The unique ligands of [Ru(bipy)2(bpda)](PF6)2 (1, BPDA=1,1′-biphenyl-2,2′-diamine) and [Ru(bipy)2(dabipy)](PF6)2 (2, DABIPY=3,3′-diamino-2,2′-bipyridine) are atropisomeric (exhibit hindered rotation about the sigma bonds that connect the two aromatic groups), so the complexes are diasteromeric with conformation isomers possible for the atropisomeric ligands and configurational isomers possible at the metal centers. Only one diastereomer is observed in the solid-state in both cases. The seven- (1) and five-membered (2) chelate ring of dabipy and bpda (the ligand is bound through its pyridyl groups) ligands are δ when the configuration at the metal is Δ. No evidence for atropisomerization is found in solution. For 1, we conclude bpda binds stereospecifically; however, the atropisomerization barrier of dabipy may be sufficiently low for 2 to preclude the observation of diastereomers by low-temperature NMR spectroscopy.  相似文献

Complexes RuCl3(PPh3)L2 (L = MeIm (1a, Im (1b)) and [RuCl2(PPh3)2(bipy)]Cl·4H2O (2) have been synthesized via the ruthenium(III) precursor RuCl3(PPh3)2 (DMA), and characterized, including an X-ray structural analysis for 1a (MeIm = N-methylimidazole, Im = imidazole, bipy = 2,2′-bipyridyl, and DMA = N, N′-dimethylacetamide). Crystals of 1a are monoclinic, space group P21/n, A = 10.5491(5), B = 20.4934(9), C = 12.8285(4) Å, β = 90.166(4)°, Z = 4. The structure, which reveals a mer configuration for the chlorides, and cis-methylimidazoles, was solved by conventional heavy atom methods and was refined by full-matrix least-square procedures to R = 0.041 and Rw = 0.042 for 3328 reflections with I 3σ(I). From the RuCl2(PPh3)3 precursor, the ruthenium(II) complexes RuCl2(PPh3)2L2 and [RuCl(PPh3)L4]Cl have been made (L = Im or MeIm), while [RuCl(dppb)Im3]Cl has been made from [RuCl2(dppb)]2(μ-dppb) (dppb = Ph2P(CH2)4PPh2).  相似文献

Fluorescence characteristics and energy transfer to a cerium complex with 2,2′-bipyridine (bpy) in methanol are described. Stoichiometries and the stability constant of the Ce(III)–bpy complex in methanol were determined by use of the molar ratio method. A fluorescence lifetime and a quantum yield have been measured for the 2:1 complex. Decay times and time-resolved (T---S) emission spectra for Ce(III)–bpy and CeCl3 were measured in methanol at room temperature. An energy transfer rate constant was determined from the luminescence lifetime and the quantum yield. The mechanism of energy transfer from the lowest excited singlet S1 to the 5d level of the Ce(III) in the Ce(III)–bpy complex system is discussed in some detail.  相似文献

We report here the synthesis of a novel series of 5′-O-carbonates of 3TC, using different aliphatic alcohols and N,N-carbonyldiimidazol. Its antiviral activity was determined in peripheral blood mononuclear cells (PBMCs) showing some carbonate derivatives with an activity similar to or better than 3TC, except 3TC-Metha and 3TC-2Pro with less activity. In vitro assays in PBMCs have demonstrated that cytotoxicity increases as the carbon chain length of the alcohol moiety increases, showing compounds with a normal chain length of n = 2–5 good selective index, compared to the parent drug. Thus, this work is an important contribution leading to the suppression of HIV replication.  相似文献

We report herein the synthesis of a novel tetracationic tris(2,2′-bipyridine) ligand 4. We show that this ligand metalated with copper(II), and in the presence of ascorbate as a reducing agent, strongly damages pUC18 plasmid DNA. Copper complex formation was demonstrated by ESI-MS (electrospray ionization-mass spectrum) at a 1:3 ligand to metal ratio. Binding of both 4 and its copper(II) complex to CT-DNA (calf thymus DNA) was characterized by viscosimetry, thermal denaturation and fluorescence-based competition assays. The viscosimetric data indicated that 4 and its copper(II) complex bind DNA through partial intercalation and thermal denaturation studies revealed a significant increase of duplex DNA stability in the presence of these species (ΔTm = 16.4 and 18.3 °C, respectively). Moreover, 4 and its copper(II) complex were found to effectively compete with ethidium bromide for the intercalative binding sites of DNA. Overall, the copper(II)-4 complex constitutes a very efficient DNA cleaving agent in the presence of ascorbate. Experiments with scavengers further suggest that the generation of Cu(I), hydrogen peroxide, superoxide, hydroxyl radical and singlet oxygen-like species contributes to the DNA breakage induced by the Cu(II) complex of 4.  相似文献

The structures of (4-bipyH)2[(μ-4-bipy)Nd2(NO3)8(H2O)4]·3(4-bipy) (4-bipy = 4,4′-bipyridine; P21/c, a = 18.723(10), b = 10.720(6), c = 18.027(10) Å, β = 94.43(5)°, Z = 2; R = 0.066 for 4931 (diffractometer data) and of a second monoclinic form of [Ho(NO3)3(H2O)3]·2(4-bipy) (P21/c, a = 15.830(10), b = 21.44(3), c = 15.70(3) Å, β = 100.4(2)°, Z = 8; R = 0.091 for 2335 film data) are reported. In the first compound pairs of Nd atoms are bridged across a crystal inversion centre by a 4-bipy ligand, and 10-coordination is completed by one monodentate NO3, three bidentate NO3, and two H2O ligands, with bond lengths Nd---N 2.70, Nd---OH2(av.) 2.44, Nd---O(NO3, av.) 2.56 Å. The second compound has a variant of the previously-reported monoclinic [Y(NO3)3(H2O)3]·2(4-bipy) structure, with doubling of the unit cell on a but with essentially no change in the geometry and orientation of the nine-coordinate complex. In both compounds the non-coordinated, non-protonated 4-bipy N atoms form hydrogen bonds with ligand H2O.  相似文献

Novel N,O-acyl chitosan (NOAC) derivatives were synthesized to examine their fungicidal activity against the gray mould fungus Botrytis cinerea (Leotiales: Sclerotiniaceae) and the rice leaf blast fungus Pyricularia oryzae (Teleomorph: Magnaporth grisea). The fungicidal activity was evaluated by the radial growth bioassay. NOAC derivatives were more active against the two plant pathogens than chitosan itself, and the effect was concentration dependent. Against B. cinerea, 4-chlorobutyryl chitosan (EC50=0.043%), decanoyl chitosan (EC50=0.044%), cinnamoyl chitosan (EC50=0.045%), and p-methoxybenzoyl chitosan (EC50=0.050%) were the most active (12-13-fold more active than chitosan). (Un)-substituted benzoyl chitosan derivatives were more active against B. cinerea than most of these with N,O-alkyl derivatives. Against P. oryzae chitosan derivatives with lauroyl, methoxy acetyl, methacryloyl and decanoyl were the most active.  相似文献

The concentration of the ruthenium-based label is determined from the rate of hydrogen peroxide production elicited by photolysis. Electron transfer quenching of the photoexcited label by methyl viologen (1,1′-dimethyl-4,4′-bipyridinium dication, MV2+) and/or oxygen in the presence of EDTA generates hydrogen peroxide. Both flow injection and direct photolysis techniques were tested, with the latter showing better results. Direct photolysis is more sensitive, faster, requires only a 20 μl sample volume, uses only 30 mW laser power and shows a smaller background. The presence of 5% normal human serum in the sample did not interfere with the measurements. Linear calibration curves were obtained in the nanomolar concentration range for goat antimouse antibody labeled with the ruthenium complex. The determination of membrane-surface-bound labeled IgG is accomplished by direct photolysis of a membrane that covers a platinum microelectrode.  相似文献

The complex [Eu(tpy)3](ClO4)3 where TPY=2,2′; 6,2″-terpyridine, has been prepared and reexamined. The complex appears to be stable in acetonitrile solution with respect to decomplexation of the ligands but the addition of water does cause partial replacement of tpy. Analogous complexes have been prepared with 3,3′; 5,3″-polymethylene bridged derivatives of tpy having two or three carbons in the bridge. The bridging enforces a cisoid geometry of the ligand and prohibits its replacement by added water. An X-ray determination was carried out for [Eu(3b)3](ClO4)3, where 3b=3,3′; 5,3″-dimethylene tpy, which crystallizes in the monoclinic space group P21/c with a=11.908(4), b=15.768(5), c=29.513(9) Å, β=93.60(2)°, μ=13.5 cm−1 and Z=4. The complex forms a tricapped trigonal prism with each of the ligands adopting the same dl conformation. Variable temperature NMR analysis of the bridged ligand complexes indicates that conformational inversion of the bound ligand is not a concerted process and barriers for inversion of individual methylene units can be estimated from coalescence of the signals from the geminal methylene protons. The luminescence properties of the bridged tpy complexes are similar to the parent unbridged system.  相似文献

Pyrazole constitutes an important heterocyclic family covering a broad range of synthetic as well as natural products that exhibit numerous chemical, biological, agrochemical and pharmacological properties. In order to explore compounds with good fungicidal activity, a series of new pyrazole derivatives containing 5-phenyl-2-furan were designed and synthesized. In vitro and in vivo fungicidal activities were evaluated and the compound ethyl-1-(5-phenylfuran-2-carbonyl)-5-propyl-1H-pyrazole-3-carboxylate (I8) displayed significant fungicidal activity against various fungi, especially against P. infestans. The structures of the novel pyrazole derivatives were confirmed by 1H NMR, 13C NMR, MS, elemental analysis and X-ray single crystal diffraction. Further study showed that compound I8 might act on the synthesis of cell walls from morphological and ultrastructural studies by SEM and TEM. The results also revealed that compound I8 could block the nutritional transportation leading to cells senescence and death. These results suggested that the novel pyrazole derivatives proved to be promising lead compounds.  相似文献

Synthesis of N- and O-acyl derivatives of DL-serine and threo-DL-phenylserine was accomplished by a regioselective acylation of the corresponding amino acid. The residues introduced into amino acid structure contain hydrophobic long chain or aromatic, namely lauroyl, myristoyl and phenylacetyl moieties. The fungicidal activity against six strains of fungi was studied. Several compounds were found to be effective against growth of fungi, and O-myristoyl-DL-serine 2 and N-phenylacetyl-threo-DL-phenylserine 8 completely inhibited the growth of the mycelium of the fungus Verticillium dahliae.  相似文献

Non-enzymatic reactions of the 3,4-oxide of 2,2′,5,5′-tetrachlorobiphenyl (TCB) with methionine or N-acetylmethionine in ethanol/neutral buffer at 37°C proceeded very slowly to yield an approx. 1 : 1 ratio of 3- and 4-methylthio-TCB. Under similar conditions reaction of TCB 3,4-oxide with cysteine proceeded about 100 times more rapidly to yield an approx. 1 : 1 ratio of 3- and 4-(cystein-S-yl)-TCB as the major products. Cystein-S-yl-3,4-dihydro-hydroxy-TCB(s) was also formed as a minor product from reaction of TCB 3,4-oxide with cysteine in dimethyl sulfoxide/neutral buffer. TCB 3,4-oxide did not react detectably with glutathione in ethanol/neutral buffer at 37°C or 70°C, but reaction in ethanol/pH 8.7 buffer at 37°C proceeded very rapidly to yield about a 1 : 1 ratio of 3- and 4-(glutathion-S-yl)-TCB and of two glutathion-S-yl-TCB precursors. Glutathion-S-yl-TCB(s) and its precursor(s) were also formed rapidly in a rat liver cytosol-catalyzed reaction of TCB 3,4-oxide with glutathione at neutral pH. The glutathion-S-yl-TCBs readily degraded upon concentration in aqueous alcohol solutions under mild conditions to yield compounds tentatively identified as [N-(5-carboxy-1-pyrrolin-2-yl)-1-glycinocystein-S-yl]-TCBs, (1-glycinocystein-S-yl)-TCBs and 2-oxopyrrolidine-5-carboxylic acid.

Rats given a single dose of TCB excreted about 0.07% of the dose in the feces during the first 4 days as 3-methylthio-TCB, 4-methylthio-TCB, 4-methylsulfonyl-TCB, methylthio-hydroxy-TCBs (tentatively identified) and mercapto-TCB(s) (tentatively identified) in about a 1 : 5 : 0.1 : 0.1 : 0.05 ratio, respectively. Rats given an equimolar dose of TCB 3,4-oxide excreted similar ratios of these fecal metabolites in approx. 10-fold greater quantities. Mice given TCB excreted about 0.1% of the dose in the feces during the first 4 days as 3-methylthio-TCB, 4-methylthio-TCB and 3-methylsulfonyl-TCB in about a 1.5 : 1 : 0.05 ratio, respectively. Methylthio-TCBs were not detected (<0.0004% of the dose) in the bile of a cannulated rat given a single dose of TCB. About 1.5% of the TCB dose was excreted in the bile as glutathion-S-yl-TCB(s) and its precursor(s). Collectively, the data indicate that TCB 3,4-oxide is a primary metabolic intermediate in the formation of methylthio-metabolites of TCB.  相似文献

We synthesized a family of 3,5-dichloropyrazin-2(1H)-one derivatives and assessed their in vitro fungicidal activity against Candida albicans. Compounds 11 and 20 were most active against C. albicans and induced accumulation of reactive oxygen species in this pathogen. Using a genome-wide approach in the yeast Saccharomyces cerevisiae, we demonstrated that genes involved in vacuolar functionality and DNA-related functions play an important role in cellular mechanisms underlying the fungicidal activity of these compounds.  相似文献

A new series of 1β-methylcarbapenems 1a–i bearing isoxazoloethenyl groups on the pyrrolidine ring has been prepared and evaluated for in vitro antibacterial activity and stability to DHP-I. Most compounds showed excellent antibacterial activity and high stability to DHP-I superior to that of meropenem. Of these new carbapenems, 1a,b,h exhibited the best combination of antibacterial activity and DHP-I stability.  相似文献

3′-Fluoro-3′-deoxy-uridine, -cytidine, -adenosine and -guanosine have been synthesized by glycosylation of the corresponding silylated bases with 1-O-acetyl-2,5-di-O-benzoyl-3-fluoro-3-deoxy-D-ribofuranose in the presence of Friedel-Crafts catalysts and were converted to the 5′- triphosphates, NTP(3′-F). It was shown that NTP(3′-F) are terminators of RNA synthesis catalyzed by DNA-dependent RNA polymerase from E. coli and may thus serve as tools for DNA sequencing.  相似文献

The disulfonic stilbene (4-acetamido-4′-isothiocyano-2,2′-disulfonic stilbene) is found to be more potent than acetazolamide as an anion transport inhibitor in the turtle bladder, but less potent than acetazolamide as a carbonic anhydrase inhibitor. The anion-dependent (HCO3-−, Cl) moeity of the short-circuiting current is eliminated by 4-acetamido-4′-isothiocyano-2,2′-disulfonic stibene, but only after its addition to the serosal bathing fluid. Whereas 4-acetmido-4′-isothiocyano-2,2′-disulfonic stilbene has no effect om Na+transport across the bladder, it is more potent than ouabain as an inhibitor of microsomal (Na++K+)-ATPase of both turtle bladder and eel electric organ.  相似文献
