 20°C with an 8 h photoperiod. In the first experimentplants were grown at 135 µ;mol m–2 s–1 photosyntheticphoton flux density (PPFD) at 415µl l–1 or 550µll–1 [CO2] with some plants transferred from the lowerto the higher [CO2] at emergence of leaf 4. In the second experimentplants were grown at 135 and 500 µmol m–2 s–1PPFD at 345 and 575 µl l–1 [CO2]. High [CO2] during growth had little effect on stomatal density,total soluble proteins, chlorophyll a content, amount of Rubiscoor cytochrome f. However, increasing [CO2] during measurementincreased photosynthetic rates, particularly in high light.Plants grown in the higher [CO2] had greater leaf extension,leaf and plant growth rates in low but not in high light. Theresults are discussed in relation to the limitation of growthby sink capacity and the modifications in the plant which allowthe storage of extra assimilates at high [CO2]. Lockyer, D. R. and Whitehead, D. C. 1986. The uptake of gaseousammonia by the leaves of Italian ryegrass.—J. exp. Bot.37: 919–927. Plants of Italian ryegrass (Lolium multiflorum Lam.) grown insoil with two rates of added 15N-labelled nitrate were exposed,in chambers, for 40 d to NH3 in the air at concentrations of16, 118 and 520 µg m–3. At the highest concentrationof NH3, this source provided 47?3% of the total nitrogen inplants grown with the lower rate of nitrate addition (100mgN kg–1 dry soil) and 35?2% with the higher rate (200mgN kg–1 dry soil) At the intermediate concentration ofNH3, the contributions to total plant N were 19?6% and 10?8%,respectively, at low and high nitrate while, at the lowest concentrationof NH3, they were 5?1% and 32%. Most of the N derived from theNH3 remained in the leaves, but some was transported to theroots. The amount of N derived from the NH3 that was presentin the leaves was not reduced by washing the leaves in waterat pH 5?0 before harvesting, indicating that the N was assimilatedby the plant and not adsorbed superficially. Rates of uptakeof NH3 per unit leaf area ranged from 1?7 µg dm–2h–1 at a concentration of 16 µg m–3 to 29?0µg dm–2 h–1 at a concentration of 520 µgm–3 and with the lower rate of nitrate addition. Increasingthe supply of nitrate to the roots slightly reduced the rateof uptake of NH3 per unit leaf area. Effects of kinetin (K), gibberellin A3 (GA3), and 2-(chloroethyl)-trimethylammoniumchloride (CCC) on levels of alanine aminotransferase (GPT) andrates of protein synthesis were studied with both intact plantsand isolated leaf segments of Lolium temulentum L. In intactplants CCC stimulated and CA3 reduced GPT activity, the effectsbsing much greater in 8.h than in 16-h photoporiods. CCC showedmaximum stimulatory effects at 10–2 M and K at 5 x 105M. No effect of GA3 could be demonstrated with concentrationsup to 10–4M. Both K and CCC retarded GPT decline in leafsections, the latter without associated effects upon pigmentbreakdown. Cycloheximide was highly effective in reducing proteinsynthesis in leaf sections. A close correlation between rateof protein synthesis and GPT activity was found over an inhibitorconcentration range from 10–6 to 10–4 M. The resultsare discussed in terms of possible methods of in vivo regulationof GPT activity.  相似文献

The influence of theconcentration of gaseous ammonia on its uptake by the leavesof Italian ryegrass, with and without an adequate supply ofnitrogen to the roots.—J. exp. Bot. 38: 818–827. Plants of Italian ryegrass (Lolium multiflorum Lam.) were grownin pots of soil with two rates of 15N-labclled nitrate, oneproviding adequate, and the other less than adequate, N formaximum growth. After 25 d in a controlled environment cabinet,the plants were transferred to chambers and exposed for 33 dto NH3in the air at one of nine concentrations ranging from14 to 709 µg NH3 m–3. Increasing the concentrationof NH3 in the air increased the dry weight of the shoots ofplants grown at the lower but not the higher rate of nitrate.The content of total N in the plant shoots (% dry weight) waslinearly related to NH3 concentration; at 709 µg NH3 andin both sets of plants it was more than double the content at14 µg NH3 m–3. Calculations, based on 15N enrichment,indicated that the amount of N taken up from the NH3 per unitleaf area increased linearly with increasing concentration ofNH3 in the air uptake (µg dm–2 h–1) = 0.1009xat the lower rate of nitrate and 0-0829x at the higher rateof nitrate, where x is the concentration of NH3 in the air expressedas µg NH3m–3. The proportion of the total plant N that was derived from theNH3 ranged from 4?0% at a concentration of 14 µg NH3 m–3with the higher rate of nitrate addition to 77?5% at a concentrationof 709 µg m–3 with the lower rate of nitrate addition.The proportions of the total N in the water-insoluble proteinof the leaf tissue that were derived from nitrate and gaseousNH3 were similar to the proportions in the whole leaf material. Two approaches to quantifying relationships between nutrientsupply and plant growth were compared with respect to growth,partitioning, uptake and assimilation of NO3 by non-nodulatedpea (Pisum sativum L. cv. Marma). Plants grown in flowing solutionculture were supplied with NO3 at relative addition rates(RAR) of 0·03, 0·06, 0·12, and 0·18d–1, or constant external concentrations ([NO3)of 3, 10, 20, and 100 mmol m–3 over 19 d. Following acclimation,relative growth rates (RGR)approached the corresponding RARbetween 0·03–0.12 d-1, although growth was notlimited by N supply at RAR =0.18 d-1. Growth rates showed littlechange with [NO3–] between 10–100 mmol m–3(RGR=0·15 –0·16 d-1). The absence of growthlimitation over this range was suggested by high unit absorptionrates of NO3, accumulation of NO3 in tissues andprogressive increases in shoot: root ratio. Rates of net uptakeof NO3 from 1 mol m–3 solutions were assessed relativeto the growth-related requirement for NO3, showing thatthe relative uptake capacity increased with RGR between 0·03–0·06d–1 , but decreased thereafter to a theoretical minimumvalue at RGR  相似文献

Net photosynthesis rate (Pn), stomatal conductance to CO2 andresidual conductance to CO2 were measured in the last six leaves(the sixth or flag leaf and the preceding five leaves) of Triticumaestivum L. cv. Kolibri plants grown in Mediterranean conditions.Recently fully expanded leaves of well-watered plants were alwaysused. Measurements were made at saturating photosynthetic photonflux density, and at ambient CO2 and O2 levels. The specificleaf area, total organic nitrogen content, some anatomical characteristics,and other parameters, were measured on the same leaves usedfor gas exchange experiments. A progressive xeromorphic adaptation in the leaf structure wasobserved with increasing leaf insertion levels. Furthermore,mesophyll cell volume per unit leaf area (Vmes/A) decreasedby 52·6% from the first leaf to the flag leaf. Mesophyllcell area per unit leaf area also decreased, but only by 24·5%.However, nitrogen content per unit mesophyll cell volume increasedby 50·6% from the first leaf to the flag leaf. This increasecould be associated to an observed higher number of chloroplastcross-sections per mm2 of mesophyll cell cross-sectional areain the flag leaf: values of 23000 in the first leaf and 48000in the flag leaf were obtained. Pn per unit leaf area remainedfairly constant at the different insertion levels: values of33·83±0·93 mg dm–2 h–1 and32·32±1·61 mg dm–2 h–1 wereobtained for the first leaf and the flag leaf, respectively.Residual conductance, however, decreased by 18·2% fromthe first leaf to the flag leaf. Stomatal conductance increasedby 41·7%. The steadiness in Pn per unit leaf area across the leaf insertionlevels could be mainly accounted for by an opposing effect betweena decrease in Vmes/A and a more closely packed arrangement ofphotosynthetic apparatus. The effects of a range of applied nitrate (NO3) concentrations(0–20 mol m3) on germination and emergence percentageof Triticum aestivum L. cv. Otane were examined at 30, 60, 90and 120 mm sowing depths. Germination percentage was not affectedby either sowing depth or applied NO3 concentration whereasemergence percentage decreased with increased sowing depth regardlessof applied NO3 concentration. Nitrate did not affectemergence percentage at 30 mm sowing depth, but at 60 to 120mm depth, emergence percentage decreased sharply with an increasedapplied NO3 concentration of 0 to 1·0 mol m–3then decreased only slightly with further increases in appliedNO3 of about 5·0 mol m–3. Root and shoot growth, NO3 accumulation and nitrate reductaseactivity (NRA) of plants supplied with 0, 1·0 and 1·0mol m–3 NO3 at a sowing depth of 60 mm were measuredprior to emergence. The coleoptile of all seedlings opened withinthe substrate. Prior to emergence from the substrate, shootextension growth was unaffected by additional NO3 butshoot fr. wt. and dry wt. were both greater at 1·0 and1·0 mol m–3 NO3 than with zero NO3.Root dry wt. was unaffected by NO3. Nitrate concentrationand NRA in root and shoot were always low without NO3.At 1·0 and 10 mol m3 NO3, NO3 accumulatedin the root and shoot to concentrations substantially greaterthan that applied and caused the induction of NRA. Regardlessof the applied NO3 concentration, seedlings which failedto emerge still had substantial seed reserves one month afterplanting. Coleoptile length was substantially less for seedlingswhich did not emerge than for seedlings which emerged, but wasnot affected by NO3. It is proposed that (a) decreasedemergence percentage with increased sowing depth was due tothe emergence of leaf I from the coleoptile within the substrateand (b) decreased emergence percentage with additional NO3was due to the increased expansion of leaf 1 within the substrateresulting in greater folding and damage of the leaf. Changes in carbon fixation rate and the levels of photosyntheticproteins were measured in fourth leaves of Lolium temulentumgrown until full expansion at 360 µmol quanta m–2s–1 and subsequently at the same irradiance or shadedto 90 µmol m–2 s–1. Ribulose-1,5-bisphosphatecarboxylase/oxygenase (Rubisco), light-harvesting chlorophylla/b protein of photosystem II (LHCII), 65 kDa protein of photosystemI (PSI), cytochrome f (Cytf) and coupling factor 1 (CF1) declinedsteadily in amount throughout senescence in unshaded leaves.In shaded leaves, however, the decrease in LHCII and the 65kDa protein was delayed until later in senescence whereas theamount of Cyt f protein decreased rapidly following transferto shade and was lower than that of unshaded leaves at the earlyand middle stages of senescence. Decreases in the Rubisco andCF1 of shaded leaves occurred at slightly reduced rates comparedwith unshaded leaves. These results indicate that chloroplastproteins in fully-expanded leaves are controlled individually,in a direction appropriate to acclimate photosynthesis to agiven irradiance during senescence. Plants of the C4 sedge Cyperus longus L. were grown at 10, 20and 30 °C. An asymptotic growth curve, the Richards function,was fitted to growth data for successive leaves. The mean rateof leaf appearance was a linear function of temperature with0.014 leaves appearing per day for every 1 °C increase intemperature. The instantaneous relative rate of leaf extensionshowed a marked ontogenetic drift which was most rapid at 30°C and slowest at 10 °C. The mean absolute extensionrate for foliage had a temperature coefficient of 0.16 cm d–1° C–1 in the range from 10 to 30 °C. The durationof leaf growth was independent of leaf number at 10 and 20 °Cbut increased linearly with leaf number at 30 °C. The smalldifferences in relative growth rate at the three temperaturesresulted in large differences in foliage area produced at theend of a 30 d growth period. This study was conducted with Lolium temulentum, Festuca pratensis,and the two hybrids L. multiflorum x F. pratensis ‘Elmet’and L. perenne x F. pratensis ‘Prior’. In a comparisonof various durations (7–42 d) of pretreatment at 4 or7 °C the highest yield of microspore-derived callus of L.temulentum was obtained after pretreatment of spikes at 7 °Cfor 28 d, conditions which also proved optimal for panicle pretreatmentwith F. pratensis. For ‘Elmet’, durations of 21–42d were optimal, and for ‘Prior’ the responses tendedto decline with increasing duration. The photosynthetic capacity of newly expanded leaves of vernalizedor non-vernalized plants of S24 perennial ryegrass (Lolium perenneL.), grown in long or short photoperiods, was measured in twoexperiments. In the first, leaves were protected from shadingduring development, while in the second, the natural shade ofneighbouring tillers in a sward was allowed. In the first experiment there was little effect of vernalization,day length or flowering, and leaves in all treatments had photosyntheticrates at 250 W m–2 of between 28 and 32 mg CO2 dm–2h–1.In the second experiment the photosynthetic rate of successiveleaves fell as sward leaf area increased. This downward trendwas reversed, however, in flowering tillers in the vernalizedlong-day treatment, while in the other treatments, which didnot flower, photosynthetic capacity continued to fall. It isconcluded that the leaves of reproductive tillers have highphotosynthetic capacities because stem extension carries themto the top of the canopy where they are well illuminated duringexpansion. Barley (Hordeum vulgare L., cvs Golf, Mette, and Laevigatum)was grown under nitrogen limitation in solution culture untilnear maturity. Three different nitrogen addition regimes wereused: in the ‘HN’ culture the relative rate of nitrate-Naddition (RA) was 0·08 d–1 until day 48 and thendecreased stepwise to, finally, 0·005 d–1 duringgrain-filling; the ‘LN’ culture received 45% ofthe nitrogen added in HN; the ‘CN’ culture was maintainedat RA 0·0375 d–1 throughout. Kinetics of net nitrateuptake were measured during ontogeny at 30 to 150 mmol m–3external nitrate. Vmax (which is argued to reflect the maximuminflux rate in these plants) declined with age in both HN andLN cultures. A pronounced transient drop was observed just beforeanthesis, which correlated in time with a peak in root nitrateconcentration. Similar, but less pronounced, trends were observedin CN. The relative Vmax (unit nitrogen taken up per unit nitrogenin plants and day) in all three cultures declined from 1·3–2·3d–1 during vegetative growth to 0·1–0·7d–1 during generative growth. These values are in HN andLN cultures 15- to more than 100-fold in excess of the demandset by growth rates throughout ontogeny. Predicted balancingnitrate concentrations (defined as the nitrate concentrationrequired to support the observed rate of growth) were below6·0 mmol m–3 in HN and LN cultures before anthesisand then decreased during ontogeny. In CN cultures the balancingnitrate concentration increased during grain-filling. Apartfrom the transient decline during anthesis, most of the effectof ageing on relative Vmax can be explained in terms of reducedcontribution of roots to total biomass (R:T). The loss in uptakeper unit root weight is largely compensated for by the declinewith time in average tissue nitrogen concentrations. The quantitativerelationships between relative Vmax and R:T in ageing plantsare similar to those observed for vegetative plants culturedat different RAs. The data support the contention that the capacity for nitrateacquisition in N-limited plants is under general growth control,rather than controlled by specific regulation of the biochemicalpathway of nitrate assimilation. The possible role in drought resistance played by sclerophyllywas studied in the Mediterranean oaks Quercus ilex, Q. suberand Q. pubescens. Studies were conducted on leaves at 30, 50and 80% of their final surface area, as well as on mature leavesof the current year's growth in June and September and on 1-year-oldleaves. Leaves of different ages of the three species showed quite differentdegrees of sclerophylly (DS). Q. ilex leaves reached the definitiveDS of 1.75 g dm–2 during leaf expansion; Q. pubescensleaves hardened at the end of their expansion, with a finalDS of 0.93 g dm–2; Q. suber showed the lowest DS of 0.76g dm–2. Leaf conductance to water vapour (g1) of 1-year-old leaves ofQ. ilex, measured in the field, showed a duration of the g1peak values about twice that of the other two species. The minimumleaf relative water content (RWC), however, was near the samein the three species, indicating that water loss was recoveredpartly by Q. ilex leaves. Diurnal temperature fluctuations induced change in soya bean-pod[Glycine max (L.) Merr.] carbon exchange rate (CER, where positiveCER represents CO2 evolution). CER appeared to depend linearlyon temperature. Linear regressions of CER on temperature interceptedthe temperature axis at 5°C (i.e. zero CER at 5°C).Slopes of these regressions (i.e. temperature sensitivity) changedover the season. Long-term effects of atmospheric carbon dioxide concentration(ambient or 700 µmol mol–1) and air temperature(simulation of field conditions or + 4 C) on leaf photosyntheticrate were examined in Lolium perenne L. cv. Vigor, exposed tonatural illumination during winter. Photosynthetic capacitywas compared over a range of air temperatures and photon fluxdensities of photosynthetically active radiation which wererepresentative of winter climate (5–15 C and 0–500µmol m–2s–1), with CO2 level during measurementsimilar to that during the experimental period. Long-term exposureto increased air temperature reduced leaf CO2 fixation capacityby 23% (averaged over all measurement conditions), resultingfrom a decline in lightsaturated uptake rate, but not in incident-lightquantum efficiency. CO2- stimulation was largely absent in plantsgrown in ambient temperature, but pronounced in plants grownunder +4 C, where it compensated for two-thirds of the 23%drop. This enhancing effect of elevated CO2 level on leaf CO2uptake rate observed in the warmer treatment, was strongly dependenton measurement temperature, increasing from 5% at 5 C, to upto 32% at 15 C. Measurements of chlorophyll fluorescence anddry matter corresponded with the observed changes in assimilationcapacity, which could not be attributed to a deteriorated nitrogenstatus of the leaves as there was a similar N content on anarea basis.
