To investigate the diversity of culturable actinobacteria and further screen for microbial pharmaceutics, seven different media were chosen to isolate actinobacteria from 87 soil samples collected from Qinghai–Tibet plateau. A total of 1930 strains was isolated and identified to belong to 11 suborders, i.e., Actinopolysporineae, Corynebacterineae, Frankineae, Glycomycineae, Kineosporiineae, Micrococcineae, Micromonosporineae, Propionibacterineae, Pseudonocardineae, Streptomycineae and Streptosporangineae, and 16 families, i.e., Nocardioidaceae, Actinopolysporaceae, Actinosynnemataceae, Dermacoccaceae, Geodermatophilaceae, Glycomycetaceae, Kineosporiaceae, Microbacteriaceae, Micromonosporaceae, Nocardiaceae, Promicromonosporaceae, Propionibacteriaceae, Pseudonocardiaceae, Streptomycetaceae, Streptosporangiaceae and Thermomonosporaceae. A primary taxonomic study showed that at least 22 genera of actinobacteria were identified from the soil samples, among which ten isolates represented hitherto unknown species. The results showed that there was abundant actinobacterial species diversity in the soil samples from the Qinghai–Tibet plateau.  相似文献

Summary A long-term experiment for increasing the traits first day litter size (LS1) and litter weight (LW1) was conducted with two populations for 33 generations. The selection plateau was reached in population DU-C (selection and estrus synchronization (h2 = 0.02±0.01); in population DU-K (selection) the plateau (h2 = 0.05±0.2) was nearly reached. Selection progress per generation was in LS1 b = 0.11±0.02; b = 0.12±0.04 (1st to 18th generation DU-K, DU-C) and b = 0.10±0.03; b = 0.07±0.05 (19th to 33rd generation, DU-K, DU-C) in LW1 b = 0.16±0.04 g; 0.19±0.07 g (DU-K, DU-C) b = 0.20±0.09 g; 0.001±0.09 g (DU-K, DU-C). Reverse and relaxe selection as well as systematic inbreeding was applied for 10 generations. Reverse selection yielded h2 = 0.28±0.11 (R-DU-K) and h2 = 0.17±0.05 (R-DU-C) and showed that there was still additive genetic variance. Relaxe selection did not cause alterations in the selection parameters, whereas inbreeding lead to inbred depressions (b = LS1 = –0.42±0.15; –0.45±0.12; b = LW1 = –1.13±0.20; –0.82±0.18 I-DU-K, I-DU-C). The plateau was based upon the heterozygote advantage. Several methods for overcoming the plateau were applied. A new selective useful variance could be created by crossing the plateau populations (h2 = 0.14±0.04). A short-term progress in overcoming the plateau (1st to 3rd generation) could be obtained by litter size standardization (LS = 388). Tandem selection (selection for body weight — BW42) as well as crossing of inbred strains were not suitable for overcoming the selection plateau. Altering the environmental conditions as a possibility for overcoming the plateau has been discussed.  相似文献

The birth of agriculture east of the Fertile Crescent, between the Iranian plateau, Central Asia and South Asia, is poorly known. This area is vast and the archaeological data relating to this period, the Neolithic, is extremely limited. Most researchers agree that plants and animals’ domestication was introduced in this area from south-west Asia, similarly to what happened in Europe. In this scenario, agriculture emerged in the Fertile Crescent, between the Levant, Anatolia and the Zagros, 12.000 to 11.000 years ago, and then spread to other regions further east, across the Iranian plateau to Central Asia and the north-western part of the Indian subcontinent, beginning between the 8th and 7th millennia BCE. This article summarizes the main available data relating to this topic with a focus on the Indo-Iranian Borderlands, between southeastern Iran and Pakistan. Currently available paleoclimatic, paleogenetic and archaeological records are presented. A synthesis and discussion is then provided in the conclusion as well as reconstructions based on these records. Issues or challenges relating to the research on the earliest farming villages are mentioned while directions for future research are evoked.  相似文献

In the present study we evaluate the population structure and potential colonization routes of the silverside Chirostoma humboldtianum through approximate Bayesian computations. Six microsatellite loci were amplified in a total of 288 individuals from six different locations covering the complete geographic distribution of the species. Additionally, two mitochondrial DNA markers, a D loop control region and cytochrome b were amplified in a subset of 107 individuals. The results found with microsatellites allow recovering well-structured populations that have experienced a drastic reduction in the effective population size. On the other hand, mtDNA sequences showed a moderate phylogeographic structure with shared haplotypes between geographic localities and signalsof a slight increase in the effective population size. Finally, the approximate Bayesian computation analysis performed with both datasets suggested a west-to-east colonization route for the species in Central Mexico.  相似文献

Using the plateau wood frog Rana kukunoris from the Hengduan Mountains as a model system, we tested whether rivers form significant genetic barriers (the riverine barrier hypothesis) to high elevation amphibians. Samples were collected from eight sites across three major river drainages, the Min, the Dadu and the Yalong Rivers, and the population genetic structure of these frogs was evaluated with data from eight microsatellite DNA loci. A large amount of genetic structure was found, and the pairwise F ST ranged from 0.022 to 0.508 and a global F ST was 0.215. Both analysis of molecular variance and isolation by distance analysis suggested that rivers, mountain ridges and geographic distances all contributed significantly to the population structure. However, no single landscape has prominent barrier effect to the plateau wood frog populations. An assignment analysis using the computer program Structure grouped the eight populations into four population clusters, and no single type of landscape can sufficiently explain the clustering. In conclusion, rivers do not appear to be the leading genetic barriers for the plateau wood frog. The strong population genetic structure is likely the consequence of attributes of the species, as opposed to environmental fragmentation, and the barrier effect of the landscapes is largely swamped by the large amount of intrinsic population structure.  相似文献

Although much research has been carried out on Australopithecus afarensis locomotion, no consensus has yet been reached. Our new critic study on femoral material brings to the fore a strictly bipedal behaviour within this taxon. Our results are based on the pertinence of human anatomical characteristics among A. afarensis and on the absence of characteristics revealing arboreal displacement. These results have emerged from our different observation and interpretation of some preceding authors concerning the anatomy of these fossil hominids. It is important to underline that apomorphic characteristics of this species are difficult to interpret. They must not however be used to support the idea of arboreal displacement simply based on the fact of a no totally human morphe. We believe that present day humans do not necessarily reflect the earliest strict bipedal anatomic model. An the other hand, it appears that the disagreement between the two locomotor hypothesis for A. afarensis that are bipedalism and arboreal displacement, facing the possibility of bipedalism associated with negligible arboreal displacement, results more from an evolutionary fact than from a real scientific conflict.  相似文献

Lilium pumilum DC. is a valuable species not only for its showy flowers but also for its edible and medicinal values. As one of the distribution areas of L. pumilum, Qinghai–Tibet plateau has unique environmental features which have high impact on the evolution of the species. No population genetic studies have been done for L. pumilum so far. To provide the first reference data for evolutionary study and understanding the influence of eco-geographic factors on the distribution of genetic variation in L. pumilum, interspecific simple sequence repeat markers were used to investigate genetic diversity and population structure of 28 populations sampled from southeast of Qinghai–Tibet plateau. Fifteen selected primers generated a total of 147 polymorphic bands. The genetic diversity was low within populations (average He = 0.173), but higher at the species level (He = 0.392). A clear population structure and high level of genetic differentiation (F ST = 0.518) were detected by unweighted pair group method for arithmetic averages, principle coordinate analysis and Bayesian clustering. All clustering approaches supported a division of the 28 populations into 4 major groups for which analysis of molecular variance confirmed a significant variation among groups (34.3 %). These population genetic parameters suggest limited gene flow among populations and evidence for isolation by distance (r = 0.272, P < 0.0001) was found in this study. Altitude, AMT and AMP explained 9.5, 11.5 and 14.0 % of the total variance among populations indicating that eco-geographic factors have a significant effect. Considering the low within-population genetic diversity, high differentiation among populations and the increasing anthropogenic pressure on the species, in situ conservation measures were recommended to preserve L. pumilum in Qinghai–Tibet plateau.  相似文献

From a 332-cm long lacustrine core taken at 60 m water depth and 25 surface sediment samples taken at different water depth sites in Nam Co lake (4,718 m a.s.l.) in the middle-south part of the Tibetan Plateau, we identified nine species of ostracods (Crustacea: Ostracoda) belonging to six genera. Using lithological data, auto-ecological information of the recovered taxa and an ostracod-based transfer function for water depth reconstruction, we distinguished three main environmental stages over the past 8,400 years: during Stage I (8,400–6,800 yr BP), the climate changed from warm-humid to cold-humid, and eventually to cold-arid. The water depth of the site was much lower than today and changed from an estimated 50 to 20–30 m. During Stage II (6,800–2,900 yr BP), environmental conditions were again warm-humid, turning into a cold-arid episode. The lake water depth initially stayed much shallower than today, but then gradually deepened to around 50 m. At the earlier period of Stage III (2,900 yr BP–present), the climate became again warm-humid from cold-arid status. There was a cold-dry event between 1,700 and 1,500 yr BP, which intensified afterward while the surface run off weakened. Early in this stage, lake depth decreased slightly, but then it continuously deepened to 60 m. Our results revealed that central Tibet experienced wavily warm toward tendency in early Holocene, a shift from warm-humid to cold-dry conditions in the middle Holocene, and from warm-humid to cold-dry conditions in the late Holocene. They also show that ostracod assemblages are not only indicative of cold-warm conditions, but are also usable to imply the dry–wet status of a lake area by the inferring water depth variations. Finally, this study provides baseline data on (natural) climate change in this mountain region against which to compare global change impacts.  相似文献

The ability of an ecosystem to retain anthropogenic nitrogen (N) deposition is dependent upon plant and soil sinks for N, the strengths of which may be altered by chronic atmospheric N deposition. Sugar maple (Acer saccharum Marsh.), the dominant overstory tree in northern hardwood forests of the Lake States region, has a limited capacity to take up and assimilate NO3. However, it is uncertain whether long-term exposure to NO3 deposition might induce NO3 uptake by this ecologically important overstory tree. Here, we investigate whether 10 years of experimental NO3deposition (30 kg N ha−1 y−1) could induce NO3 uptake and assimilation in overstory sugar maple (approximately 90 years old), which would enable this species to function as a direct sink for atmospheric NO3 deposition. Kinetic parameters for NH4+ and NO3 uptake in fine roots, as well as leaf and root NO3 reductase activity, were measured under conditions of ambient and experimental NO3 deposition in four sugar maple-dominated stands spanning the geographic distribution of northern hardwood forests in the Upper Lake States. Chronic NO3 deposition did not alter the V max or K m for NO3 and NH4+ uptake nor did it influence NO3 reductase activity in leaves and fine roots. Moreover, the mean V max for NH4+ uptake (5.15 μmol 15N g−1 h−1) was eight times greater than the V max for NO3 uptake (0.63 μmol 15N g−1 h−1), indicating a much greater physiological capacity for NH4+ uptake in this species. Additionally, NO3 reductase activity was lower than most values for woody plants previously reported in the literature, further indicating a low physiological potential for NO3 assimilation in sugar maple. Our results demonstrate that chronic NO3 deposition has not induced the physiological capacity for NO3 uptake and assimilation by sugar maple, making this dominant species an unlikely direct sink for anthropogenic NO3 deposition.  相似文献

A brick-red-pigmented strain (XTM003T) isolated from the Qinghai–Tibet plateau was investigated using a polyphasic taxonomy approach. Phylogenetic analyses based on 16S rRNA gene sequence indicated that the organism belonged to the genus Hymenobacter. The predominant menaquinone was MK7. The major fatty acids included iso-15:0, 16:1w5c and summed feature 3 (C16: 1ω7c and/or C16: 1ω6c). The G+C content of the DNA was 55.8%. In addition, DNA–DNA hybridization studies demonstrated that strain XTM003T had a relatedness value of 50.7% with the phylogenetically most closely related species Hymenobacter norwichensis DSM 15439T. Based on the results of phenotypic characteristics and DNA–DNA hybridization studies, strain XTM003T is considered to represent a novel species, for which the name Hymenobacter tibetensis sp. nov. is proposed. The type strain is XTM003T (=CCTCC AB 207089T=NRRL B-51271T).  相似文献

D2 ions produced in collisions of D ions with relative energies of 2.5–9.2 eV were detected for the first time. It is shown that the effective cross section for this reaction is no less than 1.5 × 10−14 cm2. In two mountain ecosystems at the Alptal research site in central Switzerland, pulses of 15NO3 and 15NH4 were separately applied to trace deposited inorganic N. One forested and one litter meadow catchment, each approximately 1600 m2, were delimited by trenches in the Gleysols. K15NO3 was applied weekly or fortnightly over one year with a backpack sprayer, thus labelling the atmospheric nitrate deposition. After the sampling and a one-year break, 15NH4Cl was applied as a second one-year pulse, followed by a second sampling campaign. Trees (needles, branches and bole wood), ground vegetation, litter layer and soil (LF, A and B horizon) were sampled at the end of each labelling period. Extractable inorganic N, microbial N, and immobilised soil N were analysed in the LF and A horizons. During the whole labelling period, the runoff water was sampled as well. Most of the added tracer remained in both ecosystems. More NO3 than NH4+ tracer was retained, especially in the forest. The highest recovery was in the soil, mainly in the organic horizon, and in the ground vegetation, especially in the mosses. Event-based runoff analyses showed an immediate response of 15NO3 in runoff, with sharp 15N peaks corresponding to discharge peaks. NO3 leaching showed a clear seasonal pattern, being highest in spring during snowmelt. The high capacity of N retention in these ecosystems leads to the assumption that deposited N accumulates in the soil organic matter, causing a progressive decline of its C:N ratio.  相似文献

A confined aquifer in the Malm Karst of the Franconian Alb, South Germany was investigated in order to understand the role of the vadose zone in denitrifiaction processes. The concentrations of chemical tracers Sr2+ and Cl and concentrations of stable isotope 18O were measured in spring water and precipitation during storm events. Based on these measurements a conceptual model for runoff was constructed. The results indicate that pre-event water, already stored in the system at the beginning of the event, flows downslope on vertical and lateral preferential flow paths. Chemical tracers used in a mixing model for hydrograph separation have shown that the pre-event water contribution is up to 30%. Applying this information to a conceptual runoff generation model, the values of 15N and 18O in nitrate could be calculated. Field observations showed the occurence of significant microbial denitrification processes above the soil/bedrock interface before nitrate percolates through to the deeper horizon of the vadose zone. The source of nitrate could be determined and denitrification processes were calculated. Assuming that the nitrate reduction follows a Rayleigh process one could approximate a nitrate input concentration of about 170 mg/l and a residual nitrate concentration of only about 15%. The results of the chemical and isotopic tracers postulate fertilizers as nitrate source with some influence of atmospheric nitrate. The combined application of hydrograph separation and determination of isotope values in 15N and 18O of nitrate lead to an improved understanding of microbial processes (nitrification, denitrification) in dynamic systems.  相似文献

Wide-compatibility (WC) is one of the most important traits in rice, which can overcome the fertility barrier in the indica/japonica hybrids, and hence to make it possible to utilize the higher yield potential of inter-subspecific hybrids. The S 5 n gene located on chromosome 6 has been previously reported to be responsible for the wide-compatibility in rice. Here we report the precise location of the S 5 n gene. In the first-pass mapping, the S 5 n gene was restricted within a 200 kb region by using a population of 242 isogenic lines in combination with high-density markers developed in the S 5 region. In the fine mapping, the S 5 region was further saturated with newly developed markers and more isogenic lines (549 in total) were investigated. Eventually, the S 5 n gene was mapped within a 50 kb region delimited by the left marker J13 and the right marker J17. One BAC clone screened from the BAC library of the WC rice variety 02428 covered the whole S 5 region. Sequence analysis of the 50 kb region revealed two candidate genes, coding an aspartyl protease and a hypothetical protein. This result would greatly accelerate both cloning and marker-assisted selection of this important S 5 n gene. Qing Ji and Jufei Lu have contributed equally to this paper.  相似文献

Sodium Nitroprusside (SNP) and S-Nitrosoglutathione (GSNO) differently affect mitochondrial H2O2 release at Complex-I. mM SNP increases while GSNO decreases the release induced by succinate alone or added on top of NAD-linked substrates. Stimulation likely depends on Nitric Oxide ( . NO) (released by SNP but not by GSNO) inhibiting cytochrome oxidase and mitochondrial respiration. Preincubations with SNP or high GSNO (10 mM plus DTE to increases its . NO release) induces an inhibition of the succinate dependent H2O2 production consistent with a . NO dependent covalent modification. However maximal inhibition of the succinate dependent H2O2 release is obtained in the presence of low GSNO (20–100 μM), but not with SNP. This inhibition appears independent of . NO release since μM GSNO does not affect mitochondrial respiration, or the H2O2 detection systems and its effect is very rapid. Inhibition may be partly due to an increased removal of O2.− since GSNO chemically competes with NBT and cytochrome C in O2.− detection.  相似文献

During the last years, an international debate about the concept of “Palaeolithic art” has taken place. On one hand, several specialists have critiqued the use of the concept of “art” in naming the images created by Homo sapiens during the Palaeolithic era. They claim that the use of this term implies the projection of a modern category to a world - that of prehistoric humans - which is completely different from our own. On the other hand, some archaeologists consider that the term “Palaeolithic art” does not imply an anachronistic interpretation of prehistoric representations. In presenting the historical context, which has made such a discussion possible, we consider the causes and effects of this controversy. Firstly, we analyze the traditional interpretation, which considered Palaeolithic images as “works of art”. Secondly, we examine the connections, which can be found between the debate about “Palaeolithic art” and certain polemics, which have arisen from the history of art and anthropological frameworks. Thirdly, we consider the arguments utilized by those who are for and those who are against the term “Palaeolithic art”. Finally, we emphasize the importance of this debate to better understand the categories and the concepts, which determine the scientific practice.  相似文献

A method for determining the lifetime of unstable ions is described. The method is based on measuring the decrease in the ion beam current onto a fixed detector with increasing path length of the ion beam from the ion source to the detector. The measurements performed for D? 2 and HD? molecular ions have shown that their lifetimes are 3.5 ± 0.1 and 4.4 ± 0.1 μs, respectively.  相似文献
