The fine structure of rhabdomeric photoreceptors of Cirrifera aculeuta, Monocelis fusca and Nematoplana coelogynoporoides is described. The existence of one receptor cell and one pigmented or unpigmented mantle cell is thought to be a basic feature of the Neoophora at least. The existence of a pigment shield formed by many cells is derived, in addition to one receptor and one unpigmented mantle cell, in the eyes of Monocelis fuscu.  相似文献

It is known that the bile canaliculus in the liver of almost all vertebrates is made up of membranes of two or more adjacent liver cells. Studying the liver cell ultrastructure of lasting and fed grass carp and silver carp, it was demonstrated that a bile canaliculus is formed by deep invagination of a cell membrane of one hepatocyte. The membrane forms microvilli along the bile canaliculus. The bile canaliculus is seen in the centre of liver cell cytoplasm on the cross section and stretches from the centre of the liver cell cytoplasm to the cell membrane on the longitudinal section. The bile canaliculus is connected with a small duct cell, which is distinct from a liver cell in its small size, little amount of cell organelles and the presence of cytoplasmic filaments. The terminal part of the biliary tract consists of one liver cell and one bile duct cell. The part of the tract adjacent to the terminal one is composed of two or three small bile duct cells devoid of basal membrane. Thus, the liver parenchyma is constituted of a net of numerous bile ducts. In the portal tract, there is a large bile duct, consisting of 12-13 bile duct cells, surrounded by basal membrane and connective tissue cells.  相似文献

Human papillomavirus (HPV) infection is one of the risk factors contributing to the pathogenesis of lung cancer. The aim of the study was to determine the presence of HPV in non-small cell carcinomas of the lung. The study included 40 tumors: 22 squamous cell carcinomas, 13 adenocarcinomas and 5 large cell carcinomas. HPV was found in 4 cases (10%). High risk HPV was present in 3 tumors: in one squamous cell carcinoma, one large cell carcinoma and one adenocarcinoma, while low risk HPV was detected in one adenocarcinoma.  相似文献

不对称性细胞分裂是一个母细胞通过一次分裂,产生两个不同命运的子细胞的分裂方式,是单细胞生物向多细胞生物进化的关键一步。根据现有的证据推论,不称性细胞分裂是在器官发育过程中产生细胞多样化的一种基本方式。Numb是第一个被发现决定多细胞生物不对称细胞分裂的信号蛋白。在果蝇中,Numb通过促进Notch泛素化拮抗Notch信号通路,从而决定子细胞的命运,后来的研究表明Numb是细胞内吞调节蛋白,并用通过内吞参与调节神经细胞的粘附,轴突的生长及细胞迁移等过程;并且发现Numb与肿瘤抑制基因p53、泛素化蛋白HDM2形成三聚体抑制p53的泛素化,从而调节肿瘤的恶性程度。本文系统地分析了Numb发现的历史及后来在脊椎动物中的作用和机制,重点介绍了Numb在神经发育过程中的功能。  相似文献

The cell reproduction cycle of parasitic wall-free bacteria, mycoplasma, is reviewed. DNA replication of Mycoplasma capricolum starts at a fixed site neighboring the dnaA gene and proceeds to both directions after a short arrest in one direction. The initiation frequency fits to the slow speed of replication fork and DNA content is set constant. The replicated chromosomes migrate to one and three quarters of cell length before cell division to ensure delivery of the replicated DNA to daughter cells. The cell reproduction is based on binary fission but a branch is formed when DNA replication is inhibited. Mycoplasma pneumoniae has a terminal structure, designated as an attachment organelle, responsible for both host cell adhesion and gliding motility. Behavior of the organelle in a cell implies coupling of organelle formation to the cell reproduction cycle. Several proteins coded in three operons are delivered sequentially to a position neighboring the previous organelle and a nascent one is formed. One of the duplicated attachment organelles migrates to the opposite pole of the cell before cell division. It is becoming clear that mycoplasmas have specialized cell reproduction cycles adapted to the limited genome information and parasitic life.  相似文献

We have located a novel carbohydrate epitope in the cell walls of certain single cells in embryogenic, but not in non-embryogenic, suspension cultures of carrot. Expression of this epitope, recognized by the mAb JIM8, is regulated during initiation, proliferation, and prolonged growth of suspension cultures such that changes in the abundance of JIM8-reactive cells always precede equivalent changes in embryogenic potential. Therefore, a direct correlation exists between the presence of the JIM8-reactive cell wall epitope and somatic embryo formation. The JIM8-reactive cell wall epitope is expressed in the cell walls of three types of single cells and one type of cell cluster. One of the single cell types seems able to follow one of two phytohormone-controlled developmental pathways, either a cell elongation pathway that eventually leads to cell death, or a cell division pathway that gives rise to proembryogenic masses. We demonstrate that all JIM8-reactive cell types in embryogenic carrot suspension cultures are developmentally related, and that the switch by one of them to somatic embryogenesis is accompanied by the immediate dissipation of the JIM8-reactive cell wall epitope. The cell wall carbohydrate epitope recognized by JIM8 therefore represents a cell wall marker for a very early transitional cell state in the developmental pathway to carrot somatic embryogenesis.  相似文献

Megasporogenesis in 16 species of Papilionaceae has been investigated. Archesporial development in Papilionaceae is usually hypodermal, but subhypodermal origin has been observed. The archesporial cell develops directly into the megasporocyte, or divides to produce a parietal cell and a primary sporogenous cell. The parietal cell is always oriented toward the micropylar end of the developing ovule. It may or may not divide to contribute to the nucellar mass as the primary sporogenous cell enlarges to form the megasporocyte. A single megasporocyte is produced in most species investigated. Of those species that form more than one megasporocyte, few develop more than one tetrad of megaspores, and in no case is more than one megagametophyte produced. Meiosis occurs in the megasporocyte to form a tetrad of megaspores. Several variations in tetrad patterns are explored here. Monosporic development is the prevalent condition in Papilionaceae; however, bisporic development does occur.  相似文献

Although gemcitabine is the most effective chemotherapeutic agent against pancreatic cancer, a growing concern is that a substantial number of patients acquire gemcitabine chemoresistance. To elucidate the mechanisms of acquisition of gemcitabine resistance, we developed gemcitabine-resistant cell lines from six human cancer cell lines; three pancreatic, one gastric, one colon, and one bile duct cancer. We first analyzed gemcitabine uptake using three paired parental and gemcitabine resistant pancreatic cancer cell lines (PK-1 and RPK-1, PK-9 and RPK-9, PK-59 and RPK-59) and found that uptake of gemcitabine was rapid. However, no DNA damage was induced in resistant cells. We further examined the microarray-based expression profiles of the cells to identify genes associated with gemcitabine resistance and found a remarkable reduction in the expression of deoxycytidine kinase (DCK). DCK is a key enzyme that activates gemcitabine by phosphorylation. Genetic alterations and expression of DCK were studied in these paired parental and derived gemcitabine-resistant cell lines, and inactivating mutations were found only in gemcitabine-resistant cell lines. Furthermore, siRNA-mediated knockdown of DCK in the parental cell lines yielded gemcitabine resistance, and introduction of DCK into gemcitabine-resistant cell lines invariably restored gemcitabine sensitivities. Mutation analyses were expanded to three other different paired cell lines, DLD-1 and RDLD-1 (colon cancer cell line), MKN-28 and RMKN-28 (gastric cancer cell line), and TFK-1 and RTFK -1 (cholangiocarcinoma cell line). We found inactivating mutations in RDLD-1 and RTFK-1 and decreased expression of DCK in RMKN-28. These results indicate that the inactivation of DCK is one of the crucial mechanisms in acquisition of gemcitabine resistance.  相似文献

本文描述延龄草(Trillium tschonoskii Maxim.)的大孢子发生,雌配子体的形成和雄配子体的形态。胚珠为倒生型,双珠被,厚珠心型。胎座为侧膜胎座向中轴胎座的过渡类型,胶囊发育为葱型的变异型。孢原细胞直接发生于幼胚珠的珠心表皮细胞之下,孢原细胞平周分裂,形成初生周缘细胞及初生造孢细胞。初生周缘细胞分裂先于初生造孢细胞,分裂结果与珠心表皮细胞共同形成了珠心组织。初生造孢细胞进一步发育,形成大孢子母细胞。大孢子母细胞经减数第一次分裂后,即出现壁,形成二分体。一般是珠孔端二分体细胞小于合点端二分体细胞,但偶尔也见到前者大于后者的情况。在二分体形成后珠孔端二分体细胞立即退化、或经减数第二次分裂后再退化(该次分裂多为斜向的)。合点端二分体细胞发育,经二核胚囊,四核胚囊,六核胚囊阶段至成熟胚囊。一般在珠孔端的周围淀粉粒丰富,并先于合点端的核进行分裂。珠孔端由二个助细胞,一个卵细胞构成卵器,助细胞具钩突,并具丝状器,两个极核。合点端常见多核仁的大核,成熟胚囊未见八核。成熟花粉粒为二细胞的,花药壁具变形绒毡层,花粉中充满淀粉粒。沼生目型胚乳。  相似文献

The amacrine cells in the retina of the rat are described in Golgi-stained whole-mounted retinae. Nine morphologically distinct types of cell were found: one type of diffuse cell, five types of unistratified cell, two types of bistratified cell, and one type of stratified diffuse cell. Measurements show that the largest unistratified cells have a dendritic field 2 mm across. One type of interplexiform cell is also described. Wide-field diffuse amacrine cells and unistratified amacrine cells were found with their somata located in either the inner nuclear layer or the ganglion cell layer. It is clear that there may be an amacrine cell system in the ganglion cell layer of the rat retina.  相似文献

Asymmetric stem cell division is a mechanism widely employed by the cell to maintain tissue homeostasis, resulting in the production of one stem cell and one differentiating cell. However, asymmetric cell division is not limited to stem cells and is widely observed even in unicellular organisms as well as in cells that make up highly complex tissues. In asymmetric cell division, cells must organize their intracellular components along the axis of asymmetry(sometimes in the context of extracellular architecture). Recent studies have described cell asymmetry in many cell types, and in many cases such asymmetry involves the centrosome (or spindle pole body in yeast) as the center of cytoskeleton organization. In this review, I summarize recent discoveries in cellular polarity that lead to an asymmetric outcome, with a focus on centrosome function.  相似文献

Summary The sensory epithelium of the lateral line organ of the common eel consists of two types of cells, (sensory and supporting). The sensory cell bears a kinocilium together with about 40 to 60 stereocilia on its surface. The kinocilium is situated either at rostral or at caudal margin of this cilial group. Such polarity of the cilial group of one cell is inverse to that of an adjacent cell.Two types of crystal-like inclusions exist in the sensory cells, consisting of granules 100 Å in diameter. Granules in one type are arranged regularly whereas those in the other rather irregularly.Two types of nerve endings exist at the base of sensory cells: one is predominant in number and contains few vesicles, accompanied by a dense spherical body surrounded by small vesicles in the sensory cell and the other is rare in number and contains many vesicles, accompanied by a small flat sac just beneath the plasma membrane of the sensory cell.The supporting cells contain numerous mitochondria, a well developed Golgi apparatus and rough-surfaced endoplasmic reticulum, and surround a sensory cell completely. P. godefroyae is one of the diandrous species of rather primitive orchids. The cytokinesis of PMCs conforms to simultaneous type. The arrangement of microspores in a tetrad is tetrahedral or isobilateral. The first mitosis in a pollen grain is unequal and results in the formation of two unequal cells. The small one is the generative cell and the large one, the vegetative cell. The wall material between them is callose which is easily detectable under the fluorescence microscope. When the generative cell detaches from the microspore wall and migrates into the cytoplasm of the vegetative cell, the callose wall disappears and a thin PAS-positive wall Was observed around the generative cell. The PAS-positive wall remains untill anthesis. The tapetum is of secretory type and its cells are binucleate. With the degradation of the tapetal cells, they discharge a lot of yellow, amorphous, sticky mass into the pollen sac. The pollens distribute in it to form a sticky pollen mass. The ovule has single integument and one layered nucellus around the magaspore mother cell. The mature embryo sac consists of eight or six cells and conforms to the Allium type. The interval between pollination and fertilization is about 45 days and the normal double fertilization has been observed. The primary endosperm cell undergoes one division only and results in the formation of 2 nucleate endosperm. The dormancy period of zygote lasts 45–50 days. During the development of the embryo, a suspensor consisting of a row of two to four cells is formed. It takes more than six months from the pollination to the maturation of the seed. The embryo in the mature seed is just an ellipsoidal mass of 120–140 cells without differentiation. The endosperm and suspensor are all degenerated in the mature seed.  相似文献

Following the origin of multicellularity in many groups of primitive organisms there evolved more than one cell type. It has been assumed that this early differentiation is related to size — the larger the organism the more cell types. Here two very different kinds of organisms are considered: the volvocine algae that become multicellular by growth, and the cellular slime moulds that become multicellular by aggregation. In both cases there are species that have only one cell type and others that have two. It has been possible to show that there is a perfect correlation with size: the forms with two cell types are significantly larger than those with one. Also in both groups there are forms of intermediate size that will vary from one to two cell types depending on the size of the individuals, suggesting a form of quorum sensing. These observations reinforce the view that size plays a critical role in influencing the degree of differentiation.  相似文献

The synthesis of a new family of 7-aza-rebeccamycin analogues in which the sugar moiety is attached to the nitrogen of the pyridine ring is described. The capacity of the newly synthesized compounds to bind to DNA and to inhibit topoisomerase I has been evaluated. Their cytotoxicities toward four tumor cell lines, one murine leukemia L1210 and three human tumor cell lines, one prostate carcinoma DU145, one colon carcinoma HT29, and one non-small cell lung carcinoma A549, have been determined. Their abilities to inhibit the checkpoint kinase Chk1 have been evaluated.  相似文献

Vertebrate somatic cells sometimes form unilateral furrows during cytokinesis that ingress from only one edge of the cell. In some cases after a cell initiates a normal symmetrical circumferential furrow, one of its edges stops furrowing and regresses while the furrow associated with the opposing edge continues across the cell. In cells containing two independent spindles unilateral furrows are sometimes formed that do not follow a linear path but instead sharply change their direction and wander for >40 microm through the cell. These observations reveal that the 'contractile ring' normally seen during cytokinesis is composed of multiple independent 'furrowing units' that are normally coordinated to form a symmetrical furrow around the cell, and that once formed this so-called contractile band does not function as a 'purse string' as commonly envisioned. Individual furrowing units can work independently of one another, and cytokinesis in vertebrates can be consummated by the formation of a single functional furrowing unit in a localized region of the cell cortex that is then propagated across the cell. How this propagation occurs remains an important question for the future.  相似文献

Generative and sperm cells were examined at four stages of development from generative cell formation to sperm cell maturation using serial transmission electron microscopy. The generative cell and vegetative nucleus are associated in a male germ unit association during pollen maturation and tube elongation, except for generative cell mitosis. At late stages of prophase, this association loosens; the generative cell separates from the vegetative nucleus at metaphase. Slender, unbranched, or occasionally branched projections may be found at one or both ends of the generative cell, or they may be single, blunt, and short. Slender projections are rare during anaphase and telophase. The vegetative nucleus moves back into apposition with one sperm cell at the end of mitosis. During the re-establishment of the association, the vegetative nucleus first touches the end of the leading sperm cell and then moves next to the middle of the sperm nucleus. As the sperm cells enter interphase, a conventional association is re-established between one cell and the vegetative nucleus through one or more long and slender cytoplasmic extensions; these associations are maintained throughout later passage in the pollen tube. During maturation, a significant increase occurs in the surface area of the sperm cells (particularly in the sperm cell in association with the vegetative nucleus), and a lesser increase in nuclear volume and surface area. Other sperm cell parameters, including those of heritable organelles, remain unchanged during sperm cell maturation.  相似文献

Division of labour is a marked feature of multicellular organisms. Margulis proposed that the ancestors of metazoans had only one microtubule organizing center (MTOC), so they could not move and divide simultaneously. Selection for simultaneous movement and cell division had driven the division of labour between cells. However, no evidence or explanation for this assumption was provided. Why could the unicellular ancetors not have multiple MTOCs? The gain and loss of three possible strategies are discussed. It was found that the advantage of one or two MTOC per cell is environment-dependent. Unicellular organisms with only one MTOC per cell are favored only in resource-limited environments without strong predatory pressure. If division of labour occurring in a bicellular organism just makes simultaneous movement and cell division possible, the possibility of its fixation by natural selection is very low because a somatic cell performing the function of an MTOC is obviously wasting resources. Evolutionary biologists should search for other selective forces for division of labour in cells.  相似文献

Four counting methods (two flow cytometry, one Coulter principle, one microscopic) are compared for measuring cell density and viability of batch-grown yeast. All gave adequate precision in measuring total cell density with no systematic difference between methods. However, the promise of flow cytometry as a rapid means of determining both quantity and quality of a cell population is shown.  相似文献
