Eight hundred second chromosomes were extracted from the Ishigakijima population, one of the southernmost populations of Drosophila melanogaster in Japan. Half of them were extracted in Native cytoplasm (P-type), and half in Foreign cytoplasm (M-type). Various population-genetic parameters, including the frequency of lethal-carrying second chromosomes (Q = 0.235 for the Native; 0.218 for the Foreign), the allelism rate of lethal second chromosome (Ic = 0.0217 for the Native; 0.0134 for the Foreign), the homozygous detrimental and lethal loads (D = 0.179 for the Native; 0.270 for the Foreign; L = 0.262 for the Native; 0.240 for the Foreign), the average degree of dominance of mildly deleterious mutations (?E = 0.244 for the Native; 0.208 for the Foreign), and the components of genetic variance for viability [additive (sigma A2) and dominance (sigma D2)](?igma A2 = 0.0187 for the Native; 0.0172 for the Foreign; ?igma D2 = 0.0005 for the Native; 0.0009 for the Foreign) were estimated. The data indicate that D was significantly larger and hE was significantly smaller in the Foreign cytoplasm. However, the estimates of additive and dominance variances were not significantly different between the two cytoplasms. The additive genetic variance for viability in the Ishigakijima population was greater than expected on the basis of mutation-selection balance confirming previous studies on papers of D. melanogaster in warm climates.  相似文献

Biofuels composed by fatty acid methyl esters are widely used as partly substituting fuels for diesel fossil fuels. Additionally, it is expected that the diesel biofuel norms will be extended to ethyl esters produced from bioethanol in the upcoming years. A precise knowledge of the standard enthalpy of formation is necessary for the calculation of some parameters useful for the analysis of the combustion process and emissions of a diesel engine operating with different fuels, such as the heating value, the adiabatic flame temperature or the kinetic mechanisms. However, experimental data for this property are scarce, and only available for short-chain, saturated methyl esters. In this work, four estimation methods for the calculation of the enthalpy of formation are examined and compared. Three of them are simple methods based on groups or bonds contribution, and another one is a computational method (with Gaussian 03 software). After presenting the implementation rules for each of them, conclusions are stated based on the results attained. Gaussian and Benson-Groups methods seem to be more accurate in predicting the actual values of the enthalpy of formation, both methods considering the separation between double bonds and the edge effects in the molecule. However, only the Gaussian method considers the effect of the position of the double bond in the molecule for all the unsaturated esters.  相似文献

目的:探讨死胎的病因及终止妊娠的方式。方法:收集我院住院分娩的死胎病例23 例,对孕妇一般资料、死胎发生原因、终 止妊娠方式进行统计分析。结果:①死胎发生孕妇中,年龄>36 岁者最多,共8 例,占34.8%,孕周在28-36+6 周的孕妇数最多,共 10 例,占43.5%,初产妇17 例,占73.9%,有流产史孕妇9 例,占39.1%;②在所有病因中,脐带因素占比最大,共11 例,占47.8%, 其次为胎盘因素和母体因素,各5 例,各占21.7%,胎儿原因与不明原因各2 例,各占8.7%;③在对终止妊娠方式的选择上,施行 羊膜腔穿刺引产例数最多,共13 例,占56.5%,其次为剖宫产,共5 例,占21.7%,米索流产3 例,占13.0%,自娩患者2 例,占 8.7%。结论:脐带因素为造成死胎发生的主要原因,其次为胎盘因素及母体因素,应对孕妇进行全面的孕期检查及孕期健康教育, 以减少死胎的发生,并选择合适的终止妊娠方式以减轻孕妇痛苦。  相似文献

The methods used for the sparing inactivation of highly concentrated R. prowazekii biomass and for the decrease of its infectious activity are described. These methods are recommended for use in experiments in the field of molecular biology, as well as for disinfection of different materials contaminated with rickettsiae. As conditions for complete inactivation, incubation at 50 degrees C for 1 hour without chemical disinfectants, treatment with 0.5% phenol solution at 30 degrees C for 12 hours and with 0.1% formaldehyde solution at 4 degrees C for 24 hours have been selected. Treatment with 0.5% phenol solution at 36 degrees C for 1 hour or incubation at 45 degrees C without the use of disinfectants ensures an essential decrease in the infectivity of the material if the work with viable infective agents is necessary. Ultraviolet irradiation for 1.5 hours and exposure to the action of 0.1-0.5% sodium azide are less effective.  相似文献

The otic labyrinth presents one of the most difficult objects for histological investigations as it is situated in the thick layer of the temporal bone and has a very complex structure. At the same time, a method for treating this object is not given in general hand-books on pathological techniques. The methods for histological treatment of the otic labyrinth elaborated by Wittmaack and later on modified are presented in the article. Methods for fixation, vital fixation including, decalcination, dehydratation and saturation of the objects prepared for histological investigation are described in details. Methods for the temporal bone orientation in order to obtain sections of the internal otic structures in the most advantageous plane depending on the purpose and aim of the investigation are recommended.  相似文献

Prevalence rates (PRs) for EFP (schizophrenic, schizoaffective and affective psychoses), with allowance for proband sex and age-of-onset data were studied in a subdivided population from the North-East of the European Region of the USSR. The population includes three subpopulations: a small old religious semi-isolate of Russians ("Rs"), aboriginal Komi people ("Ks")--an ethnic community of Ugro-Finnish lineage, and a mixed group of migrants ("Ms") from various regions of the USSR. The latter is mainly an urban population, while the "Rs" and "Ks" are, on the whole, rural populations. The total PR for EFP was found to be 0.97% for the "Rs", 0.63% for the "Ks" and 0.35% for the "Ms", whereas PRs-0.85-1.15% in other parts of the USSR, mainly for "panmixed" populations in large towns. The lower PRs for EFP in the "Ms" is caused by a backmigration flow involving certain groups of patients; consequently, the mean liability for "Ms" offsprings (as a whole) should also be lower. On the other hand, the lower PRs for EFP in the "Ks" is caused by underpresentation of clinically mild cases of the mental disease (mainly, pseudoneurotic schizophrenia), especially among female patients, probably due to that the so affected persons are sufficiently adapted to the cultural traditions of this rural population. It was shown that in the "Rs" the total PR for "nuclear" and paranoid schizophrenia is 0.68% versus 0.25% in a "panmixed" population. The increase is most likely caused by the high inbreeding level in the "Rs" semi-isolate, and if this is correct, we may suppose that at least one or two recessive genes are involved in the liability to the most heavy forms of schizophrenia. On the other hand, in the "Ms" (as in other "panmixed" populations) positive assortative mating among hereditary-predisposed persons is a more significant factor influencing family transmission of EFP, since the correlation between probands and their spouses is rpp = 0.31 (p less than 0.001) in the "Ms", as compared to rpp = 0.19 (p less than 0.1) in the "Rs". Thus, our general conclusion is that neither the place of inhabitance nor the life mode are the causal factors for EFP, but rather some genetic factors, more accurately, certain sets of specific genes.  相似文献

Crow's indices of the opportunity for selection and their components connected with differential mortality (Im) and differential fertility (If) were estimated for populations of Soviet Union republics and for a number of USSR ethnic groups on the basis of demographic statistics. More than 10-fold decrease in the Im value was revealed in the total population of the USSR during 1926-1987. At present, the Im values in republics vary from 0.020 to 0.094 for urban population and from 0.030 to 0.121 in rural population, the ratio of perinatal mortality in the whole structure of prereproductive mortality being higher in the republics with lower values of the Im. The range of the If values for different peoples (0.148-0.643) is wider than for the populations of the republics (0.326-0.578). Interethnic differences contribute 47% of the variance in fertility. The structure of Crow's indices is given for urban and rural populations of the republics. Genetic implications of the data presented are discussed with respect to possible manifestation of the effects of inter-group selection.  相似文献

A new method for assessing the efficiency of batteries of arbitrary numbers of tests is proposed. The posterior probability of the mutagenicity of the substances studied has been estimated using discriminant analysis. The results of tests in each test system has been presented as the probability to obtain a positive result in the given test system. This has made it possible to decrease the sample size as the number of tests in the battery increased. As a result, prognostic power may be assessed even if the matrix of results is incomplete. This approach has been used to estimate the weights of evidence for mutagenic activities of 105 chemical compounds studied by means of a battery of four tests: Ames's test, the test for chromosome aberrations in vitro, the test for cytogenetic defects in vivo, and the test for dominant lethal mutations in rodents.  相似文献

PicoGreen is a very sensitive fluorescent dye for quantitative assays of double-stranded DNA (dsDNA) in solution and is used in several analytical protocols in which sensitive and precise DNA detection is needed, also for examination of drug-DNA interactions. The data shown in this paper indicate that compounds intercalating to DNA influence the applicability of PicoGreen dye for quantitative measurements of dsDNA, and for this reason PicoGreen dye is not suitable for examination of drug-DNA interactions, especially interstrand DNA crosslinks.  相似文献

An ethnobotanical study was conducted among the Jicaque Indians of the Montana de la Flor reservation in central Honduras to collect information regarding their plant-use practices. The data, including vernacular names, ecological settings, and uses, were recorded during collecting forays with Jicaque informants. Scientific and Jicaque names have been provided for 23 plant species used for food, 11 used for medicine, 4 used for beverages, 3 used as sources of wood, 5 in artifact manufacturing, and 3 used in other ways. Also, a list of 43 species is included for which the Jicaques have a name but no known use.  相似文献

When Triton-filled lysosomes from rat liver are incubated for up to 50min at 37 degrees C, pH7.4, in 0.25m-sucrose, no loss of latency of N-acetyl-beta-glucosaminidase or p-nitrophenyl phosphatase occurs unless the incubated lysosomes are cooled to approx. 15 degrees C. 2. It is suggested that a phase change takes place in the incubated lysosomal membranes on cooling; it starts at approx. 15 degrees C and probably is not complete at 0 degrees C. 3. Incubation of the lysosomes causes an increased potential for loss of latency of the lysosomal enzymes. This potential is not fully expressed at elevated temperature (e.g. 37 degrees C), but is expressed on cooling. 4. The increase at elevated temperature in potential for loss of latency exhibits biphasic kinetics, with an initial rapid phase followed by a slower phase, which is linear with respect to time. The extra loss of latency resulting from the rapid phase in proportional to the temperature of the incubation. 5. Arrhenius plots of the increase is potential for loss of latency during the slow phase for N-acetyl-beta-glucosaminidase and p-nitrophenyl phosphatase exhibit marked deviations from linearity beginning at approx. 15 degrees C. This suggests that the increase in potential for loss of latency is affected by a phase change that occurs around this temperature. 6. Activation energies for the increase in potential for loss of latency at and above 22 degrees C are 53.1+/-5.4kJ/mol (12.7+/-1.3kcal/mol) for N-acetyl-beta-glucosaminidase and 45.2+/-7.5kJ/mol (10.8+/-1.8kcal/mol) for p-nitrophenyl phosphatase. It is postulated that these energies reflect enzymic action, the products of which cause loss of latency to occur on cooling.  相似文献

We are currently investigating the morphologic aspects of two areas of the basic cell biology of cancer: tumor-specific surface antigens as targets for immunotoxins, and the phenomenon of multidrug resistance in chemotherapy of human tumors. Colloidal gold cytochemistry has provided a useful method for the electron-microscopic cytochemical detection of materials endocytosed by cells in culture. This technique has been used to study the internalization pathway of ligands bound to the surface of cancer cells, particularly antibodies for use as immunologic targeting reagents for the construction of immunotoxins. These colloidal gold conjugates with monoclonal antibodies have demonstrated the internalization of these immunologic reagents through coated pits and receptosomes, which is a necessary step in the delivery of immunotoxins into the cell where they can mediate their cell-killing functions. Morphologic methods have been employed for the screening and selection of monoclonal antibodies reactive with the surface of human ovarian cancer cells for use as immunotoxins and have demonstrated the in vivo activity of immunotoxins made with these antibodies and Pseudomonas exotoxin in a nude mouse model system. In other studies, we have employed such reagents for the immunocytochemical detection of the surface expression of P170, the cell-surface efflux pump protein responsible for the phenotype of multidrug resistance in tumor cells, and to investigate the distribution of this protein by using immunocytochemistry in normal human tissues. These results have suggested a role for P170 in normal cell membrane transport of metabolites in various organ systems.  相似文献

The EU Tissues and Cells Directive (2004/23/EC, 2006/17/EC, 2006/86/EC) (EUTCD) provides standards for quality and safety for all aspects of banking of tissues and cells for clinical applications. Commission Directive 2006/17/EC stipulates that the complete donor record with all the medical information is assessed for suitability before releasing tissues for clinical use. The aim of this study was to investigate the medical reasons for post-procurement donor exclusion, to identify the various potential sources for gathering information about donors’ medical and behavioural history and to evaluate their contribution to maximising the safety of donations. Information was collected from the Tissue Services (TS) records of 1000 consecutive deceased donors submitted to National Health Service Blood and Transplant (NHSBT) medical officers for authorisation for release for subsequent tissue processing and then for transplantation. Of the 1000 donors 60 (6%) were excluded because they did not fulfil the donor selection requirements of the EUTCD and NHSBT donor selection guidelines. The main reasons for medical exclusion were the presence of significant local or systemic infection in 32 donors (53% of those excluded for medical reasons) and a history of past or occult malignancy in 9 donors (15% of those excluded for medical reasons) which was not identified prior to procurement. The information leading to post-procurement exclusion was obtained from autopsy reports in 35 of the 60 excluded donors for medical reasons (58%) and from the general practitioner for 10 donors (17% of those excluded for medical reasons). In summary, careful evaluation of complete donor records reduces the potential risk of disease transmission by tissue allografts and ensures compliance with regulations and guidelines. The findings may lead to changes in donor selection policies with the aim of improving efficiency without compromising safety.  相似文献

Sporulation-specific mutations in Bacillus subtilis have been mapped by transduction and transformation. The mutations caused blocks at stages 0, II, III, and IV of sporulation; more than one phenotype was found for each of these stages. On the basis of the criteria used to define a sporulation operon, a minimum estimate could be made of the number of operons activated during sporulation. Nine operons were identified for stage 0, eight for stage II, five for stage III, and six for stage IV. It is probable that several of these 28 operons are activated in groups so that the number of steps in the dependent sequence of sporulation events should turn out to be less than the number of operons.  相似文献
